Abstract Currently, antimicrobial resistance has become a global public health problem, which has made the need for new antimicrobial compounds to deal with resistant infections an emergency. However, environments that once offered so many innovative molecules, now already exhaustively exploited, do not meet this need. In this context, a geographically isolated, under-explored and extreme environment, such as Antarctica, which holds organisms with unique physiological and biochemical characteristics, assumes great importance as a potential source of new compounds with antimicrobial activity. In this patent review, we investigate the state of technological development in the field of antimicrobial compounds obtained from Antarctic organisms, highlighting the main countries and researchers active in the field, the species utilized, the compounds obtained, and their possible therapeutic applications. As results, few patent documents were found, however they encompass a wide diversity of compounds and species, indicating a great antimicrobial potential present in Antarctic biota, including compounds active against the most important human pathogenic microorganisms, such as including methicillin-resistant Staphylococcus aureus, vancomycin-resistant Enterococcus spp. and multi-resistant Mycobacterium tuberculosis. Furthermore, due to the increasing trend in patent applications, a significant rise in the number of patents in this area is expected in the coming years.

RESUMO O objetivo deste estudo foi identificar diferenças nos padrões morfogênicos de quatro cultivares de Urochloa brizantha (marandu, piatã, xaraés e paiaguás) durante o período de diferimento. Foi utilizado o delineamento inteiramente ao acaso, com três repetições, em parcelas experimentais de 9m². As avaliações ocorreram por dois anos. Os capins foram diferidos por 92 (Ano 1) e 95 (Ano 2) dias. A taxa de aparecimento foliar do capim-paiaguás foi maior, em comparação aos demais capins. No Ano 1, a taxa de alongamento do colmo do capim-xaraés foi superior aos demais capins. No final do diferimento do Ano 1, a densidade populacional de perfilho (DPP) foi maior no capim-paiaguás, intermediária nos capins xaraés e marandu e inferior no capim-piatã. No fim do período de diferimento do Ano 2, a DPP foi superior no dossel de capim-paiaguás, intermediária nos dosséis dos capins marandu e piatã, e menor no dossel de capim-xaraés. O capim-paiaguás tem maior crescimento foliar durante o período de diferimento, sendo, portanto, apropriado para uso sob pastejo diferido. O capim-xaraés apresenta elevado alongamento de colmo durante o período de diferimento, razão pela qual seu uso sob pastejo diferido deve vir acompanhado de ajustes no manejo da pastagem.

ABSTRACT The objective this study was to identify differences in the morphogenic patterns of four Urochloa brizantha cultivars (marandu, piatã, xaraés and paiaguás grasses) during the stockpiling period. A completely randomized design was used, with three replications, in experimental plots of 9m². The evaluations took place over 2 years. The grasses were stockpiled for 92 (Year 1) and 95 (Year 2) days. The leaf appearance rate of paiaguás grass was higher, compared to other grasses. In Year 1, the stem elongation rate of xaraés grass was higher than other grasses. At the end of stockpiling period of Year 1, the tiller population density (TPD) was higher in the paiaguás grass, intermediate in the xaraés and marandu grasses and lower in the piatã grass. At the end of the stockpiling period in Year 2, TPD was higher in the paiaguás grass canopy, intermediate in the marandu and piatã grasses canopies, and lower in the xaraés grass canopy. Paiaguás grass has greater leaf growth during the stockpiling period and is therefore suitable for use under stockpiled grazing. Xaraés grass has high stem elongation during the stockpiling period, which is why its use under stockpiled grazing must be accompanied by adjustments in pasture management.

RESUMO Objetivou-se identificar estratégias de rebaixamento da Urochloa brizantha cv. Marandu no início do diferimento, as quais resultem em aumento da produção e em melhoria da estrutura do dossel diferido. Foram avaliadas três estratégias de rebaixamento no início do diferimento: manutenção do capim-marandu com 15cm por quatro meses antes do início do diferimento; manutenção do capim-marandu com 30cm por quatro mese, e rebaixamento para 15cm no início do diferimento; e manutenção do capim com 45cm por quatro meses e rebaixamento para 15cm no início do diferimento. O delineamento foi inteiramente ao acaso, com quatro repetições. No fim do diferimento, o número de perfilho reprodutivo e a massa de forragem foram superiores no dossel sob 15/15cm do que nos demais; a percentagem de folha viva foi maior no dossel sob 45/15cm do que sob 15/15cm. A produção de forragem durante o diferimento foi superior no dossel sob 15/15cm e inferior no sob 45/15cm. A manutenção do capim-marandu com 15cm por quatro meses antes do período de diferimento aumenta a produção de forragem neste período. O rebaixamento do capim-marandu de 45 para 15cm no início do diferimento melhora a estrutura do dossel diferido.

ABSTRACT The objective was to identify lowering strategies for Urochloa brizantha cv. Marandu in the beginning of the stockpiling period that increase forage production and improve the structure of stockpiled canopies. Three lowering strategies were evaluated: maintenance of marandu palisade grass with 15cm four months before the stockpiling period; maintenance of palisade grass with 30cm for four months, and lowering to 15cm before the stockpiling period; and maintaining of palisade grass at 45cm for four months, and lowering to 15cm before the stockpiling period. The experimental design was completely randomized, with four replications. At the end of stockpiling, the number of reproductive tillers and forage mass were higher in the canopy under 15/15cm than in the others; the percentage of live leaf was higher in the canopy under 45/15cm than under 15/15cm. Forage production during the stockpiling period was higher in the canopy under 15/15cm and lower in the under 45/15cm. Maintaining 15cm marandu palisadegrass for four months before the stockpiling period increases forage production during this period. The lowering of the marandu palisadegrass from 45 to 15cm at the beginning of the stockpiling period improves the structure of the stockpiled canopy.

RESUMO Este trabalho foi conduzido com o objetivo de avaliar o efeito da condição da pastagem diferida de Brachiaria brizantha cv. Marandu ao final do inverno sobre o perfilhamento durante a estação de crescimento. Os tratamentos foram três condições de pastagem no final do inverno: pasto baixo, pasto alto e pasto alto/roçado. Nos meses de setembro e outubro, a taxa de aparecimento de perfilhos (TApP) e a taxa de mortalidade de perfilhos (TMoP) foram maiores na pastagem alta/roçada. Nos meses de novembro e dezembro, a pastagem alta apresentou maior TApP. De novembro a janeiro, a TMoP foi maior na pastagem alta. Os índices de estabilidade de perfilhos das pastagens baixas e altas/roçadas foram maiores em outubro. A pastagem baixa apresentou maior número de perfilhos do que as pastagens altas e altas/roçadas durante todo o período experimental. A pastagem diferida e baixa de capim-marandu no final do inverno apresenta, em geral, menor mortalidade de perfilhos e maior densidade populacional de perfilhos no início da primavera, em comparação com a pastagem alta. A roçada do capim-marandu com alta massa de forragem ao final do inverno, embora comprometa apenas temporariamente a estabilidade populacional dos perfilhos, também estimula o perfilhamento rápido a partir da primavera.

ABSTRACT This work was conducted to evaluate the effect of deferred pasture condition of Brachiaria brizantha cv. Marandu in the late winter on tillering during the growing season. The treatments were three pasture conditions at late winter: short pasture, tall pasture and tall/mown pasture. In September and October, tiller appearance rate (TApR) and tiller mortality rate (TMoR) were greater in the tall/mown pasture. In November and December, tall pasture presented greater TApR. From November to January the TMoR was greater in the tall pasture. The tiller stability index of short and tall/mown pastures were greater in October. The short pasture presented a greater tiller number than the tall and tall/mown pastures during the entire experimental period. Deferred and short pasture of marandu palisade grass at late winter presents in general lower tiller mortality and higher population density of tillers from the early spring onwards, in comparison to tall pasture. The mowing of marandu palisade grass with high forage mass at the late winter, although it only temporarily compromises the population stability of tillers, also stimulates its fast tillering from spring on.

RESUMO Objetivou-se avaliar as características estruturais e a produção de forragem do capim-piatã submetido à combinação de duas frequências de corte (95% e máxima interceptação de luz pelo dossel forrageiro, ILmax) e duas alturas pós-corte (15 e 20cm), no período de janeiro de 2012 a março de 2013. O delineamento utilizado foi em blocos ao acaso, com três repetições, em esquema fatorial 2x2, totalizando 12 unidades experimentais de 14m2 cada. Foram avaliadas as seguintes variáveis-respostas: altura do dossel no pré-corte, intervalo de corte, índice de área foliar pré e pós-corte, taxa de produção de forragem e as percentagens de colmo, folha e forragem morta na forragem produzida. A altura pré-corte decresceu durante o experimento (de 65,81 para 34,03cm), sendo a menor altura observada no dossel sob alta frequência de desfolhação (95% de IL). A taxa de produção de forragem foi maior nas épocas favoráveis ao crescimento da gramínea forrageira. As percentagens de folha, colmo e forragem morta foram semelhantes entre as frequências de corte. Assim, o capim-piatã sob manejo de desfolhação intermitente pode ser colhido com qualquer combinação de frequências de corte de 95% e ou máxima interceptação de luz, e alturas pós-corte de 15 ou 20cm.

ABSTRACT The objective of this study was to evaluate the structural characteristics and forage production of the piatã grass submitted to the combination of two cutting frequencies (95% and maximum interception of light by the forage canopy, ILmax) and two post cutting heights (15 and 20cm) in the period from January 2012 to March 2013. The design was a randomized complete block with three replications, in a 2x2 factorial scheme, totaling 12 experimental units of 14m2 each. The following variables were evaluated: pre-cut canopy height, cut interval, pre and post-cut leaf area index, forage yield rate, and percentage of stem, leaf and dead forage in the forage produced. The pre-cut height decreased during the experiment (from 65.81 to 34.03cm), with the lowest height observed in the canopy under high frequency of defoliation (95% IL). The rate of forage production was higher in the seasons favorable to forage grass growth. The percentages of leaf, stem and dead fodder were similar between cut-off frequencies. Thus, piatã grass under intermittent defoliation management can be harvested with any combination of 95% and maximum light interception cut frequencies, and post cutting heights of 15 or 20cm.

Abstract Uncaria tomentosa (UT) extracts have been shown to have promising anti-tumor activity. We hypothesized that its incorporation into nanostructured systems could improve the anticancer properties. Here, poly-e-caprolactone (PCL) and poly-d,l-lactide-co-glycolide (PLGA) were employed to generate nanoparticles loaded with UT extract in a single emulsion solvent evaporation method. The nanoparticles were characterized by particle size, zeta potential, morphology and entrapment efficiency along with stability and release profiles. The nanoparticles presented entrapment efficiencies above 60% and a mean diameter below 300nm. UT-PCL nanoparticles presented higher entrapment efficiency and mean particle size as well as a slow release rate. The UT-PLGA nanoparticles showed higher drug loading. Two prostate cancer cell-lines, LNCaP and DU145 that were derived from metastatic sites, served as model systems to assess cytotoxicity and anti-cancer activity. In vitro, both formulations reduced the viability of DU145 and LNCaP cells. Yet, the UT-PLGA nanoparticles showed higher cytotoxicity towards DU145 cells while the UTPCL against LNCaP cells. The results confirm that the incorporation of UT into nanoparticles could enhance its anti-cancer activities that can offer a viable alternative for the treatment of prostrate canner and highlights the potential of nanostructured systems to provide a promising methodology to enhance the activity of natural extracts.

RESUMO Objetivou-se avaliar o perfilhamento da Brachiaria brizantha cv. Piatã, como base para identificação das frequências e alturas pós-corte mais adequadas para essa gramínea forrageira. Em um esquema fatorial 2 x 2, foram estudadas combinações entre duas frequências de corte (95% e máxima interceptação de luz, ILmax) e duas alturas pós-corte (15 e 20cm). O delineamento foi em blocos ao acaso, com três repetições, totalizando 12 unidades experimentais de 14m2 cada. Foram avaliadas as taxas de aparecimento (TApP) e de mortalidade (TMoP) de perfilhos, o índice de estabilidade (IE) e a densidade populacional de perfilhos. A maior TApP no dossel sob IL de 95% ocorreu no Verão 1, em comparação às demais épocas. Nos dosséis manejados com ILmax, a TApP foi superior no Verão 1 e no Outono, quando comparados às outras épocas. No Verão 1 e no Final da Primavera e Verão 2, houve as maiores TMoP para as duas frequências de corte avaliadas. No Outono, a TMoP também foi alta no dossel cortado com ILmáx. O IE foi menor durante o Verão 1 no dossel sob ILmax e com 15cm de resíduo (0,995). Porém, nas demais épocas, os valores de índice de estabilidade foram maiores que uma unidade e não foram influenciados pela frequência e altura pós-corte. A densidade populacional de perfilhos foi menor no Verão 1, em comparação às demais épocas. O capim-piatã pode ser desfolhado com qualquer combinação entre as interceptações de luz, 95% e ILmax, e alturas pós-corte, 15 ou 20cm, sem prejudicar a persistência do pasto.

ABSTRACT The aim of this study was to evaluate the tillering of Brachiaria brizantha cv. Piatã, as a basis for identifying the frequencies and post-cutting heights most suitable for this grass. In a 2 x 2 factorial scheme, combinations between two cut frequencies (95% and maximum light interception, IL max ) and two post-cut heights (15 and 20cm) were studied. The experimental design was in randomized blocks with 3 replicates, totaling 12 experimental units of 14m 2 each. Rates of appearance (TAR) and mortality (TMR) of tillers, the stability index and the population density of tillers were evaluated. The highest TAR in the canopy under 95% IL occurred in Summer 1, in comparison to the other seasons. In the canopies managed with IL max , TAR was higher in summer 1 and in autumn, when compared to other seasons. In summer 1 and at the end of spring and summer 2, there were the highest TMR for the two cutoff frequencies evaluated. In autumn, TMR was also high in the canopy cut with IL max . IE was lower during summer 1 in the canopy under IL max and 15cm of residue (0.995). However, at other times, stability index values were greater than one unit and were not influenced by post-cut frequency and height. The population density of tillers was lower in Summer 1, when compared to the other seasons. The piata grass may be defoliated with any combination of light intercepts, 95% and ILmax, and post-cut heights, 15 or 20cm, without impairing the persistence of pasture.

RESUMO Este estudo teve como objetivo avaliar a ação do extrato aquoso de noni em diluente para congelação de sêmen de carneiro. Os tratamentos diferiram quanto à inclusão de extrato aquoso de noni ao meio diluidor em: T1˗ sem adição de extrato; T2˗ 24µg/mL ; T3- 72µg/mL e 120µg/mL. Por meio de vagina artificial, 16 ejaculados foram coletados, diluídos entre os quatro tratamentos e congelados. Após o descongelamento, o sêmen foi submetido ao teste de termorresistência e avaliado quanto à motilidade subjetiva, ao vigor espermático, à integridade de membrana pelo teste hiposmótico, bem como ao teste supravital, à análise de sêmen assistida por computador (CASA) e ao status de capacitação espermática e de reação acrossomal. Os dados foram submetidos a uma análise de variância, seguida pelo teste de Student-Newman-Keuls com 5% de significância. No teste de termorresistência, após duas horas de incubação, a motilidade do T4 (120µg/mL) apresentou-se inferior aos demais tratamentos. Não houve diferença significativa no teste HOS tanto para o sêmen diluído quanto para o sêmen avaliado imediatamente pós-descongelação; para as demais horas, os tratamentos apresentaram comportamento semelhante. Para os parâmetros de cinética, foi observada diferença estatística para motilidade progressiva, velocidade curvilinear, velocidade do percurso médio e amplitude de deslocamento lateral da cabeça. Quanto ao estado de capacitação espermática, observou-se diferença entre os tratamentos para espermatozoide capacitado com acrossomo intacto.

ABSTRACT The study aimed to evaluate the action of aqueous extract of noni in an extender for sheep semen freezing. Treatments differed in inclusion of aqueous extract of noni in the extender: T1 ˗ no addition; T2 ˗ 24µg/mL; T3 ˗ 72µg/mL; and T4 ˗ 120µg/mL. Ejaculates were collected, diluted in the four treatments, and frozen. After thawing, the semen was subjected to a thermoresistance test and evaluated for subjective motility, vigor, membrane integrity assessment by hypo-osmotic swelling test, live-dead assay, computer-assisted sperm analysis and the status of sperm capacitation and acrosome reaction. Data were subjected to ANOVA, and then to Student Newman Keuls’s test at 5% significance level. In the thermoresistance test after two hours of incubation, motility in T4 (120µg/mL) was lower than in the other treatments, with no differences in the HoS test in either diluted semen or in the semen evaluated immediately post-thawing, while for the other times, treatments showed similar responses. Regarding the motility parameters, a difference was observed for progressive motility, curvilinear velocity, average path velocity, and amplitude of lateral head displacement. As to the sperm capacitation status, a difference was observed between treatments for the sperm capacitated with intact acrosome.

Abstract For the past 10 years, studies have contributed greatly to knowledge regarding the composition and distribution of the planktonic cladocerans of Tupé Lake in Amazonas State, Brazil, but comparisons between these studies have not been made, and this would increase the comprehension of the ecology of these organisms and their environment. Periodically the hydrological conditions in Tupé Lake change, due to its connecting channel with the Negro River. The purpose of this study was to determine the composition, abundance and diversity of cladocerans in the limnetic zone of Tupé Lake and compare the results with previous studies performed there. Samples were taken in April, June, September, October, November/08 and February/09 in 4 stations located in different areas of the lake. Ceriodaphnia cornuta G.O. Sars, 1886, Bosminopsis deitersi Richard, 1895 and Bosmina cf. longirostris (O.F. Müller, 1785) were the most abundant and frequent species of cladocerans among the 25 recorded species. Higher abundance and richness were observed during the low water phase (October/08). A relationship between the number of samples taken, as well as the type of sampling gear used, and the species richness was observed, when comparing this and previous studies performed in the lake.

Resumo Objetivo: O presente estudo teve como objetivos: 1) analisar a associação do estado maturacional com o desempenho nas provas de coordenação motora em crianças e 2) examinar se a relação entre o estado maturacional e o desempenho no KTK é mediada por alguma medida antropométrica. Métodos: A amostra de conveniência foi composta por 73 crianças do sexo masculino com 8 anos. A antropometria considerou a estatura, massa corporal, altura sentado, o perímetro de cintura, índice de massa corporal, as estimativas de massa gorda e massa livre de gordura. A maturação biológica foi avaliada pelo percentual da estatura matura predita. A coordenação motora foi testada pela bateria Körperkoordinationstest für Kinder. Foi feita a correlação parcial entre as medidas antropométricas, z-escore da maturação e as provas de coordenação motora, com controle para idade cronológica. Por último, a análise de mediação causal foi feita. Resultados: Estatura, massa corporal, perímetro de cintura e massa gorda apresentaram correlação inversa de magnitude pequena a moderada com as provas de coordenação motora. A maturação biológica associou-se significativamente à prova de equilíbrio em marcha à retaguarda (r=-0,34). Foi identificada mediação total do perímetro de cintura na relação do estado maturacional com o equilíbrio em marcha à retaguarda (77%). Conclusões: Foi possível identificar a associação entre o estado físico maturacional e o desempenho em uma prova de bateria de coordenação motora em crianças do sexo masculino e, ainda, afirmar que há mediação do perímetro de cintura. Recomenda-se que estudos sejam feitos com indivíduos de outras idades e do sexo feminino.

Abstract Objective: The present study aimed to: 1) examine the association of biological maturation effect on performance at a motor coordination battery and 2) to assess whether the association between biological maturation and scores obtained in motor coordination tests is mediated by some anthropometric measurement. Methods: The convenience sample consisted of 73 male children aged 8 years old. Anthropometric data considered the height, body mass, sitting height, waist circumference, body mass index, fat mass and fat-free mass estimates. Biological maturation was assessed by the percentage of the predicted mature stature. Motor coordination was tested by the Körperkoordinationstest für Kinder. A partial correlation between anthropometric measurements, z-score of maturation and the motor coordination tests were performed, controlling for chronological age. Finally, causal mediation analysis was performed. Results: Height, body mass, waist circumference and fat mass showed a slight to moderate inverse correlation with motor coordination. Biological maturation was significantly associated with the balance test with backward walking (r=-0.34). Total mediation of the waist circumference was identified in the association between biological maturation and balance test with backward walking (77%). Conclusions: We identified an association between biological maturation and KTK test performance in male children and also verified that there is mediation of waist circumference. It is recommended that studies be carried out with female individuals and at other age ranges.

RESUMO O objetivo desse trabalho foi avaliar a atividade alelopática dos lixiviados das raízes e folhas secas de Asemeia extraaxillaris sobre o crescimento de corda de viola Ipomoea cordifolia em casa de vegetação, para avaliação dos efeitos sobre o estresse oxidativo. Nos bioensaios foram utilizadas três concentrações (0,25 - 5,00 e 10,00 g) e um controle, com quatro repetições de 10 plantas, cultivadas em solo durante 30 dias. Os lixiviados possuem ação alelopática e o maior efeito foi verificado para o pó das raizes. O efeito inibitório foi dose-dependente e o aumento do estresse oxidativo foi observado pelos níveis da catalase, peroxidase, superóxido dismutase, glutationa redutase, peroxidação lipídica e síntese de formazan. Concluiu-se que A. extraaxillaris possui substâncias capazes de afetar a emergência, o crescimento e o estresse oxidativo de I. cordifolia.

ABSTRACT The purpose of this article is to assess the allelopathic activity of leachates from the roots and dried leaves of Asemeia extraaxillaris on the germination and growth of corda de viola (Ipomoea cordifolia) in a greenhouse, and evaluate its effect on oxidative stress. For the bioassays, three concentrations (0.25, 5.00, and 10.00 g) and one control were used, with four replications of 10 plants, cultivated in soil for 30 days. The leachates exhibited allelopathic action, and the greatest effect was found in the root powder. The inhibitory effect was dose-dependent and the increase in oxidative stress was observed by the levels of catalase, peroxidase, superoxide dismutase, glutathione reductase, lipid peroxidation, and formazan synthesis. It was affirmed that A. extraaxillaris contains substances capable of affecting seedling emergence, growth, and oxidative stress in I. cordifolia.

A distinção de Butia capitata e de B. odorataé baseada em poucos caracteres morfológicos, o que motiva a busca por mais dados que corroborem a separação. Como dados da anatomia foliar são relevantes na circunscrição de taxa em Arecaceae, objetivou-se descrever a anatomia foliar de B. capitatae comparar com a de B. odorata. Amostras da porção mediana das pinas foram coletadas e processadas de acordo com técnicas usuais em Anatomia Vegetal. A epiderme é unisseriada e possui cutícula espessa e ceras epicuticulares em formato de filamentos em anzol. As células subsidiárias arqueiam-se sobre as células-guarda localizadas no nível da hipoderme. O mesofilo é isobilateral e compacto. Os feixes vasculares são colaterais com extensão de bainha esclerenquimática alcançando a hipoderme. Stegmatas possuem corpos silicosos esféricos e drusóides. A nervura mediana é adaxialmente projetada, apresentando espessa camada fibrosa que circunda os feixes vasculares, adjacente ao parênquima clorofiliano. Tecido de expansão estratificado ocorre na face abaxial, nos limites do mesofilo com a nervura mediana. Ráfides ocorrem apenas em B. capitata. Os feixes diminutos da nervura mediana circundam totalmente o anel fibroso somente em B. odorata. Além de diagnósticos, esses caracteres são úteis para corroborar a separação atualmente proposta.

The distinction between Butia capitata and B. odorata is based only on a few morphological characteristics, therefore there is a need for additional studies for supporting the separation of the species. As lamina anatomy characteristics are relevant in circumscribing Arecaceae taxa, this work aimed to describe B. capitata lamina anatomy and compare it with B. odorata. Samples from the middle portion of the pinnae were collected and processed in accordance with standard plant anatomy techniques. The epidermis is uniseriate and composed of a thick cuticle and epicuticular waxes into like hooked filaments. The subsidiary cells that arch over the guard cells are located at the hypodermis. The mesophyll is isobilateral and compact. The vascular bundles are collateral with a sclerenchymatic sheath extension that reaches the hypodermis. The stegmata cells have spherical and druse-like silica bodies. The midrib faces the adaxial surface with a thick fibrous layer surrounding the vascular bundles adjacent to the chlorenchyma. The stratified expansion tissue is on the abaxial surface, within the boundary between the mesophyll and midrib. Raphides are only found in B. capitata. Small bundles of the midrib fully surround the fibrous cylinder only in B. odorata. These characteristics are diagnostic and useful for supporting the proposed separation.

RESUMO Das sementes da mamona extrai-se o óleo de rícino, utilizado in natura ou em sua forma modificada nas áreas médica, farmacêutica e industrial. Um de seus derivados químicos - o Poliquilgerm® - evidencia propriedades antifúngicas sobre Candida albicans e bacteriostática/ bactericida sobre Escherichia coli no nível de 99,9%. Considerando-se essas propriedades, aplicouse o Poliquilgerm® em culturas de Leuconostoc mesenteroides, uma das espécies de bactéria contaminante dos mostos em indústrias sucroalcooleiras. Esta bactéria quando presente em mostos produz além do ácido láctico, a dextrana, que é um polímero da glicose que aumenta a viscosidade dos fluidos dos processos. No presente trabalho avaliou-se o efeito de diferentes concentrações do Poliquilgerm® sobre a viscosidade produzida por L. mesenteroides em diferentes meios de cultivo, condições de pH, e temperatura. Verificou-se 20,56% de diminuição da viscosidade quando se utilizou 1,0% do produto, além da inibição do crescimento da bactéria após 20 horas em contato com 1,0 e 0,2% de Poliquilgerm®. L. mesenteroides apresentou melhor crescimento em valores de pH 6,0, tanto a 28 como a 33º C, evidenciado pela maior produção de biomassa. Além disso, em meio de cultura neste pH verificou-se maior porcentagem de diminuição da viscosidade em ambas as concentrações utilizadas. Estas mesmas concentrações alcançaram até 100% de diminuição das UFC/mL após 24 horas em contato com o produto, evidenciado por quantificação da biomassa e confirmado mediante plaqueamento pela técnica “Pour Plate”.

ABSTRACT Castor-oil, extracted from seeds of Ricinus communis L., is normally used in natura or in its modified form in the medical, pharmaceutical and industrial areas. One of its chemical derivates – Poliquilgerm® – has shown antifungal properties on Candida albicans, and bacteriostatic/bactericide effects on Escherichia coli reaching 99.9%. Considering these features, in the present study Poliquilgerm® was applied in cultures of Leuconostoc mesenteroides, a frequent contaminant bacterium of musts in the sugar and alcohol industries. This bacterium, when a contaminant of must, produces lactic acid as well as dextran, a polymer of glucose which may increase the viscosity of fluids in the operation. The present study evaluated the effect of different concentrations of Poliquilgerm® on viscosity produced by L. mesenteroides cultures under different conditions of various culture media, pH and temperature. A 20.56% decrease in viscosity was verified when 1.0% of the product was used, and bacterium growth inhibition after 20 hours in media with 1.0 and 0.2% of Poliquilgerm®. L. mesenteroides showed a higher growth rate in pH 6.0, at both 28 and 33º C, evidenced by greater biomass production. Also, in the culture media at this pH a greater decrease in viscosity percentages was verified at both concentrations used. The media with these same concentrations reached up to 100% decrease of UFC/mL after 24 hours in the presence of the product, observed by biomass quantification and confirmed by the Pour Plate technique.

OBJETIVO: Avaliar o micrométodo analítico empregando a cromatografia líquida para quantificação de propranolol em crianças operadas de tetralogia de Fallot (TLF). MÉTODO: Requereu-se apenas volumes de 0,2mL de plasma para a realização do ensaio. Os picos foram eluídos em 8.4 (Propranolol) e 17.5 min (verapamil, padrão interno) de uma coluna C18, com fase móvel (tampão acetato 0,1 M pH 5,0 e acetonitrila, 60:40, v/v) em fluxo de 0,7 mL/min, sendo detectados em 290 nm (excitação) e em 358 nm (emissão). A cirurgia iniciou-se 776 min depois da dose administrada (8,7mg, média) e sete amostras de sangue foram coletadas de seis pacientes (4M/2F; 2,1 anos;11,5kg; 0,80m;18,9kg/m²). RESULTADOS: Os limites de confiança do método analítico evidenciaram alta seletividade e recuperação, sensibilidade (0,02ng/mL), boa linearidade (0,05-1000ng/mL), precisão de 8,6% e exatidão de 3,1%. A duração média da cirurgia foi de 283,2min, com os pacientes em circulação extracorpórea (CEC) durante 114min. Uma curva de declínio do propranolol no plasma foi obtida após a última dose na noite que precedeu o dia da intervenção. A concentração plasmática foi normalizada com o hematócrito devido à hemodiluição causada pela CEC. Por outro lado obteve-se decréscimo nas concentrações plasmáticas entre os períodos início da cirurgia para o 2º dia de pós-operatório (7,09 ng/mL e0,05 ng/mL, p<0,05 respectivamente) e do final da CEC para o 2º dia de pós-operatório (2,79ng/mL e 0,05ng/mL, p<0,05). CONCLUSÃO: O monitoramento das concentrações plasmáticas normalizadas do propranolol, em crianças com TLF, após a última dose pré-operatória revelou decaimento do início da cirurgia para o segundo pós-operatório, sugerindo que após a correção cirúrgica, uma vez restaurada a distribuição, a eliminação do fármaco foi completa.

OBJECTIVE: To evaluate the analytical micromethod using liquid chromatography for the quantification of propranolol in children submitted to surgery of tetralogy of Fallot (TLF). Methods: Only 0.2 mL of plasma is required for the assay. Peaks eluted at 8.4 (Propranolol) and 17.5 min (verapamil, internal standard) from a C18 column, with a mobile phase 0.1 M acetate buffer, pH 5.0, and acetonitrile (60:40, v/v) at flow rate 0.7 mL/min, detected at 290 nm (excitation) and 358 nm (emission). Surgery was started 776 min of drug administration (8.7mg, mean); seven blood samples were collected from six patients (4M/2F; 2.1yrs;11.5kg; 0.80m; 18.9kg/m²). RESULTS: Confidence limits of the method showed high selectivity and recovery, sensitivity of 0.02ng/mL, good linearity (0.05-1000ng/mL), precision of 8.6% and accuracy of 3.1%. The mean duration of surgery was 283.2min, with the patients remaining under cardiopulmonary bypass (CPB) for 114min. A declining curve of propranolol plasma concentration was obtained after the last dose in the night that preceded the day of surgery. Plasma concentration also was normalized with hematocrit due to the hemodilution caused by the CPB procedure. On the other hand a decrease on drug plasma concentration was obtained between periods, the beginning of surgery to the postoperative day 2 (7.09 ng/mL and 0.05 ng/mL, p<0.05 respectively) and from the end of CPB to the postoperative day 2 (2.79ng/mL e 0.05ng/mL, p<0.05). CONCLUSION: Propranolol monitoring of plasma concentrations of children (TLF) normalized after the last preoperative dose revealed a decline from the beginning of surgery to the second postoperative day, suggesting that, once redistribution was restored, propranolol washout was complete.
