Abstract Human activities on the Earth's surface change the landscape of natural ecosystems. Mining practices are one of the most severe human activities, drastically altering the chemical, physical and biological properties of the soil environment. Bacterial communities in soil play an important role in the maintenance of ecological relationships. This work shows bacterial diversity, metabolic repertoire and physiological behavior in five ecosystems samples with different levels of impact. These ecosystems belong to a historical area in Iron Quadrangle, Minas Gerais, Brazil, which suffered mining activities until its total depletion without recovery since today. The results revealed Proteobacteria as the most predominant phylum followed by Acidobacteria, Verrucomicrobia, Planctomycetes, and Bacteroidetes. Soils that have not undergone anthropological actions exhibit an increase ability to degrade carbon sources. The richest soil with the high diversity was found in ecosystems that have suffered anthropogenic action. Our study shows profile of diversity inferring metabolic profile, which may elucidate the mechanisms underlying changes in community structure in situ mining sites in Brazil. Our data comes from contributing to know the bacterial diversity, relationship between these bacteria and can explore strategies for natural bioremediation in mining areas or adjacent areas under regeneration process in iron mining areas.

RESUMO Composto orgânico produzido a partir de biomassa residual constitui uma alternativa viável para a substituição parcial ou total de fertilizantes minerais no cultivo de hortaliças. Neste estudo foram avaliados os efeitos do composto, sobre a qualidade da água de um córrego, usado principalmente para a irrigação de culturas agrícolas cultivadas em solo nas proximidades, tratado nos últimos dez anos com composto orgânico produzido a partir de carcaças, resíduos animais e vegetais. Amostras de água foram coletadas semestralmente, em 2013 e 2014, em cinco pontos ao longo do córrego. Foram analisadas variáveis físicas, algumas variáveis químicas e o número total de coliformes (Escherichia coli). As populações bacterianas foram comparadas pelo consumo de substratos de carbono. Os teores de fósforo total nas amostras de 2014 ultrapassaram 0,1 mg L-1. As concentrações das demais espécies químicas analisadas e os resultados para as variáveis fisicas ficaram de acordo com valores estabelecidos em normas nacionais e internacionais de qualidade de água. O ambiente mostrou consumo diferencial de fontes de carbono e grande diversidade de micro-organismos, mas os resultados não mostraram qualquer evidência de que o composto aplicado seja o fator responsável pela alteração da população microbiana ou sua atividade metabólica. Este estudo mostra que o uso do composto orgânico em áreas agrícolas parece não influenciar negativamente a qualidade da água superficial na área estudada. Estes resultados são importantes porque o processo de compostagem de resíduos animais e vegetais e o uso do composto obtido podem ser uma alternativa sustentável para o destino adequado desses resíduos.

ABSTRACT Organic compost from biomass residues constitutes a viable alternative for partial or total replacement of mineral fertilizers for growing vegetables. This study evaluated the effects of compost on the water quality of a stream used mainly for irrigation of agricultural crops cultivated in nearby soil that has been treated with organic compost produced by carcasses, animal and vegetable waste for the last ten years. We sampled water biannually for two years, 2013 and 2014, from five locations along the stream. Physical variables and some chemical variables were analyzed. We also analyzed the total number of coliforms (Escherichia coli). Bacterial populations were compared by carbon substrate consumption. Total phosphorus contents in the samples from 2014 exceeded 0.1 mg L-1. The concentrations of other chemical species analyzed and the results for the physical variables were in accordance with the expected values compared with national and international water quality standards. The environment showed differential carbon source consumption and a high diversity of microorganisms, but our results did not show any evidence that the applied compost is changing the microbial population or its metabolic activity. This study shows that the use of the organic compost in agricultural areas seen does not negatively influence the quality of surface water in the study area. These results are important because the process of composting animal and vegetable waste and the use of compost obtained can be an alternative sustainable for adequate destination of these wastes.

RESUMO Desde o final do século vinte, 100% do esgoto urbano da cidade de Jaboticabal é recolhido por interceptores e conduzido a uma estação de tratamento. Entre 1999 e 2000, estudos sobre o efeito desse cuidado ambiental, mostraram que a coleta de esgoto ainda não tinha sido eficiente para despoluir as águas dos dois córregos que atravessam a cidade e fluem para uma área agrícola. Neste trabalho avalia-se a influência do uso do solo na qualidade da água destes dois córregos. O estudo foi realizado 15 anos após a implantação dos interceptores de esgoto. Durante o ano de 2014, bimestralmente, coletaram-se amostras de água em oito pontos (P1 a P8), dos córregos Cerradinho e Jaboticabal, em Jaboticabal (SP, Brasil). P1 estava localizado na nascente do córrego Jaboticabal; P2 em área agrícola; P3, P4 e P5 em área urbana; P6, P7 e P8 em área agrícola. Foram determinadas variáveis físicas e químicas e a comparação metabólica da população microbiana em diversas fontes de carbono pelo método EcoPlate (Biolog). Em todos os pontos e coletas, o teor de fósforo total excedeu o limite estabelecido pela legislação brasileira, assim como valores encontrados em estudos anteriores, o que também foi observado em relação à demanda química de oxigênio. Entretanto, a população microbiana da água parece não estar sendo alterada pelas atividades urbanas e agrícolas. Os resultados das análises sinalizam à possibilidade de descarga clandestina de águas residuárias nos córregos estudados e à influência do uso agrícola do solo.

ABSTRACT Since the end of the twentieth century, 100% of the urban sewage from the city of Jaboticabal has been collected by interceptors and routed to a treatment plant. Between 1999 and 2000, studies on the effect of this environmental care showed that it did not efficiently clean up the water from the two streams that run through the city and flow into an agricultural area. This paper focuses on assessing the influence of soil use on the water quality of surface waters from these two streams. The study was conducted 15 years after the implementation of sewage interceptors. The sampling dates were bimonthly at eight points (P1 to P8) in the Cerradinho and Jaboticabal streams, in Jaboticabal (São Paulo State, Brazil). P1 was located at the source of the Jaboticabal stream, P2 was in a farming area, P3, P4 and P5 were in an urban area, and P6, P7 and P8 were in a farming area. The physical and chemical variables of the water were assessed. We compared the ability of microorganisms to metabolize different sources of carbon using the EcoPlate (Biolog). The total phosphorus (TP) concentration exceeded the limit set by the Brazilian legislation as well as values found in previous studies, which was also observed for the chemical oxygen demand. However, the bacterial metabolic profile had no association with urban or farming practices. The results of the analysis indicated the possibility of clandestine discharge of wastewater in the streams studied and the influence of the agricultural soil.

Estudos sobre a atividade microbiológica que ocorre na rizosfera de diversos vegetais levaram ao descobrimento de grupos de microrganismos importantes para o desenvolvimento vegetal. Dentre eles estão as rizobactérias que são capazes de colonizar as raízes, estimulando-a diretamente ou beneficiando o crescimento e o desenvolvimento de diversas plantas. Estas bactérias são chamadas Rizobactérias Promotoras de Crescimento em Plantas (RPCP). Este trabalho teve o objetivo de isolar, identificar, testar a capacidade da solubilização de fosfato e a produção de ácido indol acético (AIA) de bactérias que habitam a rizosfera de plantas de milho. A análise parcial do gene 16S rRNA dos 58 isolados possibilitou a identificação dos gêneros, Bacillus, Burkholderia e Azospirillum, sendo os mais frequentes totalizando 68% dos isolados, seguidos de Sphingomonas, Pseudomonas, Herbaspirillum, Pantoea , Bosea. Desses, 27 apresentaram a capacidade de solubilização do fosfato e 18 foram positivos no teste colorimétrico para detecção de produção do AIA. A partir destes resultados, selecionou-se um organismo pertencente ao gênero Sphingomonas para ser testado em casa de vegetação como promotor de crescimento com as estirpes de Azospirillum brasilense (AbV5 e AbV6). As plantas foram avaliadas quanto à altura aos vinte e setenta dias após a germinação e a massa seca da parte aérea (MSPA)e parte radicular (MSPR) foi quantificada após setenta dias no encerramento do experimento. Os resultados das análises do isolado pertencente ao gênero Sphingomonas foram estatisticamente semelhantes às estirpes AbV5 e AbV6 na planta de milho indicando que este microrganismo possui potencial para ser utilizado como RPCP

Researches about microbiological activity on many plants rizosphere led to the discovery of groups of important microrganisms for plants development. Among them there are the rizobacteria able to colonize the roots, stimulating directly or benefiting many plants. These bacteria are called Growth Promoting Rizobacteria or GPR. In view of this, the present study was performed, aiming to isolate, identify and test phosphate solubilization and acetic indol acid (AIA) production in bacteria that lives in maize roots. The partial analysis of the 16S rRNA gene of the 58 isolates enabled the identification of the genres Bacillus, Burkholderia and Azospirillum, and the most frequent totaling 86% of the isolates, followed by Sphingomonas, Pseudomonas, Herbaspirillum, Pantoea and Bosea. Out of the 58 isolates, 27 showed the capacity of phosphate solubilization and 18 were positive to the AIA production colorimetric detection test. From these results was chosen an organism belonging to the genre Sphingomonas to be tested at greenhouse as growth promoter with the strains Azospirillum brasiliense (AbV5 e AbV6). The plants were assessed twenty and seventy days after germination. Dry mass of shoot and root parts were quantified after seventy days at the end of the experiment. Statistical analysis indicated by the program Statistix v.9.0 showed that the isolates belonging to the genre Sphingomonas presented similar results as shown by strains AbV5 and AbV6 on maize plant. This microorganism may become a promising one to be used as a GPR
