RESUMO A jurubeba (Solanum paniculatum L.) é uma planta usada na medicina tradicional, para problemas hepáticos, além de ser empregada como colagoga, emenagoga, cicatrizante, febrífuga, anti-inflamatória, antipirética, tônica, descongestionante, diurética e na inapetência. Assim, objetivou-se com o presente estudo avaliar a atividade antioxidante, os teores de compostos fenólicos e de alcaloides totais dos extratos aquosos preparados a partir de folhas de jurubeba submetidas à secagem. As folhas de jurubeba foram secas em diferentes temperaturas (40, 50, 60 e 70 °C) e velocidades (0,4 e 0,8 m s-1) do ar de secagem. Os extratos aquosos das folhas, submetidas a secagem, foram avaliados por meio de leitura de absorbância em espectrofotômetro. O aumento da temperatura do ar de secagem reduziu o teor de compostos fenólicos e alcaloides totais dos extratos aquosos das folhas, enquanto que o aumento da velocidade do ar aumentou o teor dessas substâncias, já a atividade antioxidante não foi influenciada pelo incremento da temperatura do ar na velocidade de 0,4 m s-1, mas sob a velocidade do ar de 0,8 m s-1 houve redução com o aumento da temperatura. A secagem das folhas de jurubeba pode ser realizada com temperatura de 40 ou 50 °C e velocidade do ar de secagem de 0,4 m s-1.
ABSTRACT ‘Jurubeba’ (Solanum paniculatum L.) is a medicinal plant used in traditional medicine for liver problems, in addition to being used as a cholagogue, emmenagogue, healing agent, febrifuge, anti-inflammatory, antipyretic, tonic, decongestant, diuretic and against inappetence. Thus, the objective of this study was to evaluate the antioxidant activity, concentration of phenolic compounds and total alkaloids of aqueous extracts prepared from ‘Jurubeba’ leaves subjected to drying. ‘Jurubeba’ leaves were dried at different temperatures (40, 50, 60 and 70 °C) and velocities (0.4 and 0.8 m s-1) of the drying air. Aqueous extracts of the leaves, subjected to drying, were evaluated by absorbance reading in a spectrophotometer. Increase in drying air temperature reduced the contents of total phenolics and total alkaloids of the leaf aqueous extracts, whereas increment in air velocity increased the concentration of these substances. Antioxidant activity was not influenced by the increase in air temperature at air velocity of 0.4 m s-1, but under air velocity of 0.8 m s-1 there was reduction with increasing temperature. Drying of ‘Jurubeba’ leaves can be carried out with temperature of 40 or 50 °C and drying air velocity of 0.4 m s-1.

RESUMO Dentre as inúmeras plantas medicinais utilizadas pela população brasileira está a Solanum paniculatum L. (jurubeba), em que o uso de suas folhas é bastante popular, na forma de chá, como remédio contra ressacas após o consumo exagerado de álcool e comida, além de serem usadas na forma de pomadas como cicatrizantes, e, também, no tratamento de problemas hepáticos e digestivos. As folhas de plantas medicinais, normalmente, possuem elevado teor de água, necessitando a redução do mesmo por meio da secagem, para que o produto possa ser armazenado com segurança até o seu processamento. Objetivou-se com o presente estudo avaliar a cinética de secagem de folhas de jurubeba, bem como determinar o coeficiente de difusão efetiva e a energia de ativação durante a secagem. As folhas de jurubeba foram submetidas a secagem em diferentes temperaturas (30, 40, 50, 60 e 70 °C) e velocidade do ar (0,4 e 0,8 m s-1) . O modelo de Midilli ajustou-se satisfatoriamente aos dados observados da secagem das folhas de jurubeba para todas as condições. O aumento da temperatura e da velocidade do ar de secagem reduziu o tempo de secagem e aumentou o coeficiente de difusão efetiva. Para a faixa de temperatura de 40 a 70 °C, ao aumentar a velocidade do ar de secagem, reduz-se a energia de ativação para a secagem das folhas de jurubeba.
ABSTRACT Solanum paniculatum L. (jurubeba) is among the numerous medicinal plants used by the Brazilian population, and the use of its leaves is quite popular, in the form of tea, as a remedy against hangovers after excessive consumption of alcohol and food, besides being used in the form of ointments as healing agent and also in the treatment of liver and digestive problems. Leaves of medicinal plants usually have a high moisture content, requiring its reduction by drying, so that the product can be stored safely until its processing. The objective of this study was to evaluate the drying kinetics of jurubeba leaves and to determine the effective diffusion coefficient and activation energy during drying. Jurubeba leaves were dried at different air temperatures (30, 40, 50, 60 and 70 °C) and speed (0.4 and 0.8 m s-1). The Midilli model satisfactorily fitted to the observed data of drying of jurubeba leaves for all air conditions. Increasing the drying air temperature and speed reduced the drying time and increased the effective diffusion coefficient. For the air temperature range from 40 to 70 °C, as the drying air speed increases, the activation energy for the drying of the jurubeba leaves is reduced.

Piper glabratum is a plant native from South America, which has been little studied, despite its use in folk medicine. In this study, chemical composition, antioxidant and antifungal activities of essential oils (EOs) from leaves of P. glabratum were investigated. The influence of seasonality on its chemical composition was also evaluated. The chemical characterization was performed by two-dimensional gas chromatography coupled with quadrupole mass spectrometry (GC×GC/qMS). Altogether, 199 compounds were identified in the EOs and their composition varied during the seasonal cycle. The main compounds found were sesquiterpenes and their oxygenated analogues. EOs showed antioxidant activity, measured by 2,2-diphenyl-1-picrylhydrazyl-DPPH (from 28.1 to 33.4 µg mL-1) and β-carotene assays (from 57.4 to 66.4%), and inhibitory action against C. albicans. The oils provided expressive properties; however, the yield of EO from winter was not enough for activities assays, remarking the influence of the seasonality on the biological activities of this plant.

The methodology developed in this study through the acquisition of fingerprint chromatograms by liquid chromatography-diode array detector (LC-DAD), aided by principal component analysis, made it possible to assess, in a rational way, the chemical differences between six accessions of Lippia alba and to verify the influence of the method of extraction, the seasonality and the individuality of each accession on chemical composition of its extracts. Among all extracts analyzed against cancer cell lines, eight of them showed to be more promising against a human leukemia cell line (HL-60), displaying cell growth inhibition percentage ranging between 40.0 and 52.0%. In the inhibitory activity assays against acetylcholinesterase enzyme, all extracts showed weak inhibitory effect, with highlight only for six of them, which displayed inhibition ranging between 27.0 and 32.0%.

Electrochemical methods are powerful in the characterization and design of redox-modulating agents. We, herein, report the electrochemical investigation, in aprotic medium, of eleven synthetic 3-thio-substituted-nor-beta-lapachones, along with the determination of cytotoxic activity and correspondent selectivity index, against several cancer cell lines and one normal cell. Four of the quinones are novel compounds. The redox behavior is representative of two independent systems: the easy reduction of the quinone moiety and, at far more negative potential, the reductive cleavage of the C-S-C bonding; and the anodic part controlled by the oxidation of the sulfur moiety. The compounds have shown relevant cytotoxic activity, with emphasis on 3-phenyl-thio-2,2-dimethyl-2,3-dihydronaphtho[1,2-b]furan-4,5-dione (compound 2), which mechanism of molecular action was shown to be related to reactive oxygen species (ROS) release. Despite the absence of a linear correlation, there is a trend: the majority of the thionaphthoquinones, with values of first wave reduction potential, less negative than -0.65 V, were active. The less electrophilic compound (3-(cyclohexylthio)-2,2-dimethyl-2,3-dihydronaphtho[1,2-b]furan-4,5-dione, cyclohexyl derivative) is also the less cytotoxic toward cancer cells. Agents containing chalcogens and quinones can be used to attack entities with a disturbed redox balance.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Inflammatory and genetic alterations are related to the development of chronic diseases such as cancer. Schinus terebinthifolius Raddi, Anacardiaceae, is used in folk medicine to treat inflammation, wounds and tumors. This study evaluated the anti-inflammatory, immunomodulatory, chemopreventive, and wound healing potentials of the methanolic extract from the leaves of Schinus terebinthifolius. The chemical composition of the extract was characterized using preliminary analytical LC methods. The results showed that the anti-inflammatory activity of the methanolic extract was similar to that of dexamethasone for edema reduction. Also, it inhibited the leukocyte migration into the air pouch and decreased plasma extravasation. In addition, the methanolic extract showed a healing action similar to that observed with collagenase. The methanolic extract is not genotoxic nor mutagenic, and in contrast it has chemopreventive activity, which elicits a high percentage of damage reduction by comet and micronucleus assay, preferably by bioantimutagenic action. The methanolic extract induced apoptosis and enhanced splenic phagocytosis in animals treated with cyclophosphamide. The methanolic extract contents, resolved by LC, include phenolic acid and flavonoids. Our results suggest a therapeutic potential for the methanolic extract.

Achatina fulica's secretion has been recently related to antibacterial, antifungal, and cicatricial properties, and it is influenced by the kind of food offered. Therefore, this study investigated the healing potential of dressing films based on mucous secretion of Achatina fulica. Thus, surgical wounds performed in black wistar rats were dressed with films based on collagen, and on mucous secretion of A. fulica fed with Lactuca sativa; undresses worked as wounds control. After 3, 7, 14 and 21 days the animals were euthanized, and the wounds were microscopically evaluated. On the 3rd and 7th days dressing films based on mucus provided acceleration of the formation maturation of granulation tissue, better epithelization rates, and more rapid replacement of type III for type I collagen fibers. On the 14th and 21st days, these dressings induced more intense deposition and better architectural disposition of type I collagen fibers, and hastened the regeneration of cutaneous phaneros. Dressings obtained from A. fulica fed with Lactuca sativa provided the most expressive results. This study suggests that films produced with mucous secretion of A. fulica can be successfully employed as wound dressing, particularly if snails are fed with Lactuca sativa.
A la secreción mucoproteica del Achatina fulica se le han asignado propiedades antibacterianas, antifúngicas y cicatriciales que pueden estar influidas por el tipo de alimento que se ofrece a este especimen. Este estudio investigó el potencial curativo de películas basadas en la secreción mucosa de Achatina fulica alimentaba con diferentes tipos de plantas. Fueron tratadas heridas provocadas en el dorso de ratas Wistar con películas de colágeno y películas realizadas a partir de la secreción mucosa de A. fulica, alimentados con plantas de lechuga (L. sativa), utilizando como parámetro de comparación la curación del grupo de control. Después de 3,7,14 y 21 días los animales fueron sacrificados y las heridas fueron evaluadas microscópicamente. En los días 3 y 7, las heridas tratadas con moco mostraron mejores tasas de formación y maduración del tejido de granulación, epitelización, y más rápido recambio de colágeno tipos III y I. A los días 14 y 21, hubo una intensa deposición del colágeno tipo I y aceleración en la regeneración de la piel. Los resultados obtenidos a partir de A. fulica alimentados con plantas de lechuga (L. sativa) mostraron mejores resultados. Este estudio sugiere que las películas producidas con secreción mucosa de A. fulica pueden ser utilizadas con éxito como apósito, especialmente si los caracoles se alimentan con plantas de lechuga L. sativa.

Campomanesia adamantium (Cambess.) O. Berg, Myrtaceae, é uma espécie nativa. As folhas são utilizadas na medicina popular. Os extratos hexânico e acetato de etila das folhas mostraram pouca variação na sua composição química em diferentes estações do ano associado com o desenvolvimento do vegetal, enquanto o extrato etanólico mostrou alteração significativa em relação à composição química. Todos os extratos etanólicos mostraram alta atividade antioxidante frente ao método DPPH e de moderada a alta para β-caroteno/ácido linoléico.
Campomanesia adamantium (Cambess.) O. Berg, Myrtaceae, is a native species. The extracts of ethyl acetate and hexane showed little variation in their chemical composition at different seasons of the year associated with the development of the plant, while the ethanol extract showed significant change in relationship to chemical composition. All ethanol extracts showed high antioxidant activity against DPPH method and from moderate to high antioxidant activity for β-carotene/linoleic acid.

Chemical studies with aerial parts of Microgramma vacciniifolia (Langsd. & Fisch.) Copel. afforded ²-sitosterol, hopan-22-ol, 6-metoxiapinenin-7-O-²-D-allopyranoside and a mixture containing ethyl esters of carboxilic acids. The structures of the coumpounds were elucidated by spectroscopy and GC-MS analysis. The total phenolics contents of the crude extract and fractions were determined by Folin-Ciocalteau method. The antioxidant activity was evaluated using the 2,2-diphenyl-1-picrylhydrazyl (DPPH). The AcOEt fraction showed better activity in DPPH assay (9.9 ± 0.03 µg/mL), and presented also higher contents of the total phenolic (93.60 ± 1.11 µg/mg). Antimicrobial and allelopathic effects of the crude etanolic extract and fractions also were evaluated. In addition, the combination of biological activities was discussed.

A leucemia promielocítica aguda (LPA) corresponde a 10% -15% das leucemias mielóides agudas (LMA). Este tipo de leucemia (LMA-M3 de acordo com a classificação FAB) está associado, em cerca de 90% dos casos, à translocação t(15;17)(q22;q21), que resulta na fusão dos genes PML e RARalfa. A análise citogenética tradicional tem sido utilizada para confirmar o diagnóstico morfológico da LPA. Embora a t(15;17) não seja detectada em outros tipos de leucemia, podem ocorrer resultados "falso-negativos", decorrentes da análise de células que não pertencem ao clone neoplásico, da dificuldade de visualização da translocação ou, até mesmo, da existência de rearranjos crípticos que mascaram a translocação. Por outro lado, foram descritas alterações cromossômicas alternativas em pacientes com LPA e, nesses casos, o tratamento com ATRA não é eficaz. No período de julho de 1993 a dezembro de 2002 foram encaminhados para análise citogenética 47 casos com suspeita e/ou diagnóstico clínico-morfológico de LPA. Trinta e quatro pacientes (72,3%) apresentaram a t(15;17), detectada pela citogenética tradicional e/ou molecular. Em seis destes pacientes foram observadas alterações cromossômicas adicionais ou rearranjos envolvendo um terceiro cromossomo. Em cinco (10%) pacientes com características de LPA, a técnica de FISH não revelou a fusão PML/RARalfa, dado importante para a orientação do diagnóstico e da conduta terapêutica desses pacientes. O presente trabalho foi realizado com o objetivo de avaliar a importância da análise citogenética tradicional e molecular no diagnóstico de pacientes com LPA.
Acute promyelocytic leukemia (APL) accounts for 10 to 15% of acute myeloid leukemias (AML). This type of leukemia (AML-M3 according to the FAB classification) is associated, in about 90% of the cases, with a t(15;17)(q22;q21) translocation, resulting in the fusion of the PML and RARalpha genes. Traditional cytogenetic analysis has been used to confirm the morphological diagnosis of ALP. Although the t(15;17) translocation is characteristic for this kind of leukemia, "false-negative" results may occur as a result of the analysis of cells which do not belong to the neoplastic clone, of the difficulty to visualize the translocation and even of the existence of cryptic rearrangements masking the translocation. Moreover, alternative chromosome alterations were described in patients with APL and in these cases treatment with ATRA is not effective. From July 1993 to December 2002, 47 cases suspected of or being diagnosed with APL by clinical-laboratorial methods were referred for cytogenetic analysis. Thirty-four patients (72.3%) had the t(15;17) translocation, detected by traditional and/or molecular cytogenetics. In six of these patients, additional chromosome alterations or rearrangements involving a third chromosome were observed. In five patients (10%) with APL characteristics, the FISH technique did not reveal the PML/RARalpha fusion, an important finding in the process of reaching a diagnosis and of establishing a therapeutic conduct for these patients. This work was carried out with the purpose of evaluating the importance of traditional and molecular cytogenetic analysis in the diagnosis of APL.
