Predictors of mortality in critically ill patients with COVID-19 and diabetes

The COVID-19 pandemic has challenged the entire world, and patients with diabetes mellitus (DM) have been particularly affected. We aimed to evaluate predictors of mortality during the first 30 days of hospitalization in critically ill patients with COVID-19 and comorbid DM. This prospective study included 110 critically ill patients admitted with COVID-19 infection. Thirty-two (29%) patients had a previous diagnosis of DM. Clinical variables, laboratory tests, and vascular biomarkers, such as VCAM-1, syndecan-1, ICAM-1, angiopoietin-1, and angiopoeitin-2, were evaluated after intensive care unit (ICU) admission. A comparison was made between patients with and without DM. No difference in mortality was observed between the groups (48.7 vs 46.9%, P=0.861). In the multivariate Cox regression analysis, VCAM-1 levels at ICU admission (HR: 1 [1-1.001], P<0.006) were associated with death in patients with DM. Among patients with DM, advanced age (HR 1.063 [1.031-1.096], P<0.001), increased Ang-2/Ang-1 ratio (HR: 4.515 [1.803-11.308] P=0.001), and need for dialysis (HR: 3.489 [1.409-8.642], P=0.007) were independent predictors of death. Higher levels of VCAM-1 in patients with DM was better at predicting death of patients with severe COVID-19 and comorbid DM, and their cut-off values were useful for stratifying patients with a worse prognosis. Vascular biomarkers VCAM-1 and Ang-2/Ang-1 ratio were predictors of death in patients with severe COVID-19 and comorbid DM and those without DM. Additionally, kidney injury was associated with an increased risk of death.

Seizures are a disorder caused by structural brain lesions, life-threatening metabolic derangements, or drug toxicity. The present study describes the behavior related to proconvulsant activity induced by thiocolchicoside (TCC) in rats and investigates the electrocorticographic patterns of this behavior and the effectiveness of classic antiepileptic drugs used to control these seizures. Forty-nine adult male Wistar rats were used and divided into two phases of our experimental design: 1) evaluation of seizure-related behavior and electrocorticographic patterns induced by TCC and 2) evaluation of the efficacy of classical antiepileptic drugs to control the proconvulsive activity caused by TCC. Our results showed that TCC induced tonic-clonic seizures that caused changes in electrocorticographic readings, characteristic of convulsive activity, with average amplitude greater than that induced by pentylenetetrazole. Treatment with anticonvulsants, especially diazepam, reduced the electrocorticographic outbreaks induced by TCC. The results suggested that TCC caused seizures with increased power in brain oscillations up to 40 Hz and that diazepam may partially reverse the effects.

RESUMO Um cachorro-vinagre (Speothos venaticus) apresentou um nódulo cutâneo pequeno na região perianal. Histologicamente havia neoplasia cutânea de células redondas e ulcerada, constituída por mastócitos bem diferenciados, com abundantes grânulos citoplasmáticos metacromáticos na coloração de Giemsa e infiltrado eosinofílico difuso. A imuno-histoquímica demonstrou que 30% das células neoplásicas eram positivas para a proteína Kit no citoplasma e na membrana celular. As células foram negativas para MAC e CD3. Menos de 10% das células neoplásicas foram positivas para Ki67. Durante a necropsia, foram identificados outros tumores primários, como adenoma intestinal, adenoma cortical da adrenal e tumor de células intersticiais do testículo.
ABSTRACT A senile male captive bush dog (Speothos venaticus) presented a small perianal cutaneous nodule. Histologically, there was an ulcerated round cell tumor composed of well differentiated mast cells with abundant intracytoplasmic purple Giemsa-positive granules, with a diffuse eosinophilic infiltrate. Immunohistochemistry revealed that 30% of the neoplastic cells were positive for Kit in the cytoplasm and cell membrane, and all neoplastic cells were negative for MAC and CD3. Less than 10% of the neoplastic cells were positive for Ki67. At necropsy other primary tumors were identified in this animal, including an intestinal adenoma, an adrenal cortex adenoma and a testicular interstitial cell tumor.

RESUMO: O objetivo do estudo foi realizar a avaliação ultrassonográfica de órgãos do terço médio e caudal da cavidade celomática de serpentes hígidas da família Boidae. Para isto, foram avaliadas 15 serpentes adultas de cinco espécies: Boa constrictor amarali, Boa constrictor constrictor, Eunectes murinus, Epicrates assisi e Epicrates crassus. Os animais foram contidos fisicamente para a realização do exame. Após aplicação de gel acústico sobre as escamas, o transdutor foi posicionado no plano frontal da cavidade celomática. Foram utilizados transdutores microconvexo e linear em 10MHz. O exame ultrassonográfico foi conduzido no sentido craniocaudal, com cortes longitudinais e transversais para avaliar sintopia, ecogenicidade e ecotextura dos órgãos. O fígado se apresentou bilobulado, hiperecoico, ecotextura homogênea, cápsula hiperecoica, com lobo hepático medindo 1,15±0,64cm de largura, e presença de uma veia central hepática, medindo 0,44±0,21cm de diâmetro. A vesícula biliar foi caracterizada como uma estrutura anecoica de paredes ecogênicas e dimensões de 2,91±1,18cm x 1,38±0,84cm(comprimento x largura). O estômago apresentou-se com o aspecto pregueado e linear, ecogênico, com paredes hipoecoicas medindo 0,3±0,07cm de largura. O esplenopâncreasse apresentou como uma estrutura circular e ecogênica, ecotextura homogênea e medindo de 1,18±0,62cm de largura e 1,56±0,88cm de comprimento. Os rins se caracterizaram por estruturas alongadas, lobuladas, hipoecoicas, cápsula ecogênica e uma linha central ecogênica, medindo 1,05±0,7cm de largura. Os testículos foram caracterizados por estruturas fusiformes, hipoecoicas, ecotextura homogênea, medindo 2,31±0,79cm de comprimento e 0,6±0,23cm de largura. Foram visualizados folículos ovarianos em diferentes estágios, de 0,67±0,39cm de largura e 0,73±0,38cm de comprimento. Não foram encontradas diferenças entre espécies de sintopia, ecogenicidade e ecotextura dos órgãos da cavidade celomática. O exame ultrassonográfico demonstrou-se uma técnica segura, não invasiva e eficiente para caracterização dos órgãos da cavidade celomática de serpentes.
ABSTRACT: The aim of this study was the ultrasonographic evaluation of the organs in the middle third and caudal coelomic cavity of healthy snakes in the Boidae family. For such, 15 adult snakes from five species were evaluated: Boa constrictor amarali, Boa constrictor constrictor, Eunectes murinus, Epicrates assisi and Epicrates crassus. The animals were physically restrained for examination. After the application of acoustic gel over the scales the transducer was positioned in the frontal plane of the coelomic cavity. Microconvex and linear transducers at 10MHz were used. The ultrasound examination was conducted in the craniocaudal direction, with longitudinal and transversal sections to assess syntropy, echogenicity and echotexture of the organs. The liver was bilobulated, hyperechoic, homogeneous echotexture, hyperechoic capsule, with a hepatic lobe measuring 1.15±0.64cm wide, and the presence of a central hepatic vein, measuring 0.44±0.21cm in diameter. The gallbladder was characterized as an anechoic structure with echogenic walls and dimensions of 2.91±1.18cm x 1.38±0.84cm (length x width). The stomach showed a pleated and linear texture, echogenic, with hypoechoic walls measuring 0.3±0.07cm wide. The splenopancreas was a circular and echogenic structure, homogeneous echotexture and measuring 1.18±0.62cm in width and 1.56±0.88cm in length. The kidneys were characterized by elongated, lobulated, hypoechoic structures, an echogenic capsule and an echogenic central line, measuring 1.05±0.7cm in width. The testis were characterized by fusiform, hypoechoic structures, homogeneous echotexture, measuring 2.31±0.79cm in length and 0.6±0.23cm in width. Ovarian follicles were seen in different stages, 0.67±0.39cm wide and 0.73±0.38cm long. No differences were found between species of syntropy, echogenicity and echotexture of the organs of the coelomic cavity. The ultrasound examination proved to be a safe, non-invasive and efficient technique for characterizing the organs of the coelomic cavity of snakes.

Eimeria spp. infection in lambs from southern Brazil

RESUMO: As infecções por Eimeria são comuns na ovinocultura mundial. Cordeiros são mais suscetíveis a coccidiose, especialmente em condições estressantes, sendo infectados por diferentes espécies do parasito. Eimeria crandallis e Eimeria ovinoidalis são consideradas as mais patogênicas, causando redução do crescimento, desidratação, anorexia e morte. Neste estudo, a prevalência de Eimeria spp. foi avaliada em cordeiros da região sul do Estado do Rio Grande do Sul, Brasil. Amostras fecais de 248 cordeiros, provenientes de 19 fazendas, foram testadas quanto à presença de oocistos. As amostras positivas foram reexaminadas e os oocistos esporulados analisados morfometricamente para identificação das espécies de Eimeria presentes. Em 100% das fazendas avaliadas houve animais positivos para o protozoário. A frequência das espécies de Eimeria foi: E. ovinoidalis (94.74%), E. crandallis (89.47%), E. granulosa (78.95%), E. parva (68.42%), E. ahsata (63.13%), E. punctata (42.11%), E. bakuensis (36.84%), E. faurei (10.53%) e E. pallida (5.26). Infecção mista foi encontrada em 94.74% das amostras. Este trabalho descreve pela primeira vez a ocorrência de E. crandallis e E. ovinoidalis infectando cordeiros na área de estudo. A ampla distribuição desse protozoário e a alta frequência das espécies patogênicas evidenciam a importância da coccidiose ovina e os danos potenciais nos rebanhos do Rio Grande do Sul.
ABSTRACT: Eimeria infections are common in the sheep industry worldwide. Lambs are more susceptible to coccidiosis, especially in stressful conditions, being infected by different species of the parasite. Eimeria crandallis and Eimeria ovinoidalis are considered the most pathogenic, causing reduced growth, dehydration, anorexia, and death. In this study, the frequency of Eimeria species was evaluated in lambs from the southern region of the Rio Grande do Sul state, Brazil. Fecal samples from 248 lambs, from 19 farms, were tested for the presence of oocysts. The positive samples were re-examined and the sporulated oocysts analyzed morphometrically to identify the presence of Eimeria species. In 100% of the evaluated farms, there were animals positive for the protozoan. The frequency of Eimeria species was: E. ovinoidalis (94.74%), E. crandallis (89.47%), E. granulosa (78.95%), E. parva (68.42%), E. ahsata (63.13%), E. punctata (42.11%), E. bakuensis (36.84%), E. faurei (10.53%), and E. pallida (5.26%). Mixed infection was found in 94.74% of the samples. This research describes, for the first time, the occurrence of E. crandallis and E. ovinoidalis infecting lambs in the study area. The wide distribution of this protozoan and the high frequency of pathogenic species show the importance and potential damage of sheep coccidiosis in herds from Rio Grande do Sul.

RESUMO As gestações múltiplas em humanos correspondem a apenas 3% das gestações, sendo 97-98% dessas gestações gemelares. Sabe-se que a morbimortalidade fetal é maior em gêmeos monocoriônicos do que nos dicoriônicos. A espécie canina é naturalmente multípara, mas o diagnóstico gestacional de gêmeos monocoriônicos não é comum. O objetivo deste relato é descrever o diagnóstico ultrassonográfico de gêmeos monocoriônicos em duas cadelas, sendo uma da raça Pug (caso 1) e outra da raça Shih-Tzu (caso 2). Foi possível verificar a presença de uma única vesícula gestacional contendo dois fetos em cada cadela, por meio da visibilização de duas cabeças ou de dois corpos dentro de uma mesma placenta. Esses fetos apresentavam viabilidade e organogênese adequadas e o grau de desenvolvimento era similar aos demais fetos da ninhada. No caso 1, os gêmeos nasceram mortos e de tamanho menor que os outros cinco fetos nascidos vivos. Os gêmeos do caso 2 nasceram vivos, mas também eram pequenos em relação aos irmãos da ninhada. Os riscos gestacionais associados a essa condição em cadelas ainda não são conhecidos, no entanto já existem relatos de morte fetal em gestação monocoriônica nessa espécie. Portanto, o exame ultrassonográfico durante a gestação permite um diagnóstico precoce da monocorionicidade, e a monitorização da viabilidade fetal pode trazer benefícios para a saúde da matriz, assim como para o restante da ninhada.
ABSTRACT Multiple pregnancies in humans account for only 3% of pregnancies, 97-98% of which are twin pregnancies and the morbimortality is higher in the monochorionic twins when compared to dichorionic ones. The canine species is naturally multiparous, but the diagnosis of monochorionic twin pregnancy is not common. The objective of this report was to describe the ultrasonographic diagnosis of monochorionic twin pregnancies in two bitches [Pug (case 1) and Shih tzu (case 2)]. It was possible to verify the presence of one gestational vesicle containing two fetuses in each female by observing two heads or two bodies within the same placental site. These fetuses presented adequate viability and normal organogenesis. Their development was similar to the other fetuses. In case 1 they were stillborn and smaller than the other five live-born fetuses. The twins in case 2 were born alive, but they also appeared smaller when compared to the littermates. The gestational risks associated with this condition in pregnant bitches are still unknown, however, there are reports of fetal death in monochorionic pregnancies in this species. Therefore, ultrasonographic exam during pregnancy allows an early monochorionic diagnosis and monitoring the fetal viability could bring health benefits to both the female and the littermates.

RESUMO: Poucos estudos avaliaram a diversidade de ectoparasitos e a associação deles com seus hospedeiros que ocorrem no bioma Caatinga, Nordeste do Brasil. Considerando-se essa falta de conhecimento, este estudo objetivou identificar e determinar a ocorrência de carrapatos coletados de pequenos mamíferos da Estação Experimental Rafael Fernandes, no Rio Grande do Norte, Brasil. De janeiro de 2014 a fevereiro de 2015 foram capturados 52 marsupiais (38 Gracilinanus agilis e 14 Monodelphis domestica) e 10 roedores (5 Wiedomys sp., 4 Thrichomys sp. e 1 Rattus norvegicus). Foram identificados os carrapatos Amblyomma auricularium, Amblyomma parvum, Amblyomma sp., Ornithodoros mimon e Ornithodoros sp., empregando estudo morfológico, chaves taxonômicas e sequenciamento parcial do gene mitocondrial 16S rDNA de carrapatos. Todas as associações carrapato-hospedeiro encontradas neste estudo são relatadas pela primeira vez no Rio Grande do Norte e constituem novos dados ecológicos aplicáveis aos ectoparasitos de pequenos mamíferos no nordeste do Brasil.
ABSTRACT: Few studies have assessed the diversity of ectoparasites and their associated hosts occurring within the Caatinga biome in northeastern Brazil. Considering this lack of knowledge, in this study we aimed to identify and determine the occurrence of ticks collected from small mammals at the Estação Experimental Rafael Fernandes, in Rio Grande do Norte state, Brazil. From January 2014 to February 2015, we captured 52 marsupials (38 Gracilinanus agilis and 14 Monodelphis domestica) and 10 rodents (5 Wiedomys sp., 4 Thrichomys sp. and 1 Rattus norvegicus). We identified the ticks Amblyomma auricularium, Amblyomma parvum, Amblyomma sp., Ornithodoros mimon and Ornithodoros sp. by a morphological study, the use of taxonomic keys, and the partial sequencing of the tick mitochondrial 16S rDNA gene. All the tick-host associations found in this study are reported for the first time in Rio Grande do Norte and constitute new ecological data concerning ectoparasites of small mammals in northeastern Brazil.

EFFECT OF SETHOXYDIM HERBICIDE IN THE LEAF ANATOMY AND PHYSIOLOGY OF BRACHIARIA GRASS UNDER WATER STRESS

RESUMO O objetivo do presente trabalho foi analisar a anatomia foliar e o comportamento fisiológico de plantas de capim-braquiária (Urochloa decumbens) submetidas a diferentes condições hídricas e com a aplicação do herbicida sethoxydim. O delineamento experimental utilizado foi o inteiramente casualizado com quatro repetições, constituído de um fatorial 3 x 2, sendo a combinação de três manejos hídricos (-0,03, -0,07 e -1,5 MPa) com e sem a aplicação do herbicida sethoxydim + óleo mineral Assist, na dose recomendada para a espécie (184 g i.a. ha 1). Os parâmetros fisiológicos e anatômicos avaliados foram: taxa fotossintética, condutância estomática, transpiração, diferença entre temperatura ambiente e foliar, massa seca das plantas, espessura das células buliformes, da epiderme adaxial e epiderme total. Nas condições em que o experimento foi realizado, verifica-se que as folhas de capim-braquiária apresentaram epiderme unisseriada e mesofilo homogêneo, com distribuição radiada das células parenquimáticas ao redor dos feixes vasculares. A epiderme adaxial apresentou células buliformes, e os feixes vasculares são do tipo colateral e estão presentes em diversos tamanhos. O estresse hídrico influenciou negativamente na eficiência do herbicida, bem como diminuiu todos os parâmetros fisiológicos. A aplicação do herbicida ocasionou modificações anatômicas em plantas sem estresse hídrico (-0,03 MPa), como limitação no crescimento das células da epiderme e das células buliformes e na espessura total das folhas. No entanto, nos tratamentos com estresse (-0,07 e -1,5 MPa) não houve diferenças na anatomia foliar; apenas ocorreu aumento na espessura total da folha, provavelmente reflexo das condições de estresse hídrico ao qual as plantas foram submetidas.
ABSTRACT The goal of this study was to analyze the leaf anatomy and physiological behavior of Brachiaria grass plants (Urochloa decumbens) under different water conditions and with the application of sethoxydim herbicide. The used experimental design was the completely randomized one, with four replications, consisting of a 3 x 2 factorial scheme, with the combination of three water managements (-0.03, -0.07 and -1.5 MPa) with and without the application of sethoxydim herbicide + Assist mineral oil, at the recommended dose for the species (184 g a.i. ha-1). The assessed physiological and anatomicalal parameters were photosynthetic rate, stomatal conductance, transpiration, difference between leaf and room temperature, dry mass of plants, thickness of bulliform cells, adaxial and total epidermis. Under the conditions in which the experiment was conducted, it appears that Brachiaria grass leaves showed uniseriate epidermis, homogenous mesophyll, with radiated distribution of parenchymal cells around the vascular bundles. The adaxial epidermis presented bulliform cells; the vascular bundles are collateral and are present in different sizes. Water stress had a negative influence on herbicide effectiveness and decreased all physiological parameters. The application of the herbicide caused anatomical changes in plants with no water stress (-0.03 MPa), such as limitations in the growth of epidermic and bulliform cells, and in the total leaf thickness. However, in treatments with stress (-0.07 and -1.5 MPa), there were no differences in leaf anatomy, but an increase in the total thickness of leaves, probably as a result of the water stress conditions to which plants were submitted..

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A Bradypus variegatus habita florestas da América do Sul e alimenta-se de folhas, ramos e brotos de várias plantas. Em decorrência de sua dieta, bem como da escassez de literatura sobre aspectos morfológicos dos Xenarthras, avaliaram-se cinco línguas de Bradypus variegatus, que foram a óbito por causas naturais, provenientes do Museu Paraense Emílio Goeldi e doados ao Laboratório de Pesquisa Morfológica Animal (LaPMA) da UFRA, a fim de se elucidarem os diferentes tipos de papilas e a interface epitélio-tecido conjuntivo. Macroscopicamente, as línguas apresentaram formato alongado, ápice arredondado, corpo, raiz, sulco mediano do ápice à raiz e duas papilas valadas. A mucosa da língua revelou epitélio de revestimento pavimentoso estratificado queratinizado, enquanto a face ventral era delgada e lisa, desprovida de qualquer tipo de papila. Já a face dorsal apresentou-se irregular, com a presença de três tipos papilares: papilas filiformes, fungiformes e valadas. As papilas filiformes encontradas eram do tipo simples, com base circular, distribuídas irregularmente, com maior concentração e desenvolvimento no ápice e no corpo da língua. A papila fungiforme mostrou-se com superfície praticamente lisa e de formato irregular, com a presença de poros gustativos. Estas foram encontradas por toda a superfície dorsal, com maior concentração na parte rostral do ápice. Foram observadas apenas duas papilas valadas, dispostas na região da raiz da língua, com corpo rodeado por sulcos profundos e vários botões gustativos. As línguas de Bradypus variegatus apresentaram papilas linguais similares às descritas para outras espécies de Xenarthras e mamíferos selvagens.
The Bradypusvariegatus inhabits the forests of South America and feeds from leaves, branches and sprouts from different plants. Due to its diet and the lack of literature on the morphological aspect of Xenarthras, five Bradypusvariegatus tongues from animals which died from natural causes were evaluated, and they came from Pará State Museum Emílio Goeldi and were donated to the Laboratory of Animal Morphological Research (LaPMA) from UFRA, for revealing the different types of papillae and epithelial-connective tissue. Macroscopically, the tongues presented elongated shape, rounded apex, body, root, median sulcus in the root's apex, and two vallate papillae. The mucous membrane of the tongue revealed a keratinized stratified pavement epithelium, while the ventral surface of the tongue was thin and smooth, not provided with any type of papillae. However, the dorsal surface of the tongue was irregular with the presence of three types of papillae: filiform, fungiform and vallate papillae. The filiform papillae found were of a simple type, presenting a rounded base, irregularly distributed with a larger concentration and development on the tongue's apex and body. The fungiform papilla showed a practically smooth surface with irregular format, with the presence of gustatory pores; these were found all over the dorsal surface, with larger concentration at the rostral part of the apex. Only two vallate papillae were observed disposed in the root of the tongue, surrounded by a deep groove, and revealing several taste buds. The tongues from Bradypusvariegatus presented gustatory papillae similar to the ones described for other Xenarthras species and wild mammals.

Preparation and characterization of paclitaxel-loaded PLDLA microspheres

Paclitaxel (Taxol®), is a drug used to treat ovarian, breast, lung and bladder cancer. However, the low solubility of this drug in water is a major limitation in its clinical use. One strategy to overcome this limitation would be to encapsulate paclitaxel in polymeric microspheres that are biocompatible and can be used as drug carriers. The aim of this study was to use the bioresorbable, biocompatible copolymer poly-L-co-D,L-lactic acid (PLDLA) in the 70:30 rate to produce and characterize microspheres containing paclitaxel. The simple emulsion technique was used to obtain spherical microspheres that were studied by scanning electron microscopy (SEM) and atomic force microscopy (AFM). The average size of PLDLA microspheres without and with paclitaxel was 10.3 ± 1.7 μm and 12.7 ± 1.3 μm, respectively, as determined by laser light scattering (LLS). Differential scanning calorimetry (DSC) showed that pure paclitaxel had an endothermic peak corresponding to a melting point of 220 °C, which indicated its crystalline nature. The same peak was observed in a physical mixture of PLDLA + paclitaxel in which both components were present in the same proportions used to prepare the microspheres . In contrast, this peak was not observed for the drug, indicating that paclitaxel did not crystallize in PLDLA microspheres. Differential scanning calorimetry (DSC) indicated that paclitaxel was homogeneously dispersed in the PLDLA microspheres, the incorporation of paclitaxel into the microspheres did not alter the thermal properties of PLDLA. The Fourier transform infrared spectroscopy (FTIR) analysis seems to indicate the absence of chemical interaction between polymer and drugs in microspheres and the presence of drugs as a molecular dispersion in the polymer matrix. The efficiency of paclitaxel encapsulation in PLDLA microspheres was 98.0 ± 0.3%, as assessed by high performance liquid chromatography (HPLC). A kinetic study of drug release in vitro using HPLC showed an initial burst release followed by a slower release characteristic of large diameter distribution systems. PLDLA microspheres released 90 ± 4% of the drug over a 30-day period. These findings indicate that PLDLA microspheres are promising carriers for paclitaxel, with a potential for future applications in drug delivery systems.

Dye exclusion tests are used to determine the number of live and dead cells. These assays are based on the principle that intact plasma membranes in live cells exclude specific dyes, whereas dead cells do not. Although widely used, the trypan blue (TB) exclusion assay has limitations. The dye can be incorporated by live cells after a short exposure time, and personal reliability, related to the expertise of the analyst, can affect the results. We propose an alternative assay for evaluating cell viability that combines the TB exclusion test and the high sensitivity of the flow cytometry technique. Previous studies have demonstrated the ability of TB to emit fluorescence when complexed with proteins. According to our results, TB/bovine serum albumin and TB/cytoplasmic protein complexes emit fluorescence at 660 nm, which is detectable by flow cytometry using a 650-nm low-pass band filter. TB at 0.002% (w/v) was defined as the optimum concentration for distinguishing unstained living cells from fluorescent dead cells, and fluorescence emission was stable for 30 min after cell treatment. Although previous studies have shown that TB promotes green fluorescence quenching, TB at 0.002% did not interfere with green fluorescence in human live T-cells stained with anti-CD3/fluorescein isothiocyanate (FITC) monoclonal antibody. We observed a high correlation between the percentage of propidium iodide+CD3/FITC+ and TB+CD3/FITC+ cells, as well as similar double-stained cell profiles in flow cytometry dot-plot graphs. Taken together, the results indicate that a TB exclusion assay by flow cytometry can be employed as an alternative tool for quick and reliable cell viability analysis.

Myosin Va functions as a processive, actin-based motor molecule highly enriched in the nervous system, which transports and/or tethers organelles, vesicles, and mRNA and protein translation machinery. Mutation of myosin Va leads to Griscelli disease that is associated with severe neurological deficits and a short life span. Despite playing a critical role in development, the expression of myosin Va in the central nervous system throughout the human life span has not been reported. To address this issue, the cerebellar expression of myosin Va from newborns to elderly humans was studied by immunohistochemistry using an affinity-purified anti-myosin Va antibody. Myosin Va was expressed at all ages from the 10th postnatal day to the 98th year of life, in molecular, Purkinje and granular cerebellar layers. Cerebellar myosin Va expression did not differ essentially in localization or intensity from childhood to old age, except during the postnatal developmental period. Structures resembling granules and climbing fibers in Purkinje cells were deeply stained. In dentate neurons, long processes were deeply stained by anti-myosin Va, as were punctate nuclear structures. During the first postnatal year, myosin Va was differentially expressed in the external granular layer (EGL). In the EGL, proliferating prospective granule cells were not stained by anti-myosin Va antibody. In contrast, premigratory granule cells in the EGL stained moderately. Granule cells exhibiting a migratory profile in the molecular layer were also moderately stained. In conclusion, neuronal myosin Va is developmentally regulated, and appears to be required for cerebellar function from early postnatal life to senescence.

The ability of antibiotic resistant E. faecalis and E. faecium isolated from food to form biofilm at different temperatures in the absence or presence of 0.75% glucose was evaluated. A synergistic effect on biofilm at 10 °C, 28 °C, 37 °C and 45 °C and glucose was observed for E. faecalis and E. faecium.

Um estudo de vulnerabilidade para fasciolose foi conduzido na área experimental do Instituto Federal de Educação, Ciência e Tecnologia do Espírito Santo, IFES - Campus de Alegre, Espírito Santo. O Sistema de Informação Geográfica (SIG) foi utilizado para a geração de um Modelo Digital de Elevação (MDE) a partir do melhor interpolador para a área do estudo. A partir da análise do melhor interpolador, foi gerada a imagem matricial de declividade que foi reclassificada com o objetivo de discriminar apenas o intervalo representativo da classe matricial de: a) relevo plano da área de estudo (0 a 4%), convertida do formato matricial para o formato vetorial de polígono. Sobre a composição colorida normal e falsa-cor da imagem do satélite Geoeye com resolução espacial de 50 cm, foram fotointerpretadas as classes vetoriais poligonais de: b) várzea, c) corpos d'água e d) pastagem. Sobre essas 4 imagens vetoriais poligonais foi aplicada a função distância em linha reta (straight line) e a padronização linear (função Fuzzy). Após a atribuição dos pesos estatísticos de cada classe vetorial poligonal, foi utilizada a função denominada calculadora matricial (raster calculator) para geração do modelo matemático estatístico: vulnerabilidade da ação ambiental à fasciolose. O interpolador Topo to Raster foi o mais adequado neste estudo para geração do MDE. O mapa de declividade permitiu demonstrar que as áreas de coletas estudadas estão inseridas em locais que apresentaram índices baixos de declividade. Percentuais de 25 e 31% foram encontrados em áreas de altíssimo-alto risco e de alto-médio risco, respectivamente.
A study of vulnerability to fasciolosis was conducted in the experimental area of the Instituto Federal de Ciência e Tecnologia do Espírito Santo - Campus Alegre, Espírito Santo, Brazil. The Geographic Information System was used to build a digital elevation model based on the best interpolator for the study area. From the analysis of the best interpolation, the matrix slope image was generated which was reclassified in order to discriminate only the range representative of the matrix class of: a) flat terrain of the study area (0-4%), converted to vector polygon format. On the composition of normal color and false-color imaging of the Geoeye satellite with 50 cm resolution, photointerpreting was used to establish the vector polygon classes of: b) floodplain, c) water bodies and d) pasture. The (straight line) distance function and linear patterning function (fuzzy function) were applied on these 4 vector polygon images. After the allocation of the statistical weights of each vector polygon class, the raster calculator function was used to generate the statistical mathematical model: vulnerability of the environmental action to fasciolosis. The Topo to Raster interpolation was the most appropriate digital elevation model. The slope map made it possible to show that the collection areas studied are located in places with low rates of slope. Percentages of 25 and 31% were found in areas of highest/high risk and medium-high risk respectively.
