ABSTRACT The burnout variable has been studied in different professions; however, it presents challenges in its measurement. The present study provides evidence of validity based on the internal structure of the Burnout Syndrome Assessment Scale in Public Security Workers (EASB-SP) and presents the estimation of item parameters through the Item Response Theory (IRT). A total of 10,885 public security agents from all states of Brazil participated in the study. The analyses indicated a three-factor structure, with adequate reliability indices and discrimination of the items. The Professional Fulfillment factor presented a lower theta value to be endorsed. The instrument was shown to be an adequate measure, with evidence of validity for the context of Brazilian public security.

Abstract Cucurbita pepo type zucchini is an important vegetable in the Bajío region; however, its production is affected by the reduction of the populations of bee pollinators and the effect of the environmental alterations on its reproductive habits. Therefore, it is fundamental to identify parthenocarpic genotypes capable of generating fruits without any fecundation or loss of their agronomic traits. The aim of this study was to identify parthenocarpic genotypes of Cucurbita pepo type Grey Zucchini. The evaluated genotypes derived from the cross of six parent lines and two consecutive cycles of self-pollination. The genotypes were grown under controlled conditions, identifying 16 parthenocarpic genotypes. Genotypes GZS193, GZS194, GZS301, GZS148, and GZS171 presented most of the desirable traits; hence, they could be used as parents in future crossing assays to accumulate all the traits in a single genotype and keep the parthenocarpy.

This study aimed to develop a reliable stability-indicating method (SIM) for amoxicillin 500 mg capsules (DP-drug product). A literature review addressing amoxicillin (AMX; DS-drug substance) forced degradation studies and the existing SIMs was conducted to verify the most significant outcomes. Subsequently, the forced degradation of DP and DS was carried out following the Brazilian Health Surveillance Agency (ANVISA) guidelines, including thermal degradation (dry and wet heat), acidic and alkaline hydrolyses, hydrogen peroxide oxidation, reaction with copper, and photodegradation. Both DS and DP were more susceptible to 0.015 M NaOH, resulting in approximately 50% degradation. AMX DS and DP were not significantly photodegraded, but some degradation products (PDegs) showed susceptibility to light exposure. Thermodegraded samples showing ≥ 10% degradation exhibited modified profiles in thermogravimetric (TG) and differential scanning calorimetric (DSC) analyses. The X-ray powder diffraction patterns (XRPD) of DS samples exposed to dry and wet heat displayed complete amorphization of AMX, attesting to the occurrence of physical degradation concomitantly with chemical degradation, which can alter the drug’s bioavailability. In contrast, the thermodegraded DP samples exhibited intact AMX crystals interspersed with the amorphous form, perhaps partly protected by the excipient. The validated SIM was able to detect and quantify about 80 PDegs.

ABSTRACT The objective of this study was to characterize the thermal environment and evaluate the behavior of finishing pigs housed in deep bedding and conventional systems. The work was carried out in the Department of Animal Science of the Federal Institute of Education, Science and Technology of Southeast Minas Gerais, Rio Pomba campus. Three pens were used in an installation for breeding pigs in their finishing phase. Two pens contained deep bedding, with wood shavings and rice husks. The remaining pen was a conventional system. The behavior of the piglets was observed from 8 a.m. to 5:30 p.m., noting the behaviors with the use of an ethogram, while data relating to the thermal environment in the stalls were automatically collected using data loggers. Thermal environment data for all systems presented thermal stress conditions. Regarding behavioral variables, animals in conventional systems had a higher frequency of visits to feeders, while animals in the deep bedding system were more active and visited the drinking fountains more frequently. Despite the higher level of activity of the animals in the deep bedding system, it is not possible to confirm that the deep bedding provides a better degree of well-being for the animals under thermal stress conditions.

ABSTRACT The long-term outcome of acute lymphoblastic leukemia has improved dramatically due to the development of more effective treatment strategies. L-asparaginase (ASNase) is one of the main drugs used and causes death of leukemic cells by systematically depleting the non-essential amino acid asparagine. Three main types of ASNase have been used so far: native ASNase derived from Escherichia coli, an enzyme isolated from Erwinia chrysanthemi and a pegylated form of the native E. coli ASNase, the ASNase PEG. Hypersensitivity reactions are the main complication related to this drug. Although clinical allergies may be important, a major concern is that antibodies produced in response to ASNase may cause rapid inactivation of ASNase, leading to a worse prognosis. This reaction is commonly referred to as "silent hypersensitivity" or "silent inactivation". We are able to analyze hypersensitivity and inactivation processes by the measurement of the ASNase activity. The ability to individualize the ASNase therapy in patients, adjusting the dose or switching patients with silent inactivation to an alternate ASNase preparation may help improve outcomes in those patients. This review article aims to describe the pathophysiology of the inactivation process, how to diagnose it and finally how to manage it.

Abstract Leishmaniasis is caused by protozoan parasites belonging to the genus Leishmania and includes cutaneous, mucocutaneous and visceral clinical forms. Drugs currently available for leishmaniasis treatment present high toxicity, and development of parasite resistance. Plants constitute an important source of compounds with leishmanicidal potential. This study aimed to evaluate the anti-Leishmania amazonensis activity of the terpenoid fraction of Eugenia pruniformis leaves (TF-EpL). TF-EpL was active against the promastigote and intracellular amastigote forms of L. amazonensis with IC50(24h) value of 43.60μg/mL and 44.77μg/mL, respectively. TF-EpL altered the cell cycle of the parasite, increasing 2.32-fold the cells in the Sub-G0/G1 phase. TF-EpL also changed the ΔΨm and increased ROS and the number of annexin-V-PI positive promastigotes, which suggests incidental death. β-sitosterol, ursolic acid, corosolic acid and asiatic acid were isolated from TF-EpL. The results showed the antileishmanial activity of E. pruniformis terpenoids and its potential for further studies as a source of new drugs for leishmaniasis.

Abstract Aim: Our purpose was to determine whether Transcranial Direct Current Stimulation (tDCS) improves performance in untrained individuals for supramaximal intermittent exercise. Methods: In a cross-over design, 11 healthy male subjects (26.8 ± 4.6 years) performed four Wingate trials after 20 minutes of anodal or sham tDCS over the left Insular Cortex (IC). For performance indexes, Relative Peak Power (RPP), Relative Average Power (RAP) and Fatigue Index (FI) were computed. Also, a Rating of Perceived Exertion (RPE) and Electromyography (EMG) signal were used to assess central and muscle fatigue development. Results: There was a significant difference over trials on all performance indexes, but there were no significant condition x trial interactions for any of the indexes. RPE increased significantly over trials, but there was no condition x trial interaction. There was no significant difference over trials on EMG for the rectus femoris and vastus medialis muscles; however, EMG decreased over trials for the vastus lateralis muscle. Furthermore, there was no condition x trial interaction on the EMG signal for any of the muscles. Conclusion: Our findings suggest that the anodal tDCS technique has no impact on physical performance, perceived exertion nor muscle fatigue development for supramaximal intermittent exercise.

ABSTRACT In wetlands, the knowledge accumulated on the role of aquatic plants in the methane cycle focused on emergent macrophytes, to the detriment of other typologies. Herein, we evaluated whether the free-floating macrophyte Salvinia auriculata Aubl. and the floating-leaved macrophyte Eichhornia azurea (Sw.) Kunth. decrease the water column methane concentrations compared to a plant-free surface. We prepared microcosms by inserting an individual of S. auriculata or of E. azurea into chambers filled with lagoon water previously bubbled with CH4. Another set of chambers was incubated only with the prepared water, representing the plant-free surface. Half of the chambers were kept in the dark and half in sunlight to simulate a diel cycle. We observed greater loss of CH4, higher O2 uptake and lower CO2 outflow in the plants treatments. The decrease in methane concentrations in the E. azurea treatments was 93.5% in the light and 77.2% in the dark. In the S. auriculata treatments, the decreases were 74.2% and 67.4% in the light and in the dark, respectively. In plant-free surface the decrease was 58.7% in the light and 36.3% in the dark. These results indicate a role of floating aquatic macrophytes in the methane cycle in the water column. Moreover, our results suggest a diel variation of methane oxidation and methane emission, according to the differences observed in O2 uptake and CO2 outflow between dark and light conditions. Thus, future predictions of global methane budget should include the role played by floating aquatic macrophytes.

Abstract: The aim was to evaluate the use of oregano extract in Japanese quail (Coturnix coturnix japonica) diet, challenged with Escherichia coli strains, on performance, incidence of avian cellulitis and of antibody specific antigens against E. coli. Three hundred sixty Japanese quails with 90 days of age were distributed into galvanized wire cages in a conventional shed. The experimental design was completely randomized in factorial 5x2 design (oregano extract x challenged or not with E. coli), totaling ten treatments with six replicates of six birds per cage. Oregano extract levels were 0.00, 0.025, 0.050, 0.100 and 0.150%. Performance productive characteristics were evaluated, macroscopic lesions of cellulitis were measured after post-inoculation of the strains, and serum samples were collected for antibodies during experiment. The data were subjected to analysis of variance and averages compared by T test. Effect of E. coli was observed on all productive characteristics, regardless of the EO level evaluated, where challenged groups showed worse performance results. The macroscopic lesions, characteristic of cellulitis, observed only in birds of groups challenged with E. coli, were classified as mild and without bleeding. For specific antibodies, there was a higher number of birds challenged with E. coli strains in relation to unchallenged birds. It can be concluded that oregano extract supplemented in the diet was not effective against the challenge with E. coli in laying quails, and challenged birds with E. coli showed higher humoral and cellular immune response, compared with unchallenged birds, characterized by increased antibody titer and pectoral macroscopic lesion, regardless of the oregano extract levels evaluated.

Abstract Larvae and adults of certain species of predator lady beetles feed on pollen, guaranteeing their survival, and at times, reproduction in the absence of preferred prey. Palynology, therefore, may contribute in the investigation of botanical families visited by these predators in order to obtain this floral resource. There are records of the visitation of Apiaceae flowers by Coleomegilla maculata DeGeer, 1775 (Coleoptera, Coccinellidae), but not the ingestion of their pollen grains by this lady beetle. The external morphology of pollen grains of three Apiaceae aromatic species (Anethum graveolens L., Coriandrum sativum L., Foeniculum vulgare Mill.) was characterized, and it was evaluated the ingestion of these pollens by fourth instar larvae and adults of C. maculata upon confinement along with flowers of these Apiaceae for 24 and 48 hours. The pollen grains of those species presented similar external morphology. In the two times of exposure, the larvae ingested the same amount of pollen from the three Apiaceae species, and the amount of C. sativum pollen ingested was the same between larvae and adults. The amount of A. graveolens pollen grains ingested by the adults was significantly greater than the pollens of C. sativum and F. vulgare, in 24 hours, with the opposite occurring in 48 hours. In the first 24 hours, the adults ingested more A. graveolens pollen than the larvae, with the opposite occurring with F. vulgare. There was no significant difference in the amount of Apiaceae pollen ingested between larvae and adults in 48 hours. The results suggest that the pollen-eating habits of certain aphidophagous lady beetles may be crucial in their preservation within agro-ecosystems.

The Drake Passage (DP) links the Southern Pacific and Atlantic Oceans, and acts as a physical narrowing for the largest current of the World Ocean, the Antarctic Circumpolar Current (ACC). This result in a constriction of the main circumpolar fronts related to the ACC: the Subantarctic Front, the Polar Front, the Southern Antarctic Circumpolar Current Front and the Continental Water Boundary Front. Previous studies showed that the DP is a high-nutrient low-chlorophyll (HNLC) region, but studies on the interannual and seasonal macronutrients' fluctuations in relation with chlorophyll and frontal areas in the DP are scarce. This study analyzes the spatial and temporal trends of nitrate, phosphate, and silicate on surface samples from 113 oceanographic stations in the DP during summer and early fall of 2001 to 2004, in relation with the different fronts and chlorophyll-a concentration. The results revealed an increase of nutrients towards the south. Nitrate showed values from 3 µM at 55° S to 27 µM at 63° S, whereas values from 0.3 µM at 55° S to 2.6 µM at 63° S were observed for phosphate. Silicate increased considerably south of Polar Front, from 0 µM at 55° S to 74 µM at 62° S. The nutrients concentration was significantly higher in summer than in early fall and showed a trend to decrease over the studied years. A complex sum of biogeochemical processes would generate the main seasonal trends, while atmospheric-oceanic processes such as Southern Annular Mode and El Niño Southern Oscillation, would be responsible of observed nutrients interannual variability.

Abstract The objective of this study was to verify the effects of ethanol consumption and alcohol detoxification on the biomechanics, area and thickness of cortical and trabecular bone in rat femur. This was an experimental study in which 18 male Wistar rats were used, with 40 days of age, weighing 179±2.5 g. The rats were divided into three groups (n=06): CT (control), AC (chronic alcoholic), DT (detoxification). After experimental procedures, the animals were euthanized by an overdose of the anesthetic and their femurs were collected for mechanical testing and histological processing. All animals did not present malnutrition or dehydration during experimentation period. Morphometric analysis of cortical and trabecular bones in rat femurs demonstrated that AC animals showed inferior dimensions and alcohol detoxification (DT) allowed an enhancement in area and thickness of cortical and trabecular bone. Material and structural properties data of AC group highlighted the harmful effects of ethanol on bone mechanical properties. The results of this study demonstrated that chronic alcoholic rats (AC) presented major bone damage in all analyzed variables. Those findings suggested that alcohol detoxification is highly suggested in pre-operative planning and this corroborates to the success of bone surgery and bone tissue repair. Thanks to the financial support offered by PROBIC – UNIFENAS.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The objective of this study was to evaluate the efficiency of two doses of PGF associated or not to hCG on the associated reproductive parameters in dairy goats. A total of 29 goats received two doses of 30µg d-cloprostenol latero-vulvar at a 10 day intervals (Day 1 and Day 10). The does were allocated according to body weight and body condition score into two treatments, to receive hCG (250IU) or saline at estrus onset. After the second dose of PGF, estrus was monitored and ultrasound exams were performed twice daily. All does were inseminated 16h after estrus onset. Blood collection was performed every day for progesterone assay. The use of hCG at estrus onset did not affect any studied parameter and therefore the data were pooled. Estrous response rate was similar (P>0.05) after the first (75.9%, 22/29) and the second dose of PGF (79.3%, 23/29). The interval between the administration of PGF and estrus onset was greater (P<0.05) after Day 1 (75.8±53.9h) than Day 10 (47.7±10.1 h). Estrus duration was superior (P < 0.05) after Day 1 (35.4±15.9h) to Day 10 (26.8±15.0h). Ovulation rate was 79.3% (23/29) after the second dose of PGF. No differences (P>0.05) between both experimental groups were detected in the following parameters, averaging: the interval from the second dose administration to the ovulation (86.6±11.4h), interval from estrus to ovulation (39.9±12.3 h), diameter of largest follicle (7.2±1.4) and number of ovulations (1.8±0.6). At Day 1, 52.4% (11/21) of does presented progesterone concentrations <1ng/mL. At Day 10, 100% of the animals presented concentrations >1ng/mL. The results of the present study indicate that estrus can be efficiently synchronized in dairy goats with the use of two doses of PGF at a 10 day interval. Further research should be done evaluating hCG use in different doses or moments of administration.

AIM: Carbon dioxide (CO2) is an important atmospheric trace gas that is involved in both the biological carbon cycle and global warming. Inland waters - mainly lakes - contribute to C cycling and have been considered a large source of atmospheric CO2. However, scientific studies usually neglect lake morphometry and the presence of aquatic macrophytes in littoral zones, which have a great potential for CO2 absorption and for C storage. This study aimed to evaluate the importance of the littoral region on the CO2 balance in the Cabiúnas Lagoon, while also considering the contribution of the limnetic region and of Typha domingensis and Eichhornia azurea, prominent species in the area. METHODS: CO2 flux was estimated by a linear integration of CO2 concentrations measured in the internal atmospheric of a single-component static closed chambers at the studied sites. The distribution of macrophyte stands throughout lagoon surface was taken into account to evaluate the effects of macrophytes on CO2 supersaturation. Other factors were also measured throughout the sampling process to evaluate their relationship with CO2 flux data by means of Akaike model selection criterion. The area covered by aquatic macrophytes at the Cabiúnas lagoon was estimated by profiles and transects. RESULTS AND DISCUSSION: CO2 flux through the water surface ranged from - 7.39 to 17.56 mgCO2m- 2h- 1. An emission pattern predominated, suggesting that water columns are CO2 supersaturated at all sampling sites. Rates were similar among all the sampling sites, suggesting that aquatic macrophytes do not influence CO2 saturation in the water column at Cabiúnas lagoon. On the other hand, CO2 fluxes from macrophyte tissues showed a clear assimilation pattern. Influxes were higher in T. domingensis ( - 229.1 ± 320.9 mg CO2.m- 2.h- 1) than in E. azurea stands ( - 43.8 ± 39.5 mg CO2.m- 2.h- 1). Once these macrophytes covered a considerable area of the lagoon and CO2 absorption strongly overwhelmed the emission processes, then we were able to extrapolate data from the total estimated area of the evaluated sites (75% of the Cabiúnas lagoon), which resulted in a net influx of 46.6 mg CO2.m- 2.h- 1. The strong Typha domingensis contribution to CO2 absorption and other C cycling processes indicate that it is one of the most important species to Carbon cycling in the studied ecosystem. Thus, it is worth considering C cycling in lake littoral zones as a key process when estimating C balance in shallow aquatic ecosystems.
