LIQUID-LIQUID EQUILIBRIA DATA OF N-METHYL-2-HYDROXYETHYL AMMONIUM ALKYLATES WITH BUTANOL+WATER AND PENTANOL+WATER AT 293.15K, 313.15K AND 333.15K

Abstract Ionic liquids (ILs) are organic salts with low volatility and have different applications including organic synthesis, separation and extraction processes, fuel cell electrolyte, surfactants, among others. Protic ILs based on the N-methyl-2-hydroxyethyl ammonium cation and short chain carboxylic anions have simple synthesis and purification. However, as new substances few data are available for their thermodynamic properties and phase equilibria. In order to investigate the partition of these substances in alcohol-water, liquid-liquid equilibria data were determined experimentally for three ionic liquids (formed by the cation N-methyl-2-hydroxyethyl ammonium (m-2HEA) and the anions propionate (Pr), butanoate (B) and pentanoate (P)) with butanol and pentanol, at three different temperatures: 293.15K, 313.15K and 333.15K. The binodal curves are obtained by the turbidity method with gravimetric determination of all the components, curves are fitted using the Merchuk method. The tie-lines are also gravimetrically determined. The results confirm the high hydrophilicity of these ionic liquids, with partition coefficients of water-butanol and water-pentanol lower than 1. Data were correlated with UNIFAC.

The Use of Near Infrared Spectroscopy and Multivariate Calibration for Determining the Active Principle of Olanzapine in a Pharmaceutical Formulation

The aim of this study was to quantitatively determine the olanzapine in a pharmaceutical formulation for assessing the potentiality of near infrared spectroscopy (NIR) combined with partial least squares (PLS) regression. The method was developed with samples based on a commercial formulation containing olanzapine and seven excipients. Laboratory and commercial samples (n = 27 and 18, respectively) were used by defining the calibration and prediction sets. The method was validated in the range from 1.0 to 12.5 of olanzapine per 100 mg of powder (average mass 210 mg), by accuracy, precision, linearity, analytical sensitivity, limit of detection and quantification. The multivariate model developed for olanzapine was based on PLS and the determination coefficient (rc and rp), with the root mean square error of calibration and prediction being 0.95, 0.93, 3.2 × 10-3 and 4.0 × 10-3% m/m, respectively. The proposed NIR method is an effective alternative for quantification of olanzapine in the pharmaceutical industry.

Uma das chaves para manter a melhoria da alta qualidade de fabricação está no uso de cartas de controle. Neste trabalho, cartas de controle multivariada baseadas nos escores da componente principal mais significativa e no sinal analítico líquido foram desenvolvidas para monitoramento simultâneo da qualidade de dois princípios ativos (isoniazida e rifampicina) numa formulação farmacêutica (amostras produzidas no laboratório e na produção) empregando um espectrômetro portátil na região do infravermelho próximo. Os limites previstos em ambas as cartas multivariadas foram calculados a partir das especificações de qualidade da formulação farmacêutica (± 5% do valor nominal para cada princípio ativo). O uso das cartas de controle multivariadas forneceu uma simples e poderosa ferramenta para avaliar a formulação farmacêutica aliada a espectroscopia no infravermelho próximo. O procedimento é rápido e flexível para o monitoramento da produção desta formulação farmacêutica produzida na UFRN/Brasil, com o objetivo de introduzir a tecnologia analítica de processos ao tratamento de tuberculose pulmonar.
One of the keys to maintain high manufacturing quality improvement is the use of control charts. In this work, multivariate control charts based on significant principal component (PC) scores and net analytical signal (NAS) were developed to simultaneously monitor the quality of two active pharmaceutical ingredients (API) (isoniazid and rifampicin) in pharmaceutical formulation (laboratory samples and production samples) using a portable near infrared spectrometer. The limits for both multivariate charts were estimated using the quality specifications from the pharmaceutical formulation (± 5% of the nominal content of each API). The use of these multivariate control charts has provided a simple and powerful tool to evaluate the content of pharmaceutical formulations based on isoniazid and rifampicin capsules combined with near infrared spectroscopy. The procedure is rapid and adjustable for monitoring the production of the pharmaceutical preparations produced at UFRN/Brazil toward the process analytical technology (PAT) for the treatment of pulmonary tuberculosis.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Arthropod venom Hyaluronidases: biochemical properties and potential applications in medicine and biotechnology

AbstractHyaluronidases are enzymes that mainly degrade hyaluronan, the major glycosaminoglycan of the interstitial matrix. They are involved in several pathological and physiological activities including fertilization, wound healing, embryogenesis, angiogenesis, diffusion of toxins and drugs, metastasis, pneumonia, sepsis, bacteremia, meningitis, inflammation and allergy, among others. Hyaluronidases are widely distributed in nature and the enzymes from mammalian spermatozoa, lysosomes and animal venoms belong to the subclass EC 3.2.1.35. To date, only five three-dimensional structures for arthropod venom hyaluronidases (Apis mellifera and Vespula vulgaris) were determined. Additionally, there are four molecular models for hyaluronidases fromMesobuthus martensii, Polybia paulista and Tityus serrulatus venoms. These enzymes are employed as adjuvants to increase the absorption and dispersion of other drugs and have been used in various off-label clinical conditions to reduce tissue edema. Moreover, a PEGylated form of a recombinant human hyaluronidase is currently under clinical trials for the treatment of metastatic pancreatic cancer. This review focuses on the arthropod venom hyaluronidases and provides an overview of their biochemical properties, role in the envenoming, structure/activity relationship, and potential medical and biotechnological applications.

Arachnids of medical importance in Brazil: main active compounds present in scorpion and spider venoms and tick saliva

Arachnida is the largest class among the arthropods, constituting over 60,000 described species (spiders, mites, ticks, scorpions, palpigrades, pseudoscorpions, solpugids and harvestmen). Many accidents are caused by arachnids, especially spiders and scorpions, while some diseases can be transmitted by mites and ticks. These animals are widely dispersed in urban centers due to the large availability of shelter and food, increasing the incidence of accidents. Several protein and non-protein compounds present in the venom and saliva of these animals are responsible for symptoms observed in envenoming, exhibiting neurotoxic, dermonecrotic and hemorrhagic activities. The phylogenomic analysis from the complementary DNA of single-copy nuclear protein-coding genes shows that these animals share some common protein families known as neurotoxins, defensins, hyaluronidase, antimicrobial peptides, phospholipases and proteinases. This indicates that the venoms from these animals may present components with functional and structural similarities. Therefore, we described in this review the main components present in spider and scorpion venom as well as in tick saliva, since they have similar components. These three arachnids are responsible for many accidents of medical relevance in Brazil. Antiproliferative activity of Eremanthus crotonoides extracts and centratherin demonstrated in brain tumor cell lines

The genus Eremanthus is recognized by the predominance of sesquiterpene lactones from the furanoheliangolide type, a class of substances extensively tested against cancer cell lines. Thus, the species E. crotonoides (DC.) Sch. Bip., Asteraceae, obtained on "restinga" vegetation was evaluated against U251 and U87-MG glioma cell lines using the MTT colorimetric assay. Dichloromethane fraction was cytotoxic to both glioblastoma multiforme cell lines. We then conducted UPLC-PDA-ESI-MS/MS analysis of the dichloromethane fraction, which allowed the identification of the sesquiterpene lactones centratherin and goyazensolide. The isolation of centratherin was performed using chromatographic techniques and the identification of this substance was confirmed according to NMR data. Cytotoxic activity of centratherin alone was also evaluated against both U251 and U87-MG cells, which showed IC50 values comparable with those obtained for the commercial anticancer drug doxorubicin. All the tested samples showed cytotoxic activity against glioblastoma multiforme cells which suggests that E. crotonoides extracts may be important sources of antiproliferative substances and that the centratherin may serve as prototype for developing new antiglioblastoma drugs.
