Oxidative stress is involved in LLLT mechanism of action on skin healing in rats

The skin injury healing process involves the main phases of homoeostasis, inflammation, proliferation, and remodeling. The present study aimed to analyze the effects of low-level laser therapy (LLLT) on hematological dynamics, oxidative stress markers, and its relation with tissue healing following skin injury. Wistar rats were divided into control, sham, skin injury, and skin injury LLLT. The biochemical and morphological analyses were performed in the inflammatory (1 and 3 days) and regenerative phases (7, 14, and 21 days) following injury. The skin injury was performed in the dorsal region, between the intrascapular lines, using a surgical punch. LLLT (Al-Ga-In-P, λ=660 nm, energy density of 20 J/cm2, 30 mW power, and a time of 40 s) was applied at the area immediately after injury and on every following day according to the experimental subgroups. LLLT maintained hematocrit and hemoglobin levels until the 3rd day of treatment. Surprisingly, LLLT increased total leukocytes levels compared to control until the 3rd day. The effects of LLLT on mitochondrial activity were demonstrated by the significant increase in MTT levels in both inflammatory and regenerative phases (from the 1st to the 7th day), but only when associated with skin injury. The results indicated that LLLT modulated the inflammatory response intensity and accelerated skin tissue healing by a mechanism that involved oxidative damage reduction mostly at early stages of skin healing (inflammatory phase).

Abstract Biological invasion is a major threat to global biodiversity and ecosystem services. We examined the functional traits similarity between a set of native and non-native invasive tree species from the Southern Brazilian subtropical mixed forest, part of an important global hotspot for biodiversity conservation. We hypothesized that invasive species occupy marginal niche spaces. We ordered the species using the Principal Component Analysis based on their wood density, leaf area, and specific leaf area. These are all important traits that summarize essential ecological strategies associated with resource acquisition and conservation. Functional overlaps between non-native and native species were analyzed through kernel density estimation of continuous traits data. While native and non-native invasive species were distributed along the same functional gradients, the position of non-native species in the functional space is species and traits specific. We concluded that within Brazilian subtropical mixed forests, the functional dissimilarity as a key factor in invasion success could not be generalized for all species and traits.

Guarana (Paullinia cupana) is habitually ingested by people in the Amazon region and is a key ingredient in various energy drinks consumed worldwide. Extension in longevity and low prevalence of chronic age-related diseases have been associated to habitual intake of guarana. Anti-aging potential of guarana was also demonstrated in Caenorhabditis elegans; however, the mechanisms involved in its effects are not clear. Herein, we investigated the putative pathways that regulate the effects of guarana ethanolic extract (GEE) on lifespan using C. elegans. The major known longevity pathways were analyzed through mutant worms and RT-qPCR assay (DAF-2, DAF-16, SKN-1, SIR-2.1, HSF-1). The possible involvement of purinergic signaling was also investigated. This study demonstrated that GEE acts through antioxidant activity, DAF-16, HSF-1, and SKN-1 pathways, and human adenosine receptor ortholog (ADOR-1) to extend lifespan. GEE also downregulated skn-1, daf-16, sir-2.1 and hsp-16.2 in 9-day-old C. elegans, which might reflect less need to activate these protective genes due to direct antioxidant effects. Our results contribute to the comprehension of guarana effects in vivo, which might be helpful to prevent or treat aging-associated disorders, and also suggest purinergic signaling as a plausible therapeutic target for longevity studies.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Produção e composição bromatológica do sorgo (Sorghum bicolor) cultivado sob doses de nitrogênio

This study evaluated the productive response and chemical composition of sorghum hybrid BR 601 to nitrogen fertilization. The experimental design was randomized blocks with five replications. The treatments were: 0, 50, 100, 150 and 200 kg N/ha (ammonium sulfate). There was a quadratic increase of yield with N fertilization, the higher value was obtained with 143.19 kg of N/ha for fresh matter (37996 kg/ha) and with 155.67 kg of N/ha for dry matter (16 275 kg/ha). The production of leaf and dead material at the plant did not differ between treatments. The percentage of stem in the plant decreased quadratically up to a dose of 100 kg/ha and the panicle component increased quadratically to 30,96%. So, this component was the main responsible for increase in DM yield. The average plant dry matter varied from 34.84% to 38.98% in the treatment with 200 kg N/ha. The levels of MM decreased (6.41 to 5.49%) and crude protein increased linearly (from 4.99% to 6.23%) with increasing doses of N. The levels of neutral detergent fiber, acid detergent fiber and hemicellulose did not differ in relation to nitrogen fertilization. The nitrogen increases the production of dry matter and panicle without changing the fiber constituents of the plant.
Objetivou-se avaliar a resposta produtiva e a composição bromatológica do sorgo híbrido BR 601 ao incremento da adubação nitrogenada. O delineamento experimental utilizado foi em blocos casualizados com cinco repetições. Os trata-mentos foram: 0, 50, 100, 150 e 200 kg/ha de N (sulfato de amônio). A produção variou de forma quadrática ao incremento da adubação nitrogenada, o ponto de máxima para a materia verde foi obtido com 143,19 kg de N/ha (37 996 kg/ha) e para matéria seca com 155,67 kg de N/ha (16 275 kg/ha). A produção de lâmina foliar e material morto na planta não diferiu entre os tratamentos. O percentual de colmo na planta diminuiu de forma quadrática até a dose de 100 kg/ha, o componente panícula aumentou de forma quadrática em 30,96%, o que explica que este componente foi o principal responsável pelo aumento na PMS. O percentual médio de matéria seca da planta variou de 34,84% no tratamento controle a 38,98% no tratamento com 200 kg de N/ha. Os teores de MM diminuíram (6,41 a 5,49%). O teor de PB incrementara linearmente com o aumento nas doses de adubo aplicadas, com variação no teor protéico de 4,99% a 6,23%, respectivamente nos tratamentos 0 e 200 kg de N/ha. Os teores de fibra em detergente neutro, fibra em detergente ácido e hemicelulose não diferiram em função da adubação nitrogenada. A adubação nitrogenada aumenta a produção de matéria seca e panícula sem alterar os constituintes fibrosos da planta.

Aimed to evaluate the morphogenetic and structural characteristics of the pasture of Brachiaria decumbens, subjected to several intensities and frequencies of cutting and nitrogen fertilization. Experimental design was randomized blocks in split plots, whit a 2 x 2 factorial arrangement relating to combinations of two cutting heights (40 and 50 cm) and two heights of residue (15 and 25 cm) at the plots and nitrogen fertilization (with or without nitrogen) in sub-plots, with four replications. Mophogenetic variables evaluated were: leaf elongation rate (LER), stem elongation rate (SER), leaf appearance rate (LAR), leaf senescence rate (LSR) and phyllochron; and structural features were: number of basal tillers (NBT), total number of leaves (TNL), number of leaves appeared (NLAp), number of dead leaves (NDL), number of alive leaves (NAL) and final leaf length (FLL). 100 kg N/ha allowed increase (p<0.10) on LER of 31.58% and 126.32% respectively for residues 15 and 25 cm. The NBT compared to cutting heights (40 to 50 cm) were affected (p<0.10) only with the use of nitrogen fertilizer, with average of 145.62 tillers/m² in 50 cm cut height. Residue height of 25 cm combined with cut height 40 cm increase leaf elongation rate and number of basal tillers, reducing expansion time, without changing other structural characteristics of the pasture.
Objetivou-se avaliar as características morfogênicas e estruturais do pasto de Brachiaria decumbens, submetido a intensidades, frequências de corte e adubação nitrogenada. O delineamento experimental foi em blocos casualizados em esquema de parcelas subdivididas, tendo na parcela o arranjo fatorial 2 x 2 referente a combinações entre duas alturas de corte (40 e 50 cm) e duas alturas de resíduo (15 e 25 cm) e nas sub-parcelas a adubação nitrogenada (com ou sem nitrogênio), com quatro repetições. As variáveis analisadas para as características morfogênicas foram: taxa de alongamento foliar (TAlF); taxa de alongamento de colmo (TAlC); taxa de aparecimento foliar (TApF); taxa de senescência foliar (TSF) e filocrono; e para as características estruturais foram: número de perfilhos basais (NPB); número de folhas totais (NFT); número de folhas aparecidas (NFAp); número de folhas mortas (NFM); número de folhas vivas (NFV) e comprimento final da folha (CFF). A utilização de 100 kg/ha de N permitiu aumento (p<0,10) na TAlF de 31,58% e 126,32%, respectivamente para os resíduos de 15 e 25 cm. O NPB em comparação às alturas de corte (40 e 50 cm) foi afetado (p<0,10) apenas com a utilização da adubação nitrogenada, observando-se média de 145,62 perfilhos/m² para a altura de corte 50 cm. A altura de resíduo 25 cm combinada com a altura de corte 40 cm, junto à aplicação de 100 kg/ha de N, promovem maior produção de matéria seca total em menor espaço de tempo (28 dias), permitindo maiores taxas de lotação e facilitando o manejo no sistema de lotação rotacionada.

The objective of the present study was to describe motor behavioral changes in association with histopathological and hematological findings in Wistar rats inoculated intravenously with human T-cell lymphotropic virus type 1 (HTLV-1)-infected MT2 cells. Twenty-five 4-month-old male rats were inoculated with HTLV-1-infected MT2 cells and 13 control rats were inoculated with normal human lymphocytes. The behavior of the rats was observed before and 5, 10, 15, and 20 months after inoculation during a 30-min/rat testing time for 5 consecutive days. During each of 4 periods, a subset of rats was randomly chosen to be sacrificed in order to harvest the spinal cord for histopathological analysis and to obtain blood for serological and molecular studies. Behavioral analyses of the HTLV-1-inoculated rats showed a significant decrease of climbing, walking and freezing, and an increase of scratching, sniffing, biting, licking, and resting/sleeping. Two of the 25 HTLV-1-inoculated rats (8%) developed spastic paraparesis as a major behavioral change. The histopathological changes were few and mild, but in some cases there was diffuse lymphocyte infiltration. The minor and major behavioral changes occurred after 10-20 months of evolution. The long-term observation of Wistar rats inoculated with HTLV-1-infected MT2 cells showed major (spastic paraparesis) and minor motor abnormalities in association with the degree of HTLV-1-induced myelopathy.

Epidemiological aspects of astrovirus and coronavirus in poults in the South Eastern Region of Brazil

O presente estudo foi conduzido para avaliar a prevalência do Coronavirus dos perus (TCoV) e Astrovirus tipo 2 (TAstV-2) entre os meses de Fevereiro a Dezembro de 2006, em uma região produtora localizada no semi-árido a Sudeste do Brasil. Os principais fatores de risco associado a prevalência foram material clínico analisado, condições climáticas e tipo de técnica molecular empregada. Os sinais clínicos foram caracterizados como intenso fluido intestinal e baixo crescimento em aves jovens, sendo o material coletado swabs cloacais, fezes, soros, bursa de Fabrícius, segmentos do intestino delgado, timo e baço. Os dados meteorológicos (índice pluviométrico e umidade relativa) desta região, durante o período de estudo, foram de temperatura média mensal variando de 39.3 a 31.2ºC, precipitação na época chuvosa variando de 40 a 270.3mm/mês e ausência de chuva na estação fria e seca. A técnica de simplex RT-PCR resultou em valores de odds ratio (OR) que sugerem que a região do intestino delgado (junção íleo-cecal) possui alta chance (1.9 vezes) de gerar resultados positivos na amplificação de RNA viral que as fezes (1.5 vezes) analisadas. A técnica de multiplex RT-PCR demonstrou ser 3.98 vezes mais eficiente em promover resultados positivos nas fezes que nos swabs cloacais, durante a época de inverno. Os maiores fatores de risco encontrados foram baixa umidade relativa associada a altas temperaturas, durante a estação seca, o que pode permitir uma maior disseminação aérea do ambos os vírus entre os lotes estudados. A alta prevalência detectada para dois vírus sugerem que, no Brasil, estes representam os maiores responsáveis pelos surtos de enterite viral nas regiões semi-áridas, associado a baixas umidades e altas temperaturas típicas de países tropicais.
A survey of Turkey Coronavirus (TCoV) and Astrovirus (TAstV-2) prevalence was carried out from February to December during 2006 year in semiarid region of Brazil, from a turkey producer area, localized in South Eastern of Brazil. To asses the risk factor related to clinical material, climatic condition and type of RT-PCR applied, cloacal swabs (CS), faeces, sera, bursa of Fabricius (BF), thymus (TH) and spleen (SP) and ileum-caeca region were collected from 30-day-old poults suffering of enteritis episode characterized as poult enteritis mortality syndrome (PEMS). The PEMS clinical features were characterized by watery to foamy faeces, light brown-yellow in colour and low mortality rate. Meteorological data (rainfall and relative humidity) observed during along the study presented monthly average temperature ranging from 39.3 and 31.2ºC, precipitation in rainy season from 40 to 270.3 mm/month, and no rain during dry season. Simplex RT-PCR gave odds ratio (OR) values suggesting that ileum-caeca region is at higher chance (OR=1.9; p=0.9741) to have both viral RNA than faeces (OR=1.5; p=0.7319). However, multiplex RT-PCR showed 3.98 (p=0.89982) more chance to give positive results in faeces than CS at dry season. The major risk factors seem to be low rate of humidity and high temperatures at winter, probably responsible for spread, easily, the TCoV and TAstv-2 among the flocks. The positive results of both virus suggested that they can play an important role in enteric disorders, associated to low humidity and high temperatures frequently found in tropical countries.
