ABSTRACT No information regarding pathogenic variability among different isolates of Ralstonia solanacearum infecting chili from different agro-ecological zones of Pakistan with varying climatic and edaphic factors is available. Therefore, in the present study, variations were observed in biovar distribution, hypersensitive response, growth and virulence among 114 isolates of R. solanacearum collected from eight agro-ecological zones of Pakistan. Out of 114 R. solanacearum isolates, 81% were identified as Biovar III, while the remaining 19% were recognized as Biovar IV. Of all the 114 isolates of R. solanacearum, 77% showed positive hypersensitive response and mucoid growth, while 23% isolates gave negative hypersensitive response with non-mucoid growth. Out of 114 isolates of R. solanacearum consisting of Biovar III and IV, 22.8% were found avirulent, 25.4% weakly virulent, 29.8% virulent, and the remaining 21.9% were highly virulent. Variations among 114 R. solanacearum isolates were also observed in four provinces of the country. Among 92 R. solanacearum Biovar III isolates, 21.7% were identified as avirulent, 25% weakly virulent, 34.4% virulent, and 22.8% were highly virulent in the eight agro-ecological zones of the country. Similarly, out of 22 R. solanacearum Biovar IV isolates, 27.3% were detected as avirulent, weakly virulent and virulent, while 18.2% isolates were found highly virulent. The isolates having non-mucoid growth and negative hypersensitive response were found avriulent, while those with mucoid growth and positive hypersensitive response were weakly virulent to highly virulent. The information will help design control strategies accordingly and develop resistant cultivars against the bacterium.

The goal of this research was to investigate the echotextural parameters, numerical pixel values (NPVs), and pixel heterogeneity (PSDs) of the lymph node and udder parenchyma, as well as the correlation between echotextural parameters and electrical conductivity (EC), in order to develop it as an alternative to laboratory testing for mastitic animal diagnosis and prognosis. The ultrasonographic images of each quarter were processed using digital image analyses to obtain mean NPVs and mean PSDs of mammary gland and lymph node parenchyma. The mean EC increased with the progression of infection. The mean NPVs of lymph node parenchyma decreased, whereas the mean NPVs of udder parenchyma increased from healthy to subclinical and clinical cows, respectively. The mean PSDs of lymph node and udder parenchyma increased with the progression and severity of infection from healthy to subclinical and clinical cows, respectively. It was concluded that, in dairy cattle, the variation in echotextural variables (NPVs and PSDs) of mammary gland and supramammary lymph node parenchyma and electrical conductivity appeared to be good indicators for the diagnostic and prognostic evaluation of sub-clinical and clinical mastitis in animals and simultaneously assists in the evaluation of udder health status.

ABSTRACT The production of chili is seriously threatened by bacterial wilt incited by Ralstonia solanacearum in Pakistan. As the information about the distribution and prevalence of the disease and the pathogen is scarce, the present study was performed to ascertain the prevalence, occurrence and distribution of R. solanacearum in different regions of chili cultivation. The results showed differences in the incidence and prevalence of bacterial wilt all over the country. Overall, an incidence of 10% and prevalence of 76% of the disease was observed in the country. The highest incidence of bacterial wilt was observed in the province of Sindh whereas it was the lowest in Baluchistan. The prevalence of the disease followed the similar trend in the provinces. As regards agroecological zones, the highest incidence was recorded in Indus delta followed by sandy deserts whereas it was the lowest in western dry mountains. Almost the same tendency was observed regarding prevalence in the eight agroecological zones. In case of districts, the disease incidence was the maximum in Badin while it was the minimum in the district of Loralai. On the other hand, the disease prevalence was the maximum (100%) in the districts of Badin, Mirpurkhas, and Thatta and the minimum (60%) in the districts of Barkhan, Karak, and Loralai. Of the total 114 isolates of R. solanacearum, 81% were confirmed as Biovar 3 whereas the rest 19% isolates were identified as Biovar 4. The findings will help the farmers to design disease management programs accordingly to avert yield losses.

ABSTRACT For proper disease management, accurate diagnosis of the pathogen is essential. Therefore, in the present study Ralstonia solanacearum causing bacterial wilt of chili was characterized to determine the distribution of biovars of the bacterium in the eight agroecological zones with varying climatic conditions and edaphic factors. Among all the 114 isolates of R. solanacearum, 77% showed mucoid growth while 23% isolates gave non-mucoid growth. Similarly, the isolates with mucoid growth were found positive for hypersensitivity response (HR), while those with non-mucoid growth showed negative HR. All the isolates grew well at 37 °C, while none of the isolates produced its colony at 41 °C. All the 114 isolates of R. solanacearum showed positive responses for all the biochemical tests used for confirmation of the bacterium. Out of 114 R. solanacearum isolates, 81% were identified as Biovar III while the remaining 19% were recognized as Biovar IV. Biovar III constituted 37 and 70% in the provinces of Punjab and Sindh, respectively, while Biovar IV formed 19 and 30%. On the other hand, in Khyber Pakhtunkhwa and Balochistan, only Biovar III was recorded. Similarly, Biovar III was observed from all the eight agroecological zones of the four provinces of the country and found to be predominant. On the other hand, Biovar IV was recorded from four agroecological zones located in the provinces of Punjab and Sindh. All the isolates yielded a 750-bp band that corresponded to R. solanacearum. It is concluded that Biovar III is widely prevalent in the country warranting stringent control measures.

Workplace spirituality is recognised as the inner state of individuals and an aspect of their working life. In the study reported on here we aimed to unearth the effects of teachers' demographic factors (gender, age, nature of job, academic qualification, marital status, designation, teaching experience, and district) towards workplace spirituality in secondary schools. This study was a descriptive research and a cross-sectional survey research design was applied. The participants were 3,050 secondary school teachers. The participants were selected using stratified proportionate random sampling. The Workplace Spirituality Scale (WPS) developed by Petchsawang and Duchon (2009) was used along with a list of demographic variables to meet the study objectives. Different statistical techniques (t-test, mean, SD, one-way ANOVA and post hoc analysis) were applied for data analysis. The results indicate that teachers were satisfied and agreed with the practices of workplace spirituality in secondary schools. Moreover, the results reveal that the teachers' demographic factors (gender, designation, nature of job, academic qualification, marital status, teaching experience, and district) had significant effects on their workplace spirituality. From the results we recommend that the management of educational institutions should treat teachers equally and fairly without any discrimination for equal nurturing of workplace spirituality among teachers.

Abstract This study was performed to screen out the various species of ‘Cucurbitaceae’ family, musk melon (Kalash and Durga), bottle gourd (Crystal Long and Nuefield) and squash (Green Round, and Squash Malika) against the salt stress. All genotypes were treated with five different levels of NaCl (T0 = control, T1 = 1.5 dS m-1, T2 = 3.0 dS m-1, T3 = 4.5 dS m-1 and T4 = 6.0 dS m-1) and half strength of Hoagland’s nutrients solution as the base nutrient solution. Results showed that the bottle gourd varieties “Nuefield” and “Crystal Long” performed best by maintaining the highest germination (93.2% and 85.6%), number of leaves per plant (4.5 and 5.7), shoot length (16.84 cm and 16.14 cm), root length (13.48 cm and 13.00 cm), plant fresh weight (942.2 g and 918.6 g), plant dry weight (118.4 g and 107.5 g), leaf area (171.2 cm2 and 169.1 cm2), chlorophyll content (3.5 μg/cm-2 and 3.4 μg/cm-2) with low chloride (1.57 ppm and 1.59 ppm) and sodium content (0.47 ppm and 0.51 ppm) under salt stress followed by varieties of Squash (Green Round, and Squash Malika) and musk melon (Kalash and Durga). It was also found that a higher level of salinity (4.5 dS m-1 and 6.0 dS m-1) has more adverse effects on the performance of all selected genotypes. Conclusively, it can be recommended that as compared to all tested species, bottle gourd varieties “Nuefield” and “Crystal Long” have the ability to withstand against salinity stress and should be planted under salt stress conditions.

Abstract Pesticides have emerged as a global threat to human health, especially in developing countries. In this study, we examined nine major types of pesticide residues in the food commodities (8 groups) and serum samples (n = 80) of people living in the region of Southern-Punjab, Pakistan. Pesticide residues quantification was performed using gas chromatograph coupled with electron capture detector (GC-ECD). The estimated daily intakes of the selected pesticide residues through consumption of food were also calculated. The concentration of analyzed pesticide residues in food was found below the EU maximum permissible limits except for α-HCH, and β-HCH. The highest risk of cancer was found to be associated with the consumption of HCHs contaminated cereals than other products. The incidence rate of pesticide residues in the serum samples of studied subjects was 96.25%. Pearson correlation test showed no significant correlation between thyroid hormone levels of the studied population group and the analyzed pesticide residues in the food commodities (p>0.05). Findings of this study indicate that people of Southern-Punjab, Pakistan are exposed to pesticides via food chain although non-significant health implications were recorded, the levels of a few pesticides need to be curtailed.

Abstract Fenugreek leaves contains bioactive compounds, which are helpful in maintaining human health. These compounds are adversely affected in conventional drying methods. Therefore, this study was carried out to retain maximum amount of these components through microwave-assisted drying and extraction technique. The technique was optimized for simultaneous drying and extraction process for fenugreek leaves. The drying rate and drying time were influenced by the level of microwave power; time was reduced from 21 to 5 minutes, when microwave power was changed from 30 to 100 W. Unlikely, the drying rate increased from 1.79 to 4.56 ± 0.05 g/(100 g.min) with the change in power (30 to 100 W). Moreover, moisture ratio analysis determined that two term model was the best to demonstrate the fitness with experimental values. The energy consumption was lowest for 100W compared to other powers. Furthermore, comparison between microwave-based extraction and methanol-extraction indicated that the antioxidant activity was better preserved compared to conventional ones even at higher microwave power levels.

Abstract Guava (Psidium guajava) is a highly perishable fruit with a short shelf life as physico-chemical changes occur continuously and rapidly after harvest leading to heavy postharvest losses. This experiment was laid down to unravel the effects of ascorbic acid (AA) on ripened guava fruits to improve its shelf life and quality. Four different concentrations of AA namely 0, 50, 100, 200 ppm were used to treat guava fruit for five minutes followed by storage at 25±2 °C and ≥80% relative humidity (RH) for 12 days. Physiological and biochemical changes were studied, together with the specific enzymatic activities for catalase (CAT), superoxide dismutase (SOD) and peroxidase (POD). The results revealed that ascorbic acid treatments significantly reduced PWL, fruit decay percentage and suppressed pH and sugar/acid ratio than control fruits. Furthermore, soluble solid content, total acidity, total sugar, vitamin C and total phenolic contents were recorded higher in 200 ppm AA-treated fruits and maintained higher eating quality than control fruits. In addition, SOD, POD and CAT activities were observed higher in 200 ppm AA-treated fruits than control. The results clearly demonstrate that 200 ppm AA application have a potential to improve quality attributes of guava fruit.

Abstract In traditional row and strip cowpea-sorghum intercropping systems, cowpea forage yield reduces significantly due to intense competition and dominance of sorghum in acquiring growth resources. This field study evaluated novel mixed strip intercropping systems of forage cowpea and sorghum having different number of crops rows arranged under different spatial arrangements. Cowpea was intercropped with sorghum in 8, 12 and 16 rows strips with row-row spacing of 30, 45 and 60 cm. In each strip, equal number of rows of cowpea and sorghum were maintained. Factorial arrangement of randomized complete block design with three replicates was used to execute the field trials during summer seasons of 2013 and 2014. Strips having 12 rows and 60 cm row-row spacing positively affected all agronomic variables of cowpea which led to maximum forage yield (22.2 and 23.7 t ha-1 during 2013 and 2014 respectively) and dry matter biomass (6.63 and 6.94 t ha-1 during 2013 and 2014 respectively). In contrast, 8-rows strips having line spacing of 30 cm outperformed other intercropping systems by yielding the maximum herbage yield and dry matter biomass of sorghum. The intercropping system comprising of 12-rows strips with 60 cm row-row spacing remained superior in recording the maximum crude protein, fats and total ash along with the minimum fiber content of cowpea. In addition, this intercropping system under rest of spatial arrangements also remained unmatched, while 16-rows strips under all planting geometries remained inferior to other intercropping systems. Thus, cowpea intercropping with sorghum in 12-rows strips having 60 cm spacing offers biologically viable solution to improve biomass and forage quality of cowpea in intercropping with sorghum.

Resumen En los sistemas tradicionales de cultivo intercalado de frijol caupí y sorgo en franjas y filas, el rendimiento de forraje del frijol caupí se reduce significativamente debido a la intensa competencia y al dominio del sorgo en la adquisición de recursos para el cultivo. Este estudio de campo evaluó novedosos sistemas de cultivo intercalado en franjas mixtas de frijol caupí forrajero y sorgo con diferente número de filas de cultivo en diferentes disposiciones espaciales. El frijol caupí se intercaló con el sorgo en franjas de 8, 12 y 16 filas con un espaciamiento de 30, 45 y 60 cm entre las filas. En cada franja se mantuvo igual número de filas de frijol caupí y sorgo. Para la ejecución de los ensayos de campo durante las temporadas de verano de 2013 y 2014 se utilizó un diseño factorial en bloques completos aleatorizados con tres repeticiones. Las franjas con 12 filas y un espaciamiento de 60 cm entre las filas afectaron positivamente a todas las variables agronómicas del frijol caupí que condujeron al máximo rendimiento forrajero (22.2 y 23.7 t/ha en 2013 y 2014, respectivamente) y de biomasa de materia seca (6.63 y 6.94 t/ha en 2013 y 2014, respectivamente). En cambio, las franjas de 8 filas con un espaciamiento de 30 cm superaron a otros sistemas de cultivo intercalado al obtener el rendimiento máximo de hierba y de biomasa de materia seca del sorgo. El sistema de cultivo intercalado compuesto por franjas de 12 filas con un espaciamiento de 60 cm entre las filas siguió siendo superior, al registrar el contenido máximo de proteína bruta, grasas y cenizas junto con el mínimo contenido de fibra de frijol caupí. Además, este sistema de cultivo intercalado bajo el resto de las disposiciones espaciales también permaneció incomparable, mientras que las franjas de 16 filas bajo todas las geometrías de siembra permanecieron inferiores a otros sistemas de cultivo intercalado. Por lo tanto, el cultivo intercalado de frijol caupí con sorgo en franjas de 12 filas con un espaciado de 60 cm ofrece una solución biológicamente viable para mejorar la biomasa y la calidad del forraje del caupí en cultivo intercalado con sorgo.

Abstract In this era, various traditional medicinal herbs were researched for their toxicological and safety aspects. The present study focused on using Nigella sativa fixed (NSFO) and essential oil (NSEO) to mitigate the adverse consequences of oxidative stress. For the purpose, oxidative stress was induced in Sprague Dawley rats using potassium bromate injection. The effect of NSFO and NSEO on indices of serum chemistry and hematology were studied. The results indicated that potassium bromate imparted drastic changes in serum chemistry and hematology e.g. it increased the blood cholesterol and blood glucose along with decreasing the insulin secretion. Oxidative stress also influenced the hematological attributes negatively but experimental diets were slightly successful in normalizing the values. The experimental diets especially essential oil that contains significant quantities of thymoquinone modulated the adverse consequences of oxidative stress with special reference to hematology and serological attributes. In the nutshell, NSFO and NSEO hold potential to mitigate the oxidative stress, and improved various serological and hematological attributes.

Abstract Milk is a nutritious perishable product having a short shelf-life owing to the occurrence of spoilage bacteria in it. This has led to an increasing demand for ensuring safety through milk processing. Conventional techniques (such as heat treatment) increase shelf-life but damage the nutritional and sensory qualities of milk. Hence, there is a need to develop innovative, nonthermal processing techniques that increase the shelf-life while preserving its nutritional quality. This review focuses on the recent advances in assuring microbial safety of milk by using nonthermal techniques such as high-pressure processing, pulsed electric fields, ultrasound, ultraviolet irradiation and membrane microfiltration.

This study determined the effect of protease supplementation in fishmeal-based diets on growth performance and meat quality of commercial broiler birds. Four hundred and thirty-two (432) day-old broiler chicks were divided into 36 experimental units of 12 chicks each. Nine experimental diets were formulated using three levels (0%, 25%, and 50%) of the fishmeal as protein source, on protein equivalent basis, with or without alkaline protease (CIBENZA® DP100), and with overestimation of nutritional value based on the enhanced digestibility coefficient (EDC) concept because of the use of alkaline protease. Feed consumption and bodyweight were measured weekly. On the last day of the trial, two birds from each pen were picked and processed for carcass and meat quality parameters. Data were analysed by analysis of variance in a 3 x 3 factorial arrangement of completely randomized design. Weight gain and feed conversion ratio (FCR) were improved in the birds with 25% fishmeal as a replacement for soybean meal (SBM) and enzyme supplementation. Similarly, higher protein digestibility, dressing percentage and thigh meat yield were observed in birds fed diets with 25% fishmeal with added enzyme. In the blood biochemical profile, uric acid levels were lower, and cholesterol and triglyceride were higher in the group fed diets with 25% fishmeal with EDC and enzyme. The addition of protease enzyme to the diet with 25% fishmeal improved growth performance, crude protein digestibility and carcass characteristics.

Abstract To explore the protective effect of sesame seed oil (SSO) extracted from Til-06 sesame variety in Sprague-Dawley male rats against the management of hypercholesterolemia, the current two-phase study was conducted. In the first phase, oil was taken out from the sesame seeds (Til-06) and physicochemical analysis was carried out to find out the content of oil following by the evaluation of the quality attributes of extracted oil. In the second phase, efficacy was carried out to check the hyperlipidemic activity of sesame seed oil. Randomly distributed twenty (20) rats into 5 different groups with four (04) rats in each group and for 60 days fed as follow; the first group was fed on a basal diet, while the other groups were fed with 2, 4, 6 and 8% SSO plus basal diet. Blood samples were collected at the beginning and end of the study for biochemical study i.e. lipid profile which includes high-density lipoprotein (HDL-C), triglycerides (TG), low-density lipoprotein (LDL-C), total cholesterol (TC), and very-low-density lipoprotein (VLDL) and antioxidant status. Findings of the present study indicate that the oral administration of SSO significantly (P < 0.05) improved the antioxidant status with lower TC, LDL-C, TG, VLDL-C, and higher HDL-C values of hypercholesterolemic rats.

Abstract Ricinus communis L. and Withania somnifera L. have traditionally been used as analgesic and anti-inflammatory remedies. The current study was aimed to evaluate the in vitro antioxidant potential and anti-inflammatory activity of hydroalcoholic extract of R. communis leaves (RCE) and W. somnifera roots (WSE) in Wistar rats. Total phenolic and flavonoid contents were quantified and in vitro antioxidant activity of extracts was determined through DPPH* scavenging, superoxide anion scavenging and reducing power activities, while anti-inflammatory activity was observed by xylene-induced ear edema and paw edema induced by egg albumin and carrageenan. RCE and WSE demonstrated considerable antioxidant activity in DPPH* scavenging (IC50: 250.10 and 309.42 µg/mL), superoxide anion scavenging (IC50: 193.42 and 206.81 ug/mL), and reducing power (maximum absorbance: 1.47±0.01 O.D and 1.28±0.01 O.D at 500 ug/mL) activities, respectively, with high phenolic and flavonoid contents. Both extracts showed dose-dependent edema inhibition in inflammation models. A maximum ear edema inhibitions by RCE (51.49±2.54%) and WSE (49.28±1.90%) at 500 mg/kg were observed when compared to indomethacin (56.42±13.17%) in xylene-induced ear edema. RCE and WSE showed a maximum percentage of paw edema inhibitions of 46.62±8.98% and 43.00±12.44%, respectively as compared to chlorpheniramine (62.02±12.21%) after 4 h in the egg albumin model. In carrageenan-induced paw edema, RCE (72.88±13.79%) significantly inhibited paw edema in comparison to WSE (57.81±17.43%) against diclofenac (89.93±18.53%). Conclusively, both plants have shown plausible antioxidant and anti-inflammatory activities that might be due to high phenolic and flavonoid contents. Moreover, RCE demonstrated more promising effects than WSE.
