Molecular dereplication of volatile oils from Saururus cernuus L. and evaluation of anti-Trypanosoma cruzi activity

The present study reports the molecular dereplication and the evaluation of anti-Trypanosoma cruzi activity of volatile oils from inflorescences, leaves, branches, and roots of Saururus cernuus L. (Saururaceae). Chemically, the oils showed the predominance of sesquiterpenes in inflorescences (64.04%) and branches (63.82%), with β-sesquiphellandrene (25.50%) and (E)-caryophyllene (22.40%) corresponding to the main constituents of each oil. Furthermore, it was possible to detect safrole as the most predominant compound in the leaves oil (49.09%). On the other hand, the oil from roots was mainly composed by monoterpenes (84.60%), with limonene in higher concentration (38.41%), followed by α-pinene (20.19%), and camphene (14.71%). The oils from inflorescences and branches displayed higher antitrypanosomal potency with EC50 values of 7.1 and 8.8 μg mL-1, respectively, followed by the oils from the roots and inflorescences, with EC50 values of 17.3 and 30.4 μg mL-1, respectively. Additionally, branches and inflorescences oils displayed no toxicity in mammalian NCTC cells (CC50 > 200 μg mL-1). Using two PLS-DA methods, it was possible to suggest that the anti-T. cruzi activity of the tested oils could be associated with the presence of β-sesquiphellandrene, safrole, β-elemene, and α-zingiberene whereas threo-austrobailignan-5, β-sesquiphellandrene, α-humulene, germacrene D, and bicyclogermacrene play a role in the cytotoxicity against NCTC cells.

RESUMO Objetivo Disfunção cognitiva é comum em pacientes com esclerose múltipla. Por isto, a Brief Repeatable Battery of Neuropsychological Tests (BRB–N) foi desenvolvida para avaliar as funções cognitivas mais frequentemente alteradas na doença. Entretanto, estão faltando dados normativos desta bateria no Brasil. Assim, nosso objetivo foi fornecer valores normativos contínuos e discretos da BRB–N para a população brasileira. Métodos Foram recrutados 285 indivíduos sadios da comunidade em 10 centros do Brasil e aplicada a versão A em 237 e a versão B em 48 sujeitos. Normas contínuas foram calculadas com análise de regressão múltipla. Resultados Escores brutos médios e 5°percentil para cada subteste são fornecidos, estratificados por idade e nível educacional. Os escores brutos dos sujeitos sadios foram convertidos em escores de escalas e postos em regressão quanto a idade, sexo e educação, fornecendo equações que podem ser usadas para calcular escores previsíveis. Conclusão Nossos dados normativos permitem um uso mais amplo da BRB–N na prática clínica e na pesquisa, fornecendo normas para dados discretos e contínuos. Normas para dados discretos deveriam ser usadas com cuidado e escores demograficamente ajustados são geralmente preferidos quando interpretando dados neuropsicológicos.
ABSTRACT Objective Cognitive dysfunction is common in multiple sclerosis. The Brief Repeatable Battery of Neuropsychological Tests (BRB–N) was developed to assess cognitive functions most–frequently impaired in multiple sclerosis. However, normative values are lacking in Brazil. Therefore, we aimed to provide continuous and discrete normative values for the BRB–N in a Brazilian population sample. Methods We recruited 285 healthy individuals from the community at 10 Brazilian sites and applied the BRB–N version A in 237 participants and version B in 48 participants. Continuous norms were calculated with multiple–regression analysis. Results Mean raw scores and the 5th percentile for each neuropsychological measure are provided, stratified by age and educational level. Healthy participants' raw scores were converted to scaled scores, which were regressed on age, sex and education, yielding equations that can be used to calculate predicted scores. Conclusion Our normative data allow a more widespread use of the BRB–N in clinical practice and research.

ABSTRACT The food, training, and health are crucial for a good performance in sports. Intense physical activity takes the athlete to maintain a very unstable balance between energy demand and consumption of nutrients. Spirulina microalga has a nutritional profile that renders it an ideal food supplement, because has high protein content, also contains vitamins, minerals, and pigments. In this context, the study aimed to develop, characterize and evaluate the stability of foods enhanced with Spirulina, which are intended for athletes. In this study, six different supplements were developed (electrolyte replenisher, muscle enhancer, and recovery supplement), without and with Spirulina. The electrolyte replenisher with Spirulina compared to the product without the microalga, showed an increase of 0.35% (w/w) in mineral content. The carbohydrates content of the developed recovery supplement with Spirulina was 2% (w/w) higher than the muscle enhancer without Spirulina. It was not observed increased in the nutritional content of muscle recovery when added Spirulina. However, it is known that Spirulina presents active compounds with important functions for the body. Thus, the composition of the foods satisfied the nutritional needs of athletes. Regarding the stability of developed foods, the shelf life was estimated between 9 and 11 months.

ABSTRACT Prosopis juliflora is a shrub that has been used to feed animals and humans. However, a synergistic action of piperidine alkaloids has been suggested to be responsible for neurotoxic damage observed in animals. We investigated the involvement of programmed cell death (PCD) and autophagy on the mechanism of cell death induced by a total extract (TAE) of alkaloids and fraction (F32) from P. juliflora leaves composed majoritary of juliprosopine in a model of neuron/glial cell co-culture. We saw that TAE (30 µg/mL) and F32 (7.5 µg/mL) induced reduction in ATP levels and changes in mitochondrial membrane potential at 12 h exposure. Moreover, TAE and F32 induced caspase-9 activation, nuclear condensation and neuronal death at 16 h exposure. After 4 h, they induced autophagy characterized by decreases of P62 protein level, increase of LC3II expression and increase in number of GFP-LC3 cells. Interestingly, we demonstrated that inhibition of autophagy by bafilomycin and vinblastine increased the cell death induced by TAE and autophagy induced by serum deprivation and rapamycin reduced cell death induced by F32 at 24 h. These results indicate that the mechanism neural cell death induced by these alkaloids involves PCD via caspase-9 activation and autophagy, which seems to be an important protective mechanism.

Abstract The multiplication of cells in all living organisms requires a tight regulation of DNA replication. Several mechanisms take place to ensure that the DNA is replicated faithfully and just once per cell cycle in order to originate through mitoses two new daughter cells that contain exactly the same information from the previous one. A key control mechanism that occurs before cells enter S phase is the formation of a pre-replication complex (pre-RC) that is assembled at replication origins by the sequential association of the origin recognition complex, followed by Cdt1, Cdc6 and finally MCMs, licensing DNA to start replication. The identification of pre-RC members in all animal and plant species shows that this complex is conserved in eukaryotes and, more importantly, the differences between kingdoms might reflect their divergence in strategies on cell cycle regulation, as it must be integrated and adapted to the niche, ecosystem, and the organism peculiarities. Here, we provide an overview of the knowledge generated so far on the formation and the developmental controls of the pre-RC mechanism in plants, analyzing some particular aspects in comparison to other eukaryotes.

Este estudo teve como objetivo avaliar os efeitos de diferentes tipos de controle de plantas infestantes sobre a mirmecofauna em um plantio inicial de Eucalyptus grandis. Para tanto, em março de 2011 foi realizado o início da implantação da cultura, em uma área localizada no município de Santa Maria - RS. Os seis tratamentos constituíram-se em controle químico total de plantas infestantes, na linha e na entrelinha de plantio, com glifosato (T1); controle químico total de plantas infestantes, na linha de plantio, com glifosato (T2); controle químico de monocotiledôneas na linha e entrelinha de plantio, com setoxidim (T3); controle químico de dicotiledôneas na linha e entrelinha de plantio, com bentazona (T4); controle químico total de plantas infestantes em faixa de um metro paralela à linha de plantio, com glifosato, e de um metro na parte central da entrelinha, sem controle (T5); e testemunha, sem controle de plantas infestantes (T6). O levantamento da mirmecofauna foi realizado no período de um ano utilizando-se três métodos de coleta: isca atrativa, armadilha de solo e funil de Berlese, com seis repetições por tratamento, em cada data de coleta. Nesse período foram coletadas 46.675 formigas, distribuídas em 37 espécies, não sendo verificada diferença significativa entre o total de espécimens coletados. Na área do tratamento constituído pelo controle de plantas infestantes somente na linha de plantio verificou-se eficiência amostral de 99,0% e S obs = 35. Entre os índices de Diversidade de Shannon (H'), destacou-se o valor encontrado para a área do tratamento T2 (H'= 1,34) em detrimento dos valores das áreas nas quais foram instalados os tratamentos T1 (H'= 1,25) e T5 (H'= 1,23). Havendo assim, uma maior coexistência de espécies de formigas entre as áreas com estrutura florística menos alterada e entre as áreas mais simplificadas. Não tendo sido verificada correlação significativa (r = 0,0463) entre a riqueza de espécies de formigas coletadas e o número de plantas infestantes encontradas ao final do experimento. Assim, conclui-se que os efeitos indiretos da ação dos herbicidas afetam mais a composição local de espécies de formigas do que sua riqueza.
The aim of this study was to evaluated the effects of different types of weed control on the ant fauna in an initial planting of Eucalyptus grandis. Therefore, in march of 2011 was performed the implantation of culture, in an area located in the city of Santa Maria - RS. The six treatments were constituted by the total chemical control of weeds in row and interrow planting, with glyphosate (T1); total chemical control of weeds in the crop row, with glyphosate (T2); chemical control of monocotyledonous in row and interrow planting, with sethoxydim (T3); chemical control of dicotyledonous line interrow and planting, with bentazone (T4); total chemical control of weeds in a yard track parallel to the row, with glyphosate, and a meter in the central part of interrow without control (T5); and without control weeds (T6). The survey of the ant fauna was conducted in one year using three collection methods: attractive bait, trap soil and Berlese funnel, with six replicates per treatment in each collection date. In this period were collected 46,675 ants, belonging to 37 species, there being no significant difference between the total specimens collected. In the area of treatment consists of the control of weeds in the crop row only verified efficiency of 99.0% and sample S obs = 35. Among the indices of Shannon Diversity (H'), highlighted the value found for the area of treatment T2 (H'= 1.34) at the expense of the values of the areas where they were installed the T1 (H'= 1,25) and T5 (H'= 1,23). Having thus a higher co-existence of ant species between the areas with changed structure less flora, and between the areas most simplified. There were significant correlation (r = 0.0463) between the species richness of ants collected and the number of weeds found at the end of the experiment. Thus, we conclude that the indirect effects of the herbicides affect more the local composition ant species than his richness.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Aedes aegypti (L.), the main vector of dengue fever in Brazil, has been controlled with the use of massive chemical products, contributing to the development of resistance and decreasing the insect control efficiency. The control of dipterans with bioinsecticides based on Bacillus thuringiensis has been satisfactory, due to the production of insecticidal proteins denominated Cry (crystal), Cyt (cytolytic) toxins and Chi (chitinase), and to the synergistic effects among them. The present work aimed to select B. thuringiensis isolates efficient against A. aegypti larvae. A bacterial collection containing 1,073 isolates of B. thuringiensis, obtained from different locations of Brazilian territory, had the DNA isolated and submitted to PCR amplifications using specific primers for cry4Aa, cry4Ba, cry11Aa, cry11Ba, cyt1Aa, cyt1Ab, cyt2Aa and chi genes. For the LC50 and LC90 determination, the entomopathogenic isolates were evaluated by selective and quantitative bioassays. Only 45 isolates (4.2%) presented amplicons for the cry and cyt genes. The chi gene sequence was detected in 25 (54.3%) of those isolates. From the 45 isolates submitted to the selective bioassays, 13 caused 100% mortality of A. aegypti larvae. The identification of cry, cyt and chi genes of B. thuringiensis and the toxicity analysis on A. aegypti led to the selection of a set of isolates that have the potential to be used in the formulation of new bioinsecticides.

Avaliação acústica e perceptivo-auditiva da voz nos momentos pré e pós-operatório da cirurgia de implante de pré-fáscia do músculo temporal

OBJETIVO: analisar os parâmetros vocais acústicos e perceptivo-auditivos que se alteram com a cirurgia de implante de pré-fáscia do músculo temporal para sulco vocal estria maior. MÉTODOS: registraram-se as emissões vocais de 9 pacientes, 6 do sexo feminino e 3 do masculino, nas condições pré e pós-tratamento cirúrgico. As amostras vocais foram submetidas à avaliação perceptivo-auditiva por três fonoaudiólogas com experiência na área de voz, por meio de consenso utilizando a escala GRBAS e a análise acústica espectrográfica foi realizada por meio de consenso entre três estudantes do curso de Fonoaudiologia após treinamento visual com a duração de três horas, utilizando o programa GRAM 5.1. RESULTADOS: houve tendência de a freqüência fundamental apresentar-se mais grave e o tempo máximo de fonação aumentou. Por meio da análise estatística observou-se que somente o parâmetro B apresentou melhora estatisticamente significante (t>1,734), os outros parâmetros G, R e S não mostraram melhora estatisticamente significante. Na análise espectrográfica observou-se que a forma e a continuidade do traçado permaneceram estáveis, a presença de ruído diminuiu e número de harmônicos aumentou. CONCLUSÃO: os pacientes submetidos à cirurgia de implante de Pré-Fáscia do Músculo Temporal apresentaram melhora no aspecto perceptivo-auditivo de soprosidade (B), na freqüência fundamental, tempo máximo de fonação e nos aspectos espectrográficos de presença de ruído e aumento do número de harmônicos, houve também tendência da freqüência fundamental apresentar-se mais grave após a cirurgia.
PURPOSE: to analyze the impact of pre-fascia temporal muscle implantation in vocal fold for sulcus vocalis, using perceptual ratings, acoustical analysis and spectrograms analysis. METHODS: it is a retrospective study. We reviewed the records of the nine patients who underwent pre-fascia temporal muscle implantation for sulcus vocalis. Perceptual voice ratings [Grade, Roughness, Breathiness, Asthenia, and Strain (GRBAS)], acoustical analysis and spectrogram analysis were analyzed. Perceptual voice ratings was carried out by three speech-language pathologists and spectrograms analysis by three speech language pathology students after three hours of training. RESULTS: perceptual ratings (GRBAS): Statistically significant improvement was found just in parameter B (Breathiness) t>1.734. Analysis acoustical: The fundamental frequency decreased and the maximum phonation time increased after surgery. Spectrogram analysis: Harmonic form and periodicity remained without improvement after treatment. The noise of spectrogram decreased and harmonic definition increased after treatment. CONCLUSION: the patients who underwent pre-fascia temporal muscle implantation for sulcus vocalis showed improvement in perceptual rating of Breathiness (B), fundamental frequency, maximum phonation time and spectrograms analysis (noise decreased and harmonic definition increased).

O efeito do ácido S,S-etilenodiaminodi-succínico (edds) na supressão da oxidação do ácido ascórbico catalisada por metais (Mn, Fe, Co, Ni, Cu, Zn) foi testado in vitro através da oxidação da sonda fluorescente cloreto de 1,2,3-diidrorodamina. A atividade pró-oxidante do ferro não foi totalmente suprimida, mesmo sob excesso molar de quatro vezes do ligante. O efeito do meio de cultura na toxicidade dos complexos para células mononucleares do sangue periférico (PBMC) e da linhagem K562 foi estudado. A citotoxicidade de Fe-edds foi abolida na presença de Trolox ou de proteínas do soro. Os prováveis mecanismos de toxicidade celular foram investigados através do bloqueio de transportadores de monocarboxilatos (MCT) e estudos de ciclo celular por citometria de fluxo. Células tratadas com os complexos metálicos e com o ácido alfa-ciano-4-hidroxicinâmico, um conhecido bloqueador de MCT, mostraram recuperação de viabilidade, sugerindo que esses transportadores possam estar envolvidos na internalização dos complexos metal-edds. O ácido livre promoveu a parada do ciclo celular na fase G0/G1 (PBMC) ou S (K562), sugerindo dano direto ao DNA ou interferência na sua duplicação.
The effect of S,S-ethylenediaminedisuccinic acid (edds) on the quenching of metal-catalyzed (metal = Mn, Fe, Co, Ni, Cu, Zn) oxidation of ascorbic acid was tested in vitro via oxidation of the fluorescent probe 1,2,3-dihydrorhodamine dihydrochloride. The pro-oxidant activity of iron was not fully suppressed, even at a four-fold molar excess of the ligand. The effect of serum on the toxicity to peripheral blood mononuclear cells (PBMC) and K562 cells was investigated. The cytotoxic effect of Fe-edds was abrogated in the presence of Trolox or serum proteins. The probable pathways of cell toxicity were investigated through blocking of the monocarboxylate transporters (MCT) in association with cell cycle studies by flow cytometry. Cells treated with metal complexes and alpha-cyano-4-hydroxycinnamic acid, a known MCT inhibitor, showed recovery of viability, suggesting that MCT proteins may be involved in the internalization of metal-edds complexes. The free acid induced cell cycle arrest in G0/G1 (PBMC) and S (K562) phases, suggesting direct DNA damage or interference in DNA replication.

Diagnóstico do cultivo hidropônico de hortaliças na região do Distrito Federal

De outubro de 1997 a maio de 1998, foram realizadas entrevistas com produtores de hortaliças em cultivo hidropônico, na região do Distrito Federal, com o objetivo de verificar a situação atual, bem como identificar os pontos de estrangulamento no sistema de produção. Foram identificados 18 produtores em um raio de 50 km de Brasília. As principais culturas são alface e tomate, ocupando uma área de 2,7 ha e de 0,5 ha, respectivamente. Foi verificado que as estruturas são, na sua maioria, de madeira e arcos de ferro, com custo médio de R$ 26,00/m², para produção de alface e de R$ 20,00/m², para tomate. O custo por unidade de alface está em torno de R$ 0,25 e o preço médio de venda ao mercado de R$ 0,45, enquanto que o tomate está sendo vendido a R$ 1,20/kg. Devido à produção recente, os produtores de tomate não conseguiram ainda calcular os custos. As produtividades médias observadas foram de 300 t/ha/ano de alface e de 104 t/ha/ciclo de tomate. Os principais compradores são Ceasa, supermercados, restaurantes, sacolões e mercearias. Um dos produtores exporta alface para Manaus (AM). Metade dos produtores não usa agrotóxicos. Assistência técnica, ocorrência de "tipburn", análise da solução nutritiva, aquecimento interno da estrutura, doenças e pragas são os principais problemas citados pelos produtores de alface e, na produção de tomate, frutos rachados, doenças e pragas. O cultivo hidropônico de hortaliças está sendo visto como uma alternativa para viabilização do agronegócio na região, além de permitir uma sensível redução na utilização de agrotóxicos. Aproximadamente 44% dos produtores estão sem assistência técnica e produzindo por meio de tentativas, demandando uma atuação mais efetiva dos órgãos de ensino, pesquisa e extensão rural da região.
From October 1997 to May 1998, several interviews with producers of hydroponic crops took place in Distrito Federal with the objective of not only verifying but also identifying the present situation regarding the constraint points in the production system. Eighteen producers were identified in a ratio of 50km away from Brasília. The main crops are lettuce and tomato, which occupy an area of 2,7 ha and 0,5 ha, respectively. It was also verified that the majority of the structures used was made of wood and steel, which account for an average cost of R$ 26,00/m² for lettuce production, and of R$ 20,00/m² for tomato production. The unit cost for lettuce is around R$ 0,25 and the average market price is R$ 0,45, whereas the tomato is sold for R$ 1,20/kg. Due to the recent production, tomato producers haven't yet figured out the real costs. The average productivity observed was of 300t/ha/year for lettuce, and of 104t/ha/cycle for tomato. The main buyers are Ceasa, supermarkets, restaurants, vegetable stores, and green groceries stores. One producer exports lettuce to Manaus (AM). Half of the producers do not use chemicals. The main problems faced by lettuce producers are the lack of technical assistance, "tipburn", analysis of the nutrient solution, inward structure heating, diseases and pests. On the other hand, tomato producers face cracked fruits, as well as diseases and pests. Vegetables hydroponic cropping is seen as an alternative for the feasibility of the agribusiness of the region, besides allowing a substantial reduction of chemicals utilization. Approximately 44% of the producers have no technical assistance and are also operating through attempts, which demands a more effective support by universities, research institutes and extension services.
