Clinical presentation, histological findings, and prognosis in female dogs with mixed mammary neoplasms

ABSTRACT The aim of this study was to describe the clinical and morphological characteristics of mixed mammary neoplasms and verify what characteristics affect the prognosis of female dogs with carcinomas in mixed tumors and carcinosarcomas. This was a retrospective study of 67 female dogs that underwent mastectomies and were diagnosed with benign mixed tumors (n=13), carcinomas in mixed tumors (n=44) and carcinosarcomas (n=10). Data regarding the clinical and histological aspects of the neoplasms were collected and the relation with specific survival times, and hazard ratios (HR) in 24 months was calculated. In univariate analysis, the diagnosis of carcinosarcoma (HR 8.26, p=0.006), carcinomatous areas with micropapillary or solid patterns (HR 17.49; p=0. 001) and lymph node metastasis (HR 7.07;p=0.020) were associated with specific survival. In multivariable analysis, only micropapillary or solid pattern (HR=16.34; p=0.007) remained independent factor associated with lower specific survival. Micropapillary or solid carcinomatous patterns were associated with shorter specific survival time (p=0.002) among animals with carcinomas in mixed tumors. Among the carcinosarcomas, lymph node metastasis (p=0.010) was associated with a shorter specific survival time. In conclusion, carcinomas in mixed tumors and carcinosarcomas vary in prognosis depending on the carcinomatous proliferation patterns and spread of the disease.
RESUMO O objetivo deste estudo foi descrever as características clínicas e morfológicas das neoplasias mamárias mistas e verificar quais características interferem no prognóstico de cadelas com carcinomas em tumores mistos e carcinossarcomas. Este foi um estudo retrospectivo de 67 cadelas, que foram submetidas a mastectomias e diagnosticadas com tumores mistos benignos (n = 13), carcinomas em tumores mistos (n = 44) e carcinossarcomas (n = 10). Foram coletados dados sobre os aspectos clínicos e histológicos das neoplasias e a relação com o tempo de sobrevida específica e a razão de risco (OR) em 24 meses foram calculadas. Na análise univariada, o diagnóstico de carcinossarcoma (HR 8,26; p=0,006), as áreas carcinomatosas com padrão micropapilar e sólido (HR 17,49; p= 0,001) e metástase linfonodal (HR 7,07; p=0,020) foram associadas à menor sobrevida específica. Na análise multivariável, apenas o padrão micropapilar ou sólido (HR=16,34; p=0,007) permaneceu como fator independente associado à sobrevida. Proliferações carcinomatosas micropapilares ou sólidas (p = 0,002) foram associadas a tempos de sobrevida específica mais curtos entre os animais com carcinomas em tumor misto. Entre os pacientes com carcinossarcoma, metástases em linfonodos (p = 0,010) foram associadas a um menor tempo de sobrevida específica. Em conclusão, os resultados mostraram que os carcinomas em tumores mistos e os carcinossarcomas podem ter prognóstico variável, dependendo do padrão de proliferação carcinomatosa e da disseminação linfática da doença.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

Hypovolemia induced by hemorrhage is a common clinical complication, which stimulates vasopressin (AVP) secretion by the neurohypophysis in order to retain body water and maintain blood pressure. To evaluate the role of brain L-glutamate and angiotensin II on AVP secretion induced by hypovolemia we induced hemorrhage (∼25% of blood volume) after intracerebroventricular (icv) administration of AP5, NBQX, or losartan, which are NMDA, AMPA, and AT1 receptor antagonists, respectively. Hemorrhage significantly increased plasma AVP levels in all groups. The icv injection of AP5 did not change AVP secretion in response to hemorrhage. Conversely, icv administration of both NBQX and losartan significantly decreased plasma AVP levels after hemorrhage. Therefore, the blockade of AMPA and AT1 receptors impaired AVP secretion in response to hemorrhage, suggesting that L-glutamate and angiotensin II acted in these receptors to increase AVP secretion in response to hemorrhage-induced hypovolemia.

Abstract Sodium-glucose cotransporter 2 inhibitors (SGLT2i) are the latest class of drugs approved to treat type 2 DM (T2DM). Although adverse effects are often caused by a metabolite rather than the drug itself, only the safety assessment of disproportionate drug metabolites is usually performed, which is of particular concern for drugs of chronic use, such as SGLT2i. Bearing this in mind, in silico tools are efficient strategies to reveal the risk assessment of metabolites, being endorsed by many regulatory agencies. Thereby, the goal of this study was to apply in silico methods to provide the metabolites toxicity assessment of the SGLT2i. Toxicological assessment from SGLT2i metabolites retrieved from the literature was estimated using the structure and/or statistical-based alert implemented in DataWarrior and ADMET predictorTM softwares. The drugs and their metabolites displayed no mutagenic, tumorigenic or cardiotoxic risks. Still, M1-2 and M3-1 were recognized as potential hepatotoxic compounds and M1-2, M1-3, M3-1, M3-2, M3-3 and M4-3, were estimated to have very toxic LD50 values in rats. All SGLT2i and the metabolites M3-4, M4-1 and M4-2, were predicted to have reproductive toxicity. These results support the awareness that metabolites may be potential mediators of drug-induced toxicities of the therapeutic agents.

RESUMO No presente trabalho, foi avaliado o efeito imunomodulador de diferentes doses de talidomida em macrófagos associados ao tumor (TAMs), em um modelo murino de câncer de mama. Camundongos foram inoculados com células 4T1, na região do flanco esquerdo, e tratados com talidomida, uma vez ao dia, nas doses de 50, 100 e 150mg/k, por massa corporal, do quinto dia ao 28º dia de inoculação tumoral. Os tumores foram medidos, o índice de proliferação celular e a contagem de TAMs foram avaliados nos tumores primários e nos pulmões com metástases. Além disso, a taxa de metástases pulmonares também foi avaliada. A talidomida na dose de 150mg/kg diminuiu significativamente o crescimento tumoral, o índice de proliferação celular e a infiltração de TAMs nos tumores primários. Por outro lado, maior número de TAMs e menor índice de proliferação celular foram observados nos pulmões metastáticos, em camundongos tratados com 150mg/kg de talidomida. Ademais, a talidomida na dose de 150mg/kg diminuiu significativamente os nódulos metastáticos nos pulmões. Os resultados demonstraram que o tratamento com talidomida diminuiu o crescimento tumoral e as metástases pulmonares em camundongos, associado com diferentes efeitos na infiltração de TAMs nesses locais.
ABSTRACT The present work evaluated the immunomodulatory effect of thalidomide (Thal) at different doses on tumor-associated macrophages (TAMs) using a mouse model of human breast cancer. Mice were inoculated with 4T1 cells in the left flank and treated with Thal once a day at concentrations of 50, 100, and 150mg/kg body weight from the 5th day until the 28th day of tumor inoculation. The tumors were sized, proliferation index and TAMs count were evaluated in primary tumors and metastatic lungs. In addition, the metastasis rate was evaluated in the lungs. Thal at 150mg/kg significantly decreased tumor growth, proliferation index, and TAMs infiltration in primary tumors. Conversely, a higher number of TAMs and lower proliferation index were observed in metastatic lungs in mice treated with 150mg/kg of Thal. Furthermore, Thal at 150mg/kg significantly decreased the metastatic nodules in the lungs. Our findings demonstrated that Thal treatment considerably decreased the primary tumor and lung metastasis in mice associated with different TAM infiltration effects in these sites.

RESUMO O objetivo deste estudo foi descrever a técnica de venografia retrógrada podal em vacas, comparando os acessos da veia digital dorsal comum III com a digital comum II ou IV, nos membros torácicos e pélvicos, mediante a administração de dois diferentes volumes de contraste. Foram utilizados 53 membros torácicos e pélvicos de 14 vacas, com o torniquete de borracha posicionado a 5cm proximal aosparadígitos. Administraram-se 10mL do diatrizoato de meglumina em 24 membros (grupo 1), sendo 13 na veia digital dorsal comum III pelo acesso 1 (A1) e 11 na digital II ou IV no acesso 2 (A2). No grupo 2, administraram-se 20mL em 29 membros, sendo 15 pelo A1 e 19 pelo A2. Após a administração do contraste, as radiografias foram repetidas a cada 20 segundos até 120 segundos. O grau de preenchimento vascular foi maior no grupo 2, não diferindo entre membros e acessos venosos. Conclui-se que a administração de 20mL de contraste apresentou melhor preenchimento vascular e radiopacidade, não havendo diferença entre 20 e 120 segundos após a administração do contraste na qualidade radiográfica, independentemente do acesso venoso.
ABSTRACT The aim of this study was to describe the technique of retrograde venography foot in cows, comparing the approaches of the dorsal common digital vein III with the digital commons II or IV, thoracic and pelvic by administering two different volumes of contrast members. Fifty three fore and hindlimbs of 14 cows were used, a rubber tourniquet was placed at 5cm above accessory digit. Diatrizoatemeglumine was administered at 10mL to 24 members (group 1), 13 dorsal common digital vein III for access 1 (A1), and 11 digital II or IV access 2 (A2). In group 2 20mL was administered to 29 members, 15 by 19 in A1 and A2. After contrast administration, the radiographs were repeated every 20 seconds until 120 seconds. The degree of vascular filling was greater in group 2, independent of venous access, member or moment. There was no significant difference in the degree of radiopacity of radiographic images when compared to the venous access, time and a member of both groups. We conclude that administration of 20mL of contrast showed better vascular filling and radiopacity, with no difference between 20 and 120 seconds after contrast administration in independent radiographic quality venous access.

Resumo Objetivo: Investigar a associação entre uso de chupeta e mamadeira e comportamentos desfavoráveis à amamentação durante as mamadas. Métodos: Um estudo transversal foi conduzido com 427 bebês/mães. Foram coletados dados sociodemográficos, perinatais e sobre o uso de bicos artificiais (chupeta e/ou mamadeira) através de questionário. Os aspectos de amamentação referentes à posição, afetividade, adequação da sucção, respostas do bebê e anatomia das mamas foram avaliados através da observação durante a mamada. O teste qui-quadrado e a análise de regressão linear múltipla foram usados para investigar associação entre as variáveis. Resultados: Os aspectos de amamentação que apresentaram percentuais mais elevados da categoria ruim foram a adequação da sucção (22,5%) e posição (22,2%). O grupo de bebês que usavam chupeta e/ou mamadeira apresentou percentuais mais elevados nas categorias ruim e regular quando comparados com a categoria bom para todos os cinco aspectos da amamentação avaliados (p < 0,001). A análise de regressão linear revelou que o incremento do número de comportamentos desfavoráveis referentes à posição, afetividade, adequação da sucção e respostas do bebê estavam associados de forma independente tanto ao uso de chupeta quanto ao uso de mamadeira (β positivo, p < 0,05), enquanto que anatomia das mamas estava associado de forma independente apenas com o uso de mamadeira. Conclusão: Os achados sugerem que o uso de chupeta e/ou mamadeira pode estar associado a comportamentos desfavoráveis durante amamentação, em especial o uso de mamadeira.
Abstract Objective: To investigate the association between pacifier use and bottle-feeding and unfavorable behaviors during breastfeeding. Method: A cross-sectional study was conducted with 427 babies/mothers. Socio-demographic, perinatal data, and information about the use of artificial nipples (pacifier and/or bottle) were collected through a questionnaire. The breastfeeding aspects regarding position, affectivity, sucking behavior, baby responses, and breast anatomy were evaluated through observation during breastfeeding. The chi-squared test and the multiple linear regression analysis were used to investigate the association between the variables. Results: The aspects of breastfeeding that showed higher percentages of the category "poor" were sucking behavior (22.5%) and position (22.2%). The group of infants who used pacifiers and/or bottle showed higher percentages in the poor and fair categories when compared with the good category for all five breastfeeding aspects evaluated (p < 0.001). The linear regression analysis revealed that the increase in the number of unfavorable behaviors regarding position, affectivity, sucking behavior, and baby responses were independently associated with both pacifier and bottle use (β positive, p < 0.05), while breast anatomy was independently associated only with bottle use. Conclusion: The findings suggest that the use of pacifiers and/or bottle-feeding may be associated with unfavorable behaviors during breastfeeding, especially the use of bottle-feeding.

RESUMO: A presença de Salmonella spp. em produtos de origem avícola e seus subprodutos se mostra um grande desafio para a produção comercial. Dados de prevalência, dos sorotipos circulantes e do perfil de susceptibilidade antimicrobiana de cepas de Salmonella spp. no Estado do Rio de Janeiro são escassos. Portanto, objetivou-se detectar a presença Salmonella spp. em frangos vivos e carcaças em matadouros do Estados do Rio de Janeiro, identificar os sorotipos e avaliar a susceptibilidade antimicrobiana dessas cepas para fluoroquinolonas e betalactâmicos. Foram coletadas 60 amostras cloacais de frangos vivos e 60 amostras de carcaça de seis matadouros sob Inspeção Estadual (SIE). Os isolados foram sorotipificados e testados frente a oito antimicrobianos: enrofloxacina, ciprofloxacina, norfloxacina, cefalotina, ceftiofur, cefotaxima, amoxicilina/ácido clavulânico e ampicilina pelo método de difusão em disco. Os resultados mostraram uma prevalência de Salmonella spp. de 1,66% (1/60) em amostras de suabe de cloaca e de 26,66% (16/60) em carcaças. Em amostras de suabe de cloaca, somente o sorotipo Senftenberg (1,66%) foi isolado. No total, foram isolados sete sorotipos diferentes nas carcaças: Senftenberg (15%) o mais frequente, seguido por Mbandaka (8,3%), Schwarzengrund (3,3%), Cerro (3,3%), Ohio (3,3%), Minnesota (1,66%) e Tennessee (1,66%). Em relação à susceptibilidade antimicrobiana, 29 (87,87%) isolados foram sensíveis a todos os antimicrobianos testados e 4 (12,12%) isolados foram resistentes a pelo menos três antimicrobianos betalactâmicos ou mais. Não foi observada resistência às fluoroquinolonas. Os resultados encontrados demonstram uma prevalência de Salmonella spp. acima da esperada em matadouros do Estado do Rio de Janeiro, além da presença de vários sorotipos de Salmonella spp. A resistência encontrada para betalactâmicos alerta para a disseminação dessas cepas pela cadeia alimentar.
ABSTRACT: The presence of Salmonella spp. in poultry products and their by-products is a major challenge for commercial production. Data about the prevalence, the circulating serotypes and the antimicrobial susceptibility profile of Salmonella spp. strains in the State of Rio de Janeiro are scarce. Therefore, the aim of this study was to detect the presence of Salmonella spp. in live chickens and carcasses in slaughterhouses of the State of Rio de Janeiro, to identify the serotypes and to evaluate the antimicrobial susceptibility of these strains for fluoroquinolones and beta-lactams. Sixty cloacal swabs samples from broiler chickens and sixty samples of carcasses from six slaughterhouses under State Inspection were collected. The isolates were serotyped and resistance was tested to eight antimicrobials: enrofloxacin, ciprofloxacin, norfloxacin, cephalothin, ceftiofur, cefotaxime, amoxicillin/clavulanic acid and ampicillin by disc diffusion method. The results showed a prevalence of Salmonella spp. of 1.66% (1/60) in cloacal swabs samples and 26.66% (16/60) in carcasses. In cloacal swabs sample only Senftenberg (1.66%) serotype was isolated. In total, seven different serotypes were obtained from carcasses: Senftenberg (15%), followed by Mbandaka (8.3%), Schwarzengrund (3.3%), Cerro (3.3%), Ohio (3.3%), Minnesota (1.66%) and Tennessee (1.66%). Regarding antimicrobial susceptibility, 29 (87.87%) isolates were sensitive to all antimicrobials tested and 4 (12.12%) isolates were resistant to three or more beta-lactams antimicrobials. No susceptibility to fluoroquinolones was observed. These results showed a prevalence of Salmonella spp. higher than expected in slaughterhouses in the State of Rio de Janeiro, besides the presence of several serotypes of Salmonella spp. The resistance found for beta-lactams alerts to the spread of these strains through the food chain.

ABSTRACT The aim of this study was to verify the parasitologycal aspects of fish (Hoplias malabaricus), coming from São Bento city, Maranhão state. For this, 70 fish were used, verifying the parasitism indexes and coefficients. For the parasitological results nematodeos were identified, with zoonotical potential of the Contracaecum spp, Pseudoterranova spp, Eustrongylides spp; monogenetics Gyrodactirus spp, Dactylogirus spp and larva Acanthella of Acanthocephalo. Of the 70 (100%) fishes, 64 (91.4%) were sponged, with prevalence of 82.8% (58) for the nematodeo Contracaecum spp, 65.7% (46), Pseudoterranova spp, 7.1% (5), Eustrongylides spp, 71.4% (50), monogenetics and larva Acanthella of Acantocephalo, respectively. A maior amplitude de intensidade (1-30), coeficiente de variação foi verificado para o nematódeo Pseudoterranova spp., a intensidade média (7,3), índice de abundância (6,7), coeficiente de dominância (36,4) foi ao Contacaecum spp., todos os parasitos estavam no mesentério da cavidade abdominal, encistados em forma larval.The largest intensity width (1-30), variation coefficient was verified for the nematodeo Pseudoterranova spp, the medium intensity (7,3), abundance index (6,7), dominance coefficient (36,4) it went to the Contacaecum spp, all of the parasites were in the mesenterio of the abdominal cavity, encysted in larval form. The results show that fish from this region presented unsatisfactory sanitary conditions in parasitological aspect and can expose consumers to the risks of foodborne illnesses.

Avaliação do teor de antioxidantes, flavonoides e compostos fenólicos em preparações condimentares

RESUMO:Plantas condimentares são amplamente utilizadas na culinária como alimento funcional. Pacientes hipertensos, que retiram o sal de cozinha (NaCl) de sua alimentação, relatam que os alimentos "perdem" o sabor. Visando alternativas para substituir o sal e melhorar a palatabilidade das preparações alimentares de pacientes hipertensos, o objetivo deste estudo foi desenvolver temperos à base de plantas medicinais e analisar os teores de compostos fenólicos, flavonoides e atividade antioxidante destes. Plantas (coentro, salsa, orégano, alecrim, manjericão, alho, cebola) cultivadas organicamente em Viçosa (MG) foram processadas e os temperos foram desenvolvidos a partir de combinação destas espécies, de acordo com teste aceitabilidade sensorial afetivo por meio de escala hedônica. Foram definidos três temperos, dos quais foram feitos extratos metanólicos por remaceração até esgotamento. Nas dosagens utilizou-se DPPH na avaliação da atividade antioxidante; reagente de Folin-Ciocalteu para compostos fenólicos e vanilina clorídrica para flavonoides. As análises foram feitas com quatro repetições e os resultados submetidos à análise de variância e teste de média a 5% de significância. Na aceitação subjetiva dos temperos a nota média foi 7,07±1,09 e 8,0±0,93 na ação afetiva como alimento. O teor mais elevado de compostos fenólicos foi no tempero 2 (alho, salsa e manjericão) (55,04±4,307 mg/mL), a maior ação antioxidante foi no tempero 1 (alho, cebola, coentro e orégano) nos tempos 0 (48,72%) e 30 minutos (78,37%). Os teores de flavonoides foram semelhantes nos 3 temperos. Todos os temperos tiveram boa aceitação por pacientes hipertensos e mostraram-se promissores na substituição dos condimentos convencionais.
ABSTRACT:Plant herbs are widely used in cooking as functional food. Hypertensive patients, who remove salt (NaCl) from their diets, report that the food "loses" flavor. Seeking alternatives to replace salt and improve the palatability of food preparations of hypertensive patients the aim of this study was to develop spices based on medicinal plants and analyze their contents of phenolic compounds, flavonoids and antioxidant activity. Plants (cilantro, parsley, oregano, rosemary, basil, garlic, onion) grown organically in the city of Viçosa (state of Mato Grosso, Brazil) were processed and the seasonings were developed from the combination of these species, according to the affective sensory acceptability test using the hedonic scale. Three spices were defined, and methanol extracts were made from them by maceration until exhaustion. In the dosages, we used DPPH to evaluate the antioxidant activity, Folin-Ciocalteu reagent for phenolic compounds and hydrochloric vanillin for flavonoids. The analyses were performed with four replicates and the results were subjected to analysis of variance and to a 5% significance test. In the subjective acceptance of the spices, the average score was 7.07 ± 1.09 and 8.0 ± 0.93 in the affective action as food. The highest content of phenolic compounds was for treatment 2 (garlic, parsley and basil) (55.04 ± 4.307 mg/mL) and the highest antioxidant activity was measured in treatment 1 (garlic, onion, cilantro and oregano) at 0 (48.72%) and 30 minutes (78.37%). The contents of flavonoids were similar in the three treatments. All spices were well accepted by the hypertensive patients and have shown to be a promising replacement of conventional spices.

Energy partitioning and methane emission by sheep fed sorghum silages at different maturation stages

A partição de energia e a produção de metano por ovinos alimentados com silagens de três híbridos de sorgo comerciais (BRS 610, BR 700 e BRS 655), colhidos em três estádios de maturação (leitoso, pastoso e farináceo), foram avaliadas em câmaras respirométricas de circuito aberto. Utilizou-se delineamento inteiramente ao acaso, em arranjo fatorial 3 x 3 (híbridos x estádios de maturação), e as médias foram comparadas pelo teste Student-Newman-Keuls (SNK) (P<0,05). Os consumos de matéria seca, de matéria seca digestível, de energia bruta, de energia digestível e de energia metabolizável não foram afetados pelo estádio de maturação, mas foram influenciados pelo híbrido de sorgo. O consumo de energia líquida foi influenciado pela maturidade e pelo híbrido. As perdas de energia nas fezes representaram a principal fonte de perda energética como porcentagem do consumo de energia bruta (48% a 52%), seguida pelo incremento calórico (10% a 19%), pela emissão de metano (4% a 6%) e pela urina (1 a 2%). Não houve diferenças (P>0,10) entre os tratamentos para a digestibilidade aparente da energia bruta e para a metabolizabilidade (qm). Foi observada interação (P<0,05) entre híbrido de sorgo e estádio de maturação para eficiência de uso da energia metabolizável para mantença (Km), que variou entre 0,53 e 0,78. Não ocorreram diferenças (P>0,10) entre tratamentos na produção diária de metano. A emissão de metano em gramas por kg de matéria seca digestível e por kg de fibra em detergente neutro digestível foi inferior no estádio pastoso comparado ao estádio farináceo.
Energy partitioning and methane production by sheep fed silages of three commercially available sorghum hybrids (BRS 610, BR 700 and BRS 655) harvested at three maturation stages (milk, soft dough and floury) were evaluated in open circuit respiration chambers. A complete randomized design was used in a 3 × 3 (hybrids × maturity stages) factorial arrangement, and the means were compared by the Student-Newman-Keuls (SNK) test (P<0.05). The intake of dry matter, digestible dry matter, gross energy, digestible energy and metabolizable energy were not affected by maturation stage, but were influenced by hybrid. The net energy intake was influenced by maturity and sorghum genetics. The fecal output represented the main source of energy loss, as percentage of gross energy intake (48% to 52%), followed by heat increment (10% to 19%), methane emissions (4% to 6%) and urine (1% to 2%). There were no differences (P>0.10) among the treatments for the apparent digestibility of gross energy and metabolizability (qm). An interaction (P<0.05) between sorghum hybrid and maturation stages was observed for the efficiency of metabolizable energy utilization for maintenance (km), which ranged between 0.53 and 0.78. No differences (P>0.10) among treatments occurred in the daily methane production. There is substantial genetic diversity within sorghum species, determining different nutritional values. Sorghum genetics and maturity at harvest should not be an opportunity to reduce the contribution of agriculture to methane emissions.

Foi avaliada a cinética de fermentação ruminal in vitrode três híbridos comerciais de milho (BRS 1035, BRS 1031 e BRS 1001) colhidos em três estádios de maturidade (pastoso, farináceo e farináceo-duro). Após 96 horas de fermentação, não houve diferença na PCG entre as silagens dos híbridos colhidos nos diferentes estádios de maturidade (P>0,05). O potencial máximo de produção de gases das silagens de todos os híbridos reduziu com o avançar da maturidade da planta, sendo a variação encontrada de 259,3 a 280,0mL/g de matéria seca (MS). Os valores de tempo de colonização variaram de 0h06min a 1h54min. Para as silagens dos híbridos BRS 1035 e BRS 1001, houve aumento no tempo de colonização com o avançar da maturidade da planta. Para as silagens do híbrido BRS 1031, houve aumento dos valores de tempo de colonização quando a planta avançou do estádio pastoso para farináceo, e reduziu quando a maturidade alcançou o estádio farináceo-duro. A taxa fracional de produção de gases "µ" variou entre 0,03mL/h a 0,05mL/h. Com o avançar da maturidade fisiológica, as silagens dos híbridos BRS 1035 e BRS 1001 apresentaram aumento nos valores de µ; já para as silagens do híbrido BRS 1031, os valores de µ aumentaram quando o estádio de maturidade avançou de pastoso para farináceo, e reduziu para farináceo-duro. Os valores das degradabildades efetivas foram superiores para as silagens dos diferentes híbridos colhidas no estádio pastoso. As degradabilidades da MS com o tempo de fermentação de 96 horas variaram de 70,1 a 76,4%. A silagem do híbrido BRS 1035 apresentou melhor valor nutricional quando colhida no estádio pastoso de maturidade. Já a silagem do híbrido BRS 1031 apresentou valor nutricional superior no estádio farináceo, enquanto a silagem do híbrido BRS 1001 apresentou melhor valor nutricional no estádio farináceo-duro.
The in vitro ruminal fermentation kinetics of three commercial corn hybrids (BRS 1035, BRS 1031 and BRS 1001) harvested at three maturity stages (soft dough, floury and hard-floury) were evaluated. After 96 hours of fermentation there was no change in the cumulative gas production (GPC) among silages harvested for all hybrids (P>0.05). The silages maximal degradation potential for all hybrids decreased with advanced maturity stages, and ranged from 259.32 to 279.96mL/g of dry matter. The lag values ranged from 0h 06min to 1h 54min. For silages from the BRS 1035 and BRS 1001 hybrids, there was an increase in lag with advancing plant maturity. For silage from the BRS 1031 hybrid, the lag values increase when the plant moved of soft dough stage to floury, and was reduced to hard-floury stage. The fractional rate of gas production µ ranged from 0.0292mL/h to 0.0447mL/h. With advancing physiological maturity stages, the hybrids BRS 1035 and BRS 1001 showed an increase in the µ values, since for silage from the BRS 1031 hybrid, the value of µ increased when the maturity stage progressed from soft dough to floury, and reduced to hard-floury. The values of effective degradability were higher for silages from different hybrids harvested at soft dough. The dry matter degradability (DMD) in the fermentation time of 96 hours ranged from 70.14 to 76.40%. The silage from the BRS 1035 hybrid showed better nutritional value when harvested in the soft dough stage of maturity. The silage from the BRS 1031 hybrid showed highest nutritional value at the floury stage, while the silage from the BRS 1001 hybrid showed better nutritional value in the hard-floury stage.

Acute oral toxicity of Celtis iguanaea (Jacq.) Sargent leaf extract (Ulmaceae) in rats and mice

RESUMO Celtis iguanaea (Jacq.) Sargent é uma planta usada popularmente para tratar infecções do trato urinário, rim, mama, dores no corpo, reumatismo, asma, cólicas, má digestão e também é usada como diurético. Este trabalho objetivou determinar a toxicidade aguda do extrato aquoso de folhas de Celtis iguanaea (Jacq.) Sargent em roedores. Após os processos de coleta, identificação, secagem e moagem, o pó liofilizado das folhas da planta foi utilizado para produzir o seu extrato aquoso por infusão e então dissolvido em solução salina a 0.9 %. A administração foi feita por gavagem na dose de 2000 mg kg-1 em ratos e camundongos de ambos os sexos. A toxicidade oral foi determinada de acordo com o guia 423 da OECD. Sinais de toxicidade foram observados por 15 dias e tabulados de 0 a 4, respectivamente, como ausentes, raros, leves, moderados e graves. Foi acompanhado o peso dos animais e parâmetros fisiológicos tais como alimentação e excreções. Amostras do tecido de todo o animal foram coletadas para análise histológica. A toxicidade encontrada para o extrato foi incluída na classe 5 (substâncias com DL50 superior a 2000 mg kg-1 e menor que 5000 mg kg-1) sendo considerada baixa, porém, as observações histopatológicas sugerem nefrotoxicidade e cardiotoxicidade. O peso absoluto dos rins e coração de ratos e camundongos machos aumentou, porém, não houve aumento significativo no peso relativo dos órgãos dos animais.
RESUMO Celtis iguanaea (Jacq.) Sargent is popularly used to treat urinary infections, kidneys, breast, body aches, rheumatism, asthma, cramps, poor digestion and as a diuretic medicine. This study aims to determine the acute toxicity of the aqueous leaf extract of Celtis iguanaea (Jacq.) Sargent in rodents. After the collection processes, identification, drying and grinding, the lyophilized powder of the leaves produced, by infusion, the aqueous extract and it was dissolved in saline 0.9%. The administration was made by gavage at a dose of 2000 mg kg-1to rats and mice of both genders. The oral toxicity was determined according to the OECD 423 guide. Signs of toxicity were observed for 15 days and classified from 0 to 4 respectively as missing, rare, mild, moderate and severe. The weight of the animals and the physiological parameters such as food intake and excrements production were observed. All animal tissue samples were collected for histological analysis. The extract was included in Type 5 (substance with LD50 higher than 2000 mg kg-1 and less than 5000 mg kg-1), being considered of low toxicity, but the histopathologycal findings suggested nephrotoxicity and cardiotoxicity. The absolute weight of the kidneys and the heart of the male rats and mice increased, but there was no significant raise in the relative weight of the animals’ organs.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Resumo: Para determinar as concentrações plasmáticas de proteínas e metabólitos de cascavéis em cativeiro, foram utilizadas 60 serpentes adultas, sendo 30 machos e 30 fêmeas. O sangue foi coletado através de punção do seio venoso paravertebral cervical e armazenado em tubos com heparina. As análises bioquímicas foram processadas colorimetricamente em Analisador Automático de Bioquímica Chemwell (Awareness Technology®, Inc). Foram calculadas as médias e desvios padrão dos seguintes constituintes: proteínas totais, albumina, globulinas, relação albumina/globulinas, ácido úrico, creatinina, ureia, colesterol, colesterol HDL e triglicérides. Os valores obtidos foram semelhantes aos descritos na literatura para repteis e serpentes, sendo as diferenças observadas provavelmente decorrentes da diferença entre espécies, clima, estação do ano e metodologia utilizada. Não houve diferenças significativas entre machos e fêmeas para os parâmetros estudados. Estes resultados podem ser úteis no estabelecimento de valores de referência para planos de conservação destes ofídios em cativeiro.
Abstract: For determining plasma concentrations of proteins and metabolites of rattlesnakes in captivity, 60 adult snakes, 30 males and 30 females were used. Blood was collected by puncture of the cervical paravertebral venous sinus and stored in tubes with heparin. Biochemical analyzes were colorimetrically processed using an Automatic Biochemistry Analyzer Chemwell (Awareness Technology®, Inc). The mean and standard deviation were calculated for the following constituents: total protein, albumin, globulin, albumin/globulin ratio, uric acid, creatinine, urea, cholesterol, HDL cholesterol and triglycerides. The values were similar to those previously reported for reptiles and snakes, with the differences observed probably due to the difference between species, climate, season and the methodology used. There were no significant differences between males and females for the parameters studied. These results may be useful in establishing normal biochemical values for conservation plans for these snakes in captivity.
