Resumo A pitaia vermelha (Hylocereus costaricensis) é uma promissora espécie, com elevado potencial de cultivo devido às qualidades organolépticas e funcionais de seus frutos. Entretanto, a salinidade da água de irrigação pode afetar o rendimento produtivo da cultura. Diante disso, materiais ricos em substâncias orgânicas podem minimizar os danos provocados pelo excesso de sais no solo e/ou na água. Assim, o objetivo deste trabalho foi avaliar a influência de fontes de matéria orgânica como atenuante do estresse salino na produção e respostas bioquímicas de mudas de pitaia vermelha. O delineamento inteiramente casualizado em esquema fatorial 4 × 5, com cinco fontes de matéria orgânica (húmus, esterco ovino, biofertilizante, composto orgânico e areia + solo) e quatro salinidade (0.6, 2.6, 4.6 e 6.6 dS m-1), com quatro repetições e duas plantas por vaso foi utilizado. O comprimento da parte aérea, comprimento da raiz, diâmetro do cladódio, número de cladódios, número de brotos, volume da raiz, massa seca da parte aérea, massa seca da raiz e massa seca total, razão da massa seca da raiz e da parte aérea, clorofila a, b e total, aminoácidos e os açúcares solúveis foram avaliados aos 120 dias após o início da aplicação dos tratamentos. A pitaia vermelha é moderadamente tolerante a salinidade. O composto orgânico e estrume ovino atenuam os efeitos nocivos da salinidade nas mudas de pitaia vermelha. Sob condições de estresse salino, as plantas aumentam os níveis de aminoácidos e açúcares totais.

Abstract Red pitaya (Hylocereus costaricensis) is a promising species, with high cultivation potential due to the organoleptic and functional qualities of its fruits. However, irrigation water salinity can affect the crop yield. Therefore, materials rich in organic substances can minimize the damage caused by excess salts in soil and/or water. Thus, the objective of this study was to evaluate the influence of organic matter sources as attenuators of salt stress on the production and biochemical responses of red pitaya seedlings. A completely randomized design in 4 × 5 factorial scheme, with five sources of organic matter (humus, sheep manure, biofertilizer, organic compost and sand + soil) and four salinities (0.6, 2.6, 4.6 and 6.6 dS m-1) with four replicates and two plants per plot was used. The shoot length, root length, cladode diameter, number of cladodes, number of sprotus, root volume, shoot dry mass, root dry mass and total dry mass, root and shoot dry mass ratio, chlorophyll a, b and total, amino acids and soluble sugars were evaluated at 120 days after the treatments began to be applied. Red pitaya is moderately tolerant to salinity (ECw from 4.0 to 6.0 dS m-1). Organic compost and sheep manure attenuate the harmful effects of salinity on red pitaya seedlings. Under salt stress conditions, red pitaya plants increase their levels of proline, amino acids and total sugars.

In the present study, we tested the proof of concept that mobil mesoporous silica nanoparticle composition of matter (MCM) No. 48 doped with trivalent europium (nanoMCM48:Eu3+) administered systemically intravenously can act as a suitable vehicle to deliver neural cells to the brain of healthy adult Swiss mice without causing tissue damage. Moreover, we also tested the ability of this nanoparticle to release molecules in vitro, using as drug models caffeine or nicotine in phosphate buffer solution with pH 7.4 adequate to intra and extra-celular medium of the brain. The caffeine and nicotine adsorbed nanoparticles (CAF@MCM48:Eu3+ and NIC@MCM48:Eu3+) were observed in the parenchyma of the cerebral cortex and diffusely dispersed in the cellular cytoplasm. Statistical analysis using multivariate partner recognition methods indicated that there was no sign of cell damage, as it was characterized by chromatin condensation, nuclear condensation, or fragmentation. The characterization of nanoMCM48:Eu3+ as particle size, luminescent properties and release of active components were analyzed. The CAF@MCM48:Eu3+ and NIC@MCM48:Eu3+ nanoparticles were observed in the cerebral cortex parenchyma and diffusely dispersed in the cell cytoplasm, and the release of active components was analyzed. Therefore, the studies showed that mesoporous silica nanoparticles administered systemically via intravenous tissue can act as a suitable vehicle for neural cells in the brain of healthy mice without causing damage.

ABSTRACT The use of biostimulants in second-crop maize (Zea mays) can promote higher physiological activity in plants, resulting in higher grain yield. This study aimed to evaluate the effects of phytohormones and nicotinamide on growth and yield traits of second-crop maize. The statistical design used was a randomized block design arranged in a factorial scheme with two doses of phytohormones (0 and 500 mL ha-1) × five concentrations of nicotinamide (0, 200, 400, 600, and 800 mg L-1) with four replications. The use of biostimulants favored all variables evaluated for the maize crop. The isolated use of phytohormones and nicotinamide promoted grain yields of 6089.6 and 6242.5 kg ha-1, respectively, representing gains of 6.0 and 11.6%, respectively, compared with the control. The application of 800 mg L-1 nicotinamide resulted in the highest maize grain yield. The use of phytohormones and nicotinamide, isolated or associated, was favorable for the growth characteristics and grain yield of second-crop maize. A synergistic effect was noted between phytohormones and nicotinamide on the first ear insertion height and number of rows per ear.

RESUMO O uso de bioestimulantes no milho segunda safra (Zea mays) pode promover maior atividade fisiológica das plantas, resultando em maior produtividade de grãos. Assim, este trabalho teve como objetivo avaliar as características de crescimento e produtividade do milho segunda safra sob aplicação de fitormônios e nicotinamida. O delineamento estatístico utilizado foi em blocos casualizados dispostos em esquema fatorial com 2 doses de fitohormônios (0 e 500 mL ha-1) × 5 concentrações de nicotinamida (0, 200, 400, 600 e 800 mg L-1) com quatro repetições. O uso dos dois bioestimulantes favoreceu todas as variáveis avaliadas para a cultura do milho. O uso isolado de fitormônios e nicotinamida promoveu produtividades de grãos de 6.089,6 e 6.242,5 kg ha-1, representando respectivamente, ganhos de 6,0 e 11,6%, em relação à testemunha. A aplicação de 800 mg L-1 de nicotinamida proporcionou maior produtividade de grãos de milho. O uso de fitohormônios e nicotinamida, isolados ou associados, foram favoráveis às características de crescimento e produtividade do milho segunda safra. Houve efeito sinérgico de fitormônios e nicotinamida para altura de inserção da primeira espiga e o número de fileiras por espiga.

ABSTRACT Using nematode-resistant and/or -tolerant rootstock is a strategy that has maintained guava cultivation feasible in the irrigated perimeters of the Brazilian semi-arid region. The objective was to evaluate the production, gas exchange, and fruit quality of ‘Paluma’ guava with and without nematode-tolerant rootstock under different irrigation depths and production periods. The experimental design was randomized blocks, in the mixed 2 × 4 × 2 factorial scheme, corresponding to two forms of cultivation of ‘Paluma’ guava (without and with nematode-tolerant rootstock), four irrigation depths (60, 80, 100, and 120% of crop evapotranspiration - ETc), and two production periods, with four replicates. Guava plants were evaluated for physiological, production and post-harvest quality parameters. Gas exchange in ‘Paluma’ guava leaves was higher in the second production cycle, under irrigation depths above 100% ETc, not differing with the use of the rootstock. The use of the rootstock ‘BRS Guaraçá’ promotes a greater number of fruits and production with the increase of the irrigation depth, mainly in the second half of the year, with the increase of soluble solids and titratable acidity in the fruits.

RESUMO A utilização de porta enxerto resistente e ou tolerante a nematoides é uma estratégia que tem tornado viável a manutenção dos cultivos de goiabeira nos perímetros irrigados do semiárido brasileiro. Assim, o objetivo foi avaliar a produção, trocas gasosas e qualidade dos frutos da goiabeira ‘Paluma’ com e sem o porta enxerto tolerante a nematoide sob diferentes lâminas de irrigação e épocas de produção no Semiárido Brasileiro. O delineamento experimental foi em blocos ao acaso, no esquema fatorial misto 2 × 4 × 2, sendo duas formas de cultivo da goiabeira ‘Paluma’ (sem e com o porta enxerto tolerante a nematoide), quatro lâminas de irrigação (60, 80, 100 e 120% da evapotranspiração da cultura - ETc) e duas épocas de produção, com quatro repetições. Foram realizadas avaliações fisiológicas das plantas, produção e qualidade pós-colheita. As trocas gasosas nas folhas da goiabeira ‘Paluma’ foram maiores no segundo ciclo de produção, quando irrigada com lâminas de irrigação acima de 100% da ETc, não diferindo com o uso do porta enxerto. O uso do porta-enxerto ‘BRS Guaraçá’ proporciona um maior número de frutos e produção com o aumento da lâmina de irrigação, principalmente no segundo semestre do ano, bem como o aumento do teor de sólidos solúveis e acidez nos frutos.

RESUMO O uso de bioestimulantes em mudas de café pode promover ganhos em seu crescimento e na sua qualidade. Assim, o objetivo desse trabalho foi avaliar as características de crescimento e qualidade de mudas de café arábica sob o efeito da aplicação dos bioestimulantes nicotinamida e Azospirillum brasilense. O delineamento experimental utilizado foi o de blocos casualizados, em esquema fatorial 5 × 2, com quatro repetições. Os tratamentos resultaram da utilização de cinco doses de nicotinamida (0, 30, 60, 90 e 120 mg L-1 de água) combinadas com a ausência e presença de Azospirillum brasilense aplicadas em mudas de Coffea arabica da cultivar Catuaí Vermelho 144. Foram avaliadas a altura de planta, diâmetro do caule, número de folhas, área foliar, massa seca da parte aérea, massa seca da raiz, razão entre altura e diâmetro do caule, razão entre massa seca da parte aérea e massa seca da raiz, razão entre altura e massa seca da parte aérea e índice de qualidade Dickson. A utilização conjunta ou isolado do A. brasilense e da nicotinamida, até a dose de 33.5 mg L-1, aumentou características biométricas e acúmulo de massa seca. No entanto, a utilização de doses de nicotinamida entre 30 e 61,8 mg L-1 aumentou da qualidade de mudas de Coffea arabica. Foi verificado efeito sinérgico do uso de A. brasilense e nicotinamida para crescimento e qualidade de mudas.

ABSTRACT The use of biostimulants in coffee seedlings can promote gains in their growth and quality. Thus, this study aimed to evaluate the growth and quality characteristics of Coffea arabica seedlings under the effect of the nicotinamide and Azospirillum brasilense application. The experimental design was randomized blocks with treatments arranged in a 5 × 2 factorial scheme, with four replicates. The treatments resulted from the use of five doses of nicotinamide (0, 30, 60, 90, and 120 mg L-1 of water) combined with the absence and presence of Azospirillum brasilense applied to Coffea arabica seedlings from Catuaí Vermelho 144 cultivar. Plant height, stem diameter, number of leaves, leaf area, shoot dry mass, root dry mass, plant height:stem diameter ratio, shoot:root dry mass ratio, plant height:shoot dry mass ratio, and Dickson quality index were evaluated. The combined or isolated use of A. brasilense and nicotinamide, up to a dose of 33.5 mg L-1, increased the biometric characteristics and dry mass accumulation. However, using nicotinamide doses between 30 and 61.8 g L-1 increased the quality of Coffea arabica seedlings. The synergistic effect of the use of A. brasilense and nicotinamide was verified for the growth and quality of Coffea arabica seedlings.

Resumo O óleo essencial de citronela (Cymbopogon winterianus) possui diversas atividades biológicas, entre elas a ação repelente a insetos. Alguns estudos mostraram que os ésteres do ácido cinâmico podem ser aplicados como pesticidas naturais, inseticidas e fungicidas. Nesse contexto, o objetivo do presente trabalho foi avaliar a produção de ésteres a partir do óleo essencial de citronela com ácido cinâmico via esterificação enzimática. Além disso, foi investigada a toxicidade do óleo essencial antes e após a esterificação contra Artemia salina e a ação larvicida sobre Aedes aegypti. Os ésteres foram produzidos utilizando ácido cinâmico como agente acilante e óleo essencial de citronela (3: 1) em heptano e 15% em peso da enzima NS 88011 como biocatalisadores, a 70 ° C e 150 rpm. As taxas de conversão de cinamatos de citronelil e geranil foram 58,7 e 69,0% para NS 88011, respectivamente. Para a toxicidade sobre Artemia salina foram obtidos CL50 de 5,29 μg mL-1 para o óleo essencial e 4,36 μg mL-1 para os óleos esterificados com NS 88011. Na atividade inseticida contra larvas de Aedes aegypti, obteve-se CL50 de 111,84 μg mL-1 para o óleo essencial de citronela e 86,30 μg mL-1 para os óleos esterificados com a enzima NS 88011, indicando alta toxicidade dos ésteres. Os resultados demonstraram que as amostras avaliadas apresentam potencial de aplicação como bioinseticida.

Abstract The essential oil of citronella (Cymbopogon winterianus) has several biological activities, among them the insect repellent action. Some studies showed that cinnamic acid esters can be applied as natural pesticides, insecticides and fungicides. In this context, the objective of the present work was to evaluate the production of esters from citronella essential oil with cinnamic acid via enzymatic esterification. Besides, the essential oil toxicity before and after esterification against Artemia salina and larvicidal action on Aedes aegypti was investigated. Esters were produced using cinnamic acid as the acylating agent and citronella essential oil (3:1) in heptane and 15 wt% NS 88011 enzyme as biocatalysts, at 70 °C and 150 rpm. Conversion rates of citronellyl and geranyl cinnamates were 58.7 and 69.0% for NS 88011, respectively. For the toxicity to Artemia salina LC50 results of 5.29 μg mL-1 were obtained for the essential oil and 4.36 μg mL-1 for the esterified oils obtained with NS 88011. In the insecticidal activity against Aedes aegypti larvae, was obtained LC50 of 111.84 μg mL-1 for the essential oil of citronella and 86.30 μg mL-1 for the esterified oils obtained with the enzyme NS 88011, indicating high toxicity of the esters. The results demonstrated that the evaluated samples present potential of application as bioinsecticide.

Abstract A lectin from the marine sponge Haliclona (Reniera) implexiformis (HiL) was isolated by affinity chromatography on Sepharose™ matrix. HiL showed specificity for galactose and its derivatives. The glycoproteins porcine stomach mucin (PSM) and bovine stomach mucin (BSM) were potent inhibitors. Hemagglutinating activity of the lectin was maximal between pH 5.0 and 9.0. The lectin remained active until 60°C. The presence of CaCl2 and EDTA did not affect the hemagglutinating activity. In SDS-PAGE, HiL showed a single band of 20 kDa under reduced conditions, whereas in the non-reducing conditions, it showed a band of 20 kDa and one additional band of 36 kDa. The average molecular mass determined by Electrospray Ionization Mass Spectrometry (ESI-MS) was 35.874 ± 2 Da in native and non-reducing conditions, whereas carboxyamidomethylated-lectin showed 18,111 Da. These data indicated that HiL consists in a dimer formed by identical subunits linked by disulfide bonds. Partial amino acid sequence of HiL was determined by mass spectrometry, and revealed that it is a new type of lectin, which showed no similarity with any protein. Secondary structure consisted of 6% α-helice, 31% β-sheet, 18% β-turn and 45% random coil. HiL showed significant reduction in the number of viable cells of Staphylococcus biofilms.

Abstract Sugarcane cultivation stands out in Brazilian agribusiness, covering more than eight million hectares for the production of sugar, ethanol, and by-products. Fertilization is one of the limiting factors in sugarcane yield, for which filter cake is a viable solution to meet plant nutritional needs. This study aimed to assess the effect of enriched filter cake on gas exchange and yield in RB041443 sugarcane, cultivated in soils of the coastal tablelands of Paraíba, Brazil. The experiment was conducted in the Monte Alegre S/A sugarcane mill, in the municipality of Mamanguape, using a randomized blocks experimental design, with 12 treatments (T1- cake, T2- cake + MAP, T3- cake + gypsum, T4 - cake + phosphate, T5- cake + bagasse, T6- cake + MAP + gypsum, T7- cake + MAP + phosphate, T8- cake + MAP + bagasse, T9- cake + gypsum + phosphate, T10- cake + gypsum + bagasse, T11- cake + phosphate + bagasse, and T12- control (only MAP)), and 4 replications, totaling 48 plots. A significant effect (5% probability) was also observed for the variables number of leaves and tons of stem per hectare (TSH). T1- cake, T4- cake + phosphate, T6- cake + MAP + gypsum and T10- cake + gypsum + bagasse, had the best results for TSH, with yields greater than 140 t ha-1. Regarding stomatal conductance, the highest values were obtained in T6 and T8, which, together with T11, had the highest gs values. Concerning the internal carbon concentration, T1, T2, T6, and T8 stood out. T6 also had a significant effect on transpiration. From this study, it was concluded that the use of enriched filter cake as a base fertilizer in sugarcane culture contributes to increasing the yield of the RB041443 variety, generating positive responses for plant gas exchange, being T1 and T10 indicated to increase the production in the sugar-energy sector.

Resumo A cultura da cana-de-açúcar ocupa uma posição de destaque para o agronegócio brasileiro, abrangendo mais de oito milhões de hectares na produção de açúcar, etanol e subprodutos. Um dos fatores limitantes na produtividade da cana-de-açúcar é a questão da adubação, sendo a torta de filtro uma solução viável para suprir as necessidades nutricionais da planta. Com esse estudo, objetivou-se avaliar o efeito do uso da torta de filtro enriquecida sob as trocas gasosas e produtividade da variedade de cana-de-açúcar RB041443 cultivada em solos dos tabuleiros costeiros da Paraíba. O experimento foi conduzido na Usina Monte Alegre S/A, no município de Mamanguape-PB, utilizando o delineamento experimental em blocos casualizados com 12 tratamentos (T1- Torta, T2- Torta + MAP, T3- Torta + Gesso, T4 - Torta + Fosfato, T5- Torta + Bagaço, T6- Torta + MAP + Gesso, T7- Torta + MAP + Fosfato, T8- Torta + MAP + Bagaço, T9- Torta + Gesso + Fosfato, T10- Torta + Gesso + Bagaço, T11 - Torta + Fosfato + Bagaço, e T12 - Testemunha (Apenas MAP)), 4 repetições, totalizando 48 parcelas. Efeito significativo a 5% também foi observado para a variável número de folhas e tonelada de colmo por hectare. Para a variável TCH, os melhores resultados foram obtidos nos tratamentos T1- Torta, T4-Torta + Fosfato, T6- Torta + MAP + Gesso e T10- Torta + Gesso + Bagaço, com produtividades superiores a 140 t ha-1. Para a condutância estomática, resultados superiores foram obtidos no T6 e T8, que, junto ao T11, agruparam-se com os maiores valores de gs. Em relação a concentração interna de carbono, destacaram-se os tratamentos T1, T2, T6 e T8. Para a variável transpiração, o T6 apresentou influência significativa da torta de filtro. Com o referido estudo, concluiu-se que a utilização da torta de filtro enriquecida como adubação de fundação na cultura da cana-de-açúcar possibilita aumento no rendimento produtivo da variedade RB 041443, bem como respostas positivas para as trocas gasosas da planta, sendo os tratamentos T1 e T10 indicados para aumentar a produção do setor sucroenergético.

Abstract Due to the complex nature of pain and the participation of physical, cognitive, psychological and behavioral aspects, pain management has several approaches. The use of medicinal plants in developing countries is quite expressive. Seeking new options for the treatment of emerging or debilitating diseases. Therefore, the present study seeks to elucidate the effects of the monoterpene, citronellal, differentiating its activity by isomers (R)-(+) and (S)-(-) citronellal. The study used several methods to evaluate the effects of citronellal isomers on motor coordination, nociceptive response, and the involvement of opioid, glutamatergic, and transient receptor pathways. The methods included rota-rod, hot-plate, and formalin tests, as well as the use of specific inhibitors and agonists. Data were analyzed using inferential statistics with a 95% confidence level. Both isomers did not significantly affect the motor coordination of the studied animals. The isomer (S)-(-) citronellal showed better results in relation to its structural counterpart, managing to have an antinociceptive effect in the formalin and hot plate tests with a lower concentration (100 mg/kg) and presenting fewer side effects, however, the this study was not able to elucidate the mechanism of action of this isomer despite having activity in studies with substances that act on specific targets such as glutamate and capsaicin, its activity was not reversed with the use of antagonists for pathways related to nociception. While the (R)-(+) citronellal isomer, despite showing total activity only at a concentration of 150 mg/kg, was able to determine its mechanism of action related to the opioid pathway by reversing its activity by the antagonist naloxone, being this is a pathway already correlated with nociception control treatments, however, it is also related to some unwanted side effects. In this way, new studies are sought to elucidate the mechanism related to the isomer (S)-(-) citronellal and a possibility of use in other areas related to the treatment of pain or inflammation.

Resumo Devido à natureza complexa da dor e a sua participação de aspectos físicos, cognitivos, psicológicos e comportamentais, o manejo da dor possui diversas abordagens. O uso de plantas medicinais em países em desenvolvimento é bastante expressivo. Buscando novas opções para o tratamento de denças emergentes ou debilitantes. Portanto, o presente estudo busca elucidar os efeitos do monoterpeno, citronelal, diferenciando sua atividade pelos isômeros (R)-(+) e (S)-(-) citronelal. O estudo utilizou diversos métodos para avaliar os efeitos dos isômeros de citronelal na coordenação motora, resposta nociceptiva e o envolvimento de vias opioides, glutamatérgicas e de receptores transitórios. Os métodos incluíram testes de rota-rod, placa quente e formalina, além do uso de inibidores e agonistas específicos. Os dados foram analisados estatisticamente com um intervalo de confiança de 95%. Ambos os isomeros não afetaram significativamente a coordenação motora dos animais em estudo. O isômero (S)-(-) citronelal apresentou melhores resultados em relação ao seu homólogo estrutural, conseguindo ter um efeito antinociceptivo nos testes de formalina e placa quente com menor concentração (100 mg/kg) e apresentando menos efeitos colaterais, entretanto, o presente estudo não foi capaz de elucidar o mecanismo de ação deste isomero apesar de ter atividadade em estudos com substancias que agem em alvos específicos como glutamato e capsaicina, sua atividade não foi revertida com a utilização de antagonistas para as vias relacionadas à nocicepção. Enquanto o isômero (R)-(+) citronelal, apesar de apresentar de apresentar total atividade somente na concentração de 150 mg/kg, foi capaz de determinar seu mecanismo de ação relacionado à via opióide pela reversão da sua atividade pelo antagonista naloxona, sendo esta uma via já correlacionada com os tratamentos de controle da a nocicepção, no entanto, também está relacionada a alguns efeitos colaterais indesejados. Desta forma busca-se novos estudos para elucidação do mecanismo relacionados ao isomero (S)-(-) citronellal e uma possibilidade de utilização em outras areas relacionadas ao tratamento da dor ou inflamação.

Abstract Pseudomonas aeruginosa is a non-lactose fermenting Gram-negative bacteria responsible for causing numerous nosocomial infections. The present research aimed to analyze the anti-Pseudomonas aeruginosa potential of 2-Chloro-N-(4-fluoro-3-nitrophenyl)acetamide (A8). The antibacterial potential of A8 was evaluated from the Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC) and Association using the checkerboard method. MIC and MBC values were 512 µg/mL for all P. aeruginosa strains evaluated, demonstrating predominantly bactericidal activity. Furthermore, when A8 was associated with the drug ceftriaxone, pharmacological additivity and indifference were evidenced. In this sense, the synthetic amide was interesting, since it demonstrates the potential to become a possible candidate for an antimicrobial drug.

Resumo Pseudomonas aeruginosa é uma bactéria Gram-negativa não fermentadora de lactose, responsável por causar inúmeras infecções nosocomiais. A presente pesquisa teve como objetivo analisar o potencial anti-Pseudomonas aeruginosa da molécula 2-Cloro-N-(4-fluoro-3-nitrofenil)acetamida (A8). O potencial antibacteriano do A8 foi avaliado a partir da Concentração Inibitória Mínima (CIM), Concentração Bactericida Mínima (CBM) e Associação pelo método de checkboard. Os valores de CIM e CBM foram de 512 µg/mL para todas as cepas de P. aeruginosa avaliadas, demonstrando atividade predominantemente bactericida. Além disso, quando o A8 foi associado ao fármaco ceftriaxona, evidenciou-se aditividade e indiferença farmacológica. Nesse sentido, a amida sintética se mostrou interessante, pois demonstra potencial para se tornar um possível candidato a fármaco antimicrobiano.

Abstract The aim of this study was to determine the acute toxicity of the essential oils (EOs) of Aloysia triphylla, Lippia gracilis and Piper aduncum in juvenile tambaqui (Colossoma macropomum), and evaluate the possible histopathological alterations in their gills. For the acute toxicity tests, juvenile tambaqui (n=24/treatment) were distributed in six treatments with three replicates, which comprised the control and five EO concentrations of A. triphylla (60, 80, 100, 120 and 140 mg L-1), L. gracilis (35, 40, 45, 50 and 55 mg L-1) and P. aduncum (42.5, 45, 47.5, 50 and 52.5 mg L-1), with an exposure period of 4 h. The mortality rate and severity of damage to the tambaqui gills were proportional to the increase in the concentration of the EO, with LC50-4 h values estimated at 109.57 mg L -1 for A. triphylla, 41.63 mg L -1 for L. gracilis and 48.17 mg L -1 for P. aduncum. The main morphological damages observed in the gills of the tambaqui exposed to the three EOs, were Grade I: hypertrophy and hyperplasia of lamellar epithelial cells, lamellar fusion, epithelial detachment, capillary dilation and constriction, proliferation of chloride cells and mucosal cells and edema; in low frequency Grade II damage as epithelial rupture and lamellar aneurysm. Necrosis (Grade III damage) was observed only in gill lamellae exposed to P. aduncum EO (47.5, 50.0 and 52.5 mg L-1). Concentrations of EOs below LC50-4 h can be used sparingly, for short periods of exposure for the treatment of diseases in tambaqui breeding.

Resumo O objetivo deste estudo foi determinar a toxicidade aguda dos óleos essenciais (OEs) de Aloysia triphylla, Lippia gracilis e Piper aduncum em juvenis de tambaqui (Colossoma macropomum), e avaliar as possíveis alterações histopatológicas em suas brânquias. Para os testes de toxicidade aguda, juvenis de tambaqui (n=24/tratamento) foram distribuídos em 6 tratamentos, com três repetições, sendo o controle e cinco concentração do OE de A. triphylla (60, 80, 100, 120 e 140 mg L-1), L. gracilis (35, 40, 45, 50 e 55 mg L-1) e P. aduncum (42,5, 45, 47,5, 50 e 52,5 mg L-1), com exposição de 4 h. A taxa de mortalidade e a severidade dos danos nas brânquias de tambaqui foram proporcionais ao aumento da concentração do OE, com os valores de CL50-4 h estimados em 109,57 mg L-1 para A. triphylla, em 41,63 mg L-1 para L. gracilis e em 48,17 mg L-1 para P. aduncum. Os principais danos morfológicos observados nas brânquias de tambaqui, expostos aos três OEs, foram os de grau I: hipertrofia e hiperplasia das células do epitélio lamelar, fusão lamelar, descolamento epitelial, dilatação e constrição capilar, proliferação de células de cloreto e de células mucosas e edema; em baixa frequência os de grau II como ruptura epitelial e aneurisma lamelar. Necrose (dano de grau III) foi observado somente nas lamelas branquiais expostas ao OE de P. aduncum (47,5, 50,0 e 52,5 mg L-1). Concentrações do OEs abaixo dos valores de CL50-4 h podem ser utilizados com parcimônia, em curtos períodos de exposição para o tratamento de doenças na criação de tambaqui.

Abstract Schistosomiasis, caused by Schistosoma mansoni Sambon, 1907, is a severe and widely distributed parasitic disease, affecting about 200 million people worldwide. The disease is recognized by elevated mortality rates, especially among those living in areas of poor sanitation. Currently, the chemotherapeutic treatment is solely based on using the praziquantel drug. Therefore, there is a need for the discovery of new medicines for the treatment of this parasitosis. Thus, this work aimed to evaluate the schistosomicidal activity of ethanolic crude extracts from the branches, leaves, flowers, and fruits of Handroanthus impetiginosus (Mart ex DC.) Masttos and characterize its metabolic profile by UPLC-ESI-QTOF analysis. Evaluation of plant extract on S. mansoni was carried out in adult worms in vitro, in which the mortality rate was quantified, and the damages in the tegument of the worms were monitored. All extracts induced changes in the viability of adult males of S. mansoni, causing the death of the parasites, which was directly dependent of the concentration.

Resumo Schistosomíase, causada pelo Schistosoma mansoni Sambon, 1907, é uma doença parasitária severa e amplamente distribuída, afetando cerca de 200 milhões de pessoas pelo mundo. A doença é reconhecida pelo alto índice de mortalidade, especialmente dentre as populações que vivem em ambientes de pouca vigilância sanitária. Geralmente, o tratamento é apenas baseado no uso da droga praziquantel. Entretanto, há uma necessidade da descoberta de novos medicamentos para o tratamento dessa parasitose. Por isso, o presente trabalho visa avaliar a atividade schistosomicida dos extratos etanólicos cru dos galhos, folhas, flores e frutos de Handroanthus impetiginosus (Mart ex DC.) Masttos e caracterizar seu perfil metabólico por análise de UPLC-ESI-QTOF. Avaliações dos extratos vegetais em S. mansoni foram conduzidos em vermes adultos in vitro, pelos quais a taxa de mortalidade foi quantificada, e danos no tegumento dos vermes foram monitorados. Todos os extratos induziram mudanças na viabilidade de vermes adultos de S. mansoni, causando a morte desses parasitas, o que foi diretamente dependente da concentração.

Abstract A new lectin from marine sponge Ircinia strobilina, denominated IsL, was isolated by combination of affinity chromatography in Guar gum matrix followed by size exclusion chromatography. IsL was able to agglutinate native and enzymatically treated rabbit erythrocytes, being inhibited by galactosides, such as α-methyl-D-galactopyranoside, β-methyl-D-galactopyranoside and α-lactose. IsL hemagglutinating activity was stable at neutral to alkaline pH, however the lectin loses its activity at 40° C. The molecular mass determinated by mass spectrometry was 13.655 ± 5 Da. Approximately 40% of the primary structure of IsL was determined by mass spectrometry, but no similarity was observed with any protein. The secondary structure of IsL consists of 28% α-helix, 26% β-sheet, and 46% random region, as determined by dichroism circular. IsL was a calcium-dependent lectin, but no significant variations were observed by circular dichroism when IsL was incubated in presence of calcium and EDTA. IsL was not toxic against Artemia nauplii and did not have antimicrobial activity against bacterial cells. However, the IsL was able to significantly inhibit the biofilm formation of Staphylococcus aureus and Staphylococcus epidermidis.

Abstract Coffea sp. is cultivated in many tropical countries. Brazil has always adopted intensive agricultural practices, but organic coffee farming is an alternative system based on the non-use of agrochemicals and the rational management of soils. Metabarcoding 16S analysis using next-generation sequencing has been developed to identify and compare the diversity of the Coffea arabica L. rhizospheric bacterial community in two farming areas in São Paulo, Brazil. Dourado uses conventional farming, while Ribeirão Corrente uses organic. We found broad taxonomic composition, with sequences from 24 phyla, 55 classes, 61 orders, 146 families, and 337genus. The three most abundant phyla were Proteobacteria (38.27%), Actinobacteria (15.56%), and Acidobacteria (16.10%). In organic farming, the top 3 were the family Sphingomonadaceae, order Rhizobiales, genus Nocardioides, and Gp6. The genus Gp2 and the phylum Candidatus Saccharibacteria were the most abundant OTUs exclusively present in conventional farming. In the organic farming practice, Proteobacteria, Actinobacteria, and Acidobacteria were also present among the exclusive OTUs; we also found OTUs belonging to Bacteroidetes, Firmicutes, and Verrucomicrobia. Our study indicates a positive effect of organic farming on microbial communities. Fertilization may directly affect soil microbiota, suggesting that a large and active microbial community low in functional diversity might not adapt to new climatic conditions. A diverse community could provide better resilience to environmental changes, improving the productivity of this important crop.

Resumo Coffea sp. é cultivada em muitos países tropicais. O Brasil sempre adotou práticas agrícolas intensivas, mas a cafeicultura orgânica é um sistema alternativo baseado na não utilização de agrotóxicos e no manejo racional dos solos. A análise Metabarcode 16S utilizando o sequenciamento de última geração foi desenvolvida para identificar e comparar a diversidade da comunidade bacteriana rizosférica de Coffea arabica L. em duas áreas de cultivo em São Paulo, Brasil. Dourado usa agricultura convencional, enquanto Ribeirão Corrente usa agricultura orgânica. Encontramos ampla composição taxonômica, com sequências de 24 filos, 55 classes, 61 ordens, 146 famílias e 337 gêneros. Os três filos mais abundantes foram Proteobacteria (38,27%), Actinobacteria (15,56%) e Acidobacteria (16,10%). Na agricultura orgânica, os 3 primeiros foram a família Sphingomonadaceae, ordem Rhizobiales, gênero Nocardioides e Gp6. O gênero Gp2 e o filo Candidatus Saccaribacteria foram as OTUs mais abundantes exclusivamente presentes na agricultura convencional. Na prática da agricultura orgânica, Proteobacteria, Actinobacteria e Acidobacteria também estiveram presentes entre as OTUs exclusivas; também encontramos OTUs pertencentes a Bacteroidetes, Firmicutes e Verrucomicrobia. Nosso estudo indica um efeito positivo da agricultura orgânica nas comunidades microbianas. A fertilização pode afetar diretamente a microbiota do solo, sugerindo que uma grande e ativa comunidade microbiana com baixa diversidade funcional pode não se adaptar às novas condições climáticas. Uma comunidade microbiana diversificada poderia proporcionar maior resiliência às mudanças ambientais, melhorando a produtividade desta importante cultura agrícola.

Abstract The toxic potential of dithiocarbamates fungicides widely used in world agriculture is well known, among which Mancozeb is one of the most used. This study aimed to evaluate the toxicity of Mancozeb, determining the LC50% of the product and the behavioral and histological changes observed in fish of the Pacamã species through acute and sublethal toxicity tests. The first experiment was carried out on Pacamã fingerlings exposed to dosages of 0.5, 1, 2, 4, and 8mg/L of Mancozeb under the form ManzateWG®, for a total period of 96 hours in the acute experiment, and in the second experiment, fish were subjected to concentrations of 1/10 of those used in the acute experiment (0.05, 0.1, 0.2, 0.4 and 0.8mg/L, respectively), for 15 days in total. The 50% lethal concentration of ManzateWG® was calculated at the end of the acute experiment, presenting a value of 2.29mg/L at 96h for Pacamã fingerlings. A behavioral assessment was carried out through daily observation of the fish during both experiments, and an increase in mucus production was observed, as well as atypical social behavior in those exposed to the toxic agent. Histopathological evaluation was performed on livers collected after the end of the sublethal experiment, and the main hepatic alterations observed were cytoplasmic vacuolization, inflammatory infiltrate, and necrosis. Mancozeb has toxic potential and is capable of generating behavioral changes, as well as increasing the risk of liver damage in Pacamãs exposed to this compound.

Resumo O potencial tóxico dos fungicidas ditiocarbamatos amplamente usados na agricultura mundial é bem conhecido, e entre estes, o Mancozebe é um dos mais utilizados. Esse estudo teve como objetivo avaliar a toxicidade do Mancozebe, determinando a CL50% do produto e as alterações comportamentais e histológicas, observadas em peixes da espécie pacamã através de testes de toxicidade aguda e subletal. O primeiro experimento foi realizado em alevinos de pacamã expostos a dosagens de 0,5, 1, 2, 4 e 8mg/L de Mancozebe sob a forma ManzateWG®, por um período total de 96 horas no experimento agudo, e no segundo experimento os peixes foram submetidos a concentrações de 1/10 daquelas utilizadas no experimento agudo (0,05, 0,1, 0,2, 0,4 e 0,8mg/L, respectivamente), a exposição no segundo experimento foi de 15 dias. A concentração letal a 50% do ManzateWG® foi calculada ao fim do experimento agudo, apresentando um valor de 2,29mg/L às 96h para alevinos de pacamã. Avaliou-se o comportamento por meio da observação diária dos peixes, notando-se o aumento da produção de muco assim como comportamento social atípico naqueles indivíduos expostos ao agente tóxico. A análise histopatológica foi conduzida nos fígados coletados após a conclusão do experimento subletal, identificando as principais alterações hepáticas como vacuolização citoplasmática, infiltrado inflamatório e necrose. O Mancozebe apresenta potencial tóxico e é capaz de gerar alterações comportamentais, assim como aumentar o risco de lesões hepáticas em pacamãs expostos a este composto.
