ABSTRACT The present study evaluated the effect of two thermal concentration systems on bioactive compounds, the sugar content of tomato (Solanum lycopersicum L.) pulp, and the carotenoid bioaccessibility of pulp concentrate. The closed processing system ensured a higher retention of phenolic and carotenoid compounds. The bioaccessibility of lycopene in tomato pulp concentrate was relatively low (0.54 %) but higher than in raw tomato pulp (0.15 %), corroborating other results that have reported the low availability of the compound in these matrices. Carotenoid extraction from tomato residue was also evaluated through both conventional (CE) and ultrasound (UAE) extractions together with the stability of extracts over 30 days. UAE promoted a superior release of lycopene and lutein than conventional extraction. Lycopene showed less stability with a reduction of 18 % in 30 days.

RESUMO: Foi realizado um estudo comparativo com a extração convencional e assistida por ultrassom em resíduos do processamento de tomate. A extração ultrassônica exibiu teor de fenólicos e flavonóides ligeiramente maiores, bem como maior capacidade antioxidante ABTS + (4,63 mg AG.g-1, 0,96 mg RUE.g-1 e 27,90 μmol TE.g -1, respectivamente). Em ambos os extratos, uma alta porcentagem de flavonóides foi perdida durante a digestão simulada, resultando em uma bioacessibilidade de aproximadamente 13%. Os extratos apresentaram boa estabilidade durante as condições de armazenamento, o que indica uma possível aplicação tecnológica.

ABSTRACT: A comparative study was perfomed with conventional and ultrasound assisted extraction on tomato processing waste. Ultrasound extraction exhibited slightly higher phenolic and flavonoids content, as well as higher ABTS + radical scavenging capacity (4.63 mg GAE.g-1, 0.96 mg RUE.g-1 and 27.90 μmol TE.g -1 respectively). On both extracts, a high percentage of flavonoids was lost during simulated digestion, resulting on a bioacessibility of approximately 13 %. Extracts presented good stability during storage conditions, which indicates a possible technological application.

Resumo Importantes compostos funcionais presentes nas frutas são frequentemente perdidos nos processos tecnológicos e durante o armazenamento. Com a técnica de microencapsulamento, é possível manter os compostos de interesse e agregar valor ao produto usando materiais de encapsulamento funcionais. Este trabalho tem como objetivo produzir polpa de goiaba microencapsulada através da técnica de secagem por pulverização, utilizando material encapsulante funcional, uma mistura de inulina e maltodextrina. A polpa de goiaba foi analisada quanto à composição centesimal, ao teor de carotenoides e à atividade antioxidante. As microcápsulas foram analisadas quanto a retenção de carotenoides, atividade antioxidante ao longo do tempo e sua morfologia por microscopia eletrônica de varredura e difração de raios-X. Ambas as proporções utilizadas no material de revestimento foram capazes de manter a atividade antioxidante da polpa de goiaba. O microencapsulamento com maior porcentagem de inulina mostrou-se como a melhor opção, devido aos bons resultados de retenção e estabilidade da atividade antioxidante ao longo do tempo, maior retenção do conteúdo de carotenoides e microestrutura mais estável, além de a inulina poder agregar valor aos pós devido às suas propriedades funcionais inerentes. O produto obtido no estudo representa um uso inovador, interessante e de possível utilização dos materiais como agentes encapsulantes, além de poder ser considerado como um potencial ingrediente funcional.

Abstract Important functional compounds present in fruits are often lost in technological processes and during storage. Microencapsulation technique allows maintaining the compounds of interest and adding value to the product using functional encapsulating materials. This work aimed to produce microencapsulated guava pulp using the spray-drying technique and a functional encapsulant material, i.e., a mix of inulin and maltodextrin. The guava pulp was analyzed for centesimal composition, carotenoid content, and antioxidant activity. The microspheres were analyzed for retention of carotenoids, antioxidant activity over time, and morphology by Scanning Electron Microscopy (SEM) and X-ray diffraction. Two proportions of coating material could maintain the antioxidant activity of guava pulp. The microencapsulation with a higher percentage of inulin is a preferred option due to the good results of retention and stability regarding antioxidant activity over time, relevant retention of the carotenoid content, and a more stable microstructure. In addition, inulin can add value to powders owing to its inherent functional properties. The product obtained in the study is innovative and interesting, as well as may provide a capable use of these materials as encapsulated agents. In fact, it can be considered a potential functional ingredient.

Resumo O objetivo do trabalho foi avaliar as atividades antioxidante e antimicrobiana de hidrolisados proteicos de soro de leite obtidos por diferentes proteases (Alcalase e Flavourzyme) e sua mistura (proporção 50:50). Previamente à hidrólise enzimática, o soro de leite foi concentrado por ultrafiltração com membranas cerâmicas tubulares. A reação enzimática foi realizada de acordo com o método pH-stat, sendo coletadas alíquotas com diferentes graus de hidrólise ( GH), ao longo de cada hidrólise proteica. Os hidrolisados foram analisados quanto a estimativa do teor de peptídeos, atividade antioxidante pelos métodos de captura do radical livre ABTS+ e do radical peroxila (Oxygen radical antioxidant capacity - ORAC), eletroforese e atividade antimicrobiana. Os máximos GH alcançados foram 15%, 52% e 63%, para Flavourzyme, mistura das enzimas e Alcalase, respectivamente. A atividade antioxidante aumentou com o GH. Os hidrolisados proteicos obtidos pela Alcalase apresentaram maiores atividades antioxidantes que os outros hidrolisados. Entretanto, nenhum hidrolisado proteico apresentou atividade antimicrobiana contra as cepas de Salmonella choleraesuis subsp. Enteritidis (ATCC 13076) e Listeria monocytogenes (ATCC 9117).

Abstract The objective of this work was to evaluate the antioxidant and antimicrobial activities of whey protein hydrolysates obtained using different commercial proteases (Alcalase and Flavourzyme) as well as their blend (50:50 ratio). Prior to the enzymatic hydrolysis, the whey was concentrated by ultrafiltration using tubular ceramic membranes. The enzymatic reaction was carried out according to the pH-stat method, in which aliquots were collected with different degrees of hydrolysis (DH) during the protein hydrolysis. The protein hydrolysates were evaluated with respect to the estimated peptides content, antioxidant activity by capture of free radical ABTS+ and the peroxyl radical (Oxygen radical antioxidant capacity - ORAC) methods, electrophoresis and antimicrobial activity. The maximum DH values reached were: 15%, 52% and 63% for Flavourzyme, the blend of enzymes and Alcalase, respectively. The antioxidant activity increased with the DH. The protein hydrolysates obtained using Alcalase presented higher antioxidant activities than the other samples. However, no samples presented antimicrobial activity against Salmonella choleraesuis subsp. Enteritidis (ATCC 13076) or Listeria monocytogenes (ATCC 9117).

RESUMO: O objetivo deste trabalho foi desenvolver uma bebida mista do tipo smoothie utilizando o albúmen sólido do coco verde na sua composição. As formulações do smoothie foram elaboradas seguindo um planejamento experimental, fixando-se a concentração do albúmen sólido em 20% e variando-se os teores das polpas de acerola, abacaxi e água de coco, que totalizaram 80% da massa total do produto. Os parâmetros de resposta avaliados foram o teor de vitamina C, a capacidade antioxidante e a aceitação sensorial global. Foi observado que das dez formulações avaliadas, as que continham maiores concentrações de polpa de acerola, apresentaram valores superiores de compostos bioativos, porém não foram as mais aceitas sensorialmente. Por meio da função desejabilidade chegou-se a uma formulação final que consistiu de 52,8% de abacaxi, 27,2% de acerola e 20,0% de albúmen sólido de coco verde. Esta nova formulação teve boa aceitação, com nota igual a 7 (“bom”) e representa um bom aporte nutricional, sendo fonte de vitamina C, podendo contribuir para agregar valor a um coproduto da indústria de bebidas.

ABSTRACT: The present study aimed to develop a mixed smoothie drink using the solid albumen of the green coconut in its composition. Smoothie formulations were prepared following an experimental design, setting the solid albumen concentration at 20% and varying the contents of the acerola (Barbados Cherry), pineapple, and coconut water pulps, which corresponded to 80% of the total mass of the product. Response parameters evaluated were vitamin C content, antioxidant capacity, and overall sensory acceptance. Ten formulations were evaluated. The ones that contained higher concentrations of acerola pulp had higher values of bioactive compounds but were not the most sensorially accepted. By means of the desirability function, a final formulation consisted of 52.8% of pineapple, 27.2% of acerola, and 20.0% of solid albumen of green coconut. This new formulation was well accepted, with a grade (score) 7 (“good”). It represents a good nutritional contribution and a source of vitamin C which can contribute to add value to a co-product of the beverage industry.

Abstract Whey is well-known for the functional and bioactive properties of its proteins and peptides, which are of great interest to food and nutraceutical industries. The aim of this study was to investigate the effect of four different commercial proteases (Novo ProD® (NPD), Alcalase® (ALC), Pancreas Trypsin® (TRY), and Flavourzyme® (FLA)) in the generation of hydrolysates with emulsifying and antioxidant activities. Hydrolysis processes were carried out for 5 h, reaching maximum degrees of 18.5, 15.5, 9.2, and 8.7% for NPD, ALC, TRY, and FLA, respectively. All tested enzymes generated very diverse, but conservative peptide profiles when comparing the treatments along the time, with the main enzymatic actions up to 120 min-reaction. An increase in the in vitro antioxidant activity was found for all treatments, achieving 46%, 40%, 40% and 22% for ALC, TRY, NDP and FLA, respectively. TRY hydrolysate maintained or slightly increased its emulsifying capacity along the time, however, a decrease in emulsifying capacity was found for ALC and NPD hydrolysates when compared to the non-hydrolyzed whey protein concentrate (WPC). All tested enzymes generated hydrolysates with enhanced antioxidant and/or emulsifying activities, which may be used as food ingredients and the choice of the enzyme will depend on the need.

Abstract In the present study a banana, strawberry and juçara smoothie was developed using an experimental mixture design. The juçara proportion was kept constant while the banana:strawberry ratio varied and the main responses were the sensory acceptability and the antioxidant capacity. From five formulations, only the one with the highest percentage of strawberry was rejected, most likely due to its lower sugar content and higher acidity. In general, panelists chose the sweeter formulations, which were those with a higher percentage of banana. Relating to the consistency, the best formulation was the one containing the same proportion of strawberry and banana pulp, which provided a better flow to the product. Additionally, this sample showed intermediate values of phenolic compounds and antioxidant capacity, as well as desirable physicochemical characteristics. In spite of being possible to obtain four formulations with good sensory acceptance, the one with equal amounts of banana and strawberry pulp is highlighted due to its antioxidant capacity and sensory characteristics, statistically corroborated by the desirability function.

Resumo O objetivo deste estudo foi avaliar o efeito da temperatura e do tempo de secagem sobre o conteúdo de compostos fenólicos totais e a capacidade antioxidante do bagaço de uva. Os dados experimentais das cinéticas de secagem foram ajustados a seis modelos matemáticos. O modelo de Page foi selecionado como o mais adequado para representar a secagem do bagaço de uva. A melhor preservação dos compostos fenólicos e da capacidade antioxidante foi observada a 60 °C, sugerindo que as temperaturas de 40 °C e 50 °C não foram suficientes para inativar a ação da enzima polifenoloxidase, sendo estas as responsáveis pela maior degradação desses compostos.

Abstract The objective of this study was to evaluate the effect of temperature and drying time on the total phenolic content and antioxidant capacity of grape pomace. Experimental data of the drying kinetics were fitted to six mathematical models and the Page model was selected as the most suitable to represent the drying of grape pomace. The best preservation of the phenolic compounds and antioxidant capacity was observed at 60 °C, suggesting that the temperatures of 40 °C and 50 °C were not sufficient to inactivate the enzyme polyphenol oxidase, being responsible for the greater degradation of these compounds.

A amora-preta é uma fruta que vem despertando a atenção de produtores e consumidores devido ao seu sabor agradável, cor atrativa e por apresentar em sua composição elevado teor de compostos bioativos. O grande entrave para consumo e a comercialização dos frutos da amoreira é sua elevada taxa respiratória, o que reduz sua vida útil. Uma alternativa viável para o aproveitamento econômico dessas frutas consiste em sua industrialização, podendo ser congeladas, enlatadas, processadas na forma de polpa, ou na forma de sucos e geleias. O objetivo deste trabalho foi avaliar os efeitos da radiação gama na conservação da polpa de amora. A irradiação foi realizada no laboratório de irradiação gama do Centro de Desenvolvimento da Tecnologia Nuclear (CDTN), em uma fonte de cobalto 60, com as seguintes doses de radiação: 0,75 kGy; 1,5 kGy e 3 kGy, sob uma taxa de dose de 3,24 kGy.h-1 . A polpa não irradiada foi utilizada como controle. As polpas de amora irradiadas foram armazenadas à temperatura de 4ºC, sendo avaliadas nos tempos de 0; 7; 15; 30 e 60 dias. Para verificar os efeitos da radiação gama no processamento na polpa, foi realizada a caracterização físico-química e química através das análises de acidez, pH, sólidos solúveis, sólidos totais, teor de antocianinas, atividade antioxidante e cor. Também foi feita análise microbiológica de acordo com a legislação brasileira vigente. O processo de irradiação aumentou a vida útil de prateleira da polpa em até 60 dias, sendo que o tratamento com uma dose de 1,5 kGy foi a que proporcionou a melhor qualidade microbiológica.

The blackberry is a fruit that has attracted the attention of producers and consumers due to its pleasant flavor, attractive color and composition present in high content of bioactive compounds. The major deterrent to consumption and marketing of the blackberry fruits is its high respiration rate, which reduces its lifetime. A viable alternative to the economic utilization of these fruits is its industrialization, can be frozen, canned, processed in pulp form, or in the form of juices and jellies. The objective of this study was to evaluate the effects of gamma radiation on the conservation of blackberry pulp. The irradiation was performed in the laboratory of gamma irradiation in the Development Center of Nuclear Technology (CDTN) in a cobalt 60 source, with the following radiation doses : 0.75 kGy ; 1.5 kGy and 3 kGy, under a dose rate of 3.24 kGy.h-1. The non-irradiated pulp was used as control. The irradiated blackberry pulps were stored at temperature of 4 ºC and evaluated at 0, 7, 15, 30 and 60 days. To check the effects of gamma radiation on the pulp processing, it was realized physicochemical and chemical characterization through the analysis of acidity, pH , soluble solids, total solids, anthocyanins, antioxidant activity and color. Microbiological analysis according to Brazilian legislation was also done. The irradiation process increase the shelf life of the pulp within 60 days, and the treatment with a dose of 1.5 kGy was the one that provided better microbiological quality.

Este trabalho teve por objetivo avaliar o efeito de diferentes agentes carreadores (Maltodextrina 5DE, Maltodextrina 10DE, Goma arábica e Capsul®) nas características físico-químicas (umidade, densidade, solubilidade, teor de antocianinas e atividade antioxidante) da polpa de morango desidratada por atomização e armazenada à temperatura ambiente por 90 dias. A utilização dos diferentes agentes carreadores resultou em pós com valores de densidade aparente entre 0,40 e 0,52 g/cm³ e umidade menor do que 2,5%. Todas as amostras foram altamente solúveis, mas os pós obtidos com a Maltodextrina 5DE e com a Goma arábica apresentaram menor higroscopicidade. As isotermas de sorção obtidas foram do tipo III e o modelo de GAB foi o que melhor se ajustou aos dados experimentais. A amostra produzida com goma arábica apresentou a maior concentração de antocianinas e, consequentemente, maior valor de atividade antioxidante, tanto imediatamente após a secagem como ao longo de 90 dias de armazenamento. Os resultados obtidos indicaram que a goma arábica foi o agente carreador mais adequado para a produção de polpa de morango em pó.

This study aimed to evaluate the effect of different carrier agents (5DE Maltodextrin, 10DE Maltodextrin, gum Arabic and Capsul®) on the physicochemical characteristics (moisture content, density, solubility, total anthocyanins and antioxidant activity) of strawberry pulp processed by spray drying and stored at room temperature for 90 days. The use of different carrier agents resulted in powders with bulk densities ranging from 0.40 to 0.52 g/cm³ and moisture contents below 2.5%. All the samples were highly soluble, but the powders obtained with 5DE Maltodextrin and gum Arabic were less hygroscopic. The sorption isotherms were of type III and the GAB model was the model that best fitted the experimental data. The powder produced with gum Arabic had the highest anthocyanin content and consequently the greatest antioxidant activity value, both immediately after the drying process and during the storage period. The results obtained showed that gum Arabic was the most suitable carrier agent for the production of strawberry pulp powder.

This study aimed to evaluate the freeze-drying process for obtaining grape juice powder by reverse osmosis using 50% grape juice pre-concentrated (28.5 °Brix) and 50% hydrocolloids (37.5% maltodextrin and 12.5% arabic gum). The morphology of the glassy food showed the absence of crystalline structure, which was the amorphous wall that protected the contents of the powder. The samples were stored in clear and dark containers at room temperature, evaluated for their physical (X-ray diffraction) for 65 days and chemical (polyphenol content) stability for 120 days. During the storage time in plastic vessels, samples remained physically stable (amorphous) and the phenolic concentration was constant, indicating the potentiality of this technique to obtain a stable product with a high concentration of phenolic compounds. Therefore, the freeze-drying process promoted the encapsulation of concentrated grape juice increasing its stability and shelf life, as well as proving to be an applicable process to food industry

As indústrias de frutas destacam-se pelo volume de resíduo gerado e, principalmente, pela composição dos mesmos. O Brasil vem-se consolidando como um importante produtor mundial de suco de uva, gerando grande quantidade de resíduos diversos, como o bagaço, que corresponde a aproximadamente 20% da fruta processada. O bagaço constitui-se de casca e semente, e apresenta uma composição rica e heterogênea em compostos fenólicos (flavonoides e não flavonoides), razão pela qual foi empregado como matéria-prima no presente trabalho para a obtenção de um extrato bioativo. As extrações enzimática e etanólica foram avaliadas para a recuperação destes compostos para fins alimentícios. Dentre as condições avaliadas nos dois planejamentos experimentais, a extração hidroetanólica foi mais eficiente na recuperação dos compostos bioativos (p< 0,01), cujo extrato apresentou uma capacidade antioxidante de 1.965 µmol Trolox por 100 g de bagaço, 25% superior à do extrato obtido com a melhor condição enzimática (1.580 µmol Trolox por 100 g de bagaço).

Fruit industries are highlighted by the residue amount and especially, for its composition. Brazil thereby is consolidating as an important grape juice producer, generating a large amount of diverse residues like grape pomace, which corresponds around 20% of total fruit processed. Grape pomace is constituted by seeds and peel and presents rich and heterogeneous composition in phenolic compounds (flavonoids and non flavonoids), reason why it was employed as raw material to obtain a bioactive extract in this study. Enzymatic and ethanolic extractions were evaluated to recover these compounds as they are able for application in food products. Between the evaluated conditions in both experimental design, ethanolic extraction was more efficient for the recovery of bioactive compounds (p<0,01), which extract presented an antioxidant activity of 1.965 µmol Trolox per 100 g of grape pomace, 25% higher than the extract obtained in the better enzymatic extraction (1.580 µmol Trolox per 100 g of grape pomace).

O camu-camu é um fruto da região amazônica que despertou interesse da comunidade científica devido à sua elevada concentração de vitamina C. O processamento do camu-camu, para obtenção de suco, é uma alternativa para aumentar a sua conservação e incentivar o seu consumo. Processos de separação por membranas são conduzidos em condições brandas de temperatura e pressão, preservando compostos termossensíveis e, consequentemente, a qualidade do produto final. Este trabalho teve como objetivo avaliar o desempenho do processo de microfiltração para obtenção de suco de camu-camu clarificado, avaliando-se o efeito da temperatura de processo sobre o teor de vitamina C e de compostos fenólicos totais, bem como sobre o valor da atividade antioxidante total do produto obtido. O processo conduzido a 45 °C apresentou a melhor retenção de vitamina C em relação às demais temperaturas estudadas. Neste caso, o fluxo de permeado médio foi de 75 Lh-1m-2. Foi verificado que a clarificação de polpa de camu-camu requer um pré-tratamento da polpa integral para assegurar um maior fluxo de permeado e, consequentemente, aumentar o rendimento de processo. Concluiu-se também que o aumento do fator de concentração, para um mesmo intervalo de tempo, não apresentou um efeito significativo no teor dos compostos avaliados no suco clarificado.

The Amazonian fruit camu-camu called the attention of the scientific community due to its high content of vitamin C. The fruit juice consists in an alternative to increase its conservation and consumption. Membrane separation technologies are conducted in mild temperature and pressure operational conditions, preserving the thermosensitive compounds and quality of the final product. This study aimed to investigate the performance of microfiltration process for obtention of clarified camu-camu juice by evaluating the effect of the temperature on vitamin C retention, total phenolics content and total antioxidant activity of the product. The process conducted at 45 °C showed best retention of vitamin C compared to the other evaluated temperatures. In this case, the average permeate flux was 75 Lh-1m-2. It was verified that clarification of camu-camu pulp requires a pretreatment of the pulp juice to ensure a high flux of permeate, and thus increase the process efficiency. It was also observed that, if volumetric concentration factor is increased while maintaining the same processing time, the content of the evaluated compounds is not significantly affected in the clarified juice.

Disponibilidade de padrões analíticos é um aspecto crítico no desenvolvimento de métodos de análises quantitativas de antocianinas. Para a obtenção dos padrões isolados de antocianinas cianidina-3-O-glicosídeo e cianidina-3-O-rutinosídeo foram utilizadas amostras liofilizadas de açaí (Euterpe oleraceae Mart.), que é um conhecido fruto de palmeiras no Brasil de forma arredondada e roxo. O isolamento das antocianinas foi realizado por Cromatografia Líquida de Alta Eficiência (CLAE), utilizando uma válvula seletora de seis canais. A identificação de antocianinas no açaí foi baseada em dados publicados na literatura e por espectrometria de massas. Após a coleta, padrões com um alto grau de pureza foram obtidos e uma curva para padronização externa de cada antocianina foi feita.

Availability of analytical standards is a critical aspect in developing methods for quantitative analysis of anthocyanins. The anthocyanins cyanidin-3-O-glucoside and cyanidin-3-O-rutinoside were isolated from samples of freeze-dried açaí (Euterpe oleraceae Mart.), which is a round and purple well-known palm fruit in Brazil, and then used as standards. The isolation of the anthocyanins was performed by High Performance Liquid Chromatography (HPLC), using an adapted six-channel selection valve. The identification of anthocyanin pigments in açaí was based on mass spectrometric data for molecular ions and MS-MS product ions and on previous published data. After the collection procedure, standards with a high purity grade were obtained and an external standard curve of each anthocyanin was plotted.

A osmose inversa tem sido utilizada para a concentração de sucos de frutas com resultados promissores em relação à qualidade dos produtos obtidos. Este trabalho teve como objetivo a concentração de suco de abacaxi por osmose inversa. A concentração foi conduzida com membranas de poliamida em um módulo de configuração quadro e placas com área total de permeação de 0,65 m² a 60 bar de pressão aplicada à membrana. O fluxo de permeado foi de 17 L.hm-2. O teor de sólidos solúveis do suco aumentou de 11 para 31 °Brix, correspondendo ao Fator de Redução Volumétrica (FRV) de 2.9. A concentração do teor de sólidos totais e acidez aumentaram proporcionalmente ao fator de redução volumétrica. O suco concentrado e três marcas de suco de abacaxi concentrado foram avaliados em relação à preferência e intenção de compra por 79 consumidores de suco de abacaxi. O suco concentrado por osmose inversa foi o preferido entre os consumidores. Pode-se concluir que este processo pode ser considerado como uma alternativa na concentração de sucos de frutas.

Reverse osmosis has been used for the concentration of fruit juices with promising considering the quality of the obtained products. The objective of this study was to concentrate single strength pineapple juice by reverse osmosis. The concentration was carried out with polyamide composite membranes in a 0.65 m² plate and frame module at 60 bar transmembrane pressure at 20 °C. The permeate flux was 17 L.hm-2. The total soluble solid content of the juice increased from 11 to 31 °Brix corresponding to a Volumetric Concentration Factor (VCF) of 2.9. The concentration of soluble solids, total solids, and total acidity increased proportionally to FCV. The concentrated juice and three commercial concentrated pineapple juices were evaluated regarding preference and purchase intention by 79 pineapple juice consumers. The concentrated juice by reverse osmosis was the preferred among consumers. It can be concluded that this process may be considered an alternative to the pre-concentration of fruit juices.
