Abstract Schinus terebinthifolia Raddi green fruits essential oil (EO) was evaluated regarding its phytochemical profile, antimicrobial and cytotoxic activities, and toxicity. Gas chromatography with mass spectrometry was applied to identify its constituents, thereafter the minimum inhibitory concentration, minimum bactericidal and fungicidal concentrations, and its antibiofilm activity were evaluated. The EO cytotoxicity was assessed in tumor and non-tumor human cells, and in vivo toxicity was evaluated in a Galleria mellonella model. The major constituents of S. terebinthifolia EO were alpha-phellandrene and beta-phellandrene. The EO had a weak activity against all strains of Candida albicans (MIC 1000μg/mL) and had no activity against non-albicans strains, bacteria, and C. albicans biofilm. Cytostatic activity against all tumor cell lines was shown. Additionally, cell viability remained at EO concentrations up to 62.5 μg/mL. At 16 mg/mL, 50% hemolysis was observed, and it had low toxicity in vivo. Overall, the S. terebinthifolia EO was characterized by low antimicrobial and antibiofilm activities, with no evidence of toxicity to human tumor and non-tumor cells.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

ABSTRACT Reinfection by the severe acute respiratory syndrome coronavirus type 2 (SARS-COV-2) has been reported in many countries, suggesting that the virus may continue to circulate among humans despite the possibility of local herd immunity due to massive previous infections. The emergence of variants of concern (VOC) that are more transmissible than the previous circulating ones has raised particular concerns on the vaccines effectiveness and reinfection rates. The P.1 lineage was first identified in December 2020 in Manaus city and is now globally spread. We report the first case of reinfection of SARS-CoV-2 caused by the P.1 variant outside of Manaus. The potential of these new variants to escape naturally and vaccine- induced immunity highlights the need for a global vigilance.

Resumo: Embora se reconheça que depressão e ansiedade resultem em incapacidades, bem como em prejuízos laborais e para os sistemas de saúde, pesquisas de base populacional são escassas no Brasil. Este estudo avaliou a prevalência de transtornos mentais em adolescentes, jovens e adultos e sua relação com características sociodemográficas em cinco coortes de nascimento (RPS): Ribeirão Preto (São Paulo), Pelotas (Rio Grande do Sul) e São Luís (Maranhão), Brasil. Episódio depressivo, risco de suicídio, fobia social e transtorno de ansiedade generalizada foram avaliados usando-se o Mini International Neuropsychiatric Interview. Intervalos de confiança bootstrap foram estimados e prevalências estratificadas por sexo e nível socioeconômico no programa R. Foram incluídos 12.350 participantes das coortes. Episódio depressivo maior atual foi mais prevalente em adolescentes de São Luís (15,8%; IC95%: 14,8-16,8) e nos adultos de Ribeirão Preto (12,9%; IC95%: 12,0-13,9). As maiores prevalências para risco de suicídio ocorreram nos adultos de Ribeirão Preto (13,7%; IC95%:12,7-14,7), fobia social e ansiedade generalizada nos jovens de Pelotas com 7% (IC95%: 6,3-7,7) e 16,5% (IC95%: 15,4-17,5), respectivamente. As menores prevalências de risco de suicídio ocorreram nos jovens de Pelotas (8,8%; IC95%: 8,0-9,6), fobia social nos jovens de Ribeirão Preto (1,8%; IC95%: 1,5-2,2) e ansiedade generalizada nos adolescentes de São Luís (3,5%; IC95%: 3,0-4,0). Em geral, os transtornos mentais foram mais prevalentes nas mulheres e naqueles com menor nível socioeconômico, independentemente do centro e idade, reforçando a necessidade de maior investimento em saúde mental no Brasil, sem desconsiderar determinantes de gênero e socioeconômicos.

Pterodactyloid pterosaur bones from Cretaceous deposits of the Antarctic Peninsula

Abstract Fossil vertebrates from Antarctica are considerably rare, hampering our understanding of the evolutionary history of the biota from that continent. For several austral summers, the PALEOANTAR project has been carrying out fieldwork in the Antarctic Peninsula in search for fossils, particularly Cretaceous vertebrates. Among the specimens recovered so far are two bones referable to Pterosauria, more specifically to the Pterodacyloidea, the first volant reptiles from Antarctica to be fully described. MN 7800-V (part and counterpart) was recovered from a moraine at the Abernathy Flats (Santa Marta Formation, Lachman Crags Member, Santonian-Campanian) on James Ross Island. It is interpreted as the distal articulation of a first phalanx of the wing finger, representing an animal with an estimated wingspan between 3 and 4 m. The second specimen (MN 7801-V) comes from Vega Island (Snow Hill Island Formation, Maastrichtian) and is identified as a wing metacarpal IV of an animal with an estimated wingspan from 4 to 5 m. These occurrences show that pterodactyloids inhabited the Antarctic Peninsula at least during the Upper Cretaceous and demonstrate that large pterosaurs were widespread through all parts of the planet during that period.

Symptomatic ZIKV-infected individuals identified in two hospitals in Sao Paulo State, Brazil, were investigated regarding clinical characteristics, shedding in body fluids, and serodynamics. Ninety-four of 235 symptomatic patients (Site A: 58%; Site B: 16%) had Real-Time PCR-confirmed ZIKV infection; fever, headache and gastrointestinal symptoms were less frequent, and rash was more frequent compared to ZIKV-negative patients. Real-Time PCR in serum had worse performance compared to plasma, while urine had the highest sensitivity. Shedding in genital fluids and saliva was rare. IgM positivity was the highest <14 days after the symptoms onset (86%), decreasing >28 days (24%); IgG positivity increased >14 days (96%) remaining positive in 94% of patients >28 days. ZIKV prevalence varied importantly in two neighboring cities during the same transmission season. Urine Real-Time PCR can improve diagnostic sensitivity; serum testing is less useful. Accurate serological tests are needed to improve diagnosis and surveillance.

Abstract: The aim of this study was to evaluate in vitro the antifungal, antibiofilm and antiproliferative activities of the extract from the leaves of Guapira graciliflora Mart. The phytochemical characterization of the extract was performed using electrospray ionization mass spectrometry (ESI-MS). The antimicrobial activity of the extract and its fractions was evaluated using the broth microdilution method against species of Candida. The inhibition of C. albicans biofilm was evaluated based on the number of colony-forming units (CFU) and metabolic activity (MTT). The antiproliferative activity of the extract and its fraction was evaluated in the presence of human tumor and non-tumor cells, and the cytotoxicity of the extract was determined on the RAW 264.7 macrophage line – both using the sulforhodamine B method. The phytochemical characterization indicated the presence of the flavonoids rutin and kaempferol. The extract and the methanol fraction exhibited moderate antifungal activity against C. albicans, C. krusei, and C. glabrata, and strong activity against C. dubliniensis. In the biofilms at 24 and 48 hours, the concentration of 12500 µg/mL of the extract was the most effective at reducing the number of CFU s/mL (44.4% and 42.9%, respectively) and the metabolic activity of C. albicans cells (34.6% and 52%, respectively). The extract and its fractions had no antiproliferative effect on the tumor lines tested, with mean activity (log GI50) equal to or greater than 1.71 µg/mL. Macrophage cell viability remained higher than 80% for concentrations of the extract of up to 62.5 µg/mL. G. graciliflora has flavonoids in its chemical composition and demonstrates potential antifungal and antibiofilm activity, with no evidence of a significant change in the viability of human tumor and non-tumor cell lines.

Resumo: No período de janeiro de 2011 a dezembro de 2014 foram diagnosticados 9 surtos (A, B, C, D, E, F, G, H e I) de Oestrus ovis em pequenos ruminantes no estado da Bahia. No surto A obteve-se 0,5% (1/200); B 2,2% (2/90); C 0,8% (1/120); D 2% (2/100); E 1% (1/100); F 3% (1/33); G 0,6% (1/150); H 2,5% (5/200); I com 11,4% (8/70) em ovinos e 5% (2/40) em caprinos. Os sinais clínicos associados ao parasitismo pelas larvas nos surtos foram respiração ruidosa, espirro seguido de secreção nasal catarral, inquietação, movimentação excessiva da cabeça e andar em círculo. Macroscopicamente havia nos seios e conchas nasais hiperemia, edema da mucosa e presença de larvas. Todas as larvas coletadas dos cornetos e conchas nasais variavam desde o primeiro ao terceiro estágio de desenvolvimento. Algumas larvas L3 coletadas nas necropsias foram incubadas e o imago obtido das pupas mediram aproximadamente 10mm de cor acinzentada e abdômen escurecido. Realizada análise descritiva das condições climáticas, ano e positividade de casos de oestrose, demonstrou que a ocorrência tem tendência de crescimento com os anos (p˂0,001) e que houve casos com menor média de temperatura mínima (p˂0,001), possibilitando o desenvolvimento da mosca de O. ovis, demonstrando que houve a introdução da mosca enTtre o rebanho de ovinos e caprinos do estado da Bahia, e que as condições climáticas são ideais para perpetuação da espécie.
Abstract: From January 2011 to December 2014 were diagnosed 9 outbreaks of Oestrus ovis infection in small ruminants (Outbreaks A-I) in the State of Bahia. The incidence of oestrosis in sheep in outbreak A was 0.5% (1/200), in B 2.2% (2/90), in C 0.8% (1/120), in D 2% (2/100), in E 1% (1/100), in F 3% (1/33), in G 0.6% (1/150), in H 2.5% (5/200), and in I 11.42% (8/70), and 5% (2/40) in goats. Clinical signs associated with parasitism were wheezing, sneezing followed by catarrhal nasal secretion, some restlessness, excessive head movement and walking in circles. The breasts and turbinates were hyperemic, with mucosal edema and presence of O. ovis larvae. All larvae collected from the turbinates ranged from the first to the third stage of development. Some L3 larvae collected at necropsy were incubated and the gray colored Imago with dark abdomen obtained from the pupae measured about 10mm. A descriptive analysis of the climatic conditions was carried out; in the year of investigation the incidence of O. ovis infection has grown (p<0.001), and the lowest mean minimum temperature (p<0.001) caused the development the O. ovis fly, so that there was an introduction of an increased number of these flies into the sheep and goat flocks in state of Bahia with the ideal climatic conditions for their perpetuation.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Introdução: a Cecropia pachystachya Trécul., é conhecida como toré (Urticaceae), com ocorrência em todo o território brasileiro, usada na medicina popular no tratamento de infecções respiratórias e gonorreia. Métodos: o material vegetal (folhas e caule), coletado na Chapada do Araripe, foi triturado e submerso em solvente etanol PA, e submetido à destilação do solvente no aparelho evaporador rotativo para a produção dos extratos, que foi particionado e submetido ao solvente metanólico, fornecendo as frações metanólicas. Um ensaio de microdiluição foi realizado para verificar a atividade antibacteriana e as possíveis interações dos aminoglicosídeos associados às amostras estudadas, utilizando uma concentração sub-inibitória de 128 µg/mL (CIM/8). Resultados: o efeito mais representativo a associação da neomicina com o EEFCP (Extrato Etanólico das Folhas de C. pachystachya T) e sua fração com redução da CIM de 312.5 para 39,06 µg/mL e redução de 312,50 para 78,13 µg/mL, respectivamente ambos contra Staphylococcus aureus 358. Quando frente a E. coli 27, foi observado antagonismo. Conclusões: os dados obtidos indicam que o extrato e as frações metanólicas de C. pachystachya, podem ser uma alternativa relevante na pesquisa de substâncias que tenham ação contra as bactérias Staphylococcus multirresistentes. Entretanto, o mesmo também pode interferir negativamente na ação das drogas testadas. Contudo são necessários novos teste in vivo para melhor comprovar essa ação.
Introduction: Cecropia pachystachya Trécul, commonly known as 'toré' (Urticaceae) and distributed throughout Brazil, is used in folk medicine to treat respiratory and sexually transmitted infections such as gonorrhea. Objective: evaluate the antibacterial and modulatory activity on antibiotics of the ethanolic extract and methanolic fractions of C. pachystachya T. leaves and stems against standard and multiresistant bacterial strains. Methods: the plant material (leaves and stems) was collected in Araripe, crushed and put into ethanol PA as solvent. Distillation of the solvent was then performed in a rotary evaporator to produce the extracts, which were divided and subjected to the methanolic solvent to obtain the fractions. A microdilution assay was conducted to verify antibacterial activity and the possible interactions of aminoglycosides associated with the study samples. A sub-inhibitory concentration of 128 ?g/mL (MIC/8) was used. Results: the most representative effect was the association of neomycin with EEFCP (ethanolic extract of leaves of C. pachystachya T.) and its fraction, with a MIC reduction of 312.5 for 39.06 µg/mL and 312.50 for 78.13 µg/mL, respectively, against Staphylococcus aureus 358. An antagonistic effect was observed against E. coli 27. Conclusions: the data obtained indicate that C. pachystachya methanolic fractions and extract may be an important alternative in the search for substances with an action against multiresistant Staphylococcus bacteria. However, such a substance may also negatively affect the action of the drugs studied. It is necessary to conduct new experiments in vivo to verify such an action.
Introducción: Cecropia pachystachya Trécul., es conocida como "toré" (Urticaceae), presente en todo el territorio brasileño, utilizada en la medicina popular para el tratamiento de infecciones respiratorias y de transmisión sexual (gonorrea). Objetivo: evaluar la actividad antibacteriana y moduladora sobre antibióticos del extracto etanólico y fracciones metanólicas de las hojas y tallo de C. pachystachya T. contra cepas bacterianas estándar y multirresistentes. Métodos: el material vegetal (hojas y tallos), colectado en el Araripe, fue triturado y sumergido en etanol PA como solvente. Seguidamente, fue sometido a destilación del solvente en un evaporador rotativo para la producción de los extractos, que fueron divididos y sometidos al solvente metanólico, formando así las fracciones. Se llevó a cabo un ensayo de microdilución para verificar la actividad antibacteriana y las posibles interacciones de aminoglucósidos asociados con las muestras estudiadas, se utilizó una concentración sub-inhibitoria de 128 µg/mL (CIM/8). Resultados: el efecto más representativo resultó el de la asociación de neomicina con el EEFCP (Extrato Etanólico de hojas de C. pachystachya T) y su fracción con disminución de la CIM de 312,5 para 39,06 µg/mL y 312,50 para 78,13 µg/mL, respectivamente contra Staphylococcus aureus 358. Se observó un efecto antagónico contra E. coli 27. Conclusiones: los datos obtenidos indican que el extracto y fracciones metanólicas de C. pachystachya, pueden ser una alternativa importante en la búsqueda de sustancias que tienen acción contra las bacterias Staphylococcus multirresistentes. Sin embargo, puede afectar negativamente a la acción de las drogas estudiadas. No obstante, es necesario realizar nuevos experimentos in vivo para comprobar esta acción.

The aim of this study was to assess the activity of essential oil extracted from the leaves of C. blanchetianus Baill, popularly known as "marmeleiro", in inhibiting the growth and survival of pathogenic microorganisms in food by determining their survival in vitro and by observing the behaviour of Listeria monocytogenes inoculated into a food model (meat cubes) that was stored at refrigeration temperature (7 ± 1 ºC) for 4 days. The results indicated a bactericidal effect against Aeromonas hydrophila and Listeria monocytogenes and bacteriostatic action against Salmonella Enteritidis. A bacteriostatic effect on meat contaminated with L. monocytogenes was found for all concentrations of essential oils tested. These results showed that essential oil from the leaves of C. blanchetianus Baill represents an alternative source of potentially natural antimicrobial agents that may be used as a food preservative.

OBJETIVO: Dermatite atópica (DA) é uma doenc¸a inflamatória crônica com prurido intenso e características clínicas típicas. Há poucos estudos epidemiológicos a respeito da DA em adultos, bem como pouca informação disponível sobre o seu prognóstico. O objetivo do presente estudo é avaliar as características clínicas e o curso epidemiológico dos adultos com DA. MÉTODOS: Foramselecionados 80 pacientes com idade acima de 18 anos (média de idade = 29 anos, 30 homens e 50 mulheres), que foram entrevistados sobre: internações, uso de corticóide sistêmico, idade de início da DA, história pessoal e/ou familiar de atopia. A gravidade da doença foi avaliada de acordo com o SCORing Atopic Dermatitis (SCORAD). A avaliação laboratorial incluiu dosagem sérica de IgE e contagem sanguínea de eosinófilos. RESULTADOS: 71 dos 80 pacientes referiram associação com sintomas respiratórios (18: asma, 17: rinite alérgica e 36: ambas as condições); nove dos 80 indivíduos negaram qualquer sintoma respiratório. Os pacientes com DA foram divididos em DA leve (n = 25), moderada (n = 30) e grave (n = 25); destes, 56% tiveram uma ou mais internações por conta da doença. Verificou-se uma associação entre níveis séricos de IgE, contagem sanguínea de eosinófilos e gravidade da doença. CONCLUSÃO: A DA do adulto representa um desafio clínico que necessita ser melhor caracterizado, uma vez que pode ser erroneamente diagnosticada, e interfere diretamente na vida social e pessoal dos pacientes. A associação entre manifestação respiratória e cutânea é frequente, e parâmetros laboratoriais como níveis de IgE circulante e contagem sanguínea de eosinófilos podem ser úteis para acompanhar a gravidade e evolução da doença.
OBJECTIVE: Atopic dermatitis (AD) is a chronic inflammatory disease causing intense pruritus, and with typical clinical features. There are few epidemiological studies concerning AD in adults, as well as little information about its prognostic. The aim of this study was to evaluate the clinical and epidemiological course of adults with AD. METHODS: 80 patients aged above 18 years (mean age = 29 years) were selected (30 males and 50 females) and interviewed about hospitalization, systemic corticoid usage, age of AD onset, and personal and/or familial history of atopy. Disease severity was evaluated through the Scoring Atopic Dermatitis (SCORAD) tool. Laboratory examination included IgE serum levels and eosinophil blood count. RESULTS: 71 out of 80 patients referred association with respiratory symptoms (18 had asthma, 17 had rhinitis, and 36 had both conditions); nine out of 80 patients denied any respiratory disease. AD patients were divided in mild (n = 25), moderate (n = 30), and severe (n = 25); 56% had one or more hospitalizations due to AD. A positive association was found between IgE serum levels, eosinophil blood count, and disease severity. CONCLUSION: Adult AD represents a clinical challenge that needs to be better characterized, since it can be misdiagnosed and interferes with the patient's social and personal life. The association of skin and respiratory atopic disease is frequent, and laboratory parameters such as circulating IgE levels and eosinophil blood count may be helpful to assess disease severity.

Mechanism involved in the anti-inflammatory effect of Spiranthera odoratissima (Manacá)

Acetic acid-induced writhing, hot-plate, carrageenan-induced pleurisy, formalin-induced pain, croton oil-induced ear edema, vascular permeability tests and phospholipase A2 activity assay were used to study the analgesic and/or anti-inflammatory activity of the hydromethanolic fraction of ethanolic extract from Spiranthera odoratissima A. St.-Hil., Rutaceae, leaves (HMF) and its subfraction (sub-Fr10-28). HMF and sub-Fr10-28 reduced the leukocyte migration on the carrageenan-induced pleurisy test; sub-Fr10-28 reduced the pain reaction time in the second phase of formalin-induced pain, as well as the ear edema and vascular permeability. Both HMF and sub-Fr10-28 inhibited the phospholipase A2 activity. These results suggest that the analgesic effect of this plant could be, in part, due to an anti-inflammatory action produced by the inhibition of phospholipase A2 activity.

A investigação fitoquímica dos frutos maduros de Solanum caavurana Vell. (Solanaceae) forneceu um novo alcaloide esteroidal do tipo espirosolano, caavuranamida, juntamente com os alcaloides 4-tomatiden-3-ona e 5α-tomatidan-3-ona. Suas estruturas foram elucidadas através de métodos espectroscópicos. As atividades antiproliferativa e antimicrobiana para o extrato etanólico e subfrações provenientes de partição e de extração ácido-base foram investigadas. A caavuranamida apresentou atividade antibacteriana similar ao controle positivo cloranfenicol contra Rhodococcus equi.
Phytochemical investigation of the ripe fruits of Solanum caavurana Vell. (Solanaceae) afforded a novel steroidal alkaloid with spirosolane-type skeleton, named as caavuranamide, together with the alkaloids 4-tomatiden-3-one and 5α-tomatidan-3-one. Their structures were elucidated on the basis of spectroscopic methods. The antiproliferative and antimicrobial activities for the ethanolic extract, sub-fractions obtained from partition and acid-base treatment were also evaluated. Caavuranamide showed antibacterial activity similar to the chloramphenicol positive control against Rhodococcus equi.
