Anti-Zika Virus Effects, Placenta Protection and Chemical Composition of Passiflora edulis Seeds Ethanolic Extract

Various beneficial biological activities have been reported for passion fruit (Passiflora edulis), grown in tropical and subtropical regions. However, no study has yet demonstrated its action against the Zika virus (ZIKV) infection. The present study aimed to investigate the anti-ZIKV of the ethanolic passion fruit seed extract (PFSE), from which, liquid chromatography tandem mass spectrometry (LC-MS/MS) analysis allowed to putatively annotate the occurrence of piceatannol, astringin, scirpusin A, scirpusin B, isookanin-7-O-glucoside and naringenin-7-O-glucoside, two quadranguloside derivatives, tyrosine, and phenylalanine. PFSE is well tolerated by cell lines (Vero E6 and HTR-8/SVneo) and tissue explants from the human placenta. With 1- and 24-h treatments, PFSE consistently reduced the viral load and NS1 protein expression of the two strains of ZIKV (MR766 and PE243) in placental explants and cell lines. Thus, PFSE has a promising potential for developing anti-ZIKV treatments, protecting the placenta against infection caused by ZIKV, along with relevant antioxidant potential, represented by ferric reducing antioxidant power (FRAP) of 390.5 Trolox eq per g dry extract and half maximal inhibitory concentration (IC50) for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH•) of 12.1 µg mL-1, and more than 90% protection from lipid peroxidation. Although further studies need to be conducted, PFSE treatment was safe and effective in trophoblast cells and placental explants, thus representing a promising class of all-accessible products based on the reuse of industrial by-products.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

In-vitro Antibacterial and Antibiofilm Activity of Cinnamomum verum Leaf Oil against Pseudomonas aeruginosa, Staphylococcus aureus and Klebsiella pneumoniae

Abstract Phytomedicines are becoming more popular in treatment of infectious diseases worldwide. Cinnamomum verum essential oil (EO) has been used as a therapeutic alternative for various diseases. This study aimed to evaluate the antibacterial and antibiofilm activity of the C. verum leaf EO against Pseudomonas aeruginosa, Staphylococcus aureus and Klebsiella pneumoniae. Effect of EO vapor on planktonic cells was determined using microatmosphere technique. CLSI M7-A10 method was employed in Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) experiments. Effect of EO on established biofilms was quantified and visualized using XTT and Scanning Electron Microscope (SEM). In-vitro toxicity was evaluated using Human Keratinocytes (HaCaT). Chemical analysis of EO was done using Gas Chromatography- Mass Spectrometry (GC-MS). All tested strains were sensitive to cinnamon oil vapor. EO exhibited 0.5 and 1.0 mg/mL MIC and MBC against all test strains. Minimum Biofilm Inhibitory and Biofilm Eradication Concentrations (MBIC50 and MBEC) were 1.0 and 4.0 mg/mL. SEM indicated cellular shrinkages, cell wall damages, and decreased biofilm densities. Cinnamon oil didn’t show any toxicity on HaCaT cell at any concentration tested. Eugenol was the most abundant compound in C. verum oil. C. verum EO shows an antibacterial and antibiofilm activity with minimal toxicity on host.

Elevated leptin levels in healthy climacteric women from Northeastern Brazil: an effect of age or adiposity?

ABSTRACT Objective Climacterium is associated with elevated leptin levels and increased risk of cardiovascular disorders. Conflicting data diverge on whether high leptin levels in climacterium reflect increasing adipose mass or, at least partially, age-related hormonal changes. This study addresses this issue in women from a Brazilian state with a low human development index. Subjects and methods A case-control study was conducted, enrolling 136 women from the state of Maranhão, 52 (38.2%) climacteric and 84 (61.8%) non-climacteric. Biometric, biochemical, hormonal and immunological parameters were analyzed. Results Climacteric women showed a moderately increased waist/hip ratio (0.894 versus 0.834, p < 0.05), sustained body mass index (27.46 versus 28.68, p > 0.05) increased leptin levels (9.59 versus 7.13, p < 0.05) and no evidence of metabolic syndrome. No other parameters were altered. The climacteric cohort didn’t show significant body fat gains but displayed a typical age-related redistribution of adipose tissue. Even so, leptin levels were significantly elevated compared with non-climacteric women. Conclusions Altogether, these data support the hypothesis that leptin is elevated, at least partially, as a function of age and climacterium and is not necessarily correlated with metabolic dysfunction and systemic inflammation. Further studies are needed to evaluate the impact of higher leptin levels on postmenopausal women. Arch Endocrinol Metab. 2020;64(3):276-81

RESUMO Avanços significativos na compreensão e no manejo da Distrofia Muscular de Duchenne (DMD) ocorreram desde a publicação de diretrizes internacionais para o cuidado destes pacientes em 2010. Nosso objetivo foi elaborar um consenso nacional baseado em evidências para o cuidado multidisciplinar dos pacientes com DMD no Brasil. Utilizamos uma combinação da técnica de Delphi com uma revisão sistemática da literatura de 2010 a 2016 para classificarmos os níveis de evidência e graus de recomendação para o consenso. Nossas recomendações foram divididas em duas partes. A metodologia utilizada na elaboração do consenso e conceitos gerais da doença encontram-se na parte 1. Neste artigo fornecemos os resultados e recomendações sobre reabilitação e cuidados sistêmicos para DMD.
ABSTRACT Significant advances in the understanding and management of Duchenne muscular dystrophy (DMD) have occurred since the publication of international guidelines for DMD care in 2010. Our objective was to provide an evidence-based national consensus statement for multidisciplinary care of DMD in Brazil. A combination of the Delphi technique with a systematic review of studies from 2010 to 2016 was employed to classify evidence levels and grade of recommendations for the guideline. Our recommendations were divided in two parts. Guideline methodology and overall disease concept descriptions are found in Part 1. Here we present Part 2, where we provide the results and recommendations on rehabilitation and systemic care for DMD.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Objetivou-se, neste trabalho, avaliar as alterações das características químicas do solo e a resposta produtiva da alface adubada com compostos orgânicos confeccionados artesanalmente a partir de resíduos e/ou matérias-primas. Os tratamentos foram distribuídos no delineamento de blocos ao acaso no esquema fatorial 5 x 4, sendo cinco tipos e quatro doses de compostos orgânicos, com quatro repetições, aplicados em plantas crescidas em vasos por 35 dias, sob condições de casa de vegetação. No solo, as variáveis avaliadas foram: teor de matéria orgânica, teores dos nutrientes P, K+, Na+, Ca2+ e Mg2+, acidez potencial (H+ + Al3+), soma de bases (valor S), capacidade de troca de cátions (CTC), saturação por bases (V%) e pH. Avaliou-se, nas plantas, a biomassa seca da parte aérea. A adição dos compostos orgânicos aumentou a produção de matéria seca da alface e o teor de matéria orgânica no solo. A elevação de doses dos compostos orgânicos por ocasião da adubação da alface favorece o aumento não apenas dos teores de fósforo e de sódio trocável no solo, mas também dos teores de cálcio e potássio, com o uso de alguns compostos. Os compostos 3 (Resíduos orgânicos doméstico, urbano e industrial) e 4 (Composto commercial - Greenworld) melhoram sobretudo as características químicas do solo e reduzem a acidez potencial.
The objective of the present study was to evaluate changes in chemical characteristics of soil and yield response of lettuce fertilized with organic compounds from waste and/or raw materials. Treatments were arranged in a randomized block design in a 5 x 4 factorial scheme, with five types and four doses of organic compounds, with four replications, applied to plants grown in pots for 35 days under greenhouse conditions. In the soil, the evaluated variables were: organic matter content, nutrient content - P, K+, Na+, Ca2+ and Mg+2, potential acidity (H+ + Al3+), sum of bases (S value), cation exchange capacity (CEC), base saturation (V%) and pH. In plants, dry matter yield of shoot was assessed. The addition of organic compounds increased the dry matter yield of the lettuce and organic matter content in soil, favoring the increase of phosphorus and exchangeable sodium in soil, also, increase in calcium and potassium were observed, with use of some compounds. Compounds 3 (domestic, urban and industrial residues) and 4 (Commercial Composto - Greenworld), mainly, improve the chemical characteristics of soil and reduce potential acidity.

A hidradenite supurativa (HS) é uma doença crônica, recorrente e debilitante, que afeta principalmente mulheres, sobretudo na segunda e terceira décadas de vida. Incide mais nas regiões axilar, inguinal, perianal e inframamária. Dentre suas complicações crônicas, são descritos processos fistulizantes com comprometimento de estruturas importantes, como o sacro e o cóccix, aparelho esfincteriano, uretra e vasos calibrosos, como os inguinais. As proporções de alguns casos de HS, exigindo intervenções alargadas, em vários tempos e com aplicação de retalhos e enxertos justificam relatos de casos que fogem da rotina, como o atual. Os autores apresentam um caso de comprometimento extenso da região glútea e perianal, que exigiu ressecção alargada com retalho e fistulectomia em um segundo tempo cirúrgico, com bom resultado para a paciente.
Hidradenitis suppurativa (HS) is a chronic, recurrent and debilitating disease, affecting mainly women, especially in their second and third decades of life. Its most common incidence is in the axillary, inguinal, perianal and inframammary regions. Its complications include chronic fistulizing processes, with involvement of important adjacent structures, such as the sacrum and coccyx, the anal sphincter, urethra and great-caliber vessels, such as the groin vessels. The proportions of some cases of HS requiring extensive surgical procedures at several moments and the application of flaps and grafts, justify unusual cases reports, like this one. The authors present a case of extensive involvement of the perianal and gluteal regions, which required extended resection with flap in the first approach and fistulectomy in a second surgical moment, with good result for the patient.

Insecticide effects of Ruta graveolens, Copaifera langsdorffii and Chenopodium ambrosioides against pests and natural enemies in commercial tomato plantation

O objetivo deste trabalho foi avaliar o efeito inseticida dos extratos aquosos de folhas de Ruta graveolens (Rutaceae) e alcoólicos de folhas de Copaifera langsdorffii (Caesalpinaceae) e de folhas de Chenopodium ambrosioides (Chenopodiaceae) a 5%. O experimento foi em blocos casualizados com seis repetições. As parcelas tratadas tiveram redução na população de pragas, quando comparadas às parcelas sem nenhum tratamento. O extrato elaborado com C. langsdorffii apresentou maior efeito inseticida em Bemisia tabaci (Hemiptera: Aleyrodidae) e soma das pragas. Verificou-se que após 24h de pulverização, as parcelas tratadas com o extrato de C. ambrosioides apresentaram menores números de adultos de Tuta absoluta (Lepidoptera: Gelechiidae) seguido pelas parcelas tratadas com extrato de R. graveolens. Foram observados menores números do parasitoide de ovos de lepidópteros Trichogramma sp. (Hymenoptera: Trichogrammatidae) e da soma dos inimigos naturais (predadores + parasitoides) nos tratamentos que receberam as pulverizações com os extratos das plantas de C. langsdorffii e C. ambrosioides seguido por R. graveolens do que na testemunha; do parasitoide da família Eulophidae (Hymenoptera), que atacam lagartas de T. Absoluta em plantas tratadas com R. graveolens, seguido por C. langsdorffii e C. ambrosioides do que na testemunha.
The aim of this study was to evaluate the insecticide effect of watery leaf extracts of Ruta graveolens (Rutaceae), alcoholic leaf extracts of Copaifera langsdorffii (Caesalpinaceae) and Chenopodium ambrosioides (Chenopodiaceae) in the concentration of 5% under field conditions. The experiment design was randomized blocks with six replications. The parcels treated with plant extracts showed reduction in the population of pests when compared with the control parcels. The extract elaborated with C. langsdorffii presented greater insecticidal effect under Bemisia tabaci (Hemiptera: Aleyrodidae) and sum of pests. It was verified that after 24 hours of spraying, the parcels treated with the extract of C. ambrosioides presented minor numbers of adults of Tuta absoluta (Lepidoptera: Gelechiidae), followed by the parcels treated with extract of R. graveolens. There were smaller numbers of parasitoid eggs of lepidopterans Trichogramma sp. (Hymenoptera: Trichogrammatidae) and sum of natural enemies (predators + parasitoids) in the parcels that had received spraying with extracts from the plants of C. langsdorffii and C. ambrosioides, followed by R. graveolens, compared to the control. There were a smaller number of parasitoids from the family Eulophidae (Hymenoptera) attacking caterpillars of T. absoluta in plants treated with R. graveolens, followed by C. langsdorffii and C. ambrosioides than in the control.

Siolmatra brasiliensis (Cogn.) Baill., Cucurbitaceae, popularmente conhecida como "pluméria" ou "taiuiá" é utilizada na medicina popular brasileira para diversos fins terapêuticos. O estudo de toxicidade aguda do extrato bruto etanólico (EBE) de Siolmatra brasiliensis foi investigado em camundongos. Nenhuma mortalidade ou sinais de toxicidade foram observados nas doses de 10 e 100 mg/kg, entretanto em doses administradas de 1000 e 2000 mg/kg levou as diversas alterações comportamentais e mortalidade. A DL50 para o EBE foi de 1000 mg/kg. Análise macroscópica dos órgãos demonstrou alterações morfológicas no coração dos animais tratados com 1000 e 2000 mg/kg. Por meios destes resultados conclui-se que o EBE de Siolmatra brasiliensis é seguro em doses de 10 and 100 mg/kg e apresentou toxicidade nas doses de 1000 e 2000 mg/kg.
Siolmatra brasiliensis (Cogn.) Baill., Cucurbitaceae, commonly known as "pluméria" or "taiuiá", is widely used in different ways in Brazilian popular medicine to treat several diseases. Acute toxicity of Siolmatra brasiliensis crude ethanolic extract (CEE) was investigated in mice. No mortality or signs of CEE toxicity were observed at the doses of 10 and 100 mg/kg bw, but the administration of 1000 and 2000 mg/kg bw caused several adverse behavioral effects and mortality. Macroscopic inspection of the organs showed morphologic alterations in the heart of animals treated with doses of 1000 and 2000 mg/kg bw. According to our results, S. brasiliensis CEE has an LD50 of 1000 mg/kg bw. We conclude that S. brasiliensis CEE was safe at the doses of 10 and 100 mg/kg bw and presented toxicity at the doses of 1000 and 2000 mg/kg bw.

Este estudo visou à caracterização das membranas fetais em búfalas (Bubalus bubalis, Linnaeus 1758) no terço inicial da gestação. As membranas fetais foram analisadas macroscópica e microscopicamente (luz e microscopia eletrônica de transmissão). O córion possui uma camada simples de células circulares, com núcleos de forma esférica, denominadas trofobláticas; há outro tipo celular, as células trofoblásticas gigantes, com dois ou mais núcleos. Ambas possuem uma grande quantidade de vesículas no citoplasma e retículo endoplasmático à microscopia de transmissão. O alantóide possui vasos preenchidos com eritrócitos, e contêm células alongadas, que formam um epitélio estratificado simples. O âmnion é uma membrana transparente, ou esbranquiçada; constituído por epitélio estratificado simples. A diferença principal entre o alantóide e o âmnion é que o último é avascular. O saco vitelínico é uma membrana opaca que desaparece durante a gestação; é a única membrana que não está em contato com as outras e apresenta três tipos diferentes de células que dão forma a três camadas distintas (endoderma, mesotélio, mesênquima).
The aim of the study was to characterize the fetal membranes in buffalo cows (Bubalus bubalis, Linnaeus 1758) in the first 3 months of the gestation. The fetal membranes were analyzed gross and microscopically (light and ultrastructural analysis). The corion is a simple layer of circle cells, with spherical nucleus, named trofoblastics; there is another cellular type, named trophoblastic giant cells, with two or more nucleus. Both of them have many cytoplasmic and reticule endoplasmic vesicles on transmission electron microscopy. The allantoic has many vessels filled with eritrocytes, elongated cells which form a stratified simple epithelium. The amnion is transparent or sometimes whitish, consisting of a stratified simple epithelium. The main difference between the allantoic and amnion is that the latter is avascular. The yolk sac is an opaque membrane that disappears during gestation, and is the only membrane that is not in contact with the others; it presents three different types of cells which form three distinct layers (endoderm, mesothelium and mesenquimal).

O agente etiológico do míldio do sorgo, Peronosclerospora sorghi, infecta as culturas do sorgo (Sorghum spp.) e do milho (Zea mays). Esse patógeno encontra-se disseminado em muitas regiões tropicais e subtropicais do mundo e pode ocasionar danos significativos na produção de sorgo quando as condições climáticas são favoráveis à sua ocorrência e em cultivares de alta susceptibilidade. No Brasil, antes restrito aos estados da região Sul, o míldio foi registrado também nos estados da região Sudeste e Centro-Oeste, causando prejuízos principalmente em áreas de produção de sementes. O cultivo de genótipos resistentes é o método mais eficiente para o controle da doença. Entretanto, essa estratégia é dificultada pela alta variabilidade genética apresentada pelo patógeno. Essa revisão aborda aspectos da taxonomia, biologia e distribuição geográfica do míldio do sorgo e discute questões relacionadas com a sua epidemiologia e controle, enfatizando estratégias que utilizam resistência genética.
The causal agent of sorghum downy mildew, Peronosclerospora sorghi, is a pathogen of sorghum species (Sorghum spp.) and corn (Zea mays). The pathogen is disseminated in many tropical and subtropical regions all around the world, causing considerable losses when conditions are favorable for its development or susceptible cultivars are used. Within Brazil it was initially restricted to the Southern region but has now also spread to the Southeast and Middle-West, causing significant losses specifically in areas of seed production. The use of resistant genotypes is the most efficient method to control the disease. Nevertheless, a high genetic variability of the pathogen makes it difficult to use resistant cultivars. This revision considers aspects of taxonomy and biology of sorghum downy mildew and discusses questions related to geographic distribution, epidemiology and control, focusing on strategies that use genetic resistance.

OBJETIVO: Avaliar a função autonômica na cardiomiopatia hipertrófica (CMH) através da variabilidade da freqüência cardíaca (VFC) e correlacioná-la com dados ecocardiográficos. MÉTODOS: Foram estudados 2 grupos pareados por sexo, idade e freqüência cardíaca: A) 10 pacientes com CMH septal (70% não obstrutiva); B) 10 voluntários saudáveis. A VFC foi analisada durante quatro estágios sucessivos: repouso, respiração controlada, teste de inclinação e respiração controlada associada ao teste de inclinação. Compararam-se as médias das variáveis entre os grupos, intragrupos durante os estágios, e no grupo A, correlacionando com as medidas ecocardiográficas (septo interventricular e diâmetro atrial esquerdo). RESULTADOS: Não observamos diferença na VFC entre os grupos nos 3 primeiros estágios. No 4º estágio, constatamos que medidas de atividade vagal apresentaram valores maiores no grupo A [logaritmo da raiz média quadrática das diferenças entre intervalos RR (LogRMSSD) - 1,35±0,14 vs 1,17±0,16; p=0,019; logaritmo do componente alta freqüência (LogAF)-4,89±0,22 vs 4,62±0,26; p=0,032]. Durante os estágios, também verificamos que medidas vagais [proporção de pares de intervalos RR consecutivos cuja diferença > 50ms (pNN50) e LogAF] apresentaram menor redução durante o 3º estágio no grupo A, e o LogAF, um aumento no último estágio (p=0,027), indicando marcante atividade parassimpática neste grupo. A análise da VFC do grupo A não revelou diferença entre pacientes com maior hipertrofia ou diâmetro atrial. CONCLUSÃO: 1) ocorreu predomínio parassimpático durante estimulação autonômica nos pacientes com CMH; 2) não encontramos correlação entre VFC e as medidas ecocardiográficas analisadas.
OBJECTIVE: Assess the autonomic function in hypertrophic cardiomyopathy (HCM) through heart rate variability (HRV) and to correlate it to echocardiographic data. METHODS: Two groups were studied, and compared for gender, age and HR:A) Ten (10) patients reporting septal HCM (70% non-obstructive); B) ten (10) healthy volunteers. HRV was analyzed along four successive stages: at rest, under controlled breathing, while bending, and controlled breathing associated to bending. Variables means were compared between groups and intra-groups in the different stages; in Group A, variables means were correlated to echocardiographic measurements (interventricular septum and left atrial diameter). RESULTS: No HRV difference was reported among groups in the first 3 stages. In the fourth stage vagal activity was shown to be higher in Group A [quadratic mean log between RR intervals (RMSSD) - 1.35±0.14 vs 1.17±0.16; p=0.019; high frequency component logarithm (LogHF)- 4.89±0.22 vs 4.62±0.26; p=0.032]. Along the stages, vagal measurements [rate of pairs of consecutive RR intervals whose difference is > 50ms (pNN50) and LogHF] also showed lower reduction in the third stage in Group A, while LogHF showed some increase in last stage (p=0.027), thus indicating marked parasympathetic activity in that group. Group A HRV analysis showed no difference among patients reporting larger hypertrophy or atrial diameter. CONCLUSION: 1) parasympathetic prevalence was shown during autonomic stimulation in HCM patients; 2) no correlation was found between HRV and echocardiographic measurements under analysis.
