Abstract: Soybean seeds are crucial for global food production. Various factors affect the quality of soybean seeds, including mechanical damage, which can lead to reduced germination potential and decreased seedling vigor. The presence of lignin in the seed coat contributes to resistance to mechanical damage. However, the relationship between the monomeric composition of lignin and mechanical damage is unknown. To fill this gap, we evaluated the contents of monomers hydroxyphenyl (H), guaiacyl (G), and syringyl (S) in seed coats of three cultivars of soybean, namely, Doko, IAS-5, and Savana. The results revealed that the monomeric composition of lignin varied between resistant and susceptible cultivars. The levels of G and S monomers were inverse in the cultivars Doko and Savana, suggesting that the composition of lignin monomers may play a crucial role in the resistance of soybean seeds to mechanical damage. In addition, negative linear regressions between lignin and S monomer contents and S/G ratios could be helpful as an alternative to identify resistance in soybean seeds.

Resumo: Sementes de soja são cruciais para a produção global de alimentos. Vários fatores afetam a qualidade das sementes de soja, entre eles os danos mecânicos, que podem levar à redução do potencial germinativo e diminuição do vigor das plântulas. A presença de lignina no tegumento da semente contribui para a resistência aos danos mecânicos. No entanto, a relação entre a composição monomérica da lignina e o dano mecânico é desconhecida. Para preencher esta lacuna, nós avaliamos os teores de monômeros de hidroxifenil (H), guaiacil (G) e siringil (S) em tegumentos de três cultivares de soja, Doko, IAS-5 e Savana. Os resultados revelaram que a composição monomérica da lignina variou entre cultivares resistentes e suscetível. Os teores dos monômeros G e S foram inversos nas cultivares Doko e Savana, sugerindo que a composição dos monômeros de lignina pode desempenhar um papel crucial na resistência das sementes de soja aos danos mecânicos. Além disso, regressões lineares negativas entre os teores de lignina e monômero S e as relações S/G podem ser uma alternativa para identificar resistência em sementes de soja.

Glyphosate resistant transgenic soybean [Glycine max (L.) Mettill] expresses a glyphosate insensitive EPSPS (5-enolpyruvylshikimate-3-phosphate synthase). This enzyme is involved in important secondary metabolism pathways, including lignin biosynthesis. Thus, differences in lignin content and growth between susceptible (OC14) and glyphosate-resistant soybean may be observed. The objective of this study was to evaluate differences between growth and lignin content of roots in transgenic and OC14 soybean cultivated at high temperatures. Seeds from the OC14 soybean and its transgenic cultivar, CD213RR, were germinated at 25, 27.5, 30 and 32.5 °C. After 3 d, seedlings were cultivated in Hoagland half-strength nutrient solution during 12 h photoperiods and at the same germination temperature. After 4 d, roots' relative length, fresh and dry biomass, and lignin content were determined. Increasing temperatures promoted root growth. There was decreased growth and higher lignin content in roots in CD213RR soybean as compared to OC14. The transgenic soybean may present a different lignin metabolism since it showed higher lignification independently of temperatures, which is important because can be associated to a higher tolerance to drought and heat, but also with impaired growth and higher susceptibility to breakage of the stem.

La soya transgénica [Glycine max (L.) Merrill] resistente al glifosato presenta glifosato EPSPS (5-enolpiruvilsiquimato-3-fosfato sintasa) insensible. Esta enzima participa en procesos importantes del metabolismo secundario, como la biosíntesis de la lignina. Así, se pueden observar diferencias en el contenido de lignina y crecimiento entre la soya susceptible (OC14) y la resistente al glifosato. El objetivo de este estudio fue evaluar las diferencias entre el crecimiento y el contenido de lignina de las raíces de la soya transgénica y la OC14, cultivadas con temperaturas altas. Las semillas de la soya OC14 y su cultivo transgénico, CD213RR, germinaron a 25, 27.5, 30 y 32.5 °C. Después de 3 d, las plántulas se cultivaron en una solución de nutrientes Hoagland de fuerza media, durante fotoperíodos de 12 h y a la misma temperatura de germinación. Después de 4 d se determinó la longitud relativa de las raíces, la biomasa fresca y seca, y el contenido de lignina. Las temperaturas cada vez más altas promovieron el crecimiento radicular. Hubo una disminución de crecimiento y mayor contenido de lignina en las raíces de la soya CD213RR en comparación con OC14. La soya transgénica puede presentar un metabolismo de la lignina diferente, ya que mostró lignificación mayor independientemente de la temperatura, lo cual es importante porque puede estar relacionado con la tolerancia mayor a la sequía y al calor, pero también con el retraso del crecimiento y mayor susceptibilidad a la rotura del tallo.

Tocoferóis são conhecidos constituintes da vitamina E, sendo os principais antioxidantes lipofílicos na soja. Eles são substâncias antioxidantes naturais e estabilizadoras, capazes de inibir a degradação de lipídios limitando a oxidação não enzimática destes compostos durante o armazenamento das sementes, a germinação e o desenvolvimento inicial das plântulas. O objetivo deste trabalho foi determinar o conteúdo de α-tocoferol nas sementes de quarto cultivares de soja envelhecidas natural e artificialmente. As sementes de quatro cultivares de soja envelhecidas em armazenamento de zero a 180 dias em câmara seca (envelhecimento natural) e submetidas a altas temperaturas e umidade relativa (envelhecimento artificial) foram analisadas quanto ao teor de α-tocoferol. A quantificação de α-tocoferol nas sementes de soja foi efetuada por cromatografia líquida de alta eficiência (CLAE) associada ao método de extração Soxhlet. Diferenças significativas nos teores de α-tocoferol nas sementes foram observadas para todas as cultivares e tempos de armazenamento. O teor de α-tocoferol das sementes de soja mostrou resposta linear crescente com o aumento dos períodos de armazenamento. Entretanto, as sementes envelhecidas artificialmente apresentaram maior teor de α-tocoferol do que as envelhecidas naturalmente.

Tocopherols are well known constituents of vitamin E, and the main antioxidants in soybean. Are natural antioxidants and stabilizers that can inhibit lipid degradation, reducing non-enzymatic oxidation of these compounds during storage of seeds, germination and initial development of seedlings. The objective of this work was to determine the level of α-tocopherol in four soybean seeds cultivars naturally and artificially aged. Seeds of four soybean cultivars stored from 0 to 180 days in a dry chamber (natural aging) and subjected to high temperature and humidity (artificial aging) were analyzed for α-tocopherol content. The quantification of α-tocopherol in the soybean seeds was performed by high performance liquid chromatography (HPLC) combined with the Soxhlet extraction method. Significant differences in α-tocopherol levels in seeds were observed for all cultivars and storage times. The α-tocopherol contents of the soybean seeds showed linear correlations with an increasing period of storage for all the cultivars studied. However, the artificially aged seeds had a higher content of α-tocopherol than those naturally aged.

A resistência ao glifosato é conferida à soja (Glycine max L. Merrill) pela incorporação de um gene que codifica a enzima CP4-EPSP sintase, uma variante da EPSP sintase, insensível ao glifosato, que atua na via do chiquimato/corismato, importante rota metabólica envolvida na lignificação. O objetivo deste trabalho foi o de investigar o crescimento e os teores de lignina nas raízes de cultivares de soja, suscetíveis (CD 201 e OC 14) e resistentes (CD 214RR e CD 213RR) ao glifosato. Para isso, plântulas com três dias de desenvolvimento foram cultivadas em solução nutritiva de Hoagland, meia-força (pH 6,0), em câmara de crescimento (25°C, fotoperíodo de 12 h, irradiância de 280 μmol m-2 s-1) de 24 a 96h. Os resultados revelaram que as cultivares resistentes ao glifosato (CD 213RR e CD 214RR) apresentaram elevado crescimento das raízes quando comparadas com as cultivares convencionais (OC-14 e CD 201). A cultivar CD 213RR apresentou altos teores de lignina e reduzido crescimento das raízes em comparação com a cultivar convencional (OC 14). O mesmo não foi observado nas cultivares CD 214RR e CD 201. Com base nos resultados, é possível concluir que (1) uma forma diferente de EPSP sintase pode interferir na via de fenilpropanoides e, posteriormente, na síntese de lignina, e (2) outras características genéticas inerentes a cada cultivar pode afetar os conteúdos de lignina nas raízes, haja vista que a lignificação na cultivar CD 214RR não foi afetada de similar maneira que na cultivar CD 213RR.

Glyphosate resistance is conferred to soybean (Glycine max L. Merril) by incorporating a gene encoding a glyphosate-insensitive enzyme (CP4-EPSP synthase) that acts in the shikimate/chorismate pathway, an important metabolic route in the lignification process. The aim of this work was to investigate the root growth and lignin contents of susceptible (CD 201 and OC 14) and glyphosate-resistant (CD 214RR and CD 213RR) soybean cultivars. To this end, three-day-old seedlings were cultivated in half-strength Hoagland nutrient solution (pH 6.0) in a growth chamber (25°C, 12-h photoperiod, irradiance of 280 μmol m-2 s-1) for 24 to 96 hours. The results revealed that glyphosate-resistant (CD 213RR and CD 214RR) cultivars showed high root growth when compared to the conventional (OC-14 and CD 201) cultivars. CD 213RR showed high root lignin content and reduced root weight compared to the conventional (OC 14) cultivar, although CD 214RR and CD 201 did not follow the same trend. Based on these results, it is possible to conclude that (1) the different form of EPSP synthase encoded in RR soybean may interfere in phenylpropanoid pathway and further in lignin biosynthesis, and (2) other genetic characteristics inherent to each cultivar may affect roots lignin content in soybean seedlings since lignification in CD 214 RR was not affected of similar manner than cultivar CD 213 RR.

Os efeitos de naringenina, um intermediário da biossíntese de flavonóides, foram avaliados sobre o crescimento das raízes, as atividades da fenilalanina amônia liase (PAL) e peroxidases, bem como sobre os teores de compostos fenólicos e de lignina em plântulas de soja (Glycine max L. Merrill). Plântulas de três dias foram cultivadas em solução nutritiva de Hoagland, meia-força (pH 6,0), contendo ou não, naringenina 0,1 a 0,4 mM, em uma câmara de germinação (25°C, fotoperíodo de 12 h, 280 µmol m-2 s-1) durante 24 h. Efeitos inibitórios no crescimento das raízes (comprimento, massa e viabilidade celular) e nas atividades da PAL e POD solúvel foram constatados após os tratamentos com naringenina. Estes efeitos foram associados com atividade estimulatória da POD ligada à parede celular, seguido por aumento nos teores de lignina, sugerindo que a inibição do crescimento das raízes pode ser devido ao processo de lignificação.

The flavanone naringenin, an intermediate in flavonoid biosynthesis, was tested for its effect on root growth, phenylalanine ammonia-lyase (PAL) and peroxidase (POD) activities, as well as phenolic compounds and lignin contents in soybean (Glycine max L. Merrill) seedlings. Three-day-old seedlings were cultivated in half-strength Hoagland nutrient solution (pH 6.0), with or without 0.1 to 0.4 mM naringenin in a growth chamber (25°C, 12-h photoperiod, irradiance of 280 µmol m-2 s-1) for 24 h. Inhibitory effects on root growth (length, weight, cell viability), PAL and soluble POD activities were detected after naringenin treatments. These effects were associated with stimulatory activity of the cell wall-bound POD followed by an increase in the lignin contents, suggesting that naringenin-induced inhibition in soybean roots could be due to the lignification process.

O objetivo deste trabalho foi determinar o teor de metilxantinas, cafeína e teobromina, e de compostos fenólicos, ácido clorogênico e ácido cafeico, em 51 progênies de erva-mate e estimar componentes de variância e herdabilidade. As progênies de erva-mate eram oriundas de cinco municipios brasileiros: Pinhão, Ivaí, Barão do Cotegipe, Quedas do Iguaçu e Cascavel. Essas progênies foram cultivadas na localidade de Ivaí. O conteúdo dos compostos foi obtido por cromatografia líquida de alta eficiência (CLAE). Na estimativa dos componentes da variância e dos parâmetros genotípicos, utilizou-se a metodologia de modelos mistos para a obtenção da melhor predição linear não viciada (BLUP) dos efeitos genotípicos e o processo da máxima verossimilhança restrita (REML), processados pelo programa Selegen - REML/BLUP. Os conteúdos de cafeína (0,248-1,663 %) e teobromina (0,106-0.807%) foram significativamente (p<0,05) diferentes em relação à localidade de origem, com elevada herdabilidade individual ( ĥ²>0,5). Foram determinados dois diferentes grupos de progênie para ácido clorogênico (1,365-2,281%) e ácido cafeico (0,027-0,037%), e seu conteúdo não foi significativamente diferente (p<0,05) em relação à localidade de origem. Os valores de herdabilidade individual foram de baixos a médios para ácido clorogênico ( ĥ²<0,4) e ácido cafeico ( ĥ²<0,3). O conteúdo dos compostos e os valores dos parâmetros genéticos poderão subsidiar os programas de melhoramento da erva-mate.

The objective of this work was to determine the contents of methylxanthines, caffeine and theobromine, and phenolic compounds, chlorogenic and caffeic acids, in 51 mate progenies (half-sib families) and estimate the heritability of genetic parameters. Mate progenies were from five Brazilian municipalities: Pinhão, Ivaí, Barão de Cotegipe, Quedas do Iguaçu, and Cascavel. The progenies were grown in the Ivaí locality. The contents of the compounds were obtained by high performance liquid chromatography (HPLC). The estimation of genetic parameters by the restricted maximum likelihood (REML) and the prediction of genotypic values via best linear unbiased prediction (BLUP) were obtained by the Selegen - REML/BLUP software. Caffeine (0.248-1.663%) and theobromine (0.106-0.807%) contents were significantly different (p<0.05) depending on the region of origin, with high individual heritability (ĥ²>0.5). The two different progeny groups determined for chlorogenic (1.365-2.281%) and caffeic (0.027-0.037%) acid contents were not significantly different (p<0.05) depending on the locality of origin. Individual heritability values were low to medium for chlorogenic (ĥ²<0.4) and caffeic acid (ĥ²<0.3). The content of the compounds and the values of genetic parameters could support breeding programs for mate.

A proposta do presente trabalho foi investigar como os ácidos p-cumárico (p-CA) e p-hidroxibenzóico (p-HD) afetam a atividade da peroxidase (POD, EC 1.11.1.7), a peroxidação lipídica (LP) e o crescimento de raízes de soja (Glycine max (L.) Merr.). Plântulas de três dias foram cultivadas em solução nutritiva com p-CA ou p-HD (0,1 a 1 mM) por 48 horas. Após absorção, ambos os compostos (a 0,5 e 1 mM) reduziram o comprimento das raízes (RL), a biomassa fresca (FW) e a biomassa seca (DW) enquanto aumentaram a atividade da POD solúvel, a atividade da POD ligada à parede celular (com p-CA 1 mM e p-HD 0,5 mM), e a LP.

The scope of the present study was to investigate how the p-coumaric (p-CA) and p-hydroxybenzoic (p-HD) acids affect the peroxidase (POD, EC 1.11.1.7) activity, the lipid peroxidation (LP) and the root growth of soybean (Glycine max (L.) Merr.). Three-day-old seedlings were cultivated in nutrient solution containing p-CA or p-HD (0.1 to 1 mM) for 48 h. After uptake, both compounds (at 0.5 and 1 mM) decreased root length (RL), fresh weight (FW) and dry weight (DW) while increased soluble POD activity, cell wall (CW)-bound POD activity (with 1 mM p-CA and 0.5 mM p-HD) and LP.

The influence of the allelochemicals ferulic (FA) and vanillic (VA) acids on peroxidase (POD, EC 1.11.1.7) and phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) activities and their relationships with phenolic acid (PhAs) contents and root growth of soybean (Glycine max (L.) Merr.) were examined. Three-day-old seedlings were cultivated in nutrient solution containing FA or VA (0.1 to 1 mM) for 48 h. Both compounds (at 0.5 and 1 mM) decreased root length (RL), fresh weight (FW) and dry weight (DW) and increased PhAs contents. At 0.5 and 1 mM, FA increased soluble POD activity (18% and 47%, respectively) and cell wall (CW)-bound POD activity (61% and 34%), while VA increased soluble POD activity (33% and 17%) but did not affect CW-bound POD activity. At 1 mM, FA increased (82%) while VA reduced (32%) PAL activities. The results are discussed on the basis of the role of these compounds on phenylpropanoid metabolism and root growth and suggest that the effects caused on POD and PAL activities are some of the many mechanisms by which allelochemicals influence plant growth

A técnica de depleção foi usada para determinar como raízes de soja consomem os ácidos ferúlico e p-hidroxibenzóico em solução nutritiva. Os resultados mostraram que extremidades excisadas de raízes de soja consumiram rapidamente estes aleloquímicos. Entre concentrações de 0,1 e 1,0 mM, 50% ou mais de ácido ferúlico foi consumido após 6 h de depleção. Embora o consumo tenha iniciado somente após 3 h, comportamento idêntico foi encontrado com ácido p-hidroxibenzóico.

Depletion method was used to describe how soybean root consumed ferulic and p-hydroxybenzoic acids in nutrient culture. The results showed that excised cultured soybean roots consumed these allelochemicals very rapidly. At concentrations between 0.1 and 1.0 mM, roots consumed 50% or more of ferulic acid after 6-h depletion. Although consumption started only after 3 h, similar behavior was verified with p-hydroxybenzoic acid.

Plântulas de soja cultivadas em meio nutriente foram utilizadas para investigar como glicose ou metionina influenciam a depleção de ácido ferúlico. Plântulas de três dias foram cultivadas em meio hidropônico contendo ácido ferúlico e glicose ou metionina e os níveis de ácido ferúlico foram monitorados no meio nutriente. Os resultados mostraram que as raízes de soja depletaram ácido ferúlico mais rapidamente na presença dos compostos. Após 6 horas, o aumento causado pela glicose (0,01 e 0,05 mM) foi mais pronunciado que na presença de metionina, nas mesmas concentrações. Por seu lado, metionina, a 0,1 e 0,2 mM, aumentou a depleção mais significativamente que a glicose. Os resultados sugerem que ambos os compostos podem aumentar os efeitos alelopáticos do ácido ferúlico nas plântulas.

Cultured soybean seedlings were used to investigate how glucose or methionine influenced depletion of ferulic acid. Three-day-old seedlings were grown in hydroponic solution containing ferulic acid plus glucose or methionine, and the level of the phenolic acid were monitored in the nutrient culture. The results showed that ferulic acid depletion was more rapid in the presence of those compounds. After 6 h, the increase caused by glucose (0.01 and 0.05 mM) was more pronounced than methionine in the same concentrations. On the other hand, methionine (0.1 and 0.2 mM) increased depletion more significantly than glucose. Results suggested that both compounds might to increase the allelopathic effects of ferulic acid in the seedlings.
