Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

Elevated leptin levels in healthy climacteric women from Northeastern Brazil: an effect of age or adiposity?

ABSTRACT Objective Climacterium is associated with elevated leptin levels and increased risk of cardiovascular disorders. Conflicting data diverge on whether high leptin levels in climacterium reflect increasing adipose mass or, at least partially, age-related hormonal changes. This study addresses this issue in women from a Brazilian state with a low human development index. Subjects and methods A case-control study was conducted, enrolling 136 women from the state of Maranhão, 52 (38.2%) climacteric and 84 (61.8%) non-climacteric. Biometric, biochemical, hormonal and immunological parameters were analyzed. Results Climacteric women showed a moderately increased waist/hip ratio (0.894 versus 0.834, p < 0.05), sustained body mass index (27.46 versus 28.68, p > 0.05) increased leptin levels (9.59 versus 7.13, p < 0.05) and no evidence of metabolic syndrome. No other parameters were altered. The climacteric cohort didn’t show significant body fat gains but displayed a typical age-related redistribution of adipose tissue. Even so, leptin levels were significantly elevated compared with non-climacteric women. Conclusions Altogether, these data support the hypothesis that leptin is elevated, at least partially, as a function of age and climacterium and is not necessarily correlated with metabolic dysfunction and systemic inflammation. Further studies are needed to evaluate the impact of higher leptin levels on postmenopausal women. Arch Endocrinol Metab. 2020;64(3):276-81

RESUMO Objetivo: Validar a quantificação de T-cell receptor excision circles (TRECs) e kappa-deleting recombination circles (KRECs) por reação em cadeia de polimerase (polymerase chain reaction, PCR) em tempo real (qRT-PCR), para triagem neonatal de imunodeficiências primárias que cursam com defeitos nas células T e/ou B no Brasil. Métodos: Amostras de sangue de recém-nascidos (RN) e controles foram coletadas em papel-filtro. O DNA foi extraído e os TRECs e KRECs foram quantificados por reação duplex de qRT-PCR. O valor de corte foi determinado pela análise de Receiver Operating Characteristics Curve, utilizando-se o programa Statistical Package for the Social Sciences (SSPS) (IBM®, Armonk, NY, EUA). Resultados: 6.881 amostras de RN foram analisadas quanto à concentração de TRECs e KRECs. Os valores de TRECs variaram entre 1 e 1.006 TRECs/µL, com média e mediana de 160 e 139 TRECs/µL, respectivamente. Três amostras de pacientes diagnosticados com imunodeficiência grave combinada (severe combined immunodeficiency, SCID) apresentaram valores de TRECs abaixo de 4/µL e um paciente com Síndrome de DiGeorge apresentou TRECs indetectáveis. Os valores de KRECs encontraram-se entre 10 e 1.097 KRECs/µL, com média e mediana de 130 e 108 KRECs/µL, e quatro pacientes com diagnóstico de agamaglobulinemia tiveram resultados abaixo de 4 KRECs/µL. Os valores de corte encontrados foram 15 TRECs/µL e 14 KRECs/µL, e foram estabelecidos de acordo com a análise da Receiver Operating Characteristics Curve, com sensibilidade de 100% para detecção de SCID e agamaglobulinemia, respectivamente. Conclusões: A quantificação de TRECs e KRECs foi capaz de diagnosticar crianças com linfopenias T e/ou B em nosso estudo, validando a técnica e dando o primeiro passo para a implementação da triagem neonatal em grande escala no Brasil.

The cutoff values were 15 TRECs/µL and 14 KRECs/µL and were established according to the receiver operating characteristic curve analysis, with 100% sensitivity for SCID and agammaglobulinemia detection, respectively. Conclusions: Quantification of TRECs and KRECs was able to diagnose children with T- and/or B-cell lymphopenia in our study, which validated the technique in Brazil and enabled us to implement the newborn screening program for SCID and agammaglobulinemia.

Nematodes of Astyanax fasciatus (Actinopterygii: Characidae) and their parasitic indices in the São Francisco river, Brazil

Resumo A fauna endoparasitária de Astyanax fasciatus do alto rio São Francisco foi investigada e os parâmetros ecológicos e os dados morfométricos e morfológicos dos parasitos apresentados. Um total de 74 espécimes de lambaris foi coletado à jusante da barragem de Três Marias, município de Três Marias, Minas Gerais (18°12’32”S, 45°15’41”W), em janeiro de 2011 e de 2012. Onze táxons de Nematoda foram encontrados: Contracaecum sp., Hysterothylacium sp., Goezia sp., Brevimulticaecum sp., Procamallanus sp., Procamallanus (Spirocamallanus) saofranciscencis, Cystidicoloides sp., Spinitectus rodolphiheringi, Rhabdochona sp., Spiroxys sp. e Eustrongylides sp.. A fauna de A. fasciatus foi caracterizada por espécimes larvais de Contracaecum sp., Hysterothylacium sp., Brevimulticaecum sp., Cystidicoloides sp., Spiroxys sp. e por espécimes adultos de P. saofranciscencis, cuja prevalência foi maior que 10%. Assim, o lambari atua como hospedeiro intermediário de algumas espécies de nematoides larvais, especialmente, Anisakidae e Acanthocheilidae (Brevimulticaecum sp., novos hospedeiro e localidade); participa na transmissão de espécies como Rhabdochona sp., para peixes carnívoros, e atua também como hospedeiro definitivo para P. saofranciscencis e S. rodolphiheringi no alto Rio São Francisco.
Abstract The endoparasite fauna of Astyanax fasciatus from the upper São Francisco river was investigated and ecological parameters and morphological and morphometric data on the parasites are presented. A total of 74 specimens of banded astyanax were collected downstream from the Três Marias dam, municipality of Três Marias, Minas Gerais (18°12’32”S, 45°15’41”W) in January 2011 and January 2012. Eleven taxa of Nematoda were found: Contracaecum sp.; Hysterothylacium sp.; Goezia sp.; Brevimulticaecum sp.; Procamallanus sp.; Procamallanus (Spirocamallanus) saofranciscencis; Cystidicoloides sp.; Spinitectus rodolphiheringi; Rhabdochona sp.; Spiroxys sp.; and Eustrongylides sp.. The fauna of A. fasciatus consisted of by larval specimens of Contracaecum sp., Hysterothylacium sp., Brevimulticaecum sp., Cystidicoloides sp., and Spiroxys sp., and by adult specimens of P. saofranciscencis, whose prevalence was greater than 10%. Thus, this fish acts as an intermediate host of some species of larval nematodes especially, Anisakidae and Acanthocheilidae (Brevimulticaecum sp., new host record and new locality). It participates in transmitting species such Rhabdochona sp. to carnivorous fish and also acts as a definitive host for P. saofranciscencis and S. rodolphiheringi in the upper São Francisco river.

Immunomodulatory and toxicological evaluation of the fruit seeds from Platonia insignis, a native species from Brazilian Amazon Rainforest

Abstract The “bacuri” (Platonia insignis Mart., Clusiaceae) is a native tropical fruit from the Brazilian Amazon and Northeast Regions. Its seeds are used to treat inflammatory diseases, diarrhea and skin problems in traditional medical practices. Regarding its widespread medicinal uses, it is important to evaluate the biological and toxicological potential of this species. This way, the aim of this study was to investigate the in vitro cytotoxic and immunomodulatory effects of the hexanic extract of P. insignis seeds, as well as its in vivo acute oral toxicity. The biological evaluation was performed by the determination of cytotoxic (MTT and hemolysis assay) and immunomodulatory (phagocytic capacity, lysosomal volume and nitrite production) activities of EHSB in murine peritoneal macrophages. In addition, the oral acute toxicity was evaluated using female Wistar rats treated with EHSB (2.0 g/kg), in accordance with the OECD 423 Guideline. The EHSB showed low toxicity for macrophages in the MTT test (CC50 value: 90.03 µg/ml), as well as for erythrocytes, which caused only 2.5% hemolysis at the highest concentration. A strong immunomodulatory activity was observed by a markedly increase of the NO production, phagocytic ability and lysosomal volume. On the other hand, it was not observed deaths or changes in the clinical and behavioral parameters in the toxicological evaluation. This manner, the present study contributes to the knowledge about the immunomodulatory and toxicological properties of the P. insignis. This may provide perspectives for the evaluation and development of effective and safe phytomedicines created from the Brazilian local biodiversity.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Inhibition of carrageenan-induced expression of tissue and plasma prekallikreins mRNA by low level laser therapy in a rat paw edema model

CONTEXTUALIZAÇÃO: A laserterapia de baixa potência tem sido usada para o tratamento de processos inflamatórios diversos em que a calicreína tecidual e a plasmática possuem participação ativa. A pré-calicreína plasmática (PPK) é sintetizada pelos hepatócitos e secretada na corrente sanguínea, onde participa da ativação da coagulação, fibrinólise, geração de cininas e inflamação. A pré-calicreína tecidual está associada com importantes doenças (incluindo câncer, inflamação e neurodegeneração) e tem sido utilizada ou sugerida clinicamente como importante biomarcador ou alvo terapêutico. OBJETIVO: Avaliar se a laserterapia altera a expressão gênica da pré-calicreína tecidual e da plasmática no modelo de inflamação aguda induzida pela carragenina. MÉTODOS: Quarenta ratos foram separados em quatro grupos experimentais: A1 (controle), A2 (carragenina, apenas), A3 (laser 660nm, apenas) e A4 (Carragenina + laser 660nm). O edema foi medido por um pletismômetro. Tecido subplantar foi coletado para a quantificação de RNA mensageiro (RNAm) de pré-calicreínas tecidual e plasmática por PCR em tempo real. RESULTADOS: Observou-se uma diminuição significativa no volume de edema após irradiação com laser 660nm. A expressão de RNAm de pré-calicreínas tecidual e plasmática aumentou após a inoculação de carragenina, entretanto a expressão gênica das pré-calicreínas diminuiu significantemente após laserterapia de baixa potência de 660nm. CONCLUSÃO: Os resultados sugerem que a expressão de RNAm das pré-calicreínas tecidual e plasmática é modulada pela laserterapia de baixa potência, podendo ser alvo terapêutico para tratamento de processos inflamatórios.
BACKGROND: Low level laser therapy (LLLT) has been used clinically in order to treat inflammation, where tissue and plasma prekallikrein have crucial importance. Plasma prekallikrein (PPK) is synthesized by the hepatocytes and secreted into the bloodstream, where it participates in the surface-dependent activation of blood coagulation, fibrinolysis, kinin generation and inflammation. Tissue prekallikrein is associated with important disease states (including cancer, inflammation, and neurodegeneration) and has been utilized or proposed as clinically important biomarker or therapeutic target of interest. OBJECTIVE: To evaluate if LLLT modulates tissue and plasma prekallikreins mRNA expression in the carrageenan-induced rat paw edema. METHODS: Experimental groups were assigned as followed: A1 (Control-saline), A2 (Carrageenan-only), A3 (laser 660nm only) and A4 (Carrageenan + laser 660nm). Edema was measured by a plethysmometer. Subplantar tissue was collected for the quantification of prekallikreins mRNA by Real time-Polymerase Chain Reaction. RESULTS: A significantly decrease in the edema was observed after laser irradiation. Expression of prekallikreins increased after carrageenan injection. Tissue and plasma prekallikrein mRNA expression significantly decreased after LLLT's 660nm wavelength. CONCLUSION: These results suggest that expression of tissue and plasma prekallikreins is modulated by LLLT, which can be used in clinical practice due to its anti-inflammatory effects.

Siolmatra brasiliensis (Cogn.) Baill., Cucurbitaceae, popularmente conhecida como "pluméria" ou "taiuiá" é utilizada na medicina popular brasileira para diversos fins terapêuticos. O estudo de toxicidade aguda do extrato bruto etanólico (EBE) de Siolmatra brasiliensis foi investigado em camundongos. Nenhuma mortalidade ou sinais de toxicidade foram observados nas doses de 10 e 100 mg/kg, entretanto em doses administradas de 1000 e 2000 mg/kg levou as diversas alterações comportamentais e mortalidade. A DL50 para o EBE foi de 1000 mg/kg. Análise macroscópica dos órgãos demonstrou alterações morfológicas no coração dos animais tratados com 1000 e 2000 mg/kg. Por meios destes resultados conclui-se que o EBE de Siolmatra brasiliensis é seguro em doses de 10 and 100 mg/kg e apresentou toxicidade nas doses de 1000 e 2000 mg/kg.
Siolmatra brasiliensis (Cogn.) Baill., Cucurbitaceae, commonly known as "pluméria" or "taiuiá", is widely used in different ways in Brazilian popular medicine to treat several diseases. Acute toxicity of Siolmatra brasiliensis crude ethanolic extract (CEE) was investigated in mice. No mortality or signs of CEE toxicity were observed at the doses of 10 and 100 mg/kg bw, but the administration of 1000 and 2000 mg/kg bw caused several adverse behavioral effects and mortality. Macroscopic inspection of the organs showed morphologic alterations in the heart of animals treated with doses of 1000 and 2000 mg/kg bw. According to our results, S. brasiliensis CEE has an LD50 of 1000 mg/kg bw. We conclude that S. brasiliensis CEE was safe at the doses of 10 and 100 mg/kg bw and presented toxicity at the doses of 1000 and 2000 mg/kg bw.

Neste trabalho foi investigado o efeito do extrato hidroalcoólico de própolis (EHP) de Scaptotrigona aff. postica sobre o desenvolvimento do tumor de Ehrlich na forma sólida, sobre a celularidade dos órgãos linfóides dos animais portadores de tumor, bem como, sobre a produção de óxido nítrico (NO) pelos macrófagos destes animais. Camundongos Swiss foram divididos em quatro grupos: controle, EHP 0,5; EHP 5 e EHP 50, os quais foram tratados por via intraperitoneal com dose única de solução salina (NaCl 0,9%); 0,5; 5 ou 50 mg de EHP/kg de animal, respectivamente. Depois de 48 h do tratamento, os animais foram inoculados com 10(5) células do tumor de Ehrlich nas patas. Os resultados mostraram que o tratamento com EHP nas doses de 5 e 50 mg/kg inibiu de forma significativa o desenvolvimento do tumor a partir do 6º dia pós-inóculo quando comparado ao controle e ao EHP 0,5. Além disso, houve aumento significativo da celularidade do baço e da medula óssea nos grupos EHP 0,5 e EHP 5 em relação ao controle. A produção de NO estimulada com concanavalina A (ConA) apresentou uma significante diminuição nos grupos tratados com EHP em relação ao controle. Pode-se concluir que o tratamento com EHP apresentou efeito antitumoral quando administrado nas doses de 5 e 50 mg/kg, o que pode estar relacionado com a sua composição química e com a inibição da produção de NO.
It was investigated the effect of hydroalcoholic extract (HEP) of propolis from Scaptotrigona aff. postica on the solid Ehrlich tumor, on the tumor-bearing mice lymphoid organs and on the nitric oxide (NO) production. Swiss mice were divided in 4 groups: control, HEP 0.5; HEP 5 and HEP 50 that was treated by intraperitoneal route with a single dose of saline solution (NaCl 0.9%) or 0.5 or 5 or 50 mg of HEP/kg body weight, respectively. After 48 h of treatment, the animals were inoculated with 10(5) tumor cells in their footpad. The results showed that the treatment with HEP in the doses of 5 and 50 mg/kg inhibited the development of the tumor from the 6th day post inoculums when compared to the control and to the HEP 0.5 groups. Besides, there was an increase of spleen and bone marrow cell number in HEP 0.5 and HEP 5 as compared to the control. Concanavalin A (ConA)-stimulated NO production was decreased in all HEP-treated groups when compared to the control. In conclusion, the treatment with HEP had an anti-tumor effect what may be related to its chemical composition and to the inhibition of NO production.

O extrato etanólico das folhas de Paullinia elegans Cambess., Sapindaceae, e as frações n-hexano, clorofórmio, acetato de etila e hidroetanólica, obtidas de seu fracionamento, foram avaliadas quanto às suas atividades anti-edematogênica e sequestradora de radicais livres. O extrato etanólico e a fração hexano produziram inibição significativa (74,4 e 76,0%, respectivamente) do edema da orelha induzido pelo óleo de cróton em ratos, em doses de 5 mg/orelha. As frações acetato de etila e hidroetanólica mostraram atividade sequestradora de radicais livres no ensaio de DPPH, com IC50 de 36,7 e de 30,1 µg/mL, respectivamente. O fracionamento dos extratos pelo uso de métodos cromatográficos resultou no isolamento do epifriedelanol, ácido 3-O-acetil oleanólico, mistura do stigmasterol 3-β-O-glucopiranosídeo e sitosterol 3-β-O-glucopiranosídeo, canferol, canferol 3,7-O-α-diramnopiranosídeo, canferol 3-O-α-ramnopiranosídeo e 2-O-metil-chiro-inositol. Os compostos foram identificados com base na comparação de seus dados espectroscópicos de RMN com os da literatura.
Ethanol extract of the leaves of Paullinia elegans Cambess., Sapindaceae, and its hexane, chloroform, ethyl acetate, and hydroethanol fractions were evaluated for their antiedematogenic and free radical scavenging activities. The ethanol extract and the hexane fraction produced statistically significant inhibition (74.4 and 76.0%, respectively) of the ear edema induced by croton oil in mice, observed at doses of 5 mg/ear. The ethyl acetate and hydroethanol fractions showed significant radical scavenging effect in the DPPH assay, with IC50 of 36.7 and 30.1 µg/mL, respectively. Fractionation of the extracts through chromatographic methods afforded epifriedelanol, oleanolic acid 3-O-acetyl, a mixture of stigmasterol 3-β-O-glucopyranoside and sitosterol 3-β-O-glucopyranoside, kaempferol 3,7-O-α-dirhamnopyranoside, kaempeferol-3-O-α-rhamnopyranoside and 2-O-methyl-chiro-inositol. The compounds were identified on the basis of their NMR spectral data and comparison with those of literature.

Na busca de novos agentes leishmanicidas, avaliamos em culturas de promastigotas de Leishmania amazonensis o efeito dos extratos hidroalcoólicos das folhas de Tephrosia cinerea (L.) Pers. (Fabaceae), Dichorisandra sp (Commelinaceae), Syzygium jambolanum DC. (Myrtaceae), Julocroton triqueter (Lam.) Didr. var. triqueter (Euphorbiaceae), Passiflora edulis Sims(Passifloraceae), Cecropia sp (Cecropiaceae), Chenopodium ambrosioides L.(Chenopodiaceae), Pedilanhus tithymaloides (L.) Poit (Euphorbiaceae), Peristrophe angustifolia Nees(Acanthaceae) e o extrato aquoso do mesocarpo de Orbignya phalerata Mart. (Arecaceae). As promastigotas de Leishmania amazonensis foram cultivadas em presença de 31,3; 63,5; 125,0; 250,0 e 500,0 µg/mL dos extratos por 24 horas. Ao final desse período foi calculada a concentração inibitória do crescimento (CI50) em relação às culturas não tratadas com os extratos. Os extratos das espécies J. triqueter, Dichorisandra sp e T. cinerea apresentaram maior eficácia em induzir a morte das promastigotas, com CI50 de 29,5; 32,9 e 43,6 µg/mL, respectivamente. P. edulis, C. ambrosioides e S. jambolanum apresentaram eficácia moderada com CI50 de 150,1; 151,9 e 166,6 µg/mL, respectivamente. P. tithymaloides e O. phalerata apresentaram baixo efeito leishmanicida com CI50 >500 µg/mL, enquanto Peristrophe angustifolia e Cecropia spnão apresentaram efeito. Dessa forma, dos dez extratos testados, três apresentaram uma expressiva atividade leishmanicida in vitro.
Searching for new leishmanicidal agents, promastigotes forms of L. amazonensis were cultured with the hydroalcoholic extracts obtained from the Tephrosia cinerea (L.) Pers. (Fabaceae), Dichorisandra sp (Commelinaceae), Syzygium jambolanum DC. (Myrtaceae), Julocroton triqueter (Lam.) Didr. var. triqueter (Euphorbiaceae), Passiflora edulis Sims(Passifloraceae), Cecropia sp (Cecropiaceae), Chenopodium ambrosioides L.(Chenopodiaceae), Pedilanhus tithymaloides (L.) Poit (Euphorbiaceae), Peristrophe angustifolia Nees(Acanthaceae) leaves and the aqueous extract obtained from the Orbignya phalerata Mart. (Arecaceae) mesocarp flour. The in vitro assay was performed with promastigotes incubated during 24 hours with 31.3, 62.5, 125.0, 250.0 and 500.0 µg/mL of each extract. Then the inhibitory concentration of the parasite growth (IC50) was determined. The effectiveness of J. triqueter, Dichorisandra sp and T. cinerea hydroalcoholic extracts to induce promastigotes death was intense since the IC50 were 29.5; 32.9 and 43.6 µg/mL, respectively. P. edulis, C. ambrosioides and S. jambolanum extracts had moderated effectiveness since the IC50 were 150.1; 151.9 and 166.6 µg/mL, respectively. P. tithymaloides and O. phalerata extracts showed a low efficacy in comparison with IC50 >500 µg/mL. Peristrophe angustifolia and Cecropia sp extracts had no leishmanicidal effect. Thus, three of the ten extracts that were tested showed a significant in vitro leishmanicidal activity.

A classificação para linfomas não-Hodgkin (LNH) proposta pela Organização Mundial da Saúde (OMS) enfatiza a importância do imunofenótipo para o diagnóstico. O objetivo deste estudo foi avaliar a utilidade da citologia combinada a citometria de fluxo para o diagnóstico de LNH, utilizando um painel de anticorpos monoclonais e estudo do ciclo celular. O material foi obtido através de aspiração de linfonodos por agulha fina de 78 pacientes. O painel de anticorpos monoclonais para análise em citometria de fluxo foi o seguinte: CD19/CD10, CD20/CD5, CD23, CD38/CD7, CD3/CD4, CD3/CD8, kappa/lambda. O diagnóstico final foi confirmado pela histologia convencional, considerada gold standard. Em 85% dos casos a citologia associada a imunofenotipagem e porcentagem de células em fase S permitiram um diagnóstico correto. Nos demais casos foi possível diferenciar linfomas B ou T e estimar grau de agressividade. O painel, embora pequeno, foi suficiente exceto para os anaplásicos e subclassificação dos linfomas T. Nestes casos, a morfologia foi mais importante que imunofenótipo, sendo este seguro apenas para linfomas linfoblásticos. A fração de fase S mostrou-se importante para diferenciar linfomas indolentes e de alto grau. Concluímos que esta técnica é uma boa alternativa para o diagnóstico de linfomas não-Hodgkin. Permite um diagnóstico rápido, menos invasivo, podendo ser repetida quando necessário, agilizando o tratamento.
The WHO classification of non-Hodgkin's lymphoma stresses the importance of the immunophenotype for diagnosis. The aim of our study was to evaluate the use of cytology together with flow cytometric examination using a panel of monoclonal antibodies including DNA S phase analysis. Material was obtained from lymph node aspiration of 78 patients. The panel for flow cytometric analysis comprised: CD19/CD10, CD20/CD5, CD23, CD3/CD4, CD3/CD8, CD38/CD7, kappa/lambda. The final diagnosis was confirmed by lymph node biopsy. In 85% of cases, cytology combined with immuno-phenotyping and percentage of Phase S cells allowed the correct diagnosis. In the remaining cases it was possible to differentiate T or B lymphomas and estimate their aggressiveness. The panel, although small, was sufficient in all cases except for anaplastic lymphoma. S-phase fraction was important for the diagnosis of large B-cell NHL vs. Follicular NHL. In cases of T-cell lymphomas a reliable diagnosis was only possible for lymphoblastic lymphomas. In conclusions, combined cytology and cytophotometric diagnosis of lymph node aspirations is a good alternative to histologic examination, except for T-cell lymphomas. In contrast to biopsy this method is less invasive and may be repeated if necessary.
