Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

RESUMO Objetivo: Validar a quantificação de T-cell receptor excision circles (TRECs) e kappa-deleting recombination circles (KRECs) por reação em cadeia de polimerase (polymerase chain reaction, PCR) em tempo real (qRT-PCR), para triagem neonatal de imunodeficiências primárias que cursam com defeitos nas células T e/ou B no Brasil. Métodos: Amostras de sangue de recém-nascidos (RN) e controles foram coletadas em papel-filtro. O DNA foi extraído e os TRECs e KRECs foram quantificados por reação duplex de qRT-PCR. O valor de corte foi determinado pela análise de Receiver Operating Characteristics Curve, utilizando-se o programa Statistical Package for the Social Sciences (SSPS) (IBM®, Armonk, NY, EUA). Resultados: 6.881 amostras de RN foram analisadas quanto à concentração de TRECs e KRECs. Os valores de TRECs variaram entre 1 e 1.006 TRECs/µL, com média e mediana de 160 e 139 TRECs/µL, respectivamente. Três amostras de pacientes diagnosticados com imunodeficiência grave combinada (severe combined immunodeficiency, SCID) apresentaram valores de TRECs abaixo de 4/µL e um paciente com Síndrome de DiGeorge apresentou TRECs indetectáveis. Os valores de KRECs encontraram-se entre 10 e 1.097 KRECs/µL, com média e mediana de 130 e 108 KRECs/µL, e quatro pacientes com diagnóstico de agamaglobulinemia tiveram resultados abaixo de 4 KRECs/µL. Os valores de corte encontrados foram 15 TRECs/µL e 14 KRECs/µL, e foram estabelecidos de acordo com a análise da Receiver Operating Characteristics Curve, com sensibilidade de 100% para detecção de SCID e agamaglobulinemia, respectivamente. Conclusões: A quantificação de TRECs e KRECs foi capaz de diagnosticar crianças com linfopenias T e/ou B em nosso estudo, validando a técnica e dando o primeiro passo para a implementação da triagem neonatal em grande escala no Brasil.

KRECs values ranged from 10 to 1,097 KRECs/µL, with mean and median of 130 and 108 KRECs/µL. Four patients with agammaglobulinemia had results below 4 KRECs/µL. The cutoff values were 15 TRECs/µL and 14 KRECs/µL and were established according to the receiver operating characteristic curve analysis, with 100% sensitivity for SCID and agammaglobulinemia detection, respectively. Conclusions: Quantification of TRECs and KRECs was able to diagnose children with T- and/or B-cell lymphopenia in our study, which validated the technique in Brazil and enabled us to implement the newborn screening program for SCID and agammaglobulinemia.

Resumo Apresenta-se uma síntese atualizada de informações sobre algas no Brasil objetivando refinar os dados reunidos até o presente, bem como avaliar os avanços sobre o conhecimento da diversidade de algas no país desde a publicação do Catálogo de Plantas e Fungos do Brasil. Os resultados de 2015 mostraram um acréscimo de 1.250 espécies (35.7%) a um total de 4.747 em relação a 2010. As classes mais diversas em número de espécies foram Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae e Euglenophyceae. Bacillariophyceae e Cyanophyceae tiveram o maior acréscimo de espécies no intervalo de cinco anos. A região Sudeste e Sul foram as mais diversas, porém, as regiões Nordeste com os estados do Piauí e Sergipe e Centro-Oeste com os estados de Mato Grosso, Goiás e Distrito Federal destacaram-se no cenário da biodiversidade nacional. Apesar da escassez de taxonomistas e da infraestrutura limitada, os resultados obtidos evidenciaram um avanço significativo no conhecimento da diversidade de algas no país nesse período de cinco anos, iniciando uma mudança quanto as diferenças geográficas regionais.
Abstract An updated synthesis of cyanobacteria and algae information is presented for Brazil aiming to refine the data gathered to date and evaluate the progress of the biodiversity knowledge about these organisms in the country since the publication of the Catálogo de Plantas e Fungos do Brasil. The results of 2015 showed an increase of 1,250 species (35.7%) when compared to 2010, reaching a total of 4,747 species. The most diverse classes in species number were the Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae and Euglenophyceae. Bacillariophyceae and Cyanophyceae had the highest increase in species number in the five-year interval. The Southeast and South regions were the most diverse, however, the Northeast, with the states of Piauí and Sergipe, and the Central-west region, with Mato Grosso, Goiás and Distrito Federal, also stood out in the national algal biodiversity scenario. Despite the shortage of taxonomists and limited infrastructure, the results showed a significant improvement in the knowledge regarding the diversity of cyanobacteria and algae in the country during the study period, starting to even out regional geographical differences caused by subsampling.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

This study aimed to evaluate the safety of the hydroalcoholic extract (HE) of Syzygium cumini (L.) Skeels, Myrtaceae, leaves in rodents. Acute toxicity was evaluated through the determination of a LD50 in mice and rats (up to 14 days). In mice, the oral administration (p.o.) of the HE (0.1 at 6 g/kg) did not cause any death. When administered by intraperitoneal route (i.p.) the HE (0.1 at 1 g/kg) caused death of the animals (LD50 of 0.489 g/kg). In rats, the HE (0.5, 1 and 2 g/kg, p.o.) did not cause any death, while by i.p., only the 2 g/kg dose was lethal to 67% of the animals. To evaluate chronic toxicity, groups of rats daily received the HE (0.05, 0.1 and 0.25 g/kg) through p.o., during 30, 90 or 180 days and the effects on behavior, body weight, feed consumed were measured. Histology, hematology and biochemical parameters were measured at the end of the treatment. After a 30-day treatment, the HE caused changes in some biochemical parameters. Histological examination of the liver, kidneys, lungs, heart, stomach, intestine and pancreas showed normal architecture suggesting no morphological disturbances. These data may mean that the HE of S. cumini does not exert acute or chronic toxic effects by oral administration.

Crianças e adolescentes com doenças reumatológicas apresentam maior prevalência de doenças infecciosas quando comparados com a população em geral, em decorrência de atividade da doença, possível deficiência imunológica secundária à própria doença, ou uso de terapia imunossupressora. A vacinação é uma medida eficaz para a redução da morbidade e mortalidade nesses pacientes. O objetivo deste artigo foi realizar um consenso de eficácia e segurança das vacinas em crianças e adolescentes com doenças reumatológicas infantis baseadas em níveis de evidência científica. Imunização passiva para os pacientes e orientações para as pessoas que convivem com doentes imunodeprimidos também foram incluídas. Os 32 pediatras reumatologistas membros do Departamento de Reumatologia da Sociedade de Pediatria de São Paulo (SPSP) e/ou da Comissão de Reumatologia Pediátrica da Sociedade Brasileira de Reumatologia elaboraram o consenso, sendo que alguns desses profissionais estão envolvidos em pesquisas e publicações científicas nesta área. A pesquisa dos termos eficácia e/ou segurança das diferentes vacinas em crianças e adolescentes com doenças reumatológicas foi realizada nas bases de Medline e Scielo, de 1966 até março de 2009, incluindo revisões, estudos controlados e relatos de casos. O grau de recomendação e o nível científico de evidências dos estudos foram classificados em quatro níveis para cada vacina. De um modo geral, as vacinas inativadas e de componentes são seguras nos pacientes com doenças reumatológicas, mesmo em uso de terapias imunossupressoras. Entretanto, vacinas com agentes vivos atenuados são, em geral, contraindicadas para os pacientes imunossuprimidos.
Incidence of infectious diseases is higher in children and adolescents with rheumatic diseases than in the general population due to disease activity, possible immune deficiency secondary to the disease itself, or the use of immunosuppressive drugs. Vaccination is effective in reducing morbidity and mortality in those patients. The objective of this study was to establish an evidence-based consensus on the efficacy and safety of vaccination in children and adolescents with rheumatic diseases. Passive immunization of patients and guidelines for people who live with immunosuppressed patients were also included. The 32 pediatric rheumatologists of the Rheumatology Department of the Pediatrics Society of São Paulo, (SPSP, from the Portuguese), São Paulo, SP, Brazil, and/or the Commission on Pediatrics Rheumatology of the Brazilian Society of Rheumatology are responsible for this consensus; some of those professionals are involved on research and scientific publications in this field. The words efficacy and/or safety of different vaccines in children and adolescents with rheumatologic diseases were searched in Medline and Scielo data bases from 1966 to March 2009, including reviews, controlled studies, and case reports. The degree of recommendation and the scientific evidence of the studies were classified in four levels for each vaccine. As a rule, inactive and protein components vaccines are safe for patients with rheumatologic diseases, even in the presence of immunosuppressive therapy. However, live attenuated vaccines are, in general, contraindicated for immunosuppressed patients.

Foi estudada uma amostra de 3.746 pré-escolares de 15 municípios brasileiros (dez do Estado da Bahia e cinco do Estado de São Paulo), construída em três estágios (municípios, domicílios e crianças). Os dados municipais foram obtidos do censo demográfico de 1991. Os dados referentes aos indivíduos e domicílios foram coletados entre 1999 e 2001, constando de questionários estruturados e medições de peso e altura das crianças. Para análise utilizou-se o programa MLwiN. No modelo multinível final observou-se que baixo IDH e inadequada assistência pré-natal, no nível dos municípios; baixas condições ambientais, econômicas e da escolaridade materna, no nível dos domicílios; o baixo peso ao nascer, esquema vacinal incompleto, história prévia de desnutrição e ausência do aleitamento ao peito, no nível das crianças, mostraram-se fortemente associados ao déficit do crescimento linear das crianças estudadas. A variabilidade total da desnutrição explicada pelo modelo foi de 27,4%, desta, 51,3%, 33% e 15,7% foram explicadas, respectivamente, pelas variáveis dos níveis da criança, do domicílio e do município.

The sample included 3,746 preschoolers from 15 Brazilian municipalities (ten from the State of Bahia and five from São Paulo), with a three-tier analysis (municipalities, households, and children). Municipal data were obtained from the 1991 National Census. Individual and household data were collected from 1999 to 2001 using structured questionnaires and measuring children's weight and height. In the final multilevel model, low HDI and prenatal care at the municipal level, poor environmental and economic conditions and maternal schooling at the household level, and low birth weight, incomplete immunization, prior history of malnutrition, and no report of breastfeeding at the individual level were strongly associated with linear growth deficit. OBJETIVOS: descrever e analisar dados de um inquérito realizado em Londrina, Paraná, sobre práticas alimentares no primeiro ano de vida. MÉTODOS: os dados foram obtidos durante a "Campanha Nacional de Vacinação, realizada em agosto de 2002. As crianças foram selecionadas por plano de amostragem por conglomerado. Aplicou-se um questionário com questões sobre alimentação da criança nas 24 horas precedentes. Para obtenção das freqüências empregou-se análise de probitos. Pela análise de regressão logística, foram analisados os fatores de risco para interrupção da amamentação exclusiva em menores de quatro meses e desmame em menores de um ano. RESULTADOS: em menores de quatro meses, a prevalência do aleitamento materno exclusivo (AME) foi 29,3% e, para aleitamento materno predominante (AMP) 20,7%. Nos menores de seis meses, a prevalência do AME foi 21,0% e do AMP 16,5%. Para o aleitamento materno (AM) em menores de um ano, a prevalência foi 62,4%. As medianas em dias foram 11,9 para AME e 257,3 para AM. Entre os fatores de risco para interrupção do AME e do AM destacam-se: uso de chupeta, primiparidade, acompanhamento ambulatorial em serviço público, idade e trabalho materno. CONCLUSÕES: as taxas de aleitamento foram consideradas baixas, indicando a necessidade de se estabelecerem políticas públicas adequadas para alimentação da criança.
OBJECTIVES: to describe and analyze data of a survey on nutritional practices in children to one year of age carried out in Londrina, Paraná. METHOD: data were obtained during the "National Immunisition Canpaign", carried out in August 2002. Mothers were asked to answer a questionnaire on how they had fed their child in the preceding 24 hours. Children were selected following a sample planning by cluster. Probits analysis was used to know the breastfeeding frequency. Through logistic regression analysis risk factors were analyzed concerning the interruption of exclusive breastfeeding of babies younger than four months and the weaning of babies below the age of one. RESULTS: among babies younger than four months exclusive breastfeeding (EB) prevalence was 29.3% and predominant breastfeeding (PB) was 20.7%. For those younger than six months, EB prevalence was 21% and PB was 16.5%. Concerning children below the age of one, the prevalence was 62.4%. The median time was 11.9 days for EB and 257.3 for PB. Risk factors were: use of pacifiers, first child, mother's age and job. CONCLUSIONS: the breast-feeding rates found were considered low, showing the necessity of making adequate public policies.

O objetivo deste artigo é, a partir das informações disponibilizadas pela Pesquisa de Informações Básicas Municipais do IBGE, analisar três variáveis: a existência de Conselhos Municipais de Meio Ambiente, de Fundos Especiais de Meio Ambiente e de legislação sobre Áreas de Interesse Especial. Dentre outros aspectos, examina-se a incidência dos Conselhos Municipais de Meio Ambiente tendo em vista a bacia hidrográfica e o partido do prefeito.
Based on the information available on the Municipal Basic Information Research of IBGE, this article aims to analyze three variables: the existence of Municipal Councils for the Environment, Special Funds for the Environment and Legislation on Areas of Special Interest. Among other aspects, it examines the incidence of Municipal Councils for the Environment having in mind the hydrographic basin and the Mayor Political Party.

Caracterização enzimática e patogenicidade cruzada de Colletotrichum spp. associados a doenças de pós-colheita

A antracnose, causada por Colletotrichum spp., pode ocasionar grandes perdas a nível de campo e em pós-colheita sobre diversas culturas e seus produtos. O presente trabalho teve por objetivos testar a patogenicidade cruzada de isolados de C. gloeosporioides do caju (Anacardium occidentale) (CAJ), manga (Mangifera indica) (MG), mamão (Carica papaya) (MM), maracujá (Passiflora edulis) (MR) e C. musae da banana (Musa spp.) (BAJ); avaliar a produção de enzimas extracelulares (amilolítica, celulolítica, lipolítica e proteolítica) produzidas pelos isolados em substratos sólidos específicos; e detectar padrões eletroforéticos de proteínas totais e isoenzimas (alfa-esterase, beta-esterase, fosfatase ácida e leucina aminopeptidase). Na análise da patogenicidade cruzada, todos os isolados de Colletotrichum spp. induziram lesões necróticas, deprimidas sobre os frutos, exceto em maracujá que foi suscetível tão somente ao isolado MR. Quanto à produção de enzimas extracelulares hidrolíticas, os isolados de C. gloeosporioides produziram amilase, lipase, protease e celulase, sendo que esta última enzima não foi detectada em C. musae. Com relação à análise eletroforética de proteínas totais e isoenzimas, os isolados apresentaram variações no número e posição das bandas no gel de poliacrilamida em todos os sistemas, com exceção de leucina aminopeptidase, onde bandas monomórficas foram formadas, sem variação na intensidade e pouca variação na mobilidade relativa.
Anthracnose caused by Colletotrichum spp. may result in great losses in field productions and post-harvest conditions for several cultures and their products. The objective of this work was to test the pathogenicity of C. gloeosporioides isolates from cashew (Anacardium occidentale) (CAJ), mango (Mangifera indica) (MG), papaya (Carica papaya) (MM) and passion fruit (Passiflora edulis) (MR) and C. musae from banana (Musa spp.) (BAJ); to evaluate the production of extra cellular hydrolytic enzymes (amylolytic, celullolytic, lypolytic, proteolytic) by the isolates on specific solid culture media; and to detect electrophoretic patterns of total proteins and isoenzymes (alpha-esterase, beta-esterase, acidic phosphatase and leucine aminopeptidase). In crossed pathogenicity tests, all Colletotrichum spp. isolates produced necrotic and depressed lesions on fruits, except on passion fruit, which was susceptible only to the MR isolate. The isolates of C. gloeosporioides produced amylolytic, lypolytic, proteolytic, and celullolytic enzymes, and C. musae did not produce any detectable celulase. Electrophoretic analysis of total proteins and isoenzymes showed variations in the number and position of the bands among all isolates in the systems used, except for leucine aminopeptidase, which was monomorphic.
