ABSTRACT: Actinobacilluspleuropneumoniae is the causative agent of swine pleuropneumonia, a contagious respiratory disease associated with high morbidity and economic losses. While antibiotic therapy helps to control the spreading of the pathogen on the farm, resistance to several classes of antibiotics were reported, and treatment can be impaired by the bacterial ability to form biofilms. This increases the need for alternative therapy approaches, including the use of natural compounds with antimicrobial and/or antibiofilm activities. In this research we analyzed, by the broth microdilution method, the inhibitory and bactericidal activities of the essential oils obtained from eighteen Brazilian popular medicinal plants or spices against clinical isolates of Actinobacilluspleuropneumoniae. After that, sub-inhibitory concentrations of active oils were tested for their antibiofilm effects, analyzed by the crystal violet method. Among the eighteen oils tested, eight (extracted from cinnamon, coriander, peppermint, spearmint, thyme, marjoram, eucalyptus, and laurel) presented bacteriostatic and bactericidal activities against all isolates, and subinhibitory concentrations of five of them disrupted up to 80% preformed biofilms, and significantly inhibited biofilm formation. The chemical composition of such oils was assessed by gas chromatography/mass spectrometry (GC/MS) and is presented, indicating that their bactericidal and antibiofilm properties were mostly associated with the presence of monoterpenes and phenylpropanoids. To our knowledge, this is the first report of essential oils with potential to control environmental contamination and animal infection with A. pleuropneumoniae, representing an alternative to increasing levels of antibiotic resistance.

Dental trauma is a public health problem because of its high prevalence rates and treatment costs and for affecting mainly younger individuals. Aim This cross-sectional study aimed to investigate the variables associated with dental trauma and its influence on oral health-related quality of life among 7th-grade students of public elementary schools. Methods The data was collected with semi-structured and self-administered questionnaires and an intraoral clinical examination (n = 204). The variables collected regarded dental trauma, lip coverage, overjet, oral health-related quality of life (Child Perceptions Questionnaire - CPQ11-14 ISF: 8), and sociodemographic profile. Association tests and the Mann-Whitney test were performed at a 5% significance level. Results Dental trauma occurred in 12.3% of students, 94.6% had adequate lip coverage, and the mean overjet was 2.7 mm (±2.0). Sex was the only variable with a statistically significant association with dental trauma (p = 0.021). There was no statistically significant difference in the CPQ11-14 ISF: 8 score between students with and without dental trauma (p = 0.136). Conclusions Dental trauma was significantly associated with sex and did not impact the oral health-related quality of life of the students.

Abstract Background Major advances have been seen in techniques and devices for performing percutaneous coronary interventions (PCIs) for chronic total occlusions (CTOs), but there are limited real-world practice data from developing countries. Objectives To report clinical and angiographic characteristics, procedural aspects, and clinical outcomes of CTO PCI performed at dedicated centers in Brazil. Methods Included patients underwent CTO PCI at centers participating in the LATAM CTO Registry, a Latin American multicenter registry dedicated to prospective collection of these data. Inclusion criteria were procedures performed in Brazil, age 18 years or over, and presence of CTO with PCI attempt. CTO was defined as a 100% lesion in an epicardial coronary artery, known or estimated to have lasted at least 3 months. Results Data on 1196 CTO PCIs were included. Procedures were performed primarily for angina control (85%) and/or treatment of moderate/severe ischemia (24%). Technical success rate was 84%, being achieved with antegrade wire approaches in 81% of procedures, antegrade dissection and re-entry in 9%, and retrograde approaches in 10%. In-hospital adverse cardiovascular events occurred in 2.3% of cases, with a mortality rate of 0.75%. Conclusions CTOs can be treated effectively in Brazil by using PCI, with low complication rates. The scientific and technological development observed in this area in the past decade is reflected in the clinical practice of dedicated Brazilian centers.

ABSTRACT: This study applied spectroradiometry techniques with hyperspectral data to identify the correlations between sugarcane leaf reflectance and the contents of Nitrogen (N), phosphorus (P), Potassium (K), Sulfur (S), Calcium (Ca) and Magnesium (Mg). During the harvests 2019/20 and 2020/21, sugarcane was introduced to nutritional stress by the application of limestone doses. Liming was applied in a fractional way and, at the end of five years, the amounts corresponded to 0, 9, 15 and 21 t ha-1 of dolomitic limestone. The leaf hyperspectral reflectance data and the state of nutrients in the exponential growth phase of the culture were registered. The wavelengths correlated with N, P, K, S, Ca and Mg were identified using the Spearman’s correlation analysis. The test of similarity (ANOSIM) and the Principal Component Analysis (PCA) were applied to evaluate data variability, as well as the Partial Least Squares Regression (PLSR) for the prediction of the nutritional contents. The order of the degree of correlation in the region of visible was: P > K > N > Ca > S > Mg and for the region of the near infrared: P > K > Ca > N > S > Mg. P presented peaks with high correlations in the wavelengths 706-717 nm (-0.78) and 522-543 nm (-0.76). The values of the PLSR registered the best spectral responses in the region of VIS and red-edge, regions that are more sensitive to the deficiency of sulfur, potassium and phosphorus.

Abstract This study aimed to evaluate the contribution of oral and maxillofacial pathology laboratories (OMPLs) in Brazilian public universities to the diagnosis of lip, oral cavity, and oropharyngeal squamous cell carcinoma (SCC). A cross-sectional study was performed using biopsy records from a consortium of sixteen public OMPLs from all regions of Brazil (North, Northeast, Central-West, Southeast, and South). Clinical and demographic data of patients diagnosed with lip, oral cavity, and oropharyngeal SCC between 2010 and 2019 were collected from the patients’ histopathological records. Of the 120,010 oral and maxillofacial biopsies (2010-2019), 6.9% (8,321 cases) were diagnosed as lip (0.8%, 951 cases), oral cavity (4.9%, 5,971 cases), and oropharyngeal (1.2%, 1,399 cases) SCCs. Most cases were from Brazil’s Southeast (64.5%), where six of the OMPLs analyzed are located. The predominant profile of patients with lip and oral cavity SCC was Caucasian men, with a mean age over 60 years, low schooling level, and a previous history of heavy tobacco consumption. In the oropharyngeal group, the majority were non-Caucasian men, with a mean age under 60 years, had a low education level, and were former/current tobacco and alcohol users. According to data from the Brazilian National Cancer Institute, approximately 9.9% of the total lip, oral cavity, and oropharyngeal SCCs reported over the last decade in Brazil may have been diagnosed at the OMPLs included in the current study. Therefore, this data confirms the contribution of public OMPLs with respect to the important diagnostic support they provide to the oral healthcare services extended by the Brazilian Public Health System.

ABSTRACT The objective of this work was to determine the physical and mechanical properties of paricá (Schizolobium parahyba var. amazonicum (Huber x Ducke)) LVL panels bonded with urea-formaldehyde resin. Six LVL panels were produced with 17 sheets of 2.5 mm thick, 14 sheets of paricá and 3 sheets of marupá (Simarouba amara Aubl.). For the standard temperature condition of 12%, tests were carried out on moisture content, apparent and basic densities (physical characterization), resistance to resistance parallel and perpendicular to the fibers, traction parallel to the fibers, shearing in the glue line and flatwise static bending and edgewise (mechanical characterization). In addition, saturation tests parallel to the fibers and shear tests were carried out in the LVL glue line in the condition of total saturation of the wood. The results appreciated that the LVL panels fall within the D30 strength class and medium density. The brittle type failure mode was observed in all mechanical tests, except in the parallel and perpendicular compression tests to the fibers. The increase in moisture content, due to the total saturation of the wood, negatively influences the resistance to the slope parallel to the fibers and to the shear strength in the glue line. The LVL presented a mechanical performance compatible with the substitute.

Abstract Dermoid cysts (DCs) and epidermoid cysts (ECs) are uncommon developmental cysts affecting the oral cavity. This study aims to evaluate patients with oral DCs and ECs and their demographic and clinicopathologic features. A retrospective descriptive cross-sectional study was performed. A total of 105,077 biopsy records of oral and maxillofacial lesions from seven Brazilian oral pathology centers were analyzed. All cases diagnosed as oral DCs and ECs were reviewed, and clinical, demographic, and histopathological data were collected. The series comprised 32 DCs (31.4%) and 70 ECs (68.6%). Most of the DCs occurred on the floor of the mouth (n = 14; 45.2%) of women (n = 17; 53.1%) with a mean age of 34.6 ± 21.6 years. All DCs were lined partially or entirely by stratified squamous epithelium (100%). Chronic inflammatory cells, melanin pigmentation, multinucleated giant cell reaction, and cholesterol clefts were observed in the fibrous capsule . Most of the ECs affected the labial mucosa (n = 20; 31.7%) of men (n = 39; 56.5%) with a mean age of 48.0±19.8 years. Microscopically, most ECs (n = 68; 97.1%) were lined entirely by stratified squamous epithelium. Two cysts (2.9%) showed areas of respiratory metaplasia. Chronic inflammatory cells, melanin pigmentation, multinucleated giant cell reaction, and cholesterol clefts were also observed in the fibrous capsule. Conservative surgical excision was the treatment of choice in all cases. Oral DCs and ECs are uncommon and often clinically misdiagnosed lesions. Clinicians should consider DCs and ECs in the differential diagnosis of soft tissue lesions in the oral cavity, mainly located on the floor of the mouth and labial mucosa.

Abstract This study evaluated the effect of increasing the nutritional plane in finishing diets on meat quality traits of culled adult goats. Forty-one crossbred does were divided into four groups: baseline diet formulated to meet 100% of the nutritional requirements (BD); BD + 30% (BD30); + 60% (BD60); + 90% (BD90) of the nutritional level of BD. The goats received the experimental diets for 28 days. The protein profile of the longissimus lumborum muscle was evaluated using 1D SDS–PAGE and mass spectrometry. Cholesterol and albumin concentrations were highest in the BD group. There were no alterations in loin proximate composition or tissue composition. However, there was a significant reduction in the PUFA content in groups BD60 and BD90 compared with the group fed the baseline diet. Of 37 protein bands identified, 35 showed lower intensities in group BD90 than in BD and 22 bands showed lesser expression in groups BD60 and BD90 than in BD. The multivariate model identified 14 band proteins involved in group-diet segregation. Therefore, increasing the nutrient supply for finishing goats did not have marked positive effects on carcass. A lower expression of these proteins can have a profound impact on the qualitative aspects of the meat product.

ABSTRACT Rainfall erosivity (R factor) is one of the six factors of the Universal Soil Loss Equation, being calculated based on the product of rainfall kinetic energy multiplied by its 30-minute maximum intensity. However, the lack of detailed and reliable rainfall data in many parts of the world has driven the use of other methods to estimate rainfall erosivity based on daily, monthly or annual data. These methods still need to be assessed to determine if their estimates are consistent with the standard method for calculating rainfall erosivity. This study aimed to select a consistent method for such replacement in Brazilian conditions without access the rainfall intensity data. The tested methods included: modified Fournier, MF; modified Fournier by Zhang, MF-Z; modified Fournier by Men, MF-M; Rainfall Disaggregation, RD; TRMM Satellite with modified Fournier coefficient, TRMM-F; and TRMM Satellite with monthly rainfall, TRMM-M. The rainfall data were obtained from the USP Meteorological Station, referring to the period from 2009 to 2015. The analyses were performed according to the Additive Main effects and Multiplicative Interaction (AMMI) model and Scott-Knott statistical tests. Considering the 1:1 line, all methods had a good adjustment, presenting similar behavior in relation to the standard method. The methods behaved differently for monthly and annual periods. The MF method proved to be capable of consistently replacing the standard method in all aforementioned situations. Considering the driest period, any method can be used. For annual rainfall erosivity estimation, the RD, MF, TRMM-F and TRMM-M methods can be applied; highlighting that the TRMM-based methods are optimal for locations without on-site rain gauges. Additionally, it was computed that the modified Fournier by Men and the modified Fournier by Zhang underestimated and overestimated the rainfall erosivity, respectively.

Abstract Health professionals working to mitigate the COVID-19 pandemic are one of the main risk groups for the disease, being prioritized for vaccination. Considering this, the aim of this study was to analyze the immune response of these professionals immunized with CoronaVac in the first and second doses. Blood samples were collected after the first and second doses of the vaccine (CoronaVac) and used to investigate hematological and biochemical parameters, analysis of immunoglobulin production, cytokines, and gene expression profile, as well as the identification of subsets of immune cells. Post-first dose immunological phenotypic memory (CD27+) profiles (T CD4+, TCD8+ and CD19+) showed a significant increase, as did Monocyte APCs (CD80+HLA-DR+) in relation to the second dose. The cytokines IL-2, IL-6 and IFN-° showed increased values in relation to the other analyzed cytokines. The Th2/Th17 profile in the second dose was characterized by gene expression analysis. The production of IgM and IgG after vaccination showed statistically significant values in the comparison between doses. CoronaVac showed activation of APCs monocytes, memory response of T and B lymphocytes, with immunoglobulins production. This set of responses is characterized by the Th2/Th17 immunological profile.

ABSTRACT Due to the environmental problem related to the deposition of residues from the production of mineral coal in Brazil, the objective of this study was to use the residue of mineral coal in the production of mortar as a mineral addition and also as a substitute for Portland cement. For analysis of the physical-mechanical properties of mortars, 5x10 cm specimens were used. Starting from a mortar trace without addition of residue, called reference, to later be used the contents of 10 and 15% for addition, and 20 and 30% for substitution of Portland cement with mineral coal residue. The mortar samples underwent a curing process in a humid chamber and, at 28 days, density tests in the hardened state, water absorption by capillarity, compressive strength and X-ray diffractometry were performed. The results obtained indicated that the mineral coal waste increased the water absorption of mortar by 67% and 100% for the addition of 10 and 15% of coal, respectively. To replace 20 and 30%, the increases in absorption were 133% and 33%, respectively. The mechanical resistance, on the other hand, showed a reduction, for addition of 10 and 15%, of 33% and 42%, respectively. For the replacement of cement by 20 and 30%, a reduction in strength of 35% and 47% was observed.

Abstract Introduction: Next Generation Sequencing (NGS) is cost-effective and a faster method to study genes, but its protocol is challenging. Objective: To analyze different adjustments to the protocol for screening the BRCA genes using Ion Torrent PGM sequencing and correlate the results with the number of false positive (FP) variants. Material and methods: We conducted a library preparation process and analyzed the number of FP InDels, the library concentration, the number of cycles in the target amplification step, the purity of the nucleic acid, the input, and the number of samples/Ion 314 chips in association with the results obtained by NGS. Results: We carried out 51 reactions and nine adjustments of protocols and observed eight FP InDels in homopolymer regions. No FP Single-Nucleotide Polymorphism variant was observed; 67.5% of protocol variables were jointly associated with the quality of the results obtained (p<0.05). The number of FP InDels decreased when the quality of results increased. Conclusion: The Ion AmpliSeq BRCA1/BRCA2 Community Panel had a better performance using four samples per Ion-314 chip instead of eight and the optimum number of cycles in the amplification step, even when using high-quality DNA, was 23. We observed better results with the manual equalization process and not using the Ion Library Equalizer kit. These adjustments provided a higher coverage of the variants and fewer artifacts (6.7-fold). Laboratories must perform internal validation because FP InDel variants can vary according to the quality of results while the NGS assay should be validated with Sanger.

Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.
