Sperm cryopreservation of Prochilodus brevis using different concentrations of non-permeable cryoprotectants

Abstract The action of substances with non-permeable cryoprotectant potential, besides glucose, has not yet been studied for the species Prochilodus brevis. The objective of this work was to evaluate the action of four non-permeable cryoprotectants on this species sperm cryopreservation. Five pools were cryopreserved in a solution of 5% glucose and 10% dimethyl sulfoxide (Me2SO) associated or not (control) with cryoprotectants egg yolk (5, 10 or 12%), soy lecithin (2.5, 7.5 or 10%), sucrose (5, 10 or 20%) and lactose (5, 8 or 15%). After thawing, samples were evaluated for sperm kinetics (total motility, motility duration, velocities, and wobble - WOB), morphology and membrane and DNA integrity. The treatments containing egg yolk improved significantly (P<0.05) results when compared the control for the membrane integrity parameter. When compared to other treatments, egg yolk, at any concentration, presented higher results (P<0.05) for membrane integrity, total motility, curvilinear velocity (VCL) and average path velocity (VAP) parameters. Egg yolk also showed the best results for WOB, but it did not differ from 5% and 8% lactose and 5% and 20% sucrose. Soy lecithin had the lowest percentages of morphologically normal sperm (P<0.05), while the other treatments did not differ from each other. There was no difference regarding DNA integrity data. Thus, 5% egg yolk is indicated as a non-permeable cryoprotectant for P. brevis, in association with 5% glucose and 10% Me2SO.

Resumo Os óleos essenciais (OE) de Illicium verum e Pelargonium graveolens foram avaliados quanto à letalidade, inibição do desenvolvimento e eficácia residual contra a pulga Ctenocephalides felis felis. Sua composição química foi caracterizada por meio de cromatografia gasosa com detector de ionização de chama e espectrometria de massas. A mortalidade entre os diferentes estágios imaturos e pulgas adultas foi avaliada por meio de testes in vitro em papel filtro, contendo diferentes concentrações de OEs. A caracterização química do óleo volátil de I. verum mostrou que o E-anetol (79,96%) foi o constituinte majoritário, enquanto os principais compostos de P. graveolens foram citronelol (29,67%) e geraniol (14,85%). Foi observada atividade inseticida contra os estágios imaturos e adulto da pulga. O OE de I. verum teve atividade inseticida por aproximadamente 18 dias, enquanto o de P. graveolens durou 13 dias. A atividade pulicida de I. verum permaneceu acima de 70% até o 9º dia, enquanto a atividade de P. graveolens foi de 41,7% até o 2º dia. Os óleos essenciais, principalmente de I. verum, apresentam atividade inseticida para o controle de pulgas em diferentes estágios do ciclo de vida e têm potencial para o desenvolvimento de ectoparasiticidas (biopesticidas) de uso veterinário.
Abstract The essential oils (EOs) of Illicium verum and Pelargonium graveolens were evaluated for lethality, inhibition of development and residual efficacy against the flea Ctenocephalides felis felis. Their chemical composition was characterized by means of gas chromatography with a flame ionization and mass spectrometry detection. Mortality at different immature stages and among adult fleas was measured through in vitro filter paper tests at different concentrations of EOs. The chemical characterization of I. verum volatile oil showed that E-anethole (79.96%) was the major constituent, while the major compounds in P. graveolens were citronellol (29.67%) and geraniol (14.85%). Insecticidal activity against both immature and adult flea stages were observed. The EO of I. verum had insecticidal activity for approximately 18 days, while the EO activity of P. graveolens lasted for 13 days. The pulicidal activity of I. verum remained above 70% for up to 9 days, while the activity of P. graveolens was 41.7% for up to 2 days. Essential oils, especially that of I. verum, showed insecticidal activity for flea control at different life cycle stages and have potential for the development of ectoparasiticides (biopesticides) for veterinary use.

ABSTRACT In recent years, the number of cases with severe Plasmodium vivax malaria has shown an increasing trend. It is, therefore, important to identify routine laboratory markers that best characterize the acute disease phase and can serve as a tool for clinical follow-up of patients. In a cohort study, we followed 87 patients with acute P. vivax monoinfection acquired in an endemic region of the Brazilian Amazon. Forty-two different biochemical and hematological parameters frequently tested in clinical routine were evaluated at the acute phase and the convalescent phase. A total of 42 laboratory tests were performed: biochemical parameters measured were serum lipids levels, aminotransferases, bilirubin, amylase, glucose, urea, creatinine, albumin, globulin, uric acid, C-reactive protein, and alpha-1-acid glycoprotein. Hematological parameters included total and differential white blood cell and platelet counts, hemoglobin concentration, mean platelet volume, platelet width distribution, and plateletcrit. Our results show that several biochemical and hematological parameters were associated with acute phase P. vivax malaria and these parameters reverted to normal values in the convalescent phase. The use of these parameters during diagnosis and follow-up of the infection is a useful clinical tool to evaluate the clinical course and therapeutic response of patients with uncomplicated vivax malaria.

Abstract Periodontal disease is considered the main oral cavity disorder in dogs. Essential oils have the potential for use in the prevention and treatment of oral diseases. The antimicrobial activity of Schinus molle L. essential oil (SMEO) has already been reported. Chitosan, a natural product with antimicrobial activity and good biocompatibility has potential in biodental applications. In this study, we evaluated the in vitro antimicrobial activity of SMEO against bacteria associated with periodontal disease in dogs, developed and evaluated the physicochemical properties of a novel chitosan-based buccal delivery system containing SMEO. SMEO showed antimicrobial activity against Gram positive and Gram negative bacteria associated with canine periodontitis, with MIC values of 750 µg.mL-1 for Staphylococcus spp. and Streptococcus spp, 1000 µg.mL-1 for Corynebacterium spp. and 1250 µg.mL-1 for Pseudomonas spp. All formulations evaluated presented adequate physicochemical properties, good stability, and pH values close to buccal pH (5.0–7.0). Chitosan gel loaded with SMEO showed potential as a SMEO delivery system, having the ideal physicochemical and rheological properties (pseudoplastic and apparent viscosities) required for application on buccal tissue. Thus, we can conclude that formulation has the potential to be used for buccal mucosa delivery in the prevention and treatment of periodontal disease in dogs.

Abstract Contamination of water sources due to herbicide is of great concern. Clomazone is a pesticide with a high contamination potential which could possibility lixiviate to water streams. Changes caused by residual herbicide include flora modifications which are generally detrimental for some species. The lack of morphological studies performed in aquatic plants exposed to herbicide-contaminated environments has encouraged the development of our research. For the first time, we present a study that aimed to evaluate leaf injuries visible to the naked eye as well as microscopical effects which may be caused by clomazone on Pistia stratiotes. Pistia stratiotes was subjected to five concentrations of clomazone. Our analysis showed leaf injuries, especially after 15 days of clomazone application. Hormesis was observed when the water lettuce was subjected to the lower concentrations. Total leaf area showed increase following by reduction while injured until reaching the highest concentration. Although the concentrations of clomazone tested in our study are not lethal to water lettuce, such herbicide have still caused morphoanatomical damages on leaves which advocates for the use of P. stratiotes as a bioindicator of the presence of herbicides such as clomazone in water.

Abstract The molecular basis of anhydrobiosis, the state of suspended animation entered by some species during extreme desiccation, is still poorly understood despite a number of transcriptome and proteome studies. We therefore conducted functional screening by RNA interference (RNAi) for genes involved in anhydrobiosis in the holo-anhydrobiotic nematode Panagrolaimus superbus. A new method of survival analysis, based on staining, and proof-of-principle RNAi experiments confirmed a role for genes involved in oxidative stress tolerance, while a novel medium-scale RNAi workflow identified a further 40 anhydrobiosis-associated genes, including several involved in proteostasis, DNA repair and signal transduction pathways. This suggests that multiple genes contribute to anhydrobiosis in P. superbus.

Abstract The development of a biodegradable material with antimicrobial properties for local applications is required in the prevention and treatment of infectious diseases. The objective of this study was to produce blends of poly-L-lactide acid (PLLA) synthetic polymer associated with several antimicrobials, as an alternative in the prevention and treatment of infections, as well as to evaluate its cytotoxicity, release of antimicrobials and inhibit bacteria growth. Blends of PLLA added with 20% Amoxicillin, Metronidazole, Clindamycin or Azithromicyn were used to produce Films (F) or Meshs (M) by casting and electrospinning methods, respectively. Standardized discs of the films and meshs were stored in buffer solutions (pH 5 or 7.4) and aliquots were analyzed by high performance chromatography (HPLC) during 168 hours. Cytotoxicity on human gingival fibroblasts was tested after 24, 48 and 72h by MTT reaction. The antimicrobial capacity was determined against P. gingivalis and S. pyogenes. The specimens were weighed after 3 and 6 months of storage for degradation analysis. SEM was performed to control interfaces and degradation. Antimicrobials presented a continuous and exponential drug release. Analysis showed that both M and F were able to inhibit S. pyogenes and P. gingivalis growth, indicating the release of active antimicrobial agents. The products were not toxic to the fibroblasts. Amoxicillin-film showed more degradation than PLLA at both pHs (p < 0.05), whereas Azithromycin-meshes were more degraded than PLLA at pH 7.4 (p < 0.05). PLLA association with antimicrobials is biocompatible and may represent a potential tool for the local delivery of antimicrobials.

Resumo: O presente estudo teve por objetivo avaliar os efeitos da heparina sódica e Na2EDTA no sangue de Piaractus mesopotamicus (360,7±42,4g, 26,4±1,0cm). Foram amostrados vinte peixes em dois ensaios experimentais, sendo dez peixes utilizados para análise da fragilidade dos eritrócitos e dez peixes para análise dos parâmetros hematológicos e estudo bioquímico do plasma. O sangue coletado por punção veno-caudal foi aliquotado e armazenado em diferentes soluções anticoagulantes: Na2EDTA 10%, Na2EDTA 3%, heparina sódica 5.000 UI e heparina sódica 100 UI. Níveis plasmáticos de cálcio apresentados nas amostras armazenados em Na2EDTA diminuíram cerca de 80% em relação aos dois grupos armazenados com heparina. Amostras de sangue de pacus armazenados com Na2EDTA demonstraram aumento do hematócrito e VCM, e diminuição na CHCM. O efeito dose-resposta foi observado neste estudo. Estes resultados são reforçados pelos níveis mais elevados de proteína plasmática e hemólise apresentado no sangue armazenado com Na2EDTA 10%, o que confirma o efeito deletério desse tratamento anticoagulante na qualidade de amostras de sangue. Na2EDTA não é indicada para armazenar amostras de sangue de P. mesopotamicus, e heparina sódica a 100 UI é o anticoagulante mais recomendado, uma vez que este tratamento apresentou a menor taxa de alterações no sangue armazenado.
Abstract: The present study aimed to evaluate the effects of Na heparin and Na2EDTA on blood of Piaractus mesopotamicus (360.7±42.4g, 26.4±1.0cm). Twenty fishes were sampled in two experiment trials, ten for erythrocyte fragility analysis and ten for hematologic and plasma biochemical study. The blood collected by venous-caudal puncture was fractioned and stored in anticoagulants solution: Na2EDTA 10%, Na2EDTA 3%, Na heparin 5000 IU and Na heparin 100 IU. Plasmatic levels of calcium presented in the Na2EDTA stored samples were about 80% lower than both heparin groups. Blood samples of P. mesopotamicus stored with Na2EDTA demonstrated increase in the hematocrit and MCV, and decrease in MCHC. The dose-response effect was observed in this study. The results are reinforced by the higher levels of plasmatic protein and hemolysis presented in the Na2EDTA 10% stored blood, confirming the deleterious effect of this anticoagulant treatment on the quality of blood samples. Na2EDTA is not indicated to store P. mesopotamicus blood samples, but sodium heparin at 100 IU is the most recommended anticoagulant, since this treatment presented the lower rate of alterations in the stored blood.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

As elevadas taxas de mortalidade associadas com episódios de candidemia e a emergência da resistência aos antifúngicos, requerem o monitoramento da suscetibilidade de Candida spp., isoladas das candidemias, frente aos agentes antifúngicos. Os novos breakpoints, chamados “espécie-específicos,” foram recentemente aprovados (M27-S4) requerendo, pois, cuidadosa interpretação e comparações com aqueles até agora utilizados (M27-A3); ambos são propostos pelo Clinical Laboratory Standard Institute (CLSI). O presente estudo avaliou a suscetibilidade de espécies de Candida isoladas de candidemias baseando-se nestes dois breakpoints. Quatrocentos e vinte e dois isolados de Candida foram identificados e assim distribuídos: C. parapsilosis (48,68%), C. albicans (35,78%), C. tropicalis (9,71%), C. glabrata (3,55%), C. lusitaniae (1,65%), C. guilliermondii (1,65%), C. krusei (0,94%). Com base nos critérios do M27-A3, um total de 33 (7,81%) isolados foram julgados não-sensíveis, dos quais 16 (3,79%) como resistentes aos antifúngicos. De acordo com os breakpoints espécie-específicos (M27-S4) um total de 80 (18,95%) isolados foram considerados não-sensíveis, dos quais 10 (2,36%) resistentes a algum dos antifúngicos testados. Com base nos novos breakpoints espécie-específicos, o número de isolados não-sensíveis foi 2,4 vezes maior do que o número de não-sensíveis detectado pelos breakpoints do documento M27-A3. A detecção de um elevado número de isolados não-sensíveis através dos breakpoints propostos pelo M27-S4 destaca a importância dos testes de suscetibilidade, os quais trarão impactos no reconhecimento de isolados de Candida spp. não-sensíveis em episódios de candidemias, requerendo, portanto, continua avaliação.
The high mortality rates associated with candidemia episodes and the emergence of resistance to antifungal agents necessitate the monitoring of the susceptibility of fungal isolates to antifungal treatments. The new, recently approved, species-specific clinical breakpoints (SS-CBPs)(M27-S4) for evaluating susceptibility require careful interpretation and comparison with the former proposals made using the M27-A3 breakpoints, both from CLSI. This study evaluated the susceptibility of the different species of Candida that were isolated from candidemias based on these two clinical breakpoints. Four hundred and twenty-two isolates were identified and, among them, C. parapsilosis comprised 46.68%, followed by C. albicans (35.78%), C. tropicalis (9.71%), C. glabrata (3.55%), C. lusitaniae (1.65%), C. guilliermondii (1.65%) and C. krusei (0.94%). In accordance with the M27-A3 criteria, 33 (7.81%) non-susceptible isolates were identified, of which 16 (3.79%) were resistant to antifungal agents. According to SS-CBPs, 80 (18.95%) isolates were non-susceptible, and 10 (2.36%) of these were drug resistant. When the total number of non-susceptible isolates was considered, the new SS-CBPs detected 2.4 times the number of isolates that were detected using the M27-A3 interpretative criteria. In conclusion, the detection of an elevated number of non-susceptible species has highlighted the relevance of evaluating susceptibility tests using new, species-specific clinical breakpoints (SS-CBPs), which could impact the profile of non-susceptible Candida spp. to antifungal agents that require continuous susceptibility monitoring.

Statistical distribution models for migratory fish in Jacuí basin, South Brazil

O objetivo do presente estudo foi identificar o padrão de distribuição de peixes migradores da bacia hidrográfica do rio Jacuí (Rio Grande do Sul, Sul do Brasil), propondo um modelo matemático de distribuição presumida baseado em parâmetros ambientais geomorfológicos. Através de mapas de probabilidade de ocorrência, espera-se contribuir para a tomada de decisões relacionadas ao gerenciamento desta bacia hidrográfica. As espécies analisadas foram: Salminus brasiliensis (dourado), Leporinus obtusidens (piava), Prochilodus lineatus (grumatã) e Pimelodus pintado (pintado). As amostras foram obtidas a partir de entrevistas com pescadores e moradores locais, percorrendo-se a calha principal dos rios Jacuí, Taquari-Antas, Vacacaí, Vacacaí-Mirim, Pardo, Pardinho, Sinos e Caí. O programa de amostragens resultou em 204 entrevistas, sendo 187 consideradas como válidas em 155 pontos diferenciados. A probabilidade de ocorrência de peixes migradores foi ajustada utilizando-se a rotina LOGIT do software Idrisi Andes: P = e(b0 + b1 . altitude + b2 . área de bacia) . (1 + e(b0 + b1 . altitude + b2 . área de bacia))-1; onde P é a probabilidade de ocorrência da espécie (0-1) e b0, b1 e b2 são os parâmetros da equação. Pimelodus pintado foi citado como presente em 121 pontos dentre os 155 amostrados (78.06%), Prochilodus lineatus em 72 (46.45%), L. obtusidens em 62 (40.00%), e S. brasiliensis em 58 pontos (37.42%). A precisão do modelo, para a presença estimada, ficou entre 82% e 93%. Os parâmetros estimados da equação são descritos a seguir: S. brasiliensis: b0= -2.8762 ± 0.2597; b1= -1.3028 ± 0.0332; b2= 1.1487 ± 0.0301; L. obtusidens: b0 = -0.8364 ± 0.2213; b1 = -1.5564 ± 0.0462; b2 = 0.9947 ± 0.0206; Prochilodus lineatus: b0 = 0; b1 = -1.3067 ± 0.0544; b2 = 0.8128 ± 0.0177; Pimelodus pintado: b0 = -0.9487 ± 0.3688; b1 = -0.8269 ± 0.0496; b2 = 0.9255 ± 0.0304.
The aim of the present study was to identify the distribution patterns of migratory fishes in the Jacuí river basin (Rio Grande do Sul, South Brazil), proposing a statistical model of presumed distribution based on geomorphologic environmental data. Through maps of occurrence probability, we hope to contribute to decisions regarding basin environmental management. The analyzed species were: Salminus brasiliensis (dourado), Leporinus obtusidens (piava), Prochilodus lineatus (grumatã) and Pimelodus pintado (pintado). Samples were made through interviews with fishermen and local inhabitants, covering the main channel and tributaries of the rivers Jacuí, Taquari-Antas, Vacacaí, Vacacaí-Mirim, Pardo, Pardinho, Sinos, and Caí. The sampling program resulted in 204 interviews, being 187 considered as valid in 155 different sampling points. The probability of migratory fish occurrence was adjusted through the LOGIT routine of the Idrisi Andes Software: P = e(b0 + b1 . altitude + b2 . basin area) . (1 + e(b0 + b1 . altitude + b2 . basin area))-1, where P is the occurrence probability of the species (0-1) and b0, b1 and b2 are the equation parameters. Model accuracy, for estimating presence, ranged from 82% to 93%. Pimelodus pintado was cited to occur in 121 points among the 155 sampled (78.06%), Prochilodus lineatus in 72 (46.45%), L. obtusidens in 62 (40.00%) and S. brasiliensis in 58 (37.42%). Estado nutricional e prevalência de anemia em crianças que freqüentam creches em Belo Horizonte, Minas Gerais

OBJETIVO: Avaliar o estado nutricional e a prevalência de anemia em crianças de sete a 74 meses, que freqüentam creches. MÉTODOS: Estudo transversal em 25 creches da regional leste de Belo Horizonte, Minas Gerais. Realizaram-se avaliação antropométrica e dosagem de hemoglobina por hemoglobinômetro portátil. Considerou-se anemia se hemoglobina <11,0g/dL para crianças menores que 60 meses e <11,5g/dL, para maiores de 5 anos. Na avaliação do estado nutricional, utilizaram-se os índices peso/idade, estatura/idade e peso/estatura, sendo categorizados em três intervalos: abaixo de -2 escores Z (desnutrição/baixa estatura), de -2 a -1 escore Z (risco para desnutrição/baixa estatura) e maior ou igual a -1 escore Z. A análise estatística incluiu: qui-quadrado, qui-quadrado de tendência linear e análise de variância (ANOVA) seguida pelo teste de Tukey, sendo significante p<0,05. RESULTADOS: Foram avaliadas 402 crianças, com mediana de idade de 46,3 meses. A prevalência de anemia foi 28,8%, observando-se o dobro da prevalência nas crianças com idade inferior a 24 meses (70,4%), comparada às de faixa etária maior. A prevalência de desnutrição correspondeu a 5,0 e 5,5% da população para os índices peso/estatura e peso/idade, respectivamente. A prevalência de baixa estatura foi 4,2%. Em crianças menores de 5 anos, a anemia associou-se à idade e ao deficit estatural. CONCLUSÕES: A anemia em crianças matriculadas nas creches da regional leste de Belo Horizonte constitui importante problema de saúde pública, acometendo, principalmente, os menores de 24 meses. Os deficits nutricionais estiveram acima dos valores esperados.
OBJECTIVE: Assess the prevalence of anemia and the nutritional status of children with 7-74 months old, enrolled in day care centers. METHODS: This cross-sectional study enrolled children of 25 day care centers of the eastern district of Belo Horizonte city, Minas Gerais, Brazil. Anthropometric parameters were evaluated and hemoglobin count was done by a portable hemoglobinometer. The cutoff level for anemia was 11.0g/dL for children less than 60 months, and 11.5g/dL for those older than 5 years old. Nutritional status was assessed by anthropometric indexes: weight-for-age, height-for-age and weight-for-height and classified in 3 groups: below -2 Z score (malnutrition/height deficit), -2 to -1 Z score (nutritional risk for malnutrition/height deficit) and above or equal to -1 Z score. Statistical analysis included: chi-square, chi-square with linear trend, analysis of variance (ANOVA) followed by Tukey test, being significant p<0.05. RESULTS: 402 children, with a median age of 46.3 months, were assessed. The overall anemia prevalence was 28.8%. The rate of anemia in children <24 months (70.4%) was twice that observed in older children. The prevalence of malnutrition was 5,0 and 5.5%, as defined by weight-for-height and weight-for-age indexes, respectively. The prevalence of height deficit was 4.2%. Malnutrition was associated to age and height deficit in children <5 years old. CONCLUSIONS: Anemia in children enrolled in day care centers in Belo Horizonte is a serious public health problem, especially in those younger than 24 months. Nutritional deficits were beyond the expected values.
