This study aimed the optimization of culture condition and composition for production of Cryptococcus laurentii 11 biomass and lipids in cheese whey medium supplemented with sugarcane molasses. The optimization of pH, fermentation time, and molasses concentration according to a full factorial statistical experimental design was followed by a Plackett-Burman experimental design, which was used to determine whether the supplementation of the culture medium by yeast extract and inorganic salts could provide a further enhancement of lipids production. The following conditions and composition of the culture medium were found to optimize biomass and lipids production: 360 h fermentation, 6.5 pH and supplementation of (g L-1): 50 molasses, 0.5 yeast extract, 4 KH2PO4, 1 Na2HPO4, 0.75 MgSO4•7H2O and 0.002 ZnSO4•H2O. Additional supplementation with inorganic salts and yeast extract was essential to optimize the production, in terms of product concentration and productivity, of neutral lipids by C. laurentii 11. Under this optimized condition, the production of total lipids increased by 133% in relation to control experiment (from 1.27 to 2.96 g L-1). The total lipids indicated a predominant (86%) presence of neutral lipids with high content of 16- and 18- carbon-chain saturated and monosaturated fatty acids. This class of lipids is considered especially suitable for the production of biodiesel.

This work aimed to evaluate nine strains of yeast, previously identified as good producers of lipids in honey medium, for selecting the most suitable strain for the production of lipids in cheese whey medium and compared two well known extraction methods of lipids from the culture medium. The highest yield of total lipids was 1.27 g.L-1 produced by Cryptococcus laurentii 11. A comparison was made between the two culture media: cheese whey and liquid YEPG, and two lipid extraction methods: Bligh and Dyer and Folch et al. for C. laurentii. The experiments were performed with 2² full factorial design using two factors and two levels. Lipid content was higher in cheese whey and there was no difference in the extraction methods statistically. The method of Bligh and Dyer was used in preference to Folch et al. as it resulted in larger mean of total lipids.

Este trabalho objetivou estudar o efeito da vinhaça na biodegradação em solo da borra oleosa proveniente da refinaria de petróleo Replan-Petrobras. Foi utilizado o método respirométrico de Bartha para verificar a eficiência de tratamentos constituídos de solo, borra oleosa nas concentrações 7 e 14 % (m/m) e ajuste da umidade do solo com e sem vinhaça (0,11 mL/g solo seco) durante 121 dias. Embora a adição da vinhaça tenha proporcionado um aumento da população microbiana nos tratamentos, esta não se mostrou adequada para aumentar a eficiência de biodegradação da borra oleosa em solo, uma vez que não houve diferença entre o CO2 produzido nos tratamentos com ou sem vinhaça após o consumo total da vinhaça. Assim, o uso da vinhaça como agente estimulante em processos de biodegradação mostrou-se ineficiente nas condições estudadas.

This work aimed to study the effect of vinasse on the biodegradation in soil of oily sludge from the Replan-Petrobras oil refinery. The Bartha respirometric method was used to verify the efficiency of the treatments with soil, oily sludge (7 and 14% m/m) and soil moisture adjustment with or without vinasse (0.11 mL/g dry soil) for 121 days. Although an increase in the soil microbial population was obtained with the vinasse, it demonstrated not to be adequate to enhance the biodegradation efficiency of the oily sludge in soil, since there was no difference between the CO2 produced from treatments with or without vinasse after the complete consumption of the vinasse. Thus, the use of vinasse as an amendment to biodegradation processes showed not to be efficient under the studied conditions.

Neste trabalho são apresentadas duas técnicas para a avaliação da dispersão, num corpo d'água receptor, do efluente líquido de uma refinaria de petróleo. Assim, a pluma de dispersão foi caracterizada por medidas em campo de condutividade elétrica e por simulação computacional (simulador Cormix). Como caso de estudo, escolheu-se uma refinaria de petróleo cujo efluente é lançado no rio Atibaia (Paulínia/SP). O comportamento do efluente foi avaliado em um trecho de 1000 m após o ponto de lançamento. Os resultados demonstraram que a medição da condutividade elétrica é uma técnica adequada para a avaliação da dispersão de efluentes líquidos de refinaria de petróleo, pois apresentam alta condutividade elétrica e, com isso, há um forte contraste entre os valores do efluente e do rio. Além disso, outros parâmetros de qualidade da água do rio seguiram comportamento de dispersão semelhante ao da condutividade. A pluma de dispersão gerada pelo simulador computacional apresentou uma elevada concordância com os dados obtidos em campo. Nesse sentido, a simulação computacional pode ser uma ferramenta útil para a avaliação da dispersão do efluente considerando-se cenários hipotéticos, e para projetos de emissários.

In this paper, two techniques to evaluate the dispersion of the effluent of an oil refinery are presented. Thus, the dispersion plume was characterized by field measurements of electrical conductivity and by computational simulation (Cormix simulator). An oil refinery whose effluent is discharged on the Atibaia river (Paulínia/SP) is taken as study case. The behavior of the effluent was evaluated until 1000 m after the discharge. The results show that the measurement of electrical conductivity is a suitable technique to evaluate the dispersion of oil refinery wastewaters, since they have high conductivity and for this reason there is a strong contrast between the values of the wastewater and the ambient background. Furthermore, other water quality parameters had a dispersion behavior similar to that shown by the conductivity. The dispersion plume generated by the computational simulator showed high concordance with the field data. In this manner, computational simulation can be a useful tool to evaluate the dispersion of discharges considering hypothetic scenarios, as well as to design the discharge channel.

Este trabalho investigou a eficiência da técnica do bioaumento quando aplicada a solos contaminados com óleo diesel coletados em três postos de combustíveis. Experimentos de biodegradação foram realizados em frascos de Bartha (250 mL), usados para medir a produção microbiana de CO2. A eficiência de biodegradação também foi quantificada pela concentração de hidrocarbonetos. Conjuntamente aos experimentos de biodegradação, a capacidade das culturas estudadas e dos microrganismos nativos em biodegradar óleo diesel comprado de um posto de combustíveis local, foi verificada utilizando-se a técnica baseada no indicador redox 2,6 - diclorofenol indofenol (DCPIP). Resultados obtidos com esse teste mostraram que os inóculos empregados nos experimentos de biodegradação foram capazes de biodegradar óleo diesel e os testes com os microrganismos nativos indicaram que estes solos previamente apresentavam uma microbiota adaptada para degradar hidrocarbonetos. Em suma, nenhum ganho foi obtido com a adição dos microrganismos ou mesmo efeitos negativos foram observados nos experimentos de biodegradação.

This work investigated the efficiency of the bioaugmentation technique when applied to diesel oil contaminated soils collected at three service stations. Batch biodegradation experiments were carried out in Bartha biometer flasks (250 mL) used to measure the microbial CO2 production. Biodegradation efficiency was also measured by quantifying the concentration of hydrocarbons. In addition to the biodegradation experiments, the capability of the studied cultures and the native microorganisms to biodegrade the diesel oil purchased from a local service station, was verified using a technique based on the redox indicator 2,6 -dichlorophenol indophenol (DCPIP). Results obtained with this test showed that the inocula used in the biodegradation experiments were able to degrade the diesel oil and the tests carried out with the native microorganisms indicated that these soils had a microbiota adapted to degrade the hydrocarbons. In general, no gain was obtained with the addition of microorganisms or even negative effects were observed in the biodegradation experiments.

Este trabalho investigou a possibilidade de se usar a vinhaça como um agente estimulador de processos de biorremediação ex-situ. Amostras de água subterrânea e solo foram coletadas em três postos de combustíveis. A biorremediação do solo foi simulada em frascos de Bartha, usados para medir a produção de CO2, durante 48 dias, onde a vinhaça foi adicionada a uma concentração de 33 mL.Kg-1 de solo. A eficiência de biodegradação também foi medida pela quantificação de hidrocarbonetos totais de petróleo (TPH) por cromatografia gasosa. A biorremediação da água subterrânea foi realizada em experimentos laboratoriais simulando condições aeradas (bioreatores) e não aeradas (frascos de DBO). Em ambos os casos, a concentração de vinhaça foi de 5 % (v/v) e diferentes parâmetros físico-químicos foram avaliados durante 20 dias. Embora um aumento da fertilização e da população microbiana do solo foram obtidos com a vinhaça, esta estratégia não se mostrou adequada em aumentar a eficiência da biorremediação dos solos contaminados com óleo diesel. A adição de vinhaça às águas subterrâneas contaminadas teve efeitos negativos na biodegradação dos hidrocarbonetos, uma vez que a vinhaça, como uma fonte de carbono facilmente assimilável, foi preferencialmente consumida.

This work investigated the possibility of using vinasse as an amendment in ex-situ bioremediation processes. Groundwater and soil samples were collected at petrol stations. The soil bioremediation was simulated in Bartha biometer flasks, used to measure the microbial CO2 production, during 48 days, where vinasse was added at a concentration of 33 mL.Kg-1of soil. Biodegradation efficiency was also measured by quantifying the total petroleum hydrocarbons (TPH) by gas chromatography. The groundwater bioremediation was carried out in laboratory experiments simulating aerated (bioreactors) and not aerated (BOD flasks) conditions. In both the cases, the concentration of vinasse was 5 % (v/v) and different physicochemical parameters were evaluated during 20 days. Although an increase in the soil fertility and microbial population were obtained with the vinasse, it demonstrated not to be adequate to enhance the bioremediation efficiency of diesel oil contaminated soils. The addition of the vinasse in the contaminated groundwaters had negative effects on the biodegradation of the hydrocarbons, since vinasse, as a labile carbon source, was preferentially consumed.

Este trabalho objetivou avaliar a capacidade de diferentes microrganismos em degradar óleo diesel comercial em comparação com um óleo diesel intemperizado coletado da água subterrânea em um posto de combustíveis. Dois métodos microbiológicos foram usados para a avaliação da biodegradabilidade: a técnica baseada no indicador redox 2,6-diclorofenol indofenol (DCPIP) e os experimentos respirométricos usando os respirômetros de Bartha. No primeiro, testamos as culturas bacterianas Staphylococcus hominis, Kocuria palustris, Pseudomonas aeruginosa LBI, Ochrobactrum anthropi e Bacillus cereus, um inóculo comercial, consórcios obtidos do solo e da água subterrânea contaminados com hidrocarbonetos e um consórcio de uma área não contaminada. Nos experimentos respirométricos, foi avaliada a capacidade dos microrganismos nativos do solo de um posto de combustíveis em biodegradar os óleos diesel. Os experimentos com o indicador redox mostraram que apenas os consórcios, mesmo aquele de uma área não contaminada, foram capazes de biodegradar o diesel intemperizado. Em 48 dias, a remoção de hidrocarbonetos totais de petróleo (HTP) nos experimentos respirométricos foi aproximadamente 2,5 vezes maior quando o óleo diesel comercial foi usado. Esta diferença foi causada pelo consumo de hidrocarbonetos facilmente biodegradáveis, presentes em maior quantidade no óleo diesel comercial, como demonstrado pelas análises cromatográficas. Assim, resultados indicam que estudos de biodegradabilidade que não consideram o efeito de intemperização dos poluentes pode sobre estimar as taxas de biodegradação e quando o bioaumento é necessário, a melhor estratégia seria aquela baseada na injeção de consórcios, pois mesmo culturas com reconhecida capacidade de biodegradar hidrocarbonetos podem falhar quando aplicadas isoladamente.

This work aimed to evaluate the capability of different microorganisms to degrade commercial diesel oil in comparison to a weathered diesel oil collected from the groundwater at a petrol station. Two microbiological methods were used for the biodegradability assessment: the technique based on the redox indicator 2,6 - dichlorophenol indophenol (DCPIP) and soil respirometric experiments using biometer flasks. In the former we tested the bacterial cultures Staphylococcus hominis, Kocuria palustris, Pseudomonas aeruginosa LBI, Ochrobactrum anthropi and Bacillus cereus, a commercial inoculum, consortia obtained from soil and groundwater contaminated with hydrocarbons and a consortium from an uncontaminated area. In the respirometric experiments it was evaluated the capability of the native microorganisms present in the soil from a petrol station to biodegrade the diesel oils. The redox indicator experiments showed that only the consortia, even that from an uncontaminated area, were able to biodegrade the weathered diesel. In 48 days, the removal of the total petroleum hydrocarbons (TPH) in the respirometric experiments was approximately 2.5 times greater when the commercial diesel oil was used. This difference was caused by the consumption of labile hydrocarbons, present in greater quantities in the commercial diesel oil, as demonstrated by gas chromatographic analyses. Thus, results indicate that biodegradability studies that do not consider the weathering effect of the pollutants may over estimate biodegradation rates and when the bioaugmentation is necessary, the best strategy would be that one based on injection of consortia, because even cultures with recognised capability of biodegrading hydrocarbons may fail when applied isolated.

A biorremediação natural da água subterrânea contaminada com óleo diesel em um posto de combustíveis foi avaliada mediante o monitoramento de indicadores geoquímicos e ensaios laboratoriais de biodegradação. Durante um período de dois anos foram realizadas quatro coletas de água subterrânea para a avaliação de diversos parâmetros físico-químicos e a concentração de hidrocarbonetos. As análises microbiológicas consistiram na contagem de bactérias heterotróficas totais, na verificação da presença de bactérias hidrocarbonoclásticas e no teste de biodegradabilidade utilizando o indicador redox DCPIP. Foi possível identificar fatores que corroboram o emprego da técnica de biorremediação natural para o tratamento da água subterrânea, contudo, a presença de hidrocarbonetos retidos no solo representa uma fonte contínua de contaminação, por isso se faz necessário o monitoramento a longo prazo.

The natural bioremediation of the groundwater contaminated with diesel oil at a petrol station was evaluated by monitoring geochemical indicators and biodegradation tests in laboratory. During two years the groundwater was collected four times. Different physic-chemical parameters and the concentration of hydrocarbons were evaluated. Microbiological analyses consisted of total heterotrophic bacteria counting, verification of the presence of hydrocarbonoclastic bacteria and the biodegradability test using the redox indicator DCPIP. It was possible to identify factors that confirm the use of the natural bioremediation technique as a treatment to the groundwater, however, hydrocarbons retained in the soil represent a continuous source of contamination, and for this reason, a long term monitoring is necessary.

O objetivo do presente estudo foi investigar possíveis métodos para aumentar a taxa de biodegradação aeróbia de hidrocarbonetos (tratamentos ex-situ). Neste trabalho, processos de biorremediação foram aplicados a um solo arenoso com alto nível de contaminação ocasionada por um vazamento de um tanque de armazenamento de óleo diesel subterrâneo em um posto de combustíveis. Experimentos em escala laboratorial (respirômetros de Bartha) foram utilizados para avaliar a biodegradação do óleo diesel. Estímulo da biodegradação foi realizado utilizando-se as técnicas de bioestímulo (adição de soluções de nitrogênio e fósforo ou surfactante Tween 80) e de bioaumento (consórcio bacteriano isolado de um sistema de landfarming). Para investigar as interações entre os fatores otimizadores, e encontrar a melhor combinação entre esses agentes, o estudo foi baseado em um delineamento experimental fatorial completo. A eficiência de biodegradação foi simultaneamente medida com dois métodos: respirométrico (produção de CO2 microbiano) e cromatografia gasosa. Testes de toxicidade aguda com Daphnia similis foram aplicados para examinar a eficiência dos processos em termos de geração de produtos menos tóxicos. Resultados mostraram que todas as estratégias de biorremediação aceleraram a biorremediação natural do solo contaminado e os melhores resultados foram obtidos quando os tratamentos tinham adição de nutrientes. Dados respirométricos indicaram uma máxima mineralização de hidrocarbonetos de 19,8%, obtida com a combinação dos três agentes, com uma remoção de hidrocarbonetos totais de petróleo (TPH) de 45,5% em 55 dias de tratamento. No final dos experimentos, duas espécies predominantes de bactéria foram isoladas e identificadas como Staphylococcus hominis e Kocuria palustris.

The purpose of the present study was to investigate possible methods to enhance the rate of aerobic biodegradation of hydrocarbons (ex-situ treatments). In this work, the bioremediation processes were applied to a sandy soil with a high level of contamination originated from the leakage of a diesel oil underground storage tank at a petrol station. Laboratory scale experiments (Bartha biometer flasks) were used to evaluate the biodegradation of the diesel oil. Enhancement of biodegradation was carried out through biostimulation (addition of nitrogen and phosphorus solutions or Tween 80 surfactant) and bioaugmentation (bacterial consortium isolated from a landfarming system). To investigate interactions between optimizing factors, and to find the right combination of these agents, the study was based on full factorial experimental design. Efficiency of biodegradation was simultaneously measured by two methods: respirometric (microbial CO2 production) and gas chromatography. Acute toxicity tests with Daphnia similis were applied for examination of the efficiency of the processes in terms of the generation of less toxic products. Results showed that all bioremediation strategies enhanced the natural bioremediation of the contaminated soil and the best results were obtained when treatments had nutritional amendment. Respirometric data indicated a maximum hydrocarbon mineralization of 19.8%, obtained through the combination of the three agents, with a total petroleum hydrocarbons (TPH) removal of 45.5% in 55 days of treatment. At the end of the experiments, two predominant bacteria species were isolated and identified (Staphylococcus hominis and Kocuria palustris).
