Sandfly fauna (Diptera: Psychodidae) from caves in the state of Rondônia, Brazil

Resumo O objetivo deste estudo foi conhecer a fauna de flebotomíneos, e possível presença de Leishmania nestes insetos, coletados em cavernas do Estado de Rondônia. As coletas foram realizadas em oito cavernas localizadas em duas áreas diferentes do Estado. A detecção de Leishmania nos flebotomíneos foi realizada por reação em cadeia da polimerase (PCR). Este foi o primeiro trabalho com flebotomíneos em cavernas de Rondônia e um total de 1,236 indivíduos foram coletados e identificados em 24 espécies e 10 gêneros.Evandromyia georgii foi coletada pela primeira vez em Rondônia, e as espécies mais abundantes foram Trichophoromyia ubiquitalis com 448 indivíduos (36.2%) seguida por T. octavioi com 283 (22.9%) e E. georgii com 179 (14.5%). No estudo de PCR, 17 pools foram analisados, sendo cinco positivos (T. auraensis - 3, Nyssomyia shawi eN. antunesi - 1 cada). A região do kDNA foi amplificada confirmando a presença de DNA de Leishmania. A fauna de flebotomíneos nestas cavernas foi considerada diversa quando comparada com estudos semelhantes de outras regiões. É possível que algumas espécies utilizem cavernas como abrigo temporário e local de procriação e outras sejam exclusivas deste ambiente. A detecção de DNA de Leishmania indica que este patógeno está circulando no ambiente cavernícola, sendo necessários mais estudos para conhecer o risco de transmissão de leishmanioses nestes locais com alto potencial turístico.
Abstract This study had the aim of ascertaining the sandfly fauna and possible presence ofLeishmania in these insects, collected in caves in the state of Rondônia, Brazil. Collections were conducted in eight caves located in two different areas of this state. Leishmania in the sandflies collected was detected using the polymerase chain reaction (PCR). This was the first study on sandflies from caves in Rondônia and, among the total of 1,236 individuals collected, 24 species and 10 genera were identified. The speciesEvandromyia georgii was collected for the first time in Rondônia and the most abundant species were Trichophoromyia ubiquitalis with 448 individuals (36.2%), followed by T. octavioi with 283 (22.9%) and E. georgii with 179 (14.5%). For the PCR, 17 pools were analyzed and five pools were positive (forT. auraensis in three pools and for Nyssomyia shawi and N. antunesi in one pool each). The kDNA region was amplified and the presence of Leishmania DNA was confirmed. The sandfly fauna in these caves can be considered diverse in comparison with similar studies in other regions. It may be that some species use caves as a temporary shelter and breeding site, while other species live exclusively in this environment. The detection of LeishmaniaDNA indicates that this pathogen is circulating in cave environments and that further studies are needed in order to ascertain the risks of infection by leishmaniasis in these locations with high touristic potential.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

High occurrence of Entamoeba histolytica in the municipalities of Ariquemes and Monte Negro, State of Rondônia, Western Amazonia, Brazil

Introdução: Infecções por Entamoeba histolytica foram investigadas em moradores dos municípios de Ariquemes e Monte Negro, Rondônia, Brasil. Métodos: Amostras de fezes de 216 indivíduos foram processadas por microscopia óptica para detecção de cistos do complexo E. histolytica/E. dispar, seguido pelo método de imunoensaio utilizando kit de ensaio imunoenzimático para detecção específica de antígeno de E. histolytica. Resultados: Cistos de E. histolytica/E. dispar estavam presentes em 61% e 44% das amostras de Ariquemes e Monte Negro, respectivamente com diferença significativa na ocorrência da infecção entre as duas populações [p < 0,05; χ2 = 5,2; Odds relativa = 2,0 (1,1 - 3,6)]. A taxa de detecção de antígenos de E. histolytica nas amostras provenientes de Ariquemes foi de 36,6% e de 19,41% nas amostras de Monte Negro, sendo a ocorrência de amebíase significativamente maior na população de Ariquemes [p < 0,05; χ2 = 7,8; Odds relativa = 2,4 (1,2 - 4,7)]. Discussão: A elevada frequência da infecção por E. histolytica em residentes na região, bem como a indisponibilidade de avaliação clínica por testes específicos para distinção entre as duas espécies de Entamoeba, deve promover uma reflexão sobre o tratamento de infecções pelo complexo E. histolytica/E. dispar. Conclusão: Nas populações avaliadas foram detectadas elevadas ocorrências de E. histolytica.
Introduction: Entamoeba histolytica infections were investigated in residents of the Ariquemes and Monte Negro municipalities in Rondônia State, Brazil. Methods: Stool samples of 216 individuals were processed by the spontaneous sedimentation method and analyzed by microscopy for detection of the E. histolytica/E. dispar complex, followed by the immunoassay method using an enzyme-linked immunosorbent assay-based kit for the E. histolytica stool antigen. Results: E. histolytica/E. dispar cysts were present in 61% (50/82) and 44% (59/134) of the samples from Ariquemes and Monte Negro respectively, with a significant difference in the occurrence of infection between the two populations [p < 0.05; χ2 = 5.2; odds ratio = 2.0 (1.1 - 3.6)]. The E. histolytica antigen detection rate was 36.6% (30/82) for stool samples from Ariquemes, and 19.4% (26/134) for stool taken from the residents of Monte Negro. The rate of the occurrence of amoebiasis was significantly higher in the population from Ariquemes [p < 0.05; χ2 = 7.8; odds ratio = 2.4 (1.2 - 4.7)]. Discussion: Due to the high occurrence of E. histolytica infected residents diagnosed in the region and the unavailability in local clinics of a test to distinguish between the two Entamoeba species, physicians should consider treating E. histolytica/E.dispar infections. Conclusion: The results indicate that E. histolytica infection is highly endemic in the studied areas.

New resistance genes in the Zea mays: exserohilum turcicum pathosystem

The use of monogenic race-specific resistance is widespread for the control of maize (Zea mays L.) helminthosporiosis caused by Exserohilum turcicum. Inoculation of 18 Brazilian isolates of E. turcicum onto elite maize lines containing previously identified resistance genes and onto differential near-isogenic lines allowed the identification of new qualitative resistance genes. The inoculation of one selected isolate on differential near-isogenic lines, F1 generations and a BC1F1 population from the referred elite lines enabled the characterization of the resistance spectrum of three new genes, one dominant (HtP), one recessive (rt) and a third with non-identified genetic action. Three physiological races of the pathogen were also identified including two with new virulence factors capable of overcoming the resistance of one of the resistance genes identified here (rt).

Este trabalho objetivou identificar marcadores microssatélites ligados a genes de resistência a Puccinia polysora e verificar o efeito fenotípico destes nas variáveis monocíclicas número e comprimento de lesão. Foram utilizadas duas linhagens (Z-95 e Z-93) contrastantes em níveis de resistência à doença, o híbrido (Z-95 x Z-93) e uma população F2 obtida da autofecundação deste. Esses indivíduos foram fenotipados para resistência à doença em dois ensaios a campo e genotipados para 142 marcadores microssatélites. Para agilizar a genotipagem, o método de análise de segregantes agrupados (ASA) foi utilizado. Marcadores potencialmente ligados a genes de resistência identificados por este método foram utilizados para genotipar 165 indivíduos segregantes e confirmar a existência de ligação. Dois marcadores, Phi 65 e Phi 28, ambos no cromossomo 9, mostraram-se significativamente associados (p<0,000001 e p<0,000078, respectivamente) a um QRL (locos de resistência quantitaviva) a P. polysora. A associação entre QRL e marcadores explicou, respectivamente, 12,9% e 5,10% da variação fenotípica para resistência. Em um terceiro ensaio em casa de vegetação, 94 plantas foram inoculadas com suspensão de uredósporos e avaliadas 15 dias após a inoculação quanto ao número de lesões e o comprimento de dez lesões. Estas plantas foram genotipadas com os marcadores Phi 65 e Phi 28. Somente Phi 65 mostrou-se significativamente associado (P< 0,000032) à redução no número de lesões. Nenhum marcador mostrou associação significativa com a variável comprimento de lesão. Por estarem ligados ao mesmo marcador, sugere-se que o QRL identificado nos ensaios a campo seja o mesmo identificado em casa de vegetação.
The objectives of this study were to identify microsatellite markers linked to resistance gene of resistance to Puccinia polysora and to estimate the phenotypic effect of these genes on two monocyclic variables lesion, number and length. Two inbreed lines with different degrees of resistance (Z-95 and Z-93), the hybrid (Z-95 X Z-93) and F2 plants derived by selfing this hybrid were phenotyped for disease resistance, in two field trials and genotyped for 142 microsatellite markers using bulked segregant analysis (BSA). The molecular markers putatively linked to the disease resistance genes identified by this method were used to genotype 165 segregant individuals and to confirm linkage. Two markers, Phi 65 and Phi 28, both located on chromosome nine, were significantly associated (p<0.000001 and p<0.000078) with a QRL (quantitative resistance loci) to P. polysora. The association explained 12.9 and 5.10% of the phenotypic variance in resistance for Phi 65 and Phi 28, respectively. In a third trial performed in the greenhouse, 94 plants were inoculated with a uredospore suspension and evaluated 15 days after inoculation for the total number of lesions and the length of ten lesions. These plants were genotyped with Phi 65 and Phi 28. Only Phi 65 showed a significant association with lesions number (p<0.000032). No marker showed significant association with lesion length. It is suggested that the QRL identified in the field trials is the same identified in the greenhouse experiment, since they are linked to the same marker.
