Abstract This study aimed to assess the cortisol, body and reproductive development of prepubertal Holstein and Holstein-Gir ¾ heifers at 27 months of age maintained in an integrated livestock-forest (ILF) system for 60 summer days compared to the monoculture system in full sun (FS). The ILF system promoted changes (P=0.02) in the cortisol levels of Holstein-Gir ¾ heifers and did not affect weight gain in any of the breed groups studied. Animals in ILF system presented a lower (P=0.006) vulvar development for the rima height parameter and similar for the vulva width parameter. The ovarian follicular population of Holstein-Gir ¾ heifers in the ILF system was lower (P=0.004); however, for the Holstein heifers, no statistical difference was found, and numbers were higher (P=0.08) in the ILF system. None of the other ovarian parameters studied had any changes, and we also found important racial differences. Weight gain (P=0.003), vulvar development (P<0.001), and mean follicular size (P=0.008) were higher in the Holstein-Gir ¾ animals. Based on such results, the effect of the ILF system at 27 months of age on stress and reproductive parameters in the Holstein breed is considered positive, although negative effects have been detected on reproductive parameters in the Holstein-Gir ¾ breed.

Abstract This study evaluated the effect of crude protein (CP) reduction in four diets (156, 139, 132, and 127 g Kg-1 DM) maintaining constant metabolizable protein (188 g/day) on the follicular fluid and cumulus-oocyte complexes of mid-lactating Girolando cows. Twenty-two Girolando cows with average of 21.55 ±3.19 L daily milk yield, 105.30 ±22.62 days in lactation and 3.22 ±0.03 body condition score were selected. To reduce CP in diets and maintain constant metabolizable protein, urea and soybean meal were gradually replaced by lignosulfonate-treated soybean meal (SoyPass®, Cargill), resulting in an increase in rumen-undegradable protein and a reduction in rumen degradable protein. A linear and quadratic reduction was observed in the plasma and follicular fluid urea nitrogen concentration following CP reduction, with the most intense reduction occurring in the 127 g Kg-1 DM group (p<0.001). As CP reduced, there was a tendency for a linear increase in the follicular growth rate (P=0.0696), on the number and proportion of viable oocytes (P<0.09), and also a linear increase for the number (P=0.0397) and proportion (P<0.09) of grade I viable oocytes. Plus, there was a linear effect for the number of cumulus oophorus cells. Cows fed with the lowest amount of CP had cumulus-oocyte complexes with higher numbers of cumulus oophorus cells (P=0.0238). Also, the reduction of diet crude protein was followed by a decrease in the probability of oocytes’ DNA degradation. In conclusion, the reduction of CP in the diet of mid-lactating Girolando cows, reduces urea nitrogen concentration in both blood plasma and follicular fluid, and, as a consequence, increases the viability of oocytes and the number of cumulus oophorus cells while reducing oocytes’ DNA degradation of follicular included cumulus-oocyte complex. The reduction on dietary CP may improve in vivo oocytes’ embryo development impacting fertility of lactating dairy cows.

RESUMO: A eficiência reprodutiva é um dos principais elementos para qualquer sistema de produção de leite ser bem-sucedida. Neste contexto, o manejo reprodutivo deve ser planejado adequadamente e a identificação das principais desordens reprodutivas que afetam o rebanho é essencial para a tomada de decisão. O objetivo do presente estudo foi identificar os principais problemas reprodutivos que acometem rebanhos leiteiros no Rio de Janeiro (RJ), Brasil. Dados relacionados com a raça, idade, ocorrência de problema reprodutivo e o sistema de acasalamento foram individualmente coletados de vacas leiteiras criadas em 25 fazendas localizadas no RJ. A ocorrência de desordens reprodutivas foi relatada em 37% dos animais estudados. A repetição de estro (76,2%) e abortamento (15,6%) foram os problemas reprodutivos mais frequentemente relatados. A repetição de estro foi mais comum em animais mais velhos (>6 anos). Contudo, uma associação entre abortamento e idade do animal não foi observada, nem uma associação entre o sistema de acasalamento e a ocorrência abortamento. Os presentes resultados demonstraram que a repetição de estro é o principal obstáculo para a eficiência reprodutiva nos rebanhos leiteiros do RJ, e que a substituição de animais mais velhos por novilhas é um importante ponto dentro do manejo reprodutivo de um rebanho leiteiro.

ABSTRACT: Reproductive efficiency is one of the key elements for any milk production system to be successful. In this regard, reproductive management must be planned properly and the identification of the main reproductive disorders that affect the herd is primordial for decision-making. The aim of the present study was to identify the main reproductive problems that affect dairy herds on Rio de Janeiro (RJ), Brazil. Data regarding breed, age, reproductive disorder occurrence, and breeding system were individually collected from dairy cows raised at 25 farms located at RJ. The occurrence of reproductive disorders was reported in 37% of the studied animals. Estrous repetition (76.2%) and abortion (15.6%) were the most frequent reproductive problems reported. Estrous repetition was more common in older animals (>6 years old). However, an association between abortion and age of animal was not observed, nor an association between breeding system and occurrence of abortion. The present results demonstrated that estrous repetition is the main obstacle to the reproductive efficiency in dairy herds at RJ, and that the replace of older cows by heifers is an important point within reproductive management of a dairy herd.

The objective of this study was to verify the effects of GnRH on ovulation and pregnancy of ewes subjected to a short-term synchronization of estrus. Santa Inês and crossbred Santa Inês/Dorper ewes received 60 mg MAP sponges during 6 days plus 300 IU eCG and 30 µg d-cloprostenol 24 h prior to sponge withdrawal (SW). Ewes were assigned to receive 0.9% NaCl solution (Tcontrol; n = 32) or 25 µg GnRH (licerelin, T GnRH; n = 34) 24 hours after SW. Each group was assigned to intrauterine insemination by laparoscopy (n = 25) or to natural mating (n = 41). Artificial insemination was performed with a single dose of fresh semen. For controlled mating, females were exposed to males 12, 24, 36 and 48 hours after SW. Ten females per treatment were subjected to transrectal ultrasound examination at 12-hour intervals (SW to 60 hours after). Estrous response (100.0% vs 95.2%), interval from SW to estrus (32.9±7.4 vs 29.8±6.9 hours), estrous length (37.4±9.0 vs 31.5±10.4 hours), pregnancy rates (57.0% vs 41.0%), ovulation rate (100.0% vs 90.0%), number of ovulations/ewe (1.1±0.3 vs 1.2±0.4), maximum follicular diameter (6.4±0.7 vs 6.1±0.6 mm), interval from SW to ovulation (59.1±3.5 vs 58.4±3.5 hours) did not differ between Tcontrol and T GnRH, respectively. Administration of GnRH 24 hours after SW does not improve ovulation or pregnancy rate in estrous synchronization in ewes.

Avaliou-se a viabilidade de embriões bovinos cultivados in vitro, a fresco ou reaquecidos após vitrificação, depois de transportados por 6 ou 12 horas. Oócitos obtidos de folículos de ovários coletados em matadouro foram maturados, fecundados e cultivados in vitro. Após sete dias de cultivo, blastocistos com grau de qualidade I e II (segundo o manual da IETS-1998) foram selecionados, envasados em OPS (open pulled straws) e vitrificados em nitrogênio líquido. O reaquecimento foi realizado a 39ºC pela passagem em soluções de HM com concentrações decrescentes de sacarose (0,25M - 0,15M) por cinco minutos em cada solução. Foram avaliados três tratamentos - V0: embriões vitrificados, reaquecidos e cultivados in vitro (n=25); V6: embriões vitrificados, transportados por 6 horas (simulação em palhetas), reaquecidos e cultivados in vitro (n=29); e V12: embriões vitrificados, transportados por 12 horas, reaquecidos e cultivados in vitro - comparados, cada um, a um tratamento controle, com embriões a fresco-C0: embriões a fresco cultivados in vitro (n=26); C6: embriões a fresco cultivados in vitro após 6 horas de transporte (n=30); e C12: embriões a fresco cultivados in vitro após 12 horas de transporte (n=30). Os embriões foram co-cultivados com células da granulosa em microgotas de TCM 199 acrescido de SFB. Foram avaliadas as taxas de re-expansão e eclosão após 48 horas de cultivo. A análise foi realizada pelo teste do qui-quadrado. As taxas de re-expansão entre os grupos V0, V6 e V12 não diferiram, assim como as taxas de eclosão entre os embriões vitrificados e os controles. As taxas de eclosão, no entanto, diferiram entre os embriões submetidos à vitrificação e os controles. Embriões bovinos produzidos in vitro podem ser transportados a fresco ou vitrificados por períodos de até 12 horas, pois possibilitam taxas de eclosão satisfatórias.

The aim of this study was to evaluate the viability of in vitro produced bovine embryos, fresh or warmed, after submitted to different periods of transportation (6h-12h). Oocytes obtained from ovaries collected from slaughterhouse were matured, fertilized and cultured in vitro. After seven days, grades I and II blastocysts (according to IETS manual) were selected and vitrified after exposition to PBS solution with 5% fetal calf serum (HM), added with 10% ethylene glycol (EG) and 10% of dymetil sulfoxide (DMSO), for one minute, followed by HM solution with 20% EG and 20% DMSO, for 20 seconds. Embryos were loaded into open pulled straws (OPS) and plunged into liquid nitrogen. Warming was performed at 39ºC by embryo exposure to decreasing concentration of sucrose (0.25 and 0.15M), for five minutes in each step. The warmed embryos were distributed in three groups: V0: in vitro cultured after warmed; V6: embryos loaded into straws and kept for 6 hours at 35ºC, before in vitro culture; and V12: embryos loaded into straws and kept for 12 hours at 35ºC, before in vitro culture. Each group was evaluated by control groups of fresh embryos (C0, C6 and C12, respectively). The embryos were co-cultured with cumulus cells in TCM-199 micro droplets added with SFB. Re-expansion and hatching rates after 48 hours in culture were evaluated and results were compared by the Chi-square test. Re-expanded rates among groups V0, V6 and V12 as well as hatching rates among vitrified groups and among control groups did not differ. However, hatching rates were different between vitrified groups and their respective controls. The satisfactory rates of hatching suggest that it is possible to transport warmed and fresh in vitro produced embryos for periods up to 12 hours.
