Multidrug-resistant Escherichia coli isolated from a dog with a history of urolithiasis: case report

RESUMO A resistência bacteriana aos antibióticos é uma realidade, tanto na saúde humana quanto veterinária, limita o arsenal terapêutico e eleva os custos relacionados ao tratamento do paciente. Um cão, com sinais de cistite, recebeu tratamento com enrofloxacina, na dose de 5mg/kg, em três momentos seguidos, com baixa efetividade. Identificou-se presença de urólitos uretrais e foi feita uro-hidropropulsão. O animal apresentou nova obstrução, para a qual foi realizada uma cistotomia, mas continuou com sinais de infecção. Realizou-se, então, urocultura e teste de antibiograma. Foi identificada Escherichia coli, que se mostrou resistente a 13 antibióticos, sendo sensível somente à piperacilina-tazobactam e amicacina. No teste de triagem para β-lactamase, detectou-se a produção de ESβL. A qPCR indicou presença dos genes blaCTXm, blaDHA, blaOXA, blaIMP, blaTEM, blaGIM, blaSIM, blaSPM e blaSME, que podem conduzir um perfil fenotípico de resistência para ampicilina, amoxicilina-ácido clavulânico, aztreonam, cefepima, cefoxitina, cefuroxima, ceftazidima, ceftriaxona, imipenem, piperacilina-tazobactam. Este caso reafirma o valor que a análise laboratorial agrega ao diagnóstico e tratamento da cistite e da urolitíase, podendo definir o sentido de evolução do prognóstico e a velocidade em que a saúde do paciente será restabelecia.
ABSTRACT Bacterial resistance is a reality in both human and veterinary health, it limits the therapeutic arsenal and raises the costs of the patient’s treatment. A dog with signs of cystitis received treatment with 5mg/kg enrofloxacin at three consecutive times, with low effectiveness. The presence of urethral uroliths was identified and urohydropulsion was done. The animal presented a new obstruction, for which a cystotomy was performed, but continued with signs of infection. Uroculture and antimicrobial susceptibility test were then performed. Escherichia coli was identified, which was resistant to 13 antibiotics, being sensitive only to piperacillin-tazobactam and amikacin. In the screening test for β-lactamase, the production of ESβL was detected. The qPCR indicated the presence of the bla CTXm, bla DHA, bla OXA, bla IMP, bla TEM, bla GIM, bla SIM, bla SPM and bla SME genes, which may lead to a phenotypic resistance profile for ampicillin, amoxicillin-clavulanate, aztreonam, cefepime cefoxitin, cefuroxime, ceftazidime, ceftriaxone, imipenem, and piperacillin-tazobactam. This case reaffirms the value that laboratory analysis adds to the diagnosis and treatment of cystitis and urolithiasis, which can define the direction of evolution of the prognosis and the speed at which the patient's health will be restored.

Resistance patterns, ESBL genes, and genetic relatedness of Escherichia coli from dogs and owners

Abstract Antimicrobial resistance in Escherichia coli isolated from pet dogs can be considered a potential threat of infection for the human population. Our objective was to characterize the resistance pattern, extended spectrum beta-lactamase production and genetic relatedness of multiresistant E. coli strains isolated from dogs (n = 134), their owners (n = 134), and humans who claim to have no contact with dogs (n = 44, control), searching for sharing of strains. The strains were assessed for their genetic relatedness by phylogenetic grouping and pulsed-field gel electrophoresis. Multiresistant E. coli strains were isolated from 42 (31.3%) fecal samples from pairs of dogs and owners, totaling 84 isolates, and from 19 (43.1%) control group subjects. The strains showed high levels of resistance to ampicillin, streptomycin, tetracycline, trimethoprim and sulfamethoxazole regardless of host species or group of origin. The blaTEM, blaCTX-M, and blaSHV genes were detected in similar proportions in all groups. All isolates positive for bla genes were ESBL producers. The phylogenetic group A was the most prevalent, irrespective of the host species. None of the strains belonging to the B2 group contained bla genes. Similar resistance patterns were found for strains from dogs, owners and controls; furthermore, identical PFGE profiles were detected in four (9.5%) isolate pairs from dogs and owners, denoting the sharing of strains. Pet dogs were shown to be a potential household source of multiresistant E. coli strains.

Perfil hematológico de cabras Saanen e mestiças (1/2 Saanen e 1/2 Anglo-nubiana) criadas em clima tropical do Ceará

Objetivou-se verificar a influência das condições climáticas sobre o perfil hematológico de cabras Saanen e mestiça (1/2 Saanen x 1/2 Anglo-nubiana), bem como determinar valores de referência para esses animais criados no Ceará. Foram utilizadas 30 cabras que tiveram o sangue coletado mensalmente durante a época chuvosa (fevereiro a maio) e seca (agosto a novembro) para realização do eritrograma e leucograma. As médias foram comparadas pelo teste t-Student e Mann Whitney, quando constatada distribuição paramétrica e não paramétrica dos dados, respectivamente, com significância quando p<0,05. Procedeu-se ainda um estudo de correlações simples de Pearson dos parâmetros hematológicos com variáveis ambientais e fisiológicas. O número de hemácias foi maior em cabras Saanen e na época chuvosa, enquanto que o hematócrito foi maior na época seca (p<0,05). Os leucócitos e linfócitos foram maiores em cabras mestiças nas duas épocas (p<0,05). Nos dois genótipos, os leucócitos e linfócitos foram maiores na época seca e os neutrófilos segmentados maiores na época chuvosa (p<0,05). Os demais parâmetros não diferiram (p>0,05). As hemácias e neutrófilos segmentados apresentaram uma correlação negativa com a temperatura ambiente, porém positiva com a umidade do ar e a temperatura retal (p<0,05). O hematócrito correlacionou-se positivamente com a temperatura ambiente e frequência respiratória (p<0,05). Os leucócitos e linfócitos mostraram uma correlação negativa com a temperatura retal (p<0,05). Conclui-se, portanto, que fêmeas Saanen sofrem mais influência das variações climáticas e a época chuvosa tem maior impacto negativo sobre os parâmetros hematológicos. Os valores obtidos poderão servir como referência para esses genótipos no Ceará.
The aim of this study was to determine the influence of climatic conditions on the hematological profile of Saanen and mixed-breed (1/2 Saanen x 1/2 Anglo-Nubian) goats, as well as to define reference values for these animals bred in Ceará, Brazil. Thirty goats were utilized and blood samples were collected monthly during the rainy (February to May) and dry (August to November) periods to obtain an erythrogram a leukogram. The averange were compared by the t-Student test and Mann Whitney test, with parametric and non-parametric distribution of the data, respectively, where p<0.05 was considered significant. A study was carried out of simple Pearson correlations of the hematological parameters with environmental and physiological variables. The number of red blood cells (RBCs) was higher in the Saanen goats and in the rainy period, while the hematocrit was higher in the dry period (p<0.05). The leukocytes and lymphocytes were higher in the 1/2S1/2AN goats in the two periods (p<0.05). In the two genotypes, the leukocytes and lymphocytes were higher in the dry season and the segmented neutrophils higher in the rainy season (p<0.05). The other parameters did not differ (p>0.05). The RBCs and segmented neutrophils displayed a negative correlation with air temperature, but positive with relative humidity and rectal temperature (p<0.05). The hematocrit positively correlated with air temperature and respiratory rate (p<0.05). The leukocytes and lymphocytes showed a negative correlation with rectal temperature (p<0.05). It was therefore concluded that Saanen females are more affected by climatic variations and that the rainy season has a greater negative impact on hematological parameters. The values obtained could serve as a reference for these genotypes in Ceará.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Com o crescimento da indústria sucroalcooleira, buscam-se alternativas de uso dos resíduos gerados por ela. Os pequenos ruminantes de raças naturalizadas do Nordeste brasileiro mostram-se como grupos potenciais para que seja utilizado o bagaço de cana-de-açúcar (BCA) nas dietas. Nesse contexto, objetivou-se determinar a degradabilidade ruminal in situ da matéria seca (MS) e da fibra em detergente neutro (FDN) do BCA e os parâmetros ruminais em caprinos Moxotó e ovinos Morada Nova, fistulados no rúmen. Contidos em sacos de náilon, 3g de BCA foram incubados no rúmen nos tempos seis, 24 e 96 horas, determinando-se o conteúdo de MS e FDN nos resíduos obtidos. Nos tempos zero, seis e 12 horas após a primeira refeição, mediram-se no líquido ruminal pH e nitrogênio amoniacal (N-NH3) ruminal. O CMS não diferiu entre caprinos e ovinos. O potencial de máxima degradação da MS foi semelhante entre espécies, e da FDN foi superior em caprinos. Ovinos apresentaram maiores tempo de colonização, taxa de degradação e degradabilidade efetiva da MS e FDN. O pH não diferiu entre as espécies. Observou-se maior concentração de N-NH3 ruminal em caprinos, no tempo zero. Diante da maior velocidade de degradação da MS do BCA pelos ovinos, essa espécie se mostra detentora de uma microbiota ruminal com crescimento mais eficiente sobre o BCA.
With the increase of the sugar and alcohol industries, alternative uses for residue are sought, and the small ruminants from the naturalized breeds of northeastern Brazil are potential groups to make use of the sugarcane bagasse (SCB) in their diets. In this context, the objective was to determine the in situ degradability of dry matter (DM) and neutral detergent fiber (NDF) of SCB and ruminal parameters in Moxotó goats and Morada Nova sheep rumen fistulated. Three grams of SCB were placed in naylon bags and incubated in the rumen at 6, 24 and 96 hours, and the residues analyzed for DM and NDF. Rumen fluid was collected at zero, 6 and 12 hours after the first meal and pH and ammonia nitrogen (N-NH3) were determined. DMI did not differ between goats and sheep. The maximum degradation potential of DM was similar between species and the A of NDF was higher in goats. Sheep had higher colonization time, rate of degradation and effective degradability of DM and NDF. The pH did not differ between species. The highest concentration of ruminal N-NH3in goats was at time zero. Given the higher rate of degradation of bagasse dry matter by sheep, this specie shows a more efficient rumen microbial growth on SCB.

Fatores associados à ocorrência da tuberculose bovina (TB), em dados de 209 fazendas, foram avaliados por meio de análise de variância. Os dados utilizados foram provenientes do levantamento epidemiológico por demanda, realizado nos rebanhos do estado do Rio de Janeiro, no período de 1959 até 1989. As variáveis significativas na análise bivariada foram incluídas no modelo e analisadas conjuntamente pelo método do modelo linear generalizado. A taxa de prevalência da TB nos rebanhos foi de 49,8%. As variáveis detectadas no estudo como as mais importantes para a ocorrência de TB foram: aleitamento (P=0,03) e as interações densidade x aleitamento (P=0,04) e produtividade x período (P=0,02). A importância da organização econômica da produção pecuária na ocorrência da TB pode ser evidenciada pelos resultados obtidos neste estudo. Este é um fator relevante em decorrência dos danos à saúde humana e animal.
The factors associated to the occurrence of bovine tuberculosis (TB) in 209 farms were evaluated by variance analysis. Data came were colleted in an epidemiological investigation from herds in the State of Rio de Janeiro, examined from 1959 to 1989. The significant variables were analyzed and submitted to generalized linear model. The prevalence rate of TB in herds was 49.8%. The most important variations detected in this study for occurrence of TB were: sucking (P=0.03) and interactions density vs. sucking system (P=0.04) and productivity vs. sucking period (P=0.02). The influence of the economical aspects of the farming systems on the occurrence of TB was confirmed by the results of this study. This is an important factor due to human and animal health hazards.

Foram utilizadas técnicas de análise multivariada para comparar dois híbridos obtidos de linhas de frango de corte desenvolvidas pela Universidade Federal de Viçosa com dois híbridos comerciais. As características avaliadas foram: peso ao abate (PAB), peso da carcaça (PC), peso do peito (PP), peso da contra-coxa (PCC), peso da coxa (PCX) e rendimento de carcaça (RCA). A análise de variância multivariada indicou diferença significativa entre os vetores de médias das características. Pelo teste de Roy, observou-se que para PC e PP os produtos comerciais foram superiores e para PCX e RCA não houve diferenças significativas entre os produtos comercias e os cruzamentos. Quanto à função discriminante linear de Fisher, observou-se superioridade significativa dos produtos comerciais pelo teste de Roy.
Multivariate analyses were used to compare two hybrids obtained from two strains of broilers developed by Federal University of Viçosa (UFV) with two commercial hybrids. The evaluated traits were: slaughter weight (PAB), carcass weight (PC), breast weight (PP), upper thigh weight (PCC), thigh weight (PCX) and carcass yield (RCA). Multivariate analyses, showed significant differences among mean vectors of traits. The Roy test indicated the commercial products were better for PC and PP, and no differences between commercial products and UFV crosses were observed for PCX and RCA. The discriminant linear function of Fisher also indicated that commercial products were better than UFV crosses by the test of Roy.

A divergência genética entre seis linhas de aves Legorne (L1, L2, L3, L4, L5 e L6), desenvolvidas pela UFV, foi avaliada utilizando análise de variáveis canônicas e o método de agrupamento de Tocher. Foram incluídas no estudo oito características: peso corporal na 40ª semana (PC40), na 48ª semana (PC48), na 56ª semana (PC56); peso do ovo na 40ª semana (PO40), na 44ª semana (PO44), na 52ª semana (PO52), na 60ª semana (PO60) e taxa de postura da 40ª a 62ª semana (TP). Foi observada diferença entre as linhas quanto às características estudadas. A linha L4 mostrou-se divergente das demais, apresentando a menor média canônica, e foi alocada em grupo distinto das outras pelo teste de Tocher. O desempenho das diferentes linhas foi também avaliado por meio da análise de variância multivariada, usando o teste do maior autovalor de Roy, e por meio do teste de Roy para comparações múltiplas. Verificou-se divergência genética entre as linhas da UFV, sendo PC40 a característica que mais contribuiu para a divergência.
Genetic divergence among six Leghorn lines (L1, L2, L3, L4, L5 e L6), developed by Universidade Federal de Viçosa, Brazil, was evaluated using canonical variate analysis and grouping method of Tocher. Eight traits were used: body weight at 40 weeks, at 48 weeks, at 56 weeks; egg weight at 40 weeks, at 44 weeks, at 52 weeks, at 60 weeks and laying ratio from 40 to 62 weeks of age. Significant differences were observed among lines for the studied traits. The line L4 was divergent in comparison with the other ones, showed the smallest canonical mean and was allocated in a different group. The performance of different genetic groups was also evaluated by multivariate analysis of variance, using Roy test of the largest eigenvalue and the Roy principle for multiple comparisons. Genetic divergence among UFV's lines was observed and body weight at 40 weeks was the trait that more contributed for this divergence.

A divergência genética entre quatro linhas de matrizes de frango de corte (L1, L2, L3 e L4) foi estudada usando métodos de análise multivariada. Foram avaliadas quatro características de importância econômica, em três períodos (inicial, médio e total): idade ao primeiro ovo, número de ovos, peso médio do ovo e peso corporal da matriz. O estudo da divergência genética entre as linhas foi feito por meio da análise de variância multivariada, análise de agrupamento usando a distância generalizada de Mahalanobis e o método de otimização de Tocher, e análise por meio de variáveis canônicas. Verificaram-se diferenças significativas pelo teste do maior autovalor de Roy entre os vetores de médias das linhas avaliadas nos três períodos. A análise de agrupamento nos períodos inicial (L1 e L2; L3 e L4) e médio (L2 e L4; L1 e L3) agrupou os quatro genótipos em três grupos, No período total os genótipos foram agrupados em apenas dois grupos (L2, L3, L4 e L1). Na análise por meio de variáveis canônicas verificou-se que as duas primeiras variáveis canônicas explicaram mais de 96% da variação total nos três períodos, o que possibilitou representar o comportamento dos genótipos em um gráfico de dispersão bidimensional. Observou-se que a divergência genética diminuiu quando se considerou o período total de produção.
Genetic divergence among broiler lines (L1, L2, L3 and L4) was studied by multivariate analysis. Four economic important traits were evaluated in three periods (initial, medium and total): age at first egg, egg number, average egg weight and body weight. The study of the genetic divergence among lines included the following techniques:Multivariate analysis of variance, cluster analysis using Mahalanobis generalized distance and Tocher optimization method, and canonical variables analysis. Significant difference was obtained by the larget Roy eigenvalue test. The cluster analysis of traits in the initial (L1 and L2; L3 and L4) and medium (L2 and L4; L1 and L3) periods grouped the four genotypes in three groups, and in total period the genotypes were grouped in two groups (L2, L3, L4 and L1). Canonical variables analysis showed that the first two canonical variables accounted for more than 96% of the total variation in the three periods. This allowed to represent the performance of genotypes in a bidimensional dispersion graph. In the total period of production study the genetic divergence was lower.
