The search for compounds with affinity for both mu-opioid receptor (MOR) and sigma-1 receptor (σ1R) is one of the innovative directions to develop painkillers with reduced side effects. Additionally, triazole scaffolds have been extensively explored in the last two decades in medicinal chemistry. In this context, we synthesized a series of new triazole fentanyl derivatives and evaluated their affinity for both MOR and σ1R. The binding affinity of the compounds for human MOR was determined in competitive radioligand binding assays, using fentanyl as standard. For the assays with σ1R, a σ1R agonist (SKF10047) was employed. The most active analogue was 6d which moderately binds to MOR with half-maximal inhibitory concentrations (IC50) = 1.9 µM and to σ1R with IC50 = 6.9 µM. Molecular docking calculations were carried out, providing a structural elucidation for the observed values of affinity. Absorption, distribution, metabolism, and excretion toxicity (ADMET) parameters for the new compounds were simulated with the SwissADME tool.

Uma série de treze derivados triazólicos contendo grupos benzila-halogenados foi sintetizada utilizando-se como etapa chave a cicloadição azida-alcino catalisada por Cu(I) (CuAAC), transformação comumente descrita como reação click. A atividade biológica destes compostos foi avaliada, e verificou-se que estes compostos interferem na germinação e no crescimento radicular (brotos e raízes) das espécies Allium cepa (cebola), modelo de monocotiledônea, e Cucumis sativus (pepino) e Lactuca sativa (alface), modelos de dicotiledôneas. Os compostos apresentaram atividade predominantemente inibitória com relação às espécies avaliadas principalmente na concentração de 10-4 mol L-1, sendo que alguns deles foram tão ativos quanto o 2,4-D (ácido 2,4-diclorofenoxiacético), o controle positivo.

Thirteen triazole derivatives bearing halogenated benzyl substituents were synthesized using the Cu-catalyzed azide-alkyne cycloaddition (CuAAC), a leading example of the click chemistry approach, as the key step. The biological activity of the compounds was evaluated, and it was found that these compounds interfere with the germination and radicle growth (shoots and roots) of two dicotyledonous species, Lactuca sativa and Cucumis sativus, and one monocotyledonous species, Allium cepa. The compounds showed predominantly inhibitory activity related to the evaluated species mainly at the concentration of 10-4 mol L-1. Some of them presented inhibitory activity comparable to 2,4-D (2,4-dichlorophenoxyacetic acid), used as positive control.

Reações de 4-O-alil-2,3-di-O-benzil-6-desoxi-6-(3-iodobenzoilamino)-α-D-galactopiranosídeo de metila, seu epímero glico, 2,3-di-O-benzil-6-desoxi-6-(3-iodobenzoilamino)-4-O-(1-pentenil)-α-D-glicopiranosídeo de metila e seu regioisômero orto-iodobenzamida com hidreto de tri-n-butilestanho foram realizadas em diferentes condições. Dependendo das condições, os três precursores que contêm o átomo de iodo em posição meta conduziram a uma surpreendente quantidade de compostos bifenílicos, nos quais o átomo de iodo foi substituído por um grupo fenila. O substrato 2-iodobenzamida levou apenas ao produto de hidrogenólise. Nenhum produto ciclizado foi isolado de nenhuma das reações. As estruturas dos compostos bifenílicos inéditos foram elucidadas utilizando-se as espectrometrias de massa, de RMN ¹H e 13C e experimentos DEPT, COSY, HMQC e HMBC. Foram propostos mecanismos de formação dos compostos bifenílicos e hipóteses para explicar os diferentes comportamentos dos isômeros orto- e meta-iodobenzamidas na reação radicalar.

Reactions of methyl 4-O-allyl-2,3-di-O-benzyl-6-deoxy-6-(3-iodobenzoylamino)-α-D-galactopyranoside, its gluco epimer, methyl 2,3-di-O-benzyl-6-deoxy-6-(3-iodobenzoylamino)-4-O-(1-pentenyl)-α-D-glucopyranoside and its ortho-regioisomer with tri-n-butyltin hydride were performed in different conditions. Depending on reaction conditions the three meta-iodo isomers gave a surprising amount of biphenyl compounds. The 2-iodo isomer led only to the undesired but expected hydrogenolysis product. No cyclized products were isolated in all the reactions. The structures of the new biphenyl products were elucidated by ¹H and 13C NMR spectroscopy, DEPT, COSY, HMQC and HMBC experiments and ESI-MS/MS. Mechanisms for the formation of these new biphenyl derivatives and hypotheses to explain the different outcomes for radical reactions of 3- or 2-iodobenzamides were presented.

Species from genus Aspidosperma (Apocynaceae) are popularly employed to treat various diseases. This genus is characterized by the occurrence of indole alkaloids. Taking into account the various biological activities attributed to these alkaloids, the description of the chemical diversity in genus Aspidosperma is important. A review of simple carbolinic alkaloids isolated from species of various genera was published in 1979. In 1987, it was published another one dealing with the relationships between the chemical structures of the indole alkaloids and the evolution of Aspidosperma species. This work updates the information about the indole alkaloids isolated from Aspidosperma species.

A reação radicalar mediada por hidreto de tri-n-butilestanho de 2,3-di-O-benzil-4-O-trans-cinamil-6-desoxi-6-(2-iodobenzoilamino)- alfa-D-galactopyranosídeo de metila levou à formação de um produto inesperado, o 2,3-di-O-benzil-4-O-trans-cinamil-6-desoxi-6-(2-tri- n-butilestanhobenzoilamino)-alfa-D-galactopyranosídeo de metila. A estrutura do produto foi elucidada por meio das espectrometrias de RMN unidimensionais (¹H, 13C e DEPT) e bidimensionais (COSY e HMQC) e confirmada por espectrometria de massas. Foram apresentadas propostas de mecanismos para explicar a formação do derivado organoestanho.

The tri-n-butyltin hydride-mediated reaction of methyl 2,3-di-O-benzyl-4-O-trans-cinnamyl -6-deoxy-6-(2-iodobenzoylamino)-alpha-D-galactopyranoside afforded an unexpected aryltributyltin compound. The structure of this new tetraorganotin(IV) product has been elucidated by ¹H, 13C NMR spectroscopy, COSY and HMQC experiments and electrospray ionization mass spectrometry (ESI-MS). The formation of this new compound via a radical coupling reaction and a radical addition-elimination process is discussed.
