Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

This is the first report by the COVID19 Observatory - Group: Contagion Networks analyzing mortality data from the city of São Paulo. In this report, we integrated mortality data for the city of São Paulo between 04/02/2020 and 04/28/2020, with information on the flow of victims between hospitals and cemeteries/crematoriums. We included in our analyzes both confirmed and suspected deaths from COVID-19. The main objectives of this report were: (1) to describe the structure of the flow of victims between locations and (2) to suggest changes in the current flow based on geographical distances in order to avoid a potential overload of the mortuary system. We suggest that the city of São Paulo should plan for a potential overload of the mortuary system (that is, the number of burials), based on the presented results. Thus, our results reinforce the need to adopt specific planning for the management of the extraordinary number of victims of this pandemic. Our predictions are based on the structural analysis of the COVID-19 victim flow network, which shows several hotspots with high vulnerability to system overload. These hotspots concentrate with either the greatest number of deaths (hospital) or of burials (cemetery or crematorium), and therefore have high potential to become overwhelmed by receiving many bodies due to the increase in victims of the pandemic. We recommend special attention to be given to localities on the east side of São Paulo, which has both the most vulnerable hospitals in the city, and also houses cemeteries and crematoriums that have a central role in the network and / or are vulnerable. Based on our optimization analysis, we suggest logistical changes in the current flow of bodies from hospitals to cemeteries/crematoriums so as not to overload the funeral system and minimize transportation costs. In this sense, our results are potentially useful for improving the operational planning of the Municipality of São Paulo, ratifying or rectifying actions underway at the municipal level.
Este é o primeiro relatório do Observatório COVID19 - Grupo: Redes de Contágio analisando os dados de óbitos da cidade de São Paulo. Neste relatório, integramos os dados de óbitos da cidade de São Paulo entre os dias 02/04/2020 e 28/04/2020 com informações sobre o fluxo de vítimas entre os hospitais e os cemitérios e crematórios da cidade de São Paulo. Incluímos em nossas análises óbitos confirmados e óbitos suspeitos de COVID-19. Os principais objetivos deste relatório são: (1) descrever a estrutura do fluxo de vítimas entre localidades e (2) sugerir mudanças no fluxo com base em distâncias geográficas de maneira a evitar uma potencial sobrecarga do sistema funerário. Sugere-se à prefeitura da cidade de São Paulo que seja realizado um planejamento para uma potencial sobrecarga do sistema funerário (isto é, número de sepultamentos) da cidade de São Paulo com base nos resultados apresentados. Desta forma, nossos resultados reforçam a necessidade de ser adotado planejamento específico para a gestão dos casos extraordinários visualizados no contexto da pandemia. Esta previsão está baseada na análise estrutural da rede de fluxos de vítimas da COVID-19, que indica a concentração de vários locais com alta vulnerabilidade à sobrecarga do sistema. Tais locais concentram a maior quantidade de óbitos (hospitais) ou a maior concentração de sepultamentos (cemitérios ou crematórios) e tem portanto alto potencial de tornarem-se sobrecarregados por receberem muitos corpos devido ao aumento de vítimas da pandemia. Recomenda-se especial atenção à localidades da zona leste de São Paulo, que apresenta os hospitais mais vulneráveis da cidade e abriga cemitérios e crematórios que possuem papel central na rede e/ou encontram-se vulneráveis. Com base em nossa análise de otimização, sugerimos mudanças logísticas no atual fluxo de corpos de hospitais para cemitérios/crematórios de modo a não sobrecarregar o sistema funerário e minimizar os custos de transporte. Neste sentido, nossos resultados são potencialmente úteis ao aperfeiçoamento do planejamento operacional da Prefeitura Municipal de São Paulo, ratificando ou retificando ações em curso no âmbito municipal.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Rosmarinus officinalis, pertencente à família Lamiaceae, é um exemplar da flora medicinal que possui propriedades terapêuticas. No intuito de usufruir destes benefícios, desenvolveu-se uma formulação de enxaguatório bucal com seleção criteriosa de matérias-primas que atendessem os requisitos farmacotécnicos. Incorporaram-se extratos dessa planta e verificou-se a capacidade inibitória in vitro frente a micro-organismos frequentemente encontrados em periodontias. Controles foram efetuados para a avaliação da qualidade dos fármacos, quantificando-se como marcadores químicos, os fenóis e os flavonóides. Formulações de enxaguatórios contendo 0,1, 5 e 10% de extrato etanólico de R. officinalis; e 0,05, 5 e 10% da fração hexânica de R. officinalis foram preparadas. Para avaliar o sinergismo, o extrato e a fração hexânica também foram adicionados às formulações que continham fluoreto de sódio 0,05% e diguclonato de clorexidina 0,12%. Nessas formulações avaliou-se a capacidade inibitória frente a micro-organismos específicos do processo de formação de placa bacteriana, S. mutans(ATCC 25175) e C. albicans (ATCC 10231), frequentemente encontrada em quadros de infecções orais. Foi empregado o método de difusão em ágar para a avaliação da atividade inibitória dos extratos e das formulações. Todos os enxaguatórios demonstraram atividade inibitória, verificando-se maior sensibilidade a S. mutans,quando se utilizou extrato etanólico 5% + fluoreto de sódio 0,05% e sensibilidade maior a C. albicans,quando se utilizou fração hexânica a 10%. Os resultados foram caracterizados pelo aparecimento de halo de inibição de crescimento, justificando a utilização e associação dos extratos de R. officinalis.
Rosmarinus officinalis, which belongs to the Lamiaceaefamily, is a species of medicinal flora with therapeutic properties. In order to exploit the benefits of these properties, a mouthwash formulation was developed, with careful selection of raw materials to meet pharmacotechnical requirements. Extracts of the plant were incorporated into a mouthwash, which was shown to have inhibitory action in vitro against the micro-organisms commonly found in periodontics. Controls for assessing the quality of the drugs were carried out, quantifying phenols and flavonoids as chemical markers. Mouthwash solutions were formulated containing 0.1, 5 and 10% ethanol extract of R. officinalis; and 0.05, 5 and 10% of the hexane fraction of R. officinalis. In order to evaluate synergism, ethanol extract and hexane fraction were also added to formulations containing 0.05% sodium fluoride and 0.12% chlorhexidine digluconate. These formulations were assessed for inhibitory effect against the specific microorganisms involved in the process of bacterial plaque formation, S. mutans(ATCC25175) and C. albicans(ATCC 10231), frequently found in cases of oral infections. The agar diffusion method was used to evaluate the inhibitory activity of extracts and formulations. All mouthwash solutions displayed inhibitory activity having higher sensitivity to S. mutansfor the 5% ethanol extract+0.05% sodium fluoride, and greater sensitivity to C. albicansfor the 10% hexane fraction. Results were characterized by the appearance of a growth inhibition halo, justifying the utilization and association of extracts of R. officinalis.

Insecticide effects of Ruta graveolens, Copaifera langsdorffii and Chenopodium ambrosioides against pests and natural enemies in commercial tomato plantation

O objetivo deste trabalho foi avaliar o efeito inseticida dos extratos aquosos de folhas de Ruta graveolens (Rutaceae) e alcoólicos de folhas de Copaifera langsdorffii (Caesalpinaceae) e de folhas de Chenopodium ambrosioides (Chenopodiaceae) a 5%. O experimento foi em blocos casualizados com seis repetições. As parcelas tratadas tiveram redução na população de pragas, quando comparadas às parcelas sem nenhum tratamento. O extrato elaborado com C. langsdorffii apresentou maior efeito inseticida em Bemisia tabaci (Hemiptera: Aleyrodidae) e soma das pragas. Verificou-se que após 24h de pulverização, as parcelas tratadas com o extrato de C. ambrosioides apresentaram menores números de adultos de Tuta absoluta (Lepidoptera: Gelechiidae) seguido pelas parcelas tratadas com extrato de R. graveolens. Foram observados menores números do parasitoide de ovos de lepidópteros Trichogramma sp. (Hymenoptera: Trichogrammatidae) e da soma dos inimigos naturais (predadores + parasitoides) nos tratamentos que receberam as pulverizações com os extratos das plantas de C. langsdorffii e C. ambrosioides seguido por R. graveolens do que na testemunha; do parasitoide da família Eulophidae (Hymenoptera), que atacam lagartas de T. Absoluta em plantas tratadas com R. graveolens, seguido por C. langsdorffii e C. ambrosioides do que na testemunha.
The aim of this study was to evaluate the insecticide effect of watery leaf extracts of Ruta graveolens (Rutaceae), alcoholic leaf extracts of Copaifera langsdorffii (Caesalpinaceae) and Chenopodium ambrosioides (Chenopodiaceae) in the concentration of 5% under field conditions. The experiment design was randomized blocks with six replications. The parcels treated with plant extracts showed reduction in the population of pests when compared with the control parcels. The extract elaborated with C. langsdorffii presented greater insecticidal effect under Bemisia tabaci (Hemiptera: Aleyrodidae) and sum of pests. It was verified that after 24 hours of spraying, the parcels treated with the extract of C. ambrosioides presented minor numbers of adults of Tuta absoluta (Lepidoptera: Gelechiidae), followed by the parcels treated with extract of R. graveolens. There were smaller numbers of parasitoid eggs of lepidopterans Trichogramma sp. (Hymenoptera: Trichogrammatidae) and sum of natural enemies (predators + parasitoids) in the parcels that had received spraying with extracts from the plants of C. langsdorffii and C. ambrosioides, followed by R. graveolens, compared to the control. There were a smaller number of parasitoids from the family Eulophidae (Hymenoptera) attacking caterpillars of T. absoluta in plants treated with R. graveolens, followed by C. langsdorffii and C. ambrosioides than in the control.

Siolmatra brasiliensis (Cogn.) Baill., Cucurbitaceae, popularmente conhecida como "pluméria" ou "taiuiá" é utilizada na medicina popular brasileira para diversos fins terapêuticos. O estudo de toxicidade aguda do extrato bruto etanólico (EBE) de Siolmatra brasiliensis foi investigado em camundongos. Nenhuma mortalidade ou sinais de toxicidade foram observados nas doses de 10 e 100 mg/kg, entretanto em doses administradas de 1000 e 2000 mg/kg levou as diversas alterações comportamentais e mortalidade. A DL50 para o EBE foi de 1000 mg/kg. Análise macroscópica dos órgãos demonstrou alterações morfológicas no coração dos animais tratados com 1000 e 2000 mg/kg. Por meios destes resultados conclui-se que o EBE de Siolmatra brasiliensis é seguro em doses de 10 and 100 mg/kg e apresentou toxicidade nas doses de 1000 e 2000 mg/kg.
Siolmatra brasiliensis (Cogn.) Baill., Cucurbitaceae, commonly known as "pluméria" or "taiuiá", is widely used in different ways in Brazilian popular medicine to treat several diseases. Acute toxicity of Siolmatra brasiliensis crude ethanolic extract (CEE) was investigated in mice. No mortality or signs of CEE toxicity were observed at the doses of 10 and 100 mg/kg bw, but the administration of 1000 and 2000 mg/kg bw caused several adverse behavioral effects and mortality. Macroscopic inspection of the organs showed morphologic alterations in the heart of animals treated with doses of 1000 and 2000 mg/kg bw. According to our results, S. brasiliensis CEE has an LD50 of 1000 mg/kg bw. We conclude that S. brasiliensis CEE was safe at the doses of 10 and 100 mg/kg bw and presented toxicity at the doses of 1000 and 2000 mg/kg bw.

Neste trabalho foi investigado o efeito do extrato hidroalcoólico de própolis (EHP) de Scaptotrigona aff. postica sobre o desenvolvimento do tumor de Ehrlich na forma sólida, sobre a celularidade dos órgãos linfóides dos animais portadores de tumor, bem como, sobre a produção de óxido nítrico (NO) pelos macrófagos destes animais. Camundongos Swiss foram divididos em quatro grupos: controle, EHP 0,5; EHP 5 e EHP 50, os quais foram tratados por via intraperitoneal com dose única de solução salina (NaCl 0,9%); 0,5; 5 ou 50 mg de EHP/kg de animal, respectivamente. Depois de 48 h do tratamento, os animais foram inoculados com 10(5) células do tumor de Ehrlich nas patas. Os resultados mostraram que o tratamento com EHP nas doses de 5 e 50 mg/kg inibiu de forma significativa o desenvolvimento do tumor a partir do 6º dia pós-inóculo quando comparado ao controle e ao EHP 0,5. Além disso, houve aumento significativo da celularidade do baço e da medula óssea nos grupos EHP 0,5 e EHP 5 em relação ao controle. A produção de NO estimulada com concanavalina A (ConA) apresentou uma significante diminuição nos grupos tratados com EHP em relação ao controle. Pode-se concluir que o tratamento com EHP apresentou efeito antitumoral quando administrado nas doses de 5 e 50 mg/kg, o que pode estar relacionado com a sua composição química e com a inibição da produção de NO.
It was investigated the effect of hydroalcoholic extract (HEP) of propolis from Scaptotrigona aff. postica on the solid Ehrlich tumor, on the tumor-bearing mice lymphoid organs and on the nitric oxide (NO) production. Swiss mice were divided in 4 groups: control, HEP 0.5; HEP 5 and HEP 50 that was treated by intraperitoneal route with a single dose of saline solution (NaCl 0.9%) or 0.5 or 5 or 50 mg of HEP/kg body weight, respectively. After 48 h of treatment, the animals were inoculated with 10(5) tumor cells in their footpad. The results showed that the treatment with HEP in the doses of 5 and 50 mg/kg inhibited the development of the tumor from the 6th day post inoculums when compared to the control and to the HEP 0.5 groups. Besides, there was an increase of spleen and bone marrow cell number in HEP 0.5 and HEP 5 as compared to the control. Concanavalin A (ConA)-stimulated NO production was decreased in all HEP-treated groups when compared to the control. In conclusion, the treatment with HEP had an anti-tumor effect what may be related to its chemical composition and to the inhibition of NO production.

Este estudo descreve uma modificação do método de parcelas de 0.1 ha desenvolvido por Gentry, que nós adaptamos para sítios de pesquisa ecológica de longa-duração na Amazônia brasileira. Nosso objetivo foi desenvolver um método que fosse apropriado para pesquisas ecológicas de longa-duração (componente PELD), mas que permitissem inventários rápidos para avaliação da complementaridade biótica e planejamento do uso da terra na Amazônia (componente RAP). A filosofia por trás do método é a de maximizar a probabilidade de amostrar adequadamente as comunidades biológicas, para o que são necessárias áreas amostrais grandes, e ao mesmo tempo minimizar a variação nos fatores abióticos que afetam estas comunidades, o que requer amostragem de áreas pequenas. Para conseguir isto, as parcelas são longas e estreitas, com seu maior eixo orientado ao longo das curvas de nível do terreno. Este desenho minimiza a variação interna de topografia e solo em cada parcela, e permite o uso destas variáveis como preditoras das distribuições de espécies. O desenho pode ser usado para vários taxa e estágios de vida, ajustando a largura da parcela ou distribuindo sub-amostras ao longo da parcela. As parcelas são distribuídas sistematicamente em uma grade de trilhas, que é usada para amostrar taxa que não podem ser amostrados em parcelas pequenas. Cada grade de trilhas e parcelas constitui um sítio de pesquisa. A distribuição sistemática de parcelas na paisagem permite estimativas não tendenciosas da distribuição, abundância e biomassa das espécies em cada sítio, e comparações biogeográficas entre sítios. Os custos de implementação são moderados, considerando a grande produção gerada pelos estudos integrados.
Our objectives were to develop a method that would be appropriate for long-term ecological studies, but that would permit rapid surveys to evaluate biotic complementarity and land-use planning in Amazonia. The Amazon basin covers about 7 million km². Therefore, even a sparse coverage, with one sample site per 10.000 km², would require about 700 sampling sites. Financial considerations limit the number of sites and investment at each site, but incomplete coverage makes evaluation of biotic complementarity difficult or impossible (Reddy & Dávalos 2003). Our next challenge is to install similar systems throughout Amazonia. The cost, based on modification of Al Gentry's original design is moderate (less than US$ 50.000 per site if it is not necessary to immediately identify all vascular plants in plots) and we can obtain RAP results for most taxa in the short term at much lower cost. However, biological surveys will only be relevant if the local people participate and the surveys serve as much to teach the local communities about the value of their natural resources as they serve to teach the international community about biodiversity. Therefore, we want to see each site run as a long-term ecological research project by local people and institutions. Biological surveys are an important tool in land-use planning, but only the local people can implement those plans.
