BACKGROUND The high demand for adequate material for the gold standard reverse transcription real-time polymerase chain reaction (RT-qPCR)-based diagnosis imposed by the Coronavirus disease 2019 (COVID-19) pandemic, combined with the inherent contamination risks for healthcare workers during nasopharyngeal swab (NP) sample collection and the discomfort it causes patients, brought the need to identify alternative specimens suitable for the diagnosis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). OBJECTIVES The aim of this work was to compare saliva and gingival fluid swabs to NP swabs as specimens for RT-qPCR-based SARS-CoV-2 diagnosis. METHODS We compared gingival fluid swabs (n = 158) and saliva (n = 207) to the rayon-tipped NP swabs obtained from mild-symptomatic and asymptomatic subjects as specimens for RT-qPCR for SARS-CoV-2 detection. FINDINGS When compared to NP swabs, gingival fluid swabs had a concordance rate of 15.4% among positive samples, zero among inconclusive, and 100% among negative ones. For saliva samples, the concordance rate was 67.6% among positive samples, 42.9% among inconclusive, and 96.8% among negative ones. However, the concordance rate between saliva and NP swabs was higher (96.9%) within samples with lower cycle threshold (Ct) values (Ct > 10 ≤ 25). MAIN CONCLUSIONS Our data suggests that whereas gingival fluid swabs are not substitutes for NP swabs, saliva might be considered whenever NP swabs are not available or recommended.

BACKGROUND During routine Coronavirus disease 2019 (COVID-19) diagnosis, an unusually high viral load was detected by reverse transcription real-time polymerase chain reaction (RT-qPCR) in a nasopharyngeal swab sample collected from a patient with respiratory and neurological symptoms who rapidly succumbed to the disease. Therefore we sought to characterise the infection. OBJECTIVES We aimed to determine and characterise the etiological agent responsible for the poor outcome. METHODS Classical virological methods, such as plaque assay and plaque reduction neutralisation test combined with amplicon-based sequencing, as well as a viral metagenomic approach, were performed to characterise the etiological agents of the infection. FINDINGS Plaque assay revealed two distinct plaque phenotypes, suggesting either the presence of two severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) strains or a productive coinfection of two different species of virus. Amplicon-based sequencing did not support the presence of any SARS-CoV-2 genetic variants that would explain the high viral load and suggested the presence of a single SARS-CoV-2 strain. Nonetheless, the viral metagenomic analysis revealed that Coronaviridae and Herpesviridae were the predominant virus families within the sample. This finding was confirmed by a plaque reduction neutralisation test and PCR. MAIN CONCLUSIONS We characterised a productive coinfection of SARS-CoV-2 and Herpes simplex virus 1 (HSV-1) in a patient with severe symptoms that succumbed to the disease. Although we cannot establish the causal relationship between the coinfection and the severity of the clinical case, this work serves as a warning for future studies focused on the interplay between SARS-CoV-2 and HSV-1 coinfection and COVID-19 severity.

Abstract Next-generation sequencing (NGS) platforms allow the analysis of hundreds of millions of molecules in a single sequencing run, revolutionizing many research areas. NGS-based microRNA studies enable expression quantification in unprecedented scale without the limitations of closed-platforms. Yet, whereas a massive amount of data produced by these platforms is available, comparisons of quantification/discovery capabilities between platforms are still lacking. Here we compare two NGS-platforms: SOLiD and PGM, by evaluating their microRNA identification/quantification capabilities using two breast-derived cell-lines. A high expression correlation (R2 > 0.9) was achieved, encompassing 97% of the miRNAs, and the few discrepancies in miRNA counts were attributable to molecules that have very low expression. Quantification divergences indicative of artefactual representation were seen for 14 miRNAs (higher in SOLiD-reads) and another 10 miRNAs more abundant in PGM-data. An inspection of these revealed an increased and statistically significant count of uracyls and uracyl-stretches for PGM-enriched miRNAs, compared to SOLiD and to the miRBase. In parallel, adenines and adenine-stretches were enriched for SOLiDderived miRNA reads. We conclude that, whereas both platforms are overall consistent and can be used interchangeably for microRNA expression studies, particular sequence features appear to be indicative of specific platform bias, and their presence in microRNAs should be considered for database-analyses.

Este estudo teve como objetivo examinar a perceção de estudantes de Educação Física sobre o valor educativo do Modelo de Educação Esportiva (MEE) pelo confronto entre a vivência do modelo enquanto alunos e como professores estagiários. Participaram no estudo 16 estudantes do Mestrado em Ensino da Educação Física da FADEUP. Aplicou-se uma entrevista semiestruturada e de resposta aberta após a validação de construção e de conteúdo. Os participantes percepcionaram potencial elevado do MEE no desenvolvimento da formação pessoal, social e desportiva de crianças e jovens. Não obstante, as suas elevadas exigências, ao nível da organização e gestão de alunos e atividades, foram reconhecidas pelos participantes na condição de professores estagiários. A investigação futura deve se centrar na análise do modo como os futuros professores aprendem a ensinar o currículo do MEE, fornecendo indicadores para o domínio de prática e contribuindo para a eficácia pedagógica na sua implementação.

This study examined the perceptions of pre-service teachers on the educational value of the Sport Education model (SEM) by contraposition of their experiences as students during the teacher education program and as pre-service teachers. The participants were 16 senior students of the Master in Teaching Physical Educationin FADEUP. After construct and content validation a semi-structured interview containing open-ended questions was applied. The participants grasped the model as an extremely valuable educational experience that holds the potential for the young students' personal, social, and sports development. Notwithstanding, as pre-service teachers, the participants pictured SEM as highly demanding in the managerial organization of class, students and activities. In future, research should examine more thoroughly the manner by which upcoming teachers learn to teach the SEM curriculum in order to provide benchmarks for enhanced practice and pedagogical capability of novice teachers during early implementation of the model.

Objetivo Avaliar se o mês de nascimento em diferentes latitudes da América do Sul pode influenciar a presença ou gravidade da esclerose múltipla (EM) na vida. Método Neurologistas de quatro países da América do Sul trabalhando em unidades de EM coletaram os dados de seus pacientes com referência ao mês de nascimento, gênero, idade e progressão da doença. Resultados A análise dos dados mostrou que, para 1207 pacientes com EM e 1207 controles, não havia diferença significativa no mês de nascimento com relação à prevalência de EM em quatro zonas de latitude (0–10; 11–20; 21–30; e 31–40 graus). Não houve relação entre o mês de nascimento e a gravidade da doença em nenhuma destas zonas. Conclusão Os resultados deste estudo mostram que pacientes com EM nascidos de mães grávidas em diferentes latitudes sul não seguem o padrão dos resultados sazonais encontrados nas latitudes norte.

Objective To assess whether the month of birth in different latitudes of South America might influence the presence or severity of multiple sclerosis (MS) later in life. Methods Neurologists in four South American countries working at MS units collected data on their patients' month of birth, gender, age, and disease progression. Results Analysis of data from 1207 MS patients and 1207 control subjects did not show any significant variation in the month of birth regarding the prevalence of MS in four latitude bands (0–10; 11–20; 21–30; and 31–40 degrees). There was no relationship between the month of birth and the severity of disease in each latitude band. Conclusion The results from this study show that MS patients born to mothers who were pregnant at different Southern latitudes do not follow the seasonal pattern observed at high Northern latitudes.

Neste artigo apresentamos dados de um estudo mais vasto no âmbito de uma dissertação de Mestrado em Ciências da Educação - área de especialização em Avaliação, cujo objetivo consistiu em conhecer as perceções dos estudantes universitários sobre a avaliação, em particular sobre os métodos de avaliação mais utilizados e a relação entre avaliação e aprendizagem. Os dados foram recolhidos através de um inquérito por questionário, aplicado ao 3º ano de diferentes cursos (Educação, Engenharia, Medicina, Educação Básica e Psicologia) da Universidade do Minho. No total, participaram 254 estudantes. Os dados revelam que os aspetos que os estudantes mais associam à avaliação são a aprendizagem, os testes/exames, a verificação de conhecimentos e as notas/classificações, sendo o conflito, o receio/medo e a imposição os menos ligados ao processo avaliativo. Globalmente, e em relação aos métodos de avaliação, as apresentações orais em grupo, os testes e os relatórios de grupo são os mais utilizados na perspetiva dos alunos, sendo menos frequentes o ensaio individual, o ensaio em grupo e os testes orais, destacando-se, portanto, os que apelam mais ao trabalho de grupo em detrimento do individual. Neste artigo, discutem-se ainda os métodos tradicionais versus métodos alternativos de avaliação quanto à sua eficácia na perspetiva dos alunos e respetivas implicações.

This article presents data from a Master's thesis in Education - Specialization in Evaluation. It aimed at investigating students' perceptions on assessment in the context of higher education. The main goal is to get to know higher education students' views of assessment, particularly in regard to methods of assessment most used in higher education and the relationship between assessment and learning taking the perspective of students. Data were collected through a questionnaire to 3rd year undergraduate students in different programmes (Education, Engineering, Medicine, Basic Education and Psychology) at the University of Minho. In total, 254 students participated in the study. Findings suggest that the key aspects that students associate with assessment are learning, exams, verification of knowledge and marks. Data also reveal that conflict, fear and imposition are the aspects that students associate less with assessment. In regard to assessment methods, oral presentations in group, tests and group reports are the most used ones according to students. Faculty used less frequently methods such as individual essay, group essay and oral tests, which points to the use of rather collective methods in detriment to individual ones. This paper also discusses traditional versus alternative methods in terms of efficacy according to students' perspective and their implications.

Uma grande variedade de produtos tóxicos é emitida durante o processo de tratamento do couro nos curtumes que usam o cromo III para obter o couro wet blue. O resíduo sólido (raspas e aparas) contém cerca de 3 % de Cr III, que representa um sério problema ambiental e não tem, atualmente, destino adequado. O resíduo de couro após extração de Cr (colágeno) foi utilizado como fonte nitrogenada para o capim-elefante (Pennisetum purpureum Schum cv. Napier), em um Latossolo Vermelho-Amarelo, aplicando-se quatro doses do colágeno (4, 8, 16 e 32 t ha-1) e três tratamentos adicionais (testemunha; adubação com N mineral e resíduo de couro wet blue- sem a extração do Cr). O colágeno é uma boa fonte nitrogenada alternativa para crescimento do capim-elefante até a dose de 16 t ha-1. O resíduo de couro wet blue não disponibilizou N durante o cultivo de capim-elefante. Isso mostra que a extração do Cr utilizando-se o ácido fosfórico é fundamental para uso do rejeito de couro como fonte nitrogenada na agricultura. A aplicação do colágeno supriu a necessidade de N para plantas de capim-elefante, semelhantemente à adubação com N mineral.

Large amounts of solid wastes are produced in tanneries, which use chromium (III) to obtain wet blue leather hides. The solid residues (leather scraps and strips) contain around 3 % chromium (III), which represent a serious environmental problem and, presently, have no adequate destination. The leather waste after chromium extraction (collagen) was used as a nitrogen source for elephantgrass- Pennisetum purpureum Schum cv. Napier. Four doses of collagen (4, 8, 16, and 32 t ha-1) and three additional treatments (control; mineral fertilizer N; and wet blue leather residue without Cr extraction) were used. Collagen as alternative nitrogen source has a positive effect on elephantgrass growth at rates up to 16 t ha-1. Little N became available to elephantgrass plants after application of wet blue leather residues. These results show that the phosphoric acid extraction of chromium is necessary if leather waste is to be used as a nitrogen source in agriculture. The application of collagen supplied the N need of elephantgrass plants, similarly to mineral N fertilization.

The main objective of this work was to determine the effect of the use of hypophyseal raw extract to induce fish spawning in curimbatá specie. An election of females of frog bull (Rana castesbeiana) was carried through for extraction of hyphophysys, where the female donors had been those that presented an increment of their visceral socket, which was provoked by ovaries in mature phase or advanced maturation. Twenty curimbatá breeders (Prochilodus affinis) had been used for spawning. The males had an initial average weight of 170.0 g and an average length of 21.72 cm and the females with an initial weight average of 181.0 g and an average length of 6.28 cm. The experiment was carried through with 2 treatments: Treatment 1: 0.5 mg/kg and Treatment 2: 0.7mg/kg. The animals were stocked in aquarium with 1000 L capacity with constant aeration. The females received two intramuscular injections of frog bull raw extract, being the first dose of 0.5 mg/kg and the second 0.7 dose of mg/kg, with interval of 12 hours. The females with dosages of 0.5 mg/kg had obtained a superior number of larvae than those with 0,7 mg/kg. The difference was statistical significant (P>0,06). This result was probably due to different stadiums of gonadal maturation. Both dosages (0.5 and 0.7 mg/kg) of frog bull hypophyseal raw extract had allowed to induce reproduction of curimbatá females, showing an alternative in the reproductive handling.

La objetivou-se com este trabalho determinar o efeito do uso do extracto bruto de hipófisis (EBH) na indução da desova em curimbatá. Foi realizado uma seleção de fêmeas de rã touro (Rana castesbeiana) para extração da hipófise, onde as fêmeas doadoras foram aquelas que apresentavam aumento da cavidade visceral, provocado pelo ovários em estádio maduro ou de maturação avançada. Para desova foram utilizados 20 reprodutores de curimbatá (Prochilodus affinis). Os machos com peso médio inicial de 170,0 g e comprimento médio de 21,72 cm e as fêmeas com peso médios iniciais de 181,0 g e comprimento médio de 6,28 cm. O experimento foi realizado com dois tratamentos: Tratamento 1: 0,5 mg/kg e Tratamento 2: 0,7 mg/kg. Os animais foram alojados em aquário com 1000 L de água, aeração constante. Vinte fêmeas receberam duas injeções intramusculares de extrato bruto de rã touro, sendo a primeira dose de 0,5 mg/kg e a segunda dose de 0,7 mg/kg, com intervalo de 12 horas. Os curimbatás submetidos a dosagens de 0,5 mg/kg obtiveram números de larvas superior e estatisticamente significativas (P>0,06) ao tratamento 2 utilizando 0,7 mg /kg, provavelmente esse resultado pode ser em função de umas fêmeas estarem em diferentes estádios de maturação gonadal. Em curimbatá duas de doses de extrato bruto de hipófise de rã touro nas concentrações de 5 e 7 mg/kg, permitiram a indução reprodutiva das fêmeas, mostrando uma alternativa no manejo reprodutivo.

This study was carried out to compare the influence of sex on Astyanax scabripinnis morfometric and reproductive indexes. Three fingerlings (live weight 5.07 ± 0.14) were assigned to three treatment groups: T1 male only; T2 female only, and T3 both sex. Female parameters measured were trunk length, head length, pre-anal distance, and body height. Significant differences for total and body length for cultivated females was observed in presence and absence of the male in relation to the cultivated males in presence and absence of the females. Length of the trunk, length of the head, and pre-anal were not significantly different between the females. Females in the absence of males had significant differences in relation to the males cultivated in absence and presence of the females. For height of the body, significant differences among treatments were not found. The results also revealed significant changes on the gonadossomatic index of T2 females in comparison to males from T1 and T3 groups. In the other hand, the hepatossomatic index did not show any significant change amongst groups.

Objetivou-se comparar a influência do sexo na morfometria e nos índices somáticos de lambari em condições de cultivo. Utilizou-se 270 alevinos de Lambari prata, com peso médio inicial 5,07 ± 0,14 g para machos, e as fêmeas com peso médio inicial de 6,67 ± 0,07 g, com 3 tratamentos: T1-machos na ausência de fêmea e T2-fêmeas na ausência de machos e T3-machos e fêmeas juntos. Para o comprimento total e padrão foi observado diferença significativa para fêmeas cultivadas na presença e ausência do macho em relação as machos cultivados na presença e ausência das fêmeas. Já comprimento do tronco, comprimento da cabeça e distância pré-anal não foi encontrada diferença significativa entre as fêmeas. Já as fêmeas na ausência de machos apresentaram diferenças significativas em relação aos machos cultivados na ausência e na presença das fêmeas. Para altura do corpo não foi encontrada diferença estatisticamente significativa entre os tratamentos. O índice gonadossomático foi estatisticamente diferente para fêmeas cultivadas na ausência do macho em relação aos machos cultivados na ausência e na presença das fêmeas, ao contrário do observado para índice hepatossomático que não houve diferença significativa entre os tratamentos.

Summary The objective of the present study was to evaluate the influence of sex in the lambari performance. Fingerlings (300) of Lambari silver (Astyanax scabripinnis) were used with initial mean weight 5.07 ± 0.14 g and an average length of 5.88 ± 0.033 cm for males and the females had an initial weight of 6.67 ± 0.07g and a mean length of 6.28 ± 0.08 cm. A complete randomized experimental design was used with 3 treatments: T1: male separated from the female; T2: female separated from males presence, and T3: male and female together, with 3 repetition and 30 fingerlings in each aquarium, maintained in a aquarium of 1,000 l. The temperature of the morning had a mean of 24.0 ± 3.46 and 30.0 ± 0.67 ºC in the afternoon. The results demonstrate that the male cultivated alone obtained an inferior performance to the male and female together cultivated. The females cultivated alone showed a better performance in relation to the females in the presence of males, but for the total length there were no significant differences. The total length was significant in the T3 in relation to the T1.The viscerossomatic index obtained a significant result in the animals cultivated separated in relation to the animals cultivated together. For the carcass yield, the animals of the T1 resulted in higher and significant differences than the other treatments. The rate of specific growth of the animals in T2 treatment was the highest and the survival was significant higher for the T1 treatment. For the production of Lambari is recommended to cultivate lambaris males separated from the females.

O objetivo do presente estudo foi avaliar a influência do sexo no desempenho de lambari. Foram utilizados 300 alevinos de Lambari prata (Astyanax scabripinnis), com peso médio inicial 5,07 ± 0,14 g e comprimento médio de 5,88 ± 0,033 cm para machos e as fêmeas com peso inicial 6,67 ± 0,07g e comprimento médio de 6,28 ± 0,08 cm, em um experimento inteiramente casualizado, com 3 tratamentos: T1: macho separado da fêmea; T2: fêmea sem a presença do macho e T3: macho e fêmea junto, com 3 repetições e 30 alevinos que foram alojados por caixa de amianto de 1.000 l. A temperatura da manhã ficou em média em 24,0 ± 3,46 oC e da tarde ficou em média de 30,0 ± 0,67 oC. Os resultados obtidos demostram que os machos cultivados sozinhos obtiveram desempenho inferior aos machos cultivados juntos a fêmeas. As fêmeas cultivadas sozinhas obtiveram melhor desempenho em relação a fêmeas na presença de machos, mas para o comprimento total não houve diferenças estatísticas significativas. O comprimento total foi significativo no T3 em relação aos animais do T1. O índice viscerossomático obteve resultado significativo nos animais cultivados separados em relação aos animais cultivados juntos. Para rendimento de carcaça dos animais do T1 obtiveram resultado superior e significativo aos demais tratamentos. A taxa de crescimento específico dos animais do tratamento 2 foi superior e a sobrevivência foi estaticamente significativa nos animais do tratamento 1. Para a produção de Lambari, é recomendado cultivar machos de lambaris separados das fêmeas.
