ABSTRACT: The present study investigated Salmonella spp. in the feces of 200 foals up to one year of age (100 with clinical signs of diarrhea and 100 without clinical signs of diarrhea). Bacteriological culture, serotyping, antimicrobial susceptibility, and real-time PCR (qPCR SYBR® Green or a TaqMan®) for detecting the invA gene (with and without a selective pre-enrichment step in tetrathionate broth) were performed. Bacterial culture revealed 15% (n=30) of positive animals (21 animals with diarrhea and nine without diarrhea). Among the 30 isolates, 13 different serovars were identified: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multidrug resistance was found in 43.33% (n=13) of the isolates, with one isolate obtained from animals without diarrhea and 12 isolates from animals with diarrhea. All qPCR techniques used in the study classified more samples as positive for Salmonella spp. than the bacterial culture of feces. In addition, all qPCR techniques detected more positive animals in the diarrhea group than in the diarrhea-free group. The results confirm the utility of the qPCR method without the pre-enrichment step in tetrathionate as a rapid test for Salmonella spp. in carrier animals. In animals with clinical signs of diarrhea, it can be combined with bacterial culture (antimicrobial susceptibility testing and serotyping). The isolation of Salmonella spp. in nine animals without diarrhea confirms the importance of asymptomatic carrier animals in the epidemiology of the disease. The multidrug resistance observed highlights the importance of rational antimicrobial use in horses and adopting biosecurity protocols that are efficacious in controlling the spread of infections between animals and zoonotic transmission in farms.
Dentre esses 30 isolados, 13 sorovares diferentes foram identificados: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-.

BACKGROUND The parasite Giardia duodenalis infects a wide range of vertebrate hosts, including domestic and wild animals as well as humans. Giardia is genotyped into eight assemblages (A-H). Zoonotic assemblages A and B have already been identified in humans and wild and domestic animals (non-human primates and cats) from Brazilian Amazon and in the world. Due to its zoonotic/zooanthroponotic nature, surveillance initiatives and the definition of Giardia assemblages are important in order to characterise the epidemiological scenario and to implement further control measures. OBJECTIVES Determine assemblages of G. duodenalis in sloths from the Brazilian Amazon Region. METHODS Faecal parasitological examination of sloths from Amazonas State. Polymerase chain reaction (PCR) targeting the beta giardin (BG), and genes from multilocus sequence typing (MLST) scheme, amplicon sequencing and phylogenetic analysis. FINDINGS Here, we identified, by microscopy, Giardia in two northern sloths (Bradypus tridactylus). These two samples were submitted to molecular assays and it was revealed that both were infected by G. duodenalis assemblage A. Phylogenetic analysis showed that they belong to assemblage A within sequences from humans and wild and domestic animals. CONCLUSION Therefore, besides showing, by the first time, the current presence of this parasite in sloths, our findings reveals that this wild animal species would be part of the zoonotic/zooanthroponotic scenario of this parasite in the Brazilian Amazon.

Abstract Animals corpses in teaching and research institutions could be sources of infection for students and teachers when applied for dissection and surgical practice. This research aimed to evaluate cats’ corpses’ conservation using a new anatomic technique and vacuum package for seven days, aiming surgical practicing. A 150 mL/kg of alcohol with 5% glycerin and 120 mL/kg of a 20% sodium chloride, 1% nitrite, and 1% sodium nitrate solution was injected on corpses sealed in vacuum packages and put on 0 to 4°C. Skin and jejunum were collected on day 0 (fresh samples/control), and traction analysis was performed for seven consecutive days. On the last day, the liquid in the plastic bags was microbiologically analyzed. There was no statistical difference between control and conservation moments (D1 and D2) in maximal rupture force of the skin, and jejunum was similar to control in D2, D4, and D6. The microbial population did not exceed 6.0x104CFU/mL in total aerobics and 4.8x104CFU/mL in total anaerobes. Biomechanics was not significantly affected, and the microbiological count was low during conservation, demonstrating the possible effectiveness of this anatomical technique for surgery training.

Abstract A study of macro and microfacies, palynoflora and palynofacies of the non-marine Cerro Negro Formation at President Head Peninsula, Snow Island, northwest of the Antarctic Peninsula, was developed. Two assemblages were recognized: Palynofacies assemblage 1 (P1) at the base of the section with a dominance of fern spores and conifer pollen grains, and facies association consisting of a clastic layer, with the predominance of mudstones; and Palynofacies assemblage 2 (P2) at the top of the section, with remarkable abundance of AOM/Pseudoamorphous particles, associated with facies that includes tuffs. The complete section shows in some levels the presence of freshwater algae and translucent phytoclasts. The integrated data characterizes a fluvial-lacustrine environment, what is reinforced by the occurrence of freshwater algae (Botryococcus) in some levels of P1 and P2. We could verify an increase in volcanic activity towards the top of the section that apparently has played an important role in the collapse of the palynoflora. The occurrence of the spore species Muricingulisporis annulatus, Sotasporites elegans, S. triangularis, Foraminisporis wonthaggiensis, and F. asymmetricus in the Cerro Negro Formation allows the correlation with sections in South America and Australia, suggesting an Aptian age for these deposits.

Abstract Two experiments were conducted in order to evaluate the effect of dietary inclusion of guava extract on laying performance and lipid stability of egg of Japanese quails. For the first experiment, 384 Japanese quails with 42 days old were distributed in a completely randomized design with four treatments (0.0; 3.0; 6.0; 9.0 g.kg-1 guava extract), 6 replicates, and 16 birds/replicate. The experimental period lasted 84 days, divided into 4 cycles of 21 days. The parameters evaluated were: feed consumption, egg production, egg mass, feed conversion ratio, egg weight, and egg internal and external qualities. On the second experiment, 400 eggs from Japanese quails fed with diets containing four levels of guava extract were distributed in a completely randomized design with a 4x4 factorial arrangement (guava extract levels and storage time: 0, 9, 18, 27 days) with 5 replicates. Analysis of thiobarbituric acid reactive substances was performed on egg yolks. Data were submitted to regression analysis, using α=0.05. On the first experiment, the laying performance parameters were not influenced by the inclusion of guava extract in the feed, and eggshell thickness decreased linearly. On the second experiment an interaction was observed between the studied factors, affecting lipid oxidation in egg yolks. At nine days of storage, there was a quadratic effect of levels of dietary guava extract on lipid oxidation of yolk, being observed that using 4.8 g.kg-1 of guava extract on feed results in less lipid oxidation.

BACKGROUND Giardia duodenalis is a protozoan parasite that infects humans and other mammals and causes giardiasis worldwide. Giardia is genotyped into eight assemblages (A-H), with assemblages A and B considered zoonotic. OBJECTIVES The aim of this study was to determine the assemblages of G. duodenalis from individuals living in rural and urban areas of the Amazonas State. METHODS 103 human faecal specimens microscopically positive for the presence of Giardia obtained from four municipalities in Amazonas and four animal faecal specimens were genotyped based on the sequences of two genes, triosephosphate isomerase (TPI) and β-giardin (BG). FINDINGS In humans, assemblage A was the most represented with the identification of sub-assemblages AI, AII and AIII based on BG and sub-assemblages AI and AII based on TPI. Similarly, there is a diversity of sub-assemblage B considering BG (B and BIII) and TPI (B, BIII and BIV). In addition, we characterised homogeneous and heterogeneous genotypes comprising assemblages/sub-assemblages A and B in individuals from urban and rural areas. Here, for the first time, it was genotyped Giardia that infects animals from the Brazilian Amazon region. We identified sub-assemblage AI in one Ateles paniscus and two Felis catus and sub-assemblage BIV in one Lagothrix cana. MAIN CONCLUSIONS Therefore, humans and animals from the urban and rural Amazon share Giardia genotypes belonging to assemblages A and B, which are found in cosmopolitan regions around the world.

Abstract Background: Information on the carryover of herbicides applied to the destruction of cotton stalks, as well as the withdrawal period necessary to prevent the development and productivity of the crop in succession from being affected are limited in the literature. Objective: The objective was to identify the carryover effect promoted by herbicides used in the management of the destruction of cotton stalks and to estimate the host free period for sowing soybean in succession. Methods: Two individual experiments were conducted simultaneously, one for single application and the other for sequential application of herbicide treatments. The experiments were installed in a factorial scheme (15x5), in a randomized block design with four replications. The first factor evaluated was herbicide treatments and the second factor was five soybean sowing times after application (0, 30, 60, 90 and 120 days). The herbicides used in the treatments were 2,4-D, glyphosate, saflufenacil, [imazapic + imazapyr], dicamba, fluroxypyr and sulfentrazone. Results: As this work was conducted, the results provide a carryover indicator. It is concluded that the single application and the sequential application of the glyphosate + dicamba + saflufenacil treatment has great carryover potential for soybean crops, with the host free period for sowing the crop exceeding 120 days after application. Conclusions: The treatments 2,4-D, 2,4-D + glyphosate, glyphosate + saflufenacil + fluroxypyr had the lowest host free period intervals, even when in sequential application. The treatment with application of 2,4-D alone showed the lowest carryover potential for soybean.

ABSTRACT: The Labrador Retriever is among the main breeds with the greatest predisposition to obesity. Several factors, especially the interrelationships between food management, exercise and social factors; influence the likelihood of a dog becoming obese. Furthermore, genetic factors are also responsible for obesity in dogs, and in Labrador Retriever, a frameshift mutation (P187fs) in pro-opiomelanocortin (POMC) gene is strongly associated with obesity. There is no knowledge of studies that have previously evaluated the prevalence of the canine POMC deletion (P187fs) in Brazilian Labrador Retriever. Therefore, the objective of this study was to investigate this mutation in Labrador Retriever dogs in Brazil. Of the 108 Labrador Retrievers that were assessed in this study, 59 were from a previous study, composed by animals assisted in a veterinary hospital with unknown lineage, and 49 were from a prospective study, composed of 19 pet and 30 assistance/rescue Labrador Retriever dogs. The obesity risk and appetite questionnaire were applied, with some modifications, to tutors of the animals used in the prospective study. Fragments of the DNA, containing the mutation, were amplified by PCR and submitted to direct gene sequencing. The allele frequency of the mutation was 21.3% and was out of Hardy-Weinberg equilibrium (P<0.05). Using only the data of animals with known lineage, the presence of the mutated allele was higher in the Assistance/rescue Group than Pet Group (P<0.01), furthermore, the allele frequencies observed in Assistance Group (31.7%) was out of Hardy-Weinberg equilibrium (P<0.05), while that in the Pet Group (18.4%) was in equilibrium (P>0.05). Although the mutation has increased the food-motivation in the assistance/rescue dogs, other variables, especially frequent exercising, favored that these animals maintained the ideal body weight (body condition score = 5). In summary, the Hardy-Weinberg disequilibrium observed in the allele distribution of the deletion POMC_P187fs in this study, independently of the Labrador Retriever group assessed, suggesting the possibility of positive selection of the mutated allele, which may lead to the maintenance of this deleterious allele in the studied population.

ABSTRACT To characterize methicillin-resistant Staphylococcus aureus isolates from an intensive care unit of a tertiary-care teaching hospital, between 2005 and 2010. A total of 45 isolates were recovered from patients admitted to the intensive care unit in the study period. Resistance rates higher than 80% were found for clindamycin (100%), erythromycin (100%), levofloxacin (100%), azithromycin (97.7%), rifampin (88.8%), and gentamycin (86.6%). The SCCmec typing revealed that the isolates harbored the types III (66.7%), II (17.8%), IV (4.4%), and I (2.2%). Four (8.9%) isolates carried non-typeable cassettes. Most (66.7%) of the isolates were related to the Brazilian endemic clone from CC8/SCCmec III, which was prevalent (89.3%) between 2005 and 2007, while the USA100/CC5/SCCmec II lineage emerged in 2007 and was more frequent in the last few years. The study showed high rates of antimicrobial resistance among methicillin-resistant S. aureus isolates and the replacement of Brazilian clone, a well-established hospital lineage, by the USA100 in the late 2000s, at the intensive care unit under study.

Specimens of Leonotis nepetiflolia were obtained from cultivated and wild environments to verify their influences in chemical composition. Phytochemical analyses were conducted for the ethyl acetate phase obtained by partitioning the crude ethanol extract from the cultivated leaves of L. nepetifolia. In doing so, flavonoid 3',4',5-trimethoxy-6,7-dihidroxyflavone (cirsiliol), a chemotaxonomic marker of the family Lamiaceae, was isolated. The results reveal that all phases and extracts tested exhibited weak to moderate antimicrobial activity against the strains of bacteria tested. The evaluation of in vitrocytotoxic and antitumor activity showed that the ethyl acetate phases obtained from both wild and cultivated leaves exhibit high potential cytotoxic and antitumor (> 78.0% of inhibition) activity. The major component of these phases was identified by high-performance liquid chromatography-diode array detector and nuclear magnetic resonance analyses using both 1D and 2D methods. The results further indicate that the flavonoid cirsiliol is the agent responsible for the activity observed in these phases.

Abstract: In order to study copper, zinc and iron concentration in the liver of buffaloes with paratuberculosis (PTB), 13 buffalo cows above three years of age of the Murrah and Mediterranean races or their crosses were used. They originated from two farms in the municipalities of São Luís and São Mateus, state of Maranhão, Brazil. The animals were selected according to clinical signs of paratuberculosis, as diarrhea, dehydration and submandibular edema. Rectal biopsies for detection of Mycobacterium avium subsp. paratuberculosis (Map) through PCR in real time (qPCR) and Ziehl-Neelsen (ZN) stain were performed on all animals. Seven buffaloes revealed positive and six were negative for PTB. They were separated into two groups: Group 1 contained seven buffaloes positive for paratuberculosis, and Group 2 contained six animals negative for paratuberculosis. They were euthanized and postmortem examination was performed for the collection of various tissue samples to be fixed in 10% formalin for histopathology; also liver tissue samples were collected to be frozen for chemical analysis of trace minerals (Cu, Zn and Fe). Postmortem showed that all buffaloes with PTB had brown colored mesenteric lymphnodes, indicating hemosiderosis. One buffalo had brown spots on the mucosa of the small intestine. Histopathology revealed moderate to severe hemosiderosis of the spleen in buffaloes of Group 1. All animals positive for PTB showed micromineral levels below the reference values. The average Cu concentration in buffaloes with PTB was only 18.0ppm and the one of Zn only 68.6ppm. In Group 2, negative for PTB, the average Cu concentration was 113.7ppm and the one of Zn 110.0ppm. Fe concentrations in buffaloes of both groups were high (>669ppm). The analysis of trace elements in the liver confirmed copper deficiency, but also showed low levels of zinc and high levels of iron. Based on clinical and pathological findings, and the micromineral concentrations found in the liver, it is concluded that paratuberculosis could lead to mineral deficiency and worsen the clinical condition of the buffaloes.

Abstract: The objective of this study was to detect Brucella abortus in lymph nodes of buffaloes as well as to describe the lesions caused. Nineteen buffalo cows in various stages of pregnancy, serologically positive for brucellosis and subjected to culling were used. Fetal age was determined by ultrasound examination and the size of fetuses was measured at necropsy. Fragments of lymph nodes were collected for histopathology and qPCR. The detection of B. abortus DNA in the lymph nodes was checked from the fourth month of pregnancy in seven buffaloes and in a post-calving buffalo. Acute to chronic lymphadenitis was histologically diagnosed. B. abortus DNA was detected in all evaluated groups of lymph nodes; the mammary lymph nodes were the most affected.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Three sediment cores were taken from Guanabara Bay. Two of them yielded 210Pb profiles that could be applied for dating purposes. Actual sedimentation rates of approximately 1 cm year-1 (five times higher than the baseline values) were observed for both sediment cores, which agree with the reviewed existing data in the literature related to Guanabara Bay sedimentation rates based on 210Pb. Dating validation was carried out based on the chromium, copper and lead profiles, on the 210Pb flux and on the historical record of the main physical interventions occurring in the last century, such as embankment, the channeling of main rivers and the construction of express roads. At the end of this process, only one sediment core dating could be adequately validated.
