The antifungal potential of volatile organic compounds (VOCs) produced by actinobacterial strains Streptomyces sp. (ACTB-77) and Amycolatopsis sp. (ACTB-290) from the rhizosphere of Caatinga plants against Colletotrichum musae was investigated. VOCs produced by these microorganisms (axenic and co-culture) were investigated using headspace-solid phase micro-extraction (HS-SPME) and gas chromatography-mass spectrometry (GC-MS). Although no exclusive VOC peaks were observed in the co-culture with ACTB-77, the same experiment involving ACTB-290 yielded five new peaks, including two identified alcohols, suggested as bioreductive products of the corresponding ketones by the fungus. Statistical analysis revealed that co-culture ACTB-77/C. musae has a closer similarity to the fungus than to the actinobacteria, while the co culture ACTB-290/C. musae showed closer similarity to the actinobacteria. These confirmed the more pronounced antifungal activity of the ACTB-290 strain, as observed in the fungus growth inhibition experiments. The antifungal activity of ACTB-290 was associated to its sulfur-containing metabolites, while linalool was suggested as responsible for the ACTB-77 activity.

The fungus Aspergillus sp. was isolated from copper mining rejects and cultured in a liquid culture medium (potato dextrose, PD) enriched with CuSO4.5H2O and Pb(NO3)2 in different concentrations. By flame atomic absorption spectrometry (FAAS) it was observed that fungus absorbed 86% of both copper and lead, with initial concentrations of 100, 500 and 1000 mg L-1. The additional results provided by scanning electron microscopy, energy dispersive spectroscopy, and X-ray diffraction showed ability of the microorganism to act as a biosorbent as well as its capacity to transform the tested salts into other inorganic compounds. Chromatographic profile comparison of extracts obtained from the biotic, abiotic control and the cultivation enriched with the metals showed changes in the profile of the secondary metabolism. When the copper and lead salts were tested separately, there was an increase in the production of the compounds as function of the increase of the concentration and when associated promoted the decrease and disappearance of some substances produced by Aspergillus sp.

RESUMO: O objetivo deste estudo foi investigar os efeitos de folhas da espécie arbórea Poincianella pyramidalis (Tul.) L.P. Queiroz como causa espontânea de abortos, malformações e falhas reprodutivas em rebanhos caprinos no semiárido brasileiro. Foram realizadas investigações epidemiológicas, avaliações clínicas de caprinos acometidos por malformações e necropsias de fetos abortados em 12 propriedades localizadas nos municípios de Gurjão e Soledade, na Paraíba, e no município de Pau dos Ferros, Rio Grande do Norte. O estudo considerou apenas as propriedades que tinham basicamente a presença de P. pyramidalis entre as plantas teratogênicas. Na propriedade localizada no município de Pau dos Ferros foi verificada a ocorrência de grande número de abortos e o nascimento de cabritos malformados, acometendo 90% das cabras gestantes. Nas 12 propriedades pertencentes aos municípios de Gurjão e Soledade na Paraíba os casos espontâneos de mortalidade embrionária, abortos e malformações acometeram 257 cabras (41,1% das cabras dos rebanhos). A artrogripose foi a principal malformação congênita observada. Exames de necropsia e histopatológico de quatro fetos acompanhados das placentas não constataram a presença de agentes infecciosos. Os casos de perda embrionária, abortos e malformações foram concentrados predominantemente nas épocas do ano em que a pastagem estava escassa, mas havia disponibilidade de P. pyramidalis nas propriedades, devido à ocorrência das chuvas da pré-estação, que não permitiu a germinação de pasto, mas a rebrotação de P. pyramidalis. Os achados desse estudo comprovam que esta planta interfere consideravelmente na eficiência reprodutiva dos rebanhos caprinos do Semiárido.
ABSTRACT: The aim of this study was to investigate the effects of the leaves of the tree Poincianella pyramidalis (Tul.) L.P. Queiroz as a spontaneous cause of abortion, malformation and reproductive failure in goat herds in the Brazilian semiarid region. Epidemiological investigations, clinical evaluation of goats affected by malformation and necropsy of aborted fetuses was carried out on 12 farms located in the municipalities of Gurjão and Soledade, state of Paraíba, and on a farm located at the municipality of Pau dos Ferros, state of Rio Grande do Norte. The study considered only the farms where occurred predominantly P. pyramidalis among teratogenic plants. On the farm located in the municipality of Pau dos Ferros the occurrence of frequent abortion and birth of malformed goats, affecting 90% of pregnant goats, was observed. On the 12 farms belonging to the municipalities of Gurjão and Soledade, spontaneous cases of embryonic mortality, abortion and malformation occurred in 257 goats (41.1% of goats). Arthrogryposis was the main congenital malformation observed. Necropsy and histopathological exams of four fetuses and their placentas did not detect the presence of infectious agents. The cases of embryonic mortality, abortion and malformation were predominantly concentrated during the seasons of the year when pasture was scarce, but P. pyramidalis was available, due to the occurrence of pre-seasonal rains, which did not allow the pasture to germinate but P. pyramidalis to regrowth. The findings of this study confirm that this plant significantly interferes in the reproductive efficiency of semi-arid goat herds.

Diagnosis and underdiagnosis of comorbidities in psoriasis patients - need for a multidisciplinary approach

Abstract BACKGROUND: Psoriasis is an immune-mediated disease that manifests predominantly in the skin, although systemic involvement may also occur. Although associated comorbidities have long been recognized and despite several studies indicating psoriasis as an independent risk factor for cardiovascular events, little has been done in general medical practice regardind screening. In the United States, less than 50% of clinicians are aware of these recommendations. OBJECTIVE: To identify the prevalence of these comorbidities in 296 patients followed up at a university dermatology clinic. METHODS: Systematically investigated comorbidity frequencies were compared with general practitioners' registry frequencies. Clinical features correlated with comorbidities were also investigated. RESULTS: High prevalences of systematically investigated comorbidities such as hypertension (30%) and dyslipidemia (26.5%) were documented. Conversely, data from general practitioners' records showed that 33% of dyslipidemia cases were undiagnosed and indicated possible underdiagnosis of some comorbidities. Furthermore, an association was found between: the number of comorbidities and psoriasis duration, age and high body mass index an association was found between the number of comorbidities and psoriasis duration, age, high body mass index, waist circumference or waist-to-hip ratio. (p<0.05). CONCLUSION: Disease duration, age and high body mass index, waist circumference or waist-to-hip ratio are possible criteria for choosing which patients should be screened for comorbidities. Underdiagnosis of comorbidities by general practitioners highlights the need for a multidisciplinary approach in psoriasis management.

ABSTRACTThe antimicrobial activity of the myrsinoic acid A isolated from Myrsine coriacea (Sw.) R.Br. ex Roem. & Schult., Primulaceae, and a two semi-synthetics derivatives was tested against Bacillus subtilis, Escherichia coli, Salmonella enterica subsp. enterica serovar typhi, Staphylococcus aureus, Streptococcus pyogenes, Pseudomonas aeruginosa, Micrococcus luteus, Candida albicans, Candida krusei and Candida tropicalis. The microdilution method was used for the determination of the minimum inhibitory concentration during evaluation of the antimicrobial activity. The myrsinoic acid A showed no activity against the selected microorganisms but the hydrogenated and acetylated derivatives were active against B. subtilis, E. coli, S. aureus and P. aeruginosa.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

AIM: To evaluate the in vitro antimicrobial effects of photodynamic therapy (PDT). METHODS: The microorganism indicators were: Candida albicans, Pseudomonas aeruginosa,Enterococcus faecalis and Staphylococcus aureus. A microbial pool was prepared (10 8 cells/mL), from which aliquots were transferred to culture plates for carrying out the PDT using methylene blue (50 µM) and low-power laser (660 nm, 100 mW and 9 J).The effect of methylene blue alone, low power laser and the absence of treatments were evaluated. Then, aliquots of 1 µL were plated in a media culture, the number of colony forming units (CFU/mL) was obtained and the data submitted to the F test (ANOVA) with Tamhane's comparisons. RESULTS: The laser radiation in the presence of methylene blue was able to eliminate 74.90% of C. albicans, 72.41% of P. aeruginosa, 96.44% of E. faecalis and 95.42% of S. aureus, thus statistically significant differences were found among the groups (p<0.001). CONCLUSIONS: PDT was effective in reducing the number of viable cells in the studiedmicroorganisms, especially E. faecalis and S. aureus.

The phytochemical investigation of Erythroxylum pulchrum St. Hil. (Erythroxylaceae) led to the isolation of three known flavonoid glycosides quercetin-3-O-α-L-rhaminoside, ombuin-3-ruthinoside and ombuin-3-ruthinoside-5-glucoside. These flavonoids are being described for the first time in this E. pulchrum. The structures of the compounds were determined by analysis of IR, MS and NMR data, as well as by comparison with literature data. The methanolic extract of leaves from E. pulchrum inhibited the growth of the Bacillus subtilis CCT 0516, Escherichia coli ATCC 2536, Pseudomonas aeruginosa ATCC 8027, P. aeruginosa ATCC 25619, Staphylococcus aureus ATCC 6538, S. aureus ATCC 25925, Streptococcus sanguinis ATCC 15300, S. salivarius ATCC 7073, S. mutans ATCC 25175 and Streptococcus ATCC. S. aureus ATCC 25925 was the most sensitive among the other S. sanguinis while S. salivarius proved the most resistant.

In an ongoing project to evaluate natural compounds isolated from by-products or wastes from vegetables and fruits (edible plants) as modulators of antibiotic resistance, ethanol extract from mango peel was investigated using Staphylococcus aureus strains possessing efflux mechanisms of resistance to norfloxacin, erythromycin and tetracycline. The minimum inhibitory concentrations (MIC) of the antibiotics were determined by the micro dilution assay in the absence and in the presence of sub-inhibitory mango peel extract concentration. Although the extract did not display relevant antibacterial activity (MIC>2048 µg/mL), it modulated the activity of antibiotics, i.e. in combination with antibiotics (at 512 µg/mL), a four-fold reduction in the MIC values for tetracycline and erythromycin was observed. The results presented here indicates that mango peel could serve as a source of potential adjuvant of antibiotics which add value to this mango by-product.

Estudos recentes demonstram uma relação entre obesidade e inflamação crônica, confirmada através da associação de níveis elevados de fator de necrose tumoral alfa (TNF-±), interleucina seis (IL-6) e proteína C reativa, com aumento do índice de massa corporal (IMC). O estado inflamatório, nos indivíduos obesos, poderia contribuir para o desenvolvimento ou agravamento da psoríase. Fenômenos análogos já foram descritos, em outras doenças inflamatórias crônicas, como a artrite reumatóide e doença de Chrön. Estudos epidemiológicos mostram uma prevalência elevada de comorbidades cardiovasculares, secundárias às alterações metabólicas, associadas à psoríase e obesidade. Permanecem ainda não elucidados alguns aspectos desta associação, como: o impacto da obesidade (nas formas clínicas da dermatose, na associação com comorbidades e na resposta ao tratamento).
Recent studies have found a relationship between obesity and chronic inflammation, confirmed by the association of high levels of tumor necrosis factor (TNF-_), interleukin six (IL-6,) and reactive C-protein with an increase in body mass index (BMI). In obese individuals, this inflammatory condition could contribute to the development or aggravation of psoriasis. Analogous phenomena have already been described in other inflammatory chronic diseases, such as rheumatoid arthritis and Crohn's disease. Epidemiological studies have identified a high prevalence of cardiovascular comorbidities, secondary to the metabolic alterations associated with psoriasis and obesity. A few aspects of this association remain unclear, such as the impact of obesity in the clinical forms of dermatoses, in the response to treatment, and its relationship with comorbidities.

Este trabalho visou investigar a associação da coexistência da presença de granulações tóxicas com resultados de hemocultura positivas, idade dos pacientes, condições de internamento e tipos de agentes bacterianos. Foi realizada análise retrospectiva e prospectiva, cega, para a presença de granulações tóxicas em amostras sangüíneas de trezentos pacientes, de ambos os sexos, internados em hospitais da Cidade de Belém - Pará, com solicitação de hemocultura, num período de dois anos. Com os hemogramas e as hemoculturas realizadas por métodos de automação, e todos os dados submetidos à metodologia de comparação estatística pelo Qui-quadrado (método de clump). Nossos resultados mostraram a existência de associação estatística entre: (1) a presença de granulações tóxicas e os resultados de hemoculturas positivas; (2) a menor idade dos pacientes (neonatos) associadas a hemocultura positiva; (3) a condição de internamento em UTI com hemocultura positiva; e (4) a presença de granulações tóxicas e a observação de leucocitose e desvio à esquerda, em pacientes internados em UTI, com hemoculturas positivas. E que os cinco principais agentes bacterianos identificados nas hemoculturas deste estudo foram Klebsiella oxytoca (22%), Acinetobacter calcoaceticus (20%), Escherichia coli (18%), Enterobacter cloacae (14%), e Pseudomonas aeruginosa (8%).
This work aims at investigating the association of the presence of toxic granulation with positive blood cultures, age of patients, conditions of hospitalization and types of bacterial agents. Blind prospective and retrospective, analyses were carried out for the presence of toxic granulations-in blood samples of 300 patients of the both genders hospitalized in the City of Belém, Pará, Brazil. Request blood tests over a two year period were evaluated. The blood tests and cultures were performes using automated methods. All the data were statistically compared using the Qui-square test (clump method). The results show statistical associations between: (1) the presence of toxic granulations and positive blood cultures; (2) lower ages of patients (the newborn) and positive blood cultures; (3) hospitalization in the ICU and positive blood cultures and (4) toxic granulations-and the observation of leucocytosis and right-left shunts in patients hospitalized in the ICU with positive blood cultures. The commonest bacterial agents identified were klebsiella oxytoca (22%), Acinetobacter calcoaceticus (20%), Escherichia coli (18%), Enterobacter cloacae (14%), and Pseudomonas aeruginosa (8%).

A participação do sistema de histocompatibilidade humano (HLA: human leukocyte antigens) na patogênese das doenças auto-imunes é bem conhecida. Situado no braço curto do cromossomo 6, o sistema HLA se destaca por seu polimorfismo e por sua capacidade de conferir susceptibilidade ou proteção a diferentes enfermidades. Em Dermatologia, esse sistema desempenha papel importante na patogenia e história natural de várias doenças. A força e o tipo de associação variam com a dermatose e, algumas vezes, com o grupo étnico-racial estudado. O surgimento de métodos moleculares para tipificação dos alelos HLA e as recentes atualizações de sua nomenclatura têm contribuído para o melhor entendimento desse sistema. Infelizmente, essas informações não têm sido veiculadas de maneira adequada na literatura clínica, o que dificulta o entendimento da associação do HLA com as doenças cutâneas. Nesta revisão, são discutidos alguns aspectos do sistema HLA, métodos de detecção, nomenclatura e sua associação com vitiligo, pênfigo, psoríase, lúpus eritematoso, escabiose, leishmaniose cutânea, hanseníase, paracoccidioidomicose e dermatite atópica.
The participation of the human histocompatibility system (HLA: human leukocyte antigens) in the pathogenesis of autoimmune diseases is well known. Situated on the short arm of chromosome 6, the HLA system is very polymorphic and has the capacity to confer susceptibility or resistance to different diseases. In Dermatology, this system has an important participation in the pathogenesis and natural course of various diseases. The strength and type of association differ with conditions and sometimes with the ethnic-racial group studied. The discovery of molecular methods to typify HLA alleles and recent updates in its nomenclature has contributed to a better understanding of this system. Unfortunately, this information has not been adequately transmitted in the literature, hindering identification of the association of the HLA with skin diseases. In this review, some aspects of the HLA system are discussed, such as methods of detection, nomenclature and association with vitiligo, pemphigus, psoriasis, lupus erythematosus, scabies, cutaneous Leishmaniasis, leprosy, paracoccidiodomycosis and atopic dermatitis.
