Processing on the physical and physiological quality of chickpea (Cicer arietinum L.) seeds

Abstract: Processing consists of sequential operations aimed at cleaning, classifying, and treating seeds, with the goal of improving the quality of the lot. The aim of this study was to evaluate the effect of the processing stages on the physical and physiological quality of chickpea seeds, cultivar BRS Cícero. Seeds were sampled before processing and after passing through the air screen machine (upper sieve of 12 mm and lower sieve of 7.5 mm), elevators, storage silo, and gravity table (upper discharge, high intermediate, low intermediate, and lower discharge). The seeds were evaluated for moisture content, physical purity, germination test, electrical conductivity test, emergence in sand, emergence speed index, accelerated aging, and tetrazolium test. The processing resulted in increased physical purity and reduced percentage of broken seeds in the lot of chickpea seeds. Seeds collected from the upper and intermediate chutes of the gravity table showed higher germination values. Seed processing using the air screen machine and the gravity table is effective in improving the physical and physiological quality of the lot of chickpea seeds, cultivar BRS Cícero.
Resumo: O beneficiamento consiste em operações sequenciais que visam limpar, classificar e tratar as sementes, visando a melhoria da qualidade do lote. Objetivou-se com este trabalho avaliar o efeito das etapas de beneficiamento na qualidade física e fisiológica de sementes de grão-de-bico, cultivar BRS Cícero. As sementes foram amostradas antes do processamento e após a saída da máquina de ventilador e peneiras (peneira superior de 12 mm e inferior de 7,5 mm), elevadores, silo armazenador e mesa de gravidade (descarga superior, intermediária alta, intermediária baixa e inferior). As sementes foram avaliadas quanto aos teores de água, pureza física, teste de germinação, teste de condutividade elétrica, emergência em areia, índice de velocidade de emergência, envelhecimento acelerado e teste de tetrazólio. O beneficiamento promoveu o aumento da pureza física e redução do percentual de sementes quebradas no lote de sementes de grão-de-bico. As sementes coletadas nas bicas superiores e intermediárias da mesa de gravidade apresentaram valores superiores de germinação. O beneficiamento de sementes, utilizando a máquina de ar e peneiras e a mesa de gravidade, é eficiente para o aprimoramento da qualidade física e fisiológica do lote de sementes de grão-de-bico cultivar BRS Cícero.

Initiation of breeding programs for three species of Corymbia: Introduction and provenances study

Abstract The objective of this study was to establish populations to start the breeding program of three species of Corymbia that are not widely planted in Brazil yet. To this end, basic density, bark content and growth of seedlots/provenances were evaluated three years after planting. The experiments consisted of 14 treatments for Corymbia citriodora subsp. variegata (CCV), 15 for C. maculata (CM), and five for C. henryi (CH). The species exhibited high survival, indicating adaptation to the Cwa climate while average coefficients of determination of seedlots exhibited intermediate values. The CCV from the Richmond Range (28º 55’ S) exhibited the highest productivity. The mean annual increment was low (22 to 26 m3ha-1), but the basic density (573 to 613 kg m-³) was high compared to the standard for eucalypts in Brazil. The bark content was close to 15% and varied between and within species.

ABSTRACT Background Triatoma infestans (Hemiptera: Reduviidae) is a hematophagous insect and the main vector of Trypanosoma cruzi (Kinetoplastida: Trypanosomatidae). In the present study, the authors investigated whether a serine protease activity from the saliva of T. infestans has a role in vasomotor modulation, and in the insect-blood feeding by cleaving and activating protease-activated receptors (PARs). Methods T. infestans saliva was chromatographed as previously reported for purification of triapsin, a serine protease. The cleavage activity of triapsin on PAR peptides was investigated based on FRET technology. Mass spectrometry was used to analyze the sites of PAR-2 peptide cleaved by triapsin. NO measurements were performed using the DAN assay (2,3-diaminonapthalene). The vasorelaxant activity of triapsin was measured in vessels with or without functional endothelium pre-contracted with phenylephrine (3 µM). Intravital microscopy was used to assess the effect of triapsin on mouse skin microcirculation. Results Triapsin was able to induce hydrolysis of PAR peptides and showed a higher preference for cleavage of the PAR-2 peptide. Analysis by mass spectrometry confirmed a single cleavage site, which corresponds to the activation site of the PAR-2 receptor. Triapsin induced dose-dependent NO release in cultured human umbilical vein endothelial cells (HUVECs), reaching a maximum effect at 17.58 nM. Triapsin purified by gel-filtration chromatography (10-16 to 10-9 M) was applied cumulatively to mouse mesenteric artery rings and showed a potent endothelium-dependent vasodilator effect (EC30 = 10-12 M). Nitric oxide seems to be partially responsible for this vasodilator effect because L-NAME (L-NG-nitroarginine methyl ester 300 µM), a nitric oxide synthetase inhibitor, did not abrogate the vasodilation activated by triapsin. Anti-PAR-2 antibody completely inhibited vasodilation observed in the presence of triapsin activity. Triapsin activity also induced an increase in the mouse ear venular diameter. Conclusion Data from this study suggest a plausible association between triapsin activity mediated PAR-2 activation and vasodilation caused by T. infestans saliva.

Protective effects of bark ethanolic extract from Spondias dulcis Forst F. against DNA damage induced by benzo[a]pyrene and cyclophosphamide

Abstract This study evaluated the genotoxicity, mutagenicity, antigenotoxicity, and antimutagenicity effects on biochemical parameters of oxidative stress of the Spondias dulcis bark ethanolic extract on mice. The extract was evaluated in the doses of 500, 1000, and 1500 mg/kg bw via gavage. To evaluate the protective effects of the extract, benzo[a]pyrene (B[a]P) and cyclophosphamide (CP) were chosen as DNA damage inducers. Genotoxicity and antigenotoxicity were evaluated by the comet assay. Cytotoxicity, mutagenicity, and antimutagenicity were evaluated by the micronucleus test in bone marrow and peripheral blood. The biochemical parameters of oxidative stress were evaluated by the quantification of catalase activity (CAT) and reduced glutathione (GSH) in total blood, liver and kidney, and malondialdehyde (MDA), in liver and kidney. No genotoxic, cytotoxic, or mutagenic effect was found on mice exposed to the extract. The extract depleted the number of damaged nucleoids in total blood and the number of micronucleus (MN) in both cell types. The extract was able to increase CAT activity and GSH levels and decrease MDA levels after treatment with B[a]P and CP. The results indicate that the S. dulcis extract has potential to be used as preventive compound against DNA damage caused by CP and B[a]P.

Roles of the Taql and Bsml vitamin D receptor gene polymorphisms in hospital mortality of burn patients

OBJECTIVE: The aim of this study was to evaluate the roles of the Taql and Bsml vitamin D receptor gene polymorphisms in hospital mortality of burn patients. METHODS: In total, 105 consecutive burn injury patients over 18 years in age who were admitted to the Burn Unit of Bauru State Hospital from January to December 2013 were prospectively evaluated. Upon admission, patient demographic information was recorded and a blood sample was taken for biochemical analysis to identify the presence of the Taql(rs731236) and Bsml(rs1544410) polymorphisms. All of the patients were followed over their hospital stay and mortality was recorded. RESULTS: Eighteen of the patients did not sign the informed consent form, and there were technical problems with genotype analysis for 7 of the patients. Thus, 80 patients (mean age, 42.5±16.1 years) were included in the final analysis. In total, 60% of the patients were male, and 16.3% died during the hospital stay. The genotype frequencies for the Taql polymorphism were 51.25% TT, 41.25% TC and 7.50% CC; for the Bsml polymorphism, they were 51.25% GG, 42.50% GA and 6.25% AA. In logistic regression analysis, after adjustments for age, gender and total body surface burn area, there were no associations between the Taql (OR: 1.575; CI95%: 0.148-16.745; p=0.706) or Bsml (OR: 1.309; CI95%: 0.128-13.430; p=0.821) polymorphisms and mortality for the burn patients. CONCLUSIONS: Our results suggest that the Taql and Bsml vitamin D receptor gene polymorphisms are not associated with hospital mortality of burn patients.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Ventriculectomia parcial: um novo conceito no tratamento cirúrgico de cardiopatias em fase final

A melhora clínica da função cardíaca pós aneurismectomia de ventrículo esquerdo e/ou cardiomioplastia com o músculo grande dorsal parece ser, ao menos parcialmente, devida ao remodelamento do ventrículo esquerdo. Através de pesquisa em nosso laboratório experimental com carneiros, demonstramos que o aumento do diâmetro do ventrículo é mais importante que a perda de massa muscular para a deterioração da função ventricular. Sabendo-se que em miocardiopatia dilatada não ocorre aumento de massa muscular, reduzimos o diâmetro do ventrículo para o normal, em uma série consecutiva de pacientes com esta lesão. No período de 1984 a 1995, foram operados com esta nova técnica, denominada, então, "Ventriculectomia Parcial", 103 pacientes portadores de miocardiopatias complexas e insuficiência cardíaca congestiva (NYHA IV). A operação é baseada na lei de Laplace (T=P.11.D) e consistiu na remoção de uma fatia de músculo da parede lateral do ventrículo esquerdo, iniciando-se na ponta deste, estendendo-se entre os músculos papilares e terminando próximo ao anel mitral. A cirurgia é realizada sob circulação extracorpórea normotérmica e não se utiliza cardioplegia. "Todos os pacientes foram avaliados pré-operatoriamente com ecodopplercardiografia e ventriculografia digital, os quais revelaram fração de ejeção < 20%, confirmando estes pacientes como candidatos ao transplante cardíaco. A maioria era do sexo masculino (n=73) e a idade variou de 19 a 74 anos. As doenças foram: miocardite a virus (n=12); pós miocardioplastia (n=1); doença de Chagas (n=15); doença valvar (n=38); doença isquémica (n=16); idiopática (n=21). Óbitos hospitalares (ocorridos nos primeiros 30 dias da cirurgia) (n=13): embolia pulmonar (n=4); insuficiência renal (n=5); sangramento (n=4). Óbitos tardios (ocorridos depois do 30º dia de cirurgia) (n=10): arritmia (n=6); "insuficiência cardíaca" (n=2); causa desconhecida (n=2); 8 pacientes precisaram ser reoperados por sangramento. Não houve infecção e nenhum paciente precisou balão intra-aórtico. Todos saíram com nitroprussiato e 19 pacientes, com inotrópicos. A ventriculografia e a ecocardiografia pós-op mostraram melhora acentuada da FE (de 100% a 300%). Em conclusão, a nova técnrca "Ventriculectomia Parcial", com o objetivo de reduzir o diâmetro do ventrículo esquerdo, pode beneficiar pacientes em estágio final de cardiopatia dilatada. Este novo conceito pode, na nossa experiência, proporcionar ao paciente melhora clínica significativa e prolongamento de sua vida.
The clinical improvement of cardiac function post left ventricular aneurysmectomy and/or cardiomyoplasty with the latissimus dorsi muscle seems to be due to the remodeling of the left ventricle. We did some experiments with sheep in our laboratory and we concluded that the increment of ventricular diameter is more deleterious than loss of muscle mass to the ventricular function. Since there is no increment in muscle mass to the dilated hearts, we reduced their diameter to accomplish a better ventricular function in a series of very sick patients with dilated hearts. From 1984 to 1995, 103 patients with complex cardiac problems and end stage congestive heart failure (NYHA IV) with dilated cardiomyopathy underwent "Partial Ventriculectomy". The operation was based on Laplace's law (T-P.II.D) and consisted of removal of a large wedge of muscle from the lateral wall of the left ventricle beginning at the apex and extending between the papillary muscles, ending proximal to the mitral annulus. The operation was performed under normothermic cardiopulmonary bypass and no cardioplegia was used. Pre-operative left ventriculography and echocardiography showed global hypocontractility of the hearts with ejection fraction lower than 20% in all patients who were considered candidates for cardiac transplantation. Males were predominant (n=73) and the age varied from 19 to 74 years. The pathologies were: viral miocarditis (n=12); post miocardioplasty (n=1); Chagas' disease (n=15); valvular disease (n=38); ischemic disease (n=16); idiopathic (n=21). Hospital mortality (30 days post operative) (n=13): pulmonary embolus (n=4); renal failure (n=5); bleeding (n=4). Late deaths (after 30 days post-operative) (n=10): arhythmia (n=6); "cardiac failure" (n=2); unknown (n=2); 8 patients required reoperation for bleeding. There were no infection and none required IAPB. All patients had nipride and 19 needed inotropics. Post-op ventriculographies and echocardiographies showed improvement of EF (from 100% to 300%). In conclusion, this new technique "Partial Ventriculectomy", with it's purpose of reducing the ventricular diameter, may benefit patients with end stage heart failure. And this new concept may, in our experience, give the patient improvement in his clinical status and increase life span.
