Cross-amplification was tested and variability in microsatellite primers (designed for Neotropical parrots) compared, in five macaw species, viz., three endangered blue macaws (Cyanopsitta spixii [extinct in the wild], Anodorhynchus leari [endangered] and Anodorhynchus hyacinthinus [vulnerable]), and two unthreatened red macaws (Ara chloropterus and Ara macao). Among the primers tested, 84.6% successfully amplified products in C. spixii, 83.3% in A. leari, 76.4% in A. hyacinthinus, 78.6% in A. chloropterus and 71.4% in A. macao. The mean expected heterozygosity estimated for each species, and based on loci analyzed in all the five, ranged from 0.33 (A. hyacinthinus) to 0.85 (A. macao). As expected, the results revealed lower levels of genetic variability in threatened macaw species than in unthreatened. The low combined probability of genetic identity and the moderate to high potential for paternity exclusion, indicate the utility of the microsatellite loci set selected for each macaw species in kinship and population studies, thus constituting an aid in planning in-situ and ex-situ conservation.

OBJETIVO: avaliar a preservação folicular e as características celulares do tecido ovariano criopreservado, em coelhas. MÉTODOS: fez-se, sob anestesia, a ooforectomia direita de dez coelhas brancas, adultas. Dissecou-se o ovário mantendo-se o córtex com espessura de 1,5 milímetros. Fragmentou-se o tecido em pequenas secções, algumas para o estudo histológico de controle e outras destinadas à criopreservação pelo protocolo de congelamento lento. Passadas seis semanas efetuou-se o descongelamento e fez-se a avaliação histológica. As amostras do controle e do experimento, após processamento, foram coradas pela hematoxilina-eosina para identificação dos aspectos histológicos e submetidas à técnica imuno-histoquímica utilizando-se o PCNA (proliferating cell nuclear antigen) para a avaliação da viabilidade celular. Utilizaram-se os testes não paramétricos "comparação entre duas proporções" e Mann-Whitney e o teste paramétrico t de Student. RESULTADOS: observou-se que no tecido criopreservado só persistiram oócitos primordiais. Entre as alterações reversíveis identificaram-se: vacuolização citoplasmática em todas as amostras (p=0,039), lise estromal em 50% (p=0,648) e oócitos com contornos irregulares em 80% (p=0,007). Encontraram-se alterações irreversíveis como degeneração hialina e picnose em 30% das amostras (p=0,210). A análise imuno-histoquímica demonstrou os folículos, em diferentes estágios de desenvolvimento, no tecido não congelado e folículos primordiais no tecido criopreservado com positividade para o PCNA, indicando a presença de DNA ativo. CONCLUSÃO: no tecido ovariano criopreservado sobrevivem apenas os folículos primordiais; existem alterações histológicas reversíveis (vacuolização citoplasmática, lise estromal, fragmentação das células da granulosa e oócitos com contornos irregulares); alterações irreversíveis, em níveis não significantes (degeneração hialina e picnose) e presença de PCNA positivo em todos os folículos.

PURPOSE: to evaluate follicular preservation and histologic characteristics of the cryopreserved ovarian tissue and to compare with the fresh one, in rabbits. METHODS: ten adult female white rabbits were submitted to right oophorectomy. The dried ovary was dissected and the cortex was maintained with approximately 1.5 millimeter thickness. The tissue was fractionated into small sections, some reserved for control histologic study and others destined for cryopreservation. Six weeks later the ovarian tissue was thawed and evaluated histologically. After histologic processing, the control and the experimental samples were stained with hematoxylin and eosin and treated immunohistochemically by the PCNA technique for evaluation of DNA preservation. Histologic alterations present in the fresh and in the cryopreserved tissues were identified, and cryopreserved tissue viability was evaluated. RESULTS: in the cryopreserved tissue only primordial follicles persisted. Reversible alterations were identified: cytoplasmatic vacuolation (p=0,039), stromal lysis (p=0.648) and oocytes with irregular contours (p=0.007). Irreversible alterations: (hyalin degeneration and pyknosis) were found, but not at significant levels (p=0.210). The immunohistochemical analysis showed PCNA staining of follicles at different stages of development in the fresh tissue and primordial follicles in the cryopreserved tissue, indicating the presence of active DNA in both tissues. CONCLUSION: in the cryopreserved ovarian tissue the following were observed: survival of only primordial follicles; significant reversible histologic alterations (cytoplasmic vacuolation, stromal lysis and oocytes with irregular contours); irreversible alterations (hyalin degeneration and pyknosis), and PCNA staining of all follicles.

Muitas substâncias têm sido utilizadas após hepatectomias parciais a fim de conhecer como elas atuam sobre o processo de regeneração hepática. Adequado fluxo sangüíneo parece ter muita importância neste processo. Autores descreveram, após hepatectomias em ratos, elevação dos níveis plasmáticos de histamina. Substâncias inibidoras da histidina-decarboxilase e da histaminase leva a elevação da histamina endógena com vasodilatação sinusoidal e aceleração da regeneração hepática. No presente estudo busca-se conhecer a influência da histamina exógena administrada à ratos parcialmente hepatectomizados. Utilizaram-se 32 ratos Wistar que sofreram hepatectomia de aproximadamente 67% da víscera. Os animais do grupo experimento receberam por via subcutânea 0,5 mg/Kg/dia de histamina e os do grupo controle igual volume de solução salina isotônica. As aferições foram realizadas com 36 horas e 7 dias. A avaliação do peso da víscera não mostrou diferença entre os grupos. O número de figuras de mitose em 10 campos foi maior no grupo experimento com 36 horas (p=0,010). No sétimo dia o número delas era semelhante nos dois grupos. Concluiu-se que a administração de histamina exógena, talvez pela sua vida média curta, aumenta o número de figuras de mitose no início do processo, não interferindo na regeneração ao final de 7 dias.

Many substances have been used after partial hepatectomy in order to determine how they act on the process of hepatic regeneration. An adequate blood flow seems to be very important in this process. Increased plasma histamine levels after hepatectomy in rats have been reported by some investigators. Substances that inhibit histidine-decarboxylase and histaminase lead to an elevation in endogenous histamine with sinusoid vasodilation and acceleration of hepatic regeneration. The objective of the present study was to determine the effect of exogenous histamine administered to partially hepatectomized rats. Thirty two Wistar rats were submitted to approximately 67% hepatectomy. The animals in the experimental group received subcutaneously 0.5 mg/kg/day of histamine and the controls received an equal volume of isotonic saline. Determinations were made after 36 hours and after 7 days. Liver weight did not differ between groups. The number of mitotic figures in 10 fields was higher in the experimental group after 36 hours (p=0.010). On the seventh day, the number of mitotic figures was similar for both groups. We conclude that the administration of exogenous histamine, perhaps due to the short half-life of the substance, increases the number of mitotic figures at the beginning of the process but has no effect on regeneration after 7 days.
