A compreensão do mecanismo de redução de compostos com potencial atividade antichagásica pode contribuir para o melhor entendimento de sua ação biológica. Com este objetivo, a redução eletroquímica de nitrofural (nitrofurazona, NF) e seus análogos, nitrofurfurilideno tiossemicarbazona (NFS), nitrotiofeno semicarbazona (NT), nitrotiofeno tiossemicarbazona (NTS), foi estudada pelas voltametrias de pulso diferencial, cíclica e cronoamperometria em meio aquoso, utilizando eletrodo de carbono vítreo. Esses três análogos sintéticos apresentam átomos de oxigênio e de enxofre distribuídos no anel heterocíclico (furano e tiofeno, respectivamente) e no grupo carbonila (semicarbazona e tiossemicarbazona, respectivamente). A hidroxilamina é o principal produto formado em meio ácido, sendo sua formação linearmente dependente do pH. A cronoamperometria mostrou que esta redução envolve 4 elétrons. Entretanto, em meio alcalino, esta onda de redução é desdobrada e um pico reversível a potencial mais positivo foi registrado, correspondendo à formação nitro ânion radical. O nitro ânion radical sofre desproporcionamento e sua estabilidade cinética foi avaliada aplicando a razão de corrente correspondente ao par reversível R - NO2/R - NO2• - e as constantes de segunda ordem (k2) foram determinadas. A partir dos valores de k2 observou-se que o análogo NTS registrou a maior estabilidade.

The biological action of drugs with potential antichagasic activities can be better understood by knowing their reduction mechanism. For this proposal, the electrochemical reduction of nitrofurazone (NF) and its analogues, nitrofurfurilidene thiosemicarbazone (NFS), nitrothiophene semicarbazone (NT), nitrothiophene thiosemicarbazone (NTS), was studied by differential pulse and cyclic voltammetries and chronoamperometry in aqueous medium by using a glassy carbon electrode. Those three synthetic analogues have oxygen and sulfur atoms arranged between the nitroheterocyclic ring (furan and thiophene, respectively) and the carbonyl group (semicarbazone and thiosemicarbazone, respectively). The respective hydroxylamine derivatives are the main products formed in acidic medium, being its formation linearly pH dependent. Although chronoamperometric data show that this reduction involves 4 electrons, its corresponding reduction voltammetric wave, in alkaline medium, is unfolded and a reversible reduction peak at a more positive potential appears. This peak is characterized as a nitro anion radical formation, whose decay is caused by a disproportionation reaction. Its kinetic stability was studied by the current ratio values of the R - NO2/R - NO2• - redox couple and the second-order constant (k2) was determined, being NTS the analogue that registered the highest stability.

A malária é a endemia tropical mais devastadora do mundo e esse quadro é agravado pela ausência de tratamento eficaz. Entretanto, a resistência dos plasmódios à artemisinina não apresenta relevância clínica e seu mecanismo de ação está associado ao grupo heme, com formação de radicais livres e rompimento da ponte endoperóxido. O comportamento voltamétrico da artemisinina foi estudado por voltametria cíclica e voltametria de onda quadrada. O fármaco é irreversivelmente reduzido em eletrodos de carbono vítreo e os valores de potencial de pico não sofrem influência da acidez do meio, porém observou-se o maior valor de corrente em pH 6,0. O comportamento voltamétrico da artemisinina foi significativamente alterado na presença do grupo heme, provocando uma antecipação de seu pico de redução em cerca de 600 mV. Por voltametria de onda quadrada observou-se que este novo pico foi sensível à adição crescente de concentração de hemina, atingindo valor de corrente cerca de 10 vezes maior em relação ao pico original da artemisinina, numa relação de concentração de 20 mmol/L para o primeiro e 50 mmol/L do segundo. Além disso, resultados indicaram que esse processo eletrocatalítico ocorreu pela formação de Fe(II)-hemina na superfície do eletrodo, com provável processo de eletro-polimerização da hemina sobre o eletrodo de carbono vítreo. Esse efeito adsortivo foi avaliado a partir da estimativa da concentração superficial (G) de hemina sobre o eletrodo de trabalho em pH 6,0. A modificação do eletrodo de carbono vítreo por hemina mostrou que a interação entre artemisinina e o grupo heme ocorre predominantemente sobre a superfície do eletrodo e não em solução. Portanto, esclarecer o mecanismo de ação da artemisinina é importante para o planejamento e desenvolvimento de novos agentes antimaláricos.

Malaria is the tropical disease most devastating of the world and this situation is worsened by the absence of effective treatment. However, the plasmodium resistance to artemisinin does not show clinical relevance. The drug mechanism of action is associated to the heme group, with free radical formation and endoperoxide moiety breakage. The voltammetric behavior of artemisinin was studied by cyclic and square-wave voltametries. This drug was irreversibly reduced on glassy carbon electrode and the peak potential values are pH independent, however the biggest value of current peak was observed at pH 6.0. The voltammetric behavior of artemisinin was significantly changed in the heme group presence, provoking an anticipation of about 600 mV on cathodic peak. By square-wave voltammetry it was observed that this new peak was sensitive to the hemin concentration, reaching a value around 10 times larger regarding the original cathodic peak of artemisinin, being the concentration of 20 mmol/L for the former and 50 mmol/L for the latter. In addition, results indicated that this electro-catalytic process depends on the Fe(II)-hemin formation on the electrode surface, indicating the possible electro-polymerization of hemin on the glassy carbon electrode. This adsorptive effect was evaluated from the superficial concentration (G) estimation of the hemin on the working electrode at pH 6.0. The modification of the glassy carbon electrode using hemin showed that the interaction between artemisinin and the heme group predominantly occurs on the electrode surface and not in solution. Therefore, clarifying artemisinin mechanism of action is important in order to contribute for the design and development of new antimalarial agents.

A doença de Chagas é um grave problema de saúde pública para a América Latina, situação agravada pela inexistência de quimioterapia eficiente. Os dois fármacos comercialmente encontrados, benznidazol e nifurtimox, são eficazes apenas na fase aguda da doença. A nitrofurazona é ativa contra Trypanosoma cruzi, entretanto, a alta toxicidade impede seu uso na parasitose. A hidroximetilnitrofurazona é um pró-fármaco da nitrofurazona, que apresenta maior atividade contra Trypanosoma cruzi, além de ser menos tóxico. Estudou-se o comportamento voltamétrico da nitrofurazona por voltametria cíclica, varredura linear e pulso diferencial, comparando-o ao do metronidazol e do cloranfenicol. Os coeficientes de difusão dos três fármacos foram estimados aplicando-se a equação de Wilke-Chang. Este artigo também apresenta o estudo do derivado hidroximetilnitrofurazona por voltametria cíclica. A redução de nitrofurazona é pH-dependente e em meio ácido o derivado hidroxilamínico, envolvendo quatro elétrons, é o principal produto formado. Em meio alcalino e com prévio tratamento do eletrodo de carbono vítreo, a redução de nitrofurazona ocorre em duas etapas: a primeira envolve um elétron para formar o nitro-radical aniônico e a segunda etapa corresponde à formação da hidroxilamina. Hidroximetilnitrofurazona possui comportamento voltamétrico semelhante e, de forma análoga, apresentou a mesma eletroatividade e capacidade de estabilização do nitro-radical, indicando que a modificação molecular de nitrofurazona não introduziu alterações no seu comportamento voltamétrico. Uma breve discussão das diferenças de atividade biológica entre os compostos também é apresentada.

Chagas' disease is a serious health problem for Latin America. The situation is worsened by the lack of efficient chemotherapy. The two available commercial drugs, benznidazole and nifurtimox, are more effective in the acute phase of the disease. Nitrofurazone is active against Trypanosoma cruzi, however its high toxicity precludes its current use in parasitosis. Hydroxymethylnitrofurazone is a prodrug of nitrofurazone. It is more active against Trypanosoma cruzi than nitrofurazone, besides being less toxic. This work shows the voltammetric behavior of nitrofurazone and a comparison with those of metronidazole and chloramphenicol using cyclic, linear sweep and differential pulse voltammetries. For these drugs also the prediction of the diffusion coefficients using Wilke-Chang equation was performed. The reduction of nitrofurazone is pH-dependent and in acidic medium the hydroxylamine derivative, involving four electrons, is the principal product formed. In aqueous-alkaline medium and with a glassy carbon electrode pre-treatment the reduction of nitrofurazone occurs in two steps, the first involving one electron to form the nitro-radical anion and the second corresponding to the hydroxylamine derivative formation. Hydroxymethylnitrofurazone presented the same voltammetric behavior and electroactivity, indicating that the molecular modification performed in nitrofurazone did not change its capacity to be reduced. A brief discussion regarding the differences in biological activity between the two compounds is also presented.
