Abstract This work aimed to characterize the variation in the thermal regime of the active layer in a permafrost area on Fildes Peninsula, Antarctica, and relate this variability with meteorological data between 2014 and 2016. The monitoring site was installed to continuously monitor the temperature and moisture of the active layer, radiation flow on the surface, and air temperature. We used data collected to generate the indexes freezing degree-days, thawing degree-days, frost number, n-factor, apparent thermal diffusivity, and active layer thickness. The temperature of the active layer is not homogeneous, varying with depth and position in the transect, with the greatest variations in soil with better drainage and lower moisture content. Among the evaluated factors, air and soil surface temperature are the ones that most influence the thermal gradient of the active layer. We identified that near the surface there is a greater influence of albedo and cloudiness and at -35 cm depth there is a greater influence of net radiation and soil moisture. The average depth of the active layer in 2014 was -44.3 cm and in 2015 -47.7 cm and the frost number index indicates that there was a predominance of continuous permafrost in the transect during the monitoring.

Ventilation distribution as a contributor to the functional exercise capacity in patients with systemic sclerosis-associated interstitial lung disease without pulmonary hypertension

Phenotypic differences have been described between patients with systemic sclerosis (SSc)-associated interstitial lung disease (ILD) and SSc-associated pulmonary hypertension, including performance differences in the 6-min walk test (6MWT). Moreover, the correlations between the 6MWT and traditional pulmonary function tests (PFTs) are weak, indicating the need to search for new parameters that explain exercise performance. Thus, our objective was to evaluate the impact of ventilation distribution heterogeneity assessed by the nitrogen single-breath washout (N2SBW) test and peripheral muscle dysfunction on the exercise capacity in patients with SSc-ILD and limited involvement of the pulmonary parenchyma. In this cross-sectional study, 20 women with SSc-ILD and 20 matched controls underwent PFTs (including spirometry, diffusing capacity for carbon monoxide (DLco), and the N2SBW test) and performed the 6MWT and knee isometric dynamometry. The 6-min walking distance (6MWD, % predicted) was strongly correlated with the phase III slope of the single-breath nitrogen washout (phase III slopeN2SBW) (r=−0.753, P<0.0001) and reasonably correlated with the forced vital capacity (FVC) (r=0.466, P=0.008) and DLco (r=0.398, P=0.011). The peripheral oxygen saturation (SpO2) during exercise was not significantly correlated with any of the pulmonary or muscle function parameters. The phase III slopeN2SBW was the only predictive variable for the 6MWD, whereas quadriceps strength and FVC/DLco were predictive variables for SpO2. Ventilation distribution heterogeneity is one factor that contributes to a lower 6MWD in SSc-ILD patients. In addition, muscle dysfunction and abnormal lung diffusion at least partly explain the decreased SpO2 of these patients.

RESUMO: O presente estudo teve como objetivo determinar a concentração letal e efeitos estruturais e ultraestruturais causados pela exposição a formalina em juvenis de Arapaima gigas. Noventa peixes (60,1±2,5g e 20,2±0,9cm) foram expostos a 0, 22, 44, 66, 88 e 110mg L-1, para determinar a concentração letal (CL50-96h) de formalina que foi 36,4mg L-1. Os efeitos subletais foram avaliados por análises histopatológicas das brânquias e avaliação das alterações comportamentais e sinais clínicos. A CL50 de formalina para 24, 48 e 72horas foi de 88,3, 64,7 e 56,8; respectivamente. Sinais clínicos e alterações comportamentais encontradas foram: natação errática, letargia, aglomeração de peixes na superfície da água, perda de equilíbrio hidrodinâmico, espasmos e confronto agonísticos, observados apenas nas concentrações de 88 e 110mg L-1. O índice de alteração histológica (IAH) mostrou que as concentrações de 66, 88 e 100mg L-1 apresentaram diferenças significativas (p<0,05) em relação aos controles, indicando a ocorrência de danos moderados nas brânquias dos peixes expostos a formalina. Os valores médios de alteração (VMA) para as concentrações 22, 44, 66, 88 e 110mg L-1 foram 1,14, 1,29, 1,51, 1,53 e 1,60; respectivamente, e as diferenças na composição desse índice foram observados apenas na exposição com 110mg L-1 de formalina. Foi possível concluir que concentrações subletais de formalina (22,0mg L-1) não comprometem a saúde de juvenis de A. gigas. Concentrações de formalina acima da CL50-96h podem ser usadas cuidadosamente para banho de curto tempo, uma vez que o VMA para todas as concentrações testadas indicou apenas lesões localizadas que não comprometem a funcionalidade das brânquias dos peixes expostos.
ABSTRACT: This study aimed to determine the lethal concentration and the structural and ultra-structural effects caused by the formalin exposure on juveniles of Arapaima gigas. Ninety fish (60.1± 2.5g and 20.2±0.9cm) were exposed to 0, 22, 44, 66, 88 and 110mg L-1 in order to determine the lethal concentration (LC50-96h) that was 36.4mg L-1 of formalin. Sublethal effects were evaluated using histopathological analysis on the gills and assessment of behavioral alterations and clinical signs. The LC50 of formalin for 24, 48 and 72h was 88.3, 64.7 and 56.8mg L-1 respectively. Clinical signs and behavioral changes were found: erratic swimming, lethargy, crowding on the water surface, loss of hydrodynamic equilibrium, spasms and agonistic confrontation, which were observed only at 88 and 110mg L-1. The histological alteration index (HAI) showed that 66, 88 and 100mg L-1 presented significant difference (p<0.05) in relation to unexposed fish, indicating that moderate damage to the gills of fish exposed to formalin had occurred. The mean values of alteration (MVA) for 22, 44, 66, 88 and 110mg L-1 were 1.14, 1.29, 1.51, 1.53 and 1.60 respectively, and differences in this index were only observed with 110 mgL-1 of formalin. It is therefore possible to conclude that sublethal concentrations of formalin (22.0mg L-1) did not compromise the health of juveniles of A. gigas. Finally, concentrations greater than to LC50-96h may be carefully used for short-term exposure, since the MVA for all concentrations tested only indicated localized lesions that did not compromise gills functionality of exposed fish.

A comparative study of nutritional composition and potential use of some underutilized tropical fruits of Arecaceae

Neste estudo, a polpa e a amêndoa de frutos de seis espécies de Arecaceae foram submetidos à análise centesimal, determinação da composição de ácidos graxos e análise do teor de carotenoides totais. As espécies com os maiores valores de carboidratos, lipídios e proteínas foram Ptychosperma macarthurii(70,1 g/100 g na amêndoa), Syagrus cearensis(40,6 g/100 g na amêndoa) e S. coronata (20,6 g/100 g na polpa). O teor de cinzas variou de 0,61 a 7,51 g/100 g. Láurico, palmítico e oleico foram os principais ácidos graxos identificados. O teor de carotenoides totais e equivalentes de atividade de retinol foram maiores nas polpas dePinanaga kuhlii (180,3 µg/g) e Acrocomia intumescens(138,0 µg/g). Equivalente de atividade de retinol variou entre as espécies estudadas (456-1515 µg RAE/100 g). Espécies nativas como A. intumescens, S. coronatae S. cearensis são boas fontes de alimentos frescos para as populações carentes que habitam áreas pouco desenvolvidas, como a região semiárida do Brasil. P. macarthurii, uma espécie exótica, é uma excelente fonte de cinzas e carotenoides, demonstrando seu potencial como fonte de alimento e de compostos bioativos. A polpa e a amêndoa de A. intumescens, podem ser uma boa alternativa como matérias-primas para a produção de biodiesel e sabão, respectivamente.
In this study, pulp and kernel of fruits from six Arecaceae species were subjected to proximate analysis, fatty acid composition and total carotenoid content analysis. The species with the highest carbohydrate, lipid and protein values were Ptychosperma macarthurii(70.1 g/100 g in the kernel), Syagrus cearensis(40.6 g/100 g in the kernel), andS. coronata(20.6 g/100 g in the pulp). The ash content ranged from 0.61 to 7.51 g/100 g. Lauric, palmitic, and oleic acids were the major fatty acids identified. The total carotenoid contents and retinol activity equivalents were highest in the Pinanga kuhlii (180.3 µg/g) andAcrocomia intumescens (138.0 µg/g) pulp oils. Retinol activity equivalents varied between the investigated species (456 to 1515 μg RAE/100 g). Native species such as A. intumescens, S. coronata, and S. cearensis are good sources of fresh food for the underserved populations that inhabit poorly developed areas such as the semi-arid region of Brazil. P. macarthurii, an exotic species, is an excellent source of ash and carotenoids, demonstrating its potential both as a food source and as bioactive compounds. Pulp and kernel ofA. intumescens,could be a good alternative feedstock for soap and biodiesel production, respectively.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Estudos têm mostrado efeitos tóxicos da enrofloxacina em diversos tecidos. Assim, testou-se a hipótese de que a enrofloxacina pode interferir no desenvolvimento placentário e gerar efeitos adversos ao feto. A enrofloxacina (Baytril(r)) foi administrada em ratas, na dose de 5mg/kg, diariamente, IM, durante toda a gestação. As placentas foram analisadas morfologicamente, morfometricamente e imuno-histoquimicamente aos sete, 14 e 21 dias de prenhez. Os resultados mostraram que a enrofloxacina reduziu o número de sítios de implantação, o peso e a área total do disco placentário aos 14 e 21 dias de desenvolvimento, além dos elementos constituintes da placenta. A análise histoquímica não revelou alterações significativas no teor de fibras colágenas, elásticas e reticulares. O teste de Tunel mostrou atividade apoptótica apenas nas placentas com 14 dias de desenvolvimento de ambos os grupos, sendo mais intensa no grupo tratado. Não foi observado nenhum indício de malformação na cabeça, no tronco e nos membros dos neonatos. No entanto, houve uma redução significativa no número e no peso dos neonatos no grupo tratado, porém sem afetar o seu comprimento. Assim, concluiu-se que a enrofloxacina administrada na dosagem de 5mg/kg durante prenhez em ratas interfere no número de embriões implantados e no desenvolvimento placentário. Isto sugere cautela na administração da enrofloxacina durante a gestação, pois a exposição contínua a esse antibiótico pode ter reflexos na redução do número e do peso da prole.
Some studies have shown the toxic effects of enrofloxacin in various tissues. Thus, the hypothesis that enrofloxacin could interfere with placental development and generate adverse effects to the fetus was tested in this study. Enrofloxacin (Baytril(r)) was administered in the dose of 5mg/kg daily, i.m., throughout gestation in rats. The placentas were analyzed morphologically, morphometrically, and immunohistochemically on the 7, 14, and 20th days of pregnancy. The results showed that enrofloxacin reduced the number of implantation sites, weight, and placental disk total area at 14 and 20 days of development, in addition to the element components of the placenta. The histochemical analysis did not reveal significant changes in the content of collagen, reticular, and elastic fibers. The TUNEL test showed apoptosis only in placenta development at 14 days in both groups and more intense in the treated group. Head, trunk, and limb malformations were not observed in the neonates. A significant reduction in the number and weight of neonates were observed in the treated group, however, without affecting their length. Thus, it was concluded that the administration of enrofloxacin, at the dosage of 5 mg/kg during pregnancy in rats, interferes in the number of implanted embryos and placental development. This suggests caution in the administration of enrofloxacin during pregnancy because continuous exposure to this antibiotic may have adverse effects, reducing the number and weight of the offspring.

O objetivo deste estudo foi determinar o potencial probiótico in vitro de Lactobacillus spp. isolados de queijos minas artesanais da Serra da Canastra, considerando-se o antagonismo entre amostras isoladas frente a microrganismos indicadores, a susceptibilidade a antimicrobianos, a sensibilidade ao ácido gástrico e a sensibilidade a sais biliares. Todas as bactérias ácido-lácticas testadas apresentaram resistência ao ácido gástrico (pH 2,0) e aos sais biliares (0,3%), bem como atividade antagonista contra Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, Salmonella enterica var. Typhimurium, Enterococcus faecalis e bactérias ácido-lácticas isoladas dos próprios queijos - Lactobacillus plantarum (D27) e Lactobacillus rhamnosus (B25). Todas as amostras foram sensíveis à eritromicina e tetraciclina e resistentes à ciprofloxacina, gentamicina, oxacilina, estreptomicina e vancomicina. L. plantarum(B17) apresentou melhor potencial probiótico, pois obteve resultados satisfatórios em todas as propriedades avaliadas. Mais estudos são necessários para verificar a presença e a capacidade de transmissão de genes de resistência antimicrobiana a outros microrganismos e para avaliar o potencial dos microrganismos in vivo. As bactérias selecionadas poderão ser utilizadas na elaboração de queijos em que sejam mantidos o sabor e a tradição do queijo minas artesanal do estado de Minas Gerais.
The aim of this study was to determine some in vitro probiotic features of Lactobacillus spp. isolated from Minas artisanal cheese from Serra da Canastra in order to select some bacteria for future production of safer cheeses keeping their natural flavor and tradition. The evaluated properties were antagonism against indicator microorganisms, antimicrobial susceptibility and also sensitivity to gastric acid and to bile salts. All lactic acid bacteria tested were resistant to gastric acid (pH 2.0) and bile salts (0.3%). Antagonistic activities were detected against Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, Salmonella entericavar. Typhimurium, Enterococcus faecalisand other lactic bacteria isolated from the same cheese - Lactobacillus plantarum (D27) and Lactobacillus rhamnosus (B25). All samples were sensitive to the antimicrobials erythromycin, tetracycline and resistant to ciprofloxacin, gentamycin, oxacillin, streptomycin and vancomycin. L.plantarum (B17) presented the best probiotic potential. Further studies are needed to verify the presence and the capacity of transmission of antimicrobial resistance genes to other microorganisms and evaluate the in vivo probiotic potential of the selected microorganisms.

No Brasil, dados relacionados com a ocorrência de neoplasias em ruminantes e equinos são escassos. Objetivou-se com este trabalho determinar a frequência de neoplasias diagnosticadas em bovinos, caprinos, ovinos e equídeos no Laboratório de Patologia Animal da Universidade Federal de Campina Grande, Patos, Paraíba, durante o período de 1983 a 2010 e analisar os fatores de risco, mediante o teste de qui-quadrado, considerando como variáveis, espécie, raça, sexo e idade. Durante o período foram registrados 177 (5,6%) tumores de um total de 3.153 diagnósticos provenientes de biópsias e necropsias. Houve diferenças significantes (p<0,001) na frequência de tumores entre as diferentes espécies sendo mais acometidos os equinos (10,6%), seguidos pelos bovinos (6,8%), caprinos (3,3%) e ovinos (2,1%). Os tumores mais frequentes foram o carcinoma células escamosas (CCE) nos bovinos (58,3%), ovinos (80%) e caprinos (46,1%) e o sarcoide em equinos (45,3%). Quanto à localização anatômica, as neoplasias foram mais frequentes na pele em equídeos (62,5%) e ovinos (60%), olho e tecido periocular em bovinos (36,1%) e sistema reprodutor feminino (períneo e vulva) em caprinos (34,6%). Em relação ao sexo dos animais, apenas os bovinos apresentaram prevalência significativa (p<0,001). Em relação à idade apenas os caprinos apresentaram prevalência significativa (p=0,015). Não houve prevalência significativa em nenhuma espécie em relação à raça. Conclui-se que os carcinomas de células escamosas em ruminantes e o sarcoide em equídeos são os tumores mais frequentes em animais de produção no semiárido do Nordeste Brasileiro e que a espécie equina foi a mais acometida dentre as espécies estudadas.
In Brazil, data related to occurrence of tumors in livestock are scarce. The objective of this study was to determine the frequency of tumors diagnosed in equidae, cattle, sheep and goats in the Pathology Laboratory of the Federal University of Campina Grande, Patos, state of Paraíba, from 1983 to 2010, and to analyze the risk factors by the chi square test, considering variables such as species, breed, age and sex. During the period, 177 (5.6%) out of 3153 specimens from biopsies or necropsies were diagnosed as tumors. There were significant differences (p<0.001) in the frequency of tumors among different species; horses (10.6%) were more affected than cattle (6.8%), and cattle were more affected than goats (3.3%) and sheep (2.1%). Squamous cell carcinoma was most frequent tumor in cattle (58.3% of the tumors), sheep (80%) and goats (46.1%), and sarcoid (45.3%) was the most frequent in horses. Regarding the location, neoplasms were more common in the skin of horses (62.5%) and sheep (60%), in the eyes and periocular skin in cattle (36.1%), and un the female reproductive system (perineal region and vulva) of goats (34.6%). Regarding the sex of the animals, only cattle had significant prevalence (p<0.001). Regarding age only goats showed significant prevalence (p=0.015). There was no significant prevalence in any animal species in relation to race. We concluded that squamous cell carcinomas in ruminants and equine sarcoid are the most common tumors in livestock in the semiarid Brazilian Northeast, and the equidae were the most affected among the species studied.

An increased incidence of nosocomial and community-acquired infections caused by methicillin-resistant Staphylococcus aureus (MRSA) has been observed worldwide. The molecular characterization of MRSA has played an important role in demonstrating the existence of internationally disseminated clones. The use of molecular biology methods in the surveillance programs has enabled the tracking of MRSA spread within and among hospitals. These data are useful to alert nosocomial infection control programs about the potential introduction of these epidemic clones in their areas. Four MRSA blood culture isolates from patients hospitalized at two hospitals in the city of São Paulo, Brazil, were analyzed; one of them was community acquired. The isolates were characterized as SCCmec, mecA and PVL by PCR, pulsed-field gel electrophoresis (PFGE) profile and molecular sequence typing (MLST) genotyping. The isolates presented type IV SCCmec, and none proved to be positive for PVL. The isolates showed a PFGE profile similar to the pediatric clone. MLST genotyping demonstrated that the isolates belonged to clonal complex 5 (CC5), showing a new yqiL allele gene, resulting in a new sequence typing (ST) (1176). Our results showed that strains of MRSA carrying a new ST are emerging in community and nosocomial infections, including bacteremia, in São Paulo, Brazil.

Pilocarpine is a natural substance with potential application in the treatment of several diseases. In this work Fourier Transform (FT)-Raman spectrum and the Fourier Transform infra red (FT-IR) spectrum of pilocarpine hydrochloride C11H17N2O2+.Cl- were investigated at 300 K. Vibrational wavenumber and wave vector have been predicted using density functional theory (B3LYP) calculations with the 6-31 G(d,p) basis set. A comparison with experiment allowed us to assign most of the normal modes of the crystal.

Apolipoprotein E (protein: apo E; gene: APOE) plays an important role in the multifactorial etiology of both Alzheimer's disease (AD) and lipid level concentrations. The polymerase chain reaction (PCR) was used to investigate the APOE gene polymorphism in 446 unrelated Caucasians, among them 23 AD patients, and 100 Afro-Brazilians living in Porto Alegre, Brazil. The frequencies of the APOE*2, APOE*3 and APOE*4 alleles were 0.075, 0.810 and 0.115 in Caucasians and 0.075, 0.700 and 0.225 in Afro-Brazilians, respectively (c2 = 8.72, P = 0.013). A highly significant association was observed between the APOE*4 allele and AD in this population-based sample. The APOE*4 frequency in AD patients (39%) was about four times higher than in the general Caucasian population (11.5%). The influence of each of the three common APOE alleles on lipid traits was evaluated by the use of the average excess statistic. The E*2 allele is associated with lower levels of triglycerides and of total and non-HDL cholesterol in both men and women. Conversely, the E*4 allele is associated with higher levels of these traits in women only. The effect of APOE alleles was of greater magnitude in women.
