La producción de caña de azúcar (Saccharum officinarum L.) está influenciada por varios factores abióticos desfavorables, como el estrés salino. La salinidad puede inducir el estrés oxidativo, que causa daño a las biomoléculas y a las estructuras celulares. Sin embargo, estos daños se puede evitar o minimizar por los sistemas de defensa enzimático y no enzimáticos de las plantas. El objetivo del estudio fue determinar el efecto de la prolina exógena en el contenido de Na+ e K+, y la actividad de la catalasa, ascorbato peroxidasa y peroxidasa en dos genotipos de caña de azúcar (RB931011 y RB872552) cultivados in vitro bajo estrés salino. Las plantas se cultivaron durante 20 días en presencia o ausencia de NaCl 100 mM y expuestos a prolina 20 mM durante un periodo de tiempo variable. Los datos fueron sometidos a ANOVA factorial (dos genotipos y cinco tratamientos), y las medias se compararon mediante la prueba de Tukey al 5 % de probabilidad. Bajo estrés salino, ambos genotipos mostraron reducción en la integridad de la membrana, disminución de las proteínas solubles totales y el mantenimiento o incremento en el contenido de prolina endógena. La prolina exógena reduce la acumulación de Na+ en proporción al período de exposición al aminoácido. En ambos genotipos, la actividad de las enzimas antioxidantes se incrementó con la adición de NaCl. En conclusión, el genotipo RB931011 mostró mayor acumulación de prolina y un mayor aumento de la actividad de la catalasa, ascorbato peroxidasa y peroxidasa, lo que indica una mejor tolerancia al estrés salino en comparación con el genotipo RB872552.

Sugarcane (Saccharum officinarum L.) production is influenced by various abiotic stresses, including salt stress. Salinity can induce oxidative stress, which in turn damages biomolecules and cellular structures. However, such damage can be avoided or minimized by the enzymatic and non-enzymatic defense systems of plants. The aim of the study was to determine the effect of exogenous proline on Na+ and K+ content and on the activity of catalase, ascorbate peroxidase and peroxidase in two sugarcane genotypes (RB931011 and RB872552) grown in vitro under salt stress. The plants were grown for 20 days with or without 100 mM NaCl and exposed to 20 mM proline for varying period of time. The data were subjected to ANOVA factorial (two genotypes and five treatments), and the mean values were compared by Tukey test at a 5 % probability. Under salinity stress, both genotypes exhibited membrane integrity reduction, reduced total soluble protein content and unaltered or increased endogenous proline content. Exogenous proline reduced Na+ accumulation in a manner proportional to the exposition period at the amino acid. In both genotypes, antioxidant enzymes activity increased with the addition of NaCl. In conclusion, RB931011 genotype showed higher proline accumulation and increased in activity of the antioxidant enzymes, indicating better salt stress tolerance than in RB872552 genotype.

Neste trabalho foram avaliados dez genótipos diplóides de bananeira (Musa spp) quanto a tolerância à salinidade, estresse abiótico que limita a produtividade da cultura. As plantas foram cultivadas durante 21 dias, em solução acrescida ou não de 100 mol m-3 de NaCl e analisadas variáveis de crescimento que incluem área foliar, biomassa fresca e seca, alocação de biomassa e taxa de crescimento. O experimento foi conduzido em delineamento inteiramente casualizado em fatorial 10 x 2 e três repetições. Na maioria dos genótipos estudados a salinidade provocou reduções em quase todas as variáveis analisadas. O genótipo Lidi destacou-se por apresentar melhor adaptação ao estresse salino, em todas as variáveis biométricas e capacidade de manutenção, sob estresse, da biomassa seca e fresca (limbo, caule, pseudocaule + raiz), área foliar, além de taxa de crescimento absoluto, entre outros. A produção relativa da biomassa seca da parte aérea foi superior a 70%, caracterizando este genótipo como tolerante e promissor para ser integrado a programas de melhoramento. Os genótipos Ouro e Tungia, por sua vez, sofreram grande redução da taxa de crescimento absoluto e a produção relativa da biomassa seca foi inferior a 50% caracterizando este genótipo como sensivel à salinidade.

Ten diploid banana genotypes (Musa spp.) were evaluated with regard to salt tolerance, abiotic stress which limits the productivity of the crop. The plants were grown during 21 days, in a nutrient solution with and without the addition of NaCl (0 and 100 mol m-3). Growth variables including leaf area, fresh and dry biomass, biomass allocation and growth rate were analysed. The experiment was implemented in a completely randomized design in factorial (10 x 2) arrangement and three replicates per treatment. Salinity caused in most genotypes reductions in almost all variables. Genotype Lidi stood out by having better adaptation to saline stress in all measured biometric variables with the ability to maintain, under stress, the fresh and dry biomass (leaf blade, stem, pseudostem + root), leaf area, and absolute growth rate among others. The relative production of dry biomass of shoots was more than 70%, characterizing this as a tolerant genotype and promising to be integrated into breeding programs. Genotypes Ouro and Tungia, in turn, showed reduction of absolute growth rate and relative dry biomass production (less than 50%) were characterized as sensitive to salinity.

FUNDAMENTOS - A hanseníase é endêmica na América do Sul, sendo responsável por 3% do total dos casos mundiais e, particularmente, no Brasil, por 85% dos casos sul-americanos. Seu agente pode ser encontrado na mucosa oral sem qualquer alteração evidente, e apenas testes laboratoriais muito sensíveis podem detectar sua presença. OBJETIVOS - Determinar se o genoma do Mycobacterium leprae pode ser encontrado pelo teste da PCR em biópsias com punch da mucosa oral de pacientes com hanseníase. MATERIAL E MÉTODOS - Realizou-se biópsia da mucosa oral normal de sete pacientes com hanseníase multibacilar. Cinco estavam em tratamento durante o estudo, e apenas um, ainda sem tratamento, teve o diagnóstico confirmado pela hematoxilina-eosina e coloração de Fite-Faraco para M. leprae. As peças foram incluídas em parafina e submetidas à PCR para pesquisa de M. leprae. RESULTADOS - Seis dos sete casos foram positivos para M. leprae, e um para Mycobacterium sp., demonstrando-se alta sensibilidade e especificidade do método. CONCLUSÃO - A PCR é método rápido, fácil e confiável para a investigação de rotina da infecção por micobactéria, mesmo quando a doença ainda é assintomática. O diagnóstico pode ser obtido a partir de simples biópsia ambulatorial.

BACKGROUND - Hansen's disease is endemic in South America, which accounts for 3% of total world cases, and particularly in Brazil, which accounts for 85% of all South American cases. The bacteria can be found in the oral mucosa with no evident signs of infection, and only very sensitive laboratory assays can detect its presence. OBJECTIVES - The aim of this study was to ascertain if the M. leprae genome can be detected by PCR in small punch biopsy specimens from the oral mucosa of patients with Hansen's disease. METHODS - The normal oral mucosas of seven multibacillary Hansen's disease patients were biopsied. Five of them were under treatment at the time of the study. Diagnosis of Hansen's disease could be determined by conventional hematoxylineosin and Fite-Faraco staining for M. leprae in only one patient. This patient had not received treatment prior to the time of the biopsy. The paraffin embedded specimens were submitted to PCR for M. leprae. RESULTS - Six out of seven cases showed positivity for M. leprae, and the remaining case showed positivity for Mycobacterium sp., demonstrating the high sensitivity and specificity of this method. CONCLUSIONS - PCR is a fast, easy and reliable method for routine investigation of mycobacterial infection, even during nonsymptomatic periods. Diagnosis can be achieved through a simple oral biopsy at an outpatient clinic.

OBJETIVO: avaliar a preservação folicular e as características celulares do tecido ovariano criopreservado, em coelhas. MÉTODOS: fez-se, sob anestesia, a ooforectomia direita de dez coelhas brancas, adultas. Dissecou-se o ovário mantendo-se o córtex com espessura de 1,5 milímetros. Fragmentou-se o tecido em pequenas secções, algumas para o estudo histológico de controle e outras destinadas à criopreservação pelo protocolo de congelamento lento. Passadas seis semanas efetuou-se o descongelamento e fez-se a avaliação histológica. As amostras do controle e do experimento, após processamento, foram coradas pela hematoxilina-eosina para identificação dos aspectos histológicos e submetidas à técnica imuno-histoquímica utilizando-se o PCNA (proliferating cell nuclear antigen) para a avaliação da viabilidade celular. Utilizaram-se os testes não paramétricos "comparação entre duas proporções" e Mann-Whitney e o teste paramétrico t de Student. RESULTADOS: observou-se que no tecido criopreservado só persistiram oócitos primordiais. Entre as alterações reversíveis identificaram-se: vacuolização citoplasmática em todas as amostras (p=0,039), lise estromal em 50% (p=0,648) e oócitos com contornos irregulares em 80% (p=0,007). Encontraram-se alterações irreversíveis como degeneração hialina e picnose em 30% das amostras (p=0,210). A análise imuno-histoquímica demonstrou os folículos, em diferentes estágios de desenvolvimento, no tecido não congelado e folículos primordiais no tecido criopreservado com positividade para o PCNA, indicando a presença de DNA ativo. CONCLUSÃO: no tecido ovariano criopreservado sobrevivem apenas os folículos primordiais; existem alterações histológicas reversíveis (vacuolização citoplasmática, lise estromal, fragmentação das células da granulosa e oócitos com contornos irregulares); alterações irreversíveis, em níveis não significantes (degeneração hialina e picnose) e presença de PCNA positivo em todos os folículos.

PURPOSE: to evaluate follicular preservation and histologic characteristics of the cryopreserved ovarian tissue and to compare with the fresh one, in rabbits. METHODS: ten adult female white rabbits were submitted to right oophorectomy. The dried ovary was dissected and the cortex was maintained with approximately 1.5 millimeter thickness. The tissue was fractionated into small sections, some reserved for control histologic study and others destined for cryopreservation. Six weeks later the ovarian tissue was thawed and evaluated histologically. After histologic processing, the control and the experimental samples were stained with hematoxylin and eosin and treated immunohistochemically by the PCNA technique for evaluation of DNA preservation. Histologic alterations present in the fresh and in the cryopreserved tissues were identified, and cryopreserved tissue viability was evaluated. RESULTS: in the cryopreserved tissue only primordial follicles persisted. Reversible alterations were identified: cytoplasmatic vacuolation (p=0,039), stromal lysis (p=0.648) and oocytes with irregular contours (p=0.007). Irreversible alterations: (hyalin degeneration and pyknosis) were found, but not at significant levels (p=0.210). The immunohistochemical analysis showed PCNA staining of follicles at different stages of development in the fresh tissue and primordial follicles in the cryopreserved tissue, indicating the presence of active DNA in both tissues. CONCLUSION: in the cryopreserved ovarian tissue the following were observed: survival of only primordial follicles; significant reversible histologic alterations (cytoplasmic vacuolation, stromal lysis and oocytes with irregular contours); irreversible alterations (hyalin degeneration and pyknosis), and PCNA staining of all follicles.

Quatro genótipos de milho (Zea mays L.), Jatinã C3 anão, BR-201, V-190 e LW-1, foram avaliados quanto à tolerância ao estresse salino (0, 50 e 100 mM NaCl) em condições hidropônicas. As plantas submetidas a 100 mM de NaCl durante 29 dias apresentaram uma redução na produção de matéria seca superior a 50%, e uma redução na área foliar superior a 75%, quando comparadas ao tratamento controle (0 mM NaCl). Dentre os quatro genótipos estudados, a linha pura LW-1 apresentou produção de matéria seca e área foliar inferiores aos demais. O aumento dos níveis de sal na solução resultou em uma diminuição da taxa de crescimento da parte aérea e da raiz, de todos os genótipos. O sistema radicular apresentou os maiores teores de sódio e também os maiores decréscimos na taxa de crescimento relativo, evidenciando o efeito negativo da acumulação deste cátion. O número e o tamanho dos estômatos não sofreram alterações em conseqüência do estresse salino.

Four maize (Zea mays L.) genotypes, Jatinã C3 anão, BR-201, V-190 and LW-1, grown under hidroponic conditions, were evaluated for their tolerance to saline stress imposed by three salt levels (0, 50 and 100 mM NaCl). Plants submitted to 100 mM NaCl for 29 days had 50% lower dry matter production and 75% reduced leaf area, as compared to the control (0 mM NaCl) plants. Among the four genotypes studied, LW-1, a pure line, produced the least dry matter and leaf area. The increase salt levels in solution decreased the growth rate of both shoot and root in all genotypes. Roots had the largest sodium concentration and also showed the largest decrease in the relative growth rate, demonstrading the negative effect of sodium accumulation. The number and size of the stomata were not affected by saline stress.
