Acute Toxicity and Cytotoxicity Effect of Ethanolic Extract of Spondias tuberosa Arruda Bark: Hematological, Biochemical and Histopathological Evaluation

RESUMO Spondias tuberosa Arruda, popularmente conhecida como umbu, é uma espécie nativa da Caatinga amplamente utilizada na medicina popular, cujo perfil toxicológico ainda não é conhecido. Deste modo, este estudo avaliou o perfil fitoquímico, a toxicidade aguda e a citotoxicidade do extrato etanólico da casca da Spondias tuberosa Arruda (EEStb) nos parâmetros hematológicos, bioquímicos e histopatológicos. Foram utilizados ratos fêmeas Wistar (±210g) e divididos em: grupo controle (C) e tratados com uma dose única de 300mg/Kg (EEStb300) ou 2.000mg/kg do peso corporal (ESStb2.000) do EEStb. Após 24 horas e 14 dias da gavagem, os parâmetros comportamentais, hematológicos, bioquímicos e histopatológicos foram determinados. O efeito citotóxico foi avaliado em células HEp-2. Não foi observada mortalidade ou alterações de massa corporal durante os 14 dias de experimento nos grupos EEStb300 e EEStb2.000, mas EEStb2.000 provocou uma redução discreta no consumo hídrico e alimentar, assim como na atividade locomotora no 1º dia de tratamento. Não houve alterações nos parâmetros histopatológicos, macroscópicos, bioquímicos e hematológicos. EEStb2000 nas concentrações de 6.25-50μg.ml-1) não produziu efeito citotóxico. na célula HEp-2. Estes resultados sugerem que EEStb não promoveu toxicidade aguda ou citotóxica, sugerindo boa margem de segurança para uso da Spondias tuberosa Arruda.
ABSTRACT Spondias tuberosa Arruda, popularly named as umbu, is native from savanna-like vegetation and widely used for medicinal purposes, however, the toxicological profile is not available yet. This study evaluated the phytochemical profile and acute toxicity and citoxicity of Ethanolic Extract of Spondias tuberosa Arruda Bark (EEStb) in hematological, biochemical and histopathological parameters. Female Wistar rats were divided into: control (C) and animal treated single doses of 300mg/Kg (EEStb300) or 2.000mg/kg body weight (ESStb2.000) of the EEStb. After 24 hours and 14 days from gavage, the behavior, hematological, biochemical and histopathological parameters were assayed. Cytotoxicity effect was evaluated on HEp-2 cell lines. Neither EEStb300 nor EEStb2.000 produced mortality nor changes in body weight during the 14-days of observation, but EEStb2.000 reduced quietly the food and water intake as well as locomotor activity at first day. There were no changes in macroscopic, histopathological, biochemical and hematological parameters. EEStb in concentrations of 6.25- 50μg ml−1 on HEp-2 cell did not produce cytotoxic effect. These results suggest that EEStb did not cause acute toxicity and cytotoxic, suggesting a good safety rate for Spondias tuberosa Arruda.

AbstractPhytopharmaceutical products are being used in the treatment and prevention of health problems. Nowadays, the development and evaluation of novel pharmaceutical products is expensive and time consuming. A statistical approach is a good tool for optimal development processes. Nectandra falcifolia (Nees) J.A. Castigl. ex Mart. Crov. & Piccinini, Lauraceae, a Brazilian species, is reported as anti-inflammatory, anti-leishmanial and anti-microbial. However, there is little known about its chemical composition. For other species of Nectandra genus, the presence of antioxidant compounds is reported. In order to optimize the process of obtaining extract with high antioxidant activity, different extraction conditions were tested following a statistical approach. Two sequential experimental designs were used – first, a factorial 23 design, followed by central composite 22. The extracts manufactured by these experimental statistical matrixes had their antioxidant activity and phenolic contents quantified and the response surface plots were fitted in quadratic models and they predicted the best extraction condition for the best antioxidant activity. This standardized extract and its antioxidant activity were better evaluated by two complementary tests (ABTS and Burst respiratory). A topical formulation containing 1% (w/w) of standardized extract was prepared and used for an in vivo skin permeation study using a two-dose application. The photoacoustic spectroscopy was used to analyze the samples from the permeation study and the composition profile of standardized extract. In rat skin samples, the data demonstrated that for the higher dose of topical formulation (5 g/cm2), the standardized extract could cross skin and be seen in epidermis and dermis. This was not the case for the lower dose (2 g/cm2) which was only present in the epidermis. This information suggests that this novel standardized extract of N. falcifoliacould be explored for skin damage prevention or treatment for diseases developed by oxidative damage.

NOVA LEI DOS PORTOS: DESAFIOS JURÍDICOS E PERSPECTIVAS DE INVESTIMENTOS

A situação dos portos brasileiros tem sido identificada como um dos principais entraves para o desenvolvimento econômico do Brasil. Atribui-se esse problema, preponderantemente, aos altos custos e à pouca eficiência nas operações portuárias. Recentemente, com a ediçã da nova Lei dos Portos, foi estabelecido um novo marco regulatório para o setor portuário brasileiro, buscando dar maior agilidade às transações e modernizar o setor. Os arrendamentos portuários passaram a ser regulados de forma semelhante a uma concessão de serviço público, enfatizando-se parâmetros de desempenho e a regulação tarifária. A nova legislação traz importantes inovações, como o critério de julgamento nas licitações de arrendamentos, a não obrigatoriedade de pagamento de valor de outorga para a assunção das áreas e a maior facilidade para o estabelecimento de portos privados. O objetivo deste artigo é investigar os principais desafios jurídicos trazidos por essas inovações e como elas se adequam à estrutura requerida para um melhor funcionamento dos portos e para as perspectivas de investimentos no setor. Inicialmente, será abordado o processo de conversão da MP n. 595/2012 na lei n. 12.815/2013, enfatizando os principais pontos de debate e interesses em jogo que motivaram as alterações ao projeto de lei na fase do veto presidencial. Em seguida, será investigada a natureza jurídica dos arrendamentos portuários, analisando em que medida eles se aproximam do modelo de concessões de serviços públicos. No item seguinte, serão apontadas as modificações na competência do poder concedente e dos demais entes envolvidos na regulação do setor portuário. Serão analisadas, ainda, as mudanças no critério de julgamento das licitações dos arrendamentos e seus principais impactos. Por fim, serão feitas algumas observações a respeito da situação dos terminais de uso privativo. Concluiu-se que a nova Lei dos Portos, além de conferir aos arrendamentos portuários o mesmo tipo de regulação típica da concessão de serviços públicos, também prioriza a agilidade e o baixo custo das operações, trazendo perspectivas de crescimento para o setor, embora seu impacto exato seja incerto. O campo de estudo é o direito regulatório e o método utilizado foi o indutivo.
The situation of the Brazilian ports has been identified as a major constraint to economic development in Brazil. This problem is attributed mainly to the high costs and low efficiency in port operations. Recently, with the enactment of the new Law of Ports, it was established a new regulatory framework for the Brazilian port sector, aiming to provide greater flexibility to the transactions and modernize the sector. Port leases will be regulated similarly to a public service concession, emphasizing performance parameters and tariff regulation. The new legislation brings important innovations, such as the criterion of judgment in bidding for leases, the not mandatory payment of grant value for the assumption of the areas and the facilitation for establishing private ports. The aim of this paper is to investigate the main legal challenges brought by these innovations and how they conform to the required structure for a better functioning of ports and the prospects of investment in the sector. Initially, we will address the process of conversion from MP no. 595/2012 to Law no. 12.815/2013, emphasizing the main points of debate and the interests at stake that motivated changes to the bill during the veto. Then we will investigate the legal status of port leases, analyzing the extent to which they approach the public service concessions model. In the next item, we will present the changes in the competence of the granting authority and other entities involved in the regulation of the port sector. Will be further analyze the changes in the criteria for judging the bids in port leases and its major impacts. Finally, we will make some observations regarding the status of private terminals. We conclude that the new law of ports attributes to port leases the same kind of regulation typical of public service concessions, and emphasizes agility and the low cost of operations, bringing growth prospects to the sector, although its exact impact is uncertain. The field of study of this article is regulatory law and its method was the inductive.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

OBJETIVOS: Relatar um caso de transmissão vertical de dengue ocorrido durante epidemia de 2008 pelo vírus tipo II no Rio de Janeiro e revisar a literatura sobre transmissão vertical de dengue. DESCRIÇÃO: Relatamos um caso de transmissão vertical de dengue. Recém-nascido a termo do sexo feminino, peso de nascimento de 3.940 g, foi admitida na unidade de terapia intensiva neonatal com rash cutâneo, hipoatividade e febre no quinto dia de vida. O hemograma evidenciava plaquetopenia importante (38.000 plaquetas). A mãe apresentou quadro clínico compatível com dengue 3 dias antes do parto. Foram colhidos então IgM para dengue da mãe e do recém-nascido, realizados pelo método de ELISA, sendo positivos em ambos. Dengue tipo 2 foi detectado no recém-nascido através de reação em cadeia da polimerase. COMENTÁRIOS: Este relato enfatiza a importância do pediatra estar alerta para a possibilidade de transmissão vertical de dengue iniciando precocemente o tratamento.
OBJECTIVES: To report a case of vertical dengue infection in a newborn from Rio de Janeiro, Brazil, and to review the literature concerning this problem. DESCRIPTION: We report a case of vertical dengue infection. Female neonate, birth weight 3,940 g, term, was admitted to a neonatal intensive care unit on the fifth day of life with fever and erythematous rash. Her mother had had dengue fever 3 days before delivery. Her platelet count was 38,000, dropping to 15,000. She did not have any hemorrhagic episodes, including cerebral hemorrhages. Anti-dengue antibodies (IgM) were positive in the mother and infant. Dengue type 2 was detected in the infant using polymerase chain reaction. COMMENTS: This report emphasizes that pediatricians should be aware of the possibility of vertical dengue infection so that early management can be instituted.
