Ploidy estimation and pre-selection of banana autotetraploids after in vitro polyploidy induction

ABSTRACT This study aimed to evaluate the efficiency of the parameters fresh leaf mass and stomatal density in the estimation of ploidy and pre-selection of putative banana autotetraploids. Young plants of the diploid cultivar Ouro (AA), previously subjected to in vitro polyploidization with the antimitotic amiprofos-methyl - APM (0, 10, 20, 30, 40, and 60 µM L-1) and colchicine (2.5 mM L-1) were evaluated for survival, height, number of leaves, pseudostem diameter, leaf disc fresh mass, and stomatal density. Ploidy was determined by flow cytometry in a random sample of 200 plants to find the relationship between the genomic content and the analyzed variables. Spearman’s correlation analysis showed a strong correlation (0.84**) between leaf disc fresh mass and tetraploid plants, as well as an absence of significant correlation between stomatal density and ploidy levels. Thus, the pre-selection of plants was carried out using a reference value of leaf disc fresh mass and resulted in 688 plants pre-selected as putative tetraploids. Among them, 318 were confirmed as tetraploids, 291 as diploids, and 79 as mixoploids after flow cytometry analysis. The pre-selection efficiency was 46.2% of tetraploid plants from the total pre-selected. The results indicate that the use of leaf disc fresh mass is a simple, practical, and promising method to estimate ploidy when a high number of plants is obtained in in vitro banana polyploidization studies, reducing the total number of plants to have confirmed ploidy.
RESUMO Este estudo objetivou avaliar a eficiência dos parâmetros massa fresca foliar e densidade estomática na estimativa da ploidia e pré-seleção de autotetraploides putativos de bananeira. Plantas jovens da cultivar diploide Ouro (AA), previamente submetidas à poliploidização in vitro com os antimitóticos amiprofos-metil - APM (0, 10, 20, 30, 40 e 60 µM L-1) e colchicina (2,5 mM L-1) foram avaliadas quanto à sobrevivência, altura, número de folhas, diâmetro do pseudocaule, massa fresca de disco foliar e densidade estomática. Determinou-se a ploidia, via citometria de fluxo, numa amostra aleatória de 200 plantas, a fim de encontrar relação entre o conteúdo genômico e as variáveis analisadas. Por meio da análise de correlação de Spearman, verificou-se forte correlação (0,84**) entre a massa fresca de disco foliar e as plantas tetraploides, bem como ausência de correlação significativa entre a densidade estomática e os níveis de ploidia. Assim, a pré-seleção das plantas foi feita mediante um valor de referência de massa fresca de disco foliar e resultou em 688 plantas pré-selecionadas como tetraploides putativos. Destas, 318 foram confirmadas como tetraploides, 291 como diploides e 79 mixoploides, após análise de citometria de fluxo. A eficiência de pré-seleção foi de 46,2% de plantas tetraploides do total pré-selecionado. Os resultados indicam que o uso da massa fresca de disco foliar é um método simples, prático e promissor para estimar a ploidia quando é obtido um elevado número de plantas em trabalhos de poliploidização in vitro da bananeira, reduzindo o total de plantas a ter a ploidia confirmada.

Resumo Objetivo: Este estudo com acompanhamento de longo prazo visou a avaliar o quadro clínico, os achados laboratoriais, o perfil histológico, os tratamentos e os resultados de crianças e adolescentes com hepatite autoimune. Métodos: Foram analisados os prontuários médicos de 828 crianças e adolescentes com HAI. Foi usado um questionário para coletar os dados anônimos sobre o quadro clínico, os achados bioquímicos e histológicos e os tratamentos. Resultados: De todos os pacientes, 89,6% tinham hepatite autoimune-1 e 10,4% hepatite autoimune-2. O sexo feminino foi predominante nos dois grupos. A idade média no início dos sintomas foi 111,5 (6; 210) e 53,5 (8; 165) meses nos pacientes com hepatite autoimune-1 e hepatite autoimune-2, respectivamente. Foi observado início clínico agudo em 56,1% e 58,8% e sintomas insidiosos em 43,9% e 41,2% dos pacientes com hepatite autoimune-1 e hepatite autoimune-2, respectivamente. A probabilidade de insuficiência hepática foi 1,6 vezes maior para hepatite autoimune-2; 3,6% e 10,6% dos pacientes com hepatite autoimune-1 e hepatite autoimune-2, respectivamente, apresentaram insuficiência hepática fulminante; o risco foi 3,1 vezes maior para hepatite autoimune-2. Os níveis de gamaglobulina e imunoglobulina G foram significativamente maiores nos pacientes com hepatite autoimune-1, ao passo que os níveis de imunoglobulina A e C3 foram menores em pacientes com hepatite autoimune-2; 22,4% dos pacientes apresentaram cirrose e a remissão bioquímica foi atingida em 76,2%. A taxa de sobrevida atuarial foi de 93,0%. Um total de 4,6% pacientes foram submetidos a transplante de fígado e 6,9% morreram (hepatite autoimune-1: 7,5%; hepatite autoimune-2: 2,4%). Conclusões: Nesta grande série clínica de crianças e adolescentes brasileiros, a hepatite autoimune-1 foi mais frequente e os pacientes com hepatite autoimune-2 mostraram maiores taxas de remissão da doença com respostas mais rápidas aos tratamentos. Os pacientes com hepatite autoimune-1 apresentaram maior risco de óbito.
Abstract Objective: This large study with a long-term follow-up aimed to evaluate the clinical presentation, laboratory findings, histological profile, treatments, and outcomes of children and adolescents with autoimmune hepatitis. Methods: The medical records of 828 children and adolescents with autoimmune hepatitis were reviewed. A questionnaire was used to collect anonymous data on clinical presentation, biochemical and histological findings, and treatments. Results: Of all patients, 89.6% had autoimmune hepatitis-1 and 10.4% had autoimmune hepatitis-2. The female sex was predominant in both groups. The median age at symptom onset was 111.5 (6; 210) and 53.5 (8; 165) months in the patients with autoimmune hepatitis 1 and autoimmune hepatitis-2, respectively. Acute clinical onset was observed in 56.1% and 58.8% and insidious symptoms in 43.9% and 41.2% of the patients with autoimmune hepatitis-1 and autoimmune hepatitis-2, respectively. The risk of hepatic failure was 1.6-fold higher for autoimmune hepatitis-2. Fulminant hepatic failure occurred in 3.6% and 10.6% of the patients with autoimmune hepatitis-1 and autoimmune hepatitis-2, respectively; the risk was 3.1-fold higher for autoimmune hepatitis-2. The gamma globulin and immunoglobulin G levels were significantly higher in autoimmune hepatitis-1, while the immunoglobulin A and C3 levels were lower in autoimmune hepatitis-2. Cirrhosis was observed in 22.4% of the patients; biochemical remission was achieved in 76.2%. The actuarial survival rate was 93.0%. A total of 4.6% underwent liver transplantation, and 6.9% died (autoimmune hepatitis-1: 7.5%; autoimmune hepatitis-2: 2.4%). Conclusions: In this large clinical series of Brazilian children and adolescents, autoimmune hepatitis-1 was more frequent, and patients with autoimmune hepatitis-2 exhibited higher disease remission rates with earlier response to treatment. Patients with autoimmune hepatitis-1 had a higher risk of death.

RESUMO A síndrome mielodisplásica (SMD) é uma desordem clonal das células-tronco hematopoiéticas caracterizada por citopenias periféricas devido à hematopoiese ineficaz e pelo aumento do risco de evolução para a leucemia mieloide aguda (LMA). As alterações cromossômicas representam o marcador mais importante da estratificação de risco para a transformação de LMA. Quebra das cromátides (chtb) é uma descontinuidade de uma única cromátide. Relatamos o caso de um paciente com SMD, cuja análise citogenética mostrou chtb espontâneo: 46,XY,add(13)(q34),chtb(15)(q24)[3]/47,XY,chtb(2)(q22),del(5) (q35),del(7)(q32),+8,del(11q)(q23),del(q22)[cp17]. O paciente foi considerado de alto risco devido ao cariótipo complexo e à presença de chtb. Sugerimos que essa anormalidade cromossômica possa ser considerada como marcador de instabilidade.
ABSTRACT Myelodysplastic syndrome (MDS) is a clonal hematopoietic stem cell disorder characterized by peripheral cytopenias due to ineffective erythropoiesis and an increased risk for evolving into acute myeloid leukemia (AML). Chromosomal abnormalities represent the most important marker of risk stratification for AML transformation. Chromatid break (chtb) is a discontinuity of a single chromatid. We report the case of a patient with MDS whose cytogenetic analysis showed spontaneous chromatid breakage (chrb): 46,XY,add(13)(q34),chtb(15)(q24) [3]/47,XY,chtb(2)(q22),del(5)(q35),del(7)(q32),+8,del(11q)(q23),del(q22)[cp17]. He was considered a high-risk patient due to the complex karyotype and the presence of chtb. We suggest that this chromosomal abnormality may be considered as a marker of genomic instability in MDS.

Elemental composition of vegetables cultivated over coal-mining waste

ABSTRACT We assessed elemental composition of the liver in mice subjected to one-time or chronic consumption of the juice of vegetables cultivated in a vegetable garden built over deposits of coal waste. Lactuca sativa L. (lettuce), Beta vulgaris L. (beet), Brassica oleracea L. var. italica (broccoli) and Brassica oleracea L. var. acephala (kale) were collected from the coal-mining area and from a certified organic farm (control). Elemental composition was analyzed by particle-induced X-ray emission (PIXE) method. Concentrations of Mg, S, and Ca of mice subjected to one-time consumption of broccoli and concentrations of these same elements plus Si of mice receiving kale were higher in the coal-mining area. Concentrations of P, K, and Cu were increase after chronic consumption of lettuce from the coal-mining area, whereas the levels of Si, P, K, Fe, and Zn were higher in the group consuming kale from the coal-mining area. Our data suggests that people consuming vegetables grown over coal wastes may ingest significant amounts of chemical elements that pose a risk to health, since these plants contain both essential and toxic metals in a wide range of concentrations, which can do more harm than good.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Nitric oxide production, inhibitory, antioxidant and antimycobacterial activities of the fruits extract and flavonoid content of Schinus terebinthifolius

The extract of the fruits from Schinus terebinthifolius Raddi, Anacardiaceae, was obtained by exhaustive extraction with methanol. Its fractions and isolated compounds were collected by fractionation with RP-2 column chromatography. The crude extract, the flavonoid fraction and the isolated compound identified as apigenin (1), were investigated regarding its inhibitory action of nitric oxide production by LPS-stimulated macrophages, antioxidant activity by DPPH and the antimycobacterial activity against Mycobacterium bovis BCG. The samples exhibited a significant inhibitory effect on the nitric oxide production (e.g.,1, IC5019.23 ± 1.64 µg/ml) and also showed antioxidant activity. In addition, S. terebinthifolius samples inhibited the mycobacterial growth ( e.g.,1, IC50 14.53 ± 1.25 µg/ml). The necessary concentration to produce 50% of the maximum response (IC50) of these activities did not elicit a significant cytotoxic effect when compared with the positive control (100% of lysis). The antioxidant and nitric oxide inhibition activity displayed by S. terebinthifolius corroborates its ethnopharmacological use of this specie as an anti-inflammatory. In addition, our results suggest that the flavonoids of S. terebinthifolius are responsible for the activities found. We, describe for the first time the activity against Mycobacterium bovis BCG and the inhibition of nitric oxide production for S. terebinthifolius.

A calcinose enzoótica é uma enfermidade caracterizada por mineralização de artérias e tecidos moles, osteopetrose, hipercalcemia e hiperfosfatemia. Neste trabalho descreve-se uma calcinose enzoótica em ruminantes no Mato Grosso, Goiás, Minas Gerais, Tocantins e no Distrito Federal, em campos onde não foi encontrada nenhuma das plantas calcinogênicas conhecidas. Os primeiros casos foram diagnosticados em 2004, e até 2010 foram necropsiados 86 casos da doença provenientes de 42 propriedades. Trinta e três propriedades foram visitadas, e em 32 os pastos eram caracterizados por moderada a acentuada degradação e invasão por plantas daninhas. A doença foi diagnosticada em ovinos em 19 fazendas, em bovinos em 17, em caprinos em 5 e em uma fazenda foram afetados tanto caprinos quanto ovinos. Animais adultos foram mais acometidos, mas a doença foi observada, também, em animais lactentes. A enfermidade foi observada durante todo o ano, mas a maioria dos surtos ocorreu nos meses de maior índice pluviométrico. Em duas fazendas foram determinados, mensalmente, os níveis séricos de Ca e P observando-se uma elevação significativa dos mesmos durante o período de chuvas. Os principais sinais clínicos observados foram de emagrecimento progressivo, caquexia e flexão de membros anteriores, com andar rígido. As principais lesões macroscópicas e histológicas foram de mineralização das artérias, valvas cardíacas, pulmões e rins. Sugere-se que a doença é provocada por uma planta calcinogênica ainda desconhecida. Mesmo sem conhecer a causa da calcinose recomenda-se evitar o pastejo de ruminantes em áreas degradadas e reformar pastagens degradadas.
Enzootic calcinosis is a disease characterized by mineralization of soft tissues, osteopetrosis, hypercalcemia and hyperphosphatemia. From 2004 to 2010, 86 cases of enzootic calcinosis were diagnosed in 42 farms in the states of Mato Grosso, Goias, Minas Gerais and Tocantins, and in the Federal District. The pastures were inspected in 33 farms, observing that in 32 outbreaks the disease occurred in degraded pastures invaded with different weeds. Previously known calcinogenic plants were not present. The disease was diagnosed in sheep in 19 farms, in cattle in 17 farms, in goats in five farms, and in sheep and goats in one farm. Adult animals were more affected; but the disease also was observed in lactating animals. The disease occurred all over the year, but most outbreaks were diagnosed during the rainy season. Serum Ca and P concentrations determined monthly in two sheep flocks were significantly higher during the rainy season. Main clinical signs were progressive emaciation followed by cachexia, and stiff gait with flexion of the fore limbs. Gross findings were mineralization of the arteries, cardiac valves, lungs, and kidneys. Histological findings were arterial mineralization, mainly in the medial layer with cartilaginous metaplasia and colagenous deposition between smooth muscle fibers. It is suggested that the disease is caused by an unknown calcinogenic weed present in degraded pastures.

O teste mais tradicionalmente utilizado para a avaliação da qualidade de lotes de sementes é o teste de germinação. A metodologia do teste de germinação ainda não está bem definida para sementes de Tabebuia impetiginosa (ipê-roxo) e T. serratifolia (ipê-amarelo), uma vez que são encontrados na literatura poucos trabalhos referentes especificamente à definição das condições ideais para a realização do teste nessas espécies. Objetivou-se com este trabalho verificar o efeito de temperatura e luz na germinação de sementes de diferentes lotes de ipê-roxo e ipê-amarelo. Para o ipê-roxo, a seleção das temperaturas foi realizada inicialmente em mesa de termogradiente, com temperaturas variando de 20,9ºC a 34,4ºC e, posteriormente, em BODs, com temperaturas de 25ºC, 30ºC e 35ºC. Para o estudo do efeito da luz na germinação das sementes de ambas as espécies, os tratamentos foram instalados em BODs sob três regimes de luz: luz branca contínua/temperatura de 30ºC, branca alternada com fotoperíodo de 8 horas/temperatura de 20/30ºC e escuro/temperatura de 30ºC. A temperatura de 30ºC foi adequada para a germinação de sementes de ipê-roxo, independente do ano de colheita ou da qualidade fisiológica dos lotes, enquanto a resposta da germinação em relação ao regime de luz variou conforme o ano de colheita. A condição ideal para realização do teste de germinação em sementes de ipê-amarelo, independente da procedência e do ano de colheita, foi sob luz constante a 30ºC.
The most traditionally used test in order to evaluation seed lots quality is the germination test. However, the germination test methodology is not well defined for Tabebuia impetiginosa (ipê-roxo) and T. serratifolia (ipê-amarelo) seeds. This is probably due to the little attention that the species received, specifically, about the definition of the ideals conditions for germination of these species. Thus, the objective of this research was to verify the effect of temperature and light on germination of T. impetiginosa and T. serratifolia seeds. The research was performed in a thermo-gradient table with temperature varying from 20,9ºC to 34,4ºC and in BODs with temperatures adjusted at 25ºC, 30ºC and 35ºC. To study the effect of light on germination for both species the seeds were incubated in BODs at white light/temperature 30ºC, alternate white with photoperiod 8 hours/temperature 20/30ºC and in the darkness/temperature 30ºC. The temperature of 30ºC was suitable for the germination of T. impetiginosa seeds, irrespective of the year of collection or physiological quality of the batches, while the effect of light on germination varied according to the year of collection. The ideal condition for accomplishment of the germination test in T. serratifolia seeds, irrespective of the provenance and year of collection, was under constant light at 30ºC.
