The aim of this study was to assess the effect of calcitic seaweed (CS), calcitic limestone (CL) and monodicalcium phosphate (DP) fed to piglets in various combinations on their gastrointestinal tract and bones. A total of 128 piglets (21 days old, 5.50 ± 0.49 kg bodyweight (BW) were assigned to treatments in a randomized complete block design and eight replications to one of four diets: D1: with CL + DP, D2: with CL, CS + DP, D3, similar to D2, but with 30% less calcium from CS and DP, and D4: with CS + DP. Treatments affected the metatarsal bones, mineral concentration in the heart and liver, and pH in the cecum and colon. Piglets fed D4 tended to have a greater Enterobacteriaceae count in the jejunum and ileum compared with D1 (4.82 vs 4.79 CFU/g). Piglets fed D1 and D3 had a greater Enterobacteriaceae count in their cecum than D4 (4.79 and 4.80 vs 4.76). The D2 and D1 treatments produced greater crypt depth (CD) in the duodenum and ileum compared with D3 and D4, respectively. Feeding D3 resulted in a greater villus height (VH) to CD ratio in the duodenum compared with D2 and showed a 24.5% increase in heart weight compared with fed D1. In conclusion, CS could be an alternative source of calcium source for piglets. The inconsistent findings of the present study suggest the need for further studies to better understand the interplay of effects of Ca + source and level on its metabolism.

Abstract Objective: To present an updated and evidence-based guideline for the use of dual-energy x-ray absorptiometry (DXA) to assess body composition in clinical practice. Materials and methods: This Official Position was developed by the Scientific Committee of the Brazilian Association of Bone Assessment and Metabolism ( Associação Brasileira de Avaliação Óssea e Osteometabolismo , ABRASSO) and experts in the field who were invited to contribute to the preparation of this document. The authors searched current databases for relevant publications in the area of body composition assessment. In this second part of the Official Position, the authors discuss the interpretation and reporting of body composition parameters assessed by DXA and the use of DXA for body composition evaluation in special situations, including evaluation of children, persons with HIV, and animals. Conclusion: This document offers recommendations for the use of DXA in body composition evaluation, including indications, interpretation, and applications, to serve as a guiding tool in clinical practice and research for health care professionals in Brazil.

Resumo O objetivo do presente trabalho foi avaliar características qualitativas e quantitativas do sêmen de surubim do Iguaçu (Steindachneridion melanodermatum). Para as análises, utilizaram-se onze machos com peso médio de 1,76±0,48 kg e idade média de dois anos, submetidos ao tratamento de hipofisação. O volume médio de sêmen coletado foi de 1,34 ± 0,73 mL. O tempo médio de motilidade dos espermatozoides após a coleta foi de 154,4 ± 72,6 e 149,0 ± 77,5 segundos para ativação com água do tanque e água destilada, respectivamente. Para a concentração espermática, a média foi de 5,423 ± 2,155 x1010 espermatozoides/mL. Os resultados indicam que o sêmen de S. melanodermatum é facilmente obtido no período reprodutivo analisado, e que as características seminais avaliadas são adequadas para reprodução induzida.

Abstract The objective of the present work was to assess the qualitative and quantitative characteristics of semen from the surubim do Iguaçu (Steindachneridion melanodermatum). Induced spermiation was achieved in eleven males with mean weight of 1.76 ± 0.48 kg and average age of two years and semen was collected by stripping. The average volume was 1.34 ± 0.73 mL. The duration of sperm motility was 154.4 ± 72.6 and 149.0 ± 77.5 seconds after activation with hatchery water and distilled water, respectively. The sperm concentration estimated by hemocytometer was 5.423 ± 2.155 x 1010 spermatozoa/mL. The results indicate that S. melanodermatum semen is easily obtained during the spawning season and the seminal characteristics are adequate insemination and subsequent in vitro fertilization.

RESUMO Trichomycterus é um especioso gênero dentro de Trichomycterinae e exibe marcante diversidade cariotípica. No entanto, o conhecimento sobre sua estrutura genômica e localização de seqüências repetitivas ainda é restrita. Para um melhor conhecimento sobre a sua diversificação cariotípica, nós analisamos nove especies de Trichomycterus usando técnicas de citogenética clássica e molecular. Os resultados revelaram um conservado número diploide de 2n = 54 cromossomos em todas as espécies analisadas, embora diferentes marcações na distribuição da heterocromatina constitutiva tenham sido observadas. Além disso, enquanto o DNAr 18S mostrou um padrão de distribuição conservado, os sítios de DNAr 5S mostraram uma localização bastante diversa, considerando as espécies analisadas. Ambos os genes ribossomais foram co-localizados em todas as espécies, exceto em T. iheringi , sugerindo que a co-localização é provavelmente uma condição ancestral em Trichomycterus . Finalmente, três espécies analisadas mostraram cromossomos B heterocromáticos, reforçando uma intensa reoganização genômica ocorrendo em Trichomycterus . Nossos resultados mostraram que variações cromossômicas não estão restritas à diferenças na fórmula cariotípica, como proposto anteriormente, mas também às alterações a níveis de resolução estrutural.

Trichomycterus is a specious fish genus within Trichomycterinae and displays remarkable karyotype diversity. However, knowledge about their genomic structure and location of repetitive sequence is still limited. In order to better understand the karyotype diversification, we analyzed nine species of Trichomycterus using classical and molecular cytogenetic techniques. Results revealed a conserved diploid chromosome number of 2n=54 chromosomes in all analyzed species, although remarkable differences on the constitutive heterochromatin distribution were observed. In addition, while the 18S rDNA showed a conserved distribution pattern, the 5S rDNA sites showed a quite diverse location considering the analyzed species. Remarkably, both ribosomal genes were co-located in all species, except in T . iheringi , suggesting that co-localization is probably an ancestral condition in Trichomycterus . Finally, three analyzed species showed heterochromatic B chromosomes, reinforcing the intense genomic reorganization occurring in Trichomycterus . Our results showed that chromosomal variations are not restricted to differences in karyotype formula as previously proposed, but also to modifications on the microstructural level of resolution.

Para avaliar se a qualidade microbiológica da água de dessedentação intervém na morfologia intestinal de frangos de corte, foram analisados o consumo semanal de água, a microbiologia de amostras de água, a microscopia eletrônica de varredura e a histologia do intestino delgado de frangos de corte tratados com água filtrada e não filtrada. Os frangos que ingeriram água filtrada tiveram acesso ao menor número de micro-organismos fecais (2,52±0,99 Número Mais Provável (NMP) de coliformes fecais e 1,17±1,25 NMP de Escherichia coli) em relação aos que ingeriram água não filtrada (3,62±0,67 NMP e 2,53±1,13 NMP). Aos 14, 21 e 45 dias de vida, foram colhidas amostras do duodeno, jejuno e íleo de 96 aves. Após rotina laboratorial, as amostras conservadas em glutaraldeído foram eletronmicrografadas e analisadas quanto à densidade de vilos e o material mantido em solução de Bouin foi destinado à confecção de lâminas histológicas que foram analisadas morfometricamente. O duodeno das aves que receberam água não filtrada apresentou maior densidade dos vilos em resposta à qualidade microbiológica da água. Na morfometria intestinal, observou-se que aves que receberam água não filtrada apresentaram aumento na profundidade das criptas intestinais e elevada altura das vilosidades em relação às aves que ingeriram água filtrada. Infere-se que a água filtrada, oferecida aos frangos de corte em um período de vida de 45 dias, favorece a manutenção da integridade intestinal.

To evaluate if microbiological quality of drinking water has an effect on intestinal morphology of broilers, the weekly water consumption by them was verified, and microbiological analysis of water samples, scanning electron microscopy and small intestine histology of broilers treated with filtered and not filtered water was conducted. Chickens that ingested filtered water had access to fewer fecal microorganisms (2.52±0.99 Most Probable Number MPN of fecal coliforms and 1.17±1.25 MPN of Escherichia coli) compared to those who drank no filtered water (3.62±0.67 and 2.53±1.13 MPN). At 14, 21 and 45 days old, the duodenum, jejunum and ileum of 96 birds were sampled. After laboratory routine, samples preserved in glutaraldehyde were eletronmicrographed and evaluated by villous density, and the material maintained in Bouin's solution was destinated to histological slides that were analyzed morphometrically. The duodenum of birds that ingested not filtered water had the highest density of villi in response to microbiological water quality. In intestinal morphometry, were observed that birds receiving not filtered water showed increase in intestinal crypts depth and presented larger villi compared with birds that ingested filtered water. It is cocluded that filtered water, offered to broilers in a life span of 45 days, favors the maintenance of intestinal integrity.

Individual samples from five populations of the characid fish Astyanax scabripinnis (Jenyns, 1842) from the Tietê and Paranapanema river basins (Brazil) were studied. All individuals analyzed presented 50 chromosomes but three different karyotypic forms were observed. Additionally, some individuals of one karyomorph presented a macro supernumerary chromosome. No chromosomal differentiation was observed between males and females in any sample analyzed. C-banding revealed two distinct distribution patterns in these populations in which strong terminally located heterochromatic blocks were detected in the long arms of some chromosomes of the specimens from Cascatinha, Cintra, and Funari streams, whereas populations of the Capão Bonito stream and the Barbosa waterfall revealed less evident heterochromatic blocks on the chromosomes. The comparative study of the 18S rDNA localization by fluorescent in situ hybridization, and detection of active nucleolus organizing regions by silver stain (Ag-NORs) showed differences in rDNA content and expression of this gene site among the individuals analyzed. Chromosome mapping of the 5S rDNA revealed the presence of four sites located in two distinct chromosomal pairs, with no apparent differences among karyomorphs. Based on the biogeographical distribution and specific biological characteristics of the species, the data focus on the chromosome differentiation mechanisms involved in the speciation process acting on the populations of the Astyanax scabripinnis species complex.

Avaliaram-se o comprimento do infundíbulo, do magno, do istmo, do útero e da vagina e o número de pregas do magno e do istmo do oviduto de 20 marrecas Ana boschas na fase reprodutiva. O infundíbulo apresenta mucosa com pregas longitudinais e baixas, revestidas por epitélio pseudoestratificado cilíndrico ciliado, com células caliciformes. O magno, compartimento mais longo do oviduto, 25,38cm±3,20, encontra-se constituído por uma camada mucosa com pregas altas e espessas revestidas por células cilíndricas ciliadas e abundantes células caliciformes. O istmo é formado por uma mucosa com pregas estreitas e curtas e numerosas glândulas tubulares que se estendem para o interior da lâmina própria. O útero, região curta do oviduto, 5,25cm±1,26, apresenta parede com pregas e cristas baixas e numerosas glândulas tubulares enoveladas, dirigidas para o interior da lâmina própria. A vagina, um estreito tubo muscular, está constituído por oito anéis circulares, em média, e uma camada muscular altamente desenvolvida e espessa. A morfologia do oviduto da marreca apresenta características morfológicas e histológicas distintas dos galiniformes, observando-se que a vagina e a porção cranial do infundíbulo apresentam pregas e células caliciformes, respectivamente, sendo estas últimas estruturas ausentes nos galiniformes.

The length of infundibulum, magnum, isthmus, uterus, vagina, and the number of oviduct, magnum, and isthmus folds were evaluated in 20 Ana boschas female ducks in the reproductive phase. The infundibulum presented mucous membrane with longitudinal and short folds, covered by ciliated columnar pseudostratified epithelium, with goblet cells. Magnum, the longest oviduct compartment, 25.38cm±3.20, is constituted by mucous membrane with high and thick folds that are covered by ciliated columnar cells and many goblet cells. Isthmus is formed by mucous membrane with narrow and short folds, and many tubular glands that extend inside lamina propria. Uterus, an oviduct short region, 5.25cm±1.26, presented surface with short folds and crests, and numerous reeled tubular glands, that are directed inside lamina propria. Vagina, a strait muscular tube, is constituted by, approximately, eight circular rings, and a very developed and thick muscular layer. The oviduct morphology of female ducks presented different morphological and histological characteristics from Galliformes, because vagina and infundibulum cranial portion present folds and goblet cells, respectively, and the last cited structures are absent in Galliformes.

O ducto epididimário (DE) de codorna doméstica mostrou, ao longo do ano, variabilidade pequena, porém muito expressiva no outono, o qual corresponde à fase quiescente do ciclo testicular anual. A morfologia do DE na primavera foi, em termos, similar à verificada no verão e inverno. Nestas fases notaram-se aumento significante do calibre tubular do DE; estocagem intraluminal de espermatozóides e ocorrência de mitocôndrias, lamelas do RE, vesículas variáveis quanto à forma, dimensões e conteúdos e presença de alguns lisossomos localizados, principalmente, no citoplasma apical das células principais (P), no epitélio epididimário. Estas características ultra-estruturais das células P parecem ser indicativas da ocorrência de processos ativos de endocitose e de secreção micromerócrina. A quiescência outonal foi caracterizada pelo aspecto anfractuoso do DE; ausência de espermatozóides e pouco material intraluminal, observados à microscopia de luz. Características ultra-estruturais degenerativas foram verificadas ao nível do citoplasma supranuclear das células P epididimárias no outono.

Small but expressive variability was noted on the epididymidis duct (ED) of domestic quail along the year, with more evidence in autumn of the quiescent phase of the annual testis cycle in this species. Spring features of ED had a general similar pattern in summer and winter. They were characterized by enlargement of epididymis tubule, storage of spermatozoa into the luminal compartment and presence of mitochondria, ER lamellae, several variable vesicles, and lysosomes localized mainly on the apical cytoplasm of principal cells (P) of the epididymal epithelium. These P cells features indicated a process of endocytosis and perhaps protein secretion. Autumn quiescence was marked by a convolute pattern of the epididymis tubule, lacking of spermatozoa and small amount of exfoliate heterogeneous material inside the luminal compartment at light microscopy. Ultrastructural degenerative features mainly apical cytoplasmic debris were seen in the supranuclear cytoplasm of lining P cells.

O presente trabalho foi desenvolvido para testar a hipótese de que células luteínicas bovinas em cultivo, provenientes dos três terços de gestação, comportam-se da mesma maneira que células in vivo em relação à produção de P4. Foram coletadas amostras de corpos lúteos (CL) de 90 (n=3), 150 (n=3) e 210 (n=3) dias de gestação obtidos em abatedouro. Sob condições assépticas, as células foram mecanicamente dispersas e cultivadas em placas de 96 poços. Após 24 horas de cultivo foram feitas a lavagem dos poços e a adição do precursor pregnenolona. Os tratamentos foram realizados em octuplicata para cada tempo de tratamento (24, 48 e 96 horas) com três repetições de cada período gestacional. As amostras de meio de cultura e as células foram coletadas 24, 48 e 96 horas após adição do precursor e acondicionadas em freezer a -20ºC até o processamento. A progesterona foi dosada através de radioimunoensaio e o conteúdo protéico pelo método de Lowry. Os resultados foram analisados estatisticamente e considerados diferentes quando p<0.05. Foi observada maior produção de P4 aos 90 dias de gestação (35,277±0,075), posterior decréscimo aos 150 dias (28,820±0,231) e novo aumento aos 210 dias (32,777±0,099). A produção de P4 em células cultivadas por 24 horas foi maior (p<0,05) em células oriundas do grupo de 90 dias (2,912±0,047) quando comparado a 150 (2,669±0,137) e 210 dias (2,741±0,088). As 48 e 96 horas de cultivo, células luteínicas bovinas de 90 dias produziram mais P4 que células de 210 dias (2,934±0,029 e 2,976±0,121 respectivamente x 2,760±0,059 e 2,695±0,149, respectivamente; p<0,05), que por sua vez produziram mais do que células de 150 dias (2,334±0,084 para 48 horas e 2,205±0,136 para 96 horas). Aos 150 dias de gestação a produção de progesterona apresentou diminuição gradativa ao longo das 96 horas de cultivo. Essas diferenças podem ser explicadas pela expressão gênica diferencial de enzimas ou também de fatores presentes na cascata esteroidogênica de acordo com a idade gestacional. Este modelo de cultura celular luteínica poderá ser utilizado em estudos funcionais uma vez que o padrão de secreção de P4 mimetizou o que ocorre in vivo.

The aim was to test the hypothesis that cultivated bovine luteal cells from three different thirds of pregnancy behave the same way as in vivo luteal cells relative to P4 production. Corpus luteum samples from days 90 (n=3), 150 (n=3) and 210 (n=3) of pregnancy were obtained at a local slaughterhouse. Under aseptic conditions cells were mechanically dispersed and cultivated in a 96 wells-plate. After 24 hours of culture, cells were washed and the precursor pregnenolone was added. Experiments were conducted eight times for each studied time period (24, 48 and 96 h) and three times for each gestational age. Culture medium and cells were collected after 24, 48 and 96 hours of precursor addition and kept frozen at -20ºC until processing. Progesterone was measured by RIA and protein content by Lowry's method. Results were statistically analyzed and considered different when p <0.05. A higher P4 production was observed on day 90 of gestation (35.277±0.075), then this production was decreased at day 150 (28.820±0.231) and increased again at day 210 (32.777±0.099). After 24 hours of culture, luteal cells P4 production reached maximum values in the group of 90 days (2.912±0.047) when compared to 150 (2.669±0.137) and 210 days (2.741±0.088). At 48 and 96 hours of culture, bovine luteal cells from day 90 of gestation produced more P4 than cells from day 210 (2.934±0.029 and 2.976±0.121 respectively x 2.760±0.059 and 2.695±0.149, respectively; p<0.05), which in turn, produced more P4 than cells from day 150 (2.334±0.084 for 48 h and 2.205±0.136 for 96 h). Luteal cells from day 150 of gestation presented a decreasing P4 production throughout the 96 hours of culture. These differences could be explained by differential gene expression of enzymes and/or factors belonging to the esteroidogenic cascade in accordance to the gestational period. The established luteal cell culture model could be used for further functional studies once P4 secretion pattern in vitroresembled what occurs in vivo.

Astyanax altiparanae, recentemente descrita, é comum na bacia do alto rio Paraná, tendo sido registrada recentemente na bacia do rio Iguaçu. Entretanto, sua origem natural no rio Iguaçu é questionável. No presente trabalho algumas características cariotípicas e morfológicas de duas populações de Astyanax altiparanae do rio alto rio Tibagi e alto rio Iguaçu foram comparadas. Ambas as populações mostraram 2n=50 cromossomos e diferenças nas suas fórmulas de cariotípicas, cromossomos portadores das regiões organizadoras de nucléolos e localização da heterocromatina. Dados morfométricos das duas populações foram analisados para variáveis canônicas livres de tamanho. Os dados citogenéticos e morfológicos foram coincidentes e mostraram marcas exclusivas que refletem o grau de isolamento populacional. Em adição a outros dados geográficos, morfológicos e moleculares em populações de A. altiparanae do baixo rio Iguaçu e tributários do rio Paraná acima da represa de Itaipu (Sul do Brasil), os presentes resultados indicam que as duas populações analisadas neste estudo pertençam a diferentes estoques. A presença desta espécie ao longo da bacia do rio Iguaçu pode ser uma conseqüência de uma história evolutiva complexa e pouco conhecida.

The recently described fish species, Astyanax altiparanae (tetra) is common in the upper rio Paraná basin, and has been reported in the Iguaçu basin. However, its natural origin in the rio Iguaçu is questionable. In the present work, karyotypical and morphological features of two populations of Astyanax altiparanae from the upper rio Tibagi and upper rio Iguaçu were compared. Both populations showed 2n=50 chromosomes and differences in their karyotype formula, NOR-bearing chromosomes and location of heterochromatin. Morphometric data from both populations were analyzed through free-size canonical variables. Cytogenetic and morphological results were mostly coincident showing exclusive markers that reflect their degree of populational isolation. In addition to other geographic, morphological and molecular data for A. altiparanae populations from the lower rio Iguaçu and rio Paraná tributaries upstream from the Itaipu Dam (South Brazil), the present results indicate that the two populations analyzed in this study belong to different stocks. The presence of this species along the rio Iguaçu basin would be a consequence of a complex and poorly understood evolutionary history.

Light microscopy analysis of the mural structure of the common carotid artery, internal carotid artery and external carotid artery in mongrel dog revealed variability in the middle values of vascular diameter and thickness of the intimal plus medial coats and adventitial coat. The vascular diameter did not differ between the internal carotid artery and external carotid artery, but was significantly increased in the common carotid artery from which the other two arteries had origin. The increased thickness of the intimal plus medial coats of the common carotid artery revealed the constant mechanical adjustment of the arterial wall and local shearing stress that occurred among the arterial coats. The adventitial coat of the internal carotid artery was significantly thicker than those of the common carotid and external carotid arteries, with no significant difference noted between the latter arteries. These histomorphometric results were related to the qualitative observations regarding the mural structure of the three arteries analyzed, especially regarding to the increased thickness and structural complexity of the adventitial coat of the internal carotid artery. Perhaps it acted as an external protective sheath of the vascular wall during its long course until the carotid channel

El análisis de la estructura de las arterias carótidas, común, externa y interna en el perro sin raza definida, a través del microscopio fotónico mostró variabilidad en los valores promedios de los parámetros: diámetro vascular y grosor de las capas íntima, media y adventicia de cada arteria, respectivamente. El diámetro vascular no difirió significantemente entre las arterias carótidas interna y externa, pero mostró aumento significativo en la arteria carótida común, la cual da origen a los dos otros vasos sanguíneos. El grosor aumentado de las capas íntima y media de la arteria carótida común, más directamente sometida a la fuerza de la presión sistólica cardiaca, significó los ajustes constantes de la pared vascular a las alteraciones de presión y al estrés de deslizamiento controlado entre las capas de la pared arterial. El grosor de la adventicia de la arteria carótida interna fue significativamente mayor que la de las otras arterias carótidas (común y externa), las cuales, entre, sí no tuvieron diferencia con respecto de la adventicia. Estos resultados morfométricos estaban directamente correlacionados con las observaciones morfológicas de naturaleza cualitativa, especialmente, en relación al incremento relativo de la adventicia de la carótida interna ya apreciable en las microfotografías. Tal vez, la adventicia en la arteria carótida interna actúa como una capa conjuntiva protectora, durante el complejo proceso de avance de esta arteria desde su origen cervical hasta el nivel del canal carotídeo donde alcanza la cavidad craneana

Dados cariotípicos são apresentados para duas espécies simpátricas de Corydoras da Lagoa Dourada, C. ehrhadti e C. paleatus, pertencentes à bacia do alto Rio Tibagi (Ponta Grossa, Paraná, Brasil). O mesmo número diplóide e fórmula cariotípica foram observados em ambas espécies/populações. Grande similaridade foi verificada também para a distribuição da heterocromatina constitutiva e atividade das regiões organizadoras de nucléolos. O emprego da hibridação in situ com sonda fluorescente de DNAr 18S possibilitou identificar as espécies/populações por meio da localização dos sítios ribossomais.

Karyotypic data are presented for two sympatric Corydoras species of the Lagoa Dourada, namely, C. ehrhadti and C. paleatus, which are found in the upper Tibagi river basin (Ponta Grossa, State of Paraná, Brazil). The same diploid number and karyotypic formula were observed in both species/populations. A great similarity in the constitutive heterochromatin distribution and in the activity of nucleolar organizer regions was also found. The use of in situ hybridization with a fluorescent 18S rDNA probe allowed for the identification of the species/populations through the location of ribosomal sites.

The phenomenon of electromagnetically induced transparency (EIT) was discovered by Adriano Gozzini and coworkers in 1976 in Pisa. Novel schemes to investigate and exploit EIT in the optical domain have attracted much interest both in atomic and solid state systems. We discuss some of our recent theoretical results, in particular: i) a well developed EIT regime based on free exciton levels in undoped bulk crystalline Cu2O; ii) light dragging effects in moving media under EIT; iii) the coherent control of Cherenkov radiation in the EIT regime.
