ABSTRACT: Porcine enzootic pneumonia (PES), mainly caused by the bacteria Mycoplasma hyopneumoniae, is the main cause of respiratory problems in pigs. Infection by M. hyopneumoniae leads to production losses and the predisposition of affected animals to secondary infections, which may result in the condemnation of carcasses and organs due to lung lesions at the time of slaughter. The objective of the research was to evaluate the infection by M. hyopneumoniae in pigs submitted to slaughter in São Luís Island/MA, using molecular and histopathological diagnostic methods. One hundred fifty lung samples were collected from inspected (n=65) and non-inspected (n=85) slaughter pigs on São Luís Island, Maranhão, from July 2019 to August 2021. Of the 150 DNA samples collected, 121 showed an amplified product for Cyt B in the PCR assay. Thus, 121 samples were submitted to qPCR of M. hyopneumoniae, of which 44 (36.36%) showed positive results. The mean amount of bacterial load ranged from 1.20 × 101 to 7.20 × 104, with a mean of 1.73 × 104 copies. Of the reagent samples, 81.81% (36 samples) were obtained from non-inspected slaughter, while 18.18% (8 samples) were obtained from slaughterhouses. In the histopathological analysis, 44 positive qPCR samples were evaluated, of which 28 (63.63%) presented results compatible with the main inflammatory process associated with the presence of M. hyopneumoniae, that is, bronchial-associated lymphoid tissue hyperplasia (BALT). Three samples that showed the highest bacterial load (qPCR: 5.63 × 10³, 2.19 × 104 and 7.23 × 104) showed more evident lesions in this study. The microscopic findings associated with the quantifications indicated a relationship between the amount of bacterial load and the presence of microscopic lesions; higher bacterial load in lung tissue is associated with increased histopathologic staining for BALT hyperplasia. In conclusion, the results point to the circulation of the etiological agent in the sampled animals and the need for preventive measures on pig farms in Maranhão with the involvement of producers, sanitary defense and inspection agencies.

RESUMO: A pneumonia enzoótica suína (PES), causada principalmente pela bactéria Mycoplasma hyopneumoniae, é a principal causa de problemas respiratórios em suínos. A infecção por M. hyopneumoniae leva a perdas produtivas e a predisposição dos animais acometidos a infecções secundárias, o que pode resultar em condenação de carcaças e órgãos por lesões pulmonares no momento do abate. O objetivo da pesquisa foi avaliar a infecção por M. hyopneumoniae em suínos submetidos ao abate na Ilha de São Luís, por meio de métodos diagnósticos moleculares e histopatológicos. Para isso, foram coletadas 150 amostras de pulmão de suínos de abate inspecionado (n=65) e não inspecionado (n=85) na Ilha de São Luís/Maranhão, no período de julho de 2019 a agosto de 2021. Das 150 amostras de DNA coletadas, 121 apresentaram produto amplificado para Cyt B no ensaio de PCR. Assim, 121 amostras foram submetidas à qPCR de M. hyopneumoniae, das quais 44 (36,36%) apresentaram resultados positivos. A quantidade média de carga bacteriana variou de 1,20 × 101 a 7,20 × 104, com média de 1,73 × 104 cópias. Das amostras reagentes, 81,81% (36 amostras) foram obtidas de abate não inspecionado, enquanto 18,18% (8 amostras) foram obtidas em abatedouro. Na análise histopatológica, foram avaliadas 44 amostras positivas para qPCR, das quais 28 (63,63%) apresentaram resultados compatíveis com o principal processo inflamatório associado à presença de M. hyopneumoniae, ou seja, hiperplasia do tecido linfóide associado ao brônquio (BALT). Três amostras que apresentaram maior carga bacteriana (qPCR: 5,63 × 10³, 2,19 × 104 e 7,23 × 104) foram mais evidentes neste estudo. Os achados microscópicos associados às quantificações indicaram uma relação entre a quantidade de carga bacteriana e a presença de lesão microscópica; a maior carga bacteriana no tecido pulmonar está associada a maior alteração histopatológica para hiperplasia BALT. Em conclusão, os resultados obtidos sinalizam para a circulação do agente etiológico nos animais amostrados e a necessidade de medidas preventivas nas criações de suínos do estado do Maranhão com envolvimento dos produtores, órgãos de defesa sanitária e inspeção.

Maternal anxiety symptoms in the perinatal period might have long-term health effects on both the mother and the developing child. Valerian is a phytotherapeutic agent that is widely used for the treatment of anxiety. This study investigated the effects of valerian treatment in postpartum rats on maternal care, toxicity, and milk composition. Postnatal development, memory, and anxiety behavior in the offspring were also assessed. Postpartum Wistar rats received the valerian (500, 1000, or 2000 mg·kg–1·day–1) by oral gavage. Clinical and biochemical toxicity was evaluated with commercial kits. Maternal behavior was observed daily. Milk composition was analyzed by colorimetric methods. Physical and neuromotor tests were used to analyze postnatal development. Anxiolytic activity was assessed by the elevated plus maze, and memory was evaluated by the step-down inhibitory avoidance task. Maternal toxicity and care behavior were not altered by the treatment, while only the highest dose promoted a significant increase of lactose, and the doses 1000 and 2000 mg·kg–1·day–1 promoted a reduction of protein contents in milk. Postnatal development was similar in all offspring. Adult offspring did not display altered anxiety behavior, while long-term memory was impaired in the female adult offspring by maternal treatment with 1000 mg·kg–1·day–1. These results suggested that high doses of valerian had significant effects on important maternal milk components and can cause long-term alterations of offspring memory; thus, treatment with high doses of valerian is not safe for breastfeeding Wistar rat mothers.

Abstract The present study is the first report of Spiroxys larvae parasitizing stomach and intestine of three fish species in stream of Caatinga domain, Northeast, Brazil. A total of 120 fish specimens, 40 of Astyanax bimaculatus, 40 of Hoplias malabaricus, and 40 of Hoplosternum littorale were examined for nematodes. A total of 633 specimens of Spiroxys larvae were recovered for the three fish species: 227 in A. bimaculatus (prevalence (%) = 48; mean abundance = 5.78 ± 1.78 (range = 0–70); and mean intensity = 11.95 ± 2.7 (range = 2–70)), 176 in H. malabaricus (prevalence (%) = 42.5; mean abundance = 4.40 ± 1.55 (range = 0–53); and mean intensity = 10.35 ± 2.38 (range = 2–53)) and 230 in H. littorale (prevalence (%) = 42.5; mean abundance = 5.75 ± 1.98 (range = 0–52); and mean intensity = 13.53 ± 3.04 (range = 1–52)). This study expands the range of the geographic distribution of nematodes of the genus Spiroxys and increase the list of hosts, as well as contributing to the knowledge of fish parasites biodiversity in Caatinga domain.

RESUMO Diferentes tipos de queijos artesanais são produzidos, comercializados e consumidos no Brasil, o que impulsiona o constante desenvolvimento de normas por órgãos oficiais, como o Mapa. A criação do Suasa e do Sisbi-POA foi fundamental para esse setor, por permitir um sistema de equivalência na fiscalização e por ampliar a distribuição. Ainda, o Mapa passou a permitir que queijos artesanais produzidos com leite cru pudessem ser maturados em um período inferior a 60 dias, desde que comprovada sua inocuidade. A redução do tempo de maturação é um tema controverso e polêmico, já que não há critérios específicos que estudos científicos devem contemplar, o que permite múltiplas interpretações de dados. Com a criação e a regulamentação do selo Arte, a fiscalização dos produtos artesanais foi designada aos órgãos de agricultura, pecuária e de saúde pública, em complementação à atribuição já prevista pelo Mapa e pelo Sisbi-POA. Ainda, o selo Arte atribui aos órgãos de inspeção uma função orientadora, atividade que deveria ser prioritariamente executada por agências de extensão e associações. As normas que balizam a produção e comercialização de produtos artesanais devem ser frequentemente atualizadas, devido aos constantes avanços científicos na área e para assegurar a oferta de produtos com qualidade e inócuos aos consumidores.

ABSTRACT Different artisanal cheeses are produced, commercialized and consumed in Brazil, leading to a constant development of related rules by the MAPA and other official agencies. The establishment of two national programs (SUASA and SISBI-POA) allowed an equivalence in inspection system and an expanded distribution. Also, MAPA allowed ripening time lower than 60 days for artisanal raw milk cheeses, based on scientific studies that assure their safety. However, lowering the ripening period is still controversial, once there are no proper established criteria for such scientific studies, leading to potential multiple interpretation of data. The newly established ARTE certification transferred the inspection responsibilities of artisanal products to secretaries of agriculture, livestock and health, in support of what was already predicated by MAPA and SISBI-POA. Based on ARTE certification, the inspection service must also provide orientation guidance to producers, which should be done specifically by extension organs and associations. The norms that guide the production and commercialization of these artisanal products often need to be updated, but based on well-established methodologies and procedures, to ensure the distribution of suitable products to consumers.

RESUMO Caracterizaram-se fêmeas F1 Holandês x Zebu de diferentes bases maternas quanto às pelagens, despigmentações e características morfométricas. Foram utilizadas 266 fêmeas F1, progênies do cruzamento de 26 touros da raça Holandesa com fêmeas de composição genética zebuínas: Gir, Nelore, Guzonel, Nelogir. Foram aplicadas análise de distribuição de frequência para características qualitativas e medidas de dispersão e tendência central para características morfométricas, e as médias foram comparadas pelo teste de Tukey a 5% de probabilidade. Acima de 60,0% dos animais foram de pelagem preta. As vacas que tiveram origem na raça Gir apresentaram comprimento de cabeça 2,8cm maior (P<0,05) que as fêmeas da raça Nelore. O comprimento da orelha variou (P<0,05) conforme a base materna utilizada. As vacas com genes da raça Nelore são 5,0cm mais altas (P<0,05). O perímetro torácico foi a característica morfométrica que teve correlação fenotípica de elevada magnitude com o peso, acima de 0,70, para as fêmeas das bases maternas Gir, Nelore e Nelogir. A pelagem não é indicativa da base materna utilizada. As vacas F1 de base materna Gir tiveram estrutura corporal menor que as fêmeas que portam genes da raça Nelore.

ABSTRACT F1 Holstein x Zebu females from different maternal bases were characterized regarding coat, depigmentation and morphometric characteristics. A total of 266 F1 female progenies from the crossbreeding of 26 Holstein bulls with females of Zebu genetic composition were used: Gir, Nellore, Guzonel, Nellogir. Frequency distribution analysis was applied for qualitative characteristics and dispersion measures and central tendency for morphometric characteristics, and means were compared by Tukey test at 5% probability. Over 60.0% of the animals had a black coat. The cows that originated from the Gir breed had a head length of 2.8cm (P<0.05) higher than the Nellore females. Ear length varied (P<0.05) according to the maternal base used. Cows with Nelore genes were 5.0cm taller (P<0.05). The thoracic perimeter was the morphometric characteristic that had a high magnitude phenotypic correlation with weight, above 0.70, for the females of the Gir, Nellore and Nellogir maternal bases. The coat is not indicative of the maternal base used. F1 Gir-based cows had a smaller body structure than females with Nellore genes.

The aim of this study was to evaluate the effects of diets containing silages of forages that are adapted to the semi-arid environment on the characteristics of the carcass and of non-carcass components of 32 mixed-breed lambs at approximately six months old, with an average live weight of 17.61 ± 2.63 kg. Lambs were assigned to four treatments (diets containing silages of Atriplex nummularia Lind, Cenchrus ciliaris, Gliricidia sepium and Manihot sp.) in a completely randomized design with eight replicates. Animals fed the diet containing old man saltbush showed higher values for the intakes of dry matter (1.080 g/day), crude protein (190 g/day) and total digestible nutrients (740 g/day), body condition score (2.81 points), fatness (2.87 points), carcass conformation (3.19 points), live weight (29.1 kg), empty bodyweight (23.41 kg), weights of the commercial cuts neck, shoulder and ribs (0.46, 0.82 and 1.12 kg, respectively), and skin weight (1.972 kg). No differences were found between the diets for carcass dressing percentage, external and internal carcass lengths, leg length and circumference, rump circumference and width, chest width and depth, full bodyweight, empty bodyweight, cooling losses, pH before and after cooling, loin, and leg. Silages of old man saltbush, buffelgrass, gliricidia and pornunça are recommended for inclusion in lamb diets. Diets with old man saltbush silage provide the best results for characteristics of the carcass and of non-carcass components besides weight gain, which can reach up to 200 g/day depending on the formulation.

ABSTRACT Chromosome-specific probes have been widely used in molecular cytogenetics, being obtained with different methods. In this study, a reproducible protocol for construction of chromosome-specific probes is proposed which associates in situ amplification (PRINS), micromanipulation and degenerate oligonucleotide-primed PCR (DOP-PCR). Human lymphocyte cultures were used to obtain metaphases from male and female individuals. The chromosomes were amplified via PRINS, and subcentromeric fragments of the X chromosome were microdissected using microneedles coupled to a phase contrast microscope. The fragments were amplified by DOP-PCR and labeled with tetramethyl-rhodamine-5-dUTP. The probes were used in fluorescent in situ hybridization (FISH) procedure to highlight these specific regions in the metaphases. The results show one fluorescent red spot in male and two in female X chromosomes and interphase nuclei.

RESUMO: O objetivo deste estudo é avaliar as flutuações da pressão coloidosmótica (PCO) em cães adultos e idosos durante a intervenção cirúrgica. Foram utilizados 36 cães hígidos submetidos à intervenção cirúrgica, distribuídos em dois grupos de acordo com a idade e submetidos ao mesmo protocolo anestésico. Os valores de albumina, proteína plasmática total e PCO foram avaliados de amostras coletadas antes da medicação pré-anestésica, 15 minutos após e ao final do procedimento cirúrgico. Os dados obtidos foram analisados através do teste-t para comparação entre os grupos e ANOVA para medidas repetidas seguido do teste de Tukey para comparar diferentes momentos dentro do mesmo grupo. Foram considerados estastisticamente diferentes com p<0.05. Em ambos os grupos foram observados decréscimo dos valores da pressão coloidosmótica, como os valores de albumina e proteína total (p<0.001). Apesar de ligeiramente inferior, os valores de PCO para o grupo adulto não apresentaram diferença significativa. Os resultados indicam que não houve diferença nos valores da pressão oncótica ou padrão de flutuação entre adultos e idosos (p=0,124). Os resultados observados indicam significativo decréscimo da pressão coloidosmótica durante os procedimentos cirúrgicos devido à hipotensão causada pelos fármacos anestésicos e pela hemodiluição causada pela administração de fluídos, mas não houve diferença entre os grupos. Entretanto, tanto em cães adultos como idosos, essas variações retornaram gradualmente após a recuperação dos animals, através do aumento da produção de urina e da recuperação do tônus vascular, indicando restabelecimento da homeostase.

ABSTRACT: The objective this study is to evaluate colloid osmotic pressure (COP) fluctuations in adult and senile dogs during surgical interventions. Thirty-six healthy dogs to surgical interventions, distributed in two groups, A and B, according to their age, and were all subjected to the same anesthetic protocol. Values of albumin, total plasmatic protein and COP were evaluated from samples collected before pre-anesthetic medication, fifteen minutes after pre-anesthetic medication, and shortly after the end of the intervention. Results were tested using t-test to compare among groups and ANOVA for repeated measures followed by Tukey’s test to compare different moments within the same group. Statistical significance was set at p<0.05. In both groups, significant decreases were observed in colloid osmotic pressure, as well as albumin and total proteins (p<0.001). Despite slightly lower COP values for the group of adult animals, this difference was not significant as there was a high individual variation within groups. The results therefore indicate no difference in colloid osmotic pressure values or fluctuation patterns among adult and senile dogs (p=0.124). The observed results indicate that colloid osmotic pressure decreases significantly during surgical procedures, due to hypotension caused by the anesthetic drugs and to hemodilution caused by the fluid administration but there is no difference between groups. However, in both adult and senile dogs, these variables recover gradually after the animals awaken, through increased urine production and recovery of vascular tonus, indicating the successful reestablishment of homeostasis.

RESUMO: O glyphosate, herbicida mais utilizado na cultura do café, inibe indiretamente a rota de biossíntese de metabólitos secundários, responsáveis por estruturas e substâncias de defesa das plantas. Objetivou-se com este estudo avaliar a morfoanatomia foliar, fenóis totais, flavonoides e cafeína em folhas de café arábica submetidas a diferentes subdoses de glyphosate. Os tratamentos consistiram da combinação de três cultivares de café (MGS Travessia, Oeiras MG 6851 e Catuaí IAC 144) e cinco subdoses de glyphosate (0,0; 57,6; 115,2; 230,4 e 460,8 g e.a. ha-1). Aos 30 dias após a pulverização, avaliou-se a espessura total da lâmina foliar, espessura das epidermes adaxial e abaxial, parênquima paliçádico e lacunoso, além das concentrações de fenóis totais, flavonoides totais e cafeína. O cultivar Catuaí teve redução na espessura foliar total, enquanto os demais cultivares tiveram incremento. As epidermes adaxial e abaxial e o parênquima paliçádico reduziram em espessura e o parênquima lacunoso aumentou com o incremento das doses de glyphosate, independentemente do cultivar. Houve aumento de fenóis totais até 115,2 g ha-1 de glyphosate; acima desta dose as concentrações de fenóis foram reduzidas. Verificou-se redução de flavonoides até 115,2 g ha-1 nos cultivares Travessia e Oeiras; acima desta dose, a concentração de flavonides aumentou. O cultivar Catuaí teve comportamento contrário da concentração de flavonoides. Oeiras e Catuaí tiveram diminuição da cafeína até 115,2 g ha-1 de glyphosate e aumento em doses superiores. Travessia mostrou aumento de cafeína a partir de 115,2 g ha-1. Os três cultivares apresentaram maior concentração de fenóis no parênquima paliçádico após a aplicação da deriva de glyphosate. O glyphosate reduz a espessura das epidermes adaxial e abaxial e do parênquima paliçádico e aumenta a espessura de parênquima lacunoso. Doses acima de 230,4 g ha-1 de glyphosate resultam em redução da concentração de fenóis totais. A concentração de flavonoides e cafeína possui efeitos variados com aplicação de glyphosate.

ABSTRACT: The most widely used herbicide in coffee crop is glyphosate, it inhibits the biosynthesis of secondary metabolites, responsible for structures and substances of plant defense. This work aimed to evaluate the morphoanatomy, total phenols, flavonoids and caffeine in leaves of Arabica coffee submitted to different sub-doses of glyphosate. The treatments were combinations of three coffee cultivars (MGS Travessia, Oeiras MG 6851 and Catuaí IAC 144) and five sub-doses of glyphosate (0.0, 57.6, 115.2, 230.4 and 460.8 g a.e. ha-1). The total thickness of the leaf blade, thickness of adaxial and abaxial epidermis, palisade and spongy parenchyma, in addition to the total phenol, total flavonoids and caffeine were evaluated 30 days after spraying. Catuaí showed a reduction in the total leaf thickness, while the others cultivars had an increase. The thickness of adaxial and abaxial epidermis and palisade parenchyma was reduced, and the spongy parenchyma increased with increasing doses of glyphosate, regardless of the cultivars. There was an increase on the total phenols up to 115.2 g ha-1 glyphosate, and above this dose, the concentrations of phenols were reduced. The Travessia and Oeiras cultivars obtained a reduction of flavonoids up to 115.2 g ha-1; and above this dose, the concentration of flavonoids increased. The Catuaí cultivar showed an opposing behavior of the concentration of flavonoids. Oeiras and Catuaí showed a decrease of caffeine up to 115.2 g ha-1 and an increase at higher doses. Travessia showed an increase of caffeine in sub-doses above 115.2 g ha-1. The three cultivars showed a higher concentration of phenols in the palisade parenchyma after the application of glyphosate drift. Glyphosate reduces the thickness of the adaxial and abaxial epidermis and palisade parenchyma, and increases the thickness of spongy parenchyma. Doses of glyphosate above 230.4 g ha-1 result in the reduction of total phenols. The concentration of flavonoids and caffeine has varying effects with glyphosate application.

Adiponectin (APN), an adipose tissue-released adipokine with demonstrated anti-inflammatory and anti-atherogenic properties, is encoded by a gene whose polymorphisms are associated with presence of coronary artery disease (CAD). Serum APN levels are inversely related with presence and complexity of CAD. Within this context, we sought to compare levels of total APN and its high molecular weight form (HMW APN) according to clinical presentation and extent of CAD in patients undergoing elective cardiac catheterization. From March 2008 to June 2010, clinical data and blood samples for APN and HMW APN measurements were collected from 415 subjects undergoing cardiac catheterization at two tertiary centers. CAD extent was estimated by the number of coronary arteries with significant stenosis (≥70% obstruction in a major coronary artery) and by Duke Jeopardy Score (DJS). Serum APN levels were similar between groups with stable or unstable CAD (APN 9.20±5.88 vs 9.47±6.23 μg/mL, P=0.738, and HMW APN 5.31±3.72 vs 5.91±4.16 μg/mL, P=0.255), even after stratification by the number of arteries involved (single-vessel vs multivessel disease: APN 9.39±5.76 vs 9.26±6.27 μg/mL, P=0.871; HMW APN 5.29±3.79 vs 5.83±4.04 μg/mL, P=0.306) and DJS score (APN, P=0.718; HMW APN, P=0.276). We conclude that APN and HMW APN serum levels are similar across clinical presentations and different extents of CAD, despite being significantly lower in the presence of obstructive CAD.

Coronary allograft vasculopathy is an inflammatory-proliferative process that compromises the long-term success of heart transplantation and has no effective treatment. A lipid nanoemulsion (LDE) can carry chemotherapeutic agents in the circulation and concentrates them in the heart graft. The aim of the study was to investigate the effects of methotrexate (MTX) associated to LDE. Rabbits fed a 0.5% cholesterol diet and submitted to heterotopic heart transplantation were treated with cyclosporine A (10 mg·kg–1·day–1 orally) and allocated to treatment with intravenous LDE-MTX (4 mg/kg, weekly, n=10) or with weekly intravenous saline solution (control group, n=10), beginning on the day of surgery. Animals were euthanized 6 weeks later. Compared to controls, grafts of LDE-MTX treated rabbits showed 20% reduction of coronary stenosis, with a four-fold increase in vessel lumen and 80% reduction of macrophage staining in grafts. Necrosis was attenuated by LDE-MTX. Native hearts of both LDE-MTX and Control groups were apparently normal. Gene expression of lipoprotein receptors was significantly greater in grafts compared to native hearts. In LDE-MTX group, gene expression of the pro-inflammatory factors tumor necrosis factor-α, monocyte chemoattractant protein-1, interleukin-18, vascular cell adhesion molecule-1, and matrix metalloproteinase-12 was strongly diminished whereas expression of anti-inflammatory interleukin-10 increased. LDE-MTX promoted improvement of the cardiac allograft vasculopathy and diminished inflammation in heart grafts.

Doença bacteriana zoonótica, a campilobacteriose é responsável mundialmente por frequentes casos de gastroenterite humana. Campylobacter spp. apresenta fator de virulência associado à diarreia, denominado toxina citoletal distensiva (CDT), sendo codificado pelos genes do complexo cdt. Os objetivos do presente estudo foram: 1) isolar e identificar estirpes de Campylobacter spp. de 102 suabes de carcaças e 102 suabes retais de ovinos (Ovis aries) e de sete amostras de água dos efluentes, antes e depois do tratamento de desinfecção de abatedouro localizado no estado de São Paulo; e 2) detectar, pela técnica de multiplex-PCR, a presença do complexo de genes cdt. Foram isoladas e identificadas, por métodos fenotípicos e genotípicos, sete estirpes de Campylobacter coli provenientes de 4/102 (3,92%) das amostras de suabes retais, 1/102 (0,98%) de suabes de carcaças e 2/7 (28,5%) das águas dos efluentes. Dos isolados de suabes retais, em 2/7 (28,6%) estirpes foi detectada a presença dos genes cdt. Trata-se do primeiro relato de isolamento de estirpes de Campylobacter coli provenientes de abatedouro de ovinos e das estirpes portadoras do complexo de genes cdt nessa espécie animal no Brasil.

A zoonosis and bacterial disease, campylobacteriosis is responsible for frequent cases of human gastroenteritis worldwide. Campylobacter spp. presents the virulence factor called cytolethal distensive toxine (CDT), responsible for diarrhea and codified by the cdt gene. The aims of this study were: 1) to isolate and identify Campylobacter spp. strains from 102 carcass swabs and 102 rectal swabs of sheep (Ovis aries) and seven samples of wastewater, before and after the disinfection treatment, collected from the abattoir of the state of São Paulo; and 2) to detect the presence of cdt gene complex by Multiplex-PCR in strains of Campylobacter spp. Seven strains of Campylobacter coli were isolated and identified by phenotypic and genotypic methods: 4/102 (3.92%) from rectal swabs, 1/102 (0.98%) from carcass swabs and 2/7 (28.5%) from wastewater. From the rectal swab samples 2/7 (28.6%) strains were detected with the cdt gene. This is the first report on the isolation of Campylobacter coli from sheep abattoir, and of strains carrying the cdt gene complex in this animal species in Brazil.

RESUMO Zanthoxylum rhoifolium Lam. (Rutaceae) tem sido utilizada tradicionalmente no tratamento de infecções microbianas e doenças parasitárias. No presente estudo, o efeito antileishmania induzido pelo extrato etanólico de cascas do caule de Z. rhoifolium (ZR-EEtOH) e sua fração hexânica (ZR-FHEX) sobre a infecção e infectividade de macrófagos por formas promastigotas de Leishmania amazonensis foram investigados. Em diferentes experimentos, macrófagos ou promastigotas foram pré-tratadas com ZR-EEtOH ou ZR-FHEX em concentrações não-letais por 24 horas, e então os macrófagos foram submetidos à infecção pelas promastigotas. Além disso, seus efeitos sobre a ativação de macrófagos, assim como concentração de DNA, tamanho e número de promastigotas por citometria de fluxo também foram avaliados. A taxa de infecção e o número de formas amastigotas internalizadas foram consideravelmente reduzidos após os pré-tratamentos de macrófagos ou de promastigotas quando comparados com células não tratadas. O aumento na capacidade fagocítica e nas concentrações de nitrito também foi observado. Por sua vez, a redução na concentração de DNA, tamanho e número de promastigotas também foram observados. Em conclusão, ZR-EEtOH e ZR-FHEX promoveram um significativo efeito antileishmania e redução na infecção de macrófagos, provavelmente envolvendo ativação de mecanismos de defesa em macrófagos. Estes achados reforçam a potencial aplicação de Z. rhoifolium no tratamento da leishmaniose.

ABSTRACT Zanthoxylum rhoifolium Lam. (Rutaceae) has been traditionally used in the treatment of microbial infections and parasitic diseases. In the present study, the antileishmanial effect induced by the ethanol extract of stem barks from Z. rhoifolium (ZR-EEtOH) and its n-hexane fraction (ZR-FHEX) on infection and infectivity of murine macrophages by promastigote forms of Leishmania amazonensis were investigated. In different set of experiments, macrophages or promastigotes were pretreated with ZR-EEtOH or ZR-FHEX at non-lethal concentrations for 24 hours, and then macrophages were submitted to infection by promastigotes. Moreover, their effects on activation of macrophages, as well as on the DNA content, size and number of promastigotes by flow cytometry were also evaluated. The infection rate and the number of internalized amastigote forms were markedly decreased after pretreatment of macrophages or promastigotes when compared with non-treated cells. The increase in phagocytic capability and nitrite content was also observed. Furthermore, the decrease of DNA content, size and number of promastigotes was also observed. In conclusion, ZR-EEtOH and ZR-FHEX promoted a markedly significant antileishmanial effect and reduction of infection of macrophages, probably underlying defense mechanisms activation in macrophages. These findings reinforce the potential application of Z. rhoifolium in the treatment of leishmaniasis.

Devido à crescente seleção de microrganismos resistentes aos antimicrobianos atuais, tem-se valorizado a busca por alternativas naturais. O presente estudo teve por objetivo avaliar a atividade antibacteriana de extratos etanólico e de ciclohexano de flores de camomila, espécie vegetal de uso antigo pela medicina tradicional, frente às bactérias ATCC Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli e Salmonella enterica subsp. enterica sorovar Typhimurium pelas técnicas de difusão em ágar e diluição em caldo. Foi observada inibição do crescimento de P. aeruginosa frente ao extrato etanólico bruto (1g/mL) na técnica de diluição em caldo, e confirmada pela técnica de difusão em ágar (halo de inibição de 10 mm de diâmetro). Para as demais bactérias testadas, os extratos e suas diluições não apresentaram efeito bacteriostático em nenhuma das técnicas. Pode-se concluir que o extrato etanólico bruto da camomila apresentou atividade antibacteriana frente à P. aeruginosa, porém não foi eficaz frente à S. aureus, E. coli e Salmonella enterica subsp. enterica sorovar Typhimurium. Portanto, são necessários novos estudos com diferentes linhagens de microrganismos, com o intuito de corroborar e assegurar os resultados apresentados, para definir o potencial antimicrobiano do extrato da camomila.

Due to the growing selection of microorganisms resistant to antimicrobial, the search for natural alternatives has become popular. The objective of the present study was to evaluate the antibacterial activity of ethanolic and cyclohexane extracts of chamomile flowers, a plant species long used by traditional medicine, against ATCC bacteria Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Salmonella enterica subsp. enterica serovar Typhimurium by agar diffusion and broth dilution techniques. Growth of P. aeruginosa was inhibited when crude ethanolic extract (1g/mL) was used broth dilution, and was confirmed by agar diffusion, (10 mm diameter inhibition zone). For the other bacteria tested, the extracts and their dilutions did not show any bacteriostatic effect in any of the techniques. It may be concluded that pure ethanolic extract of chamomile presents antibacterial action against P. aeruginosa, and none against S. aureus, E. coli e Salmonella enterica subsp. enterica sorovar Typhimurium. However, other studies with different strains of microorganisms may be useful in order to corroborate and ensure these results, to define evaluation of the antimicrobial activity of chamomile extract.

Diante do escasso conhecimento morfofisiológico do trato digestório do Tamandua tetradactyla, este trabalho pretendeu esclarecer a morfologia do estômago dessa espécie. Utilizaram-se seis espécimes, sendo três machos e três fêmeas jovens, provenientes da área de Mina Bauxita Paragominas - PA, doados, após morte por atropelamento, ao Laboratório de Pesquisa Morfológica Animal (LaPMA). Os animais foram fixados com solução aquosa de formaldeído (10%), seguido de dissecação, avaliação da topografia do estômago, com posterior descrição, mensuração e coleta de material para microscopia. As análises topográfica e macroscópica revelaram que o estômago, unicavitário, com presença de pequena e grande curvaturas, estava localizado no antímero esquerdo da cavidade abdominal, ligado cranialmente ao esôfago e caudalmente ao duodeno, e apresentou um aumento de superfície na região pilórica (toro pilórico). Histologicamente, era constituído por epitélio colunar simples (região glandular), epitélio estratificado pavimentoso (região aglandular), lâmina própria, muscular da mucosa, submucosa, muscular circular interna, muscular longitudinal externa e serosa. A região do piloro apresentou um grande espessamento da camada muscular da mucosa, e foram observadas nesta região fossetas gástricas profundas. O estômago de T. tetradactyla revelou características morfológicas macroscópicas e histológicas semelhantes às espécies domésticas e selvagens.

Given the limited morphophysiological knowledge about the digestive tract of Tamandua tetradactyla, this study aimed to clarify the morphology of the stomach of this species. We used six young specimens, three males and three females, from the area of Paragominas bauxite mine - PA, donated after death by trampling, to the Research Laboratory Animal Morphological (LaPMA). The animals were fixed with an aqueous solution of formaldehyde (10%), followed by dissection, evaluation of the topography of the stomach, with further description, measurement and collection of material for microscopy. The topographical and macroscopic analysis showed that the stomach was located in left on-timer of the abdominal cavity, connected entirely to the esophagus and caudally to the duodenum, was unicavitary with the presence of small and large curvature and showed a surface increase in the pyloric region (torus pylorus). Histologically it was composed of simple columnar epithelium (glandular region), stratified epithelium (aglandular region), lamina propria, muscular of mucous, submucous, muscular internal circular, muscular external longitudinal and serosa. The region of the pylorus presented a great thickening of the muscular layer of mucous, and deep gastric pits were observed in this region. The stomach of T. tetradactyla revealed macroscopic and histological morphological characteristics similar to domestic and wild species.
