Abstract This work describes the circulation over the continental slope and the São Paulo Plateau in the Santos Basin during the SANSED winter 2019 survey. The cruise consisted of four legs in the period between June, 11 and August, 03 2019. The observed circulation is dominated by the Atlantic southwestern boundary current regime and remotely-generated anticyclones and cyclones. The former is composed by the Brazil Current, the Intermediate Western Boundary Current and their mesoscale meanders; the latter are 300km vortical rings with origin in the eastern side of the South Atlantic Basin. A Lagrangian scheme applied over satellite altimeter maps indicate that the origin of these rings is primarily the Cape Basin of South Africa. The interaction between the boundary currents, their cyclonic meanders, and the anticyclonic rings is complex, and varies widely. During the SANSED winter 2019 survey period, three anticyclones interacted with the Brazil Current, instabilizing it, forming dipoles with the current cyclonic meanders, leading to their downstream propagation. Ancienter cyclonic eddies within Santos Basin may interfere with the propagation of the large anticyclones further south. In addition, the continuous arrival of remotely-originated anticyclones, the larger portion over the São Paulo Plateau presented a tendency of counter-clockwise circulation during the whole cruise period.

Abstract This study aimed to develop a protocol for the cryopreservation of Pseudoplatystoma corruscans semen. For this, mature males were hormonally induced with a single dose of carp pituitary extract (5 mg/kg body weight). Semen was collected and evaluated. Two cryoprotectants were tested to compose the diluents: dimethyl acetamide (DMA) and dimethyl sulfoxide (Me2SO), in two concentrations (8% and 10%), + 5.0% glucose + 10% egg yolk. The semen was diluted in a 1: 4 ratio (semen: extender), packed in 0.5 mL straws and frozen in a dry shipper container in liquid nitrogen vapors. After thawing, sperm kinetics, sperm morphology and DNA integrity of cryopreserved sperm were evaluated. Pseudoplatystoma corruscans males produced semen with sperm motility > 80%. After thawing, all treatments provided semen with total sperm motility > 40%, with no significant difference (P < 0.05) between them, as well as between the other sperm kinetic parameters evaluated. The treatments with DMA provided a smaller fragmentation of the DNA of the gametes. Sperm malformations were identified in both fresh and cryopreserved semen, with a slight increase in these malformations being identified in sperm from thawed P. corruscans semen samples.

Resumo Este estudo teve como objetivo desenvolver um protocolo para a criopreservação do sêmen de Pseudoplatystoma corruscans. Para tal, machos maduros foram induzidos hormonalmente com uma dose única de extrato de hipófise de carpa (5 mg/kg de peso vivo). O sêmen foi coletado e avaliado. Sendo testados para compor os diluentes, dois crioprotetores: dimetil acetamida (DMA) e dimetil sulfóxido (Me2SO), em duas concentrações (8% e 10%), + 5,0% glicose + 10% gema de ovo. O sêmen foi diluído na proporção 1: 4 (sêmen: extensor), embalado em palhetas de 0,5 mL e congelado em container dryshipper em vapores de nitrogênio líquido. Após o descongelamento, foram avaliados os aspectos cinéticos espermáticos, a morfologia espermática e a integridade do DNA dos espermatozoides criopreservados. Os machos de P. corruscans produziram sêmen com motilidade espermática > 80%. Todos os tratamentos proporcionaram após o descongelamento sêmen com motilidade espermática total > 40%, sem diferença significativa (P < 0,05) entre eles, como também entre os demais parâmetros cinéticos espermáticos avaliados. Os tratamentos com DMA proporcionaram uma menor fragmentação do DNA dos gametas. Malformações espermáticas foram identificadas, tanto no sêmen fresco, como no criopreservado, sendo identificado um aumento discreto dessas malformações nos espermatozoides das amostras de sêmen descongeladas de P. corruscans.

ABSTRACT: São Paulo state is one of the country’s largest producers of beef and milk, and the midwestern region plays a key role in this production, as half of São Paulo’s cattle herd is found in this region. These numbers alone demonstrate the importance of livestock in this region. Therefore, this study aimed to describe the main epidemiological and clinical signs in cattle cases at the Large Animal Hospital at FMVZ-Unesp, located in the midwestern region of São Paulo state. The present retrospective study assessed 638 clinical cases of cattle treated from January 2010 to December 2019 (10 years). Digestive system diseases were the most prevalent, diagnosed in 30.3% of patients, followed by neurological diseases (19.1%) and respiratory diseases (10.5%). The other diseases were distributed in decreasing order as follows: musculoskeletal (7.8%), hematopoietic (6.1%), genitourinary (5.6%), metabolic and nutritional (5.5%), neonatal (4.7%), cutaneous (2.6%), poisoning (2.5%), lymphatic (2.2%), cardiovascular (1.6%) and other diseases (1.4%). Rabies, a fatal zoonotic disease, was the main cause of death in this study and the main disease associated with neurological signs (23.7%). Recognizing the main diseases of cattle in this region will promote the adoption of prophylactic measures to minimize their occurrence and manage treatment to avoid economic losses and decreased productivity of herds.

RESUMO: O estado de São Paulo é um dos maiores produtores de carne e leite bovino do país e a região centro-oeste do estado tem papel fundamental nessa produção, pois possui a metade do rebanho de bovinos do estado. Portanto, este estudo descreve os principais achados clínicos e epidemiológicos de bovinos atendidos no Hospital de Grandes Animais da FMVZ/Unesp, localizado na região centro-oeste paulista. Foi realizado um levantamento nos arquivos da Clínica de Grandes Animais do Hospital Veterinário da FMVZ-Unesp, Botucatu/SP, dos 638 casos clínicos de bovinos atendidos de janeiro de 2010 a dezembro de 2019. Enfermidades do sistema digestório foram as mais prevalentes (30,3%), seguidas das doenças neurológicas (19,1%) e respiratórias (10,5%). As demais enfermidades foram distribuídas, em ordem decrescente, em: musculoesquelético (7,8%), haematopoiético (6,1%), geniturinário (5,6%), cutânea (2,6%), linfático (2,2%) e cardiovascular (1,6%) ou foram classificadas em doenças metabólicas e nutricionais (5,5%), neonatais (4,7%), tóxicas (2,5%) ou outros distúrbios (1,4%). A raiva, uma zoonose fatal, foi a principal causa de óbito neste estudo e a principal doença neurológica (23,7%). O reconhecimento das principais doenças dos bovinos desta região permite a adoção de medidas profiláticas e de manejo para minimizar sua ocorrência e evitar perdas econômicas com o tratamento e menor produtividade do rebanho.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

Abstract Introduction: We investigated the clinical course and outcomes of patients submitted to cardiovascular surgery in Brazil and who had developed symptoms/signs of coronavirus disease 2019 (COVID-19) in the perioperative period. Methods: A retrospective multicenter study including 104 patients who were allocated in three groups according to time of positive real time reverse transcriptase-polymerase chain reaction (RT-PCR) for the SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2): group 1, patients who underwent cardiac surgery > 10 days after positive RT-PCR; group 2, patients with a positive RT-PCR within 10 days before or after surgery; group 3, patients who presented positive RT-PCR > 10 days after surgery. The primary outcome was mortality and secondary outcomes were postoperative complications, intensive care unit (ICU) length of stay, and postoperative days of hospitalization. Results: The three groups were similar with respect to age, the European System of Cardiac Operative Risk Evaluation score, and comorbidities, except hypertension. Postoperative complications and death were significantly higher in groups 2 and 3 than in group 1, and no significant difference between groups 2 and 3 was seen. Group 2 showed a high prevalence of surgery performed as an urgent procedure. Although no significant differences were observed in ICU length of stay, total postoperative hospitalization time was significantly higher in group 3 than in groups 1 and 2. Conclusion: COVID-19 affecting the postoperative period of patients who underwent cardiovascular surgery is associated with a higher rate of morbidity and mortality. Delaying procedures in RT-PCR-positive patients may help reduce risks of perioperative complications and death.

The Garças Reservoir history reveals a remarkable case of human impacts resulting from the urbanization pressure over the 20th century. It is a shallow tropical system located in the Parque Estadual das Fontes do Ipiranga, a protected area within the city of São Paulo, São Paulo State, Brazil. Based on a comprehensive dataset (1997-2017), we evaluated the water quality temporal change using a trophic state index (TSI); in addition, we also verified whether the management procedures improved the water quality. Mechanical removal of macrophytes (1999) triggered an abrupt change from eutrophic-supereutrophic to the hypereutrophic state with feedback mechanisms. After five years (2005), a slowdown in the internal P loading process occurred, followed by the installation of the secondary wastewater treatment in the city zoo two years later (2007). Both processes promoted the beginning of water quality improvement. In 2011, TSI first dropped to supereutrophic state with the temporary diversion of the sewage effluent from the São Paulo State Department of Agriculture. This effluent was finally inactivated (2014) and the system sustained the supereutrophic state. TSI responded well to the historical key events related to the control of sewage effluents. After 11 years from the permanent impact of macrophytes removal, current results have demonstrated improved health of the system. It is absolutely necessary to improve the sewage treatment from the city zoo to continue the reservoir recovery considering that since 2014 it has been the only external phosphorous loading to Garças Reservoir. The history of Garças Reservoir can offer a better understanding of wrong management strategies and the vulnerability of tropical shallow systems to eutrophication.

A história do Lago das Garças é um caso notável dos impactos humanos devidos à pressão da urbanização do século 20. Trata-se de uma represa tropical rasa localizada no Parque Estadual das Fontes do Ipiranga, área protegida situada na cidade de São Paulo, SP, Brasil. Com base em um banco de dados abrangente (1997-2017), nós avaliamos as alterações temporais da qualidade da água utilizando o índice de estado trófico (IET) e se as medidas adotadas de manejo melhoraram a qualidade da água. Em 1999, a remoção mecânica das macrófitas foi o gatilho para a mudança abrupta da represa do estado eutrófico-supereutrófico para hipertrófico, com mecanismos de retroalimentação. Após cinco anos (2005), ocorreu a desaceleração do aporte interno de P. Após dois anos (2007), houve a instalação da estação de tratamento secundário de esgoto na Fundação Parque Zoológico (2007). Ambos os eventos promoveram o início de uma melhora da qualidade da água. Em 2011, o IET passou para supereutrófico com a desativação temporária do efluente de esgotos provenientes da Secretaria de Agricultura e Abastecimento do Estado de São Paulo. Em 2014, este efluente foi totalmente desativado e o sistema manteve seu estado supereutrófico. Assim, o IET respondeu coerentemente aos eventos-chave relacionados com o controle dos efluentes de esgoto. Após 11 anos do impacto permanente da remoção das macrófitas, os resultados demonstraram a melhoria da qualidade da água da represa. Para prosseguir com a recuperação do Lago das Garças há necessidade absoluta de aperfeiçoar o sistema de tratamento secundário de esgoto proveniente do Zoológico, considerando que desde 2014 é o único efluente com aporte externo de fósforo para a represa. A história do Lago das Garças contribui para um melhor entendimento de estratégias errôneas de manejo e da vulnerabilidade de sistemas tropicais rasos à eutrofização.

A combined phytochemical, crystallographic and biological study of Worsleya procera roots was performed. Fifteen alkaloids were identified by gas chromatography mass spectrometry (GC-MS) and seven of them were isolated. The structures of the alkaloids were elucidated by spectroscopic methods, and a detailed crystallographic study of tazettine was carried out. The isolated alkaloids and the obtained extracts were tested in vitro against Plasmodium falciparum (3D7 and K1 strains) and human hepatocarcinoma cells (HepG2) to assess their antiplasmodial and cytotoxic effects, respectively. One of the isolated alkaloid derivatives, lycorine, exhibited antiplasmodial activity against both sensitive (3D7) and resistant (K1) parasite strains in the low micromolar range (half-maximal sample inhibitory concentration (IC50) values of 2.5 and 3.1 µM, respectively) and displayed a low cytotoxicity profile, with a selectivity index greater than 100. Our findings indicate that lycorine is a hit for antimalarial drug discovery.

RESUMO: O Albinismo é uma doença genética caracterizada pela deficiência na produção de melanina, o que torna os animais afetados mais susceptíveis a problemas cutâneos e influencia negativamente a criação destes animais. A mutação nonsense (c.1431G>A) no gene da tyrosinase já foi descrita como responsável pelo albinismo oculocutâneo em búfalos, entretanto estudos prévios sobre a prevalência dessa mutação ainda não foram realizados no Brasil. Portanto, o objetivo deste estudo foi avaliar a presença desta mutação em uma população de búfalos brasileiros. Foram genotipados 315 búfalos clinicamente normais, ou seja, sem o fenótipo albino evidente. Desses, 11 (3,5%) eram heterozigotos para a mutação (N/TYR) e os demais eram homozigotos selvagens (N/N). Este resultado demonstra que o alelo mutado para o albinismo em búfalo está presente no rebanho brasileiro e aponta a importância de estudos de prevalência de enfermidades hereditárias com o objetivo de prevenir a disseminação desses alelos mutados, minimizando os prejuízos.

ABSTRACT: Albinism is a genetic disease characterized by deficient melanin production making affected animals more susceptible to skin problems, negatively influencing production systems of the same. In buffalo, a nonsense mutation (c.1431G>A) in the tyrosinase gene was already described, which is responsible for the oculocutaneous albinism buffalo phenotype. However, prevalence studies have never been performed for this anomaly in Brazil. Therefore, the objective of this study was to investigate this mutation in buffalo herd in Brazil. Of the 315 buffalo tested with no albinism phenotype evident, 11 (3.5%) were heterozygous for the mutation and none were mutated homozygous, showing the existence of the albinism gene in buffalo production herds and proving the importance of prevalence studies for hereditary diseases in order to prevent the dissemination of these same genes and their negative productivity consequences.

This work reports the development of a certified reference material (CRM) of holmium(III) oxide Ho2O3 in acidic solution for the wavelength scale calibration of UV-Vis spectrophotometers. A 4.0% Ho2O3 solution was gravimetrically prepared in 10% perchloric acid and bottled in 5.0 mL glass ampoules. Homogeneity, stability and characterization were assessed according to ISO 17034 and ISO Guide 35. The material certification was undertaken by two laboratories by using UV-Vis spectrophotometry for characterization of the 14 peaks of minimum transmittance of Ho2O3, using a spectral bandwidth of 1.00 nm and a scanning speed of 30.0 nm min‒1. The material property values in wavelength (nm) were obtained for these 14 peaks, together with the expanded uncertainty (U) of 0.12 nm, for a coverage factor (k = 2.00) corresponding to a probability of approximately 95%, which took into account the standard uncertainties obtained in each study performed for the certification of the produced reference material.

RESUMO: Este trabalho teve por objetivo avaliar o proteinograma e concentrações séricas de IgG (após a padronização de teste ELISA) em potros do nascimento aos trinta dias de idade, antes e depois de mamarem colostro e serem tratados com plasma por via intravenosa. Foram utilizados 20 potros e suas respectivas mães, além de quatro animais doadores de plasma. Foram colhidas amostras de sangue dos potros em cinco momentos, logo após o nascimento e antes de mamar colostro (M1), dez horas após nascimento (M2), 24 horas após nascimento e previamente administração do plasma sanguíneo (M3), 48 horas de vida e 24 horas após administração do plasma sanguíneo (M4), e 30 dias após nascimento (M5). Foram colhidos sangue e colostro das éguas progenitoras no momento do parto. A concentração de proteína total (PT) e albumina foram determinadas em analisador bioquímico, a concentração de PT também foi avaliada em refratômetro manual. O fracionamento proteico foi realizado utilizando eletroforese em gel de agarose. A densidade do colostro foi avaliada com colostrômetros de refração BRIX e de densidade específica. A concentração de IgG total de todas as amostras foi determinada por teste ELISA. Com o sistema de ELISA aqui proposto foi possível determinar concentrações de IgG em amostras de soro, plasma e colostro equino com adequada repetibilidade. A média ± desvio padrão da concentração sérica de IgG dos potros ao nascer, foi de 15±8mg/dL, com dez horas de vida foi de 2.408±608mg/dL, se manteve em níveis semelhantes até 48 horas (2.364±784mg/dL) e diminuíram significativamente aos 30 dias de vida (1.414±586mg/dL). A concentração sérica e colostral de IgG nas éguas foi de 1.746±505mg/dL e 7.714±2.619mg/dL, respectivamente. A concentração plasmática de IgG dos doadores de plasma foi de 2.026±148mg/dL. Houve correlação positiva entre as concentrações séricas de IgG e PT (r=0,69 para refratômetro e r=0,76 para bioquímico), GT (r=0,81) e gamaglobulina (r=0,85). Dez horas após o nascimento foi possível verificar a transferência de imunidade passiva, possibilitando adotar medidas profiláticas e/ou terapêuticas em haras de criação de cavalos. Considerando que a proteína total, globulinas totais e fração γ-globulina apresentam correlação com IgG, estas determinações são úteis para monitorar os potros após mamarem o colostro. Um litro de plasma administrado às 24 horas de vida não foi suficiente para aumentar as concentrações séricas de IgG, 24 horas após transfusão, em potros com adequada transferência de imunidade passiva.

ABSTRACT: The aim of this study was to evaluate serum protein and serum IgG concentrations (after a direct enzyme immunoassay test ELISA optimization) in newborns foals from birth to thirty days of life before and after colostrum consumption and intravenous treatment with plasma. Twenty foals and their respective progenitors as well as four plasma donor’s horses were used. Blood samples were obtained from newborn foals at five time points, immediately after birth and before colostrum intake (M1), ten hours after birth (M2), 24 hours after birth and prior administration of blood plasma (M3), 48 hours after birth and 24 hours after plasma administration (M4), and 30 days after birth (M5). Blood and colostrum samples were collected from the progenitor mares immediately postpartum. Concentration of total protein (TP) and albumin were determined using a biochemical analyzer. The TP concentration was also measured by refractometer. Fractions of total serum protein were separated using agarose gel electrophoresis. Colostrum density was evaluated using BRIX refractometer and specific density colostrometer. Total IgG concentration was determined by an enzyme-linked immunosorbent assay. With the ELISA system proposed here it was possible to determine IgG concentrations in serum, plasma, and equine colostrum samples with adequate repeatability. Serum IgG concentration in foals at birth was 15±8mg/dL (mean ± standard deviation) raising at ten hours (2,408±608mg/dL) and remaining at similar levels up to 48 hours of life (2,364±784mg/dL), and decreasing significantly at 30 days of age (1,414±586mg/dL). Serum and colostrum IgG concentrations of mares were 1,746±505mg/dL and 7,714±2,619mg/dL, respectively. The plasma IgG concentrations from donor mares were 2,026±148mg/dL. Total protein, total globulins, and γ-globulin fraction showed correlation with IgG. Ten hours post birth was an adequate time to verify the transfer of passive immunity, allowing to adoption prophylactic and/or therapeutic measures in a horse farms. One liter of plasma administered at 24 hours of life was not sufficient to raise serum IgG concentrations in foals without passive immunity transfer failure.

Abstract INTRODUCTION: Pseudomonas aeruginosa, an important pathogen globally, presents several resistance mechanisms. This study aimed to investigate the presence of bla GES in clinical isolates of Pseudomonas aeruginosa obtained from various clinical specimens from patients admitted to three different hospitals in Recife, Brazil. The Guiana extended spectrum beta-lactamase (GES) enzymes are responsible for conferring broad spectrum resistance to beta-lactam drugs, including the carbapenems. METHODS: A total of 100 carbapenem-resistant P. aeruginosa isolates underwent polymerase chain reaction (PCR) testing to identify bla GES, bla KPC, bla SPM-1, bla IMP, and bla VIM. Additionally, PCR products positive for bla GES were sequenced. The clonal profiles of these same isolates were then determined by means of enterobacterial repetitive intergenic consensus (ERIC)-PCR analysis. RESULTS: PCR analysis revealed that four isolates harbored bla GES; DNA sequencing showed that two harbored bla GES-1 and two bla GES-11. Beta-lactamase genes bla SPM-1, bla IMP, bla VIM, and bla KPC were investigated; none of these genes was detected. Automated susceptibility testing methods (Vitek®2, bioMérieux) showed that the bla GES-1-positive isolates were only susceptible to polymyxin B. The patterns obtained with ERIC-PCR methods showed clonal relationship between the two isolates that harbored bla GES-11, whereas different clonal profiles were found in the isolates harboring bla GES-1. CONCLUSIONS: We detected the presence of bacterial isolates positive for two different variants of the enzyme GES in three different hospitals from Recife, Brazil. These enzymes have a great capacity for dissemination among Gram-negative bacteria and confer broad-spectrum resistance to beta-lactam antibiotics and to the carbapenems.

Abstract The molecular basis of anhydrobiosis, the state of suspended animation entered by some species during extreme desiccation, is still poorly understood despite a number of transcriptome and proteome studies. We therefore conducted functional screening by RNA interference (RNAi) for genes involved in anhydrobiosis in the holo-anhydrobiotic nematode Panagrolaimus superbus. A new method of survival analysis, based on staining, and proof-of-principle RNAi experiments confirmed a role for genes involved in oxidative stress tolerance, while a novel medium-scale RNAi workflow identified a further 40 anhydrobiosis-associated genes, including several involved in proteostasis, DNA repair and signal transduction pathways. This suggests that multiple genes contribute to anhydrobiosis in P. superbus.

ABSTRACT In this work, four secondary metabolites were isolated from leaves of Pachystroma longifolium (Euphorbiaceae): loliolide, gallic acid, methyl protocatechuate and ethyl gallate. Their structures were established by physical and spectroscopic methods. The extracts of P. longifolium and the purified compounds were tested as cathepsins inhibitors and the extracts were active.

RESUMO Neste artigo um grupo de especialistas no tratamento de cefaleia da Sociedade Brasileira de Cefaleia através de um consenso elaborou medidas estratérgicas para tratar uma crise de migrânea tanto na criança como no adulto. Uma enfase particular foi dada no tratamento da migranea na mulher, incluindo gravidez, lactação e período perimenstrual.

ABSTRACT In this article, a group of experts in headache management of the Brazilian Headache Society developed through a consensus strategic measurements to treat a migraine attack in both the child and the adult. Particular emphasis was laid on the treatment of migraine in women, including at pregnancy, lactation and perimenstrual period.

Resumo: A dermatosparaxia é uma doença autossômica recessiva do tecido conjuntivo, clinicamente caracaterizada pela fragilidade e hiperextensibilidade da pele. A dermatosparaxia em ovinos White Dorper é causada pelo polimorfismo de base única (SNP) c.421G>T no gene ADAM metalopeptidase com trombospondina tipo 1 motif, 2 (ADAMTS2). O objetivo deste estudo foi investigar a prevalência deste SNP em ovinos White Dorper no estado de São Paulo, Brasil. Foram coletadas amostras de sangue de 303 ovinos White Dorper. O DNA foi purificado destas amostras sanguíneas e utilizado em uma reação em cadeia da polimerase (PCR) para amplificação da região do gene contendo SNP c.421G>T. Os produtos das PCR foram sequenciados para determinar o genótipo dos animais. A prevalência do SNP na população estudada foi de 15,5%, estes achados indicam que medidas de controle efetivas devem ser utilizadas para prevenir a disseminação deste SNP no rebanho brasileiro de White Dorper.

Abstract: Dermatosparaxis is an autosomal recessive disorder of connective tissue; the disorder is clinically characterized by skin fragility and hyperextensibility. Dermatosparaxis in White Dorper sheep is caused by a single nucleotide polymorphism (SNP) (c.421G>T) in the ADAM metalloproteinase with thrombospondin type 1 motif, 2 (ADAMTS2) gene. The aim of this study was to investigate the prevalence of this SNP in a White Dorper herd in São Paulo state, Brazil. In this study, we collected blood DNA samples from 303 White Dorper sheep and performed polymerase chain reaction to amplify the SNP region. The samples were sequenced to determine the presence of the SNP in the ADAMTS2 gene. The SNP prevalence in the studied population was 15.5%; this finding indicates that more effective control measures should be used to prevent the inheritance of SNP c.421G>T in the ADAMTS2 gene in Brazilian White Dorper herds.
