Utilizaram-se os modelos lineares generalizados com as funções de ligação probit e logit na avaliação da prenhez precoce, e observaram-se os efeitos na variabilidade genética e na seleção de reprodutores quando diferentes idades são adotadas na definição dessa característica. A prenhez precoce foi estudada aos 15 (PP15) e 21 meses (PP21). Correlações entre os valores genéticos preditos e a porcentagem de touros em comum, considerando 10% dos touros com maiores valores genéticos (TOP10), entre a classificação dos modelos com função de ligação logit e probit e em cada modelo entre PP15 e PP21, foram calculadas. As herdabilidades para PP15 e PP21 foram próximas entre os modelos, exceto para PP15 utilizando a função de ligação probit. Aquelas entre os modelos e a TOP/10 foram altas. Os critérios de Akaike e Bayesiano apresentaram-se semelhantes entre os modelos. As correlações entre PP15 e PP21 e a TOP10, considerando o mesmo modelo, foram de média magnitude.

This study aimed to use the generalized linear models with probit and logit link function to evaluate early pregnancy, and to observe the effects on genetic variability and on sire selection when different ages are adopted in the definition of this trait. Early pregnancy was studied at 15 (EP15), and 21 (EP21) months. The analysis was done in R software. Pearson correlations (PC), between genetic predicted values and percentage of bulls in common considering only 10% of bulls with higher genetic values (TOP 10), between classification by logit and probit models and in each model among EP15 and EP21, were calculated. The heritability for EP15 and EP21 were close between models, except for EP15 using probit link function. PC and TOP10 among models were high. The Akaike and Bayesian criteria reported was similar between models. TOP10, considering the same model, among EP15-EP21 were moderated between EP15-EP21.

O trabalho teve como objetivos avaliar a morfometria dos frutos e sementes de carobinha (Jacaranda decurrens subsp. symmetrifoliolata) e o efeito de temperaturas na germinação de sementes armazenadas em ambiente sem controle de temperatura e umidade relativa. O estudo foi realizado no Laboratório de Sementes da UFGD, Dourados, MS. Em amostra de 120 frutos e de 550 sementes foi determinado: o comprimento, a largura, a espessura, o número de sementes por fruto, e a massa dos frutos. A germinação foi estudada após três períodos de armazenamento das sementes (120, 480 e 720 dias) a temperatura ambiente e sob quatro temperaturas de germinação: três constantes (18, 25 e 30ºC) com luz contínua e sob condição de tempera alternada, 20-30ºC, com fotoperíodo de 12 horas. O delineamento experimental foi inteiramente ao acaso, em arranjo fatorial 3 x 4, com quatro repetições. As variações nas medidas de frutos e sementes representam indício da alta variabilidade genética populacional. As sementes armazenadas por 120 dias e submetidas à germinação na temperatura de 25ºC resultaram em 69,4% de germinação e índice de velocidade de germinação - IVG, de 0,983. Sob temperaturas de germinação de 30ºC, constante, e de 20-30ºC, alternada, obtiveram-se 63,9% e 63,8% de germinação e 0,486 e 0,413 de IVG, respectivamente. Sob temperatura de 18ºC a germinação foi lenta e menor, 27,7% e IVG de 0,119. As sementes armazenadas por 480 e 720 dias não germinaram. Com base no comportamento fisiológico das sementes é possível classificar Jacaranda decurrens subsp. symmetrifoliolata como espécie intermediária (entre ortodoxa e recaucitrante).

This study aimed to evaluate the morphometry of fruits and seeds of Jacaranda decurrens subsp. symmetrifoliolata ("carobinha") and the effect of temperature on the germination of seeds that were stored in environment without controlled temperature and relative humidity. The study was carried out at Seed Laboratory of Federal University of Grande Dourados, Mato Grosso do Sul, Brazil. For a sample of 120 fruits and 550 seeds, the following variables were determined: length, width and thickness, besides the number of seeds per fruit and fruit biomass. Germination was studied after three seed storage periods (120, 480 and 720 days) at environment temperature and four germination temperatures: three constant temperatures (18, 25 and 30ºC) with continuous light, and one alternating temperature, 20-30ºC, with 12-hour photoperiod. Experimental design was completely randomized, in 3 x 4 factorial schemes, with four replicates. The variations in the measures of fruits and seeds evidence a high genetic variability in the population. The seeds stored for 120 days and subjected to germination test at 25ºC resulted in 69.4% germination and germination velocity index - GVI of 0.983. At 30ºC, constant temperature, and 20-30ºC, alternating temperature, germination was 63.9% and 63.8% while GVI was 0.486 and 0.413, respectively. At 18ºC, germination was slow and lower, 27.7%, and GVI was 0.119. Seeds stored for 480 and 720 days did not germinate. Based on the physiological behavior of seeds, it is possible to classify Jacaranda decurrens subsp. symmetrifoliolata as an intermediate species (between orthodox and recalcitrant).

Várias plantas têm sido consideradas produtos terapêuticos, dentre elas destacam-se os chás verde e preto, popularmente utilizados para controle da hiperglicemia e obesidade. Objetivou-se neste trabalho avaliar o potencial inibitório sobre as enzimas α-amilase, α e β-glicosidases e o teor de compostos fenólicos do chá verde e do chá preto. O teor de compostos fenólicos encontrados foram de 80,8 ± 0,43 mg g-1 no chá preto e 32,0 ± 0,12 mg g-1 no chá verde. O chá verde e o chá preto, em condições de consumo, inibiram as enzimas em estudo, porém, após a simulação do fluido gástrico o inibidor presente no chá verde perdeu a ação. O chá preto deixou de inibir a α-amilase e apresentou inibição inalterada para α e β-glicosidases. Tais resultados sugerem que o chá preto pode ser auxiliar em dietas de restrição de carboidratos.

Several plants have been considered therapeutic products, including green and black tea, popularly used to control hyperglycemia and obesity. This study aimed to evaluate the inhibitory potential of the enzymes α-amylase, α and β-glycosidases, as well as the content of phenolic compounds in green tea and black tea. The concentrations of phenolic compounds found were 80.8 ± 0.43 mg g-1 in black tea and 32.0 ± 0.12 mg g-1 in green tea. Under the tested conditions of use, green and black tea inhibited the enzyme under study. However, after simulation of the gastric fluid, the inhibitor present in green tea lost its action. Black tea no longer inhibited a-amylase and showed unaltered inhibition for α and β-glycosidases. These results suggest that black tea can be helpful in diets restricting carbohydrates.

Cinco descritores bioquímicos foram avaliados em algodoeiro infectados com o fungo causador da ramulose visando detectar respostas associadas à infecção. Sementes de quatro cultivares com diferentes níveis de resistência à doença foram semeadas em vasos plásticos, em casa de vegetação. Vinte dias após a emergência, as plântulas foram inoculadas com uma suspensão de 1 x 10(6) conídios/mL de Colletotrichum gossypii var. cephalosporioides. Folhas foram coletadas aos 3, 15 e 30 dias após a inoculação e utilizadas para determinação de prolina livre, catalase, peroxidase, carboidratos solúveis e proteínas totais. Verificou-se que os teores de prolina livre, peroxidase e catalase foram discriminadores na reação de plantas infectadas. Estes descritores apresentaram rápida resposta nos primeiros dias após a infecção das plantas, sendo mais expressivo na cv. resistente BRS Antares. Os descritores prolina livre e catalase podem ser utilizados como ferramentas auxiliares na identificação de cultivares resistentes à ramulose nos programas de melhoramento da cultura do algodão.

Five biochemical descriptors were assessed in cotton plants infected with the ramulosis-causing fungus in order to detect differences associated with the infection. Seeds from four cotton cultivars with different levels of resistance were sown in plastic containers in greenhouse conditions. At 20 days after emergence, plants were inoculated with a suspension at 1 x 10(6) conidia/mL of Colletotrichum gossypii var. cephalosporioides. Leaves were collected at 3, 15 and 30 days after inoculation and free proline, catalase, peroxidase, soluble carbohydrates and total proteins were determined. Proline, peroxidase and catalase levels discriminated the reaction of infected plants. These descriptors changed soon after infection and the change was more evident in the resistant BRS Antares variety. Free proline and catalase descriptors can be used in the identification of ramulosis-resistant cultivars in cotton breeding programs.

We present kinetic and physiological data regarding the culturing of rCHO-K1 cells on various microcarriers, to evaluate the potential of this culture strategy for mass production of these cells and expression of a recombinant disintegrin. Cultures were performed in 500 mL spinner flasks in DMEM culture medium with 10% v/v fetal calf serum, gently shaken at 37ºC, pH 7.4, in a 10% v/v CO2 atmosphere. The following values were obtained, respectively, for the adhesion time-constant Ka (h) and specific growth rate μmax (d-1) on each microcarrier: Cytodex 1 (0.91, 0.45), Cultispher S (0.28, 0.34), Immobasil FS (0.85, 0.52) and Pronectin F (5.12, 0.67). Metabolic characteristics showed some variation among the cultures with the four microcarriers, the most significant being the higher production of ammonia with microcarriers coated with adhesive molecules (Cultispher S and Pronectin F) relative to the uncoated carriers (Cytodex 1 and Immobasil FS). Experiments where the DMEM medium was gradually replaced by the serum-free medium (CHO-SFM-II) revealed important advantages over media containing serum, not only for assay purposes, but also for purification of the disintegrin. Altogether these results demonstrate that cultures on microcarriers, especially on Pronectin F, show good potential for larger scale cultures of rCHO-K1 cell
