Synthesis and Anticancer Activity of Homodimeric Morita-Baylis-Hillman Adducts Based on 3-Hydroxyindolin-2-one Core

Cancer treatment represents one of the main scientific study targets today, mainly due to the pronounced side effects arising from chemotherapy. This study reports the synthesis, characterization, and anticancer activity of ten compounds from the Morita-Baylis-Hillman reaction. Ethylene glycol diacrylate was used as a double Michael acceptor in reactions with isatin derivatives to give homodimers of 3-hydroxyindolin-2-one core, recognized in the literature for its extensive pharmacological profile. The use of 1,4-diazabicyclo[2,2,2]octane (DABCO) as a catalyst and room temperature were the optimal conditions for the study reaction. The isolated yields were up to 63%, with most reaction times inferior to 24 h, some as fast as 15 min. The anticancer potential of the synthesized dimers was evaluated in vitro against three cancer strains, resulting in average inhibitory concentrations up to 0.72 µM. It was also found that the best performing homodimers are more active than their monomeric counterparts. Considering the promising selectivity indices observed, the preliminary results obtained here act as a basis for broader tests regarding the effectiveness of homodimeric adducts against cancer cells.

Abstract Glass ceramics’ fractures in zirconia fixed dental prosthesis (FDP) remains a clinical challenge since it has higher fracture rates than the gold standard, metal ceramic FDP. Nanoindentation has been shown a reliable tool to determine residual stress of ceramic systems, which can ultimately correlate to failure-proneness. Objectives: To assess residual tensile stress using nanoindentation in veneered three-unit zirconia FDPs at different surfaces of pontics and abutments. Methodology: Three composite resin replicas of the maxillary first premolar and crown-prepared abutment first molar were made to obtain three-unit FDPs. The FDPs were veneered with glass ceramic containing fluorapatite crystals and resin cemented on the replicas, embedded in epoxy resin, sectioned, and polished. Each specimen was subjected to nanoindentation in the following regions of interest: 1) Mesial premolar abutment (MPMa); 2) Distal premolar abutment (DPMa); 3) Buccal premolar abutment (BPMa); 4) Lingual premolar abutment (LPMa); 5) Mesial premolar pontic (MPMp); 6) Distal premolar pontic (DPMp); 7) Buccal premolar pontic (BPMp); 8) Lingual premolar pontic (LPMp); 9) Mesial molar abutment (MMa); 10) Distal molar abutment (DMa); 11) Buccal molar abutment (BMa); and 12) Lingual molar abutment (LMa). Data were assessed using Linear Mixed Model and Least Significant Difference (95%) tests. Results: Pontics had significantly higher hardness values than premolar (p=0.001) and molar (p=0.007) abutments, suggesting lower residual stress levels. Marginal ridges yielded higher hardness values for connectors (DPMa, MMa, MPMp and DPMp) than for outer proximal surfaces of abutments (MPMa and DMa). The mesial marginal ridge of the premolar abutment (MPMa) had the lowest hardness values, suggesting higher residual stress concentration. Conclusions: Residual stress in three-unit FDPs was lower in pontics than in abutments. The outer proximal surfaces of the abutments had the highest residual stress concentration.

Morita-Baylis-Hillman adducts (MBHA) is a class of polyfunctional molecules that has been standing out due to their versatility and expressive biological activities. Therefore, this paper describes the synthesis and antiproliferative activity of some new MBHA/7-choroquinoline hybrids. The Michael acceptors were obtained starting from 4,7-dichloroquinoline which were submitted to the Morita-Baylis-Hillman reaction with ortho, meta and para-nitrobenzaldehyde. The in vitro screening of the synthetized MBHA against NCI-H292, HCT-116 and MCF-7 cancer cells suggests the influence of the spacer chain in its inhibition potential. The 50% inhibitory concentration (IC50) obtained in the antiproliferative assay using MCF-7, HCT-116, HL-60 and NCI-H292 cancer cells indicate expressive cytotoxic potential of the adducts containing nitro group in the ortho position, with IC50 of 4.60 µmol L-1. MBHA/7-choroquinoline hybrids were more active than MBHA described in literature, indicating the improvement of the cytotoxic effect due to 7-chloroquinoline moiety in the molecular structure, with maximum selectivity index values of 11.89.

Upconversion nanoparticles (UCNPs) are suitable materials for bioapplications due to their ability to emit visible light under near infrared (NIR) excitation, in the biological transparency range. Polycaprolactone(PCL)-based scaffolds are widely used in tissue engineering in combination with inorganic compounds to improve bioactivity and osteoconductive properties. This work proposes a 3D printed composite scaffold with upconversion property aiming at biomedical applications in therapy-stimulated bone repair and photodynamic therapy (PDT). The system combines PCL polymer, UCNPs-apatite and a PDT photosensitizer. Thermal and rheological behaviors of the composite were similar to pure PCL polymer. Mechanical properties were improved by adding UCNPs-apatite. The 3D printable composite presented upconversion property and potential for PDT application, which was demonstrated by singlet oxygen generation under 980 nm excitation. Cytotoxicity, genotoxicity and mutagenicity assays indicated no toxicological effects at low concentrations of rare earth elements. Taken together, a potential multifunctional material is proposed for biomedical applications.

A new method to identify the authenticity of Brazilian banknotes (R$) is proposed using digital image analysis by a smartphone application, Photometrix®, associated with chemometric tools. Authentic banknotes (n = 4) obtained in local bank and falsified (n = 12) by the author himself by means of three printers (laser-Deskjet®, wax-Color Qube® and inkjet-Deskjet®) were used for the development of the methodology. An experimental planning was performed to evaluate the authenticity of the banknotes determining: the best region (holographic band, tactile region in the lower left and right upper number, effigy microimpressions, lower number left, animal figure, watermark and puzzle); Region of Interest (ROI) (16x16; 32x32 and 64x64 pixels), focal length (8, 12, 15 and 20 cm) and variation of the smartphone (Motorola G5 Plus, Samsung S7 and LG K10). The best regions chosen were effigy microimpressions and round the animal. Promising results were obtained from ROI 16x16 pixels with focal length fixed at 10 cm. Thus, the application presents promising features for analyzing the authenticity of banknotes by digital images making it fast, non-destructive, portable, with low cost, and can be accessed by all society.

Composition and richness of monogonont rotifers from La Plata River Basin, South America

Resumo: Apresentamos aqui o primeiro estudo que analisou a composição e riqueza de rotíferos de toda a bacia do Rio da Prata, a segunda maior da América do Sul, com amostragens simultâneas e padronizadas. Quinze grandes reservatórios e oito trechos lóticos foram selecionados nas porções alta, média e baixa dos rios Paraná, Paraguai e Uruguai, que atuam como os principais formadores da bacia do Prata. Coletamos um total de 86 amostras (habitats de águas abertas) em 2010. Foi encontrada uma média de 27 ± 11 espécies por sub-bacia, com maior riqueza no Baixo Paraná (41 espécies), seguido por Paranapanema (40 espécies) e Baixo Uruguai (38 espécies). Uma baixa riqueza foi observada no Médio Uruguai e no Médio Paraná. Encontramos 106 espécies pertencentes a 21 famílias e duas ordens. A família com maior número de espécies foi Lecanidae (21), seguida por Brachionidae (20), Trichocercidae (9) e Synchaetidae (8). As espécies com maior ocorrência foram Conochilus dossuarius, Kellicottia bostoniensis, Keratella americana, Keratella cochlearis e Hexarthra mira. Novas ocorrências de rotíferos foram registradas para o Brasil (Colurella adriatica), Estado de São Paulo (Enteroplea lacustris) e Argentina (Gastropus hyptopus, Harringia rousseleti e Lecane thienemanni). A correlação de Spearman entre o número de espécies e as variáveis físicas e químicas demonstrou correlação positiva com clorofila e temperatura, e correlação negativa com oxigênio dissolvido. Estendemos a lista de distribuição para algumas espécies nativas (Lecane ludwigii) e não-nativas de rotíferos (K. bostoniensis). Disponibilizamos também uma lista de espécies de rotíferos Monogononta encontrados nas estações amostradas.
Abstract: We present here the first study that analyzed the composition and richness of rotifers of the entire La Plata River basin, the second largest in South America, based on simultaneous and standardized sampling. Fifteen large reservoirs and eight river stretches were selected in the upper, middle, and lower portions of the Paraná, Paraguay, and Uruguay Rivers, which are the major rivers of the La Plata basin. We took a total of 86 samples (open water habitats) in 2010. A mean of 27±11 species per sub-basin was found, with the highest richness in the Lower Paraná (41 species), followed by the Paranapanema (40 species) and Lower Uruguay (38 species). Low richness was observed in the Middle Uruguay and Middle Paraná. We found 106 species belonging to 21 families and two orders. The family with the highest number of species was Lecanidae (21), followed by Brachionidae (20), Trichocercidae (9), and Synchaetidae (8). The species with higher occurrences were Conochilus dossuarius, Kellicottia bostoniensis, Keratella americana, Keratella cochlearis and Hexarthra mira. New occurrences of rotifers were registered for Brazil (Colurella adriatica), São Paulo State (Enteroplea lacustris), and Argentina (Gastropus hyptopus, Harringia rousseleti and Lecane thienemanni). Spearman correlation between the number of species and physical and chemical variables demonstrated positive correlation with chlorophyll and temperature, and negative correlation with dissolved oxygen. We extend the distribution list for some native (Lecane ludwigii) and non-native species of rotifers (K. bostoniensis). We also list the monogonont rotifer species found at the sampling stations.

We describe the microwave-assisted synthesis of thirteen functionalized peptoids and evaluate their in vitro antileishmanial activity against forms of Leishmania (Leishmania) amazonensis promastigotes. Synthesis via the Ugi four-component reaction (Ugi 4CR) reaction furnished the compounds of interest in 55-80% yield; reactions were conducted in a microwave reactor and lasted only 10 min. We then screened the antileishmanial activity of the synthesized compounds in vitro. To determine the IC50 (inhibitory concentration necessary to inhibit the growth of 50% of parasites) values, we selected the compounds that inhibited L. (L.) amazonensis growth by more than 50%. The seven selected compounds displayed IC50 values ranging from 2.6 to 72 µM after incubation for 48 h. Three peptoids gave IC50 values between 2.6 and 7.9 µM and can be considered as bioreactive molecules (hit criteria).

RESUMO Objetivo: Comparar a composição de tecidos, o conteúdo mineral ósseo e a densidade mineral óssea totais e por regiões do corpo, a força de preensão manual estática, e força isocinética da articulação do joelho, entre um grupo de jogadores de futebol amadores e um Grupo Controle. Métodos: Estudo transversal utilizando pletismografia de ar deslocado para estimar o volume corporal, para subsequente cálculo da densidade corporal. A composição de tecidos, o conteúdo mineral ósseo e a densidade mineral óssea foram avaliados para o corpo todo e regiões padronizadas através da absorciometria de raios-X de dupla energia. A força de preensão manual estática foi avaliada por um dinamômetro ajustável. Os momentos máximos de força das ações musculares concêntricas para os extensores e flexores do joelho foram avaliados pela dinamometria isocinética (60°/s). Foi calculado o valor d-Cohen para apreciar a magnitude do efeito das diferenças entre grupos. Resultados: Os futebolistas apresentaram níveis superiores de conteúdo mineral ósseo em comparação com os adultos ativos do Grupo Controle (+651g; d=1,60; p<0,01) e obtiveram valores superiores de densidade mineral óssea (d: 1,20 a 1,90; p<0,01) para a coluna lombar, L1-L4 (+19,4%), membros superiores (+8,6%) e membros inferiores (+16,8%). Para a força de preensão (estática) a diferença foi moderada (d=0,99; p<0,01) com valores mais elevados apresentados pelo futebolistas (+5,6kg; d=0,99; p<0,01). Conclusão: A prática de futebol promove uma regulação adequada da composição corporal (tecidos magro e gordo) e ganhos na densidade mineral óssea, mais acentuada em partes do corpo com maior exposição aos impactos mecânicos da atividade motora.
ABSTRACT Objective: To compare tissue composition, total and regional bone mineral content and bone mineral density, static hand grip and knee joint isokinetic strength between amateur soccer players and Control Group. Methods: Cross-sectional study. Air displacement plethysmography was used to estimate body volume and, in turn, density. Body composition, bone mineral content and bone mineral density were assessed for the whole body and at standardized regions using dual energy X-ray absorptiometry. Static grip strength was assessed with an adjustable dynamometer, and peak torque derived from isokinetic strength dynamometer (concentric muscular knee actions at 60°/s). Magnitude of the differences between groups was examined using d-Cohen. Results: Compared to healthy active adults, soccer players showed larger values of whole body bone mineral content (+651g; d=1.60; p<0.01). In addition, differences between groups were large for whole body bone mineral density (d=1.20 to 1.90; p<0.01): lumbar spine, i.e. L1-L4 (+19.4%), upper limbs (+8.6%) and lower limbs (+16.8%). Soccer players attained larger mean values in strength test given by static hand grip protocol (+5.6kg, d=0.99; p<0.01). Conclusion: Soccer adequately regulates body composition and is associated better bone health parameters (bone mineral content and density at whole-body and at particular sites exposed to mechanical loadings).

Seed exudates of Sesbania virgata (Cav.) Pers. stimulate the asymbiotic phase of the arbuscular mycorrhizal fungus Gigaspora albida Becker & Hall

RESUMO Sesbania virgata é uma leguminosa utilizada na restauração de áreas degradadas e que forma simbiose com fungos micorrízicos arbusculares (AMF). Suas sementes exudam metabólitos secundários que podem influenciar a colonização pelos AMF. Neste trabalho, estudamos os efeitos de exsudatos de sementes (SE) e radiculares (RE) de S. virgata na fase assimbiótica de Gigaspora albida. Esporos de G. albida foram germinados em meio suplementado com diferentes concentrações de SE ou RE. Após sete dias, a germinação dos esporos foi estimulada (46,6%) com a maior concentração de SE, enquanto o crescimento micelial foi estimulado com a menor concentração de SE. Por sua vez, RE não teve efeito na fase assimbiótica de G. albida. Conluiu-se que SE exercem efeitos positivos na fase assimbiótica de G. albida e que os efeitos diferenciais entre SE e RE de S. virgata na fase de assimbiótica fúngica podem ser explicados pelo seu conteúdo distinto de metabólitos secundários.
ABSTRACT Sesbania virgata is a legume used in the restoration of degraded areas and forms a symbiosis with arbuscular mycorrhizal fungi (AMF). Its seeds exude secondary metabolites that may influence the colonization by AMF. In this work, we studied the effects of seed (SE) and root exudates (RE) of S. virgata on the asymbiotic phase of Gigaspora albida. Spores of G. albida were germinated in medium supplemented with different concentrations of SE or RE. After seven days, spore germination was stimulated (46.6%) in the medium supplemented with the highest concentration of SE, while the mycelial growth was stimulated with the lowest SE concentration. In turn, RE had no effect on the fungal asymbiotic phase. We concluded that SE exert a positive effect on the asymbiotic phase of G. albida and that the different effects between SE and RE of S. virgata can be explained by their distinct content of secondary metabolites.

RESUMO OBJETIVO: O presente estudo examinou a concordância entre os indicadores de saúde óssea e composição tecidual resultantes da aplicação de equipamentos concorrentes de absorciometria de raios X de dupla energia (DXA). MÉTODO: A amostra (n = 32), com 27,6 ± 10,1 anos de idade avaliados antropometricamente, inclui impedância bioelétrica com multifrequência e pletismografia de ar deslocado. O conteúdo mineral ósseo (CMO), a área de tecido ósseo, o tecido magro e o tecido gordo de corpo inteiro foram obtidos considerando o modo pencil beam (Lunar DPX-MD+) e o fan beam (Lunar iDXA). Para cada um dos equipamentos, foi efetuado um scanner proximal do fêmur, sendo produzida informação sobre a densidade mineral óssea (DMO) do colo, nomeadamente triângulo de Ward, trocanter e haste. Na fase de processamento, foi definida uma região de interesse (ROI; coxa direita). As análises compreenderam a diferença de médias de medidas repetidas com cálculo da magnitude de efeitos (d), coeficiente de correlação intraclasse (CCI), coeficiente de variação (CV). RESULTADOS: Foram obtidos CCI>0,900 para todas as medidas, com diferenças intraindividuais largas apenas para CMO (d = 1,312; CV = 2,7%), área de tecido ósseo (d = 1,761; CV = 2,7%), tecido gordo total (d = 1,612; CV = 11%) e tecido magro em todos os segmentos (d = 1,237-1,687; CV = 2,0-41%). A massa magra da ROI apresentou uma variaçāo intraindividual muito larga (d = 4,014; CV = 8,4%). CONCLUSÃO: Foram encontradas diferenças negligenciáveis para a DMO de corpo todo. As medidas de massa gorda e massa magra obtidas por DXA nāo devem ser tidas como critério, mas antes como referenda, muito especialmente quando se delimita uma ROI.
SUMMARY OBJECTIVE: The current study was aimed to examine intra-individual variation on indicators of bone health in addition to whole-body plus appendicular tissue measurements using two concurrent assessments based on pencil beam and fan beam dual energy X-ray absorptiometry (DXA) systems in adult athletes from several sports. METHOD: Thirty-two male participants (27.6±10.1 years) were measured on anthropometry including multifrequency bioelectric impedance and air-displacement plethysmography. Bone mineral content (BMC), bone area, fat and lean soft tissue were derived using pencil beam (Lunar DPX-MD+) and fan beam (Lunar iDXA) absorptiometry. Bone mineral density (BMD) was obtained for the femoral neck, trochanter and triangle of ward. Finally, the right thigh was defined as a region of interest (ROI). Analyses comprised intra-class correlation (ICC), Effect size (d) from mean differences of repeated measurements, coefficient of variation (CV) RESULTS: ICC were >0.900 for all measurements. Intra-individual differences were large for BMC (d=1,312; CV=2,7%), bone area (d=1,761; CV=2,7%), fat tissue (d=1,612; CV=11%) and all indicators of appendicular lean soft tissue (d=1,237-1687; CV=2,0-4,1%). A very large difference (d=4,014; CV=8.4%) was diagnosed for lean soft tissue of the ROI. CONCLUSION: Although differences among concurrent instruments for BMC and bone area, the effect size of mean differences was negligible for BMD. Fat and lean soft tissue derived from DXA should be interpreted as reference values (not criterion) due to equipment-related variation, more apparently in the ROI values.

In Vitro and In Vivo Biocompatibility Of ReOss® in Powder and Putty Configurations

Resumo Este estudo avaliou comparativamente duas configurações (pó e massa) de um biomaterial composto com base de PLGA (Poli(láctico-co-glicólico)/hidroxiapatita em nanoescala (ReOss®, Intra-Lock International) através da morfologia microscópica, citotoxicidade in vitro, biocompatibilidade e resposta in vivo como substituto ósseo. MEV e EDS caracterizaram os biomateriais antes/após o enxerto. A citocompatibilidade foi avaliada em pré-osteoblastos murinos. A osteocondutividade e a biocompatibilidade foram avaliadas em coelhos Branco da Nova Zelândia. Ambas as configurações foram implantadas na calvária de dezoito animais em defeitos não-críticos, com coágulo sanguíneo como grupo controle. Após 30, 60 e 90 dias, os animais foram eutanasiados e os fragmentos contendo os biomateriais e controles coletados. Blocos ósseos foram embebidos em parafina (n=15) destinados às análises histológica e histomorfométrica, e em resina (n=3) destinadas à MEV e EDS. Antes da implantação, a configuração massa mostrou ambas fases morfológicas porosa e fibrosa. O pó revelou partículas porosas com granulometria variável. EDS mostrou cálcio, carbono e oxigênio na configuração massa, enquanto o pó mostrou também fósforo. Após a implantação a EDS revelou cálcio, carbono e oxigênio em ambas configurações. Os materiais foram considerados citotóxicos pelo teste XTT. A análise histológica mostrou nova formação óssea e nenhuma reação inflamatória nos sítios de implante. Entretanto, a análise histomorfométrica indicou que a quantidade de osso neoformado não foi estatisticamente diferente entre os grupos experimentais. Embora ambos os materiais tenham apresentado citotoxicidade in vitro, foram biocompatíveis e osteocondutores. A configuração do ReOss® afetou as características morfológicas e a citocompatibilidade in vitro, porém não impactou a resposta biológica in vivo, como medido pelo presente modelo.
Article This study evaluated comparatively two configurations (powder and putty) of a composite biomaterial based on PLGA (Poly(lactide-co-glycolide)/nanoescale hydroxyapatite (ReOss®, Intra-Lock International) through microscopic morphology, in vitro cytotoxicity, biocompatibility and in vivo response as a bone substitute. SEM and EDS characterized the biomaterials before/after grafting. Cytocompatibility was assessed with murine pre-osteoblasts. Osteoconductivity and biocompatibility were evaluated in White New Zealand rabbits. Both configurations were implanted in the calvaria of eighteen animals in non-critical size defects, with blood clot as the control group. After 30, 60 and 90 days, the animals were euthanized and the fragments containing the biomaterials and controls were harvested. Bone blocks were embedded in paraffin (n=15) aiming at histological and histomorphometric analysis, and in resin (n=3) aiming at SEM and EDS. Before implantation, the putty configuration showed both a porous and a fibrous morphological phase. Powder revealed porous particles with variable granulometry. EDS showed calcium, carbon, and oxygen in putty configuration, while powder also showed phosphorus. After implantation EDS revealed calcium, carbon, and oxygen in both configurations. The materials were considered cytotoxic by the XTT test. Histological analysis showed new bone formation and no inflammatory reaction at implant sites. However, the histomorphometric analysis indicated that the amount of newly formed bone was not statistically different between experimental groups. Although both materials presented in vitro cytotoxicity, they were biocompatible and osteoconductive. The configuration of ReOss® affected morphological characteristics and the in vitro cytocompatibility but did not impact on the in vivo biological response, as measured by the present model.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Estudo longitudinal da infecção por enteropatógenos em bezerros neonatos, com diarreia, sob diferentes estratégias de aleitamento

Foram utilizados 17 bezerros, recém nascidos, da raça Holandesa, com o objetivo de avaliar a influência do volume de sucedâneo nos principais patógenos causadores de diarreia neonatal. Os animais foram distribuídos em dois grupos, 8 bezerros do grupo 1 e 9 bezerros do grupo 2. Os animais foram alimentados duas vezes ao dia totalizando 4 litros de sucedâneo diários para o grupo 1 e 6 litros para o grupo 2. A partir do 1° dia de chegada dos bezerros foram avaliadas as fezes diariamente após o aleitamento da manhã para a classificação das fezes em diarreicas ou não diarreicas. Do primeiro dia de diarreia até o sétimo dia, as fezes foram coletadas em dias alternados (1º, 3º, 5º e 7° dia) diretamente da ampola retal para avaliação dos enteropatógenos. Foram coletadas amostras de sangue dos bezerros com cinco dias de idade para dosagem da proteína total. A média da proteína total foi 6,33 e 6,21g/dL nos grupos 1 e 2 respectivamente. O grupo 2 apresentou tendência (p<0,1) de maior consumo de sucedâneo no período avaliado. A quantidade de sucedâneo oferecida aos animais não influenciou a incidência de diarreia e sua etiologia, ou seja, não foi observada diferença (p>0,05) na frequência das amostras positivas para cada agente entre os grupos. A frequência dos enteropatógenos nas amostras foi de 100 e 75% para Cryptosporidium spp.; 28,5 e 43,7% para Salmonella spp.; 28,5 e 15,6% para patotipos de E. coli; 3,5 e 6,2% para Rotavírus e 10,7 e 9,4% para Giardia sp. nos grupos 1 e 2 respectivamente. Foram encontrados os sorotipos de Salmonella infantis e muenster. Os patotipos de E. coli isolados foram classificados como E. coli enterohemorrágica, enteropatogênica, enterotoxigênica e produtoras de toxinas Shiga 1 e 2. Foi observada associação entre o Cryptosporidium spp. e os patotipos de E. coli em 30% das amostras do grupo 1 e Cryptosporidium spp. e Salmonella spp. em 45,5% no grupo 2. Os resultados do presente trabalho demonstraram que o fornecimento de diferentes volumes de sucedâneo não apresentou influência sobre a incidência e etiologia da diarreia neonatal. A avaliação longitudinal dos enteropatógenos durante o período de patência da diarreia demonstrou que a associação entre eles ocorre a partir do primeiro dia da doença e destacou a importância da infecção pelo Cryptosporidium spp. agente encontrado em todos os momentos e animais.
Seventeen Holstein newborn calves were used with the objective of evaluating the influence of milk replacer volume in the pattern of pathogens causing neonatal diarrhea. The animals were divided into two groups, 8 calves in group 1 and 9 calves in group 2. The calves were fed twice daily in the total of 4 liters of milk replacer to group 1 and 6 liters to group 2. From the 1st day of arrival of the calves feces were evaluated daily after the morning feeding for the classification of diarrheal feces or without diarrhea. The first day of diarrhea until the seventh day, feces were collected on alternate days (1st, 3rd, 5th and 7th day) directly from the rectum to evaluate enteropathogens. Were collected blood samples from calves with five days of age for determination of total protein. The average total protein was 6.33 and 6.21g/dL in groups 1 and 2, respectively. The group 2 tended (p<0.1) higher consumption of milk replacer during the study period. The volume of milk replacer did not influenced the incidence of diarrhea and the frequency of positive samples for each etiologic agent between the two groups (p>0.05). Also, there was no difference (p>0.05) on the pattern of the frequency of positive samples for evaluated pathogens. The frequency of pathogens in the samples was 100 and 75% for Cryptosporidium, 28.5 and 43.7% for Salmonella spp., 28.5 and 15.6% for E. coli pathotypes, 3.5 and 6.2% for Rotavirus and 10.7 and 9.4% for Giardia in groups 1 and 2, respectively. Serotypes of Salmonella infantis and muenster were found. The isolated pathotypes of E. coli isolates were classified as Escherichia coli enteropathogenic, enterotoxigenic and Shiga-toxin-producing 1 and 2. Associations between Cryptosporidium spp. and E. coli pathotypes, and between Cryptosporidium spp. and Salmonella spp. were found in 30% of the samples in group 1 and in 45.5% in group 2, respectively. Our results showed that the different volumes of milk replacer did not influence the incidence and etiology of neonatal diahrrea. Longitudinal evaluation of enteropathogens during patency demonstrated that the association between the etiologic agents starts from the first day of disease. This result highlighted the great importance of the infection by Cryptosporidium spp. which was present in every moments and animals evaluated.

A rapid and simple method for determination of 5-hydroxytryptophan in dietary supplements by capillary electrophoresis

Neste trabalho é apresentado o desenvolvimento de um método rápido e simples, empregando eletroforese capilar, para determinação de 5-hidroxitriptofano (5-HTP) em amostras de suplementos alimentares. O eletrólito de corrida utilizado foi constituído por 20 mmol L-1 de fosfato de sódio (pH 10) e 0,2 mmol L-1 de brometo de cetiltrimetilamônio (CTAB), empregado como inversor de fluxo eletrosmótico. Foram obtidos limites de detecção e quantificação de 3,1 e 10,1 µmol L-1, respectivamente. A curva analítica obtida, para concentrações do analito entre 0-500 µmol L-1, apresentou linearidade com coeficiente de correlação linear (R²) de 0,9995. A precisão instrumental intra-dia (repetibilidade), expressa em desvio padrão relativo (RSD), para a área de pico e tempo de migração do 5-HTP foram de 2,2 e 1,9% (n = 8), respectivamente. O método eletroforético foi empregado na análise de quatro diferentes amostras comerciais de suplementos alimentares e o RSD (n = 3) para as determinações variaram de 2 a 3%. A exatidão do método foi avaliada pela comparação das concentrações de 5-HTP obtidas pelo método proposto e por cromatografia líquida de alta eficiência (HPLC). Os erros relativos variaram entre 0 e 5,9% e não foram observadas diferenças significativas entre as concentrações, ao nível de 95% de confiança.
A rapid and simple capillary electrophoresis (CE) method was developed to determine 5-hydroxytryptophan (5-HTP) in samples of commercial dietary supplements. The optimized background electrolyte (BGE) was composed by 20 mmol L-1 sodium phosphate (pH 10) and 0.2 mmol L-1 cetyltrimethylammonium bromide (CTAB) for electroosmotic flow inversion. The limits of detection (LOD) and quantification (LOQ) for 5-HTP were 3.1 and 10.1 µmol L-1, respectively. The analytical curve demonstrated linearity over a concentration range of 0-500 µmol L-1 with a correlation coefficient (R²) of 0.9995. The intraday instrumental precision (repeatability) as relative standard deviation (RSD) in terms of peak area and migration time of the 5-HTP were 2.2 and 1.9% (n = 8), respectively. The CE method was applied to the analysis of four different samples of commercial dietary supplements and the RSD (n = 3) for the determinations ranged from 2 to 3%. The accuracy of the method was evaluated comparing the measured 5-HTP concentrations to those obtained by a high performance liquid chromatography (HPLC) method. The relative error ranged from 0 to 5.9% (n = 3) and no significant differences between the concentrations were observed at a confidence level of 95%.
