Abstract Objective To reproduce in an animal model the surgical technique of Masquelet used in the treatment of critical bone defects and to analyze the characteristics of the membrane formed around the bone cement. Methods A 10mm critical defect was created in the femoral shaft of 21 Sprague-Dawley rats. After resection of the central portion of the diaphysis, the defect was stabilized with a Kirschner wire introduced through the medullary canal and with the interposition of a bone cement spacer. After 2, 4, and 6 weeks of the surgical procedure, the animals were euthanized and evaluated on radiographs of the posterior limb regarding the size of the defect, alignment and stability of the osteosynthesis. The membranes formed around the spacer were subjected to histological analysis to assess thickness, connective tissue maturation and vascular density. Results Over time, the membranes initially made up of loose connective tissue were replaced by membranes represented by dense connective tissue, rich in thick collagen fibers. At six weeks, membrane thickness was greater (565 ± 208μm) than at four (186.9 ± 70.21μm, p = 0.0002) and two weeks (252.2 ± 55.1μm, p = 0.001). All membranes from the initial time showed foci of osteogenic differentiation that progressively reduced over time. Conclusion In addition to the structural and protective function of the membrane, its intrinsic biological characteristics can actively contribute to bone regeneration. The biological activity attributed by the presence of foci of osteogenesis confers to the membrane the potential of osteoinduction that favors the local conditions for the integration of the bone graft.

Resumo Objetivo Reproduzir em modelo animal a técnica cirúrgica de Masquelet utilizada no tratamento de defeitos ósseos críticos e analisar as características da membrana formada em torno do cimento ósseo. Métodos Um defeito crítico de 10mm foi realizado na diáfise femoral de 21 ratos Sprague-Dawley. Após a ressecção da porção central da diáfise o defeito foi estabilizado com fio de Kirschner introduzido pelo canal medular e com a interposição de espaçador de cimento ósseo. Após 2, 4, e 6 semanas do procedimento cirúrgico os animais foram eutanasiados e avaliados em radiografias do membro posterior quanto ao tamanho do defeito, o alinhamento e a estabilidade da osteossíntese. As membranas formadas em torno do espaçador foram submetidas a análise histológica para avaliação da espessura, da maturação do tecido conjuntivo e da densidade vascular. Resultados Ao longo do tempo as membranas inicialmente constituídas por tecido conjuntivo frouxo foram substituídas por membranas representadas por tecido conjuntivo denso, rico em fibras colágenas espessas. Com seis semanas a espessura das membranas foi maior (565 ± 208μm) do que com quatro (186,9 ± 70,21μm, p = 0,0002) e duas semanas (252,2 ± 55,1μm, p = 0,001). Todas as membranas do tempo inicial apresentaram focos de diferenciação osteogênica que reduziram progressivamente ao longo do tempo. Conclusão Além da função estrutural e protetora da membrana, suas características biológicas intrínsecas podem contribuir ativamente para a regeneração óssea. A atividade biológica atribuída pela presença de focos de osteogênese confere à membrana potencial de osteoindução que favorece as condições locais para a integração do enxerto ósseo.

BACKGROUND Trypanosoma cruzi, which causes Chagas disease (CD), is a versatile haemoparasite that uses several strategies to evade the host’s immune response, including adipose tissue (AT), used as a reservoir of infection. As it is an effective barrier to parasite evasion, the effectiveness of the drug recommended for treating CD, Benznidazole (BZ), may be questionable. OBJECTIVE To this end, we evaluated the parasite load and immunomodulation caused by BZ treatment in the culture of adipocytes differentiated from human adipose tissue-derived stem cells (ADSC) infected with T. cruzi. METHODS The ADSC were subjected to adipogenic differentiation. We then carried out four cultures in which we infected the differentiated AT with trypomastigote forms of the Y strain of T. cruzi and treated them with BZ. After the incubation, the infected AT was subjected to quantitative polymerase chain reaction (qPCR) to quantify the parasite load and transmission electron microscopy (TEM) to verify the infection. The supernatant was collected to measure cytokines, chemokines, and adipokines. FINDINGS We found elevated secretion of IL-6, CXCL-10/IP-10, CCL2/MCP-1, CCL5/RANTES, and leptin in infected fat cells. However, treatment with BZ promoted a decrease in IL-6. MAIN CONCLUSION Therefore, we believe that BZ has a beneficial role as it reduces inflammation in infected fat cells.

Résumé Ce travail présente le rapport d'une expérience de stage réalisée dans les disciplines de la Psychologie de l'éducation I et II, à la Faculté d'éducation de l'Université Fédérale de Goiás, offert aux cours de licence en Sciences Biologiques, Philosophie et Musique, au cours dans de la période académique 2020/1. L'objectif est de souligner l'importance de comprendre différentes méthodologies pour promouvoir l'apprentissage, en particulier dans le contexte de l'enseignement à distance d'urgence, en soulignant les possibilités et les difficultés du travail d'enseignement dans cette modalité, sur la base du cadre théorique de la Pédagogie Historico-Critique. L'expérience des stagiaires est rapportée, considérant le fondement théorique et les (im)possibilités d'analyse et d'intervention sur le terrain comme fondamentales pour la formation des enseignants engagés dans l'éducation et la transformation de la réalité. La conclusion est que l'expérience a été formatrice pour les stagiaires, car malgré les défis de l'enseignement à distance d'urgence, l'utilisation de méthodologies diversifiées s'est avérée bénéfique.

Abstract This work presents the report of an internship experience carried out in the subjects of Educational Psychology I and II, at the Faculty of Education of Federal University of Goiás, offered to the Licentiate degree in Biological Sciences, Philosophy and Music, at the academic period 2020/1. Our objective is to emphasize the importance of understanding different methodologies in the promotion of learning, especially in the context of Emergency Remote Teaching, punctuating possibilities and difficulties of teaching in this modality, based on the theoretical framework of the Historical-Critical Pedagogy. The experience reported considered a theoretical background and the (im)possibilities of analysis and intervention in practice as fundamental to qualificate teachers committed to education and the transformation of reality. The conclusion is that the experience was formative for the students, because despite the challenges of Emergency Remote Teaching, the use of diversified methodologies were beneficial.

Resumen Este trabajo relata una experiencia de pasantía realizada en las disciplinas de Psicología de la Educación I y II, en la Facultad de Educación de la Universidad Federal de Goiás, ofrecida a las carreras de Licenciatura en Ciencias Biológicas, Filosofía y Música, durante el período académico 2020/1. Se enfatizó en la importancia de comprender diferentes metodologías en la promoción del aprendizaje, especialmente en el contexto de Enseñanza Remota de Emergencia, puntuando las posibilidades y dificultades del trabajo docente en esta modalidade, con la base teórica no referencial de la Pedagogía Histórico-Crítica. Se expone la experiencia de las pasantes, considerando el trasfondo teórico y las (im)posibilidades de análisis y intervención en el campo como fundamentales para la formación docente comprometida con la educación y la transformación de la realidad. Se concluye que la experiencia fue formativa para las pasantes, pues el uso de metodologías diversificadas en la Enseñanza Remota de Emergencia, resultó beneficioso.

Resumo Este trabalho apresenta o relato de uma experiência de estágio, realizado nas disciplinas de Psicologia da Educação I e II, oferecidas aos cursos de Licenciatura em Ciências Biológicas, Filosofia e Música da Faculdade de Educação da Universidade Federal de Goiás, no período letivo de 2020/1. Objetiva-se ressaltar a importância da compreensão de diferentes metodologias na promoção da aprendizagem, especialmente no contexto do Ensino Remoto Emergencial, pontuando possibilidades e dificuldades do trabalho docente nesta modalidade, com base no referencial teórico da Pedagogia Histórico-Crítica. Relata-se a experiência das estagiárias, considerando a bagagem teórica e as (im)possibilidades de análise e intervenção em campo como fundamentais para a formação docente comprometida com a educação e a transformação da realidade. Conclui-se que a experiência foi formativa para as estagiárias, pois, apesar dos desafios do Ensino Remoto Emergencial, a utilização de metodologias diversificadas mostrou-se benéfica.

RESUMO Introdução: o câncer de mama é a neoplasia maligna mais comum em mulheres no mundo. A cirurgia tem sido o tratamento tradicional e, geralmente consiste em mastectomia com linfadenectomia, podendo causar dor pós-operatória. Por isso, buscamos estudar técnicas anestésicas regionais que possam minimizar esse efeito, como o bloqueio interpeitoral (PEC). Métodos: estudo controlado randomizado com 82 pacientes com câncer de mama submetidos à mastectomia com linfadenectomia de Janeiro de 2020 a Outubro de 2021, em hospital oncológico. Intervenções: dois grupos randomizados (controle - anestesia geral exclusiva e grupo PECS - anestesia geral e bloqueio PEC com levobupivacaína/ropivacaína). Aplicou-se um questionário com Escala Visual Analógica da dor 24h pós-cirurgia. Utilizamos os testes de Shapiro-Wilk, Mann-Whitney e Quiquadrado e analisamos os dados em R versão 4.0.0. Estudo registrado em Ensaios Clínicos Brasileiros (REBec). Resultados: no grupo PEC, 50% não apresentava dor 24 horas após a cirurgia enquanto no grupo controle, 42,86% negava quadro álgico. A maioria que apresentou dor classificou-a como dor leve (EVA de 1 a 3) - (42,50%) grupo PEC e (40,48%) controle (p=0,28). Apenas 17,50% consumiram opioides no grupo PEC, semelhante ao grupo controle com 21,43%. (p=0,65), (17,50%) grupo PEC e (21,43%) grupo controle (p=0,65). Houve baixo índice de complicações como PONV (náuseas, vômitos, cefaleia) em ambos os grupos. Na análise de subgrupo, não houve diferença estatística entre os grupos que usaram Levobupivacaína ou Ropivacaína quanto a dor pós-operatória e o consumo de opioides. Discussão: o grupo estudado apresentou baixa taxa de dor no pós-operatório e isso influenciou na análise estatística. Não houve diferença estatística quanto a dor pós-operatória entre grupos. Conclusão: não foi possível demonstrar melhores resultados com a associação do bloqueio PEC com analgesia intravenosa total. São necessários novos estudos para avaliar a eficácia do bloqueio anestésico no intraoperatório e pós-operatório.

ABSTRACT Objective: Breast cancer is the most common malignant neoplasm in women worldwide. Surgery has been traditional treatment and, generally, it´s mastectomy with lymphadenectomy, that can causes postoperative pain. Therefore, we seek to study regional anesthesic techniques that can minimize this effect, such as the interpectoral block (PECS). Methods: randomized controlled study with 82 patients with breast cancer who underwent mastectomy with lymphadenectomy from January 2020 to October 2021 in oncology hospital. Interventions: two randomized groups (control - exclusive general anesthesia and PECS group - received PECS block with levobupivacaine/ropivacaine and general anesthesia). We applied a questionnaire with Numeric Rating Scale for pain 24h after surgery. We used Shapiro-Wilk, Mann-Whitney and Chi-square tests, and analyzed the data in R version 4.0.0 (ReBEC). Results: in the PECS group, 50% were pain-free 24h after surgery and in the control group it was 42.86%. The majority who presented pain classified it as mild pain (VAS from 1 to 3) - (42.50%) PECS group and (40.48%) control group (p=0.28). Only 17.50% consumed opioids in the PECS group, similar to the control group with 21.43%. (p=0.65). There was a low rate of complications such as PONV in both groups. In the subgroup analysis, there was no statistical difference between the groups that used levobupivacaine or ropivacaine regarding postoperative pain and opioid consumption. Discussion: the studied group had a low rate of pain in the postoperative period and it influenced the statistical analysis. There wasn´t difference in postoperative pain in groups. Conclusion: was not possible to demonstrate better results with the association of the PECS block with total intravenous analgesia. Need further studies to assess the efficacy of the nerve block.

RESUMO A rigidez articular é a principal complicação do cotovelo, afetando o tratamento e o prognóstico. O manejo da contratura articular se baseia na sua etiologia, e a intervenção precoce deve modificar o desfecho dessa complicação. Objetivo: Revisar artigos sobre órtese articulada de cotovelo para reabilitação de rigidez articular. Métodos: Foi realizada uma revisão bibliográfica em periódicos disponíveis do PubMed, Medline e LILACS, utilizando os Descritores em Ciências da Saúde (DeCS): aparelhos ortopédicos; braquetes; cotovelo; articulação do cotovelo; contratura; e transtornos da articulação. A intenção foi estabelecer e conhecer estudos de alto poder de evidência já realizados, que tenham tido como referência a temática de órteses articuladas de cotovelo para reabilitação de rigidez articular. Resultados: Após a aplicação dos critérios de inclusão e exclusão, revisaram-se artigos indexados no PubMed, Medline e LILACS, encontrando-se quatro artigos pelo PubMed e nenhum pelo Medline ou LILACS. Dos quatro artigos, dois eram revisões sistemáticas e dois eram ensaios clínicos randomizados. Conclusão: Órteses articuladas de cotovelo podem ser benéficas no tratamento de rigidez articular, sendo sugerido que melhoram amplitude de movimentos (ADM) e dor, inclusive com efeito superior em relação às órteses não articuladas gessadas. ABSTRACT Joint stiffness is the most common complication in elbow injuries, presenting several etiologies and pathophysiological mechanisms that hinder treatment and prognosis. Prevention and treatment of joint contracture depend on the cause of stiffness, and early intervention should modify its outcomes. The methods used may be conservative or surgical, alone or in combination, according to each individual situation. Objective: Review articles on articulated elbow orthosis for joint stiffness rehabilitation. Methods: A literature review was conducted in journals available at the PubMed, Medline and LILACS databases, using the following Health Science Descriptors (DeCS): orthotic devices; braces; elbow; elbow joint; contracture; joint disorders. It sought to retrieve and analyze studies with the highest level of evidence that have already been conducted on articulated elbow orthosis for joint stiffness rehabilitation. Results: After applying the inclusion and exclusion criteria, four articles were included from PubMed and none from Medline or LILACS. Of the four PubMed articles, two were systematic reviews and two were randomized clinical trials. Conclusion: Articulated elbow orthoses can benefit joint stiffness treatment, improving range of motion and pain, showing superior effect compared to non-articulated plaster orthotics. Abstract The Bacanga River Estuary is socioeconomically important due to artisanal fishing and aquaculture. It is blocked by a dam and is under human pressure along its drainage basin, intensifying the eutrophication process. This study reports on the occurrence of phytoplankton blooms and trophic state (TSI and TRIX) at six sampling sites during the annual cycle. The estuary was divided into downstream and upstream regions. Higher salinity, turbidity, depth, and lower dissolved oxygen levels were found downstream; whereas, high levels of chlorophyll a and nutrient concentrations were observed in both regions. There were blooms of Leptocylindrus danicus (1.45 × 106 cells L-1) and Skeletonema costatum (1.89 × 106 cells L-1) downstream; whereas phytoflagellate proliferation, such as those of Chlamydomonas sp. (13.17 × 106 cells L-1), Euglena gracilis (7.84 × 106 cells L-1), and Euglena proxima (1.03 × 106 cells L-1) were recorded upstream, with Chlamydomonas sp. as the discriminant species of this zone. Both trophic indices (TSI; TRIX) indicated elevated trophic conditions for the estuary, classifying it as hypereutrophic. Nevertheless, TSI only showed a significant relationship with some specific phytoplankton blooms. Thus, TSI seems to be the trophic index with a better response in the assessment of estuarine ecological functioning.

Resumo Objetivo Verificar a capacidade de avaliação de testes de desempenho físico na identificação da baixa massa muscular (MM) em mulheres de meia-idade e idosas. Método Estudo transversal realizado com 540 mulheres de meia-idade (40-59 anos) e idosas (≥60 anos), nos municípios de Parnamirim e Santa Cruz, no Rio Grande do Norte, Brasil. Foram avaliadas MM, força de preensão palmar, extensão de joelho e velocidade da marcha. A baixa MM foi definida pelo índice de massa muscular esquelética menor que os 20% mais baixos para cada grupo de idade. Foram utilizados os testes: t de Student, qui-quadrado, análise de curva ROC para calcular a área sob a curva e ponto de corte de cada teste na discriminação das participantes com baixa MM. Foi considerado p<0,05 e IC de 95%. Resultados Para o grupo de meia-idade, as forças de preensão palmar e de extensão do joelho apresentaram sensibilidade (71,6% e 72,5%, respectivamente) e especificidade (59,4% e 56,0%, respectivamente) moderadas na identificação de baixa MM. Para as idosas, a velocidade da marcha e a força de preensão manual apresentaram boa sensibilidade (77,8% e 81,6%, respectivamente) e especificidade moderada (51,4% e 64,5%, respectivamente). A capacidade discriminatória da velocidade da marcha para as mulheres de meia-idade e da força de extensão do joelho para as mulheres idosas foi insatisfatória. Conclusão As medidas de força muscular são úteis para a triagem de baixa MM em mulheres de meia-idade, enquanto os testes de força de preensão manual e velocidade de marcha são úteis para idosas.

Abstract Objective To verify the ability to evaluate physical performance tests in the identification of low muscle mass (MM) in middle-aged and old women. Method This cross-sectional study was carried out with 540 middle-aged (40-59 years old) and old women (≥60 years old) in the municipalities of Parnamirim and Santa Cruz in the state of Rio Grande do Norte, Brazil. MM, handgrip strength, knee extension, and gait speed were evaluated. Low MM was defined by skeletal muscle mass index lower than the lower 20% for each age group. The following tests were used: Student’s t, chi-square, ROC curve analysis to calculate the area under the curve, and cutoff point of each test in the discrimination of participants with low MM. P<0.05 and CI of 95% were considered. Results For the middle-aged group, the handgrip and knee extension strength showed moderate sensitivity (71.6% and 72.5%, respectively) and specificity (59.4% and 56.0%, respectively) in the identification of low MM. For the old women, gait speed and handgrip strength showed good sensitivity (77.8% and 81.6%, respectively) and moderate specificity (51.4% and 64.5%, respectively). The discriminatory capacity of gait speed for middle-aged women and knee extension strength for old women were unsatisfactory. Conclusion Muscle strength measurements are useful for low MM screening in middle-aged women, while handgrip strength and gait speed tests are useful for older women.

Resumo O objetivo deste trabalho foi descrever os procedimentos metodológicos de um estudo longitudinal sobre a saúde de adolescentes, bem como caracterizar o perfil da amostra. Participaram desta pesquisa 302 adolescentes (13,9±1,2 anos) de Londrina – Paraná. Duas coletas de dados foram realizadas com intervalo de três anos. Foram coletados indicadores antropométricos, pressão arterial, frequência cardíaca, dor na coluna, desempenho escolar, atividade física, comportamento sedentário, hábitos alimentares, consumo de bebidas alcoólicas, tabagismo, condição socioeconômica e testes motores. Verificou-se frequência de pressão arterial elevada de 10,4%; índice de massa corporal em condições associadas com risco à saúde de 22,4%; e dor na coluna com intensidade moderada de 22,1%. A prática de esporte e/ou exercício físico (moderada à vigorosa; ≥150 minutos/semana) foi de 33,1%. O consumo elevado dos alimentos e bebidas não saudáveis variou de 21,2% (café ou chá) a 58,0% (frituras). Elevada proporção (87,1%) relatou nunca ter fumado. As prevalências de notas ≥ sete, variaram entre 21,4% (Biologia) e 71,1% (Educação Física). Apenas 33,2% tiveram desempenho escolar elevado. O maior atendimento de critérios foi no abdominal (76,4%) e o menor flexão de cotovelos (37,2%). Este trabalho possibilitará verificar a estabilidade de diferentes indicadores comportamentais e biológicos relacionados à saúde, bem como do desempenho escolar. Permitirá a identificação da associação entre desfechos em saúde e fatores de exposição de forma prospectiva, considerando a influência de potenciais variáveis de confusão. Tais informações podem contribuir para o planejamento de intervenções e políticas públicas na área da saúde.

Abstract The aim of this work was to describe the methodological procedures of a longitudinal study on adolescent health, as well as to characterize the sample profile. This research enrolled a sample of 302 adolescents from Londrina - Paraná. Two data collections were carried out with an interval of three years. Anthropometric indicators, blood pressure, heart rate, back pain, academic achievement, physical activity, sedentary behavior, eating habits, alcohol consumption, smoking, socioeconomic status, and motor tests were collected. There was a frequency of high blood pressure of 10.4%; body mass index in conditions associated with health risk of 22.4%; and spinal pain with moderate intensity of 22.1%. Sport and/or physical exercise practice (moderate-to-vigorous; ≥150 minutes/week) was 33.1%. High consumption of unhealthy foods and beverages ranged from 21.2% (coffee or tea) to 58.0% (fried foods). A high proportion (87.1%) reported never having smoked. The grades frequency ≥ seven varied between 21.4% (Biology) and 71.1% (Physical Education). Only 33.2% presented high academic achievement. The highest compliance with criteria was in the curl-up (76.4%) and the lowest in the 90º push-up (37.2%). This work will make it possible to verify the tracking of different behavioral and biological indicators related to health, as well as academic achievement. It will also allow the identification of the association between health outcomes and exposure factors prospectively, considering the influence of potential confounding variables. This information could contribute to the planning of public health interventions and policies.

Abstract INTRODUCTION: American cutaneous leishmaniasis (ACL) is a public health problem and has been associated with country’s territory. We aimed to analyze the spatial dynamics and socioeconomic factors correlated to the incidence of ACL in Pernambuco, Brazil from 2008 to 2017. METHODS: A cross-sectional, ecological study was conducted in the Brazilian municipalities. Patient data were obtained from the Health Hazard Notification System (SINAN); indicators and incidence for the total period and for quinquennium were obtained. Socioeconomic factors were analyzed to evaluate the association between the incidence of ACL and presence of bathroom and running water, garbage collection availability, inadequate water supply, sanitation, rural population, per capita income, and vulnerability to poverty. Spatial analysis considered the gross incidence; the Bayesian local empirical method and Moran spatial autocorrelation index were applied using Terra View and QGIS. RESULTS: The incidence of ACL reduced (0.29/100,000 inhabitants per year). Individuals with ACL were young adults (30.3%), men (60.2%), brown skinned (62.9%), rural residents (70.6%), and less educated (46.7%); had autochthonous transmission (78.8%); developed the cutaneous form (97.2%); had evolution to cure (82.7%); and were diagnosed using the clinical epidemiological criterion (70.5%). ACL occurred in the large part of the state and showed heterogeneous distribution, with persistence of two high priority intervention clusters covering Health Regions I, II, III, IV, and XII. CONCLUSIONS: Spatial analysis and epidemiological indicators complement each other. The combination of these methods can improve the understanding on ACL occurrence, which will help subsidize planning and enhance the quality and effectiveness of healthcare interventions.

Resumo Objetivos Avaliar os fatores que influenciam o resultado da osteossíntese pela redução fechada da fratura do colo femoral nos pacientes jovens. Métodos Foi feito um estudo retrospectivo com revisão dos dados dos pacientes operados em um hospital ortopédico de grande porte, de 2003 a 2011, com um total de 81 pacientes que atenderam aos critérios de inclusão. O intervalo de tempo entre a fratura e a cirurgia, o desvio inicial da fratura, a qualidade da redução e o posicionamento dos implantes foram os fatores avaliados. Resultados O estudo encontrou forte relação entre a qualidade da redução e o sucesso terapêutico. O grau de desvio inicial e o tempo entre o trauma inicial e a osteossíntese não influenciaramo desfecho cirúrgico emrelação à consolidação óssea. O correto posicionamento dos implantes mostrou relação com a evolução satisfatória no pós-operatório dos pacientes. Conclusão A qualidade da redução e o posicionamento dos implantes são fatores que influenciamoresultadodaosteossíntesenafraturadocolodofêmurnopacienteadultojovem.

Abstract Objectives To evaluate the factors that influence the outcome of osteosynthesis after closed reduction of the fracture of the femoral neck in young adult patients. Methods A retrospective study was conducted, reviewing the data of patients operated in a large orthopedic hospital from 2003 to 2011; a total of 81 patients met the inclusion criteria. The time interval between the fracture and the surgery, the initial fracture deviation, the quality of the reduction, and the placement of the implant were evaluated. Results The present study observed a strong relationship between the quality of the reduction and therapeutic success. The degree of the initial deviation and the time elapsed between the initial trauma and the osteosynthesis did not influence the surgical outcome regarding bone consolidation. The correct positioning of the implants was associated with a satisfactory evolution in the postoperative period. Conclusion The quality of the reduction and the positioning of the implants are factors that influence the results of osteosynthesis in fractures of the femoral neck in young adult patients.

ABSTRACT São Marcos Bay is an estuarine complex with semidiurnal tides that can reach more than 7 m during equinoctial spring tides. It is situated in the second largest Brazilian coastline and is subjected to continuous human activities holding an important port complex of Latin America. In order to contribute to the knowledge about the structure of the phytoplankton in a macrotidal systems, this study aims to evaluate the phytoplankton community and its relationship with environmental conditions in the São Marcos Bay. Five surveys in 2010 and 2011 were carried out on four sampling points during the rainy and dry seasons. Samples were taken during the flood and ebb phases at neap tides. Hydrological parameters were correlated with biological data (phytoplankton composition, abundance and biomass) using statistical analysis. The phytoplankton biomass (chlorophyll a) characterized the bay as quite productive (10.43±7.62 mg m-3) with the nanophytoplankton as the dominant fraction. Seasonal variation was observed in phytoplankton abundance with higher values (34,262±18,422 cells L-1) in the rainy season. Diatoms were the most important phytoplankton group, pointing out Nitzschia sp., Diploneis weissflogii and Synedra sp. as the dominant species. This study revealed that the composition of phytoplankton community was mainly influenced by the local dynamics, governed by macrotides, and precipitation regime that mostly contributed to the seasonal fluctuations of the environmental conditions, such as salinity, dissolved oxygen, and nutrients.

O estuário do rio Bacanga apresenta um comportamento hidrodinâmico com fluxo de marés limitado por uma barragem. Ele é considerado como um ambiente hipereutrófico que recebe diariamente altas cargas de esgoto doméstico sem tratamento. Este trabalho teve como objetivo avaliar a variação espaço-sazonal da comunidade fitoplanctônica e suas relações com parâmetros ambientais. Amostragens bimestrais foram realizadas em seis pontos fixos entre 2012 e 2013, obtendo valores dos parâmetros físico-químicos e biológicos (clorofila a, composição e abundância do fitoplâncton) para realização das análises estatísticas. Os resultados indicam que a comunidade fitoplanctônica é representada por diatomáceas, sendo Skeletonema costatum a espécie dominante responsável por pulsos de florações em abril e junho de 2012. O predomínio dessa espécie está relacionado aos elevados teores de silicato, pH e turbidez da água. Outros eventos de florações como da Euglena gracilis e Chlamydomonas sp. foram registrados em fevereiro de 2013, quando os teores de fósforo total estiveram elevados e as taxas de oxigênio dissolvido foram superiores. Os dinoflagelados, cianobactérias e a diatomácea Thallassiosira sp. apresentaram ampla distribuição no período de estiagem e estão altamente correlacionados com a salinidade, transparência da água e nutrientes. Abstract The Bacanga River Estuary has a hydrodynamic behavior and its tidal flow is limited by a dam. It is considered as a hypertrophic environment that receives daily high loads of domestic sewage without treatment. This study aimed to evaluate the spatial and temporal variation of phytoplankton community and its relationship with environmental parameters. Bi-monthly sampling campaigns were carried out at six fixed sites between 2012 and 2013. Physical-chemical and biological parameters were collected (chlorophyll a, phytoplankton composition and abundance) to perform the statistical correlations. The results indicate that phytoplankton community is mostly represented by diatoms, with Skeletonema costatum being the dominant species responsible for bloom in April and June of 2012. The dominance of this species is related to the high silicate concentrations, pH and turbidity. Other blooms events as well as the Euglena gracilis and Chlamydomonas sp. were recorded in February 2013, when the total phosphorus concentrations were high and the dissolved oxygen concentrations were higher. Dinoflagellates, cyanobacteria and diatom Thallassiosira sp. were widely distributed in the dry period and highly correlated with salinity, water transparency and nutrients. Hence, the distribution of phytoplankton community is more defined seasonally, rather than spatially.

Resumo O presente estudo descreve a ocorrência de coinfecção com Hepatozoon canis e duas espécies de Leishmania (L. infantum ou L. braziliensis) em cães. Quatro cães sorologicamente diagnosticados com leishmaniose visceral foram eutanasiados. Amostras do baço e fígado foram submetidas à histopatologia e extração de DNA. Merontes de H. canis foram observados nos quatro cães. A infecção por H. canis foi confirmada por PCR e sequenciamento de um fragmento do gene 18S rRNA. A infecção por Leishmania e tipagem foram realizadas por PCR-RFLP do região intergênica ITS1. A carga parasitária foi calculada pela qPCR quantitativa baseada no gene ssrRNA. O teste DPP - Dual Path platform foi realizado. Apenas o Cão #2 era assintomático. Os cães #1 e #4 estavam infectados com L. infantum e foram positivos no DPP. Os cães #2 e #3 estavam infectados com L. braziliensis e foram negativos no DPP. Além disso, visceralização foi observada nos cães #2 e #3, nos quais L. braziliensis foi detectada em amostras de baço e fígado. A visceralização da L. braziliensis associada a sinais clínicos sistêmicos sugerem que esta coinfecção pode ter influenciado na carga parasitária e progressão da doença.

Abstract This study describes the occurrence of dogs naturally co-infected with Hepatozoon canis and two Leishmania species: L. infantum or L. braziliensis. Four dogs serologically diagnosed with Visceral Leishmaniasis were euthanized. Liver and spleen samples were collected for histopathological analysis and DNA isolation. H. canis meronts were observed in tissues from all four dogs. H. canis infection was confirmed by PCR followed by sequencing of a fragment of 18S rRNA gene. Leishmania detection and typing was confirmed by ITS1' PCR-RFLP and parasite burden was calculated using ssrRNA quantitative qPCR. A DPP - Dual Path platform test was performed. One out (Dog #2) of four animals was asymptomatic. Dogs #1 and #4 were infected by L. infantum and were DPP test positive. Dogs #2 and #3 were infected by L. braziliensis and were DPP test negative. Furthermore, visceral dissemination was observed in Dogs #2 and #3, since L. braziliensis was detected in liver and spleen samples. The visceral dissemination of L. braziliensis associated with systemic signs suggested that this co-infection could influence the parasite burden and disease progression.

RESUMO OBJETIVOS: O tumor de células gigantes (TCG) é uma neoplasia óssea benigna relativamente rara em adultos, porém seu comportamento biológico ainda é imprevisível. A incidência de recidiva local apresenta variação entre 0-65% em estudos internacionais, porém há poucos dados referentes a essa complicação em nossa população. MÉTODOS: Foram coletadas informações sobre 155 pacientes com diagnóstico histológico confirmado de TCG, acompanhados no serviço de oncologia ortopédica da nossa instituição, de janeiro de 2000 a julho de 2014. As características demográficas foram avaliadas e comparadas entre os pacientes que apresentaram recidiva local durante o seguimento clínico. RESULTADOS: Houve recidiva local em 26 pacientes (16,7%), dos quais 22 eram do sexo feminino (84,6%). A localização mais comum de recidiva local foi o fêmur distal (38,4%). Onze pacientes apresentaram recidiva precoce, enquanto 15 casos foram diagnosticados após 15 meses, o que representa, respectivamente, 42,3% e 57,7%. Metástases foram identificadas em cinco pacientes (3,2%). CONCLUSÃO: Os fatores relacionados ao tumor não evidenciaram aumento da incidência de recidiva local de tumor de células gigantes. O tratamento cirúrgico com margem intralesional é uma opção válida no tratamento de recidivas locais e não apresenta diferença de sobrevida livre de doença entre outros tipos de procedimentos. Tratamento clínico é reservado em casos de tumores irressecáveis ou impossibilidade de tratamento cirúrgico.

ABSTRACT OBJECTIVE: Giant cell tumors are benign bone neoplasms that are relatively rare in adults and their biological behavior is still unpredictable. The incidence of local recurrence has presented variation between 0% and 65% in studies conducted worldwide, but few data are available on this complication in the Brazilian population. METHODS: Information on 155 patients with confirmed histological diagnoses of giant cell tumor who were treated in our institution's orthopedic oncology service between January 2000 and July 2014 was gathered. Demographic characteristics were evaluated and compared between patients who presented local recurrence during the clinical follow-up. RESULTS: Local recurrence was observed in 26 patients (16.7%), of whom 22 were female (84.6%). The most common site of local recurrence was the distal femur (38.4%). Eleven patients presented early recurrence, while 15 cases were diagnosed after 15 months, representing 42.3% and 57.7%, respectively. Metastases were identified in five patients (3.2%). CONCLUSION: Tumor-related factors did not show any increased incidence of local recurrence of giant cell tumors. Surgical treatment with an intralesional margin is a valid option for treating local recurrences and does not show any difference in disease-free survival in relation to other types of procedures. Clinical treatment is reserved for cases of unresectable tumors or when surgical treatment is impossible.

RESUMO OBJETIVO: O estudo avalia a diferenciação in vitro das células mesenquimais isoladas do estroma da medula óssea em tenócitos após tratamento com extrato de tendão bovino. MÉTODOS: Tendões bovinos foram usados para confecção do extrato e estocados a -80 °C. Células mesenquimais do estroma da medula óssea (BMSCs) de três doadores foram usadas para os testes de citotoxicidade por MTT e diferenciação celular por qPCR. RESULTADOS: Os dados mostram que células mesenquimais do estroma da medula óssea tratadas por até 21 dias em presença do extrato de tendão bovino diluído em concentrações crescentes (1:10, 1:50 e 1:250) promovem a ativação da expressão de biglican, colágeno tipo I e fibromodulina. CONCLUSÃO: Nossos resultados mostram que o extrato de tendão bovino é capaz de promover a diferenciação das BMSCs em tenócitos.

ABSTRACT OBJECTIVE: This study evaluated in vitro differentiation of mesenchymal stromal cells isolated from bone marrow, in tenocytes after treatment with bovine tendon extract. METHODS: Bovine tendons were used for preparation of the extract and were stored at -80 °C. Mesenchymal stromal cells from the bone marrow of three donors were used for cytotoxicity tests by means of MTT and cell differentiation by means of qPCR. RESULTS: The data showed that mesenchymal stromal cells from bone marrow treated for up to 21 days in the presence of bovine tendon extract diluted at diminishing concentrations (1:10, 1:50 and 1:250) promoted activation of biglycan, collagen type I and fibromodulin expression. CONCLUSION: Our results show that bovine tendon extract is capable of promoting differentiation of bone marrow stromal cells in tenocytes.
