Effects of protected sodium butyrate (PSB) in broiler diets with reduced metabolizable energy (ME) content on production performance, intestinal histomorphometry and nutrient metabolizability were evaluated in two experiments. In one experiment, 384 one-day-old male chicks with an average weight of 44 ± 0.2 g were distributed into four treatment groups in a completely randomized design with six replications of 16 birds. The treatments consisted of i) a basal diet without reduced ME or PSB (BD); ii) BD with a 100 kcal/kg reduction in ME, without PSB; iii) BD with a 100 kcal/kg reduction in ME + 105 g/t PSB; and iv) BD with a 100 kcal/kg reduction in ME + 225 g/t PSB. Performance and carcass yield were evaluated. In the second experiment, 280 one-day-old male chicks were also distributed into four treatment groups in a completely randomized design with seven replications of 10 birds to evaluate metabolizability coefficients and intestinal development. The birds that received diet 3 had the best feed conversion ratio (FCR), final weight (FW) and weight gain (WG). Inclusion of PSB (105 and 225 g/t) in reduced-ME diets increased the metabolizability of crude protein coefficient at 35 days old. Birds that received diet 3 also had greater jejunal villus height at 21 and 35 days old. It is recommended to feed 105 g/t of PSB with a 100 kcal/kg reduction in ME until broilers were 35 days old.

Abstract The onset of gestation is characterized by growth, morphological and functional changes of the placenta. We aim to evaluate the placental compartments in New Zealand rabbits by means of stereological methods. The fetal and maternal portion of placenta (12, 14, 18 and 20 gestational days) was randomly sampled for the stereological analysis. Histological sections were scanned to estimate fetal (labyrinth and junctional) and maternal (decidua) compartment volumes. The total volume of the placenta for the ages of 12, 14, 18 and 20 days was, respectively, 320 mm3, 340 mm3, 940 mm3 and 1300 mm3. The volume of the labyrinth was 56 mm3, 119 mm3, 231 mm3 and 481 mm3, respectively. The volume of junctional zone was 75 mm3, 76 cm3, 238 mm3 and 314 mm3, respectively. The volume of decidua was 174 mm3, 143 mm3, 469 mm3 and 504 mm3, respectively. We concluded that the rabbit´s placenta compartments varied according to the gestational period, increasing continuously over the 20 gestational days. However, on the onset of the development of the placenta the decidua presented faster growth, whereas after the 20 days of development, the labyrinth developed more quickly. This study represents an aid to the understanding of placentation in humans.

Abstract The present study consisted in optimizing the extractive method of polyphenols and total tannins of leaves of Syzygium cumini (L) Skeels assisted by microwaves to potentiate the antimicrobial activity of the dried extract of S. cumini against sensitive and resistant strains. A Box-Behnken design that consisted of 27 experimental runs coupled with a desirability function for multiple response optimization was employed to optimize the total polyphenols content and total tannins content. Antimicrobial sensitivity tests were evaluated by obtaining the minimum inhibitory concentration, minimum fungicidal concentration and minimum bactericidal concentration in 96-well petri dishes. The optimal extraction conditions were found to be 8 min of extraction, under 300 w of microwave power, using a 1:34 g/mL solid/solvent ratio and 38% of ethanol concentration as extraction solvent. The parameter with the greatest influence in the extraction was primarily the time, followed by the potency and proportion solid/solvent. This yielded a total polyphenol content of 87.37 ± 1.85 mg TAE g-1ext and a total tannin content of 79.68 ± 1.64 mg TAE g-1 ext. All tested microorganisms were sensitive to the extract, evidencing the effectiveness of the extraction method optimization.

Two experiments were conducted to evaluate the effect of including protected sodium butyrate (PSB) in the diets of laying hens of advanced age on productive performance and egg quality. In experiment 1, a total of 320 Dekalb White 61-week-old laying hens were distributed in a randomized block design to four treatments (0, 105, 210, and 300 g/ton of PSB) with eight replicates of 10 birds. The experiment was conducted over 112 days, divided into four periods of 28 days each. Productive performance and egg quality were evaluated. In experiment 2, a total of 58 000 Dekalb White 70-week-old laying hens were assigned to diets containing either 0 or 105 g/ton of PSB with two replicates of 14 500 birds. The experiment was conducted on a commercial poultry farm. Egg production and the percentages of broken eggs and dirty eggs were evaluated. The PSB levels did not influence the productive performance of laying hens. A quadratic effect was observed for eggshell thickness, percentage of eggshell and eggshell strength, reaching maxima at 193, 136 and 198 g/ton of PSB, respectively. A quadratic effect on yolk index reached a minimum at 181 g/ton of PSB. The percentages of dirty eggs and broken eggs were lower in laying hens fed 105 g/ton of PSB. In conclusion, the addition of PSB to the diets of old laying hens improved the eggshell quality and decreased the percentage of broken and dirty eggs.

The addition of protease to broiler diets may complement the action of endogenous enzymes and improve protein digestibility. Here, the authors evaluated the effect of adding protease to broiler diets that contained animal-based meal on bird performance, digestibility, and biometry of digestive tract organs. Four treatments, which contained animal- or plant-based meals with or without supplementary protease were compared, namely basal vegetable feed (BVF), BVF + protease (BFP), basal vegetable feed + animal byproduct meal (BFA), and BFA + protease. In the first experiment, 320 one-day-old Cobb 500® chicks were allocated to eight replicates with 10 birds per replicate. The experimental period was seven days, and nutrient metabolizability was evaluated. In the second experiment, 720 one-day-old Cobb 500® chicks were assigned to treatments in a similar manner, with six replicates and 30 birds per replicate, but the experimental period was 42 days. Significant differences (P <0.05) were observed between treatments for nutrient digestibility, weight gain, feed consumption, average final weight, food conversion, viability, and biometry of the pancreas. Treatments with animal-based meals had the highest digestibility. Birds fed these meals grew faster in the pre-starter phase and consumed less feed between 1 and 21 days. However, between 1 and 42 days old, broilers fed plant-based diets had better feed conversion, and the group that did not receive protease supplementation had a better liveability rate.

RESUMO: A capivara (Hydrochoerus hydrochaeris) é o maior roedor do mundo. No estado do Acre, Brasil, as populações de capivaras têm aumentado significativamente. O papel das capivaras na transmissão de certas infecções zoonóticas bacterianas não é bem compreendido, incluindo as bactérias do gênero Salmonella. Salmonella spp. geralmente causam enterite ou septicemia em mamíferos, porém muitas espécies de mamíferos podem carregar a bactéria de forma assintomática e eliminá-la em suas fezes. Para entender melhor o possível papel das capivaras como reservatórios de Salmonellaspp., realizamos um estudo para identificação de Salmonella spp. em amostras fecais de capivaras no Acre. Em uma amostra de conveniência, 54 capivaras de duas áreas urbanas e duas áreas rurais do Acre foram capturadas e mantidas por três a quatro dias para amostragem. Nenhum dos animais era sintomático de qualquer doença intestinal. Três amostras fecais foram coletadas de cada animal, durante sua permanência em cativeiro. Cada amostra foi cultivada para a presença de Salmonella spp. no Laboratório de Bacteriologia Veterinária da Universidade Federal do Acre. As amostras foram semeadas em caldo de pré-enriquecimento tetrationato e em peptona de caldo de pré-enriquecimento. Após 24 horas de incubação, todas as amostras foram semeadas em ágar MacConkey (MC) e ágar Salmonella-Shigella (SS). Colônias sugestivas foram submetidas a análises bioquímicas. Colônias compatíveis com Salmonella de acordo com o perfil bioquímico foram submetidas à sorotipagem (Sorokit para Salmonella - Probac do Brasil). Além disso, a primeira amostra de cada uma das 54 capivaras foi testada para Salmonella spp. usando PCR, visando gene hilA. Oito (5%) das 162 amostras examinadas por cultura bacteriana foram positivas para Salmonella spp. Enquanto quatro (7%) das 54 examinadas pela PCR foram positivas. Dos oito animais positivos em cultura, cinco eram de área urbana e três de área rural. Na PCR, apenas um animal positivo era de área urbana e quatro de área rural. Considerando o diagnóstico conjunto por ambos os testes, PCR e cultura, um animal foi considerado positivo. Todas as amostras foram coletadas em animais livres, sem sinais clínicos aparentes de salmonelose, indicando o potencial da capivara como reservatório nesse ecossistema.

ABSTRACT: The capybara (Hydrochoerus hydrochaeris) is the largest rodent in the world. In the state of Acre, Brazil, populations of capybaras have been increasing significantly. The role of capybaras in the transmission of certain bacterial zoonotic infections is not well understood, including bacteria of the genus Salmonella. Salmonella spp. generally cause enteritis or septicemia in mammals, however many mammalian species can carry the bacteria asymptomatically and shed it in their feces. To better understand the possible role of capybaras as reservoirs of Salmonella spp., we conducted a study of Salmonella within fecal samples from capybara in Acre. In a convenience sample, 54 capybaras from two urban and two rural areas of Acre were captured and kept for three to four days for sampling. None of the animals were symptomatic of any intestinal illness. Three separate fecal samples were collected from each animal, during their stays in captivity. Each sample was cultured for the presence of Salmonella spp. at the bacteriology laboratory of the Veterinary College of the Federal University of Acre. Samples were seeded in tetrationate pre-enrichment broth and in pre-enrichment broth peptone. After a 24 hour of incubation all samples were streaked on MacConkey Agar (MC) and Salmonella-Shigella Agar (SS). Suggestive colonies were submitted to biochemical analysis. Salmonella compatible colonies according to biochemical profile were submitted to serotyping (Sorokit for Salmonella - Probac do Brasil). In addition, the first sample from each of the 54 capybara was tested for Salmonella spp. using PCR targeting gene hilA. Eight (5%) of the 162 samples examined by bacterial culture were positive for Salmonella spp., while four (7%) of the 54 examined by PCR were positive. From the eight positive animals on culture, five were from urban area and three from rural area. On PCR, only one positive animal was from urban area and four were from rural area. Overall, by either test, one of the 54 animals was positive. All samples were collected in free - living animals with no apparent clinical signs of salmonellosis, indicating the potential of capybara as reservoir on this ecosystem.

RESUMO: A ultrassonografia com Doppler pulsado foi utilizado para avaliar o fluxo sanguíneo portal, velocidade portal e índice de congestão portal em 24 ovinos saudáveis divididos em grupos (cordeiros, borregos e ovelhas), de acordo com a idade. As medições foram realizadas no 11o espaço intercostal direito utilizando ângulo de insonação ideal e método de inclusão uniforme. Os valores médios obtidos em cada grupo foram comparados com ANOVA, seguido pelo teste post-hoc de Tukey. A velocidade portal e o fluxo de sangue portal foram estatisticamente semelhantes entre os grupos (P>0,05). A velocidade portal média foram 17,75; 17,13 e 16,75; enquanto o fluxo de sangue portal médios foram 26,65; 31,04 e 24,32 para cordeiros, borregos e ovelhas, respectivamente. O índice de congestão portal foi estatisticamente diferente entre os grupos e os valores para cordeiros, novilhos e ovelhas foram 0,009; 0,058 e 0,09, respectivamente (P<0.01). Observaram-se diferenças estatísticas nos diâmetros da veia porta, na área da veia porta e nos índices de congestão portal entre os grupos, provavelmente devido à influência do peso e não pela idade.

ABSTRACT: Pulsed Doppler ultrasound was used to evaluate portal blood flow, portal velocity and portal congestion index in 24 healthy sheep divided into groups (lambs, yearlings and ewes), according to age. Measurements were performed at the 11th right intercostal space using ideal insonation angle and uniform insonation method. Mean values obtained in each group were compared with one-way ANOVA, followed by Tukey post-hoc test. Portal velocity and portal blood flow were statistically similar between the groups (P>0.05). Mean portal velocity were 17.75; 17.13 and 16.75; while mean portal blood flow were 26.65; 31.04 and 24.32 for lambs, yearlings and ewes, respectively. Portal congestion index was statistically distinct between the groups and values for lambs, yearlings and ewes were 0.009; 0.058 and 0.09, respectively (P<0.01). Statistical differences were observed in portal vein diameter, portal vein area and portal congestion index between the groups, presumably due to influence of weight and not to age.

RESUMO Este trabalho objetivou apresentar a caracterização da morfologia do testículo de cutia (Dasyproctaprymnolopha) macho, com o intuito de colaborar com o conhecimento da morfofisiologia reprodutiva da espécie. Foram utilizados testículos de 47 animais, com idade entre um e dois anos, pesos homogêneos (2,08 ± 0,23kg), oriundos do Núcleo de Estudos e Preservação de Animais Silvestres do Centro de Ciências Agrárias da Universidade Federal do Piauí. As estruturas foram dissecadas, descritas, e fragmentos foram processados para a microscopia de luz, sendo, posteriormente avaliada a atividade gonadal. Observou-se que os testículos são órgãos elipsoides alongados, podendo ser encontrados na região inguinal ou na cavidade abdominal, não apresentando um escroto bem delimitado. Verificou-se também parênquima com característica histológica padrão para o órgão em mamíferos, com a identificação de oito tipos de associações celulares, caracterizando os estádios do ciclo do epitélio seminífero, com menor e maior frequência dos estádios 3 e 5, respectivamente.

ABSTRACT This study meant to characterize the morphology of the testicle from (Dasyprocta prymnolopha) agouti males, in order to collaborate with the knowledge of reproductive morphophysiology of the specie. Testicles were used from 47 animals aged between 1 and 2 years, homogeneous weight (2.08±0.23kg), coming from the Centre for the Study and Conservation of Wild Animals of Agricultural Sciences Center of the Federal University of Piauí. The structures were dissected, described and fragments were processed for light microscopy, and, subsequently, gonadal activity was evaluated. Testes were observed to be elongated ellipsoidal bodies that can be found in the groin or in the abdominal cavity, not having a clearly defined scrotum. We also could see parenchymal with standard histological characteristic for the mammalian body, with the identification of eight types of cell associations, characterized epithelium Seminiferous stages of the cycle, with lower and higher frequency of stages 3 and 5, respectively.

RESUMO O objetivo deste trabalho foi avaliar o efeito de níveis crescentes de substituição da silagem de sorgo por resíduo úmido de cervejaria sobre as características da carcaça e dos componentes não carcaça de cordeiros terminados em confinamento. Foram utilizados 24 cordeiros machos, não castrados, oriundos de parto simples e mantidos em baias individuais. Os tratamentos foram constituídos por quatro níveis de substituição de silagem de sorgo por resíduo úmido de cervejaria, sendo: 0%; 33,5%; 66,5% e 100% de substituição. Utilizou-se uma relação volumoso:concentrado de 50:50, com base na matéria seca. O concentrado era constituído por milho desintegrado, farelo de soja e mistura mineral. As dietas eram isoproteicas, contendo 18,81% de proteína bruta. Os cordeiros foram abatidos quando atingiram o escore de condição corporal estabelecido em 3 (escala de 1 a 5). As características de carcaça analisadas não foram influenciadas significativamente (P>0,05) pela substituição de silagem de sorgo por resíduo úmido de cervejaria, sendo obtidos valores médios de 18,92kg para peso de carcaça quente, 18,22 kg para peso de carcaça fria, 47,03% para rendimento de carcaça quente, 45,29% para rendimento de carcaça fria e 3,41% para índice de quebra ao resfriamento. Quanto às proporções dos diferentes cortes comerciais avaliados, em relação ao peso de carcaça fria, foram verificados valores médios de 31,86% para perna, 18,12% para paleta, 39,46% para costilhar e 9,08% para pescoço. As proporções de diafragma, omaso cheio e omaso vazio, em relação ao peso vivo ao abate dos cordeiros, diminuíram linearmente (P≤0,05). As demais variáveis dos componentes não carcaça avaliadas não foram influenciadas (P>0,05) pelos níveis de resíduo úmido de cervejaria das dietas. Pode-se recomendar o uso de resíduo úmido de cervejaria como fonte exclusiva de alimento volumoso quando se utiliza uma relação volumoso:concentrado de 50:50, em base de matéria seca, para terminação de cordeiros em sistema de confinamento.

ABSTRACT The objective of this study was to evaluate the effect of increasing levels of substitution of sorghum silage by wet brewery residue as forage food on carcass characteristics and non-carcass components of lambs finished in feedlot. Twenty four non castrated male lambs, Suffolk breed, single birth were maintained in individual stalls. The treatments consisted of four substitution levels of sorghum silage by wet brewery residue (0%; 33.5%; 66.5% or 100% of substitution). Roughage and concentrate were used at a 50:50 ratio, based on dry matter. The concentrate was composed of ground corn, soybean meal and mineral mixture. The diets were isoproteic containing 18.81% crude protein. The lambs were slaughtered when they reached the body condition score of 3 (ranging from 1 to 5). The carcass traits were not affected significantly (P>0,05) by substitution of sorghum silage by wet brewery residue. The average values of hot carcass weight, cold carcass weight, hot carcass yield, cold carcass yield and cooling weight losses were 18.92 kg, 18.22 kg, 47.03%, 45.29%, 3.41%, respectively. The proportion of commercial cuts in relation to the cold carcass weight, was 31.86% for leg, 18.12% for shoulder, 39.46% for ribs and 9.08% for neck. The proportion of diaphragm, omasum full and empty omasum in relation to body weight at slaughter decreased linearly (P≤0.05). The remaining variables of non-carcass components were not affected (P>0.05) by wet brewery residue inclusion. We recommend the use of wet brewery residue as exclusive source of roughage food when using roughage:concentrate at 50:50 ratio on a dry matter basis, for finishing feedlot lambs.

ABSTRACT The objective of this study was to evaluate the antiproliferative, cytotoxic and genotoxic potential of salty liquid synthetic flavorings of Butter, Cheddar Cheese and Onion. The antiproliferative potential (2.9-1500 µg/mL) was assessed by MTT assay after 72h using the human tumor lines SF-295 (glioblastoma), OVCAR-8 (ovarian), HCT-116 (colon) and HL-60 (promyelocytic leukemia) and primary cultures of murine Sarcoma 180 (S180) and peripheral blood mononuclear cells (PBMC). Allium cepa bulbs were exposed to growing respective doses (1 mL and 2 mL). Only Butter and Cheddar flavorings revealed cytotoxic activity on cancer cells, with IC50 values ranging from 125.4 µg/mL (Cheddar - HCT-116) to 402.6 µg/mL (Butter - OVCAR-8). Butter flavoring was the most cytotoxic on PBMC (136.3 µg/mL) and increased cell division rate in relation to the mitotic index but did not cause cellular aberrations. Onion and Cheddar flavorings reduced the mitotic index after 24h and 48h exposure, but only Onion flavoring resulted in cellular aberrations and mitotic spindle abnormalities, such as anaphase and telophase bridges, micronucleated cells, conchicine-metaphases and amplifications. So, Butter, Onion and/or Cheddar flavorings caused significant changes in the division of meristematic cells of A. cepa and presented cytotoxic action even on decontrolled proliferating human tumor cells.

Abstract The “bacuri” (Platonia insignis Mart., Clusiaceae) is a native tropical fruit from the Brazilian Amazon and Northeast Regions. Its seeds are used to treat inflammatory diseases, diarrhea and skin problems in traditional medical practices. Regarding its widespread medicinal uses, it is important to evaluate the biological and toxicological potential of this species. This way, the aim of this study was to investigate the in vitro cytotoxic and immunomodulatory effects of the hexanic extract of P. insignis seeds, as well as its in vivo acute oral toxicity. The biological evaluation was performed by the determination of cytotoxic (MTT and hemolysis assay) and immunomodulatory (phagocytic capacity, lysosomal volume and nitrite production) activities of EHSB in murine peritoneal macrophages. In addition, the oral acute toxicity was evaluated using female Wistar rats treated with EHSB (2.0 g/kg), in accordance with the OECD 423 Guideline. The EHSB showed low toxicity for macrophages in the MTT test (CC50 value: 90.03 µg/ml), as well as for erythrocytes, which caused only 2.5% hemolysis at the highest concentration. A strong immunomodulatory activity was observed by a markedly increase of the NO production, phagocytic ability and lysosomal volume. On the other hand, it was not observed deaths or changes in the clinical and behavioral parameters in the toxicological evaluation. This manner, the present study contributes to the knowledge about the immunomodulatory and toxicological properties of the P. insignis. This may provide perspectives for the evaluation and development of effective and safe phytomedicines created from the Brazilian local biodiversity.

Diante da importância da cultura da batata-doce, o presente estudo teve como objetivo otimizar a produção de plantas in vitro de diferentes cultivares. Segmentos nodais das cultivares ‘Abóbora', ‘Catarina', ‘Biaze', ‘Da Costa', ‘Americana' e ‘Morada Inta', foram cultivados em meio Murashige e Skoog e avaliados ao longo de oito semanas de cultivo in vitro. A cv. ‘Da Costa' também foi cultivada em meio isento de fitorreguladores, em meio acrescido de benzilaminopurina (BAP) e/ou ácido giberélico (GA3) ou em sistema de cultivo fotoautotrófico. Constatou-se que a taxa de propagação é variável de acordo com o cultivar e com o período de tempo de cultivo. O intervalo entre os subcultivos foi dependente do índice de senescência das plantas. Na cv. ‘Da Costa', uma maior produção de plantas completas foi obtida em meio nutritivo contendo BAP ou em meio isento de sacarose e em frascos vedados com algodão. As plantas oriundas de todas as cultivares apresentaram uma elevada taxa de sobrevivência durante a aclimatização.

Due to the importance of sweet potato culture, this study aimed to optimize the production of the in vitro plants of different cultivars. Nodal segments of cultivars ‘Abóbora', ‘Catarina', ‘Biaze', ‘Da Costa', ‘Americana' e ‘Morada Inta' were cultured on Murashige and Skoog medium and they were evaluated for eight weeks in an in vitro culture. The cv. ‘Da Costa' was also grown on a medium free of plant growth regulators, supplemented with benzylaminopurine (BA) and/or gibberellic acid (GA3) or in photoautotrophic culture system. It was found that the propagation rate is variable, depending on the cultivar and according to time of cultivation. The interval among subcultures was dependent on the rate of plant senescence. In cv. ‘Da Costa', a higher production of plants was obtained on medium containing BAP or on sucrose-free medium and in bottles sealed with cotton. Plants originating from all cultivars showed a high survival rate during the acclimatization.

Este trabalho foi desenvolvido com o objetivo de elaborar modelos matemáticos para previsão do crescimento e desenvolvimento da tiririca (Cyperus rotundus) com base em dias ou unidades térmicas acumuladas (graus-dia). Para isso, dois experimentos independentes foram desenvolvidos: o primeiro com fotoperíodo decrescente (março a julho) e o segundo com fotoperíodo crescente (agosto a novembro). Em cada experimento, foram realizadas dez avaliações de crescimento e desenvolvimento das plantas, quantificando-se a massa de matéria seca e a fenologia da espécie. Posteriormente, a fenologia foi ajustada a equações de primeiro grau, considerando-se os experimentos isolados e o agrupamentos destes. Da mesma forma, a massa seca total foi ajustada a modelos do tipo logístico. Em todas as regressões, avaliaram-se quatro possibilidades de escalas temporais para o eixo x: dias ou graus-dia acumulados, com temperatura basal (Tb) de 10, 12 e 15 oC. Em ambas as condições de fotoperíodo, o crescimento e o desenvolvimento da tiririca foram adequadamente ajustados a modelos matemáticos de previsão com base em unidades térmicas acumuladas, com destaque para Tb = 12 oC. Considerando-se graus-dia acumulados (x) calculados com Tb = 12 oC, a fenologia da tiririca pode ser prevista por y = 0,113x, enquanto o crescimento da espécie pode ser previsto por y = 37,678/(1+(x/509,353)-7,047).

This work was carried out with the objective of elaborating mathematical models to predict growth and development of purple nutsedge (Cyperus rotundus) based on days or accumulated thermal units (growing degree days). Thus, two independent trials were developed, the first with a decreasing photoperiod (March to July) and the second with an increasing photoperiod (August to November). In each trial, ten assessments of plant growth and development were performed, quantifying total dry matter and the species phenology. After that, phenology was fit to first degree equations, considering individual trials or their grouping. In the same way, the total dry matter was fit to logistic-type models. In all regressions four temporal scales possibilities were assessed for the x axis: accumulated days or growing degree days (GDD) with base temperatures (Tb) of 10, 12 and 15 oC. For both photoperiod conditions, growth and development of purple nutsedge were adequately fit to prediction mathematical models based on accumulated thermal units, highlighting Tb = 12 oC. Considering GDD calculated with Tb = 12 oC, purple nutsedge phenology may be predicted by y = 0.113x, while species growth may be predicted by y = 37.678/(1+(x/509.353)-7.047).

It is currently accepted that superoxide anion (O2•−) is an important mediator in pain and inflammation. The role of superoxide anion in pain and inflammation has been mainly determined indirectly by modulating its production and inactivation. Direct evidence using potassium superoxide (KO2), a superoxide anion donor, demonstrated that it induced thermal hyperalgesia, as assessed by the Hargreaves method. However, it remains to be determined whether KO2 is capable of inducing other inflammatory and nociceptive responses attributed to superoxide anion. Therefore, in the present study, we investigated the nociceptive and inflammatory effects of KO2. The KO2-induced inflammatory responses evaluated in mice were: mechanical hyperalgesia (electronic version of von Frey filaments), thermal hyperalgesia (hot plate), edema (caliper rule), myeloperoxidase activity (colorimetric assay), overt pain-like behaviors (flinches, time spent licking and writhing score), leukocyte recruitment, oxidative stress, and cyclooxygenase-2 mRNA expression (quantitative PCR). Administration of KO2 induced mechanical hyperalgesia, thermal hyperalgesia, paw edema, leukocyte recruitment, the writhing response, paw flinching, and paw licking in a dose-dependent manner. KO2 also induced time-dependent cyclooxygenase-2 mRNA expression in the paw skin. The nociceptive, inflammatory, and oxidative stress components of KO2-induced responses were responsive to morphine (analgesic opioid), quercetin (antioxidant flavonoid), and/or celecoxib (anti-inflammatory cyclooxygenase-2 inhibitor) treatment. In conclusion, the well-established superoxide anion donor KO2 is a valuable tool for studying the mechanisms and pharmacological susceptibilities of superoxide anion-triggered nociceptive and inflammatory responses ranging from mechanical and thermal hyperalgesia to overt pain-like behaviors, edema, and leukocyte recruitment.

O objetivo geral deste trabalho foi analisar o efeito da radiação vermelha de baixa intensidade sobre alguns parâmetros cinéticos do espermatozoide canino criopreservado. Ejaculados de oito cães adultos foram centrifugados, rediluídos em meio tris-gema de ovo com 6% de glicerol, e, posteriormente, fracionados em: T1: incidência de radiação vermelha (660 NM) (Fisioled - Mmoptics - 100mW) por 60 segundos, antes do resfriamento e após a descongelação; T2: incidência somente antes do resfriamento; T3: incidência somente após a descongelação; e T4: sem incidência. Após a descongelação, as amostras foram submetidas ao TTR utilizando-se Sperm Class Analyzer(r). No TTR0, TTR60 e TTR90, não houve diferença entre as variáveis analisadas pelo CASA. Somente no TTR30 os efeitos da incidência da radiação vermelha foram evidentes e significativos em T1 e T2; T1 resultou em baixa MT (12,5 + 10,6%) e T2 determinou o melhor resultado de MT 40,3 + 26,1%. De forma similar T1 apresentou maior número de espermatozoides estáticos (77,5±28,9%) em relação ao T2 (50,6±28%). Concluiu-se que a dupla incidência de radiação vermelha de baixa intensidade antes do resfriamento e após a descongelação teve efeito deletério sobre a motilidade do espermatozoide canino, expressa principalmente aos 30 minutos após descongelação.

The aim of this study was to analyze the effect of low intensity red light on some kinetic parameters of cryopreserved canine sperm. Ejaculates from 08 adult dogs were centrifuged, diluted in Tris-egg yolk with 6% glycerol, and subsequently separated into: T1: incidence of red light (660 nm) (Fisioled -MMOptics - 100mW) for 60 seconds before the cooling and after thawing, T2: just before cooling, T3: only after thawing, and T4: no incidence. After thawing the samples were subjected to TTR using Sperm analyzer(r). In TTR0, TTR60 and TTR90 there were no differences between the variables analyzed by CASA. Only in TTR30 the effects of the incidence of red light were visible and significant in T1 and T2. T1 resulted in low MT (12.5 ±10.6%) and T2 determined the best result of MT 26.1%±40.3. Similarly T1 showed a higher number of static spermatozoa (77.5±28.9%) compared to T2 (50.6±28%). We concluded that the double incidence of low intensity red light, before cooling and after thawing, had a deleterious effect on canine sperm motility expressed at 30 minutes after thawing.
