Abstract Since the first reported case of COVID-19 in Brazil, the public and private educational system started to close. Up to November 2020, scientific discussions about the return of schooling activities have been rarely performed by the national scientific community and police-makers. The great delay of school returning in Brazil contrasts with successful international strategies of school reopening worldwide and seems counterintuitive with the reopening of non-essential activities. Here, important issues to be considered before and during school reopening are reviewed and discussed. COVID-19 testing is essential to avoid disease spreading, but high cost of individual RT-qPCRs impairs an extensive testing strategy for school returning. To reduce costs and increase the speed of diagnosis, we tested the efficiency of a pooled-sample PCR strategy in a cohort of the educational staff in the city of Macaé/RJ, finding five asymptomatic individuals (0,66%) among the 754 people tested. Thus, a polled-sample PCR testing strategy of the educational staff might prevent infection spreading in schools at a reasonable cost. We discuss how our test strategy could be coupled with internationally recognized safety rules to allow for a safe school return and how countries from different world regions are dealing with educational activities during COVID-19 pandemic.

Auxin independent1 (Axi1) as an endogenous gene for copy number determination in transgenic tobacco

Abstract Determining the copy number is important because it can greatly influence the expression level and genetic stability of transgenes. This study aimed to establish a methodology that can estimate the transgene copy number in tobacco (Nicotiana tabacum) using the Auxin independent1 (Axi1) gene as an endogenous control. Tobacco c.v. Petit Havana plants were transformed via Agrobacterium tumefaciens with a pMCG1005 vector that contained Bar as a selective gene. Oligonucleotide efficiency was determined and a qPCR using the 2-ΔCt and 2-ΔΔCt methods was performed. Bar was the target gene and Axi1 was the endogenous control in five transgenic tobacco events. The results showed that the Axi1 gene was donated by the maternal parent N. sylvestris when interspecific hybridization occurred between N. sylvestris and N. tomentosiformis. The copy number results agreed with the segregation ratios for the Bar gene in T1 plants, which confirmed that Axi1 is a single copy gene that can be used as an endogenous control.

RESUMO: Objetivou-se padronizar uma reação do tipo multiplex PCR (mPCR) para detectar Microsporum canis, Microsporum gypseum e o complexo Trichophyton mentagrophytes em amostras de pelos e/ou crostas de cães e gatos. 250 amostras de pelos e/ou crostas de cães e gatos foram analisadas por meio de exame direto e cultura, o DNA das mesmas foi extraído para mPCR. Primers foram desenhados e como controle positivo da reação utilizou-se o DNA extraído de colônias de M. canis (URM 6273), M. gypseum (URM 6921) e T. mentagrophytes (URM 6211), provenientes da Coleção de Culturas (Micoteca URM), Departamento de Micologia, Centro de Ciências Biológicas da Universidade Federal de Pernambuco (CCB/UFPE). Como controles negativos de reação, utilizou-se água destilada esterilizada e DNA extraído de Alternaria sp. para verificar a especificidade dos primers. Do total de amostras analisadas, 15 (6%) foram identificadas, em cultura, como dermatófitos, e destas, 10 foram M. canis, três M. gypseum e dois T. mentagrophytes (complexo). Destas 15 amostras positivas, 11 (73,3%) foram detectadas por meio da mPCR. Além destas, seis outras, negativas em cultura, foram identificadas como M. gypseum. Verificou-se uma boa concordância entre os resultados da cultura e mPCR (Kappa: 0,66). O protocolo padronizado neste estudo pode ser utilizado como um método de triagem, por apresentar uma sensibilidade maior que a da cultura, usado paralelamente aos exames de rotina, permitindo um diagnóstico em menor tempo.
ABSTRACT: The aim of this study was to standardize a multiplex PCR (mPCR) reaction to detect Microsporum canis, Microsporum gypseum and the Trichophyton mentagrophytes complex in dog and cat fur and/or crusts. 250 fur and/or crusts samples from dogs and cats were analyzed by direct examination and culture, DNA from them was extracted for mPCR. Primers were designed and the DNA extracted from colonies of M. canis (URM 6273), M. gypseum (URM 6921) and T. mentagrophytes (URM 6211) from the Collection of Cultures - URM Micoteca - Department of Mycology, Biological Sciences Center of the Federal University of Pernambuco (CCB / UFPE). As negative controls, sterile distilled water and DNA extracted from Alternaria sp., were used to verify the specificity of the primers. Of the total samples analyzed, 15 (6%) were identified in culture as dermatophytes, and of these, 10 were M. canis, three M. gypseum and two T. mentagrophytes (complex). Of these 15 positive samples, 11 (73.3%) were detected by mPCR. Besides these, six others, negative in culture, were identified as M. gypseum. There was good agreement between culture results and mPCR (Kappa: 0.66). The protocol standardized in this study can be used as a screening method, because it has a sensitivity greater than that of the culture, used in parallel to the routine exams, allowing a diagnosis in a shorter time.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

RESUMO O objetivo deste trabalho é descrever o conhecimento popular relacionado à espinheira-santa (Maytenus ilicifolia) entre erveiros e feirantes que comercializam a planta no centro de Pelotas, Rio Grande do Sul, Brasil. Foi realizada uma pesquisa de abordagem qualitativa, exploratória e descritiva, de agosto a outubro de 2013. Entrevistas semi-estruturadas, com questões sobre os saberes e práticas relacionados à espinheira-santa, foram aplicadas a cinco erveiros e três feirantes do mercado informal de plantas medicinais. Para a análise dos dados foi utilizada a proposta operativa de Minayo, sendo Capra o referencial teórico para este estudo. Foi verificado que o saber relacionado ao uso da espinheira-santa é transmitido de geração a geração, embora existam outras fontes de conhecimento. As indicações do uso popular da espinheira-santa com finalidade terapêutica estão relacionadas a distúrbios gástricos, cicatrização e depuração do sangue. As diferentes formas dos sujeitos deste estudo interagirem com o ambiente, com sua família e em outras relações interpessoais resultam em diferentes saberes relacionados à espinheira-santa.
ABSTRACT The aim of this study was to describe the popular knowledge related to the espinheira-santa (Maytenus ilicifolia) among herbalists and marketers that sell this plant in the center of Pelotas/Rio Grande do Sul State, Brazil. A qualitative, exploratory and descriptive research was performed between August and July of 2013. Semi-structured interviews, with questions regarding the knowledge and practices related to this plant, were applied to five herbalists and three marketers inserted in the informal market of medicinal plants. The operative proposal by Minayo was used in order to analyze the data, and the theoretical framework by Capra was used in the discussion. The popular knowledge related to the use of espinheira-santa is passed on from generation to generation, although nowadays there are other sources ofinformation. The popular clueswith therapeutic purposes are related to gastric disorders, healing and blood clearance. The study participants have different forms of interaction with the environment, the family and other interpersonal relationships, resulting in different popular knowledges related to this medicinal plant.

Dengue fever and schistosomiasis are major public health issues for which vector control using larvicide and molluscicide substances present in plants provides a promising strategy. This study evaluated the potential toxicity of the extract of hydroethanol Schinopsis brasiliensis Engl., Anacardiaceae, stem bark and its chloroform, hexane, ethyl acetate, and hydromethanol fractions against Artemia salina and Aedes Aegypti larvae and snails Biomphalaria glabrata. All of the assays were performed in triplicate and the mean mortality rates were used to determine the LC50and LC90 values using the probit method. The hydroethanol hydromethanol extract and fraction were free of toxicity towards A. salina(LC50 > 1000 µg/ml), while chloroform fraction was moderately toxic (LC50313 µg/ml); ethyl acetate and hexane fractions displayed low toxicity, with LC50 557 and 582 µg/ml, respectively. Chloroform, hexane, and ethyl acetate fractions showed larvicidal potential towards A. aegypti (LC50 values of 345, 527 and 583 µg/ml, respectively), while chloroform and ethyl acetate fractions were highly toxic to B. glabrata (LC90values of 68 and 73 µg/ml, respectively). Based on these findings, ethyl acetate, chloroform, and hexane fractions should be further investigated for their potential use against the vectors of dengue and schistosomiasis.

Objetivou-se relatar características morfológicas do bulbo ocular e determinar valores de referência para testes oftálmicos selecionados em corujas-orelhudas (Asio clamator). Foram estudados 32 olhos de 16 corujas (Asio clamator), adultas e jovens, machos e fêmeas, de vida livre. Sendo compilados dados referentes a observações morfológicas do crânio, bulbo ocular e anexos, além de mensuração de testes oftálmicos, incluindo, Teste Lacrimal de Schirmer (TLS), cultura da microbiota normal da conjuntiva, estesiometria, pressão intraocular (PIO), espessura de córnea central (ECC), diâmetro horizontal da rima palpebral, diâmetro horizontal da córnea e oftalmoscopia indireta. Vinte e dois tipos de bactérias foram identificados em 12 corujas havendo predominância de microrganismos Gram-positivos. A média encontrada para o TLS foi de 5,03±3,28mm/min, para o diâmetro horizontal da rima palpebral em 16 corujas foi 21,24±1,17mm, e 15,7±2,74mm para o diâmetro horizontal da córnea. O valor médio para o teste de estesiometria foi de 0,80±0,59cm, a PIO média de 13,81±5,62mmHg e ECC média de 0,28±0,03cm. O estudo contribuiu para a caracterização da morfologia ocular e para o estabelecimento de valores de referências de testes diagnósticos oftálmicos em corujas-orelhudas, sendo necessário ainda o desenvolvimento de estudos complementares sobre histologia ocular desta espécie.
The investigation aimed to describe the most relevant morphological characteristics of the eyes of the striped owl (Asio clamator) and determine normal values for selected ophthalmic diagnostic tests. A total of 32 eyes of 16 young and adult free-ranging healthy striped owls of both sexes were investigated. Data of different morphological observations of the skull, eye globe and adnexa, clinical tests and parameters were collected, including Schirmer Tear Test (STT), normal conjunctival bacterial microbiota, esthesiometry, intraocular pressure (IOP), central corneal thickness (CCT), horizontal palpebral fissure length, horizontal, corneal diameter and fundoscopy. Twenty two types of bacteria were identified in twelve owls, the most frequent bacteriae were the Gram-positive. The mean value found for the STT was 5.03±3.28mm/min; horizontal palpebral fissure lengths was 21.24±1.17mm, and 15.7±2.74mm for horizontal corneal diameter. The mean value for the esthesiometry was 0.80±0.59cm, IOP was 13.81±5.62mmHg and CCT was 0.28±0.03cm. This study contributes to establish reference values for selected ophthalmic diagnostic tests in the striped owl, as well as for morphological characterization of the eyeball of Asio clamator; but it is still necessary to characterize furthermore its histologic features.

Avaliou-se o efeito do flavonoide quercetina na replicação do parvovírus canino in vitro por meio do ensaio de determinação da atividade virucida (ensaio 1), ensaio de determinação da atividade sobre a célula (ensaio 2) e ensaio de tempo de adição das drogas em diferentes etapas do ciclo replicativo viral (ensaio 3). A quercetina apresentou significante atividade antiviral, com valores máximos de redução do título viral de 96,3% no ensaio 1, 90% no ensaio 2 e 90% no ensaio 3. Os efeitos mais expressivos ocorreram nas etapas de adsorção e penetração viral. Os resultados deste trabalho sugerem a importância da quercetina para a medicina veterinária.
The in vitro effect of the flavonoid quercetin against canine parvovirus was evaluated. The antiviral activity of quercetin was evaluated by determining the virucidal activity (assay 1), determining the activity on the cell (assay 2) and using the time of addition assay to test the inhibition of the viral replication cycle (assay 3). Quercetin showed a significant antiviral activity, with maximum viral titer reduction of 96.3% in assay 1, 90% in assay 2 and 90% in assay 3. The most expressive effects occurred in the stages of viral adsorption and penetration. The results show the importance of quercetin for veterinary medicine.

A salmonelose permanece uma importante causa de doença diarréica em humanos no Estado de São Paulo, Brasil. Nesse estudo foram identificados 3554 isolados de Salmonella de infecções humanas, durante o período 1996-2003. Entre os 68 diferentes sorovares determinados, S. Enteritidis foi predominante em infecções gastrointestinais e extra-intestinais, responsável por 67.4% de todos os isolados. S. Typhimurium e S. enterica subsp. enterica (4,5,12:i:-) foram mais freqüentemente isolados de crianças com idade de < 1 a 4 anos, em contraste, infecções por S. Enteritidis foram predominantes em pessoas entre 20 e 50 anos. A contínua vigilância laboratorial das salmoneloses, incluindo a sorotipagem, tem evidenciado os sorovares de Salmonella causando infecções humanas em nossa área geográfica ao longo do tempo.
Salmonellosis remains an important cause of diarrheal illness in humans in São Paulo State, Brazil. In this study were identified 3554 Salmonella isolates from human infections, during the period 1996-2003. Among 68 different serovars determined, S. Enteritidis was the most frequent one in gastrointestinal and extra-intestinal infections accounting for 67.4% of all isolates. S. Typhimurium and S. enterica subsp. enterica (4,5,12:i:-) were most frequently isolated from children aged < 1-4 year-old, in contrast, people with S. Enteritidis infections were most likely to be 20-50 year-old. In our geographic area the continued laboratorial surveillance of salmonellosis, including serotyping, has showed the trends in Salmonella serovars causing infections in humans throughout the time.

O objetivo deste experimento foi avaliar o desempenho de coelhos em crescimento criados em diferentes densidades populacionais. Foram utilizados 136 coelhos, não sexados, desmamados aos 35 dias de idade, em delineamento em blocos ao acaso, com quatro tratamentos e quatro repetições. As densidades avaliadas foram 11,67; 13,34; 15 e 16,67 coelhos/m², correspondentes à ocupação de sete, oito, nove e dez coelhos/gaiola. O período experimental foi de seis semanas. Os coelhos e a ração foram pesados para obtenção do peso final, ganho diário de peso, consumo diário de ração, conversão alimentar e produção de carne total ou kg de carne por m². Não houve diferença (P>0,05) entre densidade quanto ao peso final, ganho diário de peso, consumo diário de ração e conversão alimentar. A produção em quilos de carne por metro quadrado aumentou (P<0,05) linearmente com a densidade (27,86; 31,45; 34,64 e 38,12 kg/m²). O aumento da densidade populacional não influiu no desempenho dos coelhos.
The purpose of this experiment was to evaluate the performance of growing rabbits in different stocking densities. One hundred and thirty six non-sexed rabbits, 35 days old, were used in a randomized block design, with four treatments and four repetitions. The evaluated densities were: 11.67; 13.34; 15 e 16.67 rabbits/m² corresponding to the occupation of seven, eight, nine and ten rabbits/cage. At the end of the six-week-experimental-period, rabbits and rations were weighted to obtain final body weight, daily weight gain, daily ration consumption, feed:gain ratio and total meat production or kilograms of meat per square meter. No effects (P>0.05) of stocking densities on final body weight, daily weight gain, daily ration consumption and feed:gain ratio were observed. The production in kilograms of meat per square meter increased linearly (P<0.05) in relation to stocking density (27.86; 31.45; 34.64 and 38.12 kg/m²). The increase in stocking density did not affect the performance of the rabbits.
