ABSTRACT Lead is a heavy metal and an important cause of acute or chronic toxicosis in humans, domestic, and wild animals. This report aims to describe a case of chronic lead poisoning in a jaguar (Panthera onca) kept under human care that was rescued from the wild environment. The animal was rescued in poor condition in 2004 and kept under human care at the Belo Horizonte Zoological Garden (Minas Gerais, Brazil) until 2020, when it presented with anorexia, vomiting and ataxia. Over the past years the animal had episodes of anemia and increased serum urea and creatinine. Radiography demonstrated 21 radiopaque projectiles on the right side of the face. At necropsy there were multiple projectiles surrounded by fibrous tissue in the subcutaneous of the right side of the face, fibrinous peritonitis, multiple gastric ulcers, and melena. The lead dosage was performed using the atomic absorption spectrometry technique using renal tissue collected at necropsy, with a result of 908 ppb (µg/kg). The findings of projectiles associated with the dosage of lead above the reference limits allow the diagnosis of chronic intoxication in this case.
RESUMO Chumbo é um metal pesado e uma causa importante de toxicose crônica no homem e em animais domésticos e selvagens. Este relato teve como objetivo descrever um caso de intoxicação crônica por chumbo em uma onça-pintada (Panthera onca), mantida sob cuidados humanos, que foi resgatada de seu ambiente natural em 2004, em pobre condição corporal. O animal foi encaminhado ao zoológico de Belo Horizonte (MG, Brasil), onde permaneceu até 2020, quando desenvolveu anorexia, vômitos e ataxia. Durante os últimos anos, o animal teve episódios de anemia e aumento da concentração sérica de ureia e creatinina. O exame radiográfico demonstrou 21 projéteis radiodensos na face direita. À necropsia, foram observados vários projéteis envoltos por tecido conjuntivo no subcutâneo da face esquerda, peritonite fibrinosa, múltiplas úlceras gástricas e melena. A concentração de chumbo foi determinada por espectrometria de absorção atômica em amostras de tecido renal, indicando 908 ppb (µg/kg). O achado de projéteis de chumbo associado à elevada concentração de chumbo é compatível com intoxicação crônica por chumbo neste caso.

Diabetes is associated with a worse prognosis and a high risk of morbidity and mortality in COVID-19 patients. We aimed to evaluate the main factors involved in the poor prognosis in diabetic patients. A total of 984 patients diagnosed with COVID-19 admitted to the hospital were included in this study. Patients were first divided into type-2 diabetic (DM+) and non-diabetic (DM–) groups. The participants were analyzed based on the National Early Warning Score (NEWS) and on the Quick-Sequential Organ Failure Assessment (qSOFA) to find the best prognostic risk score for our study. The DM+ and DM– groups were divided into non-severe and severe groups. Comparative and correlative analyses were used to identify the physiological parameters that could be employed for creating a potential risk indicator for DM+ COVID-19 patients. We found a poorer prognosis for the DM+ COVID-19 patients with a higher ICU admission rate, mechanical ventilation rate, vasopressor use, dialysis, and longer treatment times compared with the DM– group. DM+ COVID-19 patients had increased plasma glucose, lactate, age, urea, NEWS, and D-dimer levels, herein referred to as the GLAUND set, and worse prognosis and outcomes when compared with infected DM– patients. The NEWS score was a better indicator for assessing COVID-19 severity in diabetic patients than the q-SOFA score. In conclusion, diabetic COVID-19 patients should be assessed with the NEWS score and GLAUND set for determining their prognosis COVID-19 prognosis.

Liver fibrosis occurring as an outcome of non-alcoholic steatohepatitis (NASH) can precede the development of cirrhosis. We investigated the effects of sorafenib in preventing liver fibrosis in a rodent model of NASH. Adult Sprague-Dawley rats were fed a choline-deficient high-fat diet and exposed to diethylnitrosamine for 6 weeks. The NASH group (n=10) received vehicle and the sorafenib group (n=10) received 2.5 mg·kg-1·day-1 by gavage. A control group (n=4) received only standard diet and vehicle. Following treatment, animals were sacrificed and liver tissue was collected for histologic examination, mRNA isolation, and analysis of mitochondrial function. Genes related to fibrosis (MMP9, TIMP1, TIMP2), oxidative stress (HSP60, HSP90, GST), and mitochondrial biogenesis (PGC1α) were evaluated by real-time quantitative polymerase chain reaction (RT-qPCR). Liver mitochondrial oxidation activity was measured by a polarographic method, and cytokines by enzyme-linked immunosorbent assay (ELISA). Sorafenib treatment restored mitochondrial function and reduced collagen deposition by nearly 63% compared to the NASH group. Sorafenib upregulated PGC1α and MMP9 and reduced TIMP1 and TIMP2 mRNA and IL-6 and IL-10 protein expression. There were no differences in HSP60, HSP90 and GST expression. Sorafenib modulated PGC1α expression, improved mitochondrial respiration and prevented collagen deposition. It may, therefore, be useful in the treatment of liver fibrosis in NASH.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Objetivou-se com este trabalho descrever as medidas biométricas de cordeiros lactantes da raça Santa Inês criados em condições amazônicas; estabelecer relação entre a idade e as medidas corporais; bem como correlacionar essas medidas biométricas com o peso corporal. Utilizaram-se 75 animais criados em sistema semi-intensivo, submetidos a mensurações do peso corporal (PC), comprimento corporal (CC), altura do anterior (AA) e do posterior (AP), perímetro torácico (PT), largura da garupa (LG) e do peito (LP), comprimento da perna (CP), perímetro da perna (PP) e compacidade corporal (COMPC), ao nascer e aos 15, 30, 45 e 60 dias de idade. Somente as medidas LG e PP demonstraram regressões com R2abaixo de 0,70 em relação à idade dos cordeiros. Todas as medidas corporais utilizadas nesta pesquisa, individualmente, demonstraram r positivo com o peso corporal. As variáveis biométricas que melhor compuseram a equação de predição do peso dos cordeiros utilizados neste estudo foram CC, AP, PT, LG e PP. Concluiu-se que a idade de cordeiros pode ser utilizada como preditora do peso e de algumas medidas biométricas corporais e que o peso pode ser estimado por meio de algumas medidas biométricas.
This study aimed to 1) describe the biometric measurements of Santa Inês lambs until weaning, when raised in the Amazonia conditions; 2) establish the relationship between age and body measurements; and 3) correlate these biometric measurements to body weight. 75 animals submitted to semi-intensive breeding were used, and underwent measurements of body weight (BW), body length (BL), height of the forelimb (HF) and hindlimb (HH), thoracic perimeter (TP), width of rump (WR) and chest (WC), length of leg (LL), perimeter of leg (PL) and more body compacity (COMP), at birth and at 15, 30, 45 and 60 days of age. As a result, the WR and PL measurements demonstrated regressions with R2 below 0.70 in relation to age of lambs. All biometrical measurements used in this study individually demonstrated positive r with body weight. Biometric variables that compose the equation for predicting weight of lambs used in this study were BL, HH, TP, WR and PL. It was concluded that the age of lambs can be used as a predictor of weight and some body biometric measurements. The weight can also be estimated by some biometric measurements.

Sepsis involves a systemic inflammatory response of multiple endogenous mediators, resulting in many of the injurious and sometimes fatal physiological symptoms of the disease. This systemic activation leads to a compromised vascular response and endothelial dysfunction. Purine nucleotides interact with purinoceptors and initiate a variety of physiological processes that play an important role in maintaining cardiovascular function. The purpose of the present study was to investigate the effects of ATP on vascular function in a lipopolysaccharide (LPS) model of sepsis. LPS induced a significant increase in aortic superoxide production 16 h after injection. Addition of ATP to the organ bath incubation solution reduced superoxide production by the aortas of endotoxemic animals. Reactive Blue, an antagonist of the P2Y receptor, blocked the effect of ATP on superoxide production, and the nonselective P2Y agonist MeSATP inhibited superoxide production. Nitric oxide synthase (NOS) inhibition by L-NAME blocked vascular relaxation and reduced superoxide production in LPS-treated animals. In the presence of L-NAME there was no ATP effect on superoxide production. A vascular reactivity study showed that ATP increased maximal relaxation in LPS-treated animals compared to controls. The presence of ATP induced increases in Akt and endothelial NOS phosphorylated proteins in the aorta of septic animals. ATP reduces superoxide release resulting in an improved vasorelaxant response. Sepsis may uncouple NOS to produce superoxide. We showed that ATP through Akt pathway phosphorylated endothelial NOS and “re-couples” NOS function.

Shock and resuscitation render patients more susceptible to acute lung injury due to an exacerbated immune response to subsequent inflammatory stimuli. To study the role of innate immunity in this situation, we investigated acute lung injury in an experimental model of ischemia-reperfusion (I-R) followed by an early challenge with live bacteria. Conscious rats (N = 8 in each group) were submitted to controlled hemorrhage and resuscitated with isotonic saline (SS, 0.9% NaCl) or hypertonic saline (HS, 7.5% NaCl) solution, followed by intratracheal or intraperitoneal inoculation of Escherichia coli. After infection, toll-like receptor (TLR) 2 and 4 mRNA expression was monitored by RT-PCR in infected tissues. Plasma levels of tumor necrosis factor α and interleukins 6 and 10 were determined by ELISA. All animals showed similar hemodynamic variables, with mean arterial pressure decreasing to nearly 40 mmHg after bleeding. HS or SS used as resuscitation fluid yielded equal hemodynamic results. Intratracheal E. coli inoculation per se induced a marked neutrophil infiltration in septa and inside the alveoli, while intraperitoneal inoculation-associated neutrophils and edema were restricted to the interseptal space. Previous I-R enhanced lung neutrophil infiltration upon bacterial challenge when SS was used as reperfusion fluid, whereas neutrophil influx was unchanged in HS-treated animals. No difference in TLR expression or cytokine secretion was detected between groups receiving HS or SS. We conclude that HS is effective in reducing the early inflammatory response to infection after I-R, and that this phenomenon is achieved by modulation of factors other than expression of innate immunity components.

Estudaram-se os efeitos de soluções salinas isotônica e hipertônica em eqüinos hipovolêmicos sobre as concentrações séricas de sódio, cloreto e potássio e freqüências cardíaca (FC) e respiratória (FR). Quinze eqüinos machos com peso entre 390 e 475kg e idades entre quatro e 18 anos foram submetidos à retirada de sangue correspondente a 2% do peso corporal e distribuídos em três grupos de igual número: o grupo GSH recebeu solução hipertônica de NaCl a 7,5% em glicose a 5%; o GSI, solução isotônica de NaCl a 0,9%; e o GC não foi tratado. Os eletrólitos séricos foram avaliados antes (T0), após a retirada de sangue (T1) e após a infusão das soluções, entre 20 e 30 minutos (T2), entre 60 e 70 minutos (T3) e entre 120 e 130 minutos (T4). Após T0, houve elevação da FC e da FR, e as concentrações séricas de Na, Cl, K permaneceram inalteradas. Após a infusão, houve melhora das variáveis clínicas em GSI e GSH, em relação ao GC. Quanto a T3 e T4, os valores de Na em T2 do GSH foram maiores, e os de Cl e de K não se alteraram. As soluções hipertônica e isotônica são seguras na correção da hipovolemia induzida e não produzem alteração eletrolítica significativa.
The effect of isotonic and hypertonic solutions on serum levels of sodium, chloride and potassium and cardiac (CR) and respiratory rates (RR) of hypovolemic horses were studied. Fifteen horses weighting from 390 to 475kg, aging from four to 18-years-old were submitted to bleeding of 2% of body weight and divided in three groups: 7.5% NaCl hypertonic saline in 5% glucose (GSH), 0.9% NaCl isotonic saline and control group (GC). Serum electrolytes were evaluated before (T0) and after bleeding (T1) and after the administration of the solutions between 20 and 30 minutes (T2), 60 and 70 minutes (T3) and 120 and 130 minutes (T4). After T0, CR and RR increased while serum sodium, chloride, potassium were not affected. After the treatment, the clinical variables improved in GSI and GSH as compared to GC. The Na levels increased in GSH at T2 being higher than T3 and T4 while chloride and potassium concentrations did not change. The hypertonic and isotonic solutions safely corrected the hypovolemia of the horses, without altering significantly the electrolyte balance.

Oxidative stress and hepatic mitochondria play a role in the pathogenesis of nonalcoholic fatty liver disease. The aim of the present study was to evaluate the role of hepatic mitochondrial dysfunction and oxidative stress in the pathogenesis of the disease. Fatty liver was induced in Wistar rats with a choline-deficient diet (CD; N = 7) or a high-fat diet enriched with PUFAs-omega-3 (H; N = 7) for 4 weeks. The control group (N = 7) was fed a standard diet. Liver mitochondrial oxidation and phosphorylation were measured polarographically and oxidative stress was estimated on the basis of malondialdehyde and glutathione concentrations. Moderate macrovacuolar liver steatosis was observed in the CD group and mild liver steatosis was observed in the periportal area in the H group. There was an increase in the oxygen consumption rate by liver mitochondria in respiratory state 4 (S4) and a decrease in respiratory control rate (RCR) in the CD group (S4: 32.70 ± 3.35; RCR: 2.55 ± 0.15 ng atoms of O2 min-1 mg protein-1) when compared to the H and control groups (S4: 23.09 ± 1.53, 17.04 ± 2.03, RCR: 3.15 ± 0.15, 3.68 ± 0.15 ng atoms of O2 min-1 mg protein-1, respectively), P < 0.05. Hepatic lipoperoxide concentrations were significantly increased and the concentration of reduced glutathione was significantly reduced in the CD group. A choline-deficient diet causes moderate steatosis with disruption of liver mitochondrial function and increased oxidative stress. These data suggest that lipid peroxidation products can impair the flow of electrons along the respiratory chain, causing overreduction of respiratory chain components and enhanced mitochondrial reactive oxygen species. These findings are important in the pathogenesis of nonalcoholic fatty liver disease.

In this work two different synthesis methods of perovskites, SrCo0.5FeO3, were compared: combustion synthesis and oxides mixture aiming at evaluating their use as membranes for partial oxidation of methane. The combustion synthesis method explores an exothermic, generally very fast and self-sustaining chemical reaction between the desired metal salts and a suitable organic fuel, which is ignited at a temperature much lower than the actual phase formation temperature. The oxides mixture are based on a physical mixture of the powder oxides followed by calcination to obtain the desired phase. In order to obtain the membranes, we studied the conformation of bodies and the temperatures of sintering in the two powders synthesized. The powders were analyzed by density and grain size distribution and characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM). After conformation, in cylindrical form, the green bodies were analyzed by density. After sintering at 1150 °C, the membranes were analyzed by density and they were characterized by XRD and SEM. The powder obtained by combustion synthesis shows lower density and fine grains than the other obtained by oxides mixture. The membranes obtained present very different morphology depending on the precursor powder synthesis. The sintered membranes obtained by combustion method also present a very uniform morphology without segregation.

Determinou-se o crescimento e a extração de nutrientes pelos frutos das variedades, Haden, Sensation, Tommy-Atkins e Edward, colhidos em sete épocas distintas, de um pomar de nove anos situado sobre uma "terra roxa estruturada" em Piracicaba, SP. Os frutos foram lavados, pesados e analisados para macro e micronutrientes. O crescimento dos frutos nas variedades obedece a seguinte ordem decrescente: Edward, Haden, Tommy-Atkins e Sensation. O conteúdo total de nutrientes nas variedades foi em ordem decrescente: Haden , Tommy-Atkins, Edward e Sensation.
Quantities of nutrients removed by the harvest of mango fruits (Mangifera indica L.) of four varieties (Haden, Sensation, Tommy-Atkins and Edward) are presented. The orchard was nine years old, situated on an alfison in Piracicaba, SP, Brazil. The fruits were harvested 39, 55, 97, 108, 125 and 144 days from the initial stage of growth. The fruits were rinsed, dried, weighed, and analyzed for nutrient content by laboratory conventional methods. The authors concluded: Fruit growth followed the decreasing order: Edward, Haden, Tommy-Atkins and Sensation; The total amount of nutrients (macro and micronutrients) removed by each variety for the production of 15,000kg of fresh fruits by hectare amounted to: Haden 44kg, Sensation 29kg, Tommy-Atkins 40kg and Edward 39kg.
