ABSTRACT The aim of this study was to determine the composition of gastrointestinal content and protease and lipase activities in summer and winter as well as to evaluate the relationship between digestive enzyme activity and centesimal composition of gastrointestinal content and feeding habits of two omnivorous species, Rhamdia quelen (Quoy & Gaimard, 1824) and Pimelodus maculatus (LaCèpede, 1803), and of two detritivorous species, Loricariichthys anus (Valenciennes, 1835) and Hypostomus commersoni (Valenciennes, 1836). The activities of pepsin, trypsin, chymotrypsin, and lipase, and the levels of proteins and lipids in the gastrointestinal tract, were evaluated. The enzyme activities were not related to the centesimal composition of gastrointestinal content or feeding habits. This finding could be associated with the variations of nutrient availability over time in the environment, as was observed in the centesimal composition of food ingested by the fish in summer and winter. The analyzed enzymes exhibited a constitutive character in these species; that is, the digestive enzymes are always available in the gastrointestinal tract to digest any food that the fish may find as an adaptation to better utilize the nutrients available in the environment in winter and summer.

RESUMO: O objetivo deste estudo foi verificar a atividade de algumas enzimas digestivas em quatro espécies de peixes com hábitos alimentares diferentes. O conhecimento das atividades dessas enzimas pode nos ajudar a compreender os processos digestivos das espécies. As espécies escolhidas para este estudo foram Ctenopharyngodon idella (herbívoro), Rhamdia quelen (onívoro), Leporinus obtusidens (onívoro) e Hoplias malabaricus (carnívoro). O trato digestivo dessas espécies foi dividido em quatro partes para estimar a atividade enzimática: estômago, intestino anterior, médio e posterior. Ctenopharyngodon idella teve a maior atividade da amilase e maltase em todas as porções do intestino, seguido por L. obtusidens . A maior atividade da tripsina foi observada em todas as porções do intestino de H. malabaricus , seguido pelo intestino médio de L. obtusidens e o intestino anterior de C. idella . A maior atividade de quimotripsina foi encontrada em todas as partes do intestino do C. idella , seguida pelo intestino anterior de R. quelen, L. obtusidens e H. malabaricus . No estômago, a atividade de protease ácida foi significativamente menor em R. quelen e L. obtusidens comparado com H. malabaricus. Ctenopharyngodon idella mostrou alta atividade de enzimas que hidrolisam carboidratos, representadas neste estudo por amilase e maltase e H. malabaricus mostrou maior atividade de proteases e baixa atividade de amilase.

ABSTRACT: The aim of this study was to verify the activity of some digestive enzymes in four fish species with different feeding habits. Knowledge of these enzymatic activities can help us to understand the species' digestive processes. The species chosen for this study were Ctenopharyngodon idella (herbivore), Rhamdia quelen (omnivore), Leporinus obtusidens (omnivore) and Hoplias malabaricus (carnivore). The digestive tract of these species was divided into four portions to estimate enzymatic activity: stomach, anterior, mid and posterior intestine. Ctenopharyngodon idella had the highest amylase and maltase activities in all portions of the gut, followed by L. obtusidens . The highest trypsin activity was observed in all gut portions of H. malabaricus, followed by the mid intestine of L. obtusidens and the anterior intestine of C. idella . The highest chymotrypsin activity was found in all portions of C. idella followed by the anterior intestines of R. quelen, L. obtusidens and H. malabaricus . In the stomach, acid protease activity was significantly lower in R. quelen and L. obtusidens compared to H. malabaricus. Ctenopharyngodon idella showed high activity of enzymes that hydrolyze carbohydrates, represented in this study by amylase and maltase and H. malabaricus showed higher protease activity and low amylase activity.

Devido à intensa atividade agrícola no rio Uruguai (Sul do Brasil), há potencial para contaminação aquática por agrotóxicos. Há muitos reservatórios para atender a demanda de água de campos de arroz, formando ambientes lênticos. De acordo com estas informações, o objetivo do presente estudo foi mostrar uma análise comparativa de alguns biomarcadores como a peroxidação lipídica (TBARS), glutationa S-transferase (GST), tióis não-protéicos (NPSH), aminoácidos (AA) e teste písceo de micronúcleos (MNE) em Astyanaxjacuhiensis amostrados em ambientes lóticos e lênticos da região do médio rio Uruguai, comparando estações quentes e frias. Oito pesticidas foram encontrados em amostras de água, sendo propoxur a maior concentração encontrada em ambos os ambientes e estações. Peixes da estação quente apresentaram maiores níveis de biomarcadores bioquímicos e peixes da estação fria apresentaram maiores níveis de MNE e AA. TBARS e AA apresentaram maiores níveis nos peixes de rio, enquanto GST, NPSH, MNE e AA apresentaram níveis mais elevados em peixes da represa. Estes ambientes têm características diferentes, com potencial redox, aeração, sedimentação, estrutura trófica, a entrada de agroquímicos e outros que podem afetar as respostas fisiológicas e bioquímicas de peixe contra situação adversa.

Due to intense agricultural activity in the rio Uruguai (South Brazil), there is the potential for aquatic contamination by agrochemicals. In this region, there are many reservoirs to meet the water demand for rice fields, forming lentic environments. In line with this information, the aim of this study was to show a comparative analysis of some biomarkers, such as lipid peroxidation (TBARS), gluthatione S-transferase (GST), non-protein thiols (NPSH), amino acids (AA) and piscine micronucleus tests (MNE) in Astyanax jacuhiensis from lentic and lotic environments in the middle rio Uruguai region, comparing warm and cold seasons. Eight pesticides were found in water samples, with propoxur having the highest concentration found in both environments and seasons. Fish from the warm season showed higher levels of biochemical biomarkers, and fish from the cold season showed higher levels of MNE and AA. TBARS and AA presented higher levels in fish from the river, while GST, NPSH, MNE and AA presented higher levels in fish from dams. These environments have different characteristics in terms of redox potential, aeration, sedimentation, trophic structure, agrochemicals input and others, which may affect the physiological and biochemical responses of fish in against adverse situations.

Rhamdia quelen (jundiá) e Leporinus obtusidens (piava) foram expostos a formulação comercial Roundup(r), um herbicida a base de glifosato nas concentrações de 0,2 e 0,4 mg/L por 96h. Os efeitos do herbicida foram analisados na atividade da alanina aminotransferase (ALT), aspartato aminotransferase (AST) e glicose no plasma, glicose e proteína na camada de muco, hidrólise de nucleotídeos no cérebro e a proteína carbonil no fígado. Os parâmetros foram escolhidos devido à falta de informação com relação a análises integradas, considerando parâmetros oxidativo, danos no fígado, efeitos na composição da camada do muco e atividade da trifosfato difosfoidrolase (NTPDase). Níveis de glicose plasmática foram reduzidos em ambas às espécies, enquanto a atividade das transaminases (ALT e AST) aumentou após exposição ao herbicida. A exposição ao herbicida aumentou a proteína e níveis de glicose na camada de muco em ambas as espécies. Houve uma redução em ambas atividades de NTPDase e ecto-5'-nucleotidase no cérebro de piava, e um aumentou a atividade destas enzimas em jundiás em ambas as concentrações testadas. As espécies mostraram um aumento na proteína carbonil no fígado após exposição a ambas as concentrações de glifosato. Nossos resultados demonstraram que a exposição ao Roundup(r) causou danos no fígado, como evidenciado pelo aumento das transaminases plasmáticas e proteína carbonil no fígado em ambas as espécies de peixes estudadas. A composição do muco alterou e uma hipoglicemia foi detectada após a exposição ao Roundup(r) em ambas espécies. A hidrólise de nucleotídeos em cérebro mostrou diferente resposta para cada espécie estudada. Esses parâmetros indicam alguns importantes e indicadores potenciais da contaminação do glifosato no ecossistema aquático.

Rhamdia quelen (silver catfish) and Leporinus obtusidens (piava) were exposed to a commercial formulation Roundup(r), a glyphosate-based herbicide at concentrations of 0.2 or 0.4 mg/L for 96 h. The effects of the herbicide were analyzed on the alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities and glucose in plasma, glucose and protein in the mucus layer, nucleotide hydrolysis in the brain, and protein carbonyl in the liver. The parameters were chosen, owing to a lack of information concerning integrated analysis, considering oxidative damage parameters, liver damage, and effects on the mucus layer composition and triphosphate diphosphohydrolase (NTPDase) activities. Plasmatic glucose levels were reduced in both species, whereas the transaminase activities (ALT and AST) increased after exposure to the herbicide. Herbicide exposure increased protein and glucose levels in the mucus layer in both species. There was a reduction in both NTPDase and ecto-5'-nucleotidase activity in the brain of piava, and increased enzyme activity in silver catfish at both concentrations tested. The species showed an increase in protein carbonyl in the liver after exposure to both concentrations of the glyphosate. Our results demonstrated that exposure to Roundup(r) caused liver damage, as evidenced by increased plasma transaminases and liver protein carbonyl in both of the fish species studied. The mucus composition changed and hypoglycemia was detected after Roundup(r) exposure in both species. Brain nucleotide hydrolysis showed a different response for each fish species studied. These parameters indicated some important and potential indicators of glyphosate contamination in aquatic ecosystems.

Os efeitos do glifosato, um herbicida não seletivo (1,0 ou 5,0mg L-1), sobre a atividade de enzimas digestivas (estômago e intestino) foram avaliadas em juvenis de piava (Leporinus obtusidens) após 90 dias de exposição. A atividade da protease ácida, tripsina, quimiotripsina e amilase aumentaram com a elevação da concentração de glifosato. Esses resultados indicam que o glifosato afeta a atividade de enzimas digestivas nesta espécie e pode ser indicador da reduzida disponibilidade de nutrientes, quando peixes sobrevivem em água contaminada com este herbicida.

The effects of glyphosate, a nonselective herbicide (1.0 or 5.0mg L-1) on digestive enzymes activity (stomach and intestine) were evaluated in juveniles of piava (Leporinus obtusidens) after 90 days of exposure. The activity of acid protease, trypsin, chymotrypsin and amylase increased with the increase of glyphosate concentration. These results indicate that glyphosate affects digestive enzyme activities in this species, and may be an indicator of poor nutrient availability when fish survive in herbicide-contaminated water.

As práticas agrícolas estão diretamente relacionadas com o uso de agrotóxico, que, de forma indiscriminada e sem o devido cuidado, podem contribuir para a ocorrência de inúmeras intoxicações. Diversos estudos demonstraram a relação de determinados pesticidas e a ocorrência do estresse oxidativo e, portanto, nos anos recentes, tem-se desenvolvido métodos de análise de diversos biomarcadores de dano celular, o qual pode ser medido e quantificado. Nesse contexto, o objetivo deste estudo foi avaliar as possíveis alterações nos marcadores bioquímicos: aspartato aminotransferase (AST), alanina aminotransferase (ALT), creatinina, ureia, proteína total, e marcadores oxidativos como: a peroxidação de lipídios, danos nas proteínas e a atividade das enzimas acetilcolinesterase (AChE) e catalase (CAT), em agricultores da cidade de Ibirubá, RS, expostos a diferentes agrotóxicos por pelo menos cinco anos. Com exceção da AST e da ALT, os resultados mostraram diferença significativa entre as médias das proteínas totais, ureia e creatinina, no grupo controle, mostrando que não houve alterações na função hepática ou renal dos trabalhadores rurais. Nos parâmetros oxidativos avaliados, houve uma diminuição da atividade da AChE e da CAT em relação ao grupo controle; um aumento das proteínas carboniladas e uma diminuição dos níveis de TBARS em relação ao grupo controle. Portanto, os resultados demonstram uma alteração oxidativa nos trabalhadores rurais, comparados com o grupo controle, principalmente pela possível inibição da AChE e ocorrência do estresse oxidativo, sem demonstrar mudança nos parâmetros bioquímicos analisados.

Agricultural practices are directly related to the use of pesticides, which indiscriminately and without due care may contribute to the occurrence of numerous intoxications. Several studies have demonstrated the relationship of certain pesticides and the occurrence of oxidative stress and therefore, in recent years have developed methods of analysis of several biomarkers of cellular damage that can be measured and quantified. In this context, the objective of this study was to evaluate the possible changes in biochemical markers: glutamic aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine, urea, total protein, and oxidative markers such as lipid peroxidation, damage to proteins and the activity of the enzymes acetylcholinesterase (AChE) and catalase (CAT) in farmers exposed to different pesticides for at least five years from Ibirubá - RS city. With the exception of AST and ALT, the results showed a significant difference between the mean total protein, urea and creatinine in the control group, showing that no changes in liver or kidney function of rural workers. In the oxidative parameters, there was a decrease in AChE activity and CAT in the control group; there were an increase in protein carbonyl and a decreased on TBARS compared to control group. Therefore, the results demonstrated a change in oxidative status of rural workers compared with the control group, mainly by possible inhibition of AChE activity and the occurrence of oxidative stress without showing changes in biochemical parameters.

Verificou-se a excreção de amônia em diferentes estágios de vida (ovos, larvas e juvenis) e determinou-se a influência do tempo de jejum na excreção de amônia no jundiá, Rhamdia quelen. Ovos e larvas foram coletados das incubadoras em diferentes tempos após a fecundação e colocados em recipientes. Juvenis foram separados em duas classes de peso (2-50 e 150-320 g) e após a alimentação foram colocados em recipientes individuais. As águas foram coletadas de cada recipiente para determinação da excreção de amônia. A excreção de amônia pelos ovos foi baixa, mas iniciada a eclosão - 28h após a fecundação, ela aumentou até as 48h. Nos juvenis em jejum, houve significativa correlação negativa entre a excreção de amônia e o peso. Além disso, a excreção de amônia diminuiu significativamente após 12 e 48h de jejum nos espécimes menores e maiores, respectivamente. Portanto, a renovação da água deve ser aumentada no momento da eclosão dos ovos de jundiá para evitar aumento da concentração de amônia. Como nessa espécie a excreção de amônia aumenta após a alimentação, quando os níveis nos tanques forem elevados a alimentação deve ser suspensa para evitar aumento adicional deste metabólito e mortalidade.

This study examined ammonia excretion at different life stages (eggs, larvae and juveniles) in silver catfish (Rhamdia quelen) and determined the influence of fasting time on ammonia excretion. Eggs and larvae were collected from incubators at different times after fecundation and placed in chambers. Juveniles were separated into two weight classes (2-50 g and 150-320 g) and placed in individual chambers after feeding. Water was collected from each chamber to determine ammonia excretion. Ammonia excretion by the eggs was low, but when hatching began approximately 28h after fecundation, excretion increased until 48h after fecundation. In fasting silver catfish juveniles, there was a significant negative correlation between ammonia excretion and weight. Moreover, ammonia excretion decreased significantly after 12 and 48h of fasting (compared to 6h fasting) in the smallest and largest specimens, respectively. Consequently, during incubation of silver catfish eggs, water renovation must be increased at hatching time to avoid a build-up in the concentration of ammonia. In addition, as ammonia excretion in this species increases after feeding, feed must be discontinued when ammonia levels in the tanks are high to avoid a further increase of this metabolite and consequent mortality of silver catfish.

O objetivo do presente estudo foi determinar o efeito da amônia não ionizada (NH3) da água na sobrevivência e no crescimento de jundiá, Rhamdia quelen. Os juvenis foram expostos a 0,10; 0,22 e 0,42mg L-1 NH3 em pH 8,2 durante 45 dias. Após 15 dias, uma mortalidade significativa (33%) foi observada nos exemplares expostos a 0,42mg L-1 NH3. Depois de 20 dias, 23% e 43% de mortalidade (ambas significativas) foram registradas nos jundiás mantidos em 0,22 e 0,42mg L-1 NH3, respectivamente. Uma relação negativa significativa entre sobrevivência, comprimento, ganho de peso diário, taxa de crescimento padrão e biomassa por tanque de juvenis de jundiá, com níveis de NH3 da água, foi encontrada ao final do experimento. A relação entre a sobrevivência e a amônia da água indicou que o nível máximo de NH3 em pH 8,2 para evitar qualquer mortalidade de jundiás em uma exposição crônica deve ser 0,01mg L-1.

The aim of the present study was to determine the effects of waterborne un-ionized ammonia (NH3) on the survival and growth of silver catfish (Rhamdia quelen). Juveniles were exposed to 0.10, 0.22, and 0.42mg L-1 NH3 at pH 8.2 for 45 days. After 15 days, a significant mortality rate (33%) was observed in the fish exposed to 0.42mg L-1 NH3. After 20 days, 23% and 43% mortality (both significant) was observed in fish exposed to 0.22 and 0.42mg L-1 NH3, respectively. A significant negative relationship between survival, length, daily weight gain, standard growth rate, and biomass of juveniles of silver catfish per tank with waterborne NH3 levels was found at the end of the experiment. The resulting survival and waterborne NH3 relationship indicated that, to avoid any mortality of silver catfish, the maximum level of chronic NH3 exposure at pH 8.2 would be 0.01mg L-1.

O objetivo deste estudo foi identificar o tempo de indução e recuperação anestésica de jundiás (Rhamdia quelen) expostos ao eugenol, bem como a eficácia desse anestésico na inibição do estresse e realizar análise sensorial dos filés dos peixes expostos a essa substância. Os jundiás foram expostos ao ar por um minuto para realização da biometria, e o sangue foi coletado zero, uma e quatro horas depois. O eugenol pode ser usado na faixa de 20-50mg L-1 para a indução da anestesia em jundiás, e o tempo de recuperação da anestesia não foi afetado pela concentração do eugenol. O grupo de controle mostrou níveis significativamente mais elevados do cortisol quatro horas após a biometria que no tempo zero. Os peixes anestesiados com eugenol (50mg L-1) apresentaram níveis significativamente mais baixos do cortisol plasmático do que peixes do grupo de controle do mesmo tempo. Esses dados indicam que o eugenol inibe o aumento do cortisol no sangue. O teste sensorial demonstrou que o eugenol modifica o sabor dos filés e consequentemente é contra-indicado para a anestesia do jundiá quando o filé for destinado ao consumo humano.

The aim of this study was to identify the time of anesthetic induction and recovery of silver catfish (Rhamdia quelen) exposed to eugenol. It was also determined the efficacy of the anesthetic as a stress reducing agent and performed a sensory analysis of the fillets from fish exposed to this substance. The silver catfish were exposed to air for 1min to carry out biometry, and blood was collected at 0, 1 and 4 hours later. Eugenol can be used in the range of 20-50mg L-1 for anesthetic induction in silver catfish, and recovery time from anesthesia was not affected by eugenol concentration. The control group showed significantly higher cortisol levels 4 hours after biometry than at time zero. Fish anesthetized with eugenol (50mg L-1) presented significantly lower plasma cortisol levels than control fish at the same time. These data indicate that eugenol inhibits the rise of cortisol in the blood. The sensory analysis test demonstrated that eugenol modifies the flavor of the fillet and therefore is contra-indicated for anesthetization of silver catfish that are intended for human consumption.

As enzimas digestivas influenciam a utilização dos alimentos em peixes, e seu conhecimento é importante para otimizar a formulação de dietas. Este trabalho descreve a atividade de enzimas digestivas em juvenis de jundiá alimentados com fontes protéicas. Os peixes foram alimentados com seis dietas (90 dias): MBY (farinha de carne e ossos + levedura de cana), SY (farelo de soja + levedura de cana), S (farelo de soja), MBS (farinha de carne e ossos + farelo de soja), FY (farinha de peixe + levedura de cana) e FS (farinha de peixe + farelo de soja). A cada 30 dias, foram analisadas as enzimas digestivas (tripsina, quimiotripsina e amilase) no intestino. No estômago, foi mensurada a protease ácida. Foram estimados os índices digestivo e hepato-somático, quociente intestinal, comprimento do trato digestório e ganho em peso. As atividades de tripsina e quimiotripsina foram maiores (p < 0,0001) nos peixes alimentados com as dietas com fontes de origem animal (MBY, MBS, FY e FS). As proteases alcalinas foram afetadas negativamente pelo farelo de soja nas dietas. A atividade de amilase apresentou grandes variações. Os peixes alimentados com as dietas MBY e MBS apresentaram maior atividade de protease ácida (p < 0,0001). Os ganhos em peso foram maiores nos peixes alimentados com as dietas MBS e FS (p < 0,05). Os maiores valores (p < 0,05) de comprimento do trato digestório foram verificados nos peixes alimentados com as dietas MBS e FY. A composição da dieta influencia na atividade de enzimas digestivas do jundiá.

Digestive enzymes activity influence feed utilization by fish, and its understanding is important to optimize diet formulation. This study reports the digestive enzyme activities of jundiá juveniles fed diets with protein sources. Fish were fed six experimental diets for 90 days: MBY (meat and bone meal + sugar cane yeast), SY (soybean meal + sugar cane yeast), S (soybean meal), MBS (meat and bone meal + soybean meal), FY (fish meal + sugar cane yeast) and FS (fish meal + soybean meal), and then sampled every 30 days and assayed in two intestine sections for digestive enzymes - trypsin, chymotrypsin and amylase - activities; gastric protease was assayed in the stomach. Digestive and hepatosomatic index, intestinal quotient, digestive tract length and weight gain were also measured. Trypsin and chymotrypsin activities were higher (p < 0.0001) in fish fed diets containing animal protein sources (MBY, MBS, FY and FS diets). Alkaline proteases were negatively affected by dietary soybean meal in the SY and S diets. Amylase activity had greater variation between diets and intestine sections. Fish fed MBY and MBS diets showed higher gastric protease activity (p < 0.0001). Weight gains were higher in fish fed the MBS and FS diets (p < 0.05). Highest values (p < 0.05) of digestive tract length were observed in fish fed MBS and FY diets, but no correlation with enzymes activities was found. Hepatosomatic index (HSI) was affected by diet composition. Dietary composition influenced digestive enzymes activities of jundiá.

O conteúdo iônico (Na+, K+ e Cl-) do plasma e da bile vesicular (BV) de juvenis de jundiá, Rhamdia quelen (156,1±0,2g, 28,2±0,3cm), foi determinado em três diferentes tempos (0, 6 e 24h) após exposição a: a) controle ou alto oxigênio dissolvido (OD = 6,5mg L-1) + baixa NH3 (0,03mg L-1); b) baixo OD (3,5mg L-1) + baixa NH3; c) alto OD + alta NH3 (0,1mg L-1); e baixo OD + alta NH3 . Alta concentração de amônia ou baixo oxigênio dissolvido na água aumentaram os níveis iônicos no plasma e na BV. Aparentemente, os efeitos osmorregulatórios desses parâmetros podem estar relacionados a mecanismos distintos, pois foi detectado efeito sinérgico sobre essa alteração osmorregulatória.

Ionic contents (Na+, K+ and Cl-) of plasma and gallbladder bile (GB) of juveniles silver catfish, Rhamdia quelen (156.1±0.2g, 28.2±0.3cm), were determined in three different times (0, 6 and 24h) after exposure to: a) control or high dissolved oxygen (DO = 6.5mg L-1) + low NH3 (0.03mg L-1); b) low DO (3.5mg L-1) + low NH3; c) high DO + high NH3 (0.1mg L-1); and d) low DO + high NH3. High waterborne NH3 or low DO levels increased plasma and GB ion levels. These parameters might have followed different mechanisms to affect osmoregulation since a synergic effect of these variables was detected.

No presente estudo, avaliou-se o efeito de diferentes níveis de saturação de oxigênio e amônia sobre a atividade das enzimas NTPDase (ecto-nucleosídeo trifosfato difosfohidrolase, E.C. 3.6.1.5) e acetilcolinesterase (AChE, E.C. 3.1.1.7) em encéfalo total de jundiás (Rhamdia quelen). Os peixes foram expostos a diferentes níveis de oxigênio dissolvido e amônia, níveis altos de oxigênio (6,5 mg/L) ou baixos de oxigênio (3,5 mg/L), e níveis altos de amônia (0,1 mg/L) ou baixos de amônia (0,03 mg/L). Os peixes foram divididos em quatro diferentes grupos: (A) controle ou alto nível de oxigênio dissolvido mais baixo nível de NH3; (B) baixo nível de oxigênio dissolvido mais baixo nível de NH3; (C) alto nível de oxigênio dissolvido mais alto nível de amônia; (D) baixo nível de oxigênio dissolvido mais alto nível de NH-3. As atividades de ambas as enzimas nos grupos C e D somente foram alteradas após 24 horas de exposição. A hidrólise do ATP e ADP em encéfalo total de jundiás foi aumentada após 24h de exposição para 104% e 155% no grupo D quando comparado ao grupo controle, respectivamente. A atividade da AChE apresentou após 24h de exposição um aumento de 37% no grupo C e 27% no grupo D, ambos comparados ao grupo controle. The effects of various levels of oxygen saturation and ammonia concentration on NTPDase (ecto-nucleoside triphosphate diphosphohydrolase, E.C. 3.6.1.5) and acetylcholinesterase (AChE, E.C. 3.1.1.7) activities in whole brain of teleost fish (Rhamdia quelen) were investigated. The fish were exposed to one of two different dissolved oxygen levels, including high oxygen (6.5 mg.L-1) or low oxygen (3.5 mg.L-1), and one of two different ammonia levels, including high ammonia (0.1 mg.L-1) or low ammonia (0.03 mg.L-1) levels. The four experimental groups included the following (A) control, or high dissolved oxygen plus low NH3; (B) low dissolved oxygen plus low NH3; (C) high dissolved oxygen plus high NH3; (D) low dissolved oxygen plus high NH3. We found that enzyme activities were altered after 24 h exposure in groups C and D. ATP and ADP hydrolysis in whole brain of fish was enhanced in group D after 24 h exposure by 100% and 119%, respectively, compared to the control group. After 24 h exposure, AChE activity presented an increase of 34% and 39% in groups C and D, respectively, when compared to the control group. These results are consistent with the hypothesis that low oxygen levels increase ammonia toxicity. Moreover, the hypoxic events may increase blood flow by hypoxia increasing NTPDase activity, thus producing adenosine, a potent vasodilator.

Exposição a inseticidas em concentrações elevadas no ambiente podem ocasionar efeitos adversos subletais em organismos aquáticos. Alevinos de carpa húngara (Cyprinus carpio, Linnaeus, 1758), carpa capim (Ctenopharyngodon idella, Valenciennes, 1844) e carpa cabeça grande (Aristichthys nobilis, Richardson, 1845) foram expostos ao diafuran, um inseticida utilizado na cultura do arroz no sul do Brasil. O objetivo deste estudo foi verificar a relação entre concentração letal mediana (CL50) do diafuran e a atividade da enzima acetilcolinesterase (AChE) em cérebro e músculo dessas espécies, como um possível biomarcador inicial da exposição a este inseticida. A CL50 foi determinada com peixes expostos a concentrações de diafuran em 96 h: carpa húngara: controle; 0,5; 1,0; 1,5; 2,0; 2,5 e 3,0 mg L-1; carpa capim: controle; 1,0; 2,0; 3,0 e 3,5 mg L-1 e carpa cabeça grande: controle; 0,5; 1,0; 1,5; 2,0; 3,0 e 4,0 mg L-1, bem como a determinação da AChE em concentrações próximas da CL50 para essas espécies. Valores de CL50 (concentrações nominais) foram de 1,81 mg L-1 para carpa húngara, 2,71 mg L-1 para carpa capim e 2,37 mg L-1 para carpa cabeça grande. Todas as carpas expostas ao diafuran estavam letárgicas (menores concentrações) ou imóveis. Diafuran inibiu significativamente a atividade da AChE em cérebro (~38 %) e músculo (~50 %) de todas as espécies estudadas. Atividade da AChE em músculo para carpa cabeça grande foi mais alta que cérebro (14,44 contra 5,94 µmol min-1 g proteína-1, respectivamente). Este estudo demonstrou que concentrações de diafuran utilizadas na cultura do arroz podem afetar o comportamento de Cyprinus carpio, Ctenopharyngodon idella e Aristichthys nobilis, e a atividade da acetilcolinesterase cerebral e muscular pode ser um biomarcador inicial de toxicidade deste inseticida.

Sublethal adverse effects may result from exposure of aquatic organisms to insecticides at environmentally relevant concentrations. Fingerlings of the common carp (Cyprinus carpio, Linnaeus, 1758), grass carp (Ctenopharyngodon idella, Valenciennes, 1844), and bighead carp (Aristichthys nobilis, Richardson, 1845) were exposed to diafuran, an insecticide widely used during rice cultivation in Southern Brazil. The aim of this study was to verify the relationship between the lethal concentration (LC50) of diafuran and the acetylcholinesterase (AChE) activity in brain and muscle tissues of these species as a possible early biomarker of exposure to this insecticide. LC50 was determined for fish exposed to diafuran concentrations during 96 h (short term): common carp: control, 0.5, 1.0, 1.5, 2.0, 2.5 and 3.0 mg L-1; grass carp: control, 1.0, 2.0, 3.0 and 3.5 mg L-1 and, bighead carp: control, 0.5, 1.0, 1.5, 2.0, 3.0 and 4.0 mg L-1, as well as the determination of AChE at concentrations near LC50 for these species. LC50 values (nominal concentrations) were 1.81 mg L-1 for the common carp, 2.71 mg L-1 for the grass carp and, 2.37 mg L-1 for the bighead carp. All carps exposed to diafuran were lethargic (lower concentrations) or immobile. Diafuran inhibited the acetylcholinesterase activity in brain (~38%) and muscle (~50%) of all species. Muscle of bighead carp under control treatment showed higher specific AChE activity than brain (14.44 against 5.94 µmol min-1 g protein-1, respectively). Concentrations of diafuran used for rice cropping may affect Cyprinus carpio, Ctenopharyngodon idella and Aristichthys nobilis behaviors and the AChE activities in brain and muscle of these species may be an early biomarker of toxicity of this insecticide.
