Prevalência e susceptibilidade antimicrobiana de sorotipos de Salmonella spp. isolados de frangos vivos e carcaças no estado do Rio de Janeiro

RESUMO: A presença de Salmonella spp. em produtos de origem avícola e seus subprodutos se mostra um grande desafio para a produção comercial. Dados de prevalência, dos sorotipos circulantes e do perfil de susceptibilidade antimicrobiana de cepas de Salmonella spp. no Estado do Rio de Janeiro são escassos. Portanto, objetivou-se detectar a presença Salmonella spp. em frangos vivos e carcaças em matadouros do Estados do Rio de Janeiro, identificar os sorotipos e avaliar a susceptibilidade antimicrobiana dessas cepas para fluoroquinolonas e betalactâmicos. Foram coletadas 60 amostras cloacais de frangos vivos e 60 amostras de carcaça de seis matadouros sob Inspeção Estadual (SIE). Os isolados foram sorotipificados e testados frente a oito antimicrobianos: enrofloxacina, ciprofloxacina, norfloxacina, cefalotina, ceftiofur, cefotaxima, amoxicilina/ácido clavulânico e ampicilina pelo método de difusão em disco. Os resultados mostraram uma prevalência de Salmonella spp. de 1,66% (1/60) em amostras de suabe de cloaca e de 26,66% (16/60) em carcaças. Em amostras de suabe de cloaca, somente o sorotipo Senftenberg (1,66%) foi isolado. No total, foram isolados sete sorotipos diferentes nas carcaças: Senftenberg (15%) o mais frequente, seguido por Mbandaka (8,3%), Schwarzengrund (3,3%), Cerro (3,3%), Ohio (3,3%), Minnesota (1,66%) e Tennessee (1,66%). Em relação à susceptibilidade antimicrobiana, 29 (87,87%) isolados foram sensíveis a todos os antimicrobianos testados e 4 (12,12%) isolados foram resistentes a pelo menos três antimicrobianos betalactâmicos ou mais. Não foi observada resistência às fluoroquinolonas. Os resultados encontrados demonstram uma prevalência de Salmonella spp. acima da esperada em matadouros do Estado do Rio de Janeiro, além da presença de vários sorotipos de Salmonella spp. A resistência encontrada para betalactâmicos alerta para a disseminação dessas cepas pela cadeia alimentar.
ABSTRACT: The presence of Salmonella spp. in poultry products and their by-products is a major challenge for commercial production. Data about the prevalence, the circulating serotypes and the antimicrobial susceptibility profile of Salmonella spp. strains in the State of Rio de Janeiro are scarce. Therefore, the aim of this study was to detect the presence of Salmonella spp. in live chickens and carcasses in slaughterhouses of the State of Rio de Janeiro, to identify the serotypes and to evaluate the antimicrobial susceptibility of these strains for fluoroquinolones and beta-lactams. Sixty cloacal swabs samples from broiler chickens and sixty samples of carcasses from six slaughterhouses under State Inspection were collected. The isolates were serotyped and resistance was tested to eight antimicrobials: enrofloxacin, ciprofloxacin, norfloxacin, cephalothin, ceftiofur, cefotaxime, amoxicillin/clavulanic acid and ampicillin by disc diffusion method. The results showed a prevalence of Salmonella spp. of 1.66% (1/60) in cloacal swabs samples and 26.66% (16/60) in carcasses. In cloacal swabs sample only Senftenberg (1.66%) serotype was isolated. In total, seven different serotypes were obtained from carcasses: Senftenberg (15%), followed by Mbandaka (8.3%), Schwarzengrund (3.3%), Cerro (3.3%), Ohio (3.3%), Minnesota (1.66%) and Tennessee (1.66%). Regarding antimicrobial susceptibility, 29 (87.87%) isolates were sensitive to all antimicrobials tested and 4 (12.12%) isolates were resistant to three or more beta-lactams antimicrobials. No susceptibility to fluoroquinolones was observed. These results showed a prevalence of Salmonella spp. higher than expected in slaughterhouses in the State of Rio de Janeiro, besides the presence of several serotypes of Salmonella spp. The resistance found for beta-lactams alerts to the spread of these strains through the food chain.

Métodos de Superação da Dormência e Substratos para Germinação de Sementes de Tachigali vulgaris L.G. Silva & H. C. Lima

RESUMO O conhecimento das condições ideais como o método de superação da dormência e o substrato para que ocorra a germinação das sementes de espécies florestais é essencial para o sucesso no estabelecimento das plântulas. Este trabalho teve por objetivos definir o método adequado para a superação de dormência e avaliar o efeito de diferentes substratos na germinação de sementes de T. vulgaris. Foram testados os diferentes tratamentos pré-germinativos: testemunha; escarificação mecânica com lixa d’água; desponte do tegumento na região oposta ao hilo com auxílio de tesoura; imersão em água na temperatura de 80 °C por 24 horas e imersão das sementes em ácido sulfúrico durante 10 minutos. Após os tratamentos pré-germinativos, as sementes foram semeadas em substratos vermiculita, areia e papel mata-borrão e submetidas a 30 °C e luz contínua. O melhor tratamento foi o desponte do tegumento na região oposta ao hilo com tesoura e substrato vermiculita.
ABSTRACT The knowledge of how forest species plantules stablish successfully is essential to comprehend the conditions involving dormancy surpass and their ideal germination substrates. The present study aims to define the most efficient methods to surpass dormancy and to test different substrates that influence T. vulgaris germination. The pre-germination tests applied in this study, in addition to the control test, were: mechanic scarification (sandpaper number 100), scissor tegument cut (opposite to the hilum), 24 hour immersion in water (80 °C), and a ten minutes sulfuric acid immersion. After the pre-germinative treatments the seeds were sown in different substrates, such as: vermiculite, sand and filter paper, all of them submitted to constant light in a controlled temperature of 30 °C. The best result treatment was the scissor cut associated with vermiculite substrate.

Is there a relationship between complaints of impaired balance and postural control disorder in community-dwelling elderly women? A cross-sectional study with the use of posturography

Background: Risk of falls increases as age advances. Complaints of impaired balance are very common in the elderly age group. Objectives: The objective of this study was to investigate whether the subjective perception of impaired balance was associated with deficits in postural control (objective analysis) in elderly community-dwelling women. Method: Static posturography was used in two groups: elderly women with (WC group) and without (NC group) complaints of impaired balance. The area, mean sway amplitude and mean speed of the center of pressure (COP) in the anterior-posterior (AP) and medial-lateral (ML) directions were analyzed in three stances: single-leg stance, double-leg stance and tandem stance, with eyes open or closed on two different surfaces: stable (firm) and unstable (foam). A digital chronometer was activated to measure the time limit (Tlimit) in the single-leg stance. Kruskal-Wallis tests followed by Mann-Whitney tests, Friedman analyses followed by post hoc Wilcoxon tests and Bonferroni corrections, and Spearman statistical tests were used in the data analysis. Differences of p<0.05 were considered statistically significant. Results: The results of posturography variables revealed no differences between groups. The timed single-leg stance test revealed a shorter Tlimit in the left single-leg stance (p=0.01) in WC group compared to NC group. A negative correlation between posturography variables and Tlimit was detected. Conclusions: Posturography did not show any differences between the groups; however, the timed single-leg stance allowed the authors to observe differences in postural control performance between elderly women with and those without complaints of impaired balance.

BACKGROUND: muscle strength and power are two factors affecting balance. The impact of muscle strength and power on postural control has not been fully explored among different age strata over sixty. OBJECTIVES: the aim of the present study was to assess the muscle strength and power of elderly women in different age groups and determine their correlation with postural control. METHOD: eighty women were divided into four groups: the young 18-30 age group (n=20); the 60-64 age group (n=20); the 65-69 age group (n=20); and the 70-74 age group (n=20). The participants underwent maximum strength (one repetition maximum or 1-RM) and muscle power tests to assess the knee extensor and flexor muscles at 40%, 70%, and 90% 1-RM intensity. The time required by participants to recover their balance after disturbing their base of support was also assessed. RESULTS: the elderly women in the 60-64, 65-69, and 70-74 age groups exhibited similar muscle strength, power, and postural control (p>0.05); however, these values were lower than those of the young group (p<0.05) as expected. There was a correlation between muscle strength and power and the postural control performance (p<0.05). CONCLUSION: despite the age difference, elderly women aged 60 to 74 years exhibited similar abilities to generate strength and power with their lower limbs, and this ability could be one factor that explains the similar postural control shown by these women.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Morphological aspects and Cox-2 expression after exposure to 780-nm laser therapy in injured skeletal muscle: an in vivo study

Background: The effectiveness of low-level laser therapy in muscle regeneration is still not well known. Objective: To investigate the effects of laser irradiation during muscle healing. Method: For this purpose, 63 rats were distributed to 3 groups: non-irradiated control group (CG); group irradiated at 10 J/cm² (G10); and group irradiated at 50 J/cm² (G50). Each group was divided into 3 different subgroups (n=7), and on days 7, 14 and 21 post-injury the rats were sacrificed. Results: Seven days post-surgery, the CG showed destroyed zones and extensive myofibrillar degeneration. For both treated groups, the necrosis area was smaller compared to the CG. On day 14 post-injury, treated groups demonstrated better tissue organization, with newly formed muscle fibers compared to the CG. On the 21st day, the irradiated groups showed similar patterns of tissue repair, with improved muscle structure at the site of the injury, resembling uninjured muscle tissue organization. Regarding collagen deposition, the G10 showed an increase in collagen synthesis. In the last period evaluated, both treated groups showed statistically higher values in comparison with the CG. Furthermore, laser irradiation at 10 J/cm2 produced a down-regulation of cyclooxygenase 2 (Cox-2) immunoexpression on day 7 post-injury. Moreover, Cox-2 immunoexpression was decreased in both treated groups on day 14. Conclusions: Laser therapy at both fluencies stimulated muscle repair through the formation of new muscle fiber, increase in collagen synthesis, and down-regulation of Cox-2 expression.

CONTEXTUALIZAÇÃO: A velocidade de marcha parece estar relacionada com a capacidade aeróbica, força dos músculos dos membros inferiores e mobilidade funcional. Entretanto, não está claro se existe uma relação direta entre a melhora da força muscular e o desempenho da marcha em mulheres pós-menopausais recentes. OBJETIVO: Avaliar o efeito de exercícios de fortalecimento muscular sobre o desempenho no teste de caminhada de 6 minutos em mulheres nos primeiros cinco anos após a menopausa. MÉTODOS: As mulheres foram randomizadas em grupo controle (n=31), que não realizou exercício, e grupo exercício (n=27), que realizou um programa de exercícios. O programa de exercícios foi realizado duas vezes por semana, durante três meses, e consistiu em aquecimento, alongamento, exercícios de fortalecimento muscular para quadríceps, isquiotibiais, tríceps sural, tibial anterior, glúteo máximo e abdominais, finalizando com relaxamento. O treinamento de força iniciou com 60% de 1RM (duas séries de 10-15 repetições) e atingiu 85% (quatro séries de seis repetições) no final dos três meses. RESULTADOS: As comparações entre os grupos pré e pós-três meses não mostraram diferenças para distância percorrida, frequência cardíaca (FC) e pressão arterial (p≥0,05), mas mostraram diferenças para força muscular após três meses, com o grupo exercício apresentando maior força (p≤0,05). Nas comparações antes e após três meses intragrupos, houve diferenças para FC, forças musculares de quadríceps e isquiotibiais para o grupo intervenção. CONCLUSÃO: Os resultados sugerem que o treinamento de força muscular dos membros inferiores não melhora o teste de caminhada de 6 minutos para essa população de mulheres avaliadas.
BACKGROUND: Walking speed seems to be related to aerobic capacity, lower limb strength, and functional mobility, however it is not clear whether there is a direct relationship between improvement in muscle strength and gait performance in early postmenopausal women. OBJECTIVE: To evaluate the effect of muscle strengthening exercises on the performance of the 6-minute walk test in women within 5 years of menopause. METHODS: The women were randomized into control group (n=31), which performed no exercise, and exercise group (n=27), which performed muscle strengthening exercises. The exercises were performed twice a week for 3 months. The exercise protocol consisted of warm-up, stretching, and strengthening of the quadriceps, hamstring, calf, tibialis anterior, gluteus maximus, and abdominal muscles, followed by relaxation. Muscular strength training started with 60% of 1MR (2 series of 10-15 repetitions), reaching 85% until the end of the 3-month period (4 series of 6 repetitions each). RESULTS: The between-group comparisons pre- and post-intervention did not show any difference in distance walked, heart rate or blood pressure (p>0.05), but showed differences in muscle strength post-intervention, with the exercise group showing greater strength (p CONCLUSION: The results suggest that muscle strengthening of the lower limbs did not improve performance in the 6-minute walk test in this population of postmenopausal women.

INFLUÊNCIA DO ENVELHECIMENTO ACELERADO NO VIGOR DE SEMENTES DE Anadenanthera colubrina (Vellozo) Brenan - Mimosaceae

RESUMO A espécie Anadenanthera colubrina (Vellozo) Brenan é uma Mimosaceae, de porte arbóreo, utilizada na arborização de pastos bem como a madeira é empregada na confecção de tacos, ripas, embalagens, lenha e carvão de boa qualidade. O objetivo deste trabalho foi estudar a influência do teste de envelhecimento acelerado no vigor de sementes dessa espécie. O ensaio foi conduzido utilizando o método da câmara de envelhecimento, com temperatura de 40 ±2oC e umidade relativa de 95%, com tempo de permanência zero (T1); 24 (T2); 48 (T3); 72 (T4) e 96 horas (T5). O delineamento utilizado foi o inteiramente casualizado, com quatro repetições de 25 sementes, por tratamento. O teste de germinação foi realizado à temperatura de 30oC, com luz constante, tendo como substrato rolo de papel. A primeira contagem foi efetuada no quinto dia e a última no nono dia da instalação. A germinação das sementes frescas foi de 98% e o teor de água de 8,1%. Observou-se que a testemunha (T1), após 14 meses de armazenamento, apresentou germinação de 75% e os demais tratamentos (T2, T3, T4, T5) apresentaram percentuais variando entre 32 e 21%. O envelhecimento acelerado provocou a perda da viabilidade das sementes estudadas. Registrou-se baixa percentagem de plântulas normais e alta de sementes deterioradas. O envelhecimento acelerado provocou um declínio na velocidade de germinação das sementes da espécie.
ABSTRACT Anadenanthera colubrina (Vellozo) Brenan is a Mimosaceae tree used for pasture afforestation and its wood produces good quality parquet blocks, slats, packing, charcoal and firewood. This work aimed to study the species seed influence under accelerated aging treatments in order to evaluate seed vigour through germination test. The aging camera method was adopted with temperatures of 40 ± 2oC and humidity around 95%, with stay time zero (T1); 24 (T2); 48(T3); 72 (T4) and 96 hours (T5). Statistic design used was completely random, with four replicate and 25 seeds per treatment. Germination test was carried out under 30oC temperature and continuous light with paper filter moistened as substrate. First counting was conducted on the fifth day and the last on the ninth day after trial establishment. Fresh seed germination was 98% and water content was 8,1%. It was observed that control treatment (T1), after 14 months of storge showed 75% germination and the others treatments showed germination between 32 and 21%. Accelerated aging caused loss of viability on the observed seeds, low percentage of normal plants and high percentage of deteriorated seeds. Accelerated aging decreased the seed germination speed.
