Steinernema diaprepesi Nguyen & Duncan (Rhabditida: Steinernematidae), a new entomopathogenic nematode from Brazil

Abstract Entomopathogenic nematodes (EPNs) can control pests due to their mutual association with bacteria. The use of these biological control agents is increasing worldwide due to advances in research about its control efficiency, range of action and mass production. The identification of EPNs adapted to specific environmental and climatic conditions is important for sustainable pest suppression in integrated management (IPM) programs. The objective is to report, for the first time, the occurrence of the Steinernema diaprepesi in Brazil. Steel mesh traps with Galleria mellonella Linnaeus (Lepidoptera: Pyralidae) larvae were buried in red latosol cultivated with Eucalyptus. Infective juveniles (IJs) were isolated from dead larvae and multiplied in healthy ones of this host to confirm its pathogenicity and to start a laboratory population from the strain found in the field. The DNA of the IJs was extracted and amplified using PCR technique with the universal primers D2A and D3B. The detection of S. diaprepesi is the first report of this nematode in Brazil, increasing the knowledge about its distribution in the world and the diversity of EPNs that must be considered as agents of biological pest control in the country.

RESUMO O objetivo deste estudo foi avaliar os diferentes estágios de lactação de vacas F1 Holandês x Zebu quanto ao consumo e à digestibilidade de nutrientes, à eficiência no uso de nitrogênio, ao comportamento ingestivo e ao desempenho. Trinta e seis vacas F1 Holandês × Zebu, com peso corporal inicial (PC) de 482±43kg, foram utilizadas. Os estágios inicial, médio e final da lactação foram caracterizados após 50±13, 111,5±11,75 e 183,0±17,5 dias de lactação, respectivamente. O arranjo experimental adotado foi o delineamento inteiramente ao acaso, com três fases de lactação e 12 vacas em cada grupo de tratamento. O consumo de matéria seca (P=0,01) foi maior no período final da lactação. Na fase inicial da lactação, a produção de leite (P<0,01) foi maior em 24,17% em comparação às demais fases. Na fase intermediária da lactação, as vacas apresentaram menor peso corporal (465,63kg; P<0,01) em relação às demais fases. A eficiência alimentar foi maior em 23,36% na fase inicial da lactação (0,82kg de leite/kg de MS). Vacas F1 Holandês x Zebu aumentam o consumo de matéria seca no período final da lactação. A produção de leite e a eficiência alimentar no início da lactação foram favorecidas por mudanças no comportamento ingestivo, como o aumento do tempo de ruminação.
ABSTRACT The objective of this study was to evaluate the different lactation stages of F1 Holstein x Zebu cows on intake and digestibility of nutrients, nitrogen use efficiency, feeding behavior and performance. Thirty-six F1 Holstein × Zebu cows with initial body weight (BW) of 482±43kg were used. The early, mid and late lactation stages were characterized after 50±13, 111.5±11.75 and 183.0±17.5 days in milk, respectively. A completely randomized design with three lactation stages and 12 cows in each treatment group was used. Dry matter intake (P=0.01) was higher in late lactation. Milk yield (P<0.01) was 24.17% higher in early lactation than in other stages. Body weight was lowest in mid-lactation cows (465.63kg; P<0.01). The feed efficiency was 23.36% higher in early lactation than in other stages (0.82kg of milk/kg of DM). F1 Holstein x Zebu cows have increased dry matter intake in late lactation. Milk yield and feed efficiency in early lactation were benefited by changes in feeding behavior, such as increased rumination time.

Carbohydrate antigen 125 (CA125) has long been used as an ovarian cancer biomarker. However, because it is not specific for ovarian cells, CA125 could also be used to monitor congestion and inflammation in heart disease. Acute heart failure (HF) is used to identify patients with a worse prognosis in ST-segment elevation myocardial infarction (STEMI). We aimed to determine the association of CA125 with acute HF in STEMI and to compare CA125 with N-terminal pro brain natriuretic peptide (NTproBNP) with a cross-sectional study. At admission, patients were examined to define Killip class and then underwent coronary angioplasty. Blood samples, preferably taken in the hemodynamic ward, were centrifuged (1500 g for 15 min at ambient temperature) and stored at −80°C until biomarker assays were performed. Patients were divided into two groups according to the presence or absence of congestion. Patients in Killip class ≥II were in the congestion group and those with Killip <II in the absence of congestion group. We evaluated 231 patients. The mean age was 63.3 years. HF at admission was identified in 17.7% of patients. CA125 and NTproBNP levels were higher in patients with Killip class ≥II than those with Killip class <II (8.03 vs 9.17, P=0.016 and 772.45 vs 1925, P=0.007, respectively). The area under the receiver operator characteristic curve was 0.60 (95%CI 0.53−0.66, P=0.024) for CA125 and 0.63 (95%CI 0.56−0.69, P=0.001) for NTproBNP. There was no statistical difference between the curves (P=0.69). CA125 has similar use to NTproBNP in identifying acute HF in patients presenting with STEMI.

RESUMO: O objetivo desse trabalho foi determinar a ocorrência de anticorpos IgG para Neospora caninum bem como avaliar os fatores de risco associados à infecção em rebanhos ovinos do estado de Sergipe, Brasil. Foram coletadas, nos anos de 2011 e 2012, 1200 amostras de sangue de ovinos oriundos de sessenta propriedades distribuídas em três mesorregiões do estado para pesquisa de anticorpos para N. caninum pela Reação de Imunofluorescência Indireta (RIFI) utilizando-se como ponto de corte de 50 e as amostras diluídas na base 2. Os dados de 34 variáveis estudadas foram obtidos a partir de questionários aplicados aos proprietários e analisados para se determinar a frequências absolutas e relativas e análise dos fatores de risco pelo teste Qui-quadrado de Pearson (p≤0,05). A ocorrência de ovinos soropositivos para N. caninum foi de 39,83% (478/1200). Em relação às mesorregiões a ocorrência de animais e propriedades positivas foi de, respectivamente, 55,88% (380/680) e 88,24% (30/34) na Leste; 21,42% (60/280) e 42,85% (6/14) no Agreste e 15,83% (38/240) e 41,67% (5/12) no Sertão. Os títulos de anticorpos variaram de 50, representando 96,02% (459/478) das amostras soropositivas, a 6400 (1/478). Dentre as variáveis significantes, na analise multivariada, que foram consideradas com fatores de risco para a infecção pelo N. caninum estavam propriedades localizadas na mesorregião Leste (p=0,000, OR=4,64, IC95%=3,36-6,41), presença de fonte de água parada e corrente (p=0,000 OR=2,03, IC95%=1,41-2,92), ausência de quarentena (p=0,000 OR=2,71, IC95%=2,08-3,53), não utilização de esterqueiras (p=0,000 OR=3,14, IC95%=2,45-4,02), criações com finalidade de subsistência (p=0,000 OR=4,99, IC95%=3,15-7,92), de reprodução (p=0,002, OR=1,74, IC95%=1,22-2,49), presença de cães (p=0,000 OR=2,74, IC95%=1,73-433) e circulação de animais silvestres nos rebanhos (p=0,000 OR=3,45, IC95%=2,44-4,87). Os resultados evidenciam a ocorrência de N. caninum em rebanhos ovinos sergipanos, demonstrando o manejo e a localização dos rebanhos no estado como importantes fatores de risco.
ABSTRACT: The aim of this study was to determine the occurrence of antibodies IgG against Neospora caninumand evaluate the risk factors associated with the infection in ovine herds, State of Sergipe, Brazil. Blood samples (n=1200) were collected from sheep raised in 60 sheep run located in the three mesoregions of the State of Sergipe, between 2011 e 2012. Antibodies were investigated by Indirect Immunofluorescence Antibody Test of which cutoff point was 50 and positive samples were diluted in base 2 until the last positive titer. Data from 15 variables was obtained from questionnaires given to farmers. Absolute and relative frequencies were determined and the risk factors were analyzed by Pearson’s Qui-Square test (p≤0,05). The occurrences of serum reactive sheep were 39.83% (478/1200). The occurrences of positive sheep and sheep run were 55.88% (380/680) and 88.24% (30/34) in the Eastern region; 21.42% (60/280) e 42.85% (6/14) in dry region and 15.83% (38/240) e 41.67% (5/12) in the backwoods respectively. Antibody titers ranged from 50 (n=459), represented 96.02% (459/478) of seropositive samples to 6400 (1/478). Among the significant variables in the multivariate analysis were considered risk factors for infection with N. caninum were, sheep run located in Eastern region (p=0.000, OR=4.64, CI95%=3.36-6.41); standing and running water sources (p=0.000 OR=2.03, CI95%=1.41-2.92), absence of quarantine (p=0.000, OR=2.71, CI95%=2.08-3.53), absence of dunghill (p=0.000, OR=3.14, CI95%=2.45-4.02), presence of dogs (p=0.000, OR=2.74, CI95%=1.73-433), presence of wild animals (p=0.000, OR=3.45, CI95%=2.44-4.87) and subsistence (p=0.000, OR=4.99, CI95%=3.15-7.92) or reproduction (p=0.002, OR=1.74, CI95%=1.22-2.49) livestock were important risk factors. Our results highlight the occurrence ofN. caninumin the ovine herds from State of Sergipe. Management and location of sheep runs were important risk factors associated to the infection.

RESUMO Objetivou-se com o presente experimento avaliar o efeito de dois diferentes diluidores (leite em pó desnatado glicose - SMG ou lactose gema de ovo - LEY) sobre as características físicas e a fertilidade do sêmen asinino coletado de forma fracionada e resfriado a 5ºC. Para isso, quatro jumentos da raça Pêga foram utilizados como doadores de sêmen. A fração espermática rica do ejaculado foi diluída preparando-se doses inseminantes contendo 400 x 106 espermatozoides móveis em um volume de 22 mL, resfriadas a 5ºC e armazenadas por até 48 horas em contêiner proposto por Palhares (1997). A motilidade e o vigor espermáticos foram avaliados no sêmen fresco, após a pré-diluição, antes das inseminações, às 24 e 48 horas de armazenamento. Para avaliação de fertilidade, 44 éguas foram inseminadas com sêmen armazenado por um período entre 12 e 24 horas, em dias fixos (segundas, quartas e sextas-feiras), após a detecção de um folículo de diâmetro maior ou igual a 30mm em um dos ovários, até a ovulação. O diagnóstico de gestação foi realizado a partir de 12 dias após a ovulação, por meio de ultrassonografia transretal. O sêmen diluído em SMG apresentou motilidade espermática superior à do LEY, já a partir do tempo pré-IA. Às 48 horas de armazenamento, todos os jumentos apresentaram valores de motilidade entre 45% e 53%, quando o sêmen foi diluído em SMG, enquanto apenas um jumento apresentou motilidade superior a 30% no tratamento utilizando LEY. A taxa de concepção/ciclo das éguas inseminadas também foi superior para o sêmen diluído em SMG em relação ao diluído em LEY (56,52% versus 4,76%, respectivamente).
ABSTRACT The aim of this study was to evaluate the effect of two different extenders (Skimmed Milk Glucose - SMG or Lactose - Egg Yolk - LEY) on physical characteristics and fertility of fractionated donkey semen cooled at 5°C. For this, four Pêga donkeys were used as semen donors. The sperm rich fraction of the ejaculate was diluted preparing insemination doses containing 400 x 106 motile spermatozoa in a volume of 22 mL, cooled to 5°C and stored up to 48 hours in a container proposed by Palhares (1997). Sperm motility and vigor were assessed in fresh semen, after first semen dilution, before insemination, at 24 and 48 hours after storage. For the fertility evaluation, 44 mares were inseminated with semen stored for a period between 12 and 24 hours. The mares were inseminated on fixed days (Mondays, Wednesdays, and Fridays) after the detection of a follicle greater than a 30mm diameter in one of the ovaries through ovulation. Pregnancy diagnosis was performed on day 12 post-ovulation, using transrectal ultrasonography. Semen diluted in SMG showed superior sperm motility than LEY, at the Pre-AI evaluation (P<0.05). At 48 hours of storage, all donkeys had motility values between 45 and 53% for semen diluted in SMG, while only one donkey showed motility greater than 30% in the LEY treatment. The pregnancy rate/cycle for mares inseminated with semen diluted in SMG was superior than that obtained using LEY (56.52% vs 4.76%, respectively).

O objetivo deste estudo foi descrever o uso da elastografia ARFI (acoustic radiation force impulse) para avaliar as afecções testiculares em cães. Dezoito cães com distúrbios testiculares (36 testículos) foram avaliados. Ecotextura, tamanho, contornos e margens dos testículos foram avaliados por meio da ultrassonografia modo-B. A presença de deformidades e a rigidez tecidual (escala de cinza; homogênea ou heterogênea) foram avaliadas pela elastografia qualitativa; e a velocidade de cisalhamento foi determinada pela avaliação quantitativa. Amostras dos tecidos testiculares foram coletadas após orquiectomia para o diagnóstico histopatológico. Após ultrassonografia, orquiectomia e histopatologia, foram identificados 36 distúrbios em tecidos testiculares. Durante a elastografia qualitativa, os tecidos normais apresentaram-se homogêneos e não deformáveis; os testículos alterados demonstraram alterações na rigidez tecidual e de sua homogeneidade. Para a elastografia quantitativa, os valores obtidos foram: tecidos normais - 1,30±0,12m/s; degenerados - 0,97±0,08m/s; atrofiados - 2,00±0,35m/s; hipoplásicos - 0,82±0,2m/s; cistos - 1,32±0,18m/s; orquite - 2,68±0,42m/s; tumores de células intersticiais - 3,32±0,65m/s; sertolioma - 2,99±0,07m/s; e leydigoma - 2,73±0,37m/s. A elastografia ARFI de testículos anormais em cães demonstrou ser uma técnica aplicável e complementar para o diagnóstico de doenças testiculares nessa espécie animal.
The aim of this study was to describe the use of acoustic radiation force impulse (ARFI) elastography in the evaluation of testicular disorders in dogs. Eighteen dogs with testicular disorders (thirty-six testicles) were assessed. Echotexture, size, contours and margins of testes were analysed by ultrasonography. Deformities and tissue stiffness (greyscale and homogenous or heterogeneous) were evaluated by qualitative elastography and shear velocity was determined quantitatively. Subsequent to orchiectomy, testicular samples were collected for histopathology analysis and thirty-six disorders were identified. Qualitative elastography revealed that normal healthy testicular tissues were homogenous and not pliable while the affected testicles had alterations in tissue stiffness and homogeneity. The values obtained for quantitative elastography of the testicular tissues were: normal/healthy - 1.30±0.12 m/s; degenerated - 0.97±0.08 m/s; atrophied - 2.00±0.35 m/s; hypoplastic - 0.82±0.2 m/s; cystic - 1.32±0.18 m/s; orchitis - 2.68±0.42 m/s; interstitial cell tumours - 3.32±0.65 m/s; sertolioma - 2.99±0.07 m/s and leydigoma - 2.73±0.37. ARFI elastography of abnormal testes proved to be an applicable and complementary technique in the diagnosis of testicular disease in dogs.

RESUMO:Objetivou-se com este trabalho avaliar a produção de massa e o teor de flavonoides de plantas de marcela cultivadas com fósforo e cama de frango e colhidas em duas épocas. Estudou-se a combinação de doses de P2O5 (0, 100, 200 e 300 kg ha-1) na forma de superfosfato triplo, com e sem cama de frango (10 t ha-1), arranjados como fatorial 4x2, no delineamento blocos casualizados, com quatro repetições. Foram feitas duas colheitas das plantas, consideradas como parcelas subdivididas no tempo. A propagação foi indireta, com semeio em bandejas e transplante ao campo. As plantas foram colhidas aos 195 e 223 dias após o transplantio - DAT. A altura máxima das plantas foi de 1,14 m com uso da cama de frango e de 0,97 m sem cama, aos 195 dias após o transplantio. A produção máxima de massa fresca de parte aérea das plantas foi de 30,31 t ha-1 na colheita aos 223 DAT, enquanto a de massa seca foi de 11,38 t ha-1, ambas com o uso de 300 kg ha-1 de P2O5.A massa fresca de inflorescências foi maior (4,08 t ha-1) com adição de cama de frango ao solo e menor (3,49 t ha-1) sem cama de frango; em resposta às doses de fósforo, cresceram linearmente, sendo a máxima de 4,65 t ha-1 com uso de 300 kg ha-1 de P2O5, independente da época de colheita. A massa seca de inflorescências foi máxima de 2,38 t ha-1 com o uso de 300 kg ha-1 P2O5 na colheita aos 223 DAT. A maior produção de massa seca e o maior teor de flavonoides das inflorescências foram obtidos com o cultivo das plantas de marcela em solo com cama de frango (10 t ha-1) e com P2O5 (300 kg ha-1) na colheita aos 223 dias após o transplantio.
ABSTRACT:The aim of this experiment was to evaluate the biomass yield and flavonoid contents of Achyrocline satureioides grown under different levels of phosphate fertilizer in association with broiler litter, during two successive harvests. Four levels of P2O5 (0, 100, 200 and 300 kg ha-1), as triple superphosphate, and two levels of broiler litter (0 and 10 t ha-1) were arranged as a 4x2 factorial design in randomized blocks with four replications. Two harvests of plants, considered as split-plot, were made. Seeds were sown in polystyrene trays then transplanted to the experimental field and harvested at 195 and 223 days after transplanting (DAT). The maximum heights of the plants were 1.14 m under the addition of broiler litter and 0.97 m without it, at 195 days after transplanting. The highest yields of total fresh weight (30.31 ton ha-1) and dry weight (11.38 ton ha-1) were obtained at 223 DAT, both with the concentration of 300 kg ha-1 P2O5. The fresh weight of the inflorescences was greater under the addition of broiler litter (4.08 ton ha-1) than without it (3.49 ton ha-1). The plants grew linearly in response to the use of phosphate, and the maximum yield was 4.65 ton ha-1 under the concentration of 300 kg ha-1P2O5, regardless of the harvest season. The dry weight of the inflorescences reached maximum yield (2.38 ton ha-1) at 223 DAT, in response to the use of 300 kg ha-1P2O5. The highest yield of dry weight and the highest contents of flavonoids from the inflorescences were obtained when A. satureioides was grown under the concentration of P2O5 (300 kg ha-1) with the addition of broiler litter (10 ton ha-1) and harvested at 223 DAT.

Os objetivos deste trabalho foram estimar componentes de variância genética aditiva, fenotípica e residual e a herdabilidade para características relacionadas com a produção de mel e com a estrutura do ninho de abelhas Melipona quadrifasciata anthidioides. Sessenta colônias de diferentes regiões da Bahia foram transladadas para caixas padronizadas modelo INPA e divididas, originando as gerações parentais G1 e G2. Foram medidas as características: estimativa da produção de mel; número, largura, volume e altura dos potes de mel; número, altura e diâmetro dos potes de pólen; peso; número, largura e diâmetro dos discos de cria e estimativa da população da colônia. As medidas foram corrigidas para o efeito fixo de mês de mensuração. Os componentes de variância e herdabilidade foram estimados por meio do método de semelhança entre parentes, utilizando-se abordagem Bayesiana. As médias e os desvios padrão variaram de 2,01±0,70cm para diâmetro de potes de pólen a 2.333,0±384,1kg para o peso das caixas. Houve indicação de convergência para todas as cadeias obtidas. As estimativas de variância genética aditiva variaram de 0,02cm para as características largura dos potes de mel a 38.587,72kg para o peso. Para as estimativas de variâncias fenotípicas, os valores variaram de 0,05 para a altura dos potes de pólen a 95.136,43kg para o peso; e para as variâncias residuais, os valores encontrados variaram de 0,02 para a variável largura dos potes de mel a 56.548,71kg para o peso. As estimativas de herdabilidade variaram de 0,35 a 0,53. Os resultados demonstraram que as características avaliadas possuem variação genética aditiva que garante boa resposta à seleção.
The aim of this study was to estimate components of genetic variance, phenotypic and residual and heritability for traits related to the production of honey and the nest structure of bee Melipona quadrifasciata anthidioides. Sixty colonies from different regions of Bahia were transferred to standard INPA model boxes and divided, creating the parental generations G1 and G2. The following characteristics were measured: estimated production of honey, number, width, height and volume of the honey pots, number, height and diameter of pollen storage pots, weight, number, length and diameter of the brood combs and estimate the population of the colony. The measurements were corrected for the fixed effects of month of measurement. The variance components and heritability were estimated by the method of similarity between relatives using the Bayesian approach. The mean and standard deviations ranged from 2.01±0.70cm diameter pots for pollen to 2333.0±384.1kg to the weight of the boxes. There was indication of convergence for all the chains obtained. Estimates of the additive genetic variance ranged from 0.02cm to the width characteristics of honey pots to 38587.72kg for weight. For the estimates of phenotypic variance the values ranged from 0.05 for the height of the pollen pots to 95136.43kg for weight; and the residual variance the values varied from 0.02 for the variable width of the honey pots to 56548.71kg for weight. The results showed that the characteristics assessed have additive genetic variation that ensures good response to selection.

In vitro biological screening of the anticholinesterase and antiproliferative activities of medicinal plants belonging to Annonaceae

The aim of this research was to investigate the antiproliferative and anticholinesterase activities of 11 extracts from 5 Annonaceae species in vitro. Antiproliferative activity was assessed using 10 human cancer cell lines. Thin-layer chromatography and a microplate assay were used to screen the extracts for acetylcholinesterase (AchE) inhibitors using Ellman's reagent. The chemical compositions of the active extracts were investigated using high performance liquid chromatography. Eleven extracts obtained from five Annonaceae plant species were active and were particularly effective against the UA251, NCI-470 lung, HT-29, NCI/ADR, and K-562 cell lines with growth inhibition (GI50) values of 0.04-0.06, 0.02-0.50, 0.01-0.12, 0.10-0.27, and 0.02-0.04 µg/mL, respectively. In addition, the Annona crassiflora and A. coriacea seed extracts were the most active among the tested extracts and the most effective against the tumor cell lines, with GI50 values below 8.90 µg/mL. The A. cacans extract displayed the lowest activity. Based on the microplate assay, the percent AchE inhibition of the extracts ranged from 12 to 52%, and the A. coriacea seed extract resulted in the greatest inhibition (52%). Caffeic acid, sinapic acid, and rutin were present at higher concentrations in the A. crassiflora seed samples. The A. coriacea seeds contained ferulic and sinapic acid. Overall, the results indicated that A. crassiflora and A. coriacea extracts have antiproliferative and anticholinesterase properties, which opens up new possibilities for alternative pharmacotherapy drugs.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Avaliação de alimentadores para abelha mandaçaia (Melipona quadrifasciata anthidioides)

Three devices for feeding mandaçaia bee, for easy handling and efficiency use: Eiratama, Pernambucano and Prato, were evaluated. Five devices for treatment and 15 colonies total were used, testing the collect capacity and time spent to total consumption. The Prato device showed mean consumption of 13.55 mL, being higher than the others. For feeding mandaçaia bee, the Prato type was the most efficient.
Este trabalho teve por objetivo avaliar diferentes dispositivos para alimentação de colônias de abelha mandaçaia. Foram avaliados três alimentadores, Eiratama, Pernambucano e Prato, para a alimentação energética. Foram utilizados 5 dispositivos por tratamento, totalizando 15 colônias. Foram avaliados à capacidade de coleta e tempo até o consumo total do alimento fornecido. O Prato apresentou média para consumo de alimento de 13,55 mL, sendo superior aos demais. Concluiu-se que, para a alimentação da abelha mandaçaia o Prato foi o mais eficiente.

The influence of fish culture in floating net cages on microbial indicators of water quality

Este trabalho foi realizado para analisar os parâmetros microbiológicos da qualidade da água de um reservatório destinado a irrigação e à cultivo de tilapia do Nilo (Oreochromis niloticus) em tanques redes. Foi realizada análise de parâmetros físico-químicos, quantificação de bactérias totais aeróbias mesófilas, coliformes totais e termotolerantes, estreptococos fecais e a detecção de E. coli em amostras de água coletada em três pontos distintos do açude (pré-cultivo, cultivo, pós-cultivo). Os níveis de amônia (0,047-0,059 mg/L), nitrito (0,001-0,021 mg/L) e fósforo total (0,050-0,355 mg/L) na água não apresentaram diferenças significativas (p > 0.05) entre os pontos de coleta. As contagens de bactérias aeróbias mesófilas variaram entre 1,3 x 104 e 67,3 x 104 UFC/100 mL nas amostras de água. O NMP de coliformes termotolerantes (< 930 NMP/100 mL) se apresentou dentro do limite permitido para águas doces destinadas a piscicultura e/ou irrigação. A presença de E. coli foi constatada em 48% e de estreptococos fecais em 56% das amostras de água analisadas. O local de cultivo de tilápia em tanques-rede apresentou maior número de amostras contaminadas com E. coli e estreptococos fecais que outros pontos de amostragem.
This work was carried out to analyse the microbiological parameters of the water quality of a reservoir used for the irrigation and culture of tilapia (Oreochromis niloticus) in floating net cages. The physico-chemical parameters, counts of mesophilic total aerobic bacteria, total and thermotolerant coliforms and fecal streptococci, and the presence of Escherichia coli in samples of water collected in three sites of the reservoir (pre-culture site, culture site, post-culture site) were analysed. The levels of ammonia (0.047-0.059 mg/L), nitrite (0.001-0.021 mg/L) and total phosphorus (0.050-0.355 mg/L) in the water did not show significant differences (p > 0.05) between sampling sites. The levels of total bacteria in the water varied between 1.3 x 104 and 67.3 x 104 CFU/100 mL. The MPN values of thermotolerant coliforms (< 930 MPN/100 mL) were within values recommended for water used for fish culture and/or irrigation. The presence of E. coli and fecal streptococci were verified in 48% and 56% of analysed samples, respectively. The site with floating net cages showed more samples contaminated with E. coli and fecal streptococci than other sampling points.

This study aimed to analyze biometric parameters and productive stingless bee colonies mandaçaia (Melipona quadrifasciata anthidioides) in consecutive generations. Sixty colonies from different regions of Bahia state were transferred to INPA boxes model. After 90 to 100 days the colonies were evaluated and divided, forming the first and later the second generation. In every generation the colony biometric measurement were evaluated (brood, honey and pollen pots), queen and glossa workers morphometry. The weight of physogastric queen and length of the glossa did not differ between generations. There were differences for the number of brood disks and between honey and pollen characteristics, with variability between generations. Thus biometric and productive parameters studied were directly influenced by environmental factors. However in subsequent generations the colonies showed environment adaptation demonstrating variability that can be exploited.
O presente trabalho teve por objetivo analisar os parâmetros biométricos e produtivos em colônias de abelha mandaçaia (Melipona quadrifasciata anthidioides) em gerações consecutivas. Sessenta colônias de diferentes regiões do estado da Bahia foram transladadas para caixas modelo INPA. Passados 90 a 100 dias as colônias foram avaliadas e divididas, formando a primeira e posteriormente a segunda geração. Em todas as gerações foram realizadas mensuração dos parâmetros biométricos da colônia (discos de cria, potes de mel e potes de pólen) e de rainha, além de morfometria da glossa de operárias. O peso da rainha fisogástrica e o comprimento da glossa não apresentaram diferenças entre as gerações. Houve diferença para o número de discos de cria e entre características de produção de mel e pólen, com variabilidade entre as gerações. Os parâmetros biométricos e produtivos estudados foram influenciados diretamente por fatores ambientais. As colônias nas gerações subsequentes mostraram adaptação ao ambiente demonstrando variabilidade que pode ser explorada.

The emergence of KPC-2 producing K. pneumoniae in hospitalized patients at the intensive care unit (ICU) of a teaching hospital located in the city of João Pessoa, Paraíba, Brazil, is reported. Seven carbapenem-resistant K. pneumoniae recovered from different body sites of infection were analyzed. Most isolates showed a multidrug-resistance phenotype. Genotypic analysis demonstrated the presence of two genotypes, with the predominance of genotype A, which belongs to ST 437. These isolates also carry the encoding genes of five other betalactamases.

In this study, genotyping techniques including staphylococcal chromosomal cassette mec (SCCmec) typing, pulsed-field gel electrophoresis (PFGE), multilocus sequence typing (MLST) and restriction-modification tests were used to compare the molecular characteristics of methicillin-resistant Staphylococcus aureus (MRSA) isolates recovered at two times within a 10-year interval (1998 and 2008) from a tertiary Brazilian hospital. In addition, the antimicrobial susceptibility profiles were analyzed. All 48 MRSA isolates from 1998 and 85.7% from 2008 (48/56 isolates) displayed multidrug-resistance phenotypes and SCCmec III. All but one of the 13 representative SCCmec III isolates belonged to CC8 and had PFGE patterns similar to that of the BMB9393 strain (Brazilian epidemic clone of MRSA; BEC). In 2008, we found an increased susceptibility to rifampicin and chloramphenicol among the SCCmec III isolates. In addition, we detected the entrance of diverse international MRSA lineages susceptible to trimethoprim-sulfamethoxazole (SXT), almost all belonging to CC5. These non-SCCmec III isolates were related to the USA 300 (ST8-SCCmec IV; PFGE-type B), USA 800 (ST5-SCCmec IV; subtype D1), USA 100 (ST5-SCCmec II; subtype D2), and EMRSA-3/Cordobes (ST5-SCCmec I, type C) clones. To the best of our knowledge, this is the first report of the emergence of isolates genetically related to the EMRSA-3/Cordobes clone in southeast Brazil. In this regard, these isolates were the most common non-SCCmec III MRSA in our institution, accounting for 8.9% of all isolates recovered in 2008. Thus, despite the supremacy of BEC isolates in our country, significant changes may occur in local MRSA epidemiology, with possible consequences for the rationality of MRSA empiric therapy.
