Abstract Cancer is the leading cause of death. Melanoma skin cancer originates in melanocytes and represents 80% of the deaths associated with skin cancer. Vinblastine (VIN) is a chemotherapeutic agent used in the treatment of cancer through disrupting mitotic spindle and tumor development. Curcumin (CUR), a compound extracted from the rhizomes of the Curcuma longa plant, has beneficial effects in preventing the development and progression of cancer while modulating the immune response and oxidative stress. The expression of purinergic receptors, ecto-enzymes, and adenosine can modulate the inflammatory responses in cancer. The activity of enzymes, the markers of cell damage in oxidative stress, the generation of reactive oxygen species (ROS), and the activities of ecto-enzymes in the melanoma cell line were investigated. The human melanoma cell line was treated with curcumin (40 µM), vinblastine (VIN) (20 nM), or a combination of both for 24h. Oxidative stress enzymes and byproducts were measured and compared against the activity of ecto-enzymes. There was a marked increase in ROS production in all groups, but an increase in protein carbonylation was only detected in the VIN group. CUR had an inhibitory effect on extracellular ADP hydrolysis, as evidenced by a significant decrease in ADP substrate removal. VIN possibly increased adenosine formation, as demonstrated by an increase in ADP substrate removal. VIN (alone or in combination with CUR) reduced the activity of ADA, thus increasing the concentration of adenosine in the tumor microenvironment. CUR increased ROS generation in melanoma cells and disrupted the purinergic signaling cascade. Therefore, it may be a promising adjuvant therapy for melanoma, a cancer with a high incidence and lethality.

Introduction Candida dubliniensis, a new species of Candida that has been recovered from several sites in healthy people, has been associated with recurrent episodes of oral candidiasis in AIDS and HIV-positive patients. This species is closely related to C. albicans. The enzymatic activity of C. dubliniensis in response to oxidative stress is of interest for the development of drugs to combat C. dubliniensis. Methods Fluconazole- and amphotericin B-resistant strains were generated as described by Fekete-Forgács et al. (2000). Superoxide dismutase (SOD) and catalase assays were performed as described by McCord and Fridovich (1969) and Aebi (1984), respectively. Results We demonstrated that superoxide dismutase (SOD) and catalase activities were significantly higher (p<0.05) in the fluconazole- and amphotericin B-resistant strains of C. dubliniensis and C. albicans than in the sensitive strains. The catalase and SOD activities were also significantly (p<0.01) higher in the sensitive and resistant C. albicans strains than in the respective C. dubliniensis strains. Conclusions These data suggest that C. albicans is better protected from oxidative stress than C. dubliniensis and that fluconazole, like amphotericin B, can induce oxidative stress in Candida; oxidative stress induces an adaptive response that results in a coordinated increase in catalase and SOD activities.

Este trabalho busca, na análise de livros didáticos dos anos iniciais, as possíveis causas das lacunas explicativas que alunos dos anos finais apresentam sobre o sistema respiratório. Para isto, realizou-se a análise relativa a: espaço, conceitos, imagens, atividades práticas e exercícios propostos nas unidades referentes ao sistema respiratório de livros de ciências dos anos iniciais. Os resultados dessa análise apontam a existência de falhas explicativas em textos e imagens e a grande quantidade de imagens decorativas, o que pode comprometer a aprendizagem de conceitos. Constataram-se mudanças positivas nos livros do PNLD 2010, mas que a utilização dos mesmos deve ser feita com senso crítico pelos professores.

This work intends to analyze textbooks from the initial years for possible causes of the explanatory gaps that students have in their final grades of the respiratory system. To achieve this, we analyzed the space, concepts, images, practical activities and exercises in the units proposed for the respiratory system in science books within the initial years. The results of this analysis indicate the existence of failures in explanatory texts and images and a large quantity of decorative images, which can compromise the learning of concepts. Positive changes were noted in the books of PNLD 2010, but their use should be made with critical common sense by the teachers.

A investigação da qualidade de vida dos professores de ciências e a busca por melhorias que promovam um melhor desempenho desses profissionais no cotidiano das escolas podem ser uma forma de reduzir as desigualdades na educação no Brasil e favorecer o crescimento dos professores em nível educacional e social. Este estudo teve o objetivo de analisar indicadores físicos e mentais de qualidade de vida dos professores de ciências de uma escola pública localizada no interior do Rio Grande do Sul. Foram avaliadas informações acerca das características demográficas, econômicas, ocupacionais, atividades domésticas, esforços físicos, saúde física, saúde mental e os diagnósticos médicos mais referidos pelos professores desde o início do trabalho como docente. Como resultados, encontraram-se diversas queixas em relação à saúde e à qualidade de vida dos professores estudados, como nervosismo, rouquidão, dor nas costas, braços e pernas, além de formigamento e inchaço nas pernas. Conclui-se que o conhecimento dessas evidências pode contribuir para a construção de medidas para a reorganização da sistemática de trabalho e influenciar diretamente na qualidade de vida dos professores, gerando melhor desempenho na atividade de educar.

The investigation regarding the quality of life of science teachers and the search for improvements that ameliorate these professionals' performance in primary education schools can be a way to reduce inequalities in education in Brazil and help teachers progress, not only from the educational but also from the social viewpoint. This study aimed to analyze indicators of physical and mental quality of life among science teachers in a public school located in the interior of Rio Grande do Sul, Brazil. The information that was assessed included the demographic, economic, occupational and domestic characteristics, data on physical exertion and health, on mental health conditions, as well as the medical diagnoses most frequently mentioned by the teachers ever since they started working in the profession. The teachers brought up several complaints about their health and quality of life, such as nervousness; hoarseness; back, arm and leg pain; and tingling and swelling in the legs. It was concluded that knowledge of this evidence may contribute to preparing measures for the systematic reorganization of the work and directly influence the teachers' quality of life, affording them better performance in the activity of educating.

O objetivo deste estudo foi caracterizar o efeito do alumínio (Al) na concentração de zinco (Zn), manganês (Mn), ferro (Fe) e cobre (Cu) em quatro clones de batata (Macaca e Dakota Rose: sensíveis ao Al; e SMIC148-A e Solanum microdontum: tolerantes ao Al) crescendo em solução nutritiva (pH 4,0) com 0, 50, 100, 150 e 200mg Al L-1. A concentração de Zn e Fe em raízes diminuiu linearmente com o aumento dos níveis de Al nos clones Macaca, SMIC148-A e Dakota Rose e aumentou linearmente em S. microdontum. Na parte aérea, a concentração de Zn mostrou resposta quadrática ao Al em S. microdontum e SMIC148-A, enquanto no clone Dakota Rose houve uma resposta cúbica. Nos clones S. microdontum, SMIC148-A e Dakota Rose, a concentração de Fe mostrou resposta quadrática ao Al. A concentração de Mn em raízes diminuiu linearmente em relação ao Al nos clones Macaca e SMIC148-A e aumentou linearmente em S. microdontum. Para Dakota Rose e SMIC148-A, a concentração de Mn mostrou uma resposta quadrática em relação ao Al em raízes e parte aérea. A concentração de Mn na parte aérea aumentou de forma cúbica com os níveis de Al no clone Dakota Rose. Na parte aérea, não houve alteração na concentração de Zn e Fe na Macaca e de Mn nos clones Macaca e S. microdontum. Em raízes e na parte aérea, a concentração de Cu aumentou linearmente no clone Dakota Rose e mostrou resposta quadrática no clone Macaca. A concentração de Cu mostrou resposta quadrática com os níveis de Al em raízes dos clones S. microdontum e SMIC148-A. Na parte aérea, a concentração de Cu aumentou linearmente no clone S. microdontum e diminuiu linearmente no clone SMIC148-A com o aumento nos níveis de Al. Portanto, a acumulação excessiva de Al afetou negativamente a absorção e a distribuição de Zn, Fe, Mn e Cu nas raízes e na parte aérea dos clones de batata. A resposta da concentração de Cu na parte aérea ao Al foi menos alterada nos clones Al-tolerantes que naqueles Al-sensíveis. A tolerância ao Al em S. microdontum pode estar conectada com os maiores níveis de Zn, Fe e Mn nas raízes.

The objective of this study was to evaluate the effects of aluminum (Al) on the zinc (Zn), manganese (Mn), iron (Fe) and copper (Cu) concentrations in four potato clones (Macaca and Dakota Rose: both Al-sensitive clones; and SMIC148-A and Solanum microdontum: both Al-tolerant-clones), grown in a nutrient solution (pH 4.00) with 0, 50, 100, 150 and 200mg Al L-1. Root Zn and Fe concentrations decreased linearly with the increase of Al levels in Macaca, SMIC148-A and Dakota Rose and increased linearly in S. microdontum. Shoot Zn concentration showed a quadratic relationship with Al in S. microdontum and SMIC148-A, but a curvilinear response in Dakota Rose. Shoot Fe concentration showed a quadratic relationship with Al in S. microdontum, SMIC148-A and Dakota Rose. Root Mn concentration decreased linearly in Macaca and SMIC148-A, and increased linearly in S. microdontum with Al levels. Mn concentration showed a quadratic relationship with Al in roots of Dakota Rose and in shoot of SMIC148-A, and increased curvilinearly with Al levels in shoot of Dakota Rose. In shoot, there was no alteration in Zn, Fe and Mn in Macaca and Mn concentration in S. microdontum. Roots and shoot Cu concentration increased linearly in Dakota Rose, and showed quadratic relationship with Al in Macaca. Roots Cu concentration showed a quadratic relationship with Al levels in S. microdontum and SMIC148-A. Shoot Cu concentration increased linearly in S. microdontum, and decreased linearly in SMIC148-A. Therefore, the excessive Al accumulation affected the uptake and distribution of Zn, Fe, Mn and Cu in roots and shoot of potato clones.The response of shoot Cu concentration to Al was less altered in the Al-tolerant clones than was in Al-sensitive clones. Aluminum tolerance in S. microdontum may be connected with greater levels of Zn, Fe and Mn in the roots.

No presente estudo, avaliou-se o efeito de diferentes níveis de saturação de oxigênio e amônia sobre a atividade das enzimas NTPDase (ecto-nucleosídeo trifosfato difosfohidrolase, E.C. 3.6.1.5) e acetilcolinesterase (AChE, E.C. 3.1.1.7) em encéfalo total de jundiás (Rhamdia quelen). Os peixes foram expostos a diferentes níveis de oxigênio dissolvido e amônia, níveis altos de oxigênio (6,5 mg/L) ou baixos de oxigênio (3,5 mg/L), e níveis altos de amônia (0,1 mg/L) ou baixos de amônia (0,03 mg/L). Os peixes foram divididos em quatro diferentes grupos: (A) controle ou alto nível de oxigênio dissolvido mais baixo nível de NH3; (B) baixo nível de oxigênio dissolvido mais baixo nível de NH3; (C) alto nível de oxigênio dissolvido mais alto nível de amônia; (D) baixo nível de oxigênio dissolvido mais alto nível de NH-3. As atividades de ambas as enzimas nos grupos C e D somente foram alteradas após 24 horas de exposição. A hidrólise do ATP e ADP em encéfalo total de jundiás foi aumentada após 24h de exposição para 104% e 155% no grupo D quando comparado ao grupo controle, respectivamente. A atividade da AChE apresentou após 24h de exposição um aumento de 37% no grupo C e 27% no grupo D, ambos comparados ao grupo controle. Os resultados obtidos corroboram com a hipótese que baixos níveis de saturação de oxigênio aumentam a toxicidade da amônia. Além disso, os eventos de hipóxia podem aumentar o fluxo sanguíneo, e este evento aumenta a atividade da NTPDase produzindo adenosina, um potente vasodilatador

The effects of various levels of oxygen saturation and ammonia concentration on NTPDase (ecto-nucleoside triphosphate diphosphohydrolase, E.C. 3.6.1.5) and acetylcholinesterase (AChE, E.C. 3.1.1.7) activities in whole brain of teleost fish (Rhamdia quelen) were investigated. The fish were exposed to one of two different dissolved oxygen levels, including high oxygen (6.5 mg.L-1) or low oxygen (3.5 mg.L-1), and one of two different ammonia levels, including high ammonia (0.1 mg.L-1) or low ammonia (0.03 mg.L-1) levels. The four experimental groups included the following (A) control, or high dissolved oxygen plus low NH3; (B) low dissolved oxygen plus low NH3; (C) high dissolved oxygen plus high NH3; (D) low dissolved oxygen plus high NH3. We found that enzyme activities were altered after 24 h exposure in groups C and D. ATP and ADP hydrolysis in whole brain of fish was enhanced in group D after 24 h exposure by 100% and 119%, respectively, compared to the control group. After 24 h exposure, AChE activity presented an increase of 34% and 39% in groups C and D, respectively, when compared to the control group. These results are consistent with the hypothesis that low oxygen levels increase ammonia toxicity. Moreover, the hypoxic events may increase blood flow by hypoxia increasing NTPDase activity, thus producing adenosine, a potent vasodilator.

As fosfatases ácidas (3.1.3.2) são um grupo de enzimas amplamente distribuídas na natureza, as quais catalisam a hidrólise de uma variedade de ésteres de fosfato com uma variação de pH entre quatro e seis. Foi confirmada a presença de fosfatases ácidas em plântulas de pepino (Cucumis sativus), rabanete (Raphanus sativus) e rúcula (Eruca vesicaria) sob diferentes condições de ensaio usando uma preparação rápida e simples. Os resultados mostraram que o pH e a temperatura ótimos para todas as espécies foram 5,5 e 35°C, respectivamente. A enzima foi inibida por molibdato, fluoreto, azida, levamisole, ortovanadato, Zn2+ e Cu2+. O inibidor suramim não afetou a atividade enzimática. As fosfatases ácidas de pepino, rabanete e rúcula hidrolisaram uma ampla variedade de ésteres de fosfato e a maior atividade foi observada com PPi, ATP e GTP para pepino e rabanete e PPi, frutose-6-fosfato e GTP para rúcula. Esses resultados demonstraram que a enzima investigada neste estudo é diferente das conhecidas enzimas de plantas que clivam ésteres de fosfato (apirase e pirofosfatases inorgânicas). Desse modo, esta preparação pode ser uma ferramenta útil para futuros estudos toxicológicos e para se estudar inicialmente todas as isoformas das fosfatases ácidas.

Acid phosphatases (3.1.3.2) are a group of enzymes widely distributed in nature, which catalyze the hydrolysis of a variety of phosphate esters in the pH range of 4-6. We confirmed the presence of acid phosphatases in seedlings of cucumber (Cucumis sativus), radish (Raphanus sativus) and rocket salad (Eruca vesicaria) under different assay conditions using a rapid and simple preparation. The results showed that the optimum pH and temperature used for all species were close to 5.5 and 35°C, respectively. The enzyme was inhibited by molybdate, fluoride, azide, levamisole, orthovanadate, Zn2+ and Cu2+. Suramin had no effect on enzyme activity. The acid phosphatase from cucumber, radish and rocket salad hydrolyzed a wide variety of phosphate esters and the highest activity was observed with PPi, ATP and GTP. These results demonstrate that the enzyme investigated in this study is different from well known ester phosphate cleaving plant enzymes (apyrase and inorganic pyrophosphatases) and this preparation could be a useful tool to future toxicological studies and to study initially all isoforms of acid phosphatase.

Este estudo verificou a eficiência do extrato etanólico da casca de Syzygium cumini sobre o sistema colinérgico de ratos normais e diabéticos induzidos com aloxano. Os animais foram divididos em grupo controle (C), tratado com Syzygium cumini (TS), diabético (D) e diabético tratado com Syzygium cumini (DS). A atividade da acetilcolinesterase (AChE) foi analisada nas seguintes estruturas cerebrais: cerebelo, córtex, estriado e hipocampo. O extrato etanólico da casca de Syzygium cumini na dose de 1g.kg-1 foi administrado diariamente por um período de trinta dias. Foi verificado após este período que o extrato inibiu a atividade da AChE no cerebelo e córtex cerebral dos ratos do grupo DS (P<0,05), comparado com o TS. No estriado houve um aumento significativo na atividade da AChE nos ratos do grupo TS (P<0,01) comparado com o C, e no hipocampo não foi encontrada nenhuma variação significativa. Esses resultados indicam que o extrato da casca do Jambolão, possui um efeito inibitório da AChE no cerebelo e córtex cerebral e um efeito ativador sobre essa enzima no estriado, indicando uma possível alteração na funcionalidade do sistema colinérgico nestas estruturas cerebrais.

The present study verified the efficiency of the bark ethanol extract of Syzygium cumini on the cholinergic system of normal and alloxan induced diabetic rats. Thirty-nine female rats were divided in control (C), treated with Syzygium cumini (TS), diabetic (D) and diabetic treated with Syzygium cumini (DS). The activity of acetylcholinesterase (AChE) was analyzed in the following cerebral structures: cerebellum, cortex, striatum and hippocampus. The extract of the bark of Syzygium cumini in the dose of 1g.kg-1 was administered orally daily for a period of thirty days. After this period the extract inhibited the activity of the AChE in the cerebellum and cerebral cortex of the rats in the DS group (P<0.05) as, compared to TS. In the striatum there was a significant increase in the activity of the AChE in rats of the TS group (P<0.01) when compared to the C group, and in the hippocampus there was no significant variation. These results indicate that the bark extract of "Jambolão"has an inhibitory effect on AChE in the cerebellum and cerebral cortex and an stimulatory effect on striatum, indicating a possible alteration in the functionality of the cholinergic system in such cerebral structures.

O Syzygium cumini, também conhecido como Jambolão é amplamente utilizado na medicina popular para o tratamento da diabetes melito. Este estudo verificou a eficiência do extrato da casca de Syzygium cumini sobre os níveis glicêmicos e estresse oxidativo de ratos normais e diabéticos induzidos por aloxano. Os animais foram divididos em grupo controle (C), controle tratado (CT), diabético controle (DC) e diabético tratado (DT). A administração oral do extrato aquoso da casca de Jambolão, na dose de 1g/kg de peso vivo, por um período de trinta dias, não resultou em uma redução significativa na glicemia e nos níveis de hemoglobina glicosilada. Neste estudo, o tratamento com o extrato demonstrou um aumento dos níveis de substâncias reativas ao ácido tiobarbitúrico (TBA-RS) no plasma dos ratos do grupo DT (P<0,05), comparado com o C. A atividade da catalase nos rins dos ratos do grupo DC diminuiu significativamente (P<0,01) e no fígado houve uma elevação significativa dessa enzima no grupo DC (P<0,01). Estes resultados indicam que o extrato da casca do Jambolão não possui efeito hipoglicemiante em ratos diabéticos induzidos pelo aloxano. O efeito antioxidante desta planta não foi suficiente para diminuir significativamente a produção de TBA-RS. A diminuição da atividade da catalase nos rins pode ser devida à exaustão ou inibição desta enzima e seu aumento, no fígado, devido ao estresse oxidativo, ocasionado pelo estado diabético.

The plant Syzygium cumini, also known as Black Plum is used widely in the popular medicine for the treatment of diabetes mellitus. This study verified the efficiency of the extract of the bark of Syzygium cumini on the glycemic levels and oxidative stress of normal and diabetic rats induced by alloxan. The animals were divided in control group (C), control treatment group (CT), diabetic control group (DC) and diabetic treatment group (DT). Oral administration of the aqueous extract of the bark of Syzygium cumini, in the dose of 1g/kg live weight, for a period of thirty days, did not result in a significant reduction on glycemia or on glucosilated hemoglobin levels. The treatment with the extract demonstrated an increase in the levels of thiobarbituric acid reactive substances (TBA-RS) in the plasma of the DT group (P <0.05), compared to the C. Kidney catalase activity of rats in DC group decreased significantly (P <0.01) and, in the liver, it showed a significant elevation in group DC (P <0.01). Those results indicated that the extract of Syzygium cumini bark does not possess hypoglycemic effect in alloxan diabetic induced rats. The antioxidant effect of this plant was not enough to reduce the production of TBA-RS significantly. The decrease of catalase activity in the kidneys can be owed to its exhaustion or inhibition, and its increase in the liver could be due to oxidative stress caused by the diabetic status.
