Biodegradation of the Polycyclic Aromatic Hydrocarbon Fluoranthene by Fungi Strains from a Brazilian Tropical Peat

Polycyclic aromatic hydrocarbons (PAHs) are an important environmental issue and the identification of new biocatalysts for an efficient biodegradation of these compounds is essential for bioremediation. Therefore, fungi strains isolated for the first time from a tropical peat at Santo Amaro das Brotas (Brazil) were studied for fluoranthene biodegradation. Fusarium sp. AC-7, Penicillium sp. AC-1 and Penicillium sp. AC-6 were isolated using fluoranthene as sole carbon source. All strains were tested for biodegradation of 100 mg L−1 fluoranthene during 14 and 28 days. After 28 days of biodegradation, 64 ± 3, 60 ± 4 and 51 ± 2% biodegradation was observed for Penicillium sp. AC-1, Penicillium sp. AC-6 and Fusarium sp. AC-7, respectively. Analysis of the obtained compounds enabled the identification of four metabolites, which were common to the three employed strains: anthrone, anthraquinone, 9-methoxyanthracene and cyclopropa[1]phenanthrene. It is important to note that control experiments were performed. The obtained results clearly demonstrated the efficiency of tropical peat fungi in the transformation of fluoranthene. These findings showed the potential of tropical peats for isolation of fungi and indicated that these strains can be applied for bioremediation processes of areas contaminated with fluoranthene and other PAHs.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

The surface interactions between gold nanoparticles (AuNPs) and lipase from Candida antarctica fraction B (CALB) were investigated at macromolecular and colloidal length-scales. In order to elucidate the reciprocal effect of the interactions on the individual component’s properties, CALB was either added during the synthesis of AuNPs or added to pre-synthesized AuNPs. In both cases, it was observed that the AuNPs are spherical and stable and, by fluorescence and circular dichroism spectroscopies, changes were observed in the secondary and tertiary structure of CALB, that were dependent on the AuNPs concentration. Nevertheless, the catalytic activity of CALB was maintained, although at a lower percentage (≥ 80%), thus new bio-functionalities were inserted into AuNPs upon interaction with CALB. Using simple and straightforward approaches and state-of-the-art techniques important knowledge about CALB/AuNPs bioconjugates was gained, thus contributing to the development of new nano-biomaterials and for the safe use of AuNPs in biomedical applications.

Resistance training and hormone replacement increase MMP-2 activity, quality and quantity of bone in ovariectomized rats

Abstract AIMS The aim of the present study was to investigate the influence of resistance training (RT) and hormone replacement (HR) on MMP-2 activity, biomechanical and physical properties bone of ovariectomized (OVX) rats. METHODS Sprague-Dawley female rats were grouped into six experimental groups (n = 11 per group): sham-operated sedentary (SHAM Sed), ovariectomized sedentary (OVX Sed), sham-operated resistance training (SHAM RT), ovariectomized resistance training (OVX RT), ovariectomized sedentary hormone replacement (OVX Sed-HR), and ovariectomized resistance training hormone replacement (OVX RT-HR). HR groups received implanted silastic capsules with a 5% solution of 17β-estradiol (50 mg 17β-estradiol/ml of sunflower oil). In a 12-week RT period (27 sessions; 4-9 climbs) the animals climbed a 1.1 m vertical ladder with weights attached to their tails. Biomechanical and physical bone analyses were performed using a universal testing machine, and MMP-2 activity analysis was done by zymography. RESULTS Bone density and bone mineral content was higher in the RT and HR groups. The MMP-2 activity was higher in the RT and HR groups. The biomechanical analysis (stiffness, fracture load and maximum load) demonstrated better bone tissue quality in the RT associated with HR. CONCLUSION The RT alone as well as when it is associated with HR was efficient in increasing MMP-2 activity, biomechanical and biophysical properties bone of ovariectomized rats.

<p>Determination of urinary catecholamines and metanephrines is important for diagnosis of pheochromocytoma. Liquid chromatography tandem mass spectrometry (LC-MS/MS) method has been developed for the determination of catecholamines and metanephrines in human urine using deuterated internal standards (IS). In this method, 1000 µL of urine samples were treated by liquid-liquid extraction using ethyl acetate and subjected to LC-MS/MS analysis using positive electrospray ionization (ESI+). A BDS HYPERSILTM C18 column (125 mm × 3 mm, 3 µm) was used and mobile phase was water:methanol (98:2, v/v) with 0.25% of formic acid at 200 µ L min<sup>−1</sup>. The method has a chromatographic running time of approximately 10 min. The average of recovery was 92.9-106.1% for epinephrine, 94.6-107.7% for norepinephrine, 98.0-108.8% the dopamine, 94.2-105.1% the metanephrine, 97.0-106.0% for normetanephrine. The precision was lower than 7.0% for all analytes. Furthermore, this method has been implemented successfully in routine laboratory due to its easy execution and excellent precision.</p>

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Effect of urea on bovine serum albumin in aqueous and reverse micelle environments investigated by small angle X-ray scattering, fluorescence and circular dichroism

The influence that urea has on the conformation of water-soluble globular protein, bovine serum albumin (BSA), exposed directly to the aqueous solution as compared to the condition where the macromolecule is confined in the Aerosol-OT (AOT - sodium bis-2-ethylhexyl sulfosuccinate)/n-hexane/water reverse micelle (RM) is addressed. Small angle X-ray scattering (SAXS), tryptophan (Trp) fluorescence emission and circular dichroism (CD) spectra of aqueous BSA solution in the absence and in the presence of urea (3M and 5M) confirm the known denaturating effect of urea in proteins. The loss of the globular native structure is observed by the increase in the protein maximum dimension and gyration radius, through the Trp emission increase and maximum red-shift as well as the decrease in alpha-helix content. In RMs, the Trp fluorescence and CD spectra show that BSA is mainly located in its interfacial region independently of the micellar size. Addition of urea in this BSA/RM system also causes changes in the Trp fluorescence (emission decrease and maximum red-shift) and in the BSA CD spectra (decrease in alpha-helix content), which are compatible with the denaturation of the protein and Trp exposition to a more apolar environment in the RM. The fact that urea causes changes in the protein structure when it is located in the interfacial region (evidenced by CD) is interpreted as an indication that the direct interaction of urea with the protein is the major factor to explain its denaturating effect.
