ABSTRACT Eugenia uniflora is an American tree species with ecological, agronomical and medicinal importance. However, just few studies have focused on the in vitro propagation and production of secondary metabolites. This study investigated the explant sources and culture conditions for the in vitro callogenesis in E. uniflora towards induction of somatic embryogenesis and production of antioxidant compounds. Nodal segments, leaf sections and root segments from in vitro germinated seeds were used as explants and eight different combinations of auxins (2,4-D or NAA) and cytokinins (BAP or TDZ) were tested for the callus induction. The best callogenic response was observed in nodal segments, followed by leaf sections. Root segments presented comparatively poorer callogenic performance. Calli from nodal segments cultivated in MS medium with NAA (10 µM) + TDZ (5 µM) originated putative pro-embryogenic structures, while the culture in liquid medium using MS supplemented with NAA (10 µM) + TDZ (5 µM) revealed significantly higher content of phenols and flavonoids, as well as higher reducing capacity than the tested treatments and the control (fresh leaves). In summary, the calli obtained from nodal segments revealed competence for somatic embryogenesis induction and development as well as the production of secondary metabolites with pharmaceutical potential.

RESUMEN Eugenia uniflora es una especie arbórea americana de importancia ecológica, agronómica y medicinal. Sin embargo, pocos estudios se han centrado en la propagación y producción in vitro de metabolitos secundarios. Este estudio investigó las fuentes de explantes y las condiciones de cultivo para la callogénesis in vitro en E. uniflora para la inducción de embriogénesis somática y la producción de compuestos antioxidantes. Se utilizaron segmentos nodales, secciones de hojas y segmentos de raíz de semillas germinadas in vitro como explantes y se probaron ocho combinaciones diferentes de auxinas (2,4-D o ANA) y citoquininas (BAP o TDZ) para la formación de callos. La mejor respuesta callogénica se observó en segmentos nodales, seguidos de secciones foliares. Los segmentos de raíz presentaron un rendimiento callogénico comparativamente menor. Los callos de segmentos nodales cultivados en medio de cultivo MS con NAA (10 µM) + TDZ (5 µM) originaron estructuras proembriogénicas putativas, mientras que el cultivo en MS medio líquido con ANA (10 µM) + TDZ (5 µM) reveló un contenido significativamente mayor de fenoles y flavonoides, así como una mayor capacidad reductora que los tratamientos probados y el control (hojas frescas). En resumen, los callos obtenidos de segmentos nodales revelaron competencia para la inducción y el desarrollo de embriogénesis somática, así como para la producción de metabolitos secundarios con potencial farmacéutico.

RESUMO: No presente estudo objetivou-se avaliar a reação de genótipos de soja a Meloidogyne javanica (Est J3), Meloidogyne sp.0 (Est R0) e M. graminicola (Est VS1). No primeiro experimento, todos os genótipos (BMX Potência RR, BMX Valente RR, BMX Icone IPRO, PELBR10-6049 RR, and TECIRGA 6070 RR) testados mostraram-se suscetíveis a M. javanica, com fator de reprodução (FR) >1.0, e resistentes a Meloidogyne sp.0 (0.01 > FR < 0.15), enquanto no segundo experimento, todos os genótipos (BRS 246 RR, PELBR11-6038 RR, PELBR11-6001 RR, PELBR10-6005 RR, BMX Apolo RR, PELBR11-6028 RR, PF11651, PF103251, PELBR11-6035 RR, PELBR10-6050 RR, PELBR11-6042 RR, PELBR10-6017 RR, PELBR11-6007 RR, PELBR10-6016 RR, and PELBR10-6049 RR) testados comportaram-se como resistentes a M. graminicola (0.06 > FR < 0.43).

RESUMO Considerando a importância econômica da cultura do morangueiro no Brasil, com tendência clara de migração da produção no solo para sistemas inovadores de produção em substratos, é fundamental a avaliação das cultivares disponíveis no mercado quanto a tolerância a patógenos com potencial de infestação. Avaliou-se a resistência de oito cultivares comerciais (Festival, Camino Real, Camarosa, Oso Grande, Monterey, San Andreas, Aromas e Albion) a espécies de Meloidogyne e Pratylenchus. Cultivares de morango, mantidas em vasos individuais com solo esterilizado, foram inoculadas com 5.000 ovos + juvenis de segundo estágio de Meloidogyne arenaria, M. incognita, M. javanica ou M. hapla ou 1.000 espécimes de Pratylenchus zea ou P. brachyurus por planta. O experimento foi conduzido sob condições de casa de vegetação em delineamento inteiramente casualizado com seis repetições, utilizando-se plantas de sorgo ‘506’ e tomate ‘Rutgers’ como controles para o nematoides das galhas e das lesões radiculares, respectivamente. Noventa dias após a inoculação, foi avaliado o fator de reprodução dos nematoides (RF = população final / população inicial) para determinar a resistência dos genótipos de morango a cada espécie de fitonematoide. Todas as cultivares se comportaram como resistentes (FR<1,00) ou imunes (FR= 0,00) a M. javanica, M. incognita, P. zeae e P. brachyurus. Apenas ‘Camarosa’ foi suscetível (FR> 1,00) a M. arenaria e M. hapla e ‘Oso Grande’ comportou-se como suscetível a M. hapla; porém, as demais cultivares foram resistentes a estas duas espécies do nematoide das galhas. As cultivares de morango avaliadas neste trabalho comportaram-se como más hospedeiras dos nematoides estudados e, podem ser consideradas alternativa de cultivo em áreas infestadas com os patógenos.

ABSTRACT Considering the economic importance of strawberry in Brazil, with a clear migration trend from the soil to innovative systems on substrates, it is fundamental to evaluate the available cultivars in the market regarding tolerance to pathogens with potential for infestation. The reaction of eight commercial strawberry cultivars (Festival, Camino Real, Camarosa, Oso Grande, Monterey, San Andreas, Aromas, and Albion) to Meloidogyne and Pratylenchus species was evaluated. Strawberry cultivars were maintained in individual pots with sterilized soil and inoculated with 5,000 eggs + second stage juveniles of Meloidogyne arenaria, M. incognita, M. javanica or M. hapla or 1,000 specimens of Pratylenchus zea or P. brachyurus per plant. The experiment was carried out in a completely randomized design with six replications using tomato ‘Rutgers’ and sorghum ‘506’ plants as control to root-knot and lesion nematodes, respectively. Ninety days after inoculation, the nematode reproduction factor (RF= final population/initial population) was evaluated to determine the resistance of the strawberry genotypes to each nematode species. All cultivars behaved as resistant (FR<1.00) or immune (FR= 0.00) to M. javanica, M. incognita, P. zeae and P. brachyurus. The cultivar ‘Camarosa’ was susceptible (FR>1.00) to M. arenaria and M. hapla and ‘Oso Grande’ behaved as a good host to M. hapla; however, the other cultivars were resistant to these two Meloidogyne species. The assessed cultivars are poor hosts, being an alternative to be used in infested areas with these pests.

RESUMO: O objetivo do estudo foi caracterizar os focos de EIA identificados, entre 2009 e 2015, na região oeste do estado do Rio Grande do Sul, Brasil. Inicialmente foram identificados 26 equinos positivos em 24 propriedades, sendo que cada propriedade foi considerada um foco. Os diagnósticos foram realizados por imunodifusão em gel de ágar (AGID) na ocasião do transporte ou como medidas sanitárias, em casos de vínculo com animais infectados ou para fim de certificação do status sanitário. Um foco foi identificado em animais transportados ilegalmente da Argentina para o Brasil. Os estabelecimentos positivos eram fazendas ou estábulos e os animais infectados utilizados para trabalho, esporte ou reprodução. Quinze focos ocorreram em propriedades não cadastradas no SVO. Onze focos localizaram-se na zona urbana e 13 em propriedades rurais. Somente no ano de 2015 foram diagnosticados 12 dos 24 focos, sendo que no munícipio de São Borja ocorreram nove surtos neste período. Em duas propriedades o resultado inicial não foi confirmado no reteste, fazendo com que estes focos fossem encerrados imediatamente. Em três propriedades, durante o saneamento, identificou-se outros 12 animais positivos em três propriedades, de uma população de 1.108 susceptíveis. Assim sendo, pode-se concluir que a infecção que está presente na região, ocorre de maneira subclínica, associada a propriedades não cadastradas no SVO e animais transportados de forma ilegal, inclusive transporte internacional ilegal.

ABSTRACT: This study aimed to characterize the outbreaks of equine infectious anemia (EIA) identified, between the years 2009 and 2015, in the western region of the state of Rio Grande do Sul, Brazil. We identified 26 positive horses on 24 properties. Each positive property was considered an outbreak of the disease. The diagnoses were made using the agar gel immunodiffusion (AGID) test as a part of the sanitary checks conducted during animal transportation or certification of the horse´s sanitary status. The positive properties included farms or horse barns, and the infected animals were used for ranch work, sports, or reproduction. One outbreak was identified in animals that were being illegally transported from Argentina to Brazil. Fifteen outbreaks occurred on properties that were not registered with the Official Veterinary Service (OVS). Eleven outbreaks were identified in urban areas and 13 in rural areas. Twelve of the 24 outbreaks were diagnosed in 2015 alone, nine of which occurred in São Borja county. On two properties, a diagnosis could not be confirmed with a retest; therefore, these outbreaks were discharged. During sanitation checks on three properties, 12 additional positive animals were identified among a population of 1,108 susceptible animals. Based on these findings, we concluded that a subclinical form of the infection is present in that area, which is linked to properties that are not registered with the OVS, and that animals which are transported illegally across international borders represent a potential risk.

RESUMO A ocorrência de Fusarium spp. é um dos problemas mais limitantes em viveiros, esse patógeno pode ser transmitido via semente e causar danos em pré e pós-emergência. O presente trabalho objetivou identificar isolados de Fusarium spp. a nível de espécie e verificar a patogenicidade desses isolados em lotes de sementes de Pinus elliottii e P. taeda. Foram utilizados dois isolados do patógeno e cinco lotes de sementes de Pinus spp. Para caracterização molecular, sequenciaram-se as regiões genômicas: ITS e fator de elongação 1-α (tef1), para morfologia dos isolados utilizaram-se chaves de identificação específica para o gênero Fusarium. O teste de patogenicidade foi realizado através do contato das sementes com a cultura fúngica por 48 horas, seguida de semeadura em areia. As variáveis avaliadas foram: índice de velocidade de emergência, porcentagem de emergência, plântulas sintomáticas e tombamento de plântulas. O isolado F1UFSM foi identificado como Fusarium verticillioides e o F2UFSM como F. oxysporum. Ambos foram patogênicos a P. taeda e P. elliottii, causando redução no percentual de emergência e tombamento de plântulas.

ABSTRACT Occurrence of Fusarium spp. is one of the problems, most limiting to growth of seedlings, in nurseries. This pathogen can be transmitted via seeds and causes damages to the seedlings during pre- and post-emergence stages. The present study aimed to identify Fusarium spp. at the species level based on morphological and molecular characteristics and to verify the pathogenicity of these isolates in seeds lots of Pinus elliottii and P. taeda. For this, we used two Fusarium isolates and five lots of Pinus spp. seeds. Morphological characterization was performed based on a key, specific to Fusarium spp. identification, whereas, molecular identification was carried out by amplification and sequencing of the regions from internal transcribed spacer (ITS) and the elongation factor 1-α (tef1). The pathogenicity test was conducted through the contact of the seeds with fungal culture for 48 h, followed by sowing them in sand. The variables evaluated were emergency speed index, percentage of emergency, non-emergency seeds, symptomatic seedlings, and seedling damping-off. One isolate, F1UFSM, was identified as F. verticillioides and another isolate, F2UFSM, was identified as F. oxysporum. Both the isolates were pathogenic to the seeds of Pinus spp., causing a reduction in the percentage of emergence and seedling damping-off.

Abstract The use of tumor necrosis factor antagonists (anti-TNF) has become a usual practice to treat various inflammatory diseases. Although indicated for the treatment of psoriasis, anti-TNF may paradoxically trigger a psoriasiform condition. We present a case of a female patient who, during the use of infliximab for rheumatoid arthritis, developed psoriasis. In an attempt to switch anti-TNF class, we observed a cumulative worsening of the lesions requiring suspension of the immunobiological agent and the introduction of other drugs for clinical control. The therapeutic challenge of this paradoxical form of psoriasis is the focus of our discussion. The use of another anti-TNF in these patients is a matter of debate among experts.
